The relative toxicity of pesticides to Phyllocnistis citrella and its parasitoid Ageniaspis citricola was compared by several bioassay methods. A clip-cage bioassay measured survival of adults exposed to fresh residues at 0.25-times (0.25×), 0.5×, 1×, and 2× the lowest recommended rate of each pesticide, a water control (0×), and 24- and 48-h aged residues of oil at 1.5% (1×) rate. A one-species cylinder bioassay determined the effects of pesticides on immature stages of the citrus leafminer after treating young citrus trees with the same rates of pesticides (except for avermectin, tested at 0×, 0.01×, 0.025×, 0.05×, 0.1×, and 0.25× rates). A sublethal rate of petroleum oil (0.4%) was added as an adjuvant in some treatments. The effect of pesticides on immature stages of A. citricola was determined with a two-species cylinder bioassay after treating young citrus trees with the same rates as above. An index of IPM compatibility was developed based on the efficacy of the pesticide as a control agent of the leafminer (or other pests) and its selectivity to the parasitoid at the lowest recommended field rate (0.25× the field rate for avermectin). Azadirachtin (Neemix) + oil, diflubenzuron (Micromite) + oil, fenoxycarb (Eclipse) + oil, and oil alone (FC 435-66) were classified as IPM-compatible insecticides. Sprays of azadirachtin (Align) + oil, neem oil (Neemgard), and drenched imidacloprid (Admire) were ranked as a semi-compatible insecticides. The fungicide copper hydroxide (Kocide 101) and a fish oil-based foliar fertilizer (Zapata HFE) were considered compatible. Avermectin (Agri-Mek) + oil, ethion (Ethion), and imidacloprid (Provado) applied as a spray were IPM-incompatible insecticides. These products should be tested under field conditions to confirm these ratings.  相似文献

The cell cycle and phase times of human lymphocytes responding to PHA have been analysed with the percent labelled metaphases (PLM) technique. The range of generation times (13–18 h) and DNA synthesis times (6.5–10.5 h) reported here compare well with previous measurements in the literature. Cycle analyses of the early responding cells of the initial response, selected with partial anti-PHA serum inhibition, and of restimulated cells yield relatively well-defined PLM curves. The short cycle times measured from these curves may reflect the early cycles after stimulation or a subpopulation of responding cells. Analyses at two times during both the initial and restimulation responses suggest that cycles lengthen with time after stimulation. The poor PLM curves of the initial response and the restimulation response of cells released from anti-PHA inhibition indicate considerable intercellular variation in cycle times. Cells in the initial long G 1 phase contribute to this variation. PHA dose does not appear to affect the cycle time.  相似文献

Acute adaptation of mice to hypoxic hypoxia.   总被引:8,自引:0,他引:8  
Tolerance to hypoxia in vivo and in vitro was significantly increased by acute and repetitive exposure of mice to autoprogressive hypoxia. The average tolerance times of the successive 2nd, 3rd, 4th and 5th runs of exposure were, respectively, 2, 4, 6 and 8 times as long as that of the first exposure. The survival times under hypobaric chamber and cyanide toxification in the 4th exposure were, respectively, 10 (and even as much as 86) and 4 times those in control mice without exposure to hypoxia. Mandibular respiration and spinal reflex in vitro in hypoxia-resistant animals lasted 5-6 times as long as in control animals not previously exposed to hypoxia. Animals that received brain homogenate from hypoxia-resistant mice remained alive in a hypobaric chamber 2 times as long as those that received homogenate from controls and those that received saline. These results indicate that a kind of quickly developing adaptation with increased tolerance is achieved by acute and repetitive exposure of mice to progressive autohypoxia and some plastic or adaptive changes occur in the brain of hypoxia-resistant animals, including the production of some kind of water-soluble antihypoxic factors.  相似文献

