The nutritional requirements of 43 strains of Haemophilus influenzae isolated from clinical and normal flora sources were investigated. Two defined minimal media were developed by modifying the medium of Herriott et al. (1970):74% of the strains could grow on the minimal media and Herriott's medium; the remaining strains could not grow on any of these media.  相似文献

Bacillus thuringiensis strains HD-73 and 4412, and two spontaneous mutants termed 4412aa-ind and 4412sph-cry were studied for the ability to produce crystals of different size and shape when grown in a rich medium and in an appropriate minimal medium defined during this study. Strain 4412aa-ind showed medium-dependent variation in the crystal phenotype. Scanning electron microscopy was utilized in order to show crystal variations in size and shape. B. thuringiensis strains 4412aa-ind and 4412sph-cry grown in rich and in minimal media produced differences in crystal morphology, and SDS-PAGE gel indicated that crystal variation was only at the morphological level and not in composition of the amino acids. A further nineteen B. thuringiensis strains were tested for the ability to sporulate in two simple defined media. Of these strains thirteen were able to complete sporulation with crystal production in one or both media. All strains grew and sporulated in a medium containing the usual twenty amino acids, and no vitamins or other growth factors were required.  相似文献

The growth of two strains of Lactococcus lactis subsp. lactis from vegetable (NCDO 2118) and dairy origin (IL 1403) were compared on various culture media. Both strains grew more rapidly on a complex organic medium than on a defined synthetic medium. The best growth was obtained under nitrogen gas phase. The single omission technique was applied to each component of a non-optimized synthetic medium in order to determine the true nutritional requirements. Requirements for macro-elements, oligo-elements, bases and vitamins were identical for the two strains. As expected, the dairy strain (IL 1403) was seen to be auxotrophic for some amino acids, whereas the vegetable strain (NCDO 2118) was seen to be prototrophic for all amino acids when using the single omission technique. Growth was then characterized on progressively simplified media and the composition of the absolute minimal media for the growth of both strains was defined. Sustained growth of the vegetable strain was only possible in minimal media supplemented with six amino acids (Glu, Met, Ile, Leu, Val, Ser), indicating that the definition of prototrophy/auxotrophy is partly dependent upon the medium composition. The dairy strain showed a requirement for Arg, His and Thr in addition to the six amino acids necessary for growth of the vegetable strain. The removal of ammonium salt from the medium did not affect the growth, illustrating that the amino acids may satisfy the totality of the nitrogen requirement for biomass synthesis.  相似文献

Protoplasts of nutritionally complementary strains of Aspergillus niger producing citric acid were treated in a polyethylene glycol solution, plated onto hypertonic minimal medium and the intraspecific fusants were obtained. When transferred and cultivated on minimal medium, almost all fo the fusants continued to grow as heterokaryons. However, some fusant colonies formed sectors of the prototrophic strains (sector strains). Most of these sector strains were haploid recombinants and their properties were genetically stable when subcultivated on both minimal and supplemented media. With respect of morphological features and citric acid productivity, the recombinant strains were like either of the parental strains or an intermediate between both parental strains. The rest of the sector strains that were nonconidiation typw seemed to be haploid recombinants, although they stopped growing on the third subcultivation on both the minimal and supplemented media.  相似文献

Minimal chemically defined media for Bacillus stearothermophilus were developed at 60°C and quantitative requirements for each nutrient were determined. A prototrophic strain of B. stearothermophilus was grown in medium containing only glucose and mineral salts whereas auxotrophic strains in addition required biotin, thiamine, nicotinic acid and DL-methionine. Metabolic interaction between L-valine and L-leucine was observed with auxotrophic organisms. The presence of L-leucine in minimal medium necessitated the addition of L-valine. Growth took place in the absence of both amino acids.  相似文献

