2,4-Dichlorophenoxyacetic acid, a commonly used herbicide, increased the growth of the filamentous blue-green alga,Nostoc linckia at doses upto 100 μg /ml. The herbicidetreated N2-cultures showed enhanced heterocyst frequency and N2-growth. Thus, the herbicide stimulated algal growth at the expense of molecular nitrogen under aerobic growth conditions. Rifampicin caused chain formation of heterocysts. This was effectively counteracted by 2,4-dichlorophenoxyacetic acid, suggesting a biological interaction between them at the level of the heterocyst spacing control mechanism.  相似文献

The effect of ammonia and sulfide on rifampicin-induced heterocyst differentiation was studied in the nitrogen-fixing cyanobacteriumNostoc linckia. Aerobic growth with nitrogen gas of the cyanobacterium was greatly affected by rifampicin with formation of multiple heterocysts in chains in the filaments whereas ammonia in the medium reversed the rifampicin inhibition of growth and prevented the induction of heterocysts. In a sulfide medium the suppression exerted by rifampicin on aerobic growth with nitrogen gas and heterocyst induction was found to be considerably reduced. The results suggest two interesting points,viz. that (i) rifampicin interferes with the nitrogen-fixing function of heterocysts, and (ii) it checks the synthesis of an unknown heterocyst, inhibitor and thus permits the adjacent vegetative cells to differentiate into heterocysts in chains.  相似文献

The hetL gene from the cyanobacterium Nostoc sp. PCC 7120 encodes a 237 amino acid protein (25.6kDa) containing 40 predicted tandem pentapeptide repeats. Nostoc sp. PCC 7120 is a filamentous cyanobacterium that forms heterocysts, specialized cells capable of fixing atmospheric N(2) during nitrogen starvation in its aqueous environment. Under these conditions, heterocysts occur in a regular pattern of approximately one out of every 10-15 vegetative cells. Heterocyst differentiation is highly regulated involving hundreds of genes, one of which encodes PatS, thought to be an intercellular peptide signal made by developing heterocysts to inhibit heterocyst differentiation in neighboring vegetative cells, thus contributing to pattern formation and spacing of heterocysts along the filament. While overexpression of PatS suppresses heterocyst differentiation in Nostoc sp. PCC 7120, overexpression of HetL produces a multiple contiguous heterocyst phenotype with loss of the wild type heterocyst pattern, and strains containing extra copies of hetL allow heterocyst formation even in cells overexpressing PatS. Thus, HetL appears to interfere with heterocyst differentiation inhibition by PatS, however, the mechanism for HetL function remains unknown. As a first step towards exploring the mechanism for its biochemical function, the crystal structure of HetL has been solved at 2.0A resolution using sulfur anomalous scattering.  相似文献

Non-heterocystous, non-nitrogenfixing (het - nif-), heterocystous, non-nitrogenfixing (het + nif-) and multiple heterocystous, nitrogen-fixing (M-het + nif+) mutants of heterocystous, nitrogen-fixing (het + nif+) wild-type Nostoc muscorum and Nostoc linckia were isolated and characterized with respect to (a) nitrogenfixing activity, (b) reversion frequency, (c) ammonium repressibility of heterocyst formation, (d) heterocyst spacing pattern, and (e) action of L-methionine-DL-sulphoximine (MSO), an inhibitor of glutamine synthetase (GS), on heterocyst regulation. The mutant and revertant results suggest: (i) either involvement of a common genetic determinant in the formation of heterocyst and nitrogenase or the organization of het genes and nif genes in a single operon prone to complete inactivation by a single polar mutation, (ii) non-participation of active nitrogenase in regulation of heterocyst spacing; (iii) involvement of genetic factor(s) in the control of heterocyst spacing pattern in N. linckia, and (iv) apparently different nature of the mechanism of heterocyst inhibition by proheterocyst from that of heterocyst inhibition by NO 3 - or NH 4 + . L-Methionine-DL-sulphoximine inhibits growth and causes heterocyst formation in chains in N. linckia growing in nitrogen-free, NO 3 - , NO 2 - or NH 4 + medium, thus indicating a close physiological linkage between heterocyst and inorganic nitrogen metabolism regulation.  相似文献

