The free, esterified and glycosylated sterols and triterpene esters of Sorghum vulgare grains were analysed by GLC. 4-Demethylsterols were found free, esterified and glycosidated. 4-Monomethyl-sterols were found completely free whilst the triterpenes were completely esterified. Three quarters of the total sterols in the grain were located in the embryo whilst the triterpenes were equally distributed between the embryo and endosperm.  相似文献

Excised barley embryos cultured on a nutrient medium containing methionine-[CD3] incorporated deuterium into the newly biosynthesized sterols. Two deuterium atoms were present in 24-methylenecycloartanol, 24-methylenelophenol and campesterol and a maximum of four deuterium atoms were incorporated into 24-ethylidenelophenol, stigmasterol and sitosterol. Mevalonic acid-[2-14C(4R)4-3H1] was utilized by the barley embryos to give 28-isofucosterol with a 3H-14C atomic ratio of 3:5 and stigmasterol and sitosterol with a 3H-14C atomic ratio of 2:5. 24-Methylenelophenol and 24-ethylidenelophenol were isolated from barley seed and 24-ethylidenelophenol-[2,4-3H3] was incorporated into sitosterol by barley seedlings. These results show that in the production of sitosterol a 24-ethylidenesterol intermediate is produced and it is suggested that this is isomerized to give a Δ24,(25) sterol prior to reduction to the saturated C29 sterol side chain.  相似文献

The free, esterified and glycosylated sterols and the pentacyclic triterpene esters of developing Sorghum bicolor grains were analysed by GLC and GC-MS. All the pentacyclic triterpenes were completely esterified but were not detected until 24 days after anthesis. Lupanol, multiflorenol, α-amyrin and isoarborinol were identified in the mature grains as components of the triterpene fraction but no 4,4-dimethylsterols could be found at any stage of development. A sixfold increase in total sterol per grain occurred during development. At 8 days after anthesis, 28-isofucosterol was found to be the second most abundant steryl ester. Campesterol was the major steryl glycoside and obtusifoliol was the major 4-monomethylsterol.  相似文献

A convenient method is described for the preparation of fucosterol-[7-3H2] and 28-isofucosterol-[7-3H2]. Both of these 24-ethylidene sterols, as well as 5α-stigmasta-7,Z-24(28)-diene-3β-ol-[2,4-3H4], were converted into the 24β-ethyl sterol, poriferasterol, by cultures of the chrysophyte alga Ochromonas malhamensis. However, fucosterol-[7-3H2] was not so efficiently incorporated as the other two compounds thus indicating that the configuration of the 24-ethylidene group is of some importance. It is suggested that a 24-ethylidene sterol of the Z-configuration is produced in de novo poriferasterol synthesis and that a Δ22,24(28)-diene may be an important subsequent intermediate.  相似文献

Although all parts of the geranium plant (Pelargonium hortorum) are capable of synthesizing sterols and triterpenes and their esters in vitro from mevalonic acid-[2-14C], the aerial portions are more active than other tissues. All plant parts were shown to incorporate mevalonic acid-[2-14C] into isoprenoids for at least 3 days. The leaves and petioles had the greatest incorporation on a wet weight basis. Chopped preparations showed comparable incorporations of mevalonate whereby rootlets incorporated about one half as much as most parts; the flower petals incorporated five times the average amount. In leaves the principal sterol synthesized was sitosterol. Metabolic studies with isolated leaves indicated a fairly rapid conversion of free tetracyclic triterpenes to 4-desmethyl-sterols, while β-amyrin was synthesized at a different rate than α-amyrin. Esterified tetracyclic triterpenes exhibited only a slight amount of conversion to 4-desmethylsterols.  相似文献

Sterols were extracted from two marine phanerogames, Posidonia oceanica and Cymodocea nodosa. The two plants contain 24α-ethyl sterols, while the 24α-methyl sterols are accompanied by 24β-epimers. The most abundant components are sitosterol, cholesterol and stigmasterol.  相似文献

Nine trimethylsilylated pentacyclic triterpenes were separated by GLC on an OV-101 column employing temperature programming. Characteristic retro-Diels-Alder fragmentation was observed in their mass spectra. The fragmentation patterns allowed individual characterization except for certain isomers which, nevertheless, were resolved by GLC, thus permitting their identification. Maytenus nemerosa has been fractionated systematically by following the toxicity of extracts, partitioned solutions and column chromatographic fractions against cultured KB cells. Several compounds were isolated in this fashion, including 3-oxo-20(29)-lupen-30-al, β-amyrin, 29-hydroxyfriedelan-3-one, 30-hydroxy-20(29)-lupen-3-one, 30-hydroxyfriedelan-3-one and lup-20(29)-ene-3β,30-diol, as well as tingenone, 20-hydroxytingenone and galactitol. 3-Oxo-20(29)-lupen-30-al was shown to be cytotoxic for the first time; the structural basis of this cytotoxicity was investigated, in part, by bioassay of four products obtained by chemical transformation of single, isolated principles.  相似文献

Seedlings of Euphorbia lathyris L. were grown in the dark at 25°C. Levels of amino acids, sugars and soluble phosphate in the endosperm increased upon germination after 4 days of imbibition, while the amounts of mineral reserves Mg2+ and K+ started decreasing. Ca2+ was not translocated from the endosperm to the seedling. Maximum values for amino acids were found on day 7, and the highest amounts of sugars were present on day 10. The endosperm was completely depleted by day 12.
Before germination (days 1–3) a low level of sterol synthesis in the embryo was detected with labeled sucrose and serine and to a lesser extent with labeled pyruvate. The label of [2-14C]-mevalonate proceeded exclusively to squalene. Laticifers started the synthesis of their triterpenes upon germination (day 4), using sucrose as a main substrate. A concurrent increase of sterol synthesis outside the laticifers was traced with labeled serine. Radioactive triterpenes, 4 α-methylsterols and sterols were detected in growing seedlings after [14C]-mevalonate uptake, but most of its label accumulated in squalene. The use of labeled mevalonate in sterol synthesis in growing seedlings is discussed.  相似文献

Lenormandia prolifera and Amansia glomerata contain large amounts of 24-methylenecholesterol. These two algae belong to the Amansia group of Rhodomelaceae as do Rytiphloea tinctoria and Vidalia volubilis, which are already known to be able to alkylate sterols at C-24. Halopythis pinastroides is an exception in the Amansia group, no C-28 sterols being present in this alga.  相似文献