Since RNAs lie at the center of most cellular processes, there is a need for synthesizing large amounts of RNAs made from stable isotope-labeled nucleotides to advance the study of their structure and dynamics by nuclear magnetic resonance (NMR) spectroscopy. A particularly effective means of obtaining labeled nucleotides is to harvest these nucleotides from bacteria grown in defined minimal media supplemented with 15NH4Cl and various carbon sources. Given the high cost of carbon precursors required for labeling nucleic acids for NMR studies, it becomes important to evaluate the optimal growth for commonly used strains under standard minimal media conditions. Such information is lacking. In this study, we characterize the growth for Escherichia coli strains K12, K10zwf, and DL323 in three minimal media with isotopic-labeled carbon sources of acetate, glycerol, and glycerol combined with formate. Of the three media, the LeMaster-Richards and the Studier media outperform the commonly used M9 media and both support optimal growth of E. coli for the production of nucleotides. However, the growth of all three E. coli strains in acetate is reduced almost twofold compared to growth in glycerol. Analysis of the metabolic pathway and previous gene array studies help to explain this differential growth in glycerol and acetate. These studies should benefit efforts to make selective 13C-15N isotopic-labeled nucleotides for synthesizing biologically important RNAs.  相似文献

AIMS: The aims of the present study were to: (i) analyse a group of antimicrobial agents and to select the most active against test microbial strains; (ii) test the effect of the antimicrobial treatment on air filters in order to reduce microbial colonization. METHODS AND RESULTS: Different kinds of antimicrobial agents were analysed to assess their compatibility with the production process of air filter media. The minimal inhibitory concentration for each antimicrobial agent was determined against a defined list of microbial strains, and an antimicrobial activity assay of filter prototypes was developed to determine the most active agent among the compatible antimicrobials. Then, the most active was chosen and added directly to the filter during the production process. The microbial colonization of treated and untreated filter media was assessed at different working times for different incubation times by stereomicroscope and scanning electron microscope analysis. Some of the antimicrobial agents analysed were more active against microbial test strains and compatible with the production process of the filter media. Filter sections analysis of treated filter media showed a significantly lower microbial colonization than those untreated, a reduction of species both in density and varieties and of the presence of bacteria and fungal hyphae with reproductive structures. CONCLUSIONS: This study demonstrated the ability of antimicrobial treatments to inhibit the growth of micro-organisms in filter media and subsequently to increase indoor air quality (IAQ), highlighting the value of adding antimicrobials to filter media. SIGNIFICANCE AND IMPACT OF THE STUDY: To make a contribution to solving the problem of microbial contamination of air filters, by demonstrating the efficacy of incorporating antimicrobial agents in the filter media to improve IAQ and health.  相似文献

Defined media, both solid and liquid, that support good growth of Bacillus stearothermophilus 1503 have been developed. Data are presented which indicate that manganese is required at relatively high concentrations for growth in a defined liquid medium. Phosphate concentrations higher than 5 times 10(-3) M have been shown to inhibit colony formation on solid media. Maximum viable counts of approximately 10(9) colony-forming units per ml were obtained in both the defined and minimal liquid media. Glucose, fructose, sucrose, glycerol, and starch support the growth of this obligate thermophile in the defined media, whereas citrate, alpha-ketoglutarate, succinate, fumarate, malate, acetate, and lactate do not. The described media have been utilized to isolate several amino acid-requiring mutants of B. stearothermophilus.  相似文献

In Escherichia coli, dihydrofolate reductase is required for both the de novo synthesis of tetrahydrofolate and the recycling of dihydrofolate produced during the synthesis of thymidylate. The coding region of the dihydrofolate reductase gene, folA, was replaced with a kanamycin resistance determinant. Unlike earlier deletions, this mutation did not disrupt flanking genes. When the mutation was transferred into a wild-type strain and a thymidine-(thy) requiring strain, the resulting strains were viable but slow growing on rich medium. Both synthesized less folate than their parents, as judged by the incorporation of radioactive para-aminobenzoic acid. The derivative of the wild-type strain did not grow on any defined minimal media tested. In contrast, the derivative of the thy-requiring strain grew slowly on minimal medium with thy but exhibited auxotrophies on some combinations of supplements. These results suggest that when folates are limited, they can be distributed appropriately to folate-dependent biosynthetic reactions only under some conditions.  相似文献

Salmonella typhimurium cultured with physiological levels of calcium are significantly enhanced in their ability to penetrate HeLa cells in vitro. Increased infectivity was not observed in magnesium-supplemented media, but was demonstrated in calcium-supplemented minimal defined or calcium-supplemented cation-deficient media. Invasion enhancement was observed for a number of S. typhimurium strains and ranged from 28–390% over calcium-deficient controls. Enhanced HeLa cell infectivity was not dependent on the presence of an autonomous 60-MDa plasmid.  相似文献

