The phase transition properties of aqueous suspensions of a series of nonhydrated (not heated above room temperature) and hydrated 1,2 diacylphosphatidylethanolamines (PE's) have been examined by high sensitivity differential scanning calorimetry at scan rates of 0.02-1.0 K min-1. At all scan rates nonhydrated PE's show a single asymmetric transition curve of excess heat capacity as a function of temperature. Multilamellar dispersions of hydrated PE's, however, exhibit transitions with fine structure, which can be fitted as the sum of three two-state component transitions, at scan rates of 0.02-0.1 K min-1, but give only a single asymmetric transition at 1.0 K min-1. At all scan rates the transition(s) of hydrated samples occur at lower temperatures than those of nonhydrated samples. One of the component transitions of hydrated PE's may be analogous to the pretransition that occurs in 1,2 diacylphosphatidylcholines.  相似文献

We have previously shown that a fluorescent derivative of phosphatidic acid, 1-acyl-2-[N-(4-nitrobenzo-2-oxa-1,3-diazole) aminocaproyl]phosphatidic acid (C6-NBD-PA) is rapidly transferred from liposomes to Chinese hamster fibroblasts at 2 degrees C, resulting in intense labeling of the mitochondria, endoplasmic reticulum, and nuclear envelope, but not the plasma membrane. During this labeling, C6-NBD-PA is metabolized predominantly to fluorescent diacylglycerol (Pagano, R. E., Longmuir, K. J., Martin, O. C., and Struck, D. K. (1981) J. Cell Biol. 91, 872-877). In the present study we investigated the mechanism by which C6-NBD-PA enters cells and is translocated to intracellular membranes at low temperature. (i) When hydrolysis of C6-NBD-PA to diacylglycerol was prevented by using a nonhydrolyzable fluorescent phosphonate analog, intense labeling of the plasma membrane occurred but fluorescent lipid did not enter the cytoplasm of cells. (ii) Experiments using C6-NBD-PA and cells prelabeled with 32Pi demonstrated that some of the fluorescent diacylglycerol was rephosphorylated at 2 degrees C. (iii) When cells were treated with 1,3-[palmitoyl, N-(4-nitrobenzo-2-oxa-1,3-diazole)aminocaproyl]-glycerophosphate, the lipid was dephosphorylated to 1,3-diacylglycerol but its rephosphorylation could not be detected. Nevertheless, rapid labeling of cytoplasmic membranes occurred. (iv) Formation of fluorescent diacylglycerol at the plasma membrane by treatment of cells with fluorescent phosphatidylcholine followed by phospholipase C at 2 degrees C resulted in strong labeling of intracellular membranes. Based on these results, a working model is presented for the uptake and intracellular translocation of phosphatidic acid involving formation of diacylglycerol at the plasma membrane followed by its transbilayer movement, facilitated translocation to intracellular membranes, and rephosphorylation.  相似文献

The fluorescence energy transfer between two lipid probes, N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)-1, 2-dihexadecanoyl-sn-glycero-3-phosphoethanolamine (donor) and N-(Lissamine rhodamine B sulfonyl)-1, 2-dihexadecanoyl-sn-glycero-3-phosphoethanolamine (acceptor), incorporated into 1,2-dihexadecanoyl-sn-glycero-3-phosphocholine unilamellar and multilamellar lipid bilayers, is studied in the temperature region of the main phase transition. The two probes display different relative solubilities in the gel and fluid lipid-bilayer phases. A distinct maximum in the fluorescence intensity of the donor is observed in the transition region, indicating that the two probes are demixing and hence increasing their average separation. The observation is interpreted in terms of dynamic segregation of the two probes into coexisting gel and fluid lipid domains that are formed dynamically in the transition region due to strong density fluctuations. The interpretation of the experimental observations is supported by a detailed theoretical calculation using computer simulation of a microscopic model that takes full account of diffusion of the two probes and the fluctuations of gel and fluid lipid domains characteristic of the main phase transition.  相似文献

