The Drosophila melanogaster TRPA family member painless, expressed in a subset of multidendritic neurons embeding in the larval epidermis, is necessary for larval nociception of noxious heat or mechanical stimuli. However, the function of painless in adult flies remains largely unknown. Here we report that mutation of painless leads to a defect in male-male courtship behavior and alteration in olfaction sensitivity in adult flies. Specific downregulation of the expression of the Painless protein in the olfactory projection neurons (PNs) of the antennal lobes (ALs) resulted in a phenotype resembling that found in painless mutant flies, whereas overexpression of Painless in PNs of painless mutant males suppressed male-male courtship behavior. The downregulation of Painless exclusively during adulthood also resulted in male-male courtship behavior. In addition, mutation of the painless gene in flies caused changes in olfaction, suggesting a role for this gene in olfactory processing. These results indicate that functions of painless in the adult central nervous system of Drosophila include modulation of olfactory processing and inhibition of male-male courtship behavior.  相似文献

Mutations in the Drosophila retained/dead ringer (retn) gene lead to female behavioral defects and alter a limited set of neurons in the CNS. retn is implicated as a major repressor of male courtship behavior in the absence of the fruitless (fru) male protein. retn females show fru-independent male-like courtship of males and females, and are highly resistant to courtship by males. Males mutant for retn court with normal parameters, although feminization of retn cells in males induces bisexuality. Alternatively spliced RNAs appear in the larval and pupal CNS, but none shows sex specificity. Post-embryonically, retn RNAs are expressed in a limited set of neurons in the CNS and eyes. Neural defects of retn mutant cells include mushroom body beta-lobe fusion and pathfinding errors by photoreceptor and subesophageal neurons. We posit that some of these retn-expressing cells function to repress a male behavioral pathway activated by fruM.  相似文献

Drosophila male courtship is a complex and robust behavior, the potential for which is genetically built into specific neural circuits in the central nervous system. Previous studies using male-female mosaics and the flies with defects in particular brain structures implicated the critical central regions involved in male courtship behavior. However, their acute physiological roles in courtship regulation still largely remain unknown. Using the temperature-sensitive Dynamin mutation, shibire(ts1), here we demonstrate the significance of two major brain structures, the mushroom bodies and the central complex, in experience-independent aspects of male courtship. We show that blocking of synaptic transmission in the mushroom body intrinsic neurons significantly delays courtship initiation and reduces the courtship activity by shortening the courtship bout length when virgin females are used as a sexual target. Interestingly, however, the same treatment affects neither initiation nor maintenance of courtship toward young males that release courtship-stimulating pheromones different from those of virgin females. In contrast, blocking of synaptic transmission in a central complex substructure, the fan-shaped body, slightly but significantly reduces courtship activity toward both virgin females and young males with little effect on courtship initiation. Taken together, our results indicate that the neuronal activity in the mushroom bodies plays an important role in responding to female-specific sex pheromones that stimulate initiation and maintenance of male courtship behavior, whereas the fan-shaped body neurons are involved in maintenance of male courtship regardless of the nature of courtship-stimulating cues.  相似文献

Summary A developmental study at the electron microscopic level was conducted of the fat body cells of Hyalophora cecropia (L.). During the last larval instar the fat body increases in volume and the cells exhibit a well developed rough endoplasmic reticulum and protein bodies of diverse sizes. In the pupal fat body, the protein bodies appear to be enclosed by a double membrane and contain glycogen granules, ribosomes and mitochondrion-like structures. In addition, there are large lipid globules, cytolysomes and rough endoplasmic reticulum. The ultrastructure of the protein bodies suggests the development of large bodies by fusion of smaller protein bodies. Changes in fat body cell ultrastructure were followed during adult development and cytological evidence was obtained for the depletion of protein, glycogen and lipid in the female during this period. The female adult fat body cell contains free ribosomes, protein bodies, many mitochondria, a few lipid globules and glycogen granules. The male moth fat body cells have many mitochondria, a few glycogen granules, essentially no protein bodies, but an abundance of large lipid globules.Studies on the influence of egg maturation on the morphology of the fat body of Hyalophora gloveri (L.) revealed that ovariectomy of pupae yielded adults having more fat body than normal females, and that the fat body cells of the ovariectomized animals contained more glycogen, lipid and protein. Male pupae receiving ovarian implants developed into adults containing eggs and possessed more fat body than normal females but less than normal males. A full-length cDNA encoding a serine carboxypeptidase (designated SmSCP-1) was recovered from an ongoing salivary gland EST project of the wheat midge. The deduced 461-amino acid sequence had a putative signal sequence at the amino terminus, indicating it was a secreted protein. The protein shared homology with serine carboxypeptidases from other insects, mammals, plants, and yeasts. SmSCP-1 mRNA was expressed in all stages of development and detected in salivary gland and fat body tissues but not in midgut tissue. Expression analysis and quantitative real-time PCR assays in male and female wheat midges and the fat body tissue of adult midges revealed that SmSCP-1 was up-regulated nearly four-fold in the female midges compared to males and nearly two-fold in female fat body compared to male fat body. The wheat midge serine carboxypeptidase (SmSCP-1) most likely has a dual function. As a secreted digestive enzyme, it could play a role in mobilizing host-plant seed reserves for feeding larvae and as expressed in fat body could function as an exopeptidase in degradation of vitellogenin and/or in post-translational processing of other enzymes.  相似文献

