When microsomal membranes from maize (Zea mays L. cv. Clipper)coleoptiles were separated by isopyc-nic centrifugation on acontinuous 10–45% sucrose gradient, bafilomycin A1-inhibitedATPase activity co-localized with the activities of the tonoplastmarker-enzymes, nitrate-Inhibited ATPase and K+-dependent pyrophosphatase.Thus, bafilomycin A1 is a specific inhibitor of the vacuolarH+-ATPase of maize coleoptiles. Inhibition of the vacuolar H+-ATPaseby bafilomycin A1 was strictly dependent upon the concentrationof the enzyme present in the assay medium, suggesting a stoichiometricassociation between bafilomycin A1 and the vacuolar H+-ATPase.In tonoplast-enriched preparations, half-maximal inhibitionwas obtained at 43 pmol bafilomycin A1 mg–1 protein. BafilomycinA1 inhibited the vacuolar H+-ATPase in a simple non-competitivemanner: increasing bafilomycin A1 concentrations reduced theVmax, of the H+ -ATPase, but had no effect on its Km towardsATP. Maize (Zea mays L.) root plasma membranes purified by the aqueouspolymer two-phase technique have previously been shown to bevery low in tonoplast H+ -ATPase and H+ -PPase activities. Westernblots of a similar preparation showed that, compared to a microsomalfraction, there was practically no reaction with antibodiesto the tonoplast enzymes, but a strong reaction with an antibodyto the plasma membrane H+ -ATPase. Freeze/thaw treatment ofthe plasma membrane vesicles increased the proportion with aninsideout orientation to about 40%. This preparation was usedto demonstrate that substitution of KCl for K2S04 resulted ina 14-fold stimulation of H+ transport, but an increase in ATPaseactivity of less than 10%. In contrast to its effect on tonoplastvesicles, Cl had only a small effect on the membranepotential of plasma membrane vesicles, assayed by oxonol V fluorescencequench recovery. To account for the apparent variability inthe H+/ATP coupling ratio, it may be necessary to devise a modelthat takes into consideration the possibility of non-linearbehaviour with respect to the membrane potential of the protonleak and/or of slip in the ATPase. Young leaf segments of Zea mays L. seedlings were cultured onMurashige and Skoog's basal nutrient medium supplemented with2 mg l–1 2, 4-D and sub-cultured on medium containing8 mg l–1 2,4-D. Two types of callus tissues appeared—embryogenicand non-embryogenic. The embryogenic callus tissue producednumerous somatic embryos which on transfer to media containinglow amounts of 2,4-D or ABA produced plantlets. A field experiment was conducted to investigate the effect ofK nutrition under water stress conditions on cell membrane stabilitymeasured by the polyethylene glycol test, plant growth, internalplant water relations and solute and mineral concentrationsin maize (Zea mays L.). Water-stressed plants showed greateradaptation to water deficits at higher K levels. Cell membranestability increased, leaf water potential and osmotic potentialdecreased, turgor potential increased and stomatal resistancedecreased with increasing K nutrition. Osmotic adjustment wasevident and it may have been influenced by increased K+ concentrationsin leaf tissues with increasing K nutrition. Higher leaf thicknessand higher leaf water content were observed at higher K levels.Results suggested that higher supplies of K nutrition may increaseplant production during periods of water stress. Experiments were done to determine if the spontaneous recoveryof non-growing segments of corn root (Zea mays L.) from excisioninjury is dependent on auxin. Washing the segments with 5 runindoleacetic acid (IAA) for 2 to 4 hours gave a small but significantincrease in K+ (86Rb) influx, used here as a parameter reflectingrecovery of electrogenie H+-efflux pumping. This promotive effectwas obtained only after an hour of washing, and was sustainedby 100 nm gibberellic acid (GA3). Any early responses to auxinwere obscured by an adverse reaction of the root cells to externalIAA which resulted in a transitory inhibition of H+ pumpingand K+ influx. Pretreatment of excised root tips with 10 µM IAA in thegrinding medium protected a plasmalemma-enriched fraction ofthe microsomes during isolation, giving increased uncoupler-sensitiveATPase activity. Non-growing root tissue thus shows three responses to auxin:an adverse reaction at the outer surface of the plasmalemmawhich blocks H+ pumping; a protective or restorative effecton the H+-ATPase; an increased capacity for K+ influx duringthe developmental phase of washing, which is augmented by thepresence of GA3. Light-enhanced active pyruvate uptake into mesophyll chloroplastsof C4 plants was reported to be mimicked by either of the twotypes of cation jump: H+-jump in maize and phylogenically relatedspecies (H+-type) and Na+-jump in all the other C4 species tested(Na+-type) [Aoki, N., Ohnishi, J. and Kanai, R. (1992) PlantCell Physiol. 