The interaction of macrophages with red blood cells (RBC) displaying phosphatidylserine (PS) in their surface membranes was investigated after the transfer of an exogenously supplied fluorescent lipid analog to the RBC. Nonfluorescent (quenched) lipid vesicles were formed by ultrasonication from 1-acyl-2-[(N-4-nitro-benzo-2-oxa-1,3 diazole)aminocaproyl]phosphatidyl-serine (NBD-PS) or 1-acyl-2[(N-4-nitrobenzo-2-oxa-1,3 diazole)aminocaproyl]phosphatidylcholine (NBD-PC). The interaction of these vesicles with RBC was monitored as a function of vesicle concentration by assessment of the degree to which cell-associated lipid fluorescence was dequenched after vesicle treatment. When vesicle concentrations of less than 100 ng/ml were used, lipid fluorescence was largely dequenched, indicating that lipid transfer was the predominant mechanism of both NBD-PS and NBD-PC uptake; however, when vesicle concentrations were increased to greater than 100 ng/ml, a concentration-dependent increase in the fraction of quenched cell-associated lipid was observed, indicating that another mechanism, possibly vesicle-cell adhesion, also occurred. Using NBD-PS at concentrations at which dilution of all the phospholipid analog in the recipient cell membrane could be unequivocally confirmed, we observed that the uptake of NBD-PS-treated RBC by macrophages was increased 5-fold over that of controls, whereas the uptake of RBC containing an equivalent amount of exogenously supplied NBD-PC was unaltered. Furthermore, preincubation of macrophage monolayers with vesicles containing PS resulted in a approximately 60% inhibition in the uptake of NBD-PS-treated RBC, whereas no inhibition in the uptake of control, opsonized, or NBD-PC-treated RBC was observed. These findings suggest that PS in the outer leaflet of RBC might serve as a signal for triggering their recognition by macrophages.  相似文献

Fourier transform infrared spectroscopy was used to characterize the lamellar phases of 1,3-dipalmitoyl-sn-glycero-2-phosphocholine (1,3-DPPC), a positional isomer of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (1,2-DPPC). The molecule exists in three distinct phases over the temperature interval 0-70 degrees C. In the low-temperature (LC) phase, the spectra are indicative of acyl chains packed in an orthorhombic subcell, while the carbonyl groups and phosphate ester at the head group show evidence of only partial hydration. The transition from the low-temperature (LC) phase to the intermediate-temperature (L beta) phase at 25 degrees C corresponds to a temperature-induced head-group hydration in which the hydration of the phosphate and carbonyl ester groups results in the reorganization of the hydrocarbon chain-packing subcell from orthorhombic to hexagonal. The transition from the intermediate (L beta) to the high-temperature (L alpha) phase at 37 degrees C is a gel-to-liquid-crystalline phase transition analogous to the 41.5 degrees C transition of 1,2-DPPC. The spectra of the acyl-chain carbonyl groups show evidence of significant differences in molecular conformation at the carbonyl esters in the LC phase. In the L beta and L alpha phases, the carbonyl band contour becomes much more symmetric. However, two components are clearly present in the spectra indicating that the sn-1 and sn-3 carbonyls experience slightly different environments. The observed differences are likely due to a preferred conformation of the phosphocholine group relative to the glycerol backbone. Indications from the infrared spectra of differences in the structure of the C = O groups provide a possible explanation for the selection of the sn-1 chain of 1,3-DPPC by phospholipase A2 on the basis of a preferred head group conformation.  相似文献

