The parent Escherichia coli K-12 is constitutive for the enzymes of the glyoxylate bypass and adapts to growth on long-chain fatty acids (C(12) to C(18)). It does not utilize medium-chain (C(6) to C(11)) or short-chain (C(4), C(5)) n-monocarboxylic acids. Several mutants of this strain which grow using short- or medium-chain acids, or both, as the sole carbon source were selected and characterized. One mutant (D(1)) synthesizes the beta-oxidation enzymes constitutively and grows on medium-chain but not on short-chain acids. A second (N(3)) is partially derepressed for synthesis of these enzymes and grows both on medium-chain and on short-chain acids. Secondary mutants (N(3)V(-), N(3)B(-), N(3)OL(-)) were derived from N(3). N(3)V(-) grows on even-chain but not on odd-chain acids and exhibits a lesion in propionate oxidation. N(3)B(-) grows on odd-chain but not on even-chain acids and exhibits no crotonase activity as assayed by hydration of crotonyl-CoA. N(3)OL(-) grows on acetate and propionate but does not utilize fatty acids C(4) to C(18); it exhibits multiple deficiencies in the beta-oxidation pathway. Growth on acetate of N(3), but not of the parent strain, is inhibited by 4-pentenoate. Revertants of N(3) which are resistant to growth inhibition by 4-pentenoate (N(3)PR) exhibit loss of ability to grow on short-chain acids but retain the ability to grow on medium-chain and long-chain acids. The growth characteristics of these mutants suggest that in order to grow at the expense of butyrate and valerate, E. coli must be (i) derepressed for synthesis of the beta-oxidation enzymes and (ii) derepressed for synthesis of a short-chain fatty acid uptake system.  相似文献

Optical measurements from epithelial cells grown on clear solid surfaces (e.g., coverslips, petri dishes) are often compared with other measurements (e.g., short-circuit current; I(sc)) obtained from cells grown on opaque porous surfaces (inserts). However, the relative levels of differentiation of cells grown under the two conditions are usually unknown. To address this issue, we grew primary cultures of human tracheal epithelium on solid surfaces or on porous inserts and compared their total levels of protein and deoxyribonucleic acid, electrical properties in Ussing chambers, and ultrastructure. To measure ion transport across cells grown on solid supports, cells were grown on inserts placed on parafilm. Later, separation of insert from parafilm allowed the cells' I(sc) to be measured in Ussing chambers. Four different media were used. Cells grown in one medium showed very low levels of differentiation on all growth supports. In the other media, growth on inserts markedly enhanced differentiation as compared with solid supports. Baseline I(sc) of cells grown on either clear or opaque inserts was at least 30 times greater than that of cells grown on solid supports, though I(sc) with clear inserts averaged approximately 30% lower than that with opaque inserts. We conclude that though differentiation of cells may vary slightly depending on the insert used, cells on any type of insert are much better differentiated than cells grown on solid surfaces. Thus, it is both possible and desirable to make all functional measurements on cells grown on clear porous supports.  相似文献

The influence of tocopherol and its analogue (oxychroman) on the microviscosity of mitochondrial lipids was studied, using spin labels. The viscosity of the lipid bilayer was shown to enhance with the increase in the antioxidant content in the membrane. Small concentrations of alpha-tocopherol (10(-5)-10(-6) mol/l) were shown to increase, while large concentrations (10(-3)-10(-4) mol/l) decreased the fluidity of the lipid bilayer. The influence of alpha-tocopherol on fluidity of the lipid bilayer depending on its concentration could be realized in two ways: by direct influence on the lipid bilayer and via reception. It was shown that alterations in the viscosity of the lipid bilayer depend on chroman cycle of tocopherols, while the temperature of structural transfer and effective energy of activation depend on the lateral phytyl chain.  相似文献

