Summary Germinating spores of the fungus Botryodiplodia theobromae incorporated guanine-8-C14 into both the nuclear DNA and mitochondrial DNA fractions. Ethidium bromide inhibited the synthesis of mitochondrial DNA without having a significant effect on nuclear DNA synthesis or on the rate and extent of spore germination. Rates of leucine and uracil incorporation and of oxygen uptake were not significantly affected by ethidium bromide until germination was nearly completed. The antifungal activity of bongkrekic acid against 17 tested molds was determined. Bongkrekic acid prevented spore germination and mycelial proliferation of Aspergillus niger, Rhizopus oryzae and Penicillium italicum. The action of bongkrekic acid was fungicidal. Under these conditions, the incorporation of 14C-leucine and 14C-uracil into the perchloric acid insoluble material of germinating A. niger conidia was significantly reduced by bongkrekic acid. Respiratory activity of resting spores was not affected by bongkrekic acid. Respiratory activity of germinated spores was inhibited by bongkrekic acid to the extent of 30 to 60% of controls for A. niger, R. oryzae and P. italicum. It has been concluded that operation of adenine nucleotide translocation in mitochondria of tested fungi is obligatory both for normal spore germination and fungal growth.  相似文献

Summary The pattern of RNA synthesis during maturation and germination of pollen grains ofHyoscyamus niger was studied using3H-uridine autoradiography. Incorporation of label during pollen maturation was periodic with peak RNA synthesis occurring in the uninucleate, nonvacuolate pollen grains and in the vegetative cell of the bicellular pollen grains. During the early stages of germination, isotope incorporation occurred predominantly in the nucleus of the vegetative cell with little or no incorporation in the generative cell. With the appearance of the pollen tube, incorporation of3H-uridine in the vegetative cell nucleus decreased and completely disappeared at later stages of germination. No incorporation of isotope was observed in the sperms formed in the pollen tube by the division of the generative cell. From a comparison of the results of this study with those of previous works on RNA synthesis during pollen embryogenesis in cultured anthers ofH. niger, it is concluded that in contrast to embryogenic development, there is no requirement for sustained RNA synthesis by the generative cell nucleus for normal gametophytic development.  相似文献

The photoreactive analogs of ethidium bromide (ethidium mono- and diazide) have been developed as drug probes to determine the actual molecular details of ethidium bromide interactions with DNA. In an effort to demonstrate that the analogs in fact mimic the parent ethidium, competition experiments were designed using 3H thymidine-labeled DNA in intact Salmonella TA1538, which is reverted by the azide analogs. 14C-labeled ethidium azide analogs were used in combination with the non-labeled ethidium bromide. The results presented here demonstrate that the parent ethidium competes with the azide analogs as a DNA intercalating drug using CsCl density gradient ultracentrifugation.  相似文献

Aspergillus nigerconidia are characterized by exogenous dormancy: the first stage of their germination is accomplished in twice-distilled water. However, germ tube formation requires the availability of carbon and nitrogen sources. Exogenous dormancy in A. nigerconidia exhibits the following peculiar features: (i) nitrogen-containing substances are active stimulators of germination; (ii) temperature-dependent changes in the lipid bilayer and in the neutral lipid composition of conidia are virtually identical to those occurring in growing mycelium under temperature stress; and (iii) the spore viability threshold does not exceed 45°C; i.e., the spores are more heat-resistant than the mycelium, but they are less heat-resistant than the spores that are in the state of endogenous dormancy. According to the current classification of the types of cell metabolism arrest, the exogenous dormancy of A. nigerconidia resembles the pattern of metabolism characteristic of vegetative cells during the idiophase.  相似文献

Berberine iodide, an isoquinoline alkaloid was isolated fromFumaria indica and its efficacy was tested against spore germination of some plant parasitic as well as saprophytic fungi. The alkaloid significantly curbed spore germination ofCurvularia lunata, Erysiphe cichoracearum, E. pisi, Fusarium udum andPenicillium species. Complete inhibition (100%) of spore germination was observed inE. cichoracearum andPenicillium species at 1.5 g/L.Albugo candida andAlternaria species were not affected by the chemical.  相似文献

