All aging individuals will develop some degree of decline in cognitive capacity as time progresses. The molecular and cellular mechanisms leading to age‐related cognitive decline are still not fully understood. Through our previous research, we discovered that active neural progenitor cells selectively become more quiescent in response to aging, thus leading to the decline of neurogenesis in the aged hippocampus. Here, we further find that aging impaired dendrite development of newborn neurons. Currently, no effective approach is available to increase neurogenesis or promote dendrite development of newborn neurons in the aging brain. We found that systemically administration of 7, 8‐dihydroxyflavone (DHF), a small molecule imitating brain‐derived neurotrophic factor (BDNF), significantly enhanced dendrite length in the newborn neurons, while it did not promote survival of immature neurons, in the hippocampus of 12‐month‐old mice. DHF‐promoted dendrite development of newborn neurons in the hippocampus may enhance their function in the aging animal leading to a possible improvement in cognition.  相似文献

Evidence for contributions of airway smooth muscle (ASM) to the hyperresponsiveness of newborn and juvenile airways continues to accumulate. In our laboratory, 3 novel paradigms of hyperresponsiveness of newborn and young ASM have recently emerged using a guinea pig model of maturation in 3 age groups: 1 week (newborn), 3 weeks (juvenile), and 2-3 months (adult). The first paradigm includes evidence for a natural decline after newborn and juvenile life of the velocity of ASM shortening associated with a decrease in regulatory myosin light chain phosphorylation and a parallel decline in the content of myosin light chain kinase. Associated with the decrease in ASM shortening with age is an increase in the internal resistance to shortening. Dynamic stiffness is shown to relate inversely to the expression of myosin light chain kinase. This suggests that developmental changes in shortening relate inversely to the stiffness of the ASM early in shortening, suggesting a dynamic role for the cytoskeleton in facilitating and opposing ASM shortening. This relationship can be approximated as (dP/dt)max approximately (dP/dL)passive x (dL/dt)max (the maximal rate of increase of active stress generation approximately to the passive stiffness x the maximal shortening velocity). The second paradigm demonstrates that newborn ASM, unlike that in adults, does not relax during prolonged electric field stimulation. The impaired relaxation is related to changes in prostanoid synthesis and acetylcholinesterase function. The third paradigm demonstrates that, whereas oscillatory strain serves to transiently relax adult ASM, in newborns it induces (after the initial relaxation) a sustained potentiation of active stress. This is related to developmental changes in the prostanoid release. Together, these paradigms demonstrate that ASM contributes by multiple mechanisms to the natural hyperresponsiveness of newborn and juvenile airways. Future studies will elaborate the mechanisms and extend these paradigms to ASM hyperresponsiveness following sensitization in early life.  相似文献

The effects of hydrocortisone on newborn rat liver were studied by using biochemical assays, electron microscopy and quantitative morphometry. Hydrocortisone increased the number of lysosomes in the hepatocytes. Most of the lysosomes represented glycogen-containing autophagic vacuoles. The glucocorticoid also increased the activity of the liver glycogen-hydrolyzing acid glucosidase and the breakdown of glycogen inside lysosomes. The activity of the liver acid mannose 6-phosphatase was decreased. This may be related to the stimulation of autophagic mechanisms in the newborn rat hepatocytes.  相似文献

Cloning and developmental regulation of alpha 1 acid glycoprotein in swine   总被引:2,自引:0,他引:2  
A cDNA clone of porcine alpha 1 acid glycoprotein (alpha 1AGP) has been isolated and sequenced. Sequence homologies between porcine, human, and rat indicate that porcine alpha 1AGP is similar in structure to the rat and human proteins. RNA blots from days 40, 60, 80, and 110 fetal, newborn, and adult livers showed that alpha 1AGP mRNA is relatively abundant throughout fetal development, particularly at the later stages and in the newborn; there is a rapid decline in abundance following birth. From birth to 3 days of age, there is a three- to four-fold decline in abundance, and alpha 1AGP mRNA is approximately 100 times less abundant in the adult liver than in that of perinatal pigs. Southern blots showed that alpha 1AGP is probably a single-copy gene. The isolation of a cloned cDNA for porcine alpha 1AGP provides a tool to investigate the molecular mechanisms involved in the developmental regulation of the gene and to correlate changes in gene expression during development with fetal growth and well being.  相似文献