The display of courtship behavior has evolved in response to sexual selection driven by competition to obtain mates. Sexually dimorphic mate selection rituals are likely controlled at least in part by genes with sex-biased patterns of expression. In Drosophila melanogaster, male courtship behavior has been well described and consists of a series of stereotyped behaviors. The takeout gene is predominantly expressed in males and affects male courtship behavior. In this study, we examine the patterns of expression and evolution in takeout and the family of related proteins. We show that a number of genes in the takeout gene family show male-biased expression in D. melanogaster, largely in non-reproductive tissues. Phylogenetic analysis reveals that this gene family is conserved across insects. As expected for genes with male-biased expression, we also find evidence of positive selection in some lineages. Our results suggest that the genes in this family may have evolutionarily conserved sex specific roles in male mating behavior across insects.  相似文献

【目的】探索长足大竹象Cyrtotrachelus buqueti Guerin-Meneville成虫的交配行为规律。【方法】通过室内饲养和野外观察相结合的方法,记录了完整的交配过程,比较了不同寄主及虫体大小间交配行为的差异。【结果】长足大竹象一次完整的交配包括示爱、抱对、插入输精和配后保护4个阶段,雌雄都有多次交配行为。室内试验证明,长足大足象完整交配时间为21.71±1.84 min,其中示爱时间为0.46±0.05 min,抱对时间为6.18±0.38 min,插入输精时间为8.36±0.62 min,配后保护时间为6.71±0.79 min。雄虫初次交配时,在示爱、抱对和插入输精时间上均显著长于有交配经历的雄虫(P0.05)。长足大竹象在不同寄主植物上的交配时间存在差异,在慈竹上的示爱时间(0.46±0.05 min)和插入输精时间(8.36±0.62 min)显著长于在唐竹、撑×绿杂交竹、孝顺竹和芦竹上的时间(P0.05);在孝顺竹上的配后保护时间显著长于在其他植物上的时间(P0.05)。长足大竹象成虫会选择最适体长的配偶进行交配,当雌虫体长≥3.3 cm和3.0~3.2 cm时,与不同体长的雄虫在示爱、抱对、插入输精和配后保护时间上都显著长于其他体长的雌虫(P0.05)。林间长足大竹象的交配行为与室内观察结果基本一致。【结论】研究结果有助于了解长足大竹象交配过程,也对研究该虫的繁殖行为学及行为控制技术提供了依据。  相似文献

Sexual dimorphism is often derived from sexual selection. In sexually dimorphic Drosophila species, exaggerated male structures are used for specific behaviors in male-to-male competition or courtship toward females. In Drosophila prolongata, a member of the melanogaster species group, males have enlarged forelegs whereas females do not. However, the adaptive role of the enlarged forelegs is unclear because little is known about the behavior of D. prolongata. In this study, the courtship behavior of D. prolongata was investigated in comparison with closely related species. Males of D. prolongata use their forelegs in a specific behavior, “leg vibration”, in which the male vigorously vibrates the female’s abdomen by extending his forelegs from in front of her. Leg vibration was observed immediately before “attempting copulation”, indicating that it has an adaptive role in the mating process. In contrast, leg vibration was not observed in closely related species. Because the large forelegs are necessary to accomplish leg vibration, it was suggested that the sexual dimorphism of D. prolongata forelegs is currently under the influence of sexual selection in courtship behavior.  相似文献

1. The effects of the nutritional quality of the adult diet (primarily protein content) on testis mass, body condition and courtship vigour were studied in a Hawaiian Drosophila, D. grimshawi , a lek-forming species under strong sexual selection. The primary goals of this study were to determine whether there is a trade-off between investment in reproductive and somatic tissues, and to examine whether this trade-off is influenced by quality of the adult diet.
2. Quality of the adult diet had a major influence on male body condition, courtship vigour and testis mass, but males varied in their investment patterns even within diet treatment.
3. Body condition, a measure of phenotypic quality, was significantly related to how much males invested in testis and body tissues, but the nature of the relationships differed between males fed high- or low-quality diets.
4. Paragonia volume was significantly smaller for adult males fed low-quality diets than for males fed high-quality diets. Nutrient-deficient diets apparently forced males to trade off investment in testes and body condition, and in body condition and courtship vigour, but nutrient-rich diets did not result in severe trade-offs.
5. Collectively, the results suggest that fluctuation in adult diet quality of male D. grimshawi might influence male reproductive quality in ways that might limit female reproductive output and/or be a factor for female choice in this species.  相似文献

