The effect of sheath loss on motility and pathogenicity of the entomopathogenic nematodes, Heterorhabditis bacteriophora and Steinernema carpocapsae, was examined using both naturally and chemically exsheathed (desheathed) infective juveniles. Exsheathed S. carpocapsae showed increased motility on agar compared to sheathed nematodes. The presence of a host increased motility threefold in all S. carpocapsae treatments. These results suggest that activation of S. carpocapsae host finding may result from sheath loss in addition to host stimuli. Desheathed H. bacteriophora were significantly less motile than the sheathed or exsheathed groups. The decreased motility may be due to adverse effects of the chemical treatment for desheathment. Sheath loss did not affect the pathogenicity of either species. 相似文献

Utility of Copulatory Structures of
Selected Nematode Species
Spicules of 9 Meloidogyne, 2 Heterodera, 3 Globodera, and 12 other plant-parasitic, insect-parasitic, and free-living nematodes were excised and examined using scanning electron microscopy (SEM). Gubernacula of some of the species were also excised, and their structure was determined. The two spicules of all species examined were symmetrically identical in morphology. The spicule typically consisted of three parts: head, shaft, and blade with dorsal and ventral vela. The spicular nerve entered through the cytoplasmic core opening on the lateral outer surface of the spicule head and generally communicated with the exterior through one or two pores at the spicule tip. Spicules of Xiphinema sp. and Aporcelaimellus sp. were not composed of three typical parts, were less sclerotized, and lacked a cytoplasmic core opening and distal pores. Spicules of Aphelenchoides spp. had heads expanded into apex and rostrum and had very arcuate blades with thick dorsal and ventral edges (limbs). Gubernaculum shapes were stable within a species, but differed among species examined. The accessory structures of Hoplolaimus galeatus consisted of a tongue-shaped gubernaculum with two titillae at its distal end and a plate-like capitulum terminating distally in two flat, wing-like structures. A comparison of spicules of several species of Meloidogyne by SEM and light microscopy revealed no striking morphological differences. 相似文献

The majority of parasitological studies of terrestrial snakes in Korea have focused on zoonotic parasites. However, in the present study, we describe 3 unrecorded nematode species recovered from 5 species of snakes (n=6) in Korea. The examined snakes, all confiscated from illegal hunters, were donated by the Chungnam Wild Animal Rescue Center and Korean Broadcasting System in July 2014 and February 2015. Light and scanning electron microscopies on the shapes of spicules that are either bent or straight (kalicephalids) and the presence of the intestinal cecum (ophidascarids) figured out 3 nematodes; Kalicephalus brachycephalus Maplestone, 1931, Kalicephalus sinensis Hsü, 1934, and Ophidascaris excavata Hsü and Hoeppli, 1934. These 3 species of nematode faunas are recorded for the first time in Korea. 相似文献

Mature trees of eastern white, jack, Scotch, and shortleaf pines were inoculated with 25,000-34,000 pinewood nematodes, Bursaphelenchus xylophilus, isolated from infected Scotch pines in Missouri. Equal numbers of trees of each species inoculated with distilled water served as controls. Nine of fifteen Scotch pines died within 4 months of nematode infection or during the winter and early spring following infection. A single eastern white and shortleaf pine died. No jack pines died. A single Scotch pine control died, apparently the result of natural nematode infection. No other controls died. Mean oleoresin flow did not differ among nematode-inoculated jack and shortleaf pines and their respective controls. Oleoresin flow in nematode-inoculated eastern white and Scotch pines was significantly lower than in their controls. Oleoresin flow was temporarily reduced in mortality-resistant eastern white and Scotch pines following nematode infection. Thus a sublethal impact of nematode infection on mortality-resistant host trees was documented. 相似文献

Water from a major irrigation canal and water from a deep well was used to irrigate plants growing in methyl bromide fumigated screenhouse ground beds. Nematode populations in these beds were compared during three seasons of continuous cropping to alfalfa, bean, eggplant, mint, sugarbeet, or wheat. Beds irrigated with canal water became heavily infested with a variety of plant parasitic nematodes while those receiving well water did not. 相似文献

The effects of perennial peanut (Arachis glabrata) ground cover on the nematode community in a citrus orchard were examined. Samples were taken from two different ground cover treatments (perennial peanut or bare ground) at each of three distances from the tree trunk. Richness, measured as total numbers of nematode genera per sample, and total numbers of nematodes were greatest in the perennial peanut treatment (P < 0.05). Abundance of many genera of bacterivores, fungivores, and omnivores were increased by the perennial peanut ground cover. Total numbers of plant parasites were greater in perennial peanut treatments on three of the five sampling dates (P < 0.05), mainly due to trends in numbers of Mesocriconema. Distance from a tree trunk and the interaction of ground cover treatments and proximity to a tree trunk were most influential for Belonolaimus and Hoplolaimus. Although differences among treatments were observed for nematode genera and trophic groups, ecological indices were not consistently sensitive to treatments. Among several ecological indices evaluated, richness was most often affected by ground cover treatment. 相似文献