Ecosystem services are the numerous, essential processes that natural ecosystems provide free to human societies. Examples include the maintenance of breathable air; the movement, storage, and purification of water; the breakdown of wastes; and the provision of food, building materials, and medicines. However, the exponential increases in human population and concomitant environmental destruction make it likely that the level of ecosystem services available per capita will decline. There are three possible scenarios. First, if present practices continue, ecosystem services per capita will surely decline. Second, if a no-net-loss policy is implemented for habitats and species, ecosystem services per capita will still decline due to increases in human population, but the declines will be less precipitous. Third, if habitat is restored (including concomitant ecosystem services) at a rate exceeding that of destruction, then, perhaps the current level of ecosystem services per capita can be maintained, or even expanded to provide increased levels of ecosystem services per capita to more of the world's people.  相似文献

Electrical activity of the stellate ganglion was studied in newborn, 10-, 20-, 30-day-old, two- and six-month-old kittens using the spectral analysis. The development of sympathetic activity patterns was different during ontogenesis. The amplitude of discharges increased from the period of birth until the second month of kittens' life. In newborn and 10-day-old kittens, synchronous discharges of postganglionic fibers were represented by slow and low frequency impulses with frequencies of breathing and heart rate. ppears in 20-day-old kittens. Actin, one of the myofibrillar proteins of muscle, has one molecule of ADP very firmly bound to each subunit. In this study the firmly bound nucleotide has been used as a marker for actin synthesis in vivo , Myofibrils were prepared from limb muscles of mice of different ages, injected with 2.0 uCi/g body weight 3-adenosine and the specific activity of the bound ADP was determined following removal of unbound radioactivity by repeated washing. The adult rate of incorporation was low compared with the newborn rate. The results suggest that the rate of actin synthesis is high in the newborn but decreases markedly as the animal grows older. The amount of RNA associated with the myofibrils was also measured and was found to decline with age in a similar way to the rate of incorporation of label into actin.  相似文献

A serious shortage of nurses has developed since 1984 despite a growing number of employed nurses and a substantial decline in the number of hospital inpatient days. The evidence suggests that the shortage is the result of an increased demand for nurses, not a decline in supply. The increased demand in large part has resulted from the substitution of registered nurses for licensed practical nurses, aides, and other patient services personnel. The substitution was feasible because nurses'' wages have been depressed compared with those of other hospital employees. The shortage is likely to abate if nurses'' wages increase, making substitution more costly. Even in the absence of continuing wage increases, hospitals could ease the shortage by restructuring patient services and enabling nurses to spend a greater portion of their time in direct patient care.  相似文献

If climate change affects pollinator‐dependent crop production, this will have important implications for global food security because insect pollinators contribute to production for 75% of the leading global food crops. We investigate whether climate warming could result in indirect impacts upon crop pollination services via an overlooked mechanism, namely temperature‐induced shifts in the diurnal activity patterns of pollinators. Using a large data set on bee pollination of watermelon crops, we predict how pollination services might change under various climate change scenarios. Our results show that under the most extreme IPCC scenario (A1F1), pollination services by managed honey bees are expected to decline by 14.5%, whereas pollination services provided by most native, wild taxa are predicted to increase, resulting in an estimated aggregate change in pollination services of +4.5% by 2099. We demonstrate the importance of native biodiversity in buffering the impacts of climate change, because crop pollination services would decline more steeply without the native, wild pollinators. More generally, our study provides an important example of how biodiversity can stabilize ecosystem services against environmental change.  相似文献

Pollinators are key agents for ecosystems and humankind concerning biodiversity, agriculture, climate change adaptation, and all other ecosystem services. Particularly in industrialized countries pollinator diversity is in decline. The bulk of research is on entomological or plant‐pollinator network related topics, but the broad range of impacts of pollinator loss on coupled human and natural systems is not yet studied. As 87% of all flowering plants depend on pollinators, they are basic for all ecosystem services to some extent. Therefore, pollinator loss might cause simultaneous degradation of ecosystem services inducing counterproductive human responses and interlinked poverty spirals. The interaction of climate change, a main risk factor for pollinators, and unadvised human responses to pollinator decline are rarely studied. Tipping points of pollinator loss are not yet identified. Can counterproductive human responses to pollinator deficiency upscale pollinator decline toward a pollinator‐loss syndrome in the course of climate change? The article argues for research on the impacts of pollinator loss on other ecosystem services, useful and counterproductive human strategies on pollinator‐loss induced degradation, and the integration of pollinator protection into all terrestrial restoration efforts.  相似文献