Conidia of a new pathotype of Bipolaris zeicola (Stout) Shoemaker, which causes Helminthosporium corn leaf spot (HCLS) on inbreds derived from B73, are morphologically atypical on potato dextrose agar (PDA). The average conidial length on PDA (31.9 μm) is half that on naturally infected leaf (65.2 μm). Conidiogenuous cells terminally and subterminally located on short conidiophores produce new conidia which behave as initial conidia, i.e. they immediately elongate or germinate. Sometimes, they appear in chains. Based on conidial morphology on leaf materials and on wheat straw agar (WSA), the investigated fungus was identified as B. zeicola.  相似文献

研究了化学杀虫剂对6种不同类型棉田的害虫、天敌种群数量及其相互作用的影响。结果表明,化学防治使棉花苗期棉蚜数量下降,但不能抑制中后期棉蚜数量;除豆间棉田外,其他类型棉田化防区棉红蜘蛛和棉铃虫数量低于未防区。化学防治使各类型棉田天敌数量减少,以单作棉田天敌数量减少最明显,间套作棉田天敌数量下降幅度较小;此外,棉田不同天敌种群所受的影响存在一定差异,化学防治对瓢虫、捕食蝽类的影响显著,数量下降剧烈,但蜘蛛类群下降幅度比前两者小。化学防治对棉田害虫与天敌的相互关系的影响随棉田类型或害虫、天敌种类的不同而有所差异。  相似文献

Abstract:  The influence of three solanaceous plants (tomato, sweet pepper and eggplant) on the functional response of the predatory bug Picromerus bidens to densities of fourth-instar larvae of the beet armyworm Spodoptera exigua was assessed. Logistic regression indicated a type II functional response on all host plants. Over all prey densities, P. bidens killed significantly fewer fourth instars of S. exigua on tomato than on sweet pepper or eggplant (1.96 ± 0.17 vs. 4.37 ± 0.19 and 3.90 ± 0.15 larvae per predator per 24 h respectively). A higher theoretical maximum predation rate was estimated on sweet pepper (11.1 prey larvae per day) and eggplant (7.4) than on tomato (5.4). The mean number of prey killed per day by P. bidens females ranged from 0.78 at a density of one prey on tomato to 8.45 at a density of 24 prey on sweet pepper. The data indicated that the estimates of handling time ( T h) and attack rate ( a ) were highly affected by host plant. Based on asymptotic 95% confidence intervals, a lower attack coefficient was found on tomato (0.02 h−1) than on sweet pepper or eggplant (0.07 and 0.11 h−1 respectively). On the other hand, handling times were significantly longer on tomato (4.42 h) and eggplant (3.23 h) than on sweet pepper (2.15 h). This laboratory study suggests that plant characteristics influence the ability of P. bidens to respond to changes in prey density.  相似文献

Samples of maternal and fetal placental tissues were obtained from cows on Days 100 (N = 4), 150 (N = 5), 200 (N = 6) and 250 (N = 6) of gestation and incubated for 24 h. Conditioned media from caruncular explants were mitogenic for bovine aortic endothelial cells (BAEC) on all days of gestation. Media from intercaruncular endometrium were stimulatory for proliferation of BAEC on Day 100 but inhibitory on Days 150, 200 and 250. Media from cotyledonary and intercotyledonary tissues inhibited proliferation of BAEC on all days. Caruncular-conditioned media stimulated migration of BAEC on Days 150, 200 and 250. Cotyledonary-conditioned media inhibited migration of BAEC on all days. Effects of media from intercaruncular and intercotyledonary tissues on migration of BAEC varied with stage of gestation. Angiogenic activity of media from caruncular (all stages) and intercaruncular (Day 100) tissues appeared to have an Mr greater than 100,000. In cows, therefore, the maternal placentome (caruncle) appears to be the primary source of placental angiogenic activity throughout gestation. The fetal placentome (cotyledon) secretes activity which inhibits two major components of angiogenesis (proliferation and migration of endothelial cells) throughout gestation. Intercaruncular and intercotyledonary tissues may modulate placental angiogenesis throughout gestation. Placental vascular development in the cow is therefore probably controlled by an interaction between stimulatory and inhibitory factors produced by the placenta itself.  相似文献

