Diamine oxidase activity against putrescine and agmatine is investigated in young decotylized Glycine max plants growing, in the light, under sterile conditions on a liquid medium in the presence of ammonium chloride or nitrates. Optimal conditions for the enzyme activity are determined. Both amines are oxidised more slowly in plants cultivated on ammonium chloride than in plants grown on nitrates.  相似文献

A comparative study has been made of phenylalanine ammonia lyase (PAL) activity in plants sensitive or resistant to various herbicides (piclorame, methylchloro-phenoxyacetic acid (MCPA), atrazine). Piclorame, a herbicide with hormonal activity caused a large decrease in PAL activity of sensitive plants (Nicotiana tabacum), even at low concentrations (5 × 10-9M) whilst in resistant plants (Triticum aestivum) its effect is negligible; MCPA, also a herbicide with hormonal activity, similarly affects the activity of PAL, but only at higher concentrations. On the contrary, the action of atrazine, which has no hormonal activity, is lower and weaker, probably being only a secondary effect. Determinations of PAL activity during the photoperiod following piclorame application indicate that this herbicide influences principally the photodependent enzyme activity.  相似文献

Fate of Putrescine-1,4-14C in Glycine max Putrescine-1,4-14C was supplied to young decotylized Glycine max (L.) Merr. cv. Chippewa plants growing under aseptic conditions on a liquid medium with nitrogen supplied either as ammonium chloride or nitrates. Whatever the source of nitrogen the diamine was quickly transformed to γ-aminobutyric acid, succinic acid and malic acid; only a very minor part was utilized for the synthesis of polyamines. In the presence of ammonium chloride the putrescine catabolism may be slower than in the presence of nitrates. The results are explained by a weak isotopic dilution or by a diamine oxidase activity lower in “ammonium” plants than in “nitrate” plants; the two causes might co-exist. The possibility of a diamine compartmentation has to be considered.  相似文献

We have investigated the influence of juvenile hormone (JH) on the intestinal epithelium of G. mellonella, in vivo and in vitro. The larvae undergoing a supernumerary instar present a typical larval epithelium with columnar (CL) and globlet (CF) cells; the spinning period is characterized by a delay and a loss of synchronism in the process of differentiation of intermediates cells (Ci) typical of the pharate pupa. The larval-pupal intermediates show true mosaïcs in which Ci and CF are juxtaposed; however, the ratio of Ci in the epithelium progressively increases.The injection of JH at the beginning of spinning induces the appearance of CF just as Ci should normally grow. Hormone administration during the second half of the spinning period modifies the differentiation of epithelial cells: they become taller. We consider them to be cells engaged in pharate pupal differentiation, and which have then been partially oriented toward larval differentiation.These results show that the intestinal epithelium is a competent tissue, the sensibility of which to JH, is higher than that of the epidermis. The basal cell plasticity is very important and the action of JH on their differentiation may lead to CL or CF, to tall cells, and to Ci, depending on hormonal rate. In vitro, the experiments show that the action of JH is probably direct on the target tissue. The fact that JH can act very late as a modifier of the differentiation of the growing epithelial cells exclude the possibility that the hormone exercises its control through DNA replication.  相似文献

Pea Plants ( Pisum sativaum L. ev. Little Marvel) were grown in N-free medium and when well nodulated (28 days) were supplied for 8 days with nitrate or ammonium. Over the 8 days of nitrate treatment, total amino and amide N in sap declined, and the proportion of aspartate relative to the other amino acids increased. After 8 days of treatment, nitrogenase (EC 1.18.2.1) activity in nitrate-treated plants declined to about 30% of the activity in controls even though nodules were not directly in contact with nutrient solution. Nitrogenase activity was also decreased by the addition of ammonium chloride (10 m M ). With addition of nitrate or ammonium. clear signs of senescence began to show in the nodules after 4 days. Nitrate reductase (EC 1.6.6.1) activity was induced in roots by nitrate, but decreased sharply in nodules. In response to nitrate addition, newly formed root tissues showed 3- to 5-times higher glutamine synthetase (GS. EC 6.3.1.4) activity than newly formed tissues of control plants, expressed on a protein or weight basis. In complementary experiments, when ammonium salts were used instead of nitrates, the increase in GS activity was significantly lower. GS activity decreased in nodules of treated plants and total extractable protein was 3 times lower in nodules of nitrate-treated plants than in controls at day 8 of treatment.  相似文献