Fatigue Profile, a new numerical method for characterising fatigue in isokinetic cycle ergometry is presented and compared with the conventional fatigue index (FI). The new method describes the temporal development of muscle fatigue based on the decline of peak power output throughout a whole trial. The advantage of this method is demonstrated by the analysis of two 25 s maximum trials, separated by 90 s recovery, performed by a well-trained athlete at a pedal frequency of 120 revolutions per minute. A fourth degree polynomial was fitted to model the peak power data. Using the polynomial model coefficients the first derivative represented the rate of changing peak power which represented the Fatigue Profile. The conventional FI was calculated as -35 Ws(-1) and -32 Ws(-1) for trials 1 and 2 respectively, indicating minor differences in fatigue between trials. In contrast the Fatigue Profile revealed important numeric and temporal differences between the trials. For trial 1 a maximum rate of peak power decline of -65 Ws(-1) was reached at approximately 6 s into the trial. In marked contrast, in trial 2, maximum rate of peak power decline (-146 Ws(-1)) occurred immediately. The Fatigue Profile approach allows the characterisation of the temporal development of fatigue under different experimental conditions and in combination with other techniques may yield further insight into the underlying mechanisms of fatigue.  相似文献

The harbour seal population in Orkney, off the north coast of Scotland, has reduced by 65% between 2001 and 2010. The cause(s) of this decline are unknown but must affect the demographic parameters of the population. Here, satellite telemetry data were used to test the hypothesis that increased pup mortality could be a primary driver of the decline in Orkney. Pup mortality and tag failure parameters were estimated from the duration of operation of satellite tags deployed on harbour seal pups from the Orkney population (n = 24) and from another population on the west coast of Scotland (n = 24) where abundance was stable. Survival probabilities from both populations were best represented by a common gamma distribution and were not different from one another, suggesting that increased pup mortality is unlikely to be the primary agent in the Orkney population decline. The estimated probability of surviving to 6 months was 0.390 (95% CI 0.297 – 0.648) and tag failure was represented by a Gaussian distribution, with estimated mean 270 (95% CI = 198 – 288) and s.d. 21 (95% CI = 1 – 66) days. These results suggest that adult survival is the most likely proximate cause of the decline. They also demonstrate a novel technique for attaining age-specific mortality rates from telemetry data.  相似文献

Pollinators for animal pollinated crops can be provided by natural and semi-natural habitats, ranging from large vegetation remnants to small areas of non-crop land in an otherwise highly modified landscape. It is unknown, however, how different small- and large-scale habitat patches interact as pollinator sources. In the intensively managed Argentine Pampas, we studied the additive and interactive effects of large expanses (up to 2200 ha) of natural habitat, represented by untilled isolated "sierras", and narrow (3-7 m wide) strips of semi-natural habitat, represented by field margins, as pollinator sources for sunflower (Helianthus annus). We estimated visitation rates by feral honey-bees, Apis mellifera, and native flower visitors (as a group) at 1, 5, 25, 50 and 100 m from a field margin in 17 sunflower fields 0-10 km distant from the nearest sierra. Honey-bees dominated the pollinator assemblage accounting for >90% of all visits to sunflower inflorescences. Honey-bee visitation was strongly affected by proximity to the sierras decreasing by about 70% in the most isolated fields. There was also a decline in honey-bee visitation with distance from the field margin, which was apparent with increasing field isolation, but undetected in fields nearby large expanses of natural habitat. The probability of observing a native visitor decreased with isolation from the sierras, but in other respects visitation by flower visitors other than honey-bees was mostly unaffected by the habitat factors assessed in this study. Overall, we found strong hierarchical and interactive effects between the study large and small-scale pollinator sources. These results emphasize the importance of preserving natural habitats and managing actively field verges in the absence of large remnants of natural habitat for improving pollinator services.  相似文献