33: 805]. In this study, medium and stromal pH was monitored in the suspensionof C4 mesophyll chloroplasts. Medium alkalization lasting for5 to 10 seconds after pyruvate addition was detected by a pHelectrode and observed only in the light and only in mesophyllchloroplasts from H+-type species, Zea mays L. and Coix lacryma-jobiL., but not in those from Na+-type species Panicum miliaceumL., Setaria italica (L.) Beauv. and Panicum maximum Jacq. Theinitial rate of H+ consumption showed good correlation with[14C]pyruvate uptake measured by silicone oil filtering centrifugation,both being inhibited by N-ethylmaleimide and 7-chloro-4-nitrobenzo-2-oxa-l,3-diazole to the same degree. The ratio of the rate of H+ uptaketo that of pyruvate uptake was always about 1. Pyruvate-inducedacidification of the stroma was observed in maize mesophyllchloroplasts. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. Comparative analysis of the transport activity of proton pumps (plasmalemma H+-ATPase, vacuolar H+-ATPase, and vacuolar H+-pyrophosphatase) in the membrane preparations obtained from coleoptile cells of etiolated maize seedlings (Zea mays L.) was carried out. The highest level of vacuolar pyrophosphatase activity was observed during the early development of coleoptile cells under growth intensification through the elongation. The role of ATPase pumps of tonoplast and plasmalemma in the transport of hydrogen ions increases during further development. The plasmalemma activity in this process is higher. When the growth stops, the activity of proton pumps becomes significantly lower. Nevertheless, their substrate specificity and sensitivity to proton pump inhibitors do not change, which can be an evidence of physiological significance of pumps in the maintenance of cell homeostasis.  相似文献

The effects of NO-3 and NH+4 nutrition on hydroponically grownwheat (Triticum aestivum L.) and maize (Zea mays L.) were assessedfrom measurements of growth, gas exchange and xylem sap nitrogencontents. Biomass accumulation and shoot moisture contents ofwheat and maize were lower with NH+4 than with NO-3 nutrition.The shoot:root ratios of wheat plants were increased with NH+4compared to NO-3 nutrition, while those of maize were unaffectedby the nitrogen source. Differences between NO-3 and NH+4-fedplant biomasses were apparent soon after introduction of thenitrogen into the root medium of both wheat and maize, and thesedifferences were compounded during growth. Photosynthetic rates of 4 mM N-fed wheat were unaffected bythe form of nitrogen supplied whereas those of 12 mM NH+4-fedwheat plants were reduced to 85% of those 12 mM NO-3-fed wheatplants. In maize supplied with 4 and 12 mM NH+4 the photosyntheticrates were 87 and 82% respectively of those of NO-3-fed plants.Reduced photosynthetic rates of NH+4 compared to NO-3-fed wheatand maize plants may thus partially explain reduced biomassaccumulation in plants supplied with NH+4 compared to NO-3 nutrition.Differences in the partitioning of biomass between the shootsand roots of NO-3-and NH+4-fed plants may also, however, arisefrom xylem translocation of carbon from the root to the shootin the form of amino compounds. The organic nitrogen contentof xylem sap was found to be considerably higher in NH+4- thanin NO-3-fed plants. This may result in depletion of root carbohydrateresources through translocation of amino compounds to the shootin NH+4-fed wheat plants. The concentration of carbon associatedwith organic nitrogen in the xylem sap of maize was considerablyhigher than that in wheat. The clone encoded a 105-kDa polypeptide(956 amino acids) that was 79–85% identical in terms ofamino acid sequence to other plant H+-ATPases. Proton transport catalyzed by the nitrate-insensitive, vanadate-sensitive H+-ATPase in microsomes from maize (Zea mays L.) roots washed with 0.25 molar KI decreased as a function of time at 0 to 4°C. The rate of proton transport was approximately one-half of that by freshly isolated microsomes after 6 to 18 hours of cold storage. The decrease in proton transport coincided with losses in membrane phosphatidylcholine and was not associated with a change in vanadate-sensitive ATP hydrolysis. A technique based on a protocol developed for the reconstitution of Neurospora crassa plasma membrane H+-ATPase (DS Perlin, K Kasamo, RJ Brooker, CW Slayman 1984 J Biol Chem 259: 7884-7892) was employed to restore proton transport activity to maize microsomes. These results indicated that the decline in proton transport by maize root membranes during cold storage was not due to degradation of the protein moiety of the H+-ATPase, but was due to the loss of phospholipids.  相似文献