The frequency of plant transformation can be improved by addition of various chemical into transformation media. In the past, we showed that exposure of tobacco, wheat and triticale explants to ammonium nitrate, cerium and lantanium chloride and potassium chloride resulted in an increase in the frequency of transformation. Here, we tested whether a combination of increased concentrations of the aforementioned salts yielded a higher transformation frequency. We found that exposure to 61.8 mM ammonium nitrate caused a 5.0-fold increase in transformation frequency, whereas exposure to 1.0 μM cerium chloride or 47.0 mM potassium chloride resulted in 1.2- and 2-fold increases, respectively. Exposure to 61.8 mM ammonium nitrate and 1.0 μM cerium chloride led to a 4.8-fold increase in transformation frequency, whereas exposure to 61.8 mM ammonium nitrate and 47.0 mM potassium chloride let to a 5.2-fold increase. Finally, exposure to 61.8 mM ammonium nitrate, 1.0 μM cerium chloride and 47.0 mM potassium chloride produced a 5.1-fold increase. The analysis of the intactness of T-DNA borders showed that plants exposed to ammonium nitrate and a combination of ammonium nitrate with other salts had the more intact right borders and the less intact left borders. The best results were observed when all three salts (ammonium nitrate, potassium chloride and cerium chloride) were used. Thus, we concluded that the addition of cerium chloride and potassium chloride does not substantially improve the transformation rate beyond the improvement observed upon treatment with 61.8 mM ammonium nitrate, but may slightly improve the intactness of T-DNA borders.  相似文献

Starved Cerianthus lloydi Gosse were fed experimentally with Crangon muscles ground and impregnated with Bromocresol Green powder. A phagocytosis of the muscular fibres was detected in the cells of the endoderm layer of the sterile septa. In addition, through feeding with chicken blood, a phagocytosis became evident in the endoderm of sterile septa and of tentacles.The use of Ferritin allowed us to prove an uptake in all the endoderm cells of Cerianthus lloydi with an intensity which varies from one tissue to another. Furthermore, there is an unexpected pinocytosis in the ectoderm layer of the peristome, of pharynx, and the labial tentacles.Results obtained with bacteria and labelled amino acids are less conclusive. A mixture of C28 and C29 sterols have been isolated from Leptosphaeria typhae grown in vitro on “oat water” and characterized by GLC and MS. Mono-, di- and tri-unsaturated sterols are present in the extracts of fungi cultivated both in the dark and in the light but the sterol composition is different. The influence of “oat water” on sterol structure has been determined by comparison with the sterols of the same fungus grown on synthetic medium in the dark.  相似文献

The effect of low pH and aluminum on nitrogen uptake and metabolism was studied in roots of Lotus japonicus grown in hydroponic cultures. The low pH slightly suppressed root elongation, and this effect was accompanied by the suppression of nitrate and ammonia uptake, as well as the nitrate reductase activity. In spite of high resistance of young Lotus plants to short-term Al application, the one-day treatment of Al strongly reduced nitrate uptake and also the activity of nitrate reductase (NRA) in the apical parts of roots. The glutamine synthetase activity was also suppressed by Al treatment, but in lower extent. On the other hand, the ammonium uptake and nitrite reductase activity stayed unchanged by Al treatment and the values were practically the same as in control plants. These results support the view that nitrate uptake and nitrate reduction might be the main processes responsible for Al induced growth retardation in Lotus plants grown in mineral acid soils.  相似文献