2,4-Dichlorophenoxyacetic acid (2,4-D) stimulated growth and heterocyst differen- tiation ofNostoc linckia in nitrogen-free medium at lower concentrations (100 μ.g/mL) while its higher concentrations inhibited both processes and 1500 μg/mL proved to be lethal. Dry mass and specific growth rate of the alga declined with increasing concentration of 2,4-D in the range of 100–1500 μg/mL. Glucose slightly increased the heterocyst frequency without any lag in their differentiation. Tryptophan promoted growth of the alga, and formation of heterocysts (nearly three-fold). Tryptophan (50 μg/mL) complex medium with 1 mg 2,4-D per mL did not produce mature heterocysts. The filaments were fragmented at the point of hererocyst development and detached heterocysts germinatedin situ. Glucose and tryptophan protected the alga, its growth and heterocyst differentiation even at the lethal concentration of the herbicide. Effect in vitro of benthiocarb, an organocarbamate herbicide on neonate rat (3 day old) brain was studied to understand the interaction of benthiocarb with Na+K+-ATPase. Na+K+-ATPase of the developing rat brain was selected as an index enzyme since alterations in the Na+K+-ATPase activity leads to neuronal dysfunction. The assay of Na+K+-ATPase in the presence of 1-8 mu moles of benthiocarb showed decreased activity and a concentration dependent inhibition of Na+K+-ATPase was noticed upto 7 mu moles of benthiocarb. Based on IC50 values (median concentration), 50% inhibition of the enzyme was observed with 5 mu moles of benthiocarb. Norepinephrine (NE) was selected to study the modulation of benthiocarb inhibited enzyme. Maximum increase (76.7%) of Na+K+-ATPase was noticed with 35 mu moles of NE and effective concentration (EC50) of NE which produced 50% activation of the enzyme was found to be 20 mu moles. This study suggests that NE acts as a protective agent in reversing the benthiocarb in vitro inhibited neonate rat brain Na+K+-ATPase.  相似文献

A characteristic of N2-fixing cyanobacteria in symbiotic associations appears to be release of N2-derived NH4+. The specific activity of the primary ammonium-assimilating enzyme, glutamine synthetase (GS), was found to be three- to fourfold lower in Nostoc sp. strain 7801 grown in symbiotic association with the bryophyte Anthoceros punctatus than in free-living Nostoc sp. strain 7801. Quantitative immunological assays with antisera against GS purified from Nostoc sp. strain 7801 and from Escherichia coli indicated that similar amounts of the GS protein were present in symbiotic (50 micrograms mg-1) and free-living (68 micrograms mg-1) cultures. The conclusion from these experiments is that GS is regulated by a posttranslational mechanism in Anthoceros-associated Nostoc sp. strain 7801. However, the results of comparative catalytic and immunological experiments between N2- and NH4+-grown free-living Nostoc sp. strain 7801 implied control of GS synthesis. A correlation was not observed between the level of GS expression and the extent of symbiotic heterocyst differentiation in Nostoc sp. strain 7801 associated with A. punctatus.  相似文献

The filamentous cyanobacterium Anabaena (Nostoc) sp. strain PCC 7120 responds to starvation for fixed nitrogen by producing a semiregular pattern of nitrogen-fixing cells called heterocysts. Overexpression of the hetY gene partially suppressed heterocyst formation, resulting in an abnormal heterocyst pattern. Inactivation of hetY increased the time required for heterocyst maturation and caused defects in heterocyst morphology. The 489-bp hetY gene (alr2300), which is adjacent to patS (asl2301), encodes a protein that belongs to a conserved family of bacterial hypothetical proteins that contain an ATP-binding motif.  相似文献