Summary Plant-uptake and yield data for ryegrass in a greenhouse experiment are used to estimate the theoretical fertilizer phosphate requirement (Pf) of 24 Sherborne soils. Pf is shown to be a function of three parameters: (i) quantity of P required by the plant (Pr) for optimum yield; (ii) quantity of soil P (Qr) required to maintain a non-limiting soil solution concentration (Ir); (iii) quantity of labile soil P (Q). Because of its large effect on Qr and Ir, the phosphate buffer capacity has an important effect on Pf. However Pf cannot be directly related to phosphate buffer capacity if Q is ignored. On soils of similar Q, increasing buffer capacity will always have a positive effect on Pf, but on soils of the same I, it may have a positive or negative effect on Pf. Many physiological functions of the stomach depend on an intact mucosal integrity; function reflects structure and vice versa. Histamine in the stomach is synthesized by histidine decarboxylase (HDC), stored in enterochromaffin-like (ECL) cells, and released in response to gastrin, acting on CCK(2) receptors on the ECL cells. Mobilized ECL cell histamine stimulates histamine H(2) receptors on the parietal cells, resulting in acid secretion. The parietal cells express H(2), M(3), and CCK(2) receptors and somatostatin sst(2) receptors. This review discusses the consequences of disrupting genes that are important for ECL cell histamine release and synthesis (HDC, gastrin, and CCK(2) receptor genes) and genes that are important for "cross-talk" between H(2) receptors and other receptors on the parietal cell (CCK(2), M(3), and sst(2) receptors). Such analysis may provide insight into the functional significance of gastric histamine.  相似文献

Significantly more species of freshwater hyphomycetes colonized bait leaves of alder (16 species) and willow (14 species) during early submersion period (1 wk) than those of oak (8 species). Production of conidia was also higher on alder and willow leaves than on oak leaves. Higher amounts of dry mass were lost from the alder (28.6%) and willow leaves (29.4%) than from the oak leaves (18.7%). Rapid loss of dry mass was accompanied by rapid development of a fungal community. Fungal species took a longer time (6 wk) to reach their peak of occurrence on oak than on willow and alder leaves (3–4 wk).Flagellospora curvula dominated the assemblages of freshwater hyphomycetes on alder and willow leaves, andLunulospora curvula dominated the assemblage on oak leaves. The freshwater hyphomycete community showed higher values of species diversity (H value) on oak leaves than on alder and willow leaves.  相似文献

Laboratory studies with Neomegalotomus parvus(Westwood) (Hemiptera: Alydidae) with one nymph per Petri dish in multiple-choice tests indicated that seeds of pigeon pea [Cajanus cajan(L.) Mills.], lablab (Dolichos lablabL.), and soybean [Glycine max(L.) Merrill] were visited before seeds of common bean (Phaseolus vulgarisL.) and rice (Oryza sativaL.). The percentage of individuals engaging in dabbing/antennation resulting in probing, and percentage probing resulting in feeding, were higher on common bean (97%) and pigeon pea (87%) seeds than on lablab (55%), soybean (50%), or rice (5%) seeds. No significant differences were found in preference (number of flanges) among pigeon pea, common bean, and lablab, and preference (insects on foods) varied throughout the assessment period (5 d). In tests using 10 nymphs per dish, pigeon pea was the preferred food (number of flanges and insects on plants) throughout the period (5 d). In no-choice tests, the average duration of a feeding session and the longest feeding session were greater on lablab and common bean than on pigeon pea, soybean, or rice seeds. The number of feeding sessions was greater on seeds of common bean, pigeon pea, and soybean than on those of lablab or rice. Laboratory tests with N. parvusadults indicated that pigeon pea seeds were located faster, followed by common bean, soybean, and rice. When pods were tested, dabbing/antennation time was shorter on pigeon pea than on soybean, and probing time was longer on soybean than on pigeon pea or common bean. On pigeon pea, 100% of the insects probed the host, while on common bean and soybean pods, and on rice panicles, these values dropped to 71.8%, 46.0%, and 10.5%, respectively. Adults showed similar feeding times on pigeon pea, common bean, and soybean pods, but did not feed on rice panicles. Electronmicroscopical analysis showed the presence of two apical lobes with 12 peg sensilla on the labial tip. Sensillum tips were stained with silver nitrate solution, indicating a permeability of the cuticle and, therefore, their function as taste receptors.  相似文献

