Techniques for the preparation of haptoral sclerites of Gyrodactylus species for scanning electron microscopy are described. Enzymatic digestion using pepsin allows fresh, frozen and alcohol-fixed material to be examined, but some sclerites, such as the dorsal and ventral bars, are lost. Sonication of fresh and frozen material retained these accessory bars, including the ventral bar membrane, the guard or sickle filament loop on the marginal hooks and even on some occasions ligaments associated with the sclerites. The sclerites of Gyrodactylus spp. from Gasterosteus aculeatus (L.) (three-spined stickleback), Oncorhynchus mykiss (Walbaum) (rainbow trout) and Salvelinus alpinus (L.) (charr) are described from SEM observations following the use of these preparation techniques, and the articulation between the hamuli and bars is commented upon.  相似文献

Introduced and allopatric populations of brown trout Salmo trutta and rainbow trout Oncorhynchus mykiss were sampled in Slovenia for stable isotope analysis to assess dietary niche shifts through ontogeny and estimate the propensity for cannibalism. Both S. trutta and O. mykiss are cannibals, with higher average relative contribution of conspecific assimilated energy for S. trutta (27·9%) compared with O. mykiss (7·7%). The smallest cannibal was 166 mm in the S. trutta population and 247 mm in the O. mykiss population.  相似文献

Wild and cultured winter flounder Pseudopleuronectes americanus (Walbaum) from Passamaquoddy Bay were surveyed for species of Gyrodactylus Nordmann, 1832. Two species were found: G. pleuronecti Cone, 1981 and G. aideni n. sp, both members of Malmberg’s ‘groenlandicus group’. Although the hard parts in the haptor are very similar in the two species, hamuli of G. aideni are consistently shorter than those of G. pleuronecti. The two species differed by 35 base pairs in the ITS 1, 5.8 and ITS 2 region. A BLAST search identified a variety of species of Gyrodactylus from marine fishes in the Atlantic Ocean as closest matches, indicating the ‘groenlandicus group’ is part of a major marine lineage within Gyrodactylus (sensu lato) that has successfully radiated among coastal percid, pleuronectid, cottid and anarhichadid fishes. Exposure experiments suggested that winter flounder is the primary host of both species of parasites and that three other pleuronectid species in the bay may potentially serve only as occasional transport hosts.  相似文献

Juvenile Atlantic salmon Salmo salar from three allopatric populations (LaHave, Sebago and Saint‐Jean) were placed into artificial streams with combinations of four non‐native salmonids: brown trout Salmo trutta, rainbow trout Oncorhynchus mykiss, Chinook salmon Oncorhynchus tshawytscha and coho salmon Oncorhynchus kisutch. Non‐additive effects, as evidenced by lower performance than predicted from weighted summed two‐species competition trials, were detected for S. salar fork length (LF) and mass, but not for survival, condition factor or riffle use. These data support emerging theory on niche overlap and species richness as factors that can lead to non‐additive competition effects.  相似文献

In the present study, the authors investigated the effect of acclimation duration (up to 4 h) on behavioural characteristics of taxonomically and functionally different fish species, i.e., the migratory rheophilic salmonids rainbow trout (Oncorhynchus mykiss) and Atlantic salmon (Salmo salar), and the non-migratory eurytopic European perch (Perca fluviatilis) and three-spined stickleback (Gasterosteus aculeatus). Specifically, the authors explored fish behavioural patterns based on specific endpoints (average, maximum and angular velocity) during the acclimation period, and determined the acclimation period suitable for the tested fish species. The performed behavioural data analysis showed that the minimum time needed to adjust fish activity to a more stable (baseline) level should be at least 2 h for O. mykiss and S. salar and 1 h for G. aculeatus. Nonetheless, P. fluviatilis behaviour did not show significant changes during the 4 h acclimation. The results of this study revealed that the effect of the acclimation duration on such rheophilic species as O. mykiss and S. salar was greater than that on the eurytopic species P. fluviatilis and G. aculeatus, indicating that acclimation period is important in managing fish stress before behavioural observations. For all species, the highest variability was found in the endpoint of maximum velocity, and the lowest in that of angular velocity. This study showed that before starting actual toxicity testing experiments, it is important to determine an appropriate, species-specific acclimation period.  相似文献