Amino acid uptake and utilization of various nitrogen sources (amino acids, nitrite, nitrate and ammonia) were studied in Nostoc ANTH and i ts mu tant (Het(-)Nif(-)) isolate defective in heterocyst formation and N2-fixation. Both parent and its mutant grew at the expense of glutamine, asparagine and arginine as a source of fixed-nitrogen. Growth was better in glutamine-and asparagine-media as compared to that in arginine media. Glutamine and asparagine repressed heterocyst formation, N2-fixation and nitrate reduction in Nostoc ANTH, but arginine did so only partially. The poor growth in arginine-medium was not due to poor uptake rates, since the uptake rates were not significantly different from those for glutamine or asparagine. The glutamine synthetase activity remained unaffected during cultivation in media containing any one of the three amino acids tested. The uptake of amino acids was substrate-inducible, energy-dependent and required de novo protein synthesis. Nitrate and ammonium repressed ammonium uptake, but did not repress uptake of amino acids. In N2-medium (BG-11(0)), the uptake of ammonium and amino acids in the mutant was significantly higher than its parent strain. This was apparently due to nitrogen limitation since the mutant was unable to fix N2 and the growth medium lacked combined-N.  相似文献

The effect of different pesticides (fungicide, herbicide and insecticide) on the mycelial growth and formation and germination of sclerotia of sheath blight pathogen of rice, Rhizoctonia solani Kühn was studied in nutrient medium and in natural soil. Among the herbicides and insecticides, fenitrothion (insecticide) and thiobencarb (herbicide) were the most effective in inhibiting the mycelial growth and sclerotial formation. These pesticides effected significant inhibition of both mycelialgrowth and sclerotial formation at 7.5 to 10μg/ml concentration while fungicides carbendazim and tolclofosmethyl effected almost complete inhibition at 2.5 μg/ml. Interestingly, a synergistic increase in the toxicity to mycelial growth, sclerotial formation and sclerotial germination was noticed when the fungicide, mancozeb and the herbicide, thiobencarb were applied at subtoxic concentration in combination. Such synergistic interactions between pesticides in a combination leading to increased toxicity suggest that under the current practice of applying fungicides, herbicides and insecticides either simultaneously or in rotation, certain combinations may help in reducing the dose of a fungicide in disease control.  相似文献

The blue-green alga Nostoc linckia was treated with nitrosoguanidine and two classes of morphological mutant clones were isolated. One class shows certain abnormal phenotypic features of vegetative cells, spores, and heterocysts. It has increased heterocyst frequency and impaired growth rate. The other class exhibits an altered heterocyst spacing pattern. Both classes of mutants have reduced nitrogenase activity.  相似文献

We show that high levels of both carbon sources (sugars) and cations (notably magnesium) in the growth medium enhance the frequency of heterocyst formation by three-fold over the control in Nostoc sp. Such an alteration in the frequency and in enzyme activity (hydrogenase and nitrogenase), induced a proportional increase in hydrogen evolution capacity under a variety of conditions. Nitrogenase and hydrogenase activity also showed an increase in the enriched cultures. Our results therefore suggest that enriching the Nostoc culture with some selected carbon source or a low cost inorganic source like magnesium would result in a high yield of heterocysts in heterocystous cyanobacteria which would be helpful in the production of sustainable bio-fuels.  相似文献

The effects of two amino acid analogues, viz., L-methionine-DL-sulphoximine and L-methyl-DL-methionine on growth, heterocyst differentiation and nitrogen fixation in the blue-green algaNostoc linckia have been studied with special reference to heterocyst spacing pattern. L-methionine-DL-sulphoximine strongly inhibited growth but produced an unusual number of heterocysts with changed heterocyst spacing pattern in both nitrogen-free and ammonium-containing media. L-methyl-DL-methionine was less effective than L-methionine-DL-sulphoximine. An attempt was also made to counteract the toxic effects of these analogues by supplying amino acids. Glutamine and methionine reversed the inhibitory effect of L-methionine-DL-sulphoximine while only methionine reversed the inhibitory effect of L-methyl-DL-methionine. Production of changed heterocyst spacing pattern in nitrogen-free and ammonium-containing media when supplemented with L-methionine-DL-sulphioximine suggests that ammonia may not be the inhibitor of heterocyst spacing pattern.  相似文献