We investigated whether Acetobacter diazotrophicus (syn.Gluconacetobacter diazotrophicus) could be recovered only from sugarcane plants either with low or no application of fertiliser N. We report here the enrichment and enumeration of A. diazotrophicus from high N-fertilised samples where high heterotrophic populations reduce the numbers of A. diazotrophicus ultimately diminshing its isolation frequency as reported earlier. The growth medium of micropropagated sugarcane seedlings of the varieties Co 8021, Co 86249, Co 86010, Co 86032, and Co 87025 was amended with potassium nitrate, ammonium nitrate, ammonium chloride and urea. The colonisation and AR activity of A. diazotrophicus were affected in the presence of high levels (25 mM) of ammonium chloride and ammonium nitrate but remained unaffected in low levels of N (i.e 1/10th of MS liquid medium) and with high levels of potassium nitrate (25 mM) and urea (500 ppm). A. diazotrophicus was detected in the inoculated plants both at low and high levels of N based on the amplification of a specific 16S rRNA gene fragment using PCR based method targeting a stretch of 445 bp with primers AC and DI. High levels of N in the growth medium induced morphological changes on A. diazotrophicus cells resulting in long pleomorphic cells. The percentage of pleomorphic cells was in the decending order from NH4NO3, NH4Cl, KNO3, and urea. These changes were more prominent in ammonium chloride and ammonium nitrate than potassium nitrate, urea and N free medium. The morphological changes and the increased heterotrophic populations may play a role on the survival ofA. diazotrophicus in high N-fertilised samples/environments.  相似文献

High-pressure liquid chromatographic determination of benfurodil hemisuccinate in blood and urineA sensitive and reliable method for quantitative determination of benfurodil hemisuccinate and benfurodil in plasma by high-pressure liquid chromatography on a Zorbax SIL column with a mean particle size of 7 μm and UV detection at 254 nm is described.Benfurodil hemisuccinate is stable in plasma but not in aqueous solutions. This is explained by its great fixation to plasma proteins which has been shown by equilibrium dialysis.  相似文献

When grown on a synthetic medium containing Zn2+ and Cd2+ in toxic concentrations and a high concentration of Mg2+, the mycelium of Aspergillus niger ATCC 9029 is pigmented yellow. Four pigments have been chromatographically separated. Two are still unknown; the others are asperenone and asperrubrol. Asperrubrol is the methyl ester of a dimethyl 13-phenyl-3-ol-2,4,6,8,10,12-tridecahexaenoic acid. One of the methyl groups is on C2 and the other is probably at C8. Asperrubrol has been obtained as the enol of the all trans molecule; ketoe nolisation can be obtained by action of heat or alkalis. When grown on the same medium, 13 of 17 strains of A. niger produced asperrubrol and asperenone.  相似文献

Analysis of macromolecular carbohydrates performed by gas liquid chromatography on Gregarina blaberae haemolymph showed the presence of hexoses (galactose, mannose, glucose), methyl-pentose (fucose), and hexosamines (N-acetylgalactosamine, N-acetylglucosamine). No trace of pentose or sialic acid or uronic acid was found. Mannose was the main neutral sugar. A change in the molar ratio of mannose consisting of an enrichment of female haemolymph occurred during larval-adult ecdysis. There was a parallel increase in glycoprotein staining with periodate-fuchsin after cellulose acetate electrophoresis of female haemolymph.  相似文献

It has been shown previously that 16-14C palmitic acid, 1-14C cetyl alcohol and 16,17-14C dotriacontane dissolved in sea water are rapidly incorporated into the oyster Ostrea gryphea L. and the sea anemone Calliactis parasitica Couch. In the present communication, we establish that palmitic acid and cetyl alcohol are slowly metabolized, but the hydrocarbon dotriacontane is unchanged after 48 h. The results are discussed.  相似文献

It has previously been shown that in Carausius the mechanisms ensuring normal, i.e. female, differentiation of the genital rudiment can be deflected towards male differentiation. A study of bicephalic monsters and microsurgical experiments show that the genital anlage is not affected by a lack or an excess of neurosecretory or allatal components. Reduced nicotinamide adenine dinucleotide phosphate-dependent nitrate reductase activity in crude extracts of Trichoderma virde was significantly inhibited by physiological concentrations of ammonium chloride, sodium chloride, and potassium chloride, but not by ammonium or sodium sulfate.