The ether-linked phospholipid 1,2-dihexadecylphosphatidylethanolamine (DHPE) was studied as a function of hydration and in fully hydrated mixed phospholipid systems with its ester-linked analogue 1,2-dipalmitoylphosphatidylethanolamine (DPPE). A combination of differential scanning calorimetry (DSC) and X-ray diffraction was used to examine the phase behavior of these lipids. By DSC, from 0 to 10 wt % H2O, DHPE displayed a single reversible transition that decreased from 95.2 to 78.8 degrees C and which was shown by X-ray diffraction data to be a direct bilayer gel to inverted hexagonal conversion, L beta----HII. Above 15% H2O, two reversible transitions were observed which stabilized at 67.1 and 92.3 degrees C above 19% H2O. X-ray diffraction data of fully hydrated DHPE confirmed the lower temperature transition to be a bilayer gel to bilayer liquid-crystalline (L beta----L alpha) phase transition and the higher temperature transition to be a bilayer liquid-crystalline to inverted hexagonal (L alpha----HII) phase transition. The lamellar repeat distance of gel-state DHPE increased as a function of hydration to a limiting value of 62.5 A at 19% H2O (8.6 mol of water/mol of DHPE), which corresponds to the hydration at which the transition temperatures are seen to stabilize by DSC. Electron density profiles of DHPE, in addition to calculations of the lipid layer thickness, confirmed that DHPE in the gel state forms a noninterdigitated bilayer at all hydrations. Thermotropic and lyotropic mesomorphism in the polymerizable lecithin 1,2-ditricosa-10,12-diynoyl-sn-glycero-3-phosphocholine and its saturated analogue, 1,2-ditricosanoyl-sn-glycero-3-phosphocholine, has been investigated by wide- and low-angle X-ray diffraction of both powder and oriented samples and by differential scanning calorimetry. Previous studies have shown that the hydrated diacetylenic lipid forms novel microstructures (tubules and stacked bilayer sheets) in its low-temperature phase. The diffraction results indicate that at low temperatures fully hydrated tubules and sheets have an identical lamellar repeat size (d001 = 66.4 A) and crystalline-like packing of the acyl chains. Chain packing in the lamellar crystalline phase is hydration independent. A model for the polymerizable lecithin with (1) fully extended all-trans methylene segments, (2) a long-axis tilt of 32 degrees, and (3) minimal chain interdigitation seems most reasonable on energetic grounds, is consistent with the diffraction data (to 3.93-A resolution), and is likely to support facile polymerization. Above the chain "melting" transition the lamellar repeat of the polymerizable lipid increases to 74 A. The conformational similarity between tubules, sheets, and the dry powder is corroborated by calorimetry, which reveals a cooling exotherm at the same temperature where tubules form upon cooling hydrated sheets. The data suggest that although a high degree of conformational order is a pertinent feature of tubules, this character alone is not sufficient to account for tubule formation. The conformation of the corresponding saturated phosphatidylcholine appears to be similar to that of other saturated phosphatidylcholines in the lamellar gel phase. Furthermore, above the main transition temperature, the dry, saturated lipid shows evidence of a P delta phase (112 degrees C), whereas the diacetylenic lipid appears to exhibit a centered rectangular phase, R alpha (55 degrees C).  相似文献

In previous studies it was shown that cannabinoids (CBs) bearing a phenolic hydroxyl group modify the thermal properties of lipid bilayers more significantly than methylated congeners. These distinct differential properties were attributed to the fact that phenolic hydroxyl groups constitute an anchoring group in the vicinity of the head-group, while the methylated analogs are embedded deeper towards the hydrophobic region of the lipid bilayers. In this work the thermal effects of synthetic polyphenolic stilbenoid analogs and their methylated congeners have been studied using differential scanning calorimetry (DSC). Molecular dynamics (MD) simulations have been performed to explain the DSC results. Thus, two of their phenolic hydroxyl groups orient in the lipid bilayers in such a way that they anchor in the region of the head-group. In contrast, their methoxy congeners cannot anchor effectively and are embedded deeper in the hydrophobic segment of the lipid bilayers. The MD results explain the fact that hydroxystilbenoid analogs exert more significant effects on the pretransition than their methoxy congeners, especially at low concentrations. To maximize the polar interactions, the two phenolic hydroxyl groups are localized in the vicinity of the head-group region, directing the remaining hydroxy group in the hydrophobic region. This topographical position of stilbenoid analogs forms a mismatch that explains the significant broadening of the width of the phase transition and lowering of the main phase-transition temperature in the lipid bilayers. At high concentrations, hydroxy and nonhydroxy analogs appear to form different domains. The correlation of thermal effects with antioxidant activity is discussed.  相似文献

Raman spectra of aqueous dispersions of 1,2-dipalmitoyl-phosphatidylcholine (DPPC) have been measured as a function of pressure (up to 46 kbar) for samples incubated at 2°C and for nonincubated DPPC samples subjected to equally high pressure. The nature of the transition from the GII gel phase of the hydrated lipid into the subgel phase on incubation is entirely different from that of the transition from the GII gel phase into the GIII gel phase of the nonincubated lipid. The GIII gel phase has a monoclinic interchain packing, while the subgel phase exhibits a triclinic interchain structure. It is shown that pressure cannot induce the transition from the GII gel phase to the subgel phase; however, it does stabilize the subgel phase above the subtransition temperature. The mechanism for the formation of the subgel phase and the complex phase behavior of the gel phase of DPPC are rationalized in terms of the dynamic properties of the acyl chains of the lipid molecule.  相似文献