Replication banding technique was applied to the chromosomes of Salmo salar, Salmo trutta, and Oncorhynchus mykiss. The in vitro technique has proved more advantageous than the in vivo technique due to a higher number of bands obtained. The comparison of these replication banding patterns has revealed that some chromosomes of Salmo trutta karyotype appeared associated with Salmo salar and Oncorhynchus mykiss karyotypes by single chromosomal rearrangements.  相似文献

Thirty populations of potato cyst nematode (Globodera spp.) from the Island of Tenerife and two populations from the UK were assessed for several morphometric and non-morphometric characters thought to discriminate between the species G. rostochiensis and G. pallida. Also 200 cysts from each population were analysed by isoelectric focusing of soluble proteins. Correlation analysis, analysis of variance and principal component analysis were used to investigate relationships between the morphometric characters, how the relationships varied between species and between populations, and which characters were most useful for discriminating between species. The two species differed significantly for each of the four morphometric characters: stylet length, fenestra length, anus-fenestra distance and the number of ridges. The stylet length and fenestra length also showed differences between populations of G. rostochiensis while stylet length and number of ridges showed differences between populations of C. pallida. In general, populations of G. pallida showed greater variation than populations of C. rostochiensis. Principal component analysis of the population means indicated that over 73% of the variation in the characters could be explained by the contrast of stylet and fenestra lengths against the anus-fenestra distance and number of ridges. A plot of the first two principal components separated the two species. Stepwise discriminant analysis provided a linear combination of these four variables which discriminated between the species. Stylet length was found to be the most useful characteristic for distinguishing the species whilst anus-fenestra distance was the least useful.  相似文献

Gyrodactylus jennyae n. sp. is described from the body surface and mouthparts of tadpoles of the bullfrog Rana catesbeiana Shaw imported presumably from Missouri, USA, into a federal government facility in Moncton, New Brunswick, Canada. Its morphology resembles most closely that of G. chologastris Mizelle, Whittaker & McDougal, 1969 described from two amblyopsids (blind cave fishes) in Kentucky and North Carolina. Both species have long slender hamuli, a ventral bar with a relatively long membrane and small anterolateral processes, a cirrus with two rows of small spines and marginal hooks with a well-developed sickle heel and short handle. The two species differ morphologically; G. jennyae has a marginal hook sickle with a more pronounced heel than that found in G. chologastris. A BLAST search using a 945 base pair sequence that included the nuclear ribosomal DNA internal transcribed spacers 1 and 2 and the 5.8S rRNA gene from G. jennyae n. sp. showed that the overall similarity with other Gyrodactylus sequences on GenBank was relatively low. The ITS1 region was similar to that of G. misgurni Ling, 1962; however, no ITS2 and 5.8S rRNA sequences are available for that species. A separate search using 5.8S sequences revealed that G. markakulensis Gvosdev, 1950 and G. laevis Malmberg, 1957 were the closest to G. jennyae (1 and 2 bp differences, respectively). These species are parasites of cyprinids (or their predators) and are similar to G. jennyae and G. chologastris in having a double row of small hooks on the cirrus and overall similar morphologies of the haptoral hard parts. There are now five species of Gyrodactylus described exclusively from amphibians and this appears to have involved at least three separate host-switches from fishes.  相似文献

An Atlantic salmon (Salmo salar L.) expressed sequence tag (EST) database consisting of 58 146 ESTs was screened for microsatellite sequences. Subsequent development of 75 polymorphic EST‐associated microsatellite markers in this species is described together with cross‐species amplification results of 133 gene‐associated tandem repeat markers in five salmonid species (Salmo trutta, Oncorhynchus mykiss, Salvelinus aplinus, Thymallus thymallus, Coregonus lavaretus). The number of alleles among EST‐linked microsatellites in Atlantic salmon ranged from two to 41 with an average of 12 alleles per locus. Cross‐species amplification resulted in detection of a total of 111 polymorphic locus‐species combinations (12–32 loci per species).  相似文献

