Wing polyphenism, which is the ability of a single genome to produce winged and wingless castes in a colony in response to environmental cues, evolved just once and is a universal feature of ants. The gene network underlying wing polyphenism, however, is conserved in the winged castes of different ant species, but is interrupted at different points in the network in the wingless castes of these species. We previously constructed a mathematical model, which predicts that a key gene brinker (brk) mediates the development and evolution of these different "interruption points" in wingless castes of different ant species. According to this model, brk is upregulated throughout the vestigial wing discs of wingless ant castes to reduce growth and induce apoptosis. Here, we tested these predictions by examining the expression of brk, as well as three other genes up- and downstream of brk-decapentaplegic (dpp), spalt (sal), and engrailed (en)-in the winged reproductive and wingless soldier castes in the ant Pheidole morrisi. We show that expression of these genes is conserved in the wing disc of winged castes. Surprisingly, however, we found that brk expression is absent throughout development of the vestigial soldier forewing disc. This absence is correlated with abnormal growth of the soldier forewing disc as revealed by En expression and morphometric analyses. We also discovered that dpp and sal expression change dynamically during the transition from larval-to-prepupal development, and is spatiotemporally correlated with the induction of apoptosis in soldier forewing disc. Our results suggest that, contrary to our predictions, brk may not be a key gene in the network for suppressing wings in soldiers, and its absence may function to disrupt the normal growth of the soldier forewing disc. Furthermore, the dynamic changes in network interruptions we discovered may be important for the induction of apoptosis, and may be a general feature of gene networks that underlie polyphenism.  相似文献

Abstract Many ants have independently evolved castes with novel morphology as well as function, such as soldiers and permanently wingless (ergatoid) queens. We present a conceptual model, based on modularity in morphology and development, in which evolutionary innovation is facilitated by the ancestral ant polyphenism of winged queens and wingless workers. We suggest that novel castes evolved from rare intercastes, anomalous mosaics of winged queens and workers, erratically produced by colonies through environmental or genetic perturbations. The colonial environment is highly accommodating and buffers viable intercastes from individual selection. Their cost is limited because they are diluted by the large number of nestmates, yet some can bring disproportionate benefits to their colonies in the context of defense or reproduction (e.g., wingless intercastes able to mate). Useful intercastes will increase in frequency as their morphology is stabilized through genetic accommodation. We show that both soldiers and ergatoid queens are mosaics of winged queens and workers, and they are strikingly similar to some intercastes. Modularity and developmental plasticity together with winged/wingless polyphenism thus allow for the production of highly variable mosaic intercastes, and colonies incubate the advantageous mosaics.  相似文献

Aphids display divergent adult phenotypes, depending on environmental conditions experienced during their embyonic and nymphal stages in their complex life cycles. The plastic developmental mode is an extreme case of phenotypic plasticity, so-called “polyphenism”, in which discrete multiple phenotypes are produced based on a single genome. For example, winged and wingless adult females are derived from a single genotype. However, the developmental mechanisms producing these polyphenic traits according to the extrinsic stimuli, such as density conditions, still remain unknown. In this study, to analyze the developmental processes underlying the wing polyphenism, we extensively observed and compared wing development in the winged and wingless individuals in parthenogenetic generations of the aphid Acyrthosiphon pisum (Harris), using scanning electron microscopy and histological sectioning. At the first-instar stage, the wing primordia were observed both in the future winged (W) and wingless (WL) nymphs. Developmental differences can be seen from the second-instar stage, when wing primordia degenerate in the WL nymphs, while they develop and become more thickened in the W nymphs, suggesting that the developmental programs should be launched prior to this stage. Furthermore, during the third- to fifth-instar stages, wing buds and flight muscles were well developed in the W nymphs, while wing primordia completely disappeared in the WL ones. In addition, the observation on the detailed developmental process of wing primordia during the third-instar W nymphs showed that the wing buds become swollen especially at the basal part, even during the intermolt period. This was caused by the development of wing epithelia under the cuticle of this instar nymph. Actually on the surface of the cuticle of wing-bud bases, there were numerous furrows, which gradually expand during the intermolt period. The similar situation was also observed at the forth-instar nymphs, in which the wings are formed in the complicated manner inside the wing pads. Furthermore, the developmental process of flight muscles was also described in detail. These dynamic developmental differences between the wing morphs should be regulated under the gene expression cascades that switch according to environmental stimuli.  相似文献