The dependence of the gel-to-fluid phase transition temperature of dimyristoyl- and dipalmitoylphosphatidylserine bilayers on pH, NaCl concentration, and degree of hydration has been studied with differential scanning calorimetry and with spin-labels. On protonation of the carboxyl group (pK2app = 5.5), the transition temperature increases from 36 to 44 degrees C in the fully hydrated state of dimyristoylphosphatidylserine (from 54 to 62 degrees C for dipalmitoylphosphatidylserine), at ionic strength J = 0.1. In addition, at least two less hydrated states, differing progressively by 1 H2O/PS, are observed at low pH with transition temperatures of 48 and 52 degrees C for dimyristoyl- and 65 and 68.5 degrees C for dipalmitoylphosphatidylserine. On deprotonation of the amino group (pK3app = 11.55) the transition temperature decreases to approximately 15 degrees C for dimyristoyl- and 32 degrees C for dipalmitoylphosphatidylserine, and a pretransition is observed at approximately 6 degrees C (dimyristoylphosphatidylserine) and 21.5 degrees C (dipalmitoylphosphatidylserine), at J = 0.1. No titration of the transition is observed for the fully hydrated phosphate group down to pH less than or equal to 0.5, but it affinity for water binding decreases steeply at pH greater than or equal to 2.6. Increasing the NaCl concentration from 0.1 to 2.0 M increases the transition temperature of dimyristoyphosphatidylserine by approximately 8 degrees C at pH 7, by approximately 5 degrees at pH 13, and by approximately 0 degrees C at pH 1. These increases are attributed to the screening of the electrostatic titration-induced shifts in transition temperature. On a further increase of the NaCl concentration to 5.5 M, the transition temperature increases by an additional 9 degree C at pH 7, 13 degree C at pH 13, approximately 7 degree C in the fully hydrated state at pH 1, and approximately 4 and approximately 0 degree C in the two less hydrated states. These shifts are attributed to displacement of water of hydration by ion binding. From the salt dependence it is deduced that the transition temperature shift at the carboxyl titration can be accounted for completely by the surface charge and change in hydration of approximately 1 H2O/lipid, whereas that of the amino group titration arises mostly from other sources, probably hydrogen bonding. The shifts in pK (delta pK2 = 2.85, delta pK3 = 1.56) are consistent with a reduced polarity in the head-group region, corresponding to an effective dielectric constant epsilon approximately or equal to 30, together with surface potentials of psi congruent to -100 and -150 mV at the carboxyl and amino group pKs, respectively. The transition temperature of dimyristoylphosphatidylserine-water mixtures decreases by approximately 4 degree C each water/lipid molecule added, reaching a limiting value at a water content of approximately 9-10 H2O/lipid molecule.  相似文献

Phosphatidylethanolamines are known to exhibit asymmetric phase transitions with a low temperature shoulder. However, in this work we demonstrate that suspensions of dielaidoylphosphatidylethanolamine can be prepared which exhibit very sharp and only slightly asymmetric phase transitions. Such preparations are made either by isolating a rapidly sedimenting fraction of a vortexed suspension of this lipid or by dialyzing a suspension which had been hydrated at pH 9.2 to pH 7.2. Smaller aggregates of the lipid can be isolated from the supernate of a vortexed suspension of dielaidoylphosphatidylethanolamine after removal of the rapidly sedimenting fraction or it can be produced by sonication of a sample at pH 9.2 followed by dialysis to pH 7.2 Such preparations exhibit very broad transitions and the transition temperature is shifted to lower values. These results demonstrate that the shape of the phase transition of dielaidoylphosphatidylethanolamine is particularly sensitive to the method of sample preparation. Furthermore, an asymmetric phase transition with a low temperature shoulder is not necessarily an intrinsic property of phosphatidylethanolamines.  相似文献