背景: 番茄(Solanum lycopersicum)是广泛种植的最具价值的果蔬之一,番茄晚疫病会导致其产量和品质降低。microRNAs(miRNAs)是一类内源性非编码RNA,广泛参与基因转录后调控。已有研究表明miR399家族可参与调控植物抗病过程。目的:探究番茄miR399(sly-miR399)对番茄抗晚疫病的影响。方法:构建sly-miR399的过表达和沉默载体,利用农杆菌介导在番茄叶片中瞬时表达;实时荧光定量PCR(qRT-PCR)技术检测相关基因表达水平;台盼蓝染色并统计分析瞬时过表达和瞬时沉默sly-miR399的番茄叶片的病斑情况。结果:瞬时过表达sly-miR399导致其靶基因UBC24的表达量下降了75%,病程相关蛋白(pathogenesis related proteins,PRs)SlPR1SlPR2SlPR3SlPR5的表达量分别上升了3.6倍、2.2倍、2.3倍和6.4倍,茉莉酸(JA)信号通路相关基因SlJA1SlLOX1SlLOX2的表达量分别上升了1.3倍、2.5倍和1.5倍,JA转录抑制因子SlJAZ1的表达量下降了50%,接种晚疫病菌后,叶片上相对病斑面积明显减小;瞬时沉默导致其靶基因的表达量上升了1.8倍,PRs基因的表达量分别下降了65%、82%、52%和80%,SlJA1SlLOX1SlLOX2的表达量分别下降了84%、50%和65%,SlJAZ1的表达量上升了1.8倍,病菌接种后叶片上相对病斑面积明显变大。结论:初步揭示了sly-miR399在番茄抗晚疫病过程中的正调控因子作用。  相似文献

Patch clamp recordings of excitatory postsynaptic currents (EPSCs) in central neurons reveal large fluctuations in amplitudes and decay times of AMPA-receptor-mediated EPSCs. By using Monte Carlo simulations of synaptic transmission in brainstem interneurons, we tested several hypothesis that could account for the observed variability. The coefficient of variation (CV) of 0.5 for miniature amplitudes cannot be explained by fluctuations in vesicle content or receptor distribution, but is traced to variations in receptor number, which is estimated as 77+/-39 receptors per bouton. As the variability of rise times reflects fluctuations in size of the post-synaptic density and heterogeneity of the receptor distribution, the relatively small CV=0.37 of experimentally determined values points to a homogeneous arrangement of receptors. Within our model the large variability of decay times (CV=0.49) can only be explained by fluctuations in the transmitter time course (mean residence times of 0.4+/-0.13 ms), presumably resulting from heterogeneities in synaptic morphology. Hence, our simulations indicate that different noise sources control the variability of amplitudes, rise and decay times. In particular, the distribution of decay times yields information about the synaptic transmission process, which cannot be obtained from other observables.  相似文献

The Phaseolus vulgaris isolectins L4,L3E1, L2E2, L1E3, and E4 were isolated by affinity and ion exchange chromatography. Pure isolectins were radiolabeled by the chloramine-T method with Na125IO4 and their binding to human erythrocytes was studied. A normal erythrocyte has approximately 8 times 10(5) receptor sites for each isolectin; however, the association constants (Ka) of binding increased from 1.1 times 10(7) M-1 to 3.8 times 10(8) M-1, with increasing number of E subunits per tetrameric isolectin molecule. Isolectin to erythrocyte binding reached equilibrium rapidly and was reversed by fetuin. All isolectins competed with 125I-E4 for erythrocyte binding sites, with a constant (KI) similar to the Ka calculated for each respective radiolabeled isolectin. When isolectin binding at 0 degrees C, 4 degrees C, or 8 degrees C was compared to that at 25 degrees C, there was no reduction in the number of binding sites per cell, but the Ka of E4 was reduced to 3 times 10(7) M-1. Fixed erythrocytes displayed similar isolectin binding characteristics.  相似文献