Biological activity of lipid fractions of actinomycetes was examined on microorganisms and laboratory animals. The anabolic effect of the acetone fraction of Actinomyces aurigineus 2377 (on Wistar female rats), anabolic effect of sterols of Actinomyces griseus 15 (on chicks), and the high growth stimulating effect (up to 40-60%) of sterols of Actinomyces canosus 89 (on rats) were noted. The estrogenic effect of sterols of Act. griseus 15, Act. griseus 20 and Act. albodenitrificans 13a on the sex system was shown. The antimicrobial properties of lipid fractions differed in their selectivity with respect to the test organisms and depended also on the actinomycete species and cultivation conditions.  相似文献

TRH (10 and 1000 micrograms/kg body weight (BW] was injected ip into pregnant rats daily from day 0 to 20 of pregnancy, and the pituitary-thyroid axis of their pups (Mat-TRH rats) was examined on days 0, 4, 10, 21 and 90 after birth. The pituitary TSH content of male Mat-TRH rats was significantly lower on day 4, and higher on day 10 than that of control rats. The serum TSH was significantly higher on day 10 (except female 10 micrograms/kg group). An exaggerated TSH response to exogenous TRH (10 micrograms/kg BW; ip) was observed on day 10 (males, 1000 micrograms/kg group). The serum T4 level of female Mat-TRH rats was low on day 4 (1000 micrograms/kg group), and higher on day 10. On days 21 and 90, the levels of pituitary TSH, serum TSH and T4 in Mat-TRH rats were similar to those in controls, but the TSH response to TRH was still exaggerated (1000 micrograms/kg group). No significant difference between control and TRH-treated mothers was seen on days 10 and 90 postpartum except for a decreased pituitary TSH content on day 10 in the 1000 micrograms/kg group. It is concluded that repeated administration of TRH to pregnant rats shows an effect on the pituitary-thyroid axis function of their progeny in later life.  相似文献

A modifying effect of a catecholamine precursor (L-DOPA, 20 mg/kg, intraperitoneally) on various stages (initiation and promotion) of hepatocarcinogenesis, induced by N-nitrosodiethylamine (NDEA, 85 mg per 1 litre of tap water) was studied in chronic experiments on 150 male rats. L-DOPA administration prior to NDEA (influence on initiation) stimulated hepatocarcinogenesis considerably, while its administration after NDEA (influence on promotion) inhibited the realization of a carcinogenic effect. A statistically significant decrease in noradrenaline hypothalamus content was identified during early stages of chemically induced neoplastic transformation of hepatic cellular elements (stages of diffuse and focal proliferation). The results are discussed in terms of the regulatory role of the tone of the adrenergic autonomous nervous system component at the level of presynaptic inhibitory receptors in realization of the chemical carcinogenic effect.  相似文献