SUMMARY Many taxa of winged insects have independently lost the ability to fly and often possess reduced wings. Species exhibiting natural variation in wing morphology provide opportunities to investigate the genetics and developmental processes underlying the evolution of alternative wing morphs. Although many wing dimorphic species of beetles are known, the underlying mechanisms of variation are not well understood in this insect order. Here, we examine wing development of wild type and natural wingless morphs of the two-spot ladybird beetle, Adalia bipunctata . We show that both pairs of wings are distally truncated in the wingless adults. A laboratory population of the wingless morph displays heritable variation in the degree of wing truncation, reflecting reduced growth of the larval wing discs. The coexistence of variable wingless morphs supports the idea that typical monomorphic wingless insects may be the result of a gradual evolution of wing loss. Gene expression patterns in wing discs suggest that the conserved gene network controlling wing development in wild-type Adalia is disrupted in the dorsoventral patterning pathway in the wingless morphs. Previous research on several species of ant has revealed that the anteroposterior wing patterning pathway is disrupted in wingless workers. Future investigations should confirm whether interruptions in both taxa are limited to the patterning pathways found thus far, or whether there are also shared interruption points. Nevertheless, our results highlight that diverse mechanisms of development are likely to underlie the evolution of wingless insects.  相似文献

Phenotypes of organisms are not determined completely genetically, but vary according to environmental factors (phenotypic plasticity). Some organisms express several discrete adaptive phenotypes (polyphenism). Social insects possess a few types of individuals (castes) in their colonies, to which specific tasks are allocated. Here, I review studies on caste polyphenism in ants and termites, in terms of the developmental mechanisms of caste-specific characters, such as alate wings and soldier mandibles. In ants, the developmental fate of caste is probably determined by the pattern-formation genes in the early stage of postembryonic development, but apoptotic degeneration occurs in the wing primordia of future workers. As apoptotic wing degeneration has been observed in two phylogenetically distant groups of ants, this phenomenon is suggested to be conserved in many ant species. On the other hand, all termite species possess distinct sterile soldiers with specific morphologies suitable for defense. Recent studies using molecular techniques isolated genes related to soldier differentiation and analyzed the expression profiles of those genes in order to understand the mechanism of caste differentiation and the link between molecular and social evolution. In this review, I focus on these studies, in terms of the alteration of body plan in response to environmental signals, and discuss the evolutionary process of the interaction between ontogeny and environment.  相似文献

Many polyphenisms are examples of adaptive phenotypic plasticity where a single genotype produces distinct phenotypes in response to environmental cues. Such alternative phenotypes occur as winged and wingless parthenogenetic females in the pea aphid (Acyrthosiphon pisum). However, the proportion of winged females produced in response to a given environmental cue varies between clonal genotypes. Winged and wingless phenotypes also occur in males of the sexual generation. In contrast to parthenogenetic females, wing production in males is environmentally insensitive and controlled by the sex-linked, biallelic locus, aphicarus (api). Hence, environmental or genetic cues induce development of winged and wingless phenotypes at different stages of the pea aphid life cycle. We have tested whether allelic variation at the api locus explains genetic variation in the propensity to produce winged females. We assayed clones from an F2 cross that were heterozygous or homozygous for alternative api alleles for their propensity to produce winged offspring. We found that clones with different api genotypes differed in their propensity to produce winged offspring. The results indicate genetic linkage of factors controlling the female wing polyphenism and male wing polymorphism. This finding is consistent with the hypothesis that genotype by environment interaction at the api locus explains genetic variation in the environmentally cued wing polyphenism.  相似文献

Wing polymorphisms observed in many Insecta are important topics in developmental biology and ecology; these polymorphisms are a consequence of trade-offs between flight and other abilities. The pea aphid, Acyrthosiphon pisum, possesses 2 types of wing polymorphisms: One is a genetic wing polymorphism occurring in males, and the other is an environmental wing polyphenism seen in viviparous females. Although genetic and environmental cues for the 2 wing polymorphisms have been studied, differences in their developmental regulation have not been elucidated. In particular, there is little knowledge regarding the developmental processes in male wing polymorphism. Therefore, in this study, the development of flight apparatuses and external morphologies was compared among 3 male wing morphs (winged, wingless, and intermediate). These male developmental processes were subsequently compared with those of female wing morphs. Developmental differences between the male and female polymorphisms were identified in flight muscle development and degeneration but not in wing bud development. Furthermore, the nymphal periods of wingless and intermediate males were significantly shorter than that of winged males, indicating the adaptive significance of male winglessness. Overall, this study indicates that the male and female wing polymorphisms are based on different regulatory systems for flight apparatus development, which are probably the result of different adaptations under different selection pressures.  相似文献