In many lipid systems, polymorphic and mesomorphic behavior depends on sample thermal history. To establish unequivocally the structural origin of endothermic and exothermic events in such systems, we have performed simultaneous calorimetry and time-resolved x-ray diffraction (SCALTRD). To this end, aluminum calorimetry crucibles were used to contain the hydrated lipid sample, and the calorimeter was mounted with the base of the crucible oriented perpendicular to a synchrotron-derived focused monochromatic x-ray beam for SCALTRD data collection. Measurements were made with hydrated monoelaidin and 1,2-dielaidoyl-sn-glycero-3-phosphoethanolamine (DEPE) contained in hermetically sealed crucibles. Time-resolved x-ray diffraction (TRXRD) data were collected using an x-ray image intensifier/video system and a streak camera containing an x-ray sensitive image plate and/or film. SCALTRD analysis of the lamellar gel to lamellar liquid crystalline phase transition in hydrated monoelaidin gives identical progress curves by calorimetry and TRXRD at a scan rate of 1 degree C/min. At faster rates, calorimetry shows a broader phase transition that starts at a lower and ends at a higher temperature than is observed by TRXRD. The disparity arises in part because the x-ray beam used in TRXRD interrogates only a small portion of the sample, whereas the calorimeter responds to the entire sample volume. Because data collection times are relatively long, radiation damage is an important potential problem for SCALTRD measurements. Such an effect was observed with DEPE/water in that TRXRD shows the lamellar gel to lamellar liquid crystalline phase transition occurring at a lower temperature than observed by calorimetry. We speculate that the sample accumulates impurities locally as a result of radiation damage that has the effect of lowering the phase transition temperature at the site of interrogation by the x-ray beam. This "methods-in-combination" SCALTRD approach facilitates the direct correlation of structure rearrangements and thermal events in the same sample under identical conditions of thermal history. The information content of the data so derived far surpasses that available from either method used in isolation.  相似文献

The thermotropic properties of N-(alpha-hydroxyacyl)-sphingosine (CER[AS]) in dry and hydrated state were studied by means of X-ray powder diffraction and FT-Raman spectroscopy. The polymorphic states of the CER[AS]/water mixture (lamellar crystalline, lamellar hexagonal gel, liquid crystalline) depend on the thermal pre-treatment of the sample. Only by heating the CER[AS]/water mixture above the melting chain transition can the system be hydrated. At room temperature, both dry and hydrated states form lamellar structures, which differ in their repeat distance and packing of hydrocarbon chains. Above the melting chain transition, hydrated CER[AS] forms a liquid crystalline hexagonal phase, whereas anhydrous CER[AS] forms an isotropic liquid phase. The various phases of hydrated CER[AS] are distinguished on the basis of the corresponding Raman spectra.  相似文献

The effects of anesthetic and nonanesthetic steroids on dipalmitoylphosphatidylcholine liposomes were studied by use of high sensitivity scanning calorimetry and Raman spectroscopy. Calorimetric measurements indicated that both anesthetic and nonanesthetic steroids depressed and broadened the gel to liquid-crystalline phase transition. There was no correlation between the perturbations by the steroids on the primary gel to liquid-crystalline phase transition temperature and anesthetic potency. The magnitudes of the steroid-induced transition broadening and lowering of the pretransition temperature, however, correlated well with anesthetic potency. This effect appeared to arise from the projection from the plane of the D ring of substituents at the C(17) position of the steroid nucleus. Raman spectroscopic measurements demonstrated that the steroid molecule is localized within the acyl region of the bilayer and that effects of the steroid do not extend to either the head-group or interface regions of the lamellae. The data are consistent with unitary hypotheses relating general anesthesia to lipid perturbations. For model systems, perturbations to the subtle structural and dynamical properties of the bilayer pretransition may provide a more sensitive marker than the main phase transition in assessing the significance of lipid mediation in inducing anesthetic action.  相似文献

The metabolism and intracellular distribution of a fluorescent analog of phosphatidylinositol (PI), 1,2-[oleoyl,N-(6-[(7-nitrobenz-2-oxa-1,3-diazo-4-yl) aminocaproyl)]-PI (C6-NBD-PI), was examined in monolayer cultures of Swiss 3T3 cells following its insertion into the plasma membrane. Evidence is presented that the exogenously supplied C6-NBD-PI was hydrolyzed by a calcium-dependent PI-specific phospholipase C (PI-PLC) at the external cell surface and that this PI-specific phospholipase C may play a role in the density-dependent inhibition of cell growth: (i) When confluent monolayer cultures were incubated with C6-NBD-PI for 60 min at 7 degrees C, the lipid spontaneously transferred to the cells, and prominent labeling of intracellular membranes was observed. Lipid extraction and analysis demonstrated that more than 60% of the fluorescent lipid in these cells was fluorescent diacylglycerol (DAG). However, when the corresponding fluorescent analogs of phosphatidylcholine or phosphatidylethanolamine were used, the fluorescent lipids readily transferred to cells, but no hydrolysis to fluorescent DAG occurred. (ii) Both intracellular labeling and hydrolysis of C6-NBD-PI to -DAG were inhibited in the presence of [ethylenebis(oxyethylenenitrilo)]tetraacetic acid. (iii) When myo-[2-3H]inositol-labeled C6-NBD-PI was incubated with cells, [3H]inositol phosphate was released into the incubation medium, but no water-soluble 3H-labeled products were found associated with the cells. (iv) The level of C6-NBD-PI hydrolysis increased dramatically with increasing density of 3T3 cells in monolayer culture.  相似文献