Behavioral changes during early development provide useful insights into the internal mechanisms that generate complex behavior expressed by mature individuals. At the same time, social conditions during early adult phase can influence behavior in later stages of development even in holometabolous insects. In this study, age-dependent changes in courtship behavior and the effect of social conditions were examined in a fruit fly, Drosophila prolongata. Younger males showed lower mating activity and simpler courtship behavior. Mating activity reached a maximum level by 5 days after eclosion, whereas expression of complex courtship behavior was not yet fully developed at that time, suggesting that they are controlled by different mechanisms. When two males were maintained in the same vial, not only mating activity but also courtship complexity was reduced, demonstrating for the first time that preceding social experience, not current social conditions, influenced the complexity of male courtship. The effect of social experience was completely erased by 1 day of isolation, however, showing that social experience did not suppress or promote behavioral development itself. Rather, these results suggest that the observed effect of social experience was a plastic response of males that reduced investment in courtship effort by anticipating increased male–male competition.  相似文献

Male courtship behavior and displays influence female mating decisions, and therefore affect mating success in a diverse range of organisms. While there is substantial evidence confirming that females prefer males who invest more in courtship, less attention has been paid to the relative importance of individual behaviors, and the discrete sequences of courtship that result in mating success. The small hairy maggot blow fly, Chrysomya varipes, performs stereotyped courtship behaviors, involving orienting, tapping, waving, arching, wing vibration and mounting. This study aimed to quantify male investment in specific courtship behaviors, and compare courtship investment and behavioral transitions between males who gained mating success (successful males) and those that did not (unsuccessful males). Our results show that mating success was influenced by the behaviors orienting, tapping, arching and mounting. Behavioral transitions revealed a distinct pattern of behaviors leading to a mating attempt, and some differences were observed between successful and unsuccessful males. Overall, our findings suggest that female mating decisions were based on differences in specific male courtship behaviors. This detailed observational study has quantified multiple courtship behaviors for the first time in C. varipes, and highlights the importance of considering multiple behavioral traits when exploring the influence of male courtship on mating success.  相似文献

The water-soluble proteins of abdominal cuticle of larvae, pupae and adults subjected to electrophoresis, revealed a specific pattern for each stage. An antibody against an adult specific 18 kdaltons protein was produced. Electrophoresis analysis of SDS slab gel and Western-blots revealed that the 18 kdaltons adult protein is not present in the water-soluble fraction from haemolymph or fat body of pupae which secreted pre-ecdysial adult cuticle. Immuno-histochemical methods showed that the cross-reaction of the immune serum was limited to the apical part of the adult epidermis and to the cuticle lamellae which are not stabilized. Therefore, the 18 kdaltons protein is specific for the adult cuticle and originates in the epidermis.  相似文献

Dopamine is an important neuromodulator in animals and its roles in mammalian sexual behavior are extensively studied. Drosophila as a useful model system is widely used in many fields of biological studies. It has been reported that dopamine reduction can affect female receptivity in Drosophila and leave male-female courtship behavior unaffected. Here, we used genetic and pharmacological approaches to decrease the dopamine level in dopaminergic cells in Drosophila, and investigated the consequence of this manipulation on male homosexual courtship behavior. We find that reduction of dopamine level can induce Drosophila male-male courtship behavior, and that this behavior is mainly due to the increased male attractiveness or decreased aversiveness towards other males, but not to their enhanced propensity to court other males. Chemical signal input probably plays a crucial role in the male-male courtship induced by the courtees with reduction of dopamine. Our finding provides insight into the relationship between the dopamine reduction and male-male courtship behavior, and hints dopamine level is important for controlling Drosophila courtship behavior.  相似文献

Experimentally naive male Drosophila melanogaster respond to sexually immature males with intense courtship. However, this response decreases markedly in a short period of time, and "experienced" males then avoid further courtship with immature males for 4 hr. This subsequent inhibition of the courtship response is specific to immature males; the response to virgin females remains intact. This experience-dependent modification in courtship behavior is designated as "conditioned courtship." Seven mutant strains isolated for their inability to express avoidance conditioning (on criteria independent of courtship) were all found to be mutant with respect to expression of conditioned courtship. The potential application of this phenomenon to mosaic analysis of these mutations is posed. Other results indicate that immature males constitutively release a chemical signal that is sufficient for the expression of conditioned courtship. The interpretation of conditioned courtship as a component of fitness is discussed.  相似文献

为揭示和探讨中国大鲵(Andrias davidianus)繁殖前期的冲凉行为及其意义,在仿生态养殖环境条件下,采用定时定点观察法和全事件记录法,对雄性成鲵的冲凉行为过程及姿势进行了观察记录,并分析了冲凉行为与年龄、求偶行为、繁殖成功率之间的关系.繁殖前期雄性成鲵表现出冲凉行为,冲凉行为与求偶行为之间存在极显著的正相关,表现出求偶行为的6尾雄性成鲵有4尾成功参与了繁殖.冲凉行为可能促进了雄性成鲵精巢的发育,引发求偶行为,最终达到成功繁殖.  相似文献