The extent by which salinity affects plant growth depends partlyon the ability of the plant to exclude NaCl. To study the uptakeof NaCl into excised roots of Zea mays L. cv. ‘Tanker’,two different techniques were applied. A root pressure probewas used to record steady state as well as transient valuesof root (xylem) pressure upon exposure of the root to mediacontaining NaCl and KCl as osmotic solutes. In treatments withNaCl, pressure/time responses of the root indicated a significantuptake of NaCl into the xylem. NaCl induced kinetics were completelyreversible when the NaCl solution was replaced by an isosmoticKCl solution. This indicated a passive movement of Na+-saltsacross the root cylinder. Root samples were taken at differenttimes of exposure to NaCl and prepared for X-ray microanalysis(EDX analysis). Radial profiles of ion concentrations (Na+,K+, Cl) were measured in cell vacuoles and xylem vesselsalong the root axis. Na+ appeared rapidly in mature xylem (earlymetaxylem) and living xylem (late metaxylem) before it was detectablein vacuoles of the root cortex. EDX results confirmed that thekinetics observed by the pressure probe technique correspondedmainly to an influx of Na+-salts into early metaxylem. In latemetaxylem, the uptake of Na+ was associated with a decline ofK+. The Na+/K+ exchange indicated a mechanism to reduce sodiumfrom the transpiration stream. Intact Zea mays L. kernels attached to cob tissue develop tomaturity when grown in vitro. This experiment was designed todetermine if it is possible to prolong kernel growth by refreshingthe culture medium. Blocks of maize kernels were grown in vitroon media containing several concentrations of sucrose. Kernels,at all concentrations of sucrose, developed to maturity at 30–35d post-pollination, indicating that it is not possible to extendthe kernel growth phase by supplying a carbohydrate source.Kernels grown on media containing 80 g 1–1 or higher sucroseconcentration had a significantly greater percentage of kernelsthat developed to maturity, and had greater weight and starchcontent per seed. Fusicoccin was shown to stimulate the ATP-driven, intravesicular acidification of liposomes reconstituted with crude fusicoccin receptors and the H+-translocating ATPase, both solubilized from maize (Zea mays L.) plasma membrane. The present paper reports optimal conditions for dual reconstitution and fusicoccin activation as well as the biochemical characterization of the effect of fusicoccin on this system. Fusicoccin stimulation of proton pumping was dependent on pH and fusicoccin concentration. Its specificity was demonstrated by the positive effect of two cotylenins that have a high affinity for fusicoccin receptors and by the negative response to 7,9-epideacetylfusicoccin, an inactive fusicoccin derivative. Kinetic measurements at different ATP concentrations showed that fusicoccin increases the Vmax of the enzyme. Fusicoccin stimulation of maize H+-ATPase was also maintained when receptors from maize were substituted by those from spinach (Spinacia oleracea L.).  相似文献

Exocytic insertion of H+-ATPase into the apical membrane of inner medullary collecting duct (IMCD) cells is dependent on a soluble N-ethylmaleimide-sensitive factor (NSF) attachment protein target receptor (SNARE) complex. In this study we determined the role of Munc-18 in regulation of IMCD cell exocytosis of H+-ATPase. We compared the effect of acute cell acidification (the stimulus for IMCD exocytosis) on the interaction of syntaxin 1A with Munc-18-2 and the 31-kDa subunit of H+-ATPase. Immunoprecipitation revealed that cell acidification decreased green fluorescent protein (GFP)-syntaxin 1A and Munc-18-2 interaction by 49 ± 7% and increased the interaction between GFP-syntaxin 1A and H+-ATPase by 170 ± 23%. Apical membrane Munc-18-2 decreased by 27.5 ± 4.6% and H+-ATPase increased by 246 ± 22%, whereas GP-135, an apical membrane marker, did not increase. Pretreatment of IMCD cells with a PKC inhibitor (GO-6983) diminished the previously described changes in Munc-18-2-syntaxin 1A interaction and redistribution of H+-ATPase. In a pull-down assay of H+-ATPase by glutathione S-transferase (GST)-syntaxin 1A bound to beads, preincubation of beads with an approximately twofold excess of His-Munc-18-2 decreased H+-ATPase pulled down by 64 ± 16%. IMCD cells that overexpress Munc-18-2 had a reduced rate of proton transport compared with control cells. We conclude that Munc-18-2 must dissociate from the syntaxin 1A protein for the exocytosis of H+-ATPase to occur. This dissociation leads to a conformational change in syntaxin 1A, allowing it to interact with H+-ATPase, synaptosome-associated protein (SNAP)-23, and vesicle-associated membrane protein (VAMP), forming the SNARE complex that leads to the docking and fusion of H+-ATPase vesicles. soluble N-ethylmaleimide-sensitive factor attachment protein target receptor; cell pH; acid secretion  相似文献