The ant genus Cardiocondyla is characterized by an extraordinary male polyphenism, with winged disperser males and wingless, territorial ergatoid males. Winged males are produced only after the colony has experienced stressful environmental conditions, e.g., a drastic temperature decrease. We investigated the proximate basis of male polyphenism and caste dimorphism in C. obscurior. The critical stage for both morph and caste determination is the end of the second of three instars. Larval development as well as duration of the pupal stage are extended both in winged males and winged females and winged reproductives need on average 8.8 days longer for the development from egg to adult than wingless ergatoid males and workers. Treatment of first and second instar larvae with methoprene, a juvenile hormone analogue, led to the expression of the winged morph, suggesting an important role of juvenile hormone in both sexes. Although queens are produced year-round in contrast to winged males, the proximate basis of variation in morphology is likely to be the same in both sexes. Whereas the larvae themselves appear to be insensitive to the environmental changes, behavioral observations revealed that workers react to stress by changing their behavior towards larvae and in this way trigger them to develop into winged males.  相似文献

Antibiotics, primary symbionts and wing polyphenism in three aphid species   总被引:1,自引:0,他引:1  
The possible role of the primary Buchnera symbionts in wing polyphenism is examined in three aphid species. Presumptive winged aphids were fed on antibiotic-treated beans to destroy these symbionts. As previously reported, this leads to inhibited growth and low/zero fecundity. When such treatment is applied to the short-day-induced gynoparae (the winged autumn migrant) of the black bean aphid, Aphis fabae, it also causes many insects to develop as wingless or winged/wingless intermediate adult forms (apterisation). However, whilst antibiotic treatment of crowd-induced, long-day winged forms of the pea aphid, Acyrthosiphon pisum (a green and a pink clone) and the vetch aphid, Megoura viciae has similar effects on size and fecundity, it does not affect wing development. Food deprivation also promotes apterisation in A. fabae gynoparae but not in the crowd-induced winged morphs of the other two species. Thus, it appears that apterisation in A. fabae is not a direct effect of antibiotic treatment or a novel role for symbionts but is most likely related to impaired nutrition induced by the loss of the symbiont population.  相似文献

Discrete variation in wing morphology is a very common phenomenon in insects and has been used extensively in the past 50 years as a model to study the ecology and evolution of dispersal. Wing morph determination can be purely genetic, purely environmental, or some combination of the two. The precise genetic determinants of genetically based wing morph variation are unknown. Here we explore the genetic basis of wing polymorphism in the pea aphid, which can produce either winged or wingless males. We confirm that three types of pea aphid clones coexist in natural populations, those producing winged males only, those producing wingless males only, and those producing a mixture of both. A Mendelian genetic analysis reveals that male wing polymorphism in pea aphids is determined by a single locus, two alleles system. Using microsatellite loci of known location, we show that this locus is on the X chromosome. The existence of a simple genetic determinism for wing polymorphism in a system in which genetic investigation is possible may help investigations on the physiological and molecular mechanisms of genetically-based wing morph variation. This locus could also be used in the search for genes involved in the wing polyphenism described in parthenogenetic females and to investigate the interplay between polymorphisms and polyphenisms.  相似文献

Over the last decade, it has become clear that organismal form is largely determined by developmental and evolutionary changes in the growth and pattern formation of tissues. Yet, there is little known about how these two integrated processes respond to environmental cues or how they evolve relative to one another. Here, we present the discovery of vestigial wing imaginal discs in worker larvae of the red imported fire ant, Solenopsis invicta. These vestigial wing discs are present in all worker larvae, which is uncommon for a species with a large worker size distribution. Furthermore, the growth trajectory of these vestigial discs is distinct from all of the ant species examined to date because they grow at a rate slower than the leg discs. We predicted that the growth trajectory of the vestigial wing discs would be mirrored by evolutionary changes in their patterning. We tested this prediction by examining the expression of three patterning genes, extradenticle, ultrabithorax, and engrailed, known to underlie the wing polyphenism in ants. Surprisingly, the expression patterns of these three genes in the vestigial wing discs was the same as those found in ant species with different worker size distributions and wing disc growth than fire ants. We conclude that growth and patterning are evolutionarily dissociated in the vestigial wing discs of S. invicta because patterning in these discs is conserved, whereas their growth trajectories are not. The evolutionary dissociation of growth and patterning may be an important feature of gene networks that underlie polyphenic traits.  相似文献