Strains of Saccharomyces cerevisiae transformed with a multicopy expression vector bearing both the Escherichia coli beta-galactosidase gene under the control of the upstream activating sequence of the GAL1-10 genes and the GAL4 activator gene release part of beta-galactosidase in the growth medium. This release is due to cell lysis of the older mother cells; the enzyme maintains its activity in buffered growth media. Fermentation studies with transformed yeast strains showed that the release of beta-galactosidase allowed an efficient growth on buffered media containing lactose as carbon source as well as on whey-based media. The transformed strains utilized up to 95% of the lactose and a high growth yield was obtained in rich media. High productions of ethanol were also observed in stationary phase after growth in lactose minimal media.  相似文献

ABSTRACT: BACKGROUND: Filamentous fungi are versatile cell factories and widely used for the production of antibiotics, organic acids, enzymes and other industrially relevant compounds at large scale. As a fact, industrial production processes employing filamentous fungi are commonly based on complex raw materials. However, considerable lot-to-lot variability of complex media ingredients not only demands for exhaustive incoming components inspection and quality control, but unavoidably affects process stability and performance. Thus, switching bioprocesses from complex to defined media is highly desirable. RESULTS: This study presents a strategy for strain characterization of filamentous fungi on partly complex media using redundant mass balancing techniques. Applying the suggested method, interdependencies between specific biomass and side-product formation rates, production of fructooligosaccharides, specific complex media component uptake rates and fungal strains were revealed. A 2-fold increase of the overall penicillin space time yield and a 3-fold increase in the maximum specific penicillin formation rate were reached in defined media compared to complex media. CONCLUSIONS: The newly developed methodology enabled fast characterization of two different industrial Penicillium chrysogenum candidate strains on complex media based on specific complex media component uptake kinetics and identification of the most promising strain for switching the process from complex to defined conditions. Characterization at different complex/defined media ratios using only a limited number of analytical methods allowed maximizing the overall industrial objectives of increasing both, method throughput and the generation of scientific process understanding.  相似文献

Glycine dependent Staphylococcus aureus var. bovis strains (TSCHAPE and RISCHE 1971) were tested for their ability to grow on glycine containing and glycine deprived media. We observed glycine dependence only in minimal media. In complete media the strains grew in absence of glycine. Testing the ability of some glycine precursors we found that in minimal media glycine could be replaced by threonine and in some cases by serine. One mutant (Gly 100 Glyox.r.) was able to metabolize glyoxylate instead of glycine. Aspartic acid was metabolized in presence of glycine. Using 14C-aspartic acid we detected 14C-glycine and 14C-threonine. Presumably the bacteria metabolize aspartic acid to glycine via threonine.  相似文献

Conventional complex media are routinely used to grow auxotrophic strains under the assumption that they can compensate the latter's nutritional deficiencies. We here demonstrate that this is not always true. This study compares the growth parameters of Saccharomyces cerevisiae (S288C) and its derived auxotrophic strains FY1679-14C and BY4741 in synthetic minimal medium (SD), standard YPD medium from two of the most commonly used suppliers, or modified YPD medium. Maximum specific growth rates of auxotrophic strains were slightly lower than the prototrophic case in all growth conditions tested. Also, the biomass production of auxotrophic strains in synthetic medium was slightly less than the prototrophic case. However in both of the two standard YPD media used, the biomass production of both auxotrophic strains was markedly lower than that of the prototrophic one. The extent of the differences depended on the medium used. Indeed in one of the two YPD media, the lower biomass production of auxotrophic strains was evident even at the diauxic shift. Uracil seems to be the main limiting growth factor for both auxotrophic strains growing in the two standard YPD medium tested. No YPD media or specific supplement was able to compensate for the effect of the auxotrophic mutations in the multiple auxotrophic marker strain BY4741. The fact that auxotrophic strains grew poorly on YPD when compared to their prototrophic counterpart indicates that standard YPD medium is not sufficient to overcome the effect of auxotrophic mutations.  相似文献

The organism requires 10 amino acids and one vitamin for growth, whilst three other vitamins are growth stimulatory. L-α-hydroxy acid analogues can replace eight, and fatty acid analogues four, of these amino acids. The organism may generate free energy by a variety of Stickland reactions. Most Stickland acceptors, but not glycine, stimulate the growth of this organism on glucose. Nonetheless, cells grown in the presence of glycine will reductively deaminate it. The media described support the growth of several other strains of this species. The simplified growth media which we have developed permit quantitative studies of the physiology of this organism.  相似文献