Field experiments were conducted in 1982 and 1983 to assess interactions between Heterodera glycines and Pratylenchus scribneri on soybean in southern Illinois. Soybean cyst nematode susceptible cultivar Williams 79 and resistant cultivar Fayette were treated or not treated with aldicarb 15G. Initial population densities were 35 H. glycines cysts containing eggs, 100 P. scribneri, 30 Helicotylenchus pseudorobustus, 225 Paratylenchus projectus, and 85 Tylenchorhynchus martini per 250 cm³ soil in 1982, whereas in 1983 populations were 11 H. glycines cysts, 330 P. scribneri, and 620 H. pseudorobustus. In both years H. glycines populations increased on nontreated Williams 79, decreased on both treated and nontreated Fayette, and remained at initial levels on treated Williams 79. Recovery of P. scribneri per gram dry root was different between nontreated cultivars in 1982 but not in 1983. Aldicarb treatment suppressed soil and root populations of P. scribneri on both cultivars in both years. Populations of H. pseudorobustus, P. projectus, and T. martini at harvest indicated little population increase on either nontreated cultivar. In 1982 H. glycines caused yield suppression but P. scribneri did not, as differences in yield occurred between cultivars but not between aldicarb treatments. In 1983, however, there were no yield differences between cultivars, but aldicarb application resulted in yield increase in both cultivars. In 1983 the yield increase resulting from P. scribneri control was approximately 25%. No synergistic effect on yield was observed between H. glycines and P. scribneri. 相似文献

We have adapted the Sherlock® Microbial Identification system for identification of plant parasitic nematodes based on their fatty acid profiles. Fatty acid profiles of 12 separate plant parasitic nematode species have been determined using this system. Additionally, separate profiles have been developed for Rotylenchulus reniformis and Meloidogyne incognita based on their host plant, four species and three races within the Meloidogyne genus, and three life stages of Heterodera glycines. Statistically, 85% of these profiles can be delimited from one another; the specific comparisons between the cyst and vermiform stages of H. glycines, M. hapla and M. arenaria, and M. arenaria and M. javanica cannot be segregated using canonical analysis. By incorporating each of these fatty acid profiles into the Sherlock® Analysis Software, 20 library entries were created. While there was some similarity among profiles, all entries correctly identified the proper organism to genus, species, race, life stage, and host at greater than 86% accuracy. The remaining 14% were correctly identified to genus, although species and race may not be correct due to the underlying variables of host or life stage. These results are promising and indicate that this library could be used for diagnostics labs to increase response time. 相似文献

Nine species of mosquitoes and several species of non-target aquatic organisms were tested for susceptibility to the mernaithid nematode, Strelkovimermis spiculatus. All species of Anopheles, Aedes, Culex, and Toxorhynchites exposed to S. spiculatus were susceptible. Of the nine mosquito species tested, C. pipiens quinquefasciatus had the greatest tolerance to initial invasion and the highest percent infection of those that survived. High levels of infection were also achieved with Aedes taeniorhynchus and A. albopictus, but these mosquitoes were significantly less tolerant to parasitism than C. pipiens quinquefasciatus. Temperature gradient fluctuations that occur naturally as a result of heating and cooling of the soil surface were reproduced within 15-cm-d, 15-cm-long acrylic tubes filled with moist sand. Sunny and rainy periods during the late summer in eastern Texas were simulated. Five ecologically different nematode species were adapted to fluctuating temperatures for 20-36 hours at a simulated depth of 12.5 cm before being injected simultaneously into the centers of tubes at that depth. When heat waves were propagated horizontally to eliminate gravitational effects, the movement of Ditylenchus phyllobius, Steinernema glaseri, and Heterorhabditis bacteriophora relative to the thermal surface was rapid and largely random. However, Rotylenchulus reniformis moved away from and Meloidogyne incognita moved toward the thermal surface. When heat waves were propagated upward or downward, responses to temperature were the same as when propagated horizontally, irrespective of gravity. The initial direction of movement 1.5 hours after introduction to 20-era-long tubes at five depths at five intervals within a 24-hour cycle indicated that M. incognita moved away from and R. reniformis moved toward the temperature to which last exposed. Differences in movement of the five species tested relative to gravity appeared related to body length, with the smallest nematodes moving downward and the largest moving upward. 相似文献