Young seedlings of Capsicum annum L., Commelina communis L.and maize (Zea mays L.) were subjected to a mild water-stressingtreatment and/or treated with abscisic acid (ABA). Plants rootedin soil received a soil-drying treatment and their leaves weresprayed with a 10–4 M solution of ABA. Plants grown insolution culture were stressed by the addition of polyethyleneglycol (PEG) to the rooting medium and ABA was also added tothe rooting medium, either with or without PEG. The effectsof both treatments on the growth of roots and shoots and theultimate root: shoot dry weight ratio were very similar. Shootgrowth was limited both by water stress and by ABA application;while there was some evidence that mild water stress and/orABA application may have resulted in a stimulation of root growth.More severe water stress reduced the growth of roots but theoverall effect of stress was to increase the ratio of rootsto shoots. The role of abscisic acid (ABA) in controlling growth and developmenthas been studied in maize (Zea mays L.) coleoptile segments.Application of ABA reduces the elongation rate by about 50%and affects ion fluxes. In particular, proton extrusion is decreasedwhile potassium efflux is greatly enhanced. Apparently, ABAdoes not: seem to influence calcium influx from the apoplastinto the cytosol, but more likely it influences its efflux.Alteration of cytosolic calcium concentration may also be obtainedby increasing its release from internal stores. This possibilitymight be sustained by the increased hydrolysis of phosphatidylinositolupon ABA application. Change in the balance of ion fluxes shouldresult from regulation of transport mechanisms at the membranelevel and should produce changes in the transmembrane electricalpotential. The H+- ATPase and the ATP-dependent calcium transportactivities are both influenced by the treatment with ABA, –55%and –40%, respectively. Maize plants, subjected to 0, 80, 120 and 160 meq l–1salinity using NaCl, showed adverse effects on viability, germinationand tube growth of pollen, besides enhancing the bursting ofpollen. The endogenous levels of various metabolites in pollenwere also affected. Pollen grains from salinized plants hadmore soluble carbohydrates, free amino acids, especially proline,phenols and DNA and less starch, protein and RNA compared tothe non-saline controls. Salinity also resulted in the accumulationof ions such as Na+, K+ and Cl while it caused a reductionin the boron content of pollen. These metabolic disturbancespossibly lead to decreased viability, germination and tube growthof pollen thereby resulting into a reduction in reproductivecapacity of the plants under salt stress. To study the effect of salt (NaCl) on root elongation we developeda device that measures this effect by means of a Linear VariableDifferential Transformer (LVDT). To test the efficacy of thedevice we performed experiments demonstrating that (a) ratesof elongation of primary maize (Zea mays L.) roots were comparableto elongation rates of primary roots growing freely in solutionculture; and (b) chilling and low O2 concentrations of the solutionelicited the expected responses. Inhibition of root elongation by 75 mol m–3 NaCl was gradual.At an iso-osmotic concentration, mannitol did not inhibit rootgrowth, suggesting that the inhibition was not due to osmoticfactors but rather to effects of salt on metabolism. The additionof supplemental Ca (10 mol m–3) ameliorated this stressfulcondition. Timing of the application of Ca was critical. Treatmentwith Ca after addition of NaCl only partially restored growth,but pretreatment with Ca completely prevented the inhibitionof growth by salt stress. The radial movements of Rb+ and Sr2+ as analogues for K+ andCa2+ were followed by electron probe X-ray microanalysis inaerenchymatous roots of maize (Zea mays L. cv. LG 11). The objectwas to determine the possible pathways by which ions can reachthe xylem when degeneration and lysis of much of the root cortexhas taken place during formation of numerous gas-filled spacesin aerenchymatous roots. After 1, 6 and 24 h uptake of Rb+ andSr2+ from a full strength nutrient solution containing K+ andCa2+, the distribution of these elements was examined. Transverseroot sections, prepared by cryostat sectioning and lyophilization,were used for electron probe X-ray microanalysis. If the cellwalls lining gas-filled spaces become suberized (as reportedin earlier literature), this development might be expected toretard ion movement, but we found that Rb+ or Sr2+ could migrateas readily in the radial wall residues remaining after corticalcell lysis, as in the walls of intact cortical cells. The distributionsof Rb+ and Sr2+ differed, however. The lack of a concentrationgradient for Rb+ across the root was compatible with its transportmainly in the symplast, constituted by occasional files of intactcortical cells bridging the gas-filled spaces. The evident concentrationgradient for Sr2+ was accounted for by its movement to the endodermisby the apoplastic pathway constituted by the walls of intactand lysed cells.