We have examined the phase behavior of positional isomers of a polymerizable diacetylenic phospholipid, 1,2-di(heptacosadiynoyl)-sn-glycero-3-phosphocholine which has the diacetylene in varying position along the acyl chains. Upon cooling multilamellar vesicles (MLVs) through the liquid-crystalline to gel phase transition, all isomers examined spontaneously formed hollow, cylindrical microstructures (or tubules). Differential scanning calorimetry (DSC) and Fourier-transform infrared spectroscopy (FTIR) have been used to characterize positional isomers of this lipid in an effort to understand the effect of diacetylenic position on the molecular characteristics of tubule formation. Calorimetric results indicate that moving the position of the diacetylene along the acyl chain results in the alternation of the exotherm observed for the hydrated transition temperature associated with tubule formation, with higher transition temperatures (Tm) observed from isomers with an even number of methylenes between the diacetylene groups and the glycerol backbone. As the diacetylene is moved toward either end of the acyl chain, even with the observed alternation, the Tm was observed to increase. Calorimetric results of dry members of this series reveal an exotherm during cooling, the same temperature at which fully hydrated samples form tubules. This suggests that there is little difference in the phase behavior observed upon cooling the hydrated tubules and the dry diacetylenic material. FTIR results support the high degree of conformational order observed in tubules of this isomer series as a very strong CH2 wagging progression is observed between 1375 and 1200 cm-1. In addition, the C-H stretch region (3000 cm-1 to 2800 cm-1) indicates tight acyl chain packing with many all-trans segments. These results provide further evidence that tubules are uniquely crystalline microstructures and that this inherent crystallinity, and the formation of tubules is not affected by diacetylenic position.  相似文献

The phase transition of hydrated brain sphingomyelin occurs at around 35 degrees C, which is close to the physiological temperature. Freeze-fracture electron microscopy is used to characterize different gel state morphologies in terms of solid-ordered and liquid-ordered phase states, according to the occurrence of ripples and other higher-dimensional bilayer deformations. Evidently, the natural mixed-chain sphingomyelin does not assume the flat L beta, phase but instead the rippled P beta, phase, with symmetric and asymmetric ripples as well as macroripples and an egg-carton pattern, depending on the incubation conditions. An unexpected difference was observed between samples that are hydrated above and below the phase transition temperature. When the lipid is hydrated at low temperature, a sponge-like network of bilayers is formed in the gel state, next to some normal lamellae. The network loses its ripples during cold-incubation, which indicates the formation of a liquid-ordered (lo) gel phase. Ripples re-appear upon warming and the sponge-like network disintegrates spontaneously and irreversibly into small vesicles above the phase transition.  相似文献

The lateral distribution of N-[10(1-pyrenyl)decanoyl]-sphingomyelin (PyrSPM) and N-[10(1-pyrenyl)decanoyl]-glucocerebroside (PyrGlcCer) was studied in multilamellar vesicles of 1,2-dipalmitoyl-, 1,2-dimyristoyl-, and 1-palmitoyl-2-oleoyl-phosphatidylcholine (DPPC, DMPC, and POPC, respectively) under anaerobic conditions by determining the excimer-to-monomer fluorescence intensity ratio (E/M) as a function of temperature. The E/M(T) curves for PyrSPM and PyrGlcCer in the three phosphatidylcholine matrices are qualitatively similar to the curves reported for 1-palmitoyl-2-[10-(1-pyrenyl)decanoyl]-phosphatidylcholine (PyrPC) in the same three matrix phospholipids (Hresko, R. C., I. P. Sugár, Y. Barenholz, and T. E. Thompson, 1986, Biochemistry, 25:3813-3823). However, there is independent evidence to suggest that sphingomyelin and glucocerebroside are organized in POPC, DPPC, and DMPC in a more complex manner than is PyrPC. In an effort to examine further the relationship between the lateral distribution of the labeled lipid and the shape of an E/M(T) curve, E/M vs. temperature simulations were carried out together with an analysis of the equation that relates E/M to the system parameters. The results indicate that information about the lateral distribution of the pyrene-labeled lipid can be obtained from an E/M(T) curve only for those systems in which the gel to liquid crystalline phase transition temperature of the matrix lipid is higher than that of the pyrene-labeled lipid. However, very little can be known about the system from an E/M(T) curve if the matrix lipid has the lower phase transition temperature.  相似文献

