Summary The uptake of C14O2 by the roots of intact tomato plants from solution containing Na2C14O3 was studied at different light intensities as well as in darkness.Where plants had previously been starved for CO2 for 12 hours, a higher rate of C14 uptake was observed than with plants which had been transferred directly from the soil to the radioactive solution.In general, the C14 content of the roots was slightly higher than that of the shoots. At light intensities under the compensation point and in darkness the C14 content of the shoots relative to the roots decreased. This was accompanied by release of C14O2 during respiration, indicating that the absorbed C14 was readily translocated upwards and released as C14O2 under these conditions. At light intensities above the compensation point no C14O2 was released.  相似文献

1. Three modified horse haemoglobins have been prepared: (i) alpha(c) (2)beta(c) (2), in which both the alpha-amino groups of the alpha- and beta-chains have reacted with cyanate, (ii) alpha(c) (2)beta(2), in which the alpha-amino groups of the alpha-chains have reacted with cyanate, and (iii) alpha(2)beta(c) (2), in which the two alpha-amino groups of the beta-chain have reacted with cyanate. 2. The values of n (the Hill constant) for alpha(c) (2)beta(c) (2), alpha(2)beta(c) (2) and alpha(c) (2)beta(2) were (respectively) 2.5, 2.0 and 2.6, indicating the presence of co-operative interactions between the haem groups for all derivatives. 3. In the alkaline pH range (about pH8.0) all the derivatives show the same charge as normal haemoglobin whereas in the acid pH range (about pH6.0) alpha(c) (2)beta(c) (2) differs by four protonic charges and alpha(c) (2)beta(2), alpha(2)beta(c) (2) by two protonic charges from normal haemoglobin, indicating that the expected number of ionizing groups have been removed. 4. alpha(c) (2)beta(2) and alpha(c) (2)beta(c) (2) show a 25% decrease in the alkaline Bohr effect, in contrast with alpha(2)beta(c) (2), which has the same Bohr effect as normal haemoglobin. 5. The deoxy form of alpha(c) (2)beta(c) (2) does not bind more CO(2) than the oxy form of alpha(c) (2)beta(c) (2), whereas alpha(c) (2)beta(2) and alpha(2)beta(c) (2) show intermediate binding. 6. The results reported confirm the hypothesis that, under physiological conditions, haemoglobin binds CO(2) through the four terminal alpha-amino groups and that the two terminal alpha-amino groups of alpha-chains are involved in the Bohr effect.  相似文献

The pCO2 distribution in thesurface waters of the Elbe estuary and German Bight was investigated during high river discharge in April 1997. pCO2 values of about 1100 atm foundin the upper part of the estuary are, comparedto other European rivers, only moderate. In theouter estuary, the enhanced river dischargecreates a pronounced river plume reflected in decreased surface salinity.However, pCO2 in this area(250 atm) is below atmospheric values dueto primary production. Comparison with resultswe obtained during other measuringcampaigns shows that in the outer estuarysupersaturation of CO2 with respect to theatmosphere occurs only in early spring duringthe time of highest river discharge. In latespring and summer, intensive primary productionwithin the surface layer leads to a pronouncedundersaturation (141 atm), despite atemperature increase of more than 20 °Cfrom winter to summer. Undersaturation isenforced by stable stratification of the watercolumn during the productive seasons thatdiminishes the entrainment of CO2from below the thermocline into the surfacewater. In winter, CO2 is close toequilibrium with the atmosphere and controlledby physical processes. Our data suggest thatthe Elbe estuary represents a sink foratmospheric CO2 over the investigatedseasons and that carbon cycling in this areais dominated by biological processes.  相似文献

Combined gas chromatography-mass spectrometry of malic acid derivatives has been used to show unequivocally that malic acid, synthesized during active acid accumulation in the dark by Kalanchoë daigremontiana Hammet et Perrier in the presence of 13CO2 is produced by a pathway involving a single carboxylation. The significance of the finding that crassulacean malate synthesized in the dark and in the presence of 14CO2 often contains 66% of the total carboxyl label in carbon atom 4, which has previously been taken to indicate the operation of a double carboxylation pathway or has been dismissed as an artefact, is discussed.  相似文献