Factors associated with conidial germination and appressoria formation ofGlomerella cingulata causing the brown blight disease of tea (Camellia sinensis) were studiedin vitro. Spore germination and appressoria formation were optimum at a temperature of 25°C, pH 5.0, 7 h light/day regime and a 24-h incubation period. At a concentration of conidia of 1200/μL 10-d-old culture,G. cingulata exhibited a maximum germination and appressoria formation. A maximum production of lesions was also evident on detached tea leaves at this spore concentration and in diffuse light. Diffusates of a phenolie nature collected from tea varieties susceptible and resistant toG. cingulata inhibited spore germination and appressoria formation. Diffusates from resistant varieties were more fungitoxic than those from susceptible varieties. Some phenolics known to be present in tea leaves, when testedin vitro, exhibited varying degrees of fungitoxicity. Pyrogallol totally inhibited spore germination, while pyrocatechol and phloroglucinol completely inhibited appressoria formation.  相似文献

Roots and aerial shoots ofZygophyllum coccineum L.,Z. album L. andZ. dumosum Boiss were extracted with solvents of increasing polarity and tested against spore germination ofVerticellium albo-atrum andFusarium oxysporum f.sp.lycopersici. The effectiveness of each extract depends on the solvent, plant species, organ and test fungus. Petroleum ether extracts were ineffective as fungistatics while the methanol extracts possessed a high inhibitory effect toward spore germination suppression, depending on the fungus and plant species. Water or ethyl acetate root extracts ofZ. album caused a marked suppression of spore germination of the test fungi. Six saponins were isolated from the effective extracts on silica gel and Sephadex columns. They were identified by acid hydrolysis, alkaline saponification, IR and FAB MS. Quinovic acid was the dominant aglycone. Kaempferol was isolated from the flavonoid fraction ofZ. dumosum for the first time. Some of the isolated saponins and kaempferol provided successful control against the test plant pathogens under greenhouse experiment when used as seed treatment.  相似文献

We have isolated a single step spontaneous mutant of S. cerevisiae resistant simultaneously to oligomycin, venturicidin, chloramphenicol, cycloheximide and triethyltin. This multiple drug resistance results from the interaction of two genetic factors showing both chromosomal location and episomal characteristics. One factor (π) confers oligomycin resistance, the other (τ) confers the other resistances. π can be lost spontaneously while τ can be completely eliminated with ethidium bromide. All π+ strains, whether grande or petite, τ+ or τ?, carry a covalently closed circular DNA while π? strains are devoid of it. We hypothesise that this circular DNA may play an informational role in the biogenesis and/or function of membranes.  相似文献

The initiating mechanism in the germination of Bacillus thiaminolyticus spores was studied with 14C-L -alanine. A characteristic pattern of incorporation of L -alanine into the spores was observed during the early stages of germination with two incorporation peaks, one occurred just after contact with L -alanine (first incorporation) and the other 5 min later (second incorporation). L -Glutamine, L -valine, or L -serine substituted for the incorporation of L -alanine during the first stage of germination. Although, L -alanine taken up during the first incorporation phase was extractable with trichloroacetic acid (TCA), that taken up during the second incorporation phase was not extractable. The distribution of radioactivity showed that incorporated L -alanine was located in the spore coat, mainly in the paracrystal fraction. The radioactive material which remained in the germination medium or was extractable from the spore coat fraction with TCA treatment or pronase digestion was identified as alanine. Significance of incorporation of L -alanine and its location in the spore in reference to the initiation of germination is discussed.  相似文献

Summary Evans' tube and cellophane agar-disc techniques were used to evaluate the colonization capacity of the soil microbes and fungistasis, respectively. Spore germination ofAlternaria tenuis, Curvularia geniculata, Helminthosporium rostratum andPestalotia sp. was studied on a natural soil. Aspergillus fumigatus was the pioneer colonizer, followed in the order of decreasing colonizing capacity by,A. flavus, A. niger, A. terreus andPenicillium nigricans. Sensitivity of test spores to soil fungistasis declined in the order,C. geniculata → Pestalotia sp. →A. tenuis → H. rostratum. The fungistatic level of the soil samples decreased gradually from side arms 1 to 7 of the Evans tube, inoculated with unsterilized soil from one end. Studies conducted with an inoculum of four soil fungi (A. fumigatus, A. flavus, A. niger, andP. nigricans) as well as those involving fumigation of Evans tube by CS2 also proved the greater colonizing capacity ofA. fumigatus. The level of fungistasis followed closely the colonization pattern of this fungus. When added individually to the soil, all fungi were able to inhibit spore germination of test fungi to some degree or other.  相似文献