The polymorphic phase behavior of a homologous series of n-saturated 1,2-diacyl phosphatidylethanolamines was investigated by differential scanning calorimetry, 31P-nuclear magnetic resonance, and Fourier transform infrared spectroscopy. Upon heating, aqueous dispersions of dried samples of the short- and medium-chain homologues (n < or = 17) exhibit single, highly energetic transitions from a dry, crystalline form to the fully hydrated, liquid-crystalline bilayer at temperatures higher than the lamellar gel-liquid-crystalline phase transition exhibited by fully hydrated samples. In contrast, the longer chain homologues (n > or = 18) first exhibit a transition from a dehydrated solid form to the hydrated L beta gel phase followed by the gel-liquid-crystalline phase transition normally observed with fully hydrated samples. The fully hydrated, aqueous dispersions of these lipids all exhibit reversible, fairly energetic gel-liquid-crystalline transitions at temperatures that are significantly higher than those of the corresponding phosphatidylcholines. In addition, at still higher temperatures, the longer chain members of this series (n > or = 16) exhibit weakly energetic transitions from the lamellar phase to an inverted nonlamellar phase. Upon appropriate incubation at low temperatures, aqueous dispersions of the shorter chain members of this homologous series (n < or = 16) form a highly ordered crystal-like phase that, upon heating, converts directly to the liquid-crystalline phase at the same temperature as do the aqueous dispersions of the dried lipid. The spectroscopic data indicate that unlike the n-saturated diacyl phosphatidylcholines, the stable crystal-like phases of this series of phosphatidylethanolamines describe an isostructural series in which the hydrocarbon chains are packed in an orthorhombic subcell and the headgroup and polar/apolar interfacial regions of the bilayer are effectively immobilized and substantially dehydrated. Our results suggest that many of the differences between the properties of these phosphatidylethanolamine bilayers and their phosphatidylcholine counterparts can be rationalized on the basis of stronger intermolecular interactions in the headgroup and interfacial regions of the phosphatidylethanolamine bilayers. These are probably the result of differences in the hydration and hydrogen bonding interactions involving the phosphorylethanolamine headgroup and moieties in the polar/apolar interfacial regions of phosphatidylethanolamine bilayers.  相似文献

The synthesis of a new bis-(D-glucopyranosid-2-yl)oxamides via the key intermediate, N-acetyl N-(methyl 3,4,6-tri-O-acetyl-alpha-D-glucopyranosid-2-yl) oxamic acid chloride (2alpha) is described. Treatment of compound 2alpha with methyl 3,4,6-tri-O-acetyl-2-amino-2-deoxy-beta-D-glucopyranoside afforded N-(methyl 3,4,6-tri-O-acetyl-alpha-D-glucopyranosid-2-yl)-N'-(methyl 3,4,6-tri-O-acetyl-beta-D-glucopyranosid-2-yl)-oxamide. Reaction of 2alpha with 1,2-diaminoethane afforded 1,2-bis-[N,N'-(methyl 3',4',6'-tri-O-acetyl-alpha-D-glucopyranosid-2'-yl)]ethyloxamide as a main product, while 2-N-[N'-(methyl 3',4',6'-tri-O-acetyl-alpha-D-glucopyranosid-2'-yl)oxamide]-ethyl acetamide was formed as a side product. Reaction of 2alpha with 1,3-diamino-2-hydroxypropane gave only 1,3-bis-N,N-[N'-(methyl 3',4',6'-tri-O-acetyl-2'-deoxy-alpha-D-glucopyranosid-2'-yl)-oxamido]-2-propanol.  相似文献