SUMMARY In wing polyphenisms that produced alternative wing morphs depending on environmental conditions, the developmental regulations to balance between flight and reproductive abilities should be important. Many species of aphids exhibit wing polyphenisms, and the development of wing and flight muscles is thought to incur costs of reproductive ability. To evaluate the relationship between flight and reproduction, the fecundity and the wing- and ovarian development in the parthenogenetic generations were compared between winged and wingless aphids in the vetch aphid Megoura crassicauda . Although no differences in offspring number and size were detected, the onset of larviposition after imaginal molt was delayed in winged adults. The comparison of growth in flight apparatus revealed that, after the second-instar nymphs, the flight-apparatus primordia of presumptive wingless aphids were degenerated while those of winged nymphs rapidly developed. In the ovaries of winged line, the embryo size was smaller and the embryonic stages were delayed from third to fifth instars, although these differences had disappeared by the time of larviposition. It is therefore likely that the delay in larviposition in winged aphids is due to the slower embryonic development. The correlation between embryo size and developmental stage suggests that the embryos of winged aphids are better developed than similarly sized embryos in wingless aphids. These heterochronic shifts would facilitate the rapid onset of larviposition after the dispersal flight. This developmental regulation of embryogenesis in the aphid wing polyphenism is suggested to be an adaptation that compensates the delay of reproduction caused by the wing development.  相似文献

【目的】前期研究发现麦长管蚜Sitobion avenae孤雌蚜有翅和无翅个体中存在很多差异表达的微小RNA(microRNA, miRNA),本研究旨在进一步明确这些miRNA在豌豆蚜Acyrthosiphon pisum中发挥作用的发育阶段,探索miRNA调控孤雌蚜翅两型性分化的机制。【方法】选择在麦长管蚜有翅蚜和无翅蚜中显著差异表达,且靶基因为蜕皮激素、胰岛素信号通路及翅型发育关键基因的5个miRNA(Let-7,miR-92a, miR-92b, miR-92a-1-p5和miR-277),利用qPCR检测这些miRNA及其靶标基因在豌豆蚜3-4龄若蚜和成虫有翅和无翅个体中的表达谱;同时利用双荧光素酶活性检测法对上述miRNA的靶基因进行验证。【结果】表达谱分析发现,这5个miRNA在豌豆蚜成虫中表达量均高于其在若蚜中的表达量,而其预测的靶基因在4龄若蚜中的表达量均高于其在成虫中的表达量,表明miRNA对其靶基因的调控作用可能集中在成虫阶段。分析豌豆蚜有翅和无翅个体中5个miRNA的表达情况发现,在成虫有翅个体中5个miRNA的表达量均高于无翅个体中的,其中miR-277表达差异最显著,成虫有翅个体中的表达量是无翅个体中表达量的7.5倍;其次为Let-7,表达差异达3倍。而Let-7在3龄有翅若蚜和无翅若蚜中表达差异最显著,有翅个体中的表达量是无翅个体中的37.8倍;其次为miR-277,表达差异达7.6倍。比较5个miRNA与其靶基因在豌豆蚜3-4龄若蚜及成虫有翅和无翅个体中的表达发现,miRNA Let-7和miR-92b的表达趋势分别与其靶基因abrupt和Foxo的基本相反。荧光素酶活性检测结果显示,Let-7的真实靶标为abrupt,共转染Let-7模拟物后与对照相比,荧光素酶活性下降53%,达极显著水平。其他miRNA与靶标基因的互作不显著。【结论】首次发现miRNA对豌豆蚜孤雌蚜翅型分化相关基因的调控可能发生在成虫阶段。Let-7可能通过调控abrupt基因参与孤雌蚜翅型分化。该研究为进一步探索miRNA参与孤雌蚜翅两型性分化的机制奠定了基础。  相似文献