Summary The effect of spore concentration on spore germination and germtube growth ofTrichoderma hamatum on water agar and on potato dextrose agar (PDA) was studied. Increasing inoculum size up to 109 spores/plate on PDA and up to 107 spores/plate on water agar shortened the incubation period required for germtubes emergence and increased germination rate. However, on water agar germination was inhibited at 108 and was completely arrested at 109 spores/plate. Inhibition in germination of 107 spores/plate was observed on water agar when the plates were preincubated with 109 spores/plate for 5 h or more. Addition of glucose and ammonium nitrate to the water agar medium allowed only 25% of the spores to germinate at 109 as compared to 78% at 107 spores/plate after 8 h of incubation. Addition of polysaccharides to the C+N supplemented medium, significantly increased germination up to 84% as compared to 100% on PDA, after 8 h of incubation. Germlings ofTrichoderma hamatum phialospores exhibited positive autotropism and anastamosis on both media. The phenomenon was positively related to inoculum size, being most pronounced at 107 spores/plate.  相似文献

Summary Chloroplast activities of dark-imbibed (non-germinating) and photoinduced (germinating) spores of the sensitive fern,Onoclea sensibilis were compared to gain insight into the germination process. There were no changes in the number of chloroplasts or in the chlorophyll contents of the spore during dark-imbibition and during the early phase of germination. Levels of increase in the Chloroplast DNA content of dark-imbibed and photoinduced spores were nearly the same and were associated with autoradiographic incorporation of [3H]thymidine into the cytoplasm. However, incorporation of the label into the nucleus occurred only during photoinduction of spores. Analysis of Chloroplast and nuclear DNA contents by dot-blot hybridization with labeled gene-specific probes has confirmed that chloroplast DNA content of the spore increases during dark-imbibition and photoinduction, while increase in nuclear DNA occurs only in photoinduced spores. Chloroplasts isolated from dark-imbibed and photoinduced spores incorporated [3H]TTP into an acid-insoluble fraction identified as DNA. The results show that physiological activities of chloroplasts of dark-imbibed and photoinduced spores ofO. sensibilis are similar and support an exclusive role for nuclear DNA synthesis in spore germination.  相似文献

To test the suitability of DNA stable isotope probing (DNA-SIP) for characterizing bacterial spore populations in soils, the properties of Bacillus subtilis cells and spores intensely labeled with [13C]glucose were characterized. Spore germination, vegetative growth rates, and sporulation efficiency were indistinguishable on glucose versus [13C]glucose, as were spore wet heat and UV resistance. Unlabeled and 13C-labeled spores contained 1.0989 and 74.336 at.% 13C, and exhibited wet densities of 1.356 and 1.365 g/ml, respectively. Chromosomal DNAs containing 12C versus 13C were readily separated by their different buoyant densities in cesium chloride/ethidium bromide gradients.  相似文献

Pretreatment with ethidium bromide (5 μg/ml) followed by a water wash had no effect on unheated Bacillus subtilis spores, but the viability of these spores after heating was much lower than that of similarly heated spores exposed to water alone. The fate of water- or ethidium bromide-treated spores, unheated or heated, was followed by allowing them to germinate and outgrow in a minimal or a complex liquid medium. Spores exposed to ethidium bromide and then heated (85°C, 10 min) exhibited a developmental block during germination and outgrowth. Many of them were blocked at the stage when the bacterium emerged from the germinated spore. When 0.35 μg of ethidium bromide per ml was added to heated spores in the germination-growth medium, the outgrowth of heated spores was inhibited to the same extent as were pretreated spores. Ethidium bromide acted in the first hour of germination of heated spores since addition after this time was ineffective in inhibiting recovery events. Repair of heat-damaged spore DNA was detected during the first 2 h of germination. The addition of ethidium bromide (final concentration, 0.35 μg/ml) inhibited DNA repair during early outgrowth. Increased sensitivity of spores to heat after pretreatment with sublethal concentrations of ethidium bromide was due to the inhibition of the repair of heat-damaged DNA.  相似文献

The surface structure of conidia and arthrospores ofMicrosporon 2)racemosum andTrichophyton winacei is described. Apart fromMicrosporon macroconidia, the spore surfaces displayed little or no ornamentation. The arrangement of verrucae on the surface ofMicrosporon conidia appears to be variable, but may be of little taxonomic importance other than as a characteristic of the genus.
Zusammenfassung Die Oberflächenstruktur der Konidien und Arthrosporen vonMicrosporon racemosum undTrichophyton erinacei sind beschrieben. Abgesehen von den Makrokonidien vonMicrosporon zeigten die Oberflächen der Sporen geringe oder keine Ornamentation. Die Anordnung der Warzen an der Oberfläche von Makrosporenkonidien erscheint wechselvoll und sie mag von geringer Wichtigkelt sein, außer der Charakterisierung der Gattung.