Fifty cultures belonging to the genus Clostridium were isolated from digesting sewage sludge and some of their hydrolytic and fermentative properties studied. The isolates could be divided into four loosely defined groups. Organisms belonging to the first group possessed little or no hydrolytic properties but produced acetic and iso -valeric acids in media containing minimal amounts of carbohydrate. The second (largest) group produced butyric acid in carbohydrate and non-carbohydrate media and were able to hydrolyse starch. The third group comprised non-hydrolytic strains which fermented carbohydrate to butyric acid; acetic, iso -butyric and iso -valeric acids were produced in media containing no additional carbohydrate. The final group, proteolytic organisms, produced very large quantities of several iso -acids in non-carbohydrate media. It is suggested that these organisms fulfil an important role in the ecosystem and that the properties described help to define that role.  相似文献

Two strains of Ruminococcus flavefaciens were studied. Each grew in a chemically defined minimal medium containing: minerals; ammonium sulfate as a nitrogen source; amino acids as a nitrogen source, a growth promotant(s) or as both; cellobiose as an energy and carbon source; isobutyric acid, isovaleric acid, carbonic acid, and bicarbonate as additional carbon sources; and biotin, thiamine, and tetrahydrofolic acid as vitamins. Tetrahydrofolic acid (5 ng/ml) served as a replacement for rumen fluid that was required in previous media tested for the growth of these bacteria. The present bacteria differ from many of the ruminococci previously studied in that they do not require either p-amino-benzoic acid or folic acid but do require tetrahydrofolic acid for maximum growth. Dihydrofolic acid and 5-methyltetrahydrofolic acid can substitute for tetrahydrofolic acid in minimal chemically defined medium. Thus, there must be extensive metabolic interaction between the microbes inhabitating the rumen, because the R. flavefaciens isolated had complex requirements for growth and yet was among the predominant bacteria in the rumen of cattle fed a simple vitamin B-deficient, nonprotein nitrogen, high-fiber, purified diet.  相似文献

Cell extracts of Pseudomonas aeruginosa strain PAO were found to contain pyruvate carboxylase activity. Specific activities were minimal when cells were grown on Casamino Acids, acetate, or succinate, but were three- to fourfold higher when cells were grown in glucose, gluconate, glycerol, lactate, or pyruvate minimal media. The reaction in crude cell extracts and in partially purified preparations was dependent on pyruvate, adenosine 5'-triphosphate, and Mg(2+), but was not affected by either the presence or absence of acetyl coenzyme A. Activity was nearly totally inhibited by avidin and this inhibition was substantially blocked by free biotin in incubation mixtures. Cell extracts were shown to fix (14)CO(2) in a reaction that had these same characteristics. Eight pleiotropic, carbohydrate-negative mutant strains of the organism were isolated after nitrosoguanidine mutagenesis. Each mutant strain grew normally in acetate, succinate, and citrate minimal media but failed to utilize glucose, gluconate, 2-ketogluconate, mannitol, glycerol, lactate, and pyruvate as sole sources of carbon and energy. These strains were found by quantitative transductional analysis with phage F116 to form a single linkage group. Cell extracts of each mutant strain were either lacking or severely deficient in pyruvate carboxylase activity. Spontaneous revertants of five of the eight strains were isolated and found to recover simultaneously both pyruvate carboxylase activity and the ability to utilize each of the C(6) and C(3) compounds. A second linkage group of similar mutant strains that grew on the C(3) compounds was found to contain normal levels of pyruvate carboxylase activity, but each strain was deficient in an enzyme of the Entner-Doudoroff pathway.  相似文献

A defined minimal sporulation medium for Clostridium acetobutylicum P262, which produces high levels of solvents, is described. The overall sporulation sequence was similar to that of other endospore-forming bacteria. However, we observed a presporulation stage, during which swollen phase-bright cells which contained large amounts of granulose formed. During sporulation, the initiation of spore coat formation occurred before the onset of cortex formation. Other Clostridium strains tested showed marked variations in ability to grow and sporulate in various minimal media.  相似文献