This paper is the second in a series studying procedures for estimating and calibrating features of nematodes from digital images. Two kinds of features were analyzed for recognition: those with a directional component and those with a textural component. Features that have a directional component (lateral field and annules) were preprocessed with classic algorithms and modified by directional filters. Three Ektaphelenchus species with unusual spicules are redescribed. Ektaphelenchus obtusus and E. riograndensis are redescribed from newly discovered material in the collection of the USDA Forest Service RMRS. Lectotype slides are designated for both species. Ektaphelenchus obtusus is characterized by a set-off head, lips of unequal size, double rows of oocytes and spermatocytes, and a hooked spicule terminus. Ektaphelenchus riograndensis is distinguished by a rounded head with six equal lips, and spicules with a large, rounded apex, and recurved terminus. A bursa and gubernaculum are absent in both species. Ektaphelenchus scolyti is redescribed from type material deposited in the collection of Rothamsted Experimental Station. It is characterized by a well set-off head, double rows of oocytes and spermatocytes, and spicules with a hooked terminus. A bursa and gubernaculum are absent. 相似文献

Field experiments were conducted in 1989 and 1990 to examine the population fluctuation patterns of Tylenchorhynchus nudus, Criconemella curvata, and Helicotylenchus cornurus in mixed bentgrass and annual bluegrass putting greens on two golf courses near Chicago, Illinois, to determine if fluctuation patterns could be extrapolated to unsampled greens. Fenamiphos-treated and untreated plots were established on seven putting greens on two golf courses. Greens were sampled intensively five times during the growing season, and statistical comparisons of population levels per gram of root were made among dates for each green. Population levels per gram of root changed significantly on all greens in both years for each of the three nematode populations. Within a putting green in either year, population fluctuation patterns in fenamiphos-treated and untreated plots were similar. Population fluctuation patterns were different between years, however. Within a year, population fluctuation patterns among greens showed similarities indicating that carefully monitoring a few locations may allow extrapolation of population fluctuation data to other locations within that year. 相似文献

A survey was conducted to determine the assemblage and abundance of plant-parasitic nematodes and their associations with soil factors in organically farmed fields in Minnesota. A total of 31 soil samples were collected from southeast (SE), 26 samples from southwest (SW), 28 from west-central (WC), and 23 from northwest (NW) Minnesota. The assemblage and abundance of plant-parasitic nematodes varied among the four regions. The soybean cyst nematode, Heterodera glycines, the most destructive pathogen of soybean, was detected in 45.2, 88.5, 10.7, and 0% of organically farmed fields with relative prominence (RP) values of 10.3, 26.5, 0.6, and 0 in the SE, SW, WC, and NW regions, respectively. Across the four regions, other common genera of plant-parasitic nematodes were Helicotylenchus (42.6, RP value, same below), Pratylenchus (26.9), Tylenchorhynchus and related genera (9.4), Xiphinema (5.6), and Paratylenchus (5.3). Aphelenchoides, Meloidogyne, Hoplolaimus, Mesocriconema, and Trichodorus were also detected at low frequencies and/or low population densities. The similarity index of plant-parasitic nematodes between two regions ranged from 0.44 to 0.71 and the similarity increased with decreasing distance between regions. The densities of most plant-parasitic nematodes did not correlate with measured soil factors (organic matter, pH, texture). However, the densities of Pratylenchus correlated negatively with % sand, and Xiphinema was correlated negatively with soil pH. 相似文献

Two hundred soil samples from the Ap horizon of a reed canarygrass field overlaying several different but related soils in northern Minnesota were analyzed for plant-parasitic nematodes and 22 edaphic factors. Pratylenchus penetrans was the predominant nematode taxon. Others were Aglenchus agricola, Tylenchorhynchus spp., Heterodera trifolii, Paratylenchus spp., Tylenchus maius, and Criconemella sp. Five nematode taxa, P. penetrans, A, agricola, Tylenchorhynchus spp., H. trifolii, and Paratylenchus spp., were correlated with particle size, Tylenchus maius and Criconemella sp. were correlated with effective cation exchange capacity. Nematode field spatial arrangements were related to a combination of statistically significant positive and negative soil factor effects on the nematode populations. Contour maps derived by geostatistical techniques were used to visually validate statistically significant correlations of nematode and soil data. Contour mapping to supplement traditional statistical techniques can be used to achieve a more holistic approach to studies of nematode-soil interrelationships. 相似文献