This investigation is concerned with assignments of Lewis(a) (Le(a)) and Le(x) analogs on linear and branched di- to hexasaccharide backbones as components of the recognition motifs for E-selectin. The influence of the location of fucose residue(s) was investigated using 14 structurally defined and variously fucosylated oligosaccharides in biotinylated form or as neoglycolipids in static binding assays, in microwells, and on thin-layer chromatograms. Results of the two assay systems were in agreement overall and showed that the recognition motifs for E-selectin include 4-fucosyl-lacto (Le(a)) and 3-fucosyl-neo-lacto (Le(x)) sequences strictly at capping positions and not Le(x) at an internal position as a part of VIM-2 antigen sequence. There is greater potency of the Le(a) over the Le(x) series. Additional fucose residues alpha1-2-linked to neighboring galactoses or alpha1-3-linked to inner N-acetyglucosamines or to reducing-terminal glucose residues of the tetrasaccharide backbone had little or no effect on the selectin binding. E-selectin binding to the Le(a) or Le(x )capping motif on a 3-linked branch was equivalent to the binding on the corresponding linear backbone. A lack of E-selectin binding to the Le(x) motif capping a 6-linked branch and to the Le(x) trisaccharide linked to biotin via a nine-carbon spacer indicates that the -GlcNAcbeta1-3Gal- sequence on the oligosaccharide backbone adjoining the Le(x) is a part of recognition motif for E-selectin. These findings contribute to understanding the molecular basis of E-selectin recognition and could influence future designs of selectin antagonists as possible therapeutic substances.  相似文献

Initially reserved for rehabilitation programs, unstable surface training (UST) has recently grown in popularity in strength and conditioning and general exercise scenarios. Nonetheless, no studies to date have examined the effects of UST on performance in healthy, trained individuals. The purpose of this study was to determine the effects of 10 weeks of lower-body UST on performance in elite athletes. Nineteen healthy, trained members (ages 18-23 years) of a National Collegiate Athletic Association Division I collegiate men's soccer team participated. The experimental (US) group (n = 10) supplemented their normal conditioning program with lower-body exercises on inflatable rubber discs; the control (ST) group (n = 9) performed the same exercises on stable surfaces. Bounce drop jump (BDJ) and countermovement jump (CMJ) heights, 40- and 10-yard sprint times, and T-test (agility) times were assessed before and after the intervention. The ST group improved significantly on predicted power output on both the BDJ (3.2%) and CMJ (2.4%); no significant changes were noted in the US group. Both groups improved significantly on the 40- (US = -1.8%, ST = -3.9%) and 10-yard sprint times (US = -4.0%, ST = -7.6%). The ST group improved significantly more than the US group in 40-yard sprint time; a trend toward greater improvement in the ST group was apparent on the 10-yard sprint time. Both groups improved significantly (US = 2.9%, ST = -4.4%) on T-test performance; no statistically significant changes were apparent between the groups. These results indicate that UST using inflatable rubber discs attenuates performance improvements in healthy, trained athletes. Such implements have proved valuable in rehabilitation, but caution should be exercised when applying UST to athletic performance and general exercise scenarios.  相似文献

Tunicamycins (TCMs) and liposidomycins (LPMs) are naturally occurring inhibitors of the bacterial translocase (MraY). Based on structure-activity relationship (SAR) studies, a molecular model has been proposed for their inhibitory mechanism. This study points out the importance of the nucleoside moiety of liposidomycins in the inhibition of MraY. A simplified molecule (I) based on the liposidomycin core structure has been synthesised and tested on MraY. The compound displayed a moderate inhibitory activity (IC50 = 50 microM). The validation of the molecular model was then performed by synthesising higher homologues of I, containing an additional stereocentre in the 5' position (XIV and XV). In agreement with the prediction, only the (S) isomer XV showed significant activity against MraY (IC50 = 5 microM).  相似文献

在25℃恒温条件下,以盆栽玉米、小麦、高粱、水稻、谷子、大麦及虮子草为食料,研究了食物对二点叶蝉实验种群生长发育及繁殖的影响.结果表明,二点叶蝉在虮子草上不能完成世代发育.在其余6种寄主植物上各虫态的发育历期、存活率、雌成虫寿命及单雌平均产卵量存在显著差异.从卵到若虫期的发育历期在高粱上最短(24.1d),玉米上次之(24.2d),小麦上最长(25.5d);取食水稻时。若虫的存活率(40.8%)最低,成虫寿命最短(12.2d),单雌平均产卵量(12.3粒)也显著低于其它5种寄主植物.用生命表参数综合评价表明,6种寄主植物中,谷子和玉米最适合二点叶蝉生长发育及繁殖。其次分别为高粱、大麦、小麦、水稻。  相似文献