Gazella is one of the most species‐rich genera within horned ruminants. Despite overall similarity in body size and morphology, gazelles show variability in coloration and horn morphology. Unfortunately, however, species differentiation based on these characters, or on discrete skull characters, is very difficult due to high intraspecific variability. Furthermore, most species have fragmented and allopatric distributions, so that species boundaries were hard to define in the past. Mitochondrial DNA sequences have proven useful for investigating gazelle taxonomy in recent years, but especially for old museum material, i.e. type specimens, destructive sampling is often impossible. We provide a comprehensive morphometric framework for the genus Gazella based on linear skull measurements reconciled with results from molecular phylogenetic analysis based on the largest dataset available so far. In particular for males, the skull morphology shows interspecific differences concurrent with DNA data and provides a reliable tool for species identification. Based on morphometric data we synonymize G. karamii with G. marica, and confirm the identification of the G. arabica and G. a. rueppelli type skulls from analyses of mitochondrial DNA sequences. © 2013 The Linnean Society of London  相似文献

Morphological species of freshwater microalgae often have broad geographic distribution. However, traditional species concepts have been challenged by the results of molecular phylogenetic analyses that mostly indicate higher diversity than was previously recognized by purely morphological approaches. A degree of phenotypic differentiation or different geographic distribution of species defined by molecular data remains largely unknown. In this study, we analyzed a pair of well-known and widely distributed desmid species (Micrasterias fimbriata and M. rotata) and tested for their phylogenetic and morphological homogeneity as well as their geographic distribution. Geometric morphometric and morphological attributes of cells were used in combination with genetic analysis of the trnG ucc sequences of 30 strains isolated from a variety of European locations and obtained from culture collections. Micrasterias rotata proved to be phylogenetically homogenous across Europe while M. fimbriata turned out to be composed of two firmly delimited lineages, differing by molecular as well as by morphometric and morphological data. Published records of traditional M. fimbriata were also included in the classification discrimination analysis and were placed into the newly identified lineages upon comparison to the morphometric data collected from living material. Largely disparate geographic patterns were revealed within traditional M. fimbriata. One phylogenetic lineage is frequent in central and eastern Europe, but occurs also in the British Isles. A second lineage has been recorded in North America and in Western Europe, where its distribution is possibly limited to the west of the Rhine River. Interestingly, the morphometric analyses of the published records illustrated that the geographic differences have remained largely unchanged since the 1850s indicating a previously unknown distributional stability among microalgal species groups such as the desmids.  相似文献

A new species of Gyrodactylus von Nordmann, 1832 and the first monogenean reported from Botswana is described from the cichlid Pseudocrenilabrus philander philander,bringing the number of gyrodactylids described from the African Continent to 18. Gyrodactylus thlapi n. sp. can be readily discriminated from the other described species almost exclusively by the shape of its hamuli, which have short roots and large, elongate ventral bar attachment points with the dorsal bar attachment point, demarcating the junction between the hamulus shaft and the root, positioned at the anterior terminus of the attachment plane. The marginal hooks of G. thlapi n. sp., which most closely resemble those of G. groschafti Ergens, 1973, are compared and discussed.  相似文献

Species of the genus Gambierdiscus Adachi & Fukuyo, in particular G. toxicus Adachi & Fukuyo are known producers of neurotoxins associated with ciguatera fish poisoning (CFP). In this study live samples were collected from seaweed beds of the east coast of Sabah, Malaysian Borneo and a strain of Gambierdiscus was isolated and cultured. Examination of the thecal fine morphology was undertaken using light, epifluorescence, and scanning electron microscopy. Observed morphological features and their associated morphometric information enabled identification to Gambierdiscus belizeanus Faust. This represents the first report for the occurrence of G. belizeanus in the Asia Pacific region.  相似文献

We introduce a new statistical method to select which morphological characters are most useful to identify monogenean species. The method estimates the average size overlap of candidate diagnostic structures among a set of species to individuate those that mostly differ between the species. To demonstrate our approach, we report a comprehensive analysis conducted on two of the most species‐rich monogenean genera: Dactylogyrus Diesing, 1850 and Gyrodactylus von Nordmann, 1832. We demonstrate that, in contrast to common taxonomic practice, very few but highly diagnostic measurements are necessary to correctly identify a specimen. In particular, we found that most of Dactylogyrus and Gyrodactylus species can be classified on the basis of the width of the supplementary connecting bar and of the length of the hook sickle, respectively.  相似文献