The palm weevil, Rhynchophorus palmarum (L.), was collected in cocoons from red ring-diseased coconut palms (Cocos nucifera L.) in Trinidad and Tobago. Juveniles of five species of nematodes were extracted from the genitalia and macerated bodies of newly emerged adults of the palm weevil: Rhadinaphelenchus cocophilus (Cobb) Goodey (the red ring nematode), Teratorhabditis sp., Diplogasteritus sp., Mononchoides sp., and Bursaphelenchus sp. Over 90% of newly emerged weevil females and males were infested internally with red ring nematode juveniles, and over 47% of the weevils contained more than 1,000 red ring nematodes each. There was no significant correlation between weevil body length and the number of red ring nematodes carried internally by each weevil. Teratorhabditis sp. and Diplogasteritus sp. were extracted from over 50% of the palm weevils, and Monochoides sp. and Bursaphelenchus sp. were found in a small proportion of the weevils. Field-collected adult weevils were also internally and externally infested with a Rhabditis sp., which was not observed in or on weevils allowed to emerge from field-collected cocoons. 相似文献

A study was conducted to determine the vertical distribution of Tylenchorhynchus nudus, Criconemella curvata, and Helicotylynchus cornurus in the upper 5 cm of bentgrass (Agrostis palustris cv. Penncross) putting green turf. The effect of fenamiphos on the vertical distribution of these species also was examined. Experimental design was a split-plot in which whole-plots were fenamiphos treated (0.11 kg a.i./100 m²) or untreated, and sub-plots were two strata (depths of 0-2.5 crn and 2.5-5.0 cm). Soil samples were collected during the growing season for 2 years after treatment to determine root weight and number of nematodes. Root weight was greater in the upper stratum on all sampling dates in both years. When differences between strata in population density were observed, T. nudus, C. curvata, and H. cornurus were more concentrated in the upper stratum. Vertical distribution of T. nudus, C. curvata, and H. cornurus was similar to the distribution of root weight. The difference in population density of H. cornurus between upper and lower strata was affected by fenamiphos on some dates, whereas differences between strata were unaffected for T. nudus and C. curvata. Double arcsine transformed proportions of the total populations of T. nudus, C. curvata, and H. cornurus in the upper stratum on each sampling date indicated no differences between fenamiphos treated and untreated plots in 1989 or 1990. 相似文献

The Mi-1.2 resistance gene in tomato (Solanum lycopersicum) confers resistance against several species of root-knot nematodes (Meloidogyne spp.). This study examined the impact of M. javanica on the reproductive fitness of near-isogenic tomato cultivars with and without Mi-1.2 under field and greenhouse conditions. Surprisingly, neither nematode inoculation or host plant resistance impacted the yield of mature fruits in field microplots (inoculum=8,000 eggs/plant), or fruit or seed production in a follow-up greenhouse bioassay conducted with a higher inoculum level (20,000 eggs/plant). However, under heavy nematode pressure (200,000 eggs/plant), greenhouse-grown plants carrying Mi-1.2 had more than ten-fold greater fruit production than susceptible plants and nearly forty-fold greater estimated lifetime seed production, confirming prior reports of the benefits of Mi-1.2. In all cases Mi-mediated resistance significantly reduced nematode reproduction. These results indicated that tomato can utilize tolerance mechanisms to compensate for moderate levels of nematode infection, but that the Mi-1.2 resistance gene confers a dramatic fitness benefit under heavy nematode pressure. Interbreeding potential, chromosome number, and host range were compared among several isolates and species of Bursaphelenchus from diverse geographic areas. Some isolates from North America, Japan, and France had a wide-ranging interbreeding potential, whereas others were restricted in their potential to hybridize with other isolates. Although interbreeding occurred in the laboratory between some "M" and "R" forms of B. xylophilus, interbreeding of B. xylophilus and B. mucronatus was rare. The hybrids had the pathogenicity of the parent with the broader host range. This fact suggests that virulence may be inherited as a dominant character or that increased virulence may have resulted from differences in hybrid vigor. The haploid chromosome number of the different isolates separated the isolates into three groups and distinguished B. xylophilus from B. mucronatus. The findings suggest that the pinewood nematode species complex consists of sibling species that have evolved by reproductive isolation, that the French isolate is a new species, and that B. xylophilus and B. mucronatus have evolved from a common ancestor. 相似文献

Xiphinema llanosum n. sp. and Trophurus vultus n. sp. are described and illustrated from grass soils in Llanos Orientales, Colombia. Xiphinema llanosum is a bisexual species. The female body length is 2.3-2.7 mm, odontostyle 86-96 μm, and odontophore 58-65 μm long; vulva at 42-47%; anterior ovary is absent; the anterior uterus and oviduct are similar to the posterior branch but slightly reduced; and the tail is dorsally convex-conoid with a blunt hemispherical terminus. Male body length is 2.06-2.96 mm; spicules are 40-44 μm long; and four (rarely three or five) anterior ventromedian supplementary papillae are present. Trophurus vultus females are 0.52-0.67 mm long; vulva at 56-60%; stylet is 10.5-13.5 μm long; isthmus is as long as the basal esophageal bulb; the tail is subclavate, 1.6-2.2 times anal body width long; and the terminal cuticle thickness is about one-sixth of the tail length. 相似文献