The newborn of 4 pregnant female rats were collected within 24 h of birth. Twelve neonates were used for the study of chromaffin reaction, by both the direct and the indirect methods. The medullary cells could be demonstrated at this stage; however, the demarcation between noradrenaline and adrenaline cells was not quite sharp. Application of formaldehyde-induced fluorescence to the newborn rat adrenal gland was highly significant. A clear distinction could be demonstrated between noradrenaline and adrenaline cells, the former giving off strong, white-green fluorescence and the latter being weakly green-fluorescent. A third type giving off moderate green fluorescence was interposed between the cells and possibly represented an equivocal cell type at this stage. Sympathetic nerves could be demonstrated in relation to blood vessels and medullary cells.  相似文献

The stellate ganglion neurons' transmitter composition in the rat siblings of different age (newborn, 10-, 20-, 30-, 60-day old) revealed that the greatest amount of the neurons in all the rats contained thyrosinhydroxylase. Most cholinacetyltransferasepositive. In 30- and 60-day old rats, just a few cells contained both above enzymes. Since birth, the percentage of cells containing thyrosinhydroxylase and neuropeptide Y has been growing. Along with that, the share of somatostatin-positive neurons was decreasing. The percentage of the VIP-positive cells and the neurons containing cholinacetyltransferase was increasing until the 10th day of life and then dropped again. The somatostatin-positive neurons in all the rat siblings were represented by small cells, whereas the neurons containing cholinacetyltransferase were represented by large cells. The maturation of the neurotransmitter set in the rat stellate ganglion terminates by the end of the second month of life.  相似文献

选取白洋淀8个采样点于2005年9月—2006年12月对轮虫群落进行定性定量分析,共检出轮虫110种,采蒲台种类数最多,其次是枣林庄,南刘庄最少;从季节来看,春、秋轮虫种类数量最多,夏季其次,冬季最少;根据优势种比较,臂尾轮虫科、迈氏三肢轮虫、针簇多肢轮虫、龟甲轮虫属等甲型-中污性和乙型-中污性水体种类分布在污染较严重的南刘庄,而单趾轮属、腔轮属、鬼轮属等清洁种类则分布在采蒲台、枣林庄、烧车淀等淀区。除端村外,各采样点轮虫密度和生物量于2006年4月出现最大值。根据Q(B/T)指数评价白洋淀各采样点水质,南刘庄属富营养,圈头属寡营养,其它样点属中营养水质。白洋淀轮虫的Shannon-Wiener指数以及均匀度指数则在夏秋季较高,春季较低,而物种多样性指数并不能完全反映白洋淀各采样点的水质。  相似文献

One of the central justifications for the conservation of biodiversity is the notion that species diversity is essential for the maintenance of ecosystem services. However, an important observation overlooked by proponents of this argument is that most ecosystem services are provided not by whole ecosystems, but by any group of species that fulfils certain basic functional criteria. Distinguishing between services that are resilient in response to species decline, and those that are not, is a far less challenging task than identifying the precise influence on ecosystem functioning of rare species. Conservationists have been almost unanimous in their failure to acknowledge this distinction between resilient and sensitive ecosystem services. Not only does this threaten the credibility of conservation science, but also increases the likelihood that natural area management becomes hijacked by the demand that ecosystem service provision be made the dominant management criteria.  相似文献

The aging gut     
A spectrum of changes occurs in the function of the gastrointestinal tract throughout the life of the animal. With aging, there is a decline towards newborn levels of the villus surface area and brush border membrane markers, but the absorption of some nutrients continues to increase (such as glucose and vitamin), whereas the absorption of some nutrients falls (such as cholesterol) and fatty acids). Clearly there is a distinction between age-related alterations in intestinal form and function, and some of the enzyme- or carrier-mediated changes are substrate specific. With aging, the structure and brush border membrane composition of the intestine tends to regress towards newborn levels. It remains to be clarified whether these changes are a benefit to the animal, whether they represent a degeneration of normal function, and whether these changes are the cause or the effect of various age-related alterations in metabolic and physiological function.  相似文献