Zusammenfassung Für Infektionsversuche und biologische Bek?mpfung k?nnen Massenkulturen vonBeauveria bassiana entweder Konidien oder Blastosporen liefern. Die qualitative Bewertung solcher Sporen und Sporenpr?parate und ebenso deren Standardisierung setzen voraus, dass auch das Keimverhalten der Sporen bekannt ist. Deshalb wurden die zeitabh?ngigen Keimungsprozente und Keimungskurven der Blastosporen und Konidien eines Stammes vonB. bassiana auf Malzextrakt-Pepton-Agar bei 24 und 18°C ermittelt und damit auch die Unterschiede, die durch Temperatur und Sporenart bedingt sind. Für die Bewertung und Standardisierung von Sporen und Sporenpr?paraten k?nnen die Keimprozente nach 24 und 36 Stunden oder die Zeit für 50% Keimung Kriterien sein und mit den hier wiedergegebenen Standardwerten verglichen werden.
Summary Mass cultures ofBeauveria bassiana provide either conidia or blastospores for laboratory experiments and biological control measures. For qualitative evaluation of spores and spore preparations it is of primary importance to know the germination behaviour of spores. Therefore, the percentage germination of blastospores and conidia in relation to time (0–36 hours) and temperature (24 and 18°C) have been determined on malt-extract peptone agar (Table 1) and demonstrated by graphs (Fig. 1). The periods of time necessary for 50% germination of blastospores and conidia show differences depending on temperature and type of spore (Table 2). Pronounced differences of percentage germination as caused by the two temperatures become obvious with blastospores after 6–10, with conidia after 15–20 hours; after 24 hours, such differences are relatively low (Table 3). For evaluation and standardization of spores and spore preparations ofB. bassiana the percentage germination at 24°C as obtained after 24 and 36 hours or the period of time for 50% germination can be used as criteria and compared with the corresponding standard values presented.

The growth of the filamentous fungus Aspergillus niger, a common food spoilage organism, is inhibited by the weak acid preservative sorbic acid (trans-trans-2,4-hexadienoic acid). Conidia inoculated at 105/ml of medium showed a sorbic acid MIC of 4.5 mM at pH 4.0, whereas the MIC for the amount of mycelia at 24 h developed from the same spore inoculum was threefold lower. The MIC for conidia and, to a lesser extent, mycelia was shown to be dependent on the inoculum size. A. niger is capable of degrading sorbic acid, and this ability has consequences for food preservation strategies. The mechanism of action of sorbic acid was investigated using 31P nuclear magnetic resonance (NMR) spectroscopy. We show that a rapid decline in cytosolic pH (pHcyt) by more than 1 pH unit and a depression of vacuolar pH (pHvac) in A. niger occurs in the presence of sorbic acid. The pH gradient over the vacuole completely collapsed as a result of the decline in pHcyt. NMR spectra also revealed that sorbic acid (3.0 mM at pH 4.0) caused intracellular ATP pools and levels of sugar-phosphomonoesters and -phosphodiesters of A. niger mycelia to decrease dramatically, and they did not recover. The disruption of pH homeostasis by sorbic acid at concentrations below the MIC could account for the delay in spore germination and retardation of the onset of subsequent mycelial growth.  相似文献

Oxygen uptake by the spores ofFusarium moniliforme, F. oxysporum, F. semitectum, F. solani, Mucor racemosus andTrichoderma viride was increased in the presence of volatile substances extracted, fromOriganum majorana andOcimum basilicum. This increase was greater in the presence of volatile substances fromO. basilicum thanO. majorana, except in the case ofF. semitectum where the reverse was true. A drop in the RQ of all the germinating spores was observed in the presence of these substances. Volatile substances fromO. majorana reduced the spore germination ofM. racemosus whereas the spores ofT. viride were stimulated to germinate. Volatile substances fromO. basilicum stimulated the spore germination ofM. racemosus whereasT. viride spores were not affected.  相似文献

The morphology of spore germination in Didymium nigripes was studied using scanning electron microscopy and Nomarski phase optics. First, the outer spore wall splits, revealing a fibrillar inner wall. Remnants of the inner wall continue to cover the newly emerged amoeba. A single nucleus and a prominent vacuole are visible throughout germination. Germination is more rapid in glucose-peptone-yeast extract than in phosphate buffer. Germination is completely inhibited at 4°C, and is very slow at 18°C. Germination is most rapid at 26°C; at 21°C or 32°C it is slightly slower. Germination is reversibly inhibited by 20 μ/ml cycloheximide, but not by 200 μ/ml 5-fluoro uracil or 200 μ/ml proflavin. It is completely inhibited by 10-3 M Na azide.  相似文献