Reaction times were determined for monkeys and humans who made wrist flexion and extension movements in response to vibratory and visual cues. Humans initiated movements approximately 50 msec sooner in response to vibratory as compared to visual cues. For monkeys, this difference was approximately 100 msec. Mean daily reaction times for monkeys and humans improved with practice until they reached a steady level of performance. Increased differences between vibratory and visual reaction times were weakly correlated with increased age of humans. The increase in the differences appeared to result from decreased reaction times by older subjects for vibratory-cued movements; reaction times for visually cued movements did not consistently vary across the age range of subjects tested (19-36 years). The results obtained using this novel paradigm suggest that it may be a useful tool for simultaneously testing behavioral performance or neurological function during somatosensorimotor and visuomotor tasks.  相似文献

The significance of the phase of circadian rhythmicity for the diagnosis of sleep disturbance was investigated in a group of 80 chronic insomniacs (59 females; mean age 34.8, range 18-59 years). In order to stay close to common clinical practice, data were collected by means of two-week sleep diaries in combination with repeated measurements of subjective alertness and oral temperature. Special measures were taken to minimize the impact of masking upon the temperature measurements. In addition, wrist activity was monitored for an overlapping period of 11 days. Measurements of oral temperature and subjective alertness were fitted with 3 (rd) -degree polynomials, for which the peak times (times of maximum) were identified. Principal Components Analysis of these peak times and the times of bed-in and wake-up for all subjects revealed that the phase estimates for the alertness and the sleep-wake rhythms had a strong interrelationship, which was independent from the temperature phase. Using the 25- and the 75-percentiles of the frequency distribution of the temperature peak times as boundaries, the subjects were classified into early (N = 18), middle (N = 37) and late (N = 19) temperature phase subgroups, which had mean peak times of 14:08 h, 17:43 h and 20:09 h, respectively. Comparisons between the early phase and the late phase subgroups showed that a significant overall MANOVA effect was mainly due to differences in total sleep time (early < late) as calculated from the log, and to differences in the mean nocturnal actigraphic count (early> late). Moreover, the subjective estimates of sleep latency (early < late) and wake after sleep onset (early > late) tended to differ between the two subgroups. The main result of this study, i.e., that insomniacs with a relatively advanced temperature phase had a relatively shorter and more restless sleep, while insomniacs with a relatively delayed temperature phase tended to experience a relatively long sleep latency, supports the conclusion that the addition of oral temperature measurements to a sleep/wake log extends its diagnostic and therapeutic applicability.  相似文献

This paper is motivated by a case study to reduce the throughput times for radiotherapy treatment. The goal is to find a cost-effective way to meet future throughput targets. A combination of queuing theory and computer simulation was used. First, computer simulation to detect the bottleneck(s) in a multi-step radiotherapy process. Despite, the investment in an additional linear accelerator, the main bottleneck turned out to be the outpatient department (OPD). Next, based on queuing theory, waiting times were improved by reducing the fluctuations in the OPD capacity. Computer simulation was used again to quantify the effect on the total throughput time of a radiotherapy patient. The results showed a reduction in both access times as well as waiting times prior to the consecutive steps: the preparation phase and actual treatment. The paper concludes with practical suggestions on how to reduce the fluctuations in capacity, and seems of interest for other radiotherapy departments or other multi-step situations in a hospital.  相似文献

The root cell walls of the resistant cultivars of the date palm were more resistant to the action of the cell wall-degrading enzymes (CWDE) of Fusarium oxysporum f. sp. albedinis than those of the susceptible cultivars. Date palm roots contain four cell wall-bound phenolics identified as p-hydroxybenzoic acid, p-coumaric acid, ferulic acid and sinapic acid. The contents of p-coumaric acid and ferulic acid in the resistant cultivars (IKL, SLY, BSTN) were about 2 times higher than those in the susceptible cultivars (BFG, JHL, BSK). The contents of p-hydroxybenzoic acid and sinapic acid in the resistant cultivars were 8.4 and 4.5 times, respectively, higher than those in the susceptible cultivars. The lignin contents in roots of the resistant cultivars were 1.8 times higher than those of the susceptible cultivars. The cell wall-bound phenols accumulated particularly in resistant cultivars reduced strongly the mycelial growth and the CWDE production in vitro.  相似文献