Summary The effect of graded concentrations of four common ricefield herbicides (Arozin, Butachlor, Alachlor, 2,4-D) on diazotrophic growth, macromolecular contents, heterocyst frequency and tolerance potentials of Ca-alginate immobilized diazotrophic cyanobacterial isolates Nostoc punctiforme, N. calcicola, Anabaena variabilis, Gloeocapsasp., Aphanocapsa sp. and laboratory strain N. muscorum ISU (Anabaena ATCC 27893) was studied and compared with free-living cultures. Cyanobacterial isolates showed progressive inhibition of growth with increasing dosage of herbicides in both free and immobilized states. There were significant differences in the relative toxicity of the four herbicides. Arozin proved to be more growth toxic in comparison to Alachlor, Butachlor and 2,4-D. Growth performance of the immobilized cyanobacterial isolates under herbicide stress showed a similar diazotrophic growth pattern to free cells with no difference in lethal and sub-lethal dosages. However, at lethal concentrations of herbicides, the immobilized cells exhibited prolonged survivability of 14–16 days as compared to their free-living counterparts (8–12 days). The decline in growth, macromolecular contents and heterocyst frequency was found to be similar in both the states in graded dosages of herbicides. Of the test organisms, A. variabilis showed maximum natural tolerance towards all the four herbicides tested. Evidently immobilization by Ca-alginate seems to provide protection to the diazotrophic cyanobacterial inoculants to a certain extent against the growth-toxic action of herbicides.  相似文献

The N2, NO 3 , NO 2 , NH 4 + and glutamine growing cultures of parentNostoc muscorum are found more or less equally sensitive to azide inhibition of growth. A mutant strain resistant to sodium azide was isolated from the parent strain in NO 3 medium and the two strains were compared with regard to their heterocyst formation and nitrogenase activity in NO 3 , NO 2 , NH 4 + and glutamine media. While the parent strain stops production of both heterocyst and nitrogenase in all the fixed nitrogen media, the azide resistant strain forms both in the fixed inorganic nitrogen media but only heterocyst and no nitrogenase in the glutamine medium. Clearly a single genetic determinant of regulatory nature appears to mediate azide-resistance as well as relief of heterocyst and nitrogenase formation from inhibition by the fixed inorganic nitrogen source. The results of glutamine effect on the heterocyst and nitrogenase formation of the two strains indicate the operation of two levels of glutamine-sensitive regulation, one which operates through the common genetic determinant of heterocyst and nitrogenase regulation and the other exclusive to nitrogenase regulation. The in vivo functional nitrogenase does not appear to be the reason for azide-resistance and neither ammonia nor glutamine or its close metabolic product seems to function in the control of heterocyst spacing.  相似文献

Effect of two photosynthetic inhibitor herbicides, atrazine (both purified and formulated) and [3-(3,4-dichlorophenyl)-1,1-dimethyl urea] (DCMU), on the growth, macromolecular contents, heterocyst frequency, photosynthetic O2 evolution and dark O2 uptake of wild type and multiple herbicide resistant (MHR) strain of diazotrophic cyanobacterium A. variabilis was studied. Cyanobacterial strains showed gradual inhibition in growth with increasing dosage of herbicides. Both wild type and MHR strain tolerated < 6.0 mg L(-1) of atrazine (purified), < 2.0 mg L(-1) of atrazine (formulated) and < 0.4 mg L(-1) of DCMU indicating similar level of herbicide tolerance. Atrazine (pure) (8.0 mg L(-1)) and 4.0 mg L(-1) of atrazine (formulated) were growth inhibitory concentrations (lethal) for both wild type and MHR strain indicating formulated atrazine was more toxic than the purified form. Comparatively lower concentrations of DCMU were found to be lethal for wild type and MHR strain, respectively. Thus, between the two herbicides tested DCMU was more growth toxic than atrazine. At sublethal dosages of herbicides, photosynthetic O2 evolution showed highest inhibition followed by chlorophyll a, phycobhiliproteins and heterocyst differentiation as compared to carotenoid, protein and respiratory O2 uptake.  相似文献