A new species of Gyrodactylus was described on the body surface of zebrafish (Danio rerio) in China. Basing on morphological characteristics and ITS sequence, we identified the parasite as a new member of the Gyrodactylus-wageneri group. Morphologically, Gyrodactylus sp. nov. is greatly similar to “G. zebrae”, another species parasitic on zebrafish: both have moderately stout hamulus, marginal hook sickle with a prominent heel and toe, as well as a curved blade. However, distinct haptoral shape differences were detected between these two species. The dorsal bar is straight in Gyrodactylus sp. nov. but strongly curved in “G. zebrae”, and the sickle shaft in Gyrodactylus sp. nov. is approximately perpendicular to the base, but in “G. zebrae” it is slanted downwards. The ITS1-5.8S-ITS2 sequence also indicate that Gyrodactylus sp. nov. exhibits the highest similarity to “G. zebrae”: 95.7% sequence identity suggests interspecific differentiation. Phylogenetic analysis of the ITS1-ITS2 sequence showed that Gyrodactylus sp. nov. formed a sister clade with “G. zebrae”, and exhibited a relatively close phylogenetic relationship with G. kobayashii, G. gurleyi, and G. longoacuminatus, all of which parasitise on goldfish, Carassius auratus. To test the susceptibility of zebrafish and goldfish to the Gyrodactylus sp. nov., ten gyrodactylids were inoculated to the caudal fin of zebrafish and goldfish. The gyrodactylids exhibited the ability to attach themselves to the goldfish, and some gyrodactylids reproduced a few days after the inoculation. On day 9, however, the mean abundance sharply decreased to zero on goldfish and increased to more than 30 on zebrafish. The result suggested that golfish is an unsuitable host for Gyrodactylus sp. nov. Therefore, on the basis of morphology, molecular sequence similarity, and host susceptibility, we conclude that the gyrodactylid found on the zebrafish is a new species, which we named Gyrodactylus banmae.  相似文献

Studies were carried out on Gyrodactylus salaris Malmberg, 1957 on rainbow trout Oncorhynchus mykiss (Walbaum, 1792) from a fish farm in Lake Tyrifjorden, south-eastern Norway. The anchors were larger, and the shape of the anchors and the ventral bar differed slightly, as compared with the same parts of G. salaris on parr of Atlantic salmon Salmo salar L. in northern and north-western Norwegian rivers. At different water temperatures (0.8°C, 10.0°C, 18.0°C), the opisthaptoral hard parts of G. salaris on rainbow trout showed considerable variation in size, but varied only slightly in shape. It was found that the total length of the anchors of G. salaris on rainbow trout may considerably exceed the previously reported maximum of 80 m for the species. The spread of G. salaris to south-eastern Norway is described and discussed.  相似文献

Where the distribution ranges of the waterstriders Limnoporus notabilis and L. dissortis meet in western Canada, extensive hybridization and introgression occurs. Multivariate ordination analyses of genetic and morphometric data by principal component analysis revealed that a single axis separating the two parent species could account for nearly all the variation in both data sets. Maps of principal component scores for both data sets revealed geographical patterns of variation reflecting specific topographic features in the region. Comparisons of morphometric data from some of the samples collected in the 1980s and from the same sites revisited in the 1990s revealed substantial changes. An ‘island’ of dissortis‐like populations inside the range of L. notabilis in interior British Columbia expanded, and a marked local protrusion of notabilis‐like phenotypes into the range of L. dissortis on the east slope of the Rocky Mountains diminished during the decade between collections. We conclude that introgressive hybridization between these two species of waterstriders is a spatially complex and highly dynamic process.  相似文献

Abstract

This integrated study provides new insights into pollen and seed morphology and pollen heteromorphism of four closely related annual taxa of Viola sect. Melanium. The plant material, both fresh and dried, was collected in Italy and studied using light and scanning electron microscope. Palynological data for V. hymettia together with a detailed comparative analysis of seed morphology and micromorphology of the four species are reported for the first time. Results of this work highlight some pollen and seed features as useful diagnostic characters. The pollen size proves to be of diagnostic value to easily separate V. kitaibeliana, having the smallest pollen grains, from the others, especially from V. arvensis with the largest ones. Exine ornamentation is microreticulate, showing no relevant differences among species. We can partially confirm the diagnostic value of the prevailing pollen morph as it can be useful only for V. arvensis (five-aperturate) versus V. tricolor and V. hymettia (four-aperturate). The macro- and micromorphology of seeds provide additional useful distinguishing characters. Particularly, seed size was found to be a good delimitating character, especially to distinguish V. kitaibeliana (with the smallest seeds) from V. arvensis, and easy to be measured with no need of particular equipment.  相似文献