Arginine kinase (AK), a primary enzyme in cell metabolism and adenosine 5′-triphosphate (ATP)-consuming processes, plays an important role in cellular energy metabolism and maintaining constant ATP levels in invertebrate cells. In order to identify genes that are differentially expressed between larvae and adults, queens and workers, and female alates (winged) and queens (wingless), AK cDNA was obtained from the red imported fire ant. The cDNA sequence of the gene has open reading frames of 1065 nucleotides, encoding a protein of 355 amino acid residues that includes the substrate recognition region, the signature sequence pattern of ATP:guanidino kinases, and an “actinin-type” actin binding domain. Northern blot analysis and protein activity analysis demonstrated that the expression of the AK gene and its protein activity were developmentally, caste specifically, and tissue specifically regulated in red imported fire ants with a descending order of worker> alate (winged adult) female> alate (winged adult) male> larvae> worker pupae ≈ alate pupae. These results suggest a different demand for energy-consumption and production in the different castes of the red imported fire ant, which may be linked to their different missions and physiological activities in the colonies. The highest level of the AK gene expression and activity was identified in head tissue of both female alates and workers and thorax tissue of workers, followed by thorax tissue of female alates and abdomen tissue of male alates, suggesting the main tissues or cells in these body parts, such as brain, neurons and muscles, which have been identified as the major tissues and/or cells that display high and variable rates of energy turnover in other organisms, play a key role in energy production and its utilization in the fire ant. In contrast, in the male alate, the highest AK expression and activity were found in the abdomen, suggesting that here energy demand may relate to sperm formation and reproduction.  相似文献

Caste polyphenism in social insects provides us with excellent opportunities to examine the plasticity and robustness underlying developmental pathways. Several ant species have evolved unusual castes showing intermediate morphologies between alate queens and wingless workers. In some low-temperature habitats, the ant Myrmecina nipponica produces such intermediate reproductives (i.e. ergatoids), which can mate and store sperm but cannot fly. To gain insight into the developmental and evolutionary aspects associated with ergatoid production, we conducted morphological and histological examinations of the post-embryonic development of compound eyes, gonads and wings during the process of caste differentiation. In compound eyes, both the queen-worker and ergatoid-worker differences were already recognized at the third larval instar. In gonads, queen-worker differentiation began at the larval stage, and ergatoid-worker differentiation began between the prepupal and pupal stages. Wing development in ergatoids was generally similar to that in workers throughout post-embryonic development. Our results showed that the developmental rate and timing of differentiation in body parts differed among castes and among body parts. These differences suggest that the rearrangement of modular body parts by heterochronic developmental regulation is responsible for the origination of novel castes, which are considered to be adaptations to specific ecological niches.  相似文献

Laboratory experiments were conducted to test the hypothesis that widelyforaging generalist lizard,Eumeces okadae, visually discriminates palatable queen ants from unpalatable worker ants. The workers ofCamponotus japonicus andLasius niger were rejected on sight, while the queens of both species were eaten with little prior chemical examination by tongue flicks or licks. Comparison of the lizards' responses towards the workers and wingless queens of similar size indicated that neither body size nor presence or absence of wings accounted for difference in responses toward the 2 ant castes. The lizards probably discriminated different ant castes by body proportions.  相似文献

红火蚁Solenopsis invicta是火蚁属重要的入侵蚂蚁,与其近缘种黑火蚁S.richteri和杂交蚁S.invicta×S.richteri形态相似,难以区分。为了快速准确鉴定3种火蚁属近缘种,本研究利用气相色谱-质谱联用仪(GC-MS),解析3种火蚁的工蚁、有翅雌蚁、有翅雄蚁的表皮碳氢化合物种类和含量,并进行主成分分析、判别分析及聚类分析。结果表明:3种火蚁共检测到62种表皮碳氢化合物,主要包括一甲基烷烃、二甲基烷烃和正构烷烃等;红火蚁、黑火蚁及杂交蚁不同品级的表皮碳氢化合物种类及含量存在显著的种间差异,红火蚁不同地理种群的表皮碳氢化合物种类及含量相似度较高;建立3种火蚁相应品级的分类判别函数,可准确区分各品级下的3种火蚁。因此,表皮碳氢化合物组成分析可用于红火蚁及其近缘种的分类鉴定,为口岸火蚁属蚂蚁的快速检疫鉴定提供新技术。  相似文献