Summary This study presents the effects of Cr, Pb, Ni and Ag on growth, pigments, protein, DNA, RNA, heterocyst frequency, uptake of NH4 + and N03 , loss of electrolytes (Na+ and K+), nitrate reductase and glutamine synthetase activities ofNostoc muscorum. The statistical tests revealed a direct positive correlation between the metal concentration and inhibition of different processes. Ni was found to be more toxic against growth, pigments and heterocyst differentiation compared to the other metals. Inhibition of pigment showed the following trend: chlorophyll > phycocyanin > carotenoid. No generalized trend for inhibition of macromolecules was observed. The loss of K+ and Na+ as affected by Cr, Ni and Pb was similar but more pronounced for K+ than Na+. The inhibition of physiological variables depicted the following trend: Na+ loss > K+ loss > glutamine synthetase > NH4 uptake > growth > N03 uptake > nitrate reductase > heterocyst frequency. This study therefore suggests that loss of electrolytes can be used as a first signal of metal toxicity in cyanobacteria. However, further study is needed to confirm whether the abnormality induced by nickel (branch formation) is a physiological or genetic phenomenon.  相似文献

Sensitivity of L-forms of group A streptococci to 5 antibiotics such as erythromycin, lincomycin, tetracycline, gentamicin and chloramphenicol was studied in an artificial nutrient medium and cell cultures i.e. human fibroblast diploid cells and transplantable human heart cells (Girardi). In vitro investigation of the antibiotic effect on the streptococcal L-forms revealed their sensitivity to erythromycin (MIC, 0.4 micrograms/ml), lincomycin (MIC, 0.08 microgram/ml) and tetracycline (MIC, 2 micrograms/ml). The streptococcal L-forms were slightly sensitive to gentamicin (MIC, 6 micrograms/ml) and chloramphenicol (MIC, 30 micrograms/ml). Complete inhibition of the growth of the L-forms in the Girardi cells on the 1st day of the experiment after the antibiotics administration in single doses was induced by lincomycin, 5 micrograms/ml, erythromycin, 10 micrograms/ml, and tetracycline, 100 micrograms/ml. In the diploid cells, the respective figures were 50, 100 and 200 micrograms/ml. Chloramphenicol and gentamicin had an inhibitory effect on the growth of the L-forms but produced no sanative effect.  相似文献

The potential of Nostoc calcicola and its bicarbonate resistant mutant as bioameleorating agent was investigated, using laboratory simulation experiments, in terms of their growth potential, glutamine synthetase (GS) activity, heterocyst frequency and effect on pH of soil. Nostoc calcicola, exhibited a tendency to lower the pH of ‘usar’ soil significantly and showed better growth and pigment content at 20% soil extract as compared to basal medium. The bicarbonate resistant mutant (HCO3 −R) exhibited a better ability to grow at higher percentage of soil extract (60%), besides bringing about a more significant change in soil pH as compared to wild type. The heterocyst frequency was much higher in the mutant strain, which was not significantly affected by growth in various concentrations of soil extract. The mutant strain holds promise as a potential bioameliorant for ‘usar’ soil after further evaluation of its reclamative properties at field level.  相似文献

Lead decreased the growth rates, total cell mass, heterocyst frequency, total cell protein, nitrogenase activity, glutamine synthetase (GS) and glutamate synthase (GOGAT) activities in N:muscorum. However, lead at 0.01 and 10 micrograms ml-1 conc. enhanced nitrogenase as well as GS activity of the cells. On transfer to excess lead (100 micrograms ml-1), nitrogenase and GS activities ceased almost after 24 hr in the cyanobacterium. It is deduced that lead has a two step effect on stimulation and inhibition of metabolic activity at 0.01 and 10 micrograms ml-1 concentration and 0.1 and 100 micrograms ml-1 concentration respectively indicating a close interaction between nitrogen fixation and GS activity. However, GOGAT activity is an exception to this two step stimulation and inhibition process.  相似文献