Experiments were conducted on guinea pigs and Papio hamadryas; it was shown that a reduction of the intensity of postvaccinal immunity occurred at various periods after a single vaccination. In inhalation method of immunization in guinea pigs it decreased in 6 months 135 times, in monkeys in one year--133 times. However, at the mentioned periods vaccination provided protection of 50% of the animals from infection with Past. pestis in a dose constitutin 20 to 25 aerosol LD50 for nonimmunized animals. Despite the more pronounced (57--640 times) reduction of the intensity of immunity than in the animals vaccinated by inhalation, in the subcutaneously vaccinated guinea pigs in subcutaneously infected with Past. pestis protection level remained high (resistance index in 3 and 6 months constituted 37.10(6) and 3-3-10(6), respectively).  相似文献

Rhesus mothers (Macaca mulatta) may hold their own infant together with the infant of another female in a so-called double-hold. This behavior was observed 240 times in 23 individual females in two independent captive groups. Approximately nine out of ten times, the second infant was from a dominant matriline. Almost one-third of Australians report having made errors at work that are related to sleep issues. While there is significant literature investigating the role of sleep in workplace health and safety in shiftworking and nightwork operations, long working hours, work-family conflict, and commute times getting longer also impact day workers’ sleep behaviors and opportunities. The aim of this study was to examine the relationship between sleep duration and disorders, sleep health and hygiene factors, work-related factors and errors at work in Australian workers. From a sample of 1011 Australian adults, age-adjusted binary logistic regression analyses were conducted in 512 workers who provided responses to the question “Thinking about the past three months, how many days did you make errors at work because you were too sleepy or you had a sleep problem?” A number of sleep behaviors and poor sleep hygiene factors were linked with work errors related to sleepiness or sleep problems, with age-adjusted odds of errors (confidence intervals) up to 11.6 times higher (5.4–25.1, p < 0.001) in those that snored, 7.7 (4.6–12.9) times higher in those reporting more than three sleep issues (p < 0.001), 7.0 times higher (3.4–14.8) in short (≤5 hours/night) sleepers (p < 0.021), 6.1 times higher (2.9–12.7) in those staying up later than planned most nights of the week (p< 0.001) and 2.4 times higher (1.6–3.7) in those drinking alcohol ≥3 nights/week before bed (p < 0.001). More than 40% of participants working non-standard hours reported making errors at work, and they were more likely to be young (compared to the main sample of workers) and more likely to engage in work activities in the hour before bed. Sleep factors (other than clinical sleep disorders) were associated with an increased likelihood of sleep-related work errors. Both day workers and those working non-standard hours engage in work, sleep and health behaviors that do not support good sleep health, which may be impacting safety and productivity in the workplace through increased sleepiness-related errors.  相似文献

Escherichia coli mutants lacking exonuclease III (xthA) are exceptionally sensitive to hydrogen peroxide. They are killed by H2O2 at 20 times the rate of wild-type bacteria and at 3 to 4 times the rate of recA cells. This is the first clear phenotypic sensitivity reported for xth- E. coli and should aid in clarifying peroxide-induced lethality and the in vivo role of exonuclease III.  相似文献

A variety of luminol-based substrates and either an autoradiographic film or a charge-coupled device (CCD) imaging system were used for chemiluminescence detection of c-type cytochromes. The Pierce Femto peroxidase substrate was at least 10 times more sensitive when using film than the highly cited 3,3('),5,5(')-tetramethylbenzidine (benzidine derivative) staining method and 50 times more sensitive when using a CCD imaging system. Film or CCD imaging result in highly sensitive and quantitative signals. The quantitative detection of c-type cytochromes from single colonies or from less than 1ml of a bacterial culture is possible.  相似文献

以山西省吉县蔡家川流域为对象,研究了植被覆盖类型对流域水文的影响.结果表明:不同植被覆盖的流域年径流系数分别为:林地流域1.6%~2.3%,以农、牧为主的流域3.1%~3.9%;各流域基流系数差异显著,人工林流域为零,次生林为主的流域1.0%~1.5%,以农、牧为主的流域2.5%~2.8%;在雨季人工林流域的径流总量是次生林流域的3.37倍、农地流域的1.9倍,而农地流域的基流量是次生林流域的2.2倍;短历时高强度降雨条件下,人工林流域、次生林流域地表径流量分别是农地流域的10.8倍和2.2倍;在历时较长的暴雨条件下,人工林流域单位面积上的洪峰流量是农地流域的3.4倍,次生林流域的6.9倍;在长历时、大雨量条件下,农地流域的径流量是次生林流域的1.8倍.水平梯田的水源涵养功能与次生林植被相当,次生林植被的水源涵养功能远好于人工植被,在水资源短缺的黄土高原应提倡植被的自然恢复.  相似文献

We have developed a general method for solving transient kinetic equations using Laplace transforms. Laplace transforms can be used to transform systems of differential equations that describe pre-steady-state kinetics to systems of linear algebraic equations. The general form of the pre-steady-state solution is (formula; see text) where I(t) is the time dependence of the physically observed property of the system, n is the number of intermediates, lambda i are the observed rate constants (reciprocals of the relaxation times), t is time, and Ii are the amplitude coefficients associated with each observed rate constant. We have written a program in compiled BASIC to run on a personal computer to evaluate Ii and lambda i. The program will evaluate the rate constants and coefficients of a mechanism with eight intermediates and seven relaxation times in 4 s on an 8-MHz PC-XT equipped with a math coprocessor. The most complex mechanism that we have solved, a mechanism containing 20 intermediates and 19 relaxation times, required approximately 5 min. We believe that this method will be useful to evaluate the differences in transient properties of complex biochemical mechanisms.  相似文献

The consequences of parental exposure to epidermal growth factor (EGF), for progeny cell cycle times was investigated. Slowly dividing mouse 3T3 fibroblasts were exposed to EGF for 8 hr, the EGF was withdrawn, and the cell cycle times of parental and progeny cells were measured by time-lapse video microscopy. It was observed that exposure to EGF induced a round of cell division following a lag phase of approximately 8 hr. The progeny of these cells exhibited accelerated cell cycle times compared to cells that had not been exposed to EGF. Parental cell division time was significantly correlated with progeny cell cycle time. Sibling progeny cell cycle times were also significantly correlated. EGF can therefore apparently exert an effect on the cell cycle times of more than one generation of cells.  相似文献

Glycogen stores (liver and carcass) have been studied in lean and obese Zucker rats. The animals were submitted to one of three feeding conditions: ad libitum, a 48-h fast, or a 48-h fast and food ad libitum for 24 h, and to two environmental conditions, either thermoneutrality or an acute cold exposure (2 days at 4-7 degrees C). After a 2-day fast at 25 degrees C, the liver glycogen store was reduced by 45 times in the lean rats, while it was decreased by only 3 times in the obese rats. Under these conditions, the liver glycogen store was 45 times higher in the obese than in the lean rats. After 2 days in the cold, liver glycogen store was 4.4 times higher in obese rats than in lean rats. After a 2-day fast in the cold, the liver glycogen store in the obese rats was 30 times higher than in the lean rats. In comparison to fasting at thermoneutrality, fasting in the cold did not lead to a further reduction in hepatic glycogen in obese Zucker rats. The differences observed in the mobilization of the hepatic glycogen store between obese and lean rats have not been found in the mobilization of the carcass glycogen store. Drastic conditions, such as a 2-day fast in the cold, did not exhaust the glycogen store in obese Zucker rats. The present observations point out that obese Zucker rats cannot mobilize the entire hepatic glycogen store, as seen in lean control rats. The role of this abnormality in the high hyperlipogenesis that maintains the obese state is still to be evaluated.  相似文献
