Abstract

A scientific reserve of 200 ha has been created on Pitt Island, Chatham Islands for some 300 sheep from a flock of merino origin which has been feral for about 70 years. The reserve has a history of burning-off, and of grazing by cattle, sheep, and pigs. Transects have been established to follow changes in the vegetation. Although the original sheep were white, about 90% of the feral sheep have pigmented fleeces. Self-shedding of the wool is common in the population. The contribution that feral mammals can make to genetic conservation, and the problems that they pose to other conservation priorities, are discussed.  相似文献

Feral livestock offer an excellent opportunity lo study factors affecting fertility as the physiology of their husbanded relatives is well known and social and environmental influences can be studied free of man's interference. This is so in the population of about 1250 sheep of the primitive Soay breed on the islands of St Kilda, Scotland. There is a high coincidence of oestrus amongst the ewes in mid November, and the breeding season is constant within a few days, from year to year. The breeding season in other British breeds of sheep demonstrates a strong correlation with latitude. On St Kilda, mortality rates in male sheep are higher than in females so that the ratio of rams to ewes at mating time is about 1:5. Competition amongst rams is intense. The optimal time for mating and the place of ram lambs in the social system are discussed.  相似文献

Genetic diversity of 50 isolates of Neofusicoccum parvum, the predominant species of the Botryosphaeriaceae recovered from grapevines displaying symptoms of dieback and decline in New Zealand, was compared to that of isolates from Australia, South Africa, and California. The eight universally primed polymerase chain reaction (UP-PCR) primers distinguished 56 genotypes, with only four clonal pairs found. Seven main groups were identified in a neighbour-joining (NJ) tree with isolates from different regions and vineyards of New Zealand, Australia, and California distributed in different groups, indicating a high level of intra and intervineyard genetic variation. All of the South African isolates were positioned in a separate UP-PCR group, indicating that these isolates were less related to the other N. parvum isolates. When compared to fungi that reproduce sexually the genetic diversity and Shannon diversity indices were low (0.076-0.249; 0.109-0.367, respectively), genetic identity levels were high (0.76-0.95), and genetic distance levels were low (0.04-0.27). The large number of genotypes and the low number of clones in the New Zealand N. parvum populations may be explained by parasexual recombination as anastomosis was observed between nonself pairings. Pathogenicity tests using isolates from different UP-PCR groups inoculated onto either green shoots or 1-y-old grapevines detected virulence diversity, indicating intra and intervineyard variation between isolates, however, no correlation was detected between UP-PCR group and virulence.  相似文献

Background

Elodea canadensis, Egeria densa and Lagarosiphon major are dioecious clonal species which are invasive in New Zealand and other regions. Unlike many other invasive species, the genetic variation in New Zealand is very limited. Clonal reproduction is often considered an evolutionary dead end, even though a certain amount of genetic divergence may arise due to somatic mutations. The successful growth and establishment of invasive clonal species may be explained not by adaptability but by pre-existing ecological traits that prove advantageous in the new environment. We studied the genetic diversity and population structure in the North Island of New Zealand using AFLPs and related the findings to the number of introductions and the evolution that has occurred in the introduced area.

Results

Low levels of genetic diversity were found in all three species and appeared to be due to highly homogeneous founding gene pools. Elodea canadensis was introduced in 1868, and its populations showed more genetic structure than those of the more recently introduced of E. densa (1946) and L. major (1950). Elodea canadensis and L. major, however, had similar phylogeographic patterns, in spite of the difference in time since introduction.

Conclusions

The presence of a certain level of geographically correlated genetic structure in the absence of sexual reproduction, and in spite of random human dispersal of vegetative propagules, can be reasonably attributed to post-dispersal somatic mutations. Direct evidence of such evolutionary events is, however, still insufficient.  相似文献

The diets of feral pigs and feral goats shot on the main Auckland Island in 1989 are described from analyses of stomach and rumen contents. Feral goats ate at least 50 species of plants, but only three, Metrosideros umbellata, Chionochloa antarctica, and Durvillea antarctica made up over 50% by dried weight of the food eaten. Feral pigs ate a mixed plant and animal diet, of which plants made up 61% of the diet, with the megaherb Anisotome antipoda being the largest dietary item at 38% by dried weight. The animals eaten by pigs were mostly annelids, at 26% by dry weight of their diet, and there was little evidence of predation or scavenging on birds. Goats have subsequently been eradicated from the island, but feral pigs remain arguably a potential threat to current conservation values and certainly an impediment to any restoration of the island's natural values.  相似文献

Although the antiestrogen agent tamoxifen has long been used to treat women with hormone receptor positive invasive breast carcinoma, the mechanisms of its action and acquired resistance to tamoxifen during treatment are largely unknown. A number of studies have revealed that over-activation of some signaling pathways can cause tamoxifen resistance; however, very little information is available regarding the genes whose loss-of-function alternation contribute to tamoxifen resistance. Here we used a forward genetic approach in vitro to generate tamoxifen resistant cells from the tamoxifen sensitive breast cancer cell line ZR-75-1, and further identified the disrupted gene in different tamoxifen resistant clones. Retinol binding protein 7, DNA polymerase-transactivated protein 3, γ-glutamyltransferase-like activity 1,slit-robo RhoGTPase-activating protein, tetraspan NET-4, HSPC194, amiloride-sensitive epithelial sodium channel gene,and Notch2, were the eight mutated genes identified in different tamoxifen resistant clones, suggesting their requirement for tamoxifen sensitivity in ZR-75-1 cells. Since the functions of these genes are not related to each other, it suggests that multiple pathways can influence tamoxifen sensitivity in breast cancer ceils.  相似文献

Fertilisation is a highly programmed process by which two radically different cells, sperm and egg, unite to form a zygote, a cell with somatic chromosome numbers. Development of the zygote begins immediately after sperm and egg haploid pronuclei come together, pooling their chromosomes to form a single diploid nucleus with the parental genes. Mammalian fertilisation is the net result of a complex set of molecular events which allow the capacitated spermatozoa to recognise and bind to the egg's extracellular coat, the zona pellucida (ZP), undergo the acrosome reaction, and fuse with the egg plasma membrane. Sperm-zona (egg) interaction leading to fertilisation is a species-specific carbohydrate-mediated event which depends on glycan-recognising proteins (glycosyltransferases/glycosidases/lectin-like molecules) on sperm plasma membrane (receptors) and their complementary glycan units (ligands) on ZP. The receptor-ligand interaction event initiates a signal transduction pathway resulting in the exocytosis of acrosomal contents. The hydrolytic action of the sperm glycohydrolases and proteases released at the site of sperm-egg interaction, along with the enhanced thrust generated by the hyperactivated beat pattern of the bound spermatozoon, are important factors regulating the penetration of egg investments. This review focuses on sperm molecules believed to be important for the interaction with the female genital tract, passage through cumulus oophorus and attachment to ZP, induction of the acrosome reaction, secondary binding events, and passage through the ZP. An understanding of the expression and modifications of molecules thought to be important in multiple events leading to fertilisation will allow new strategies to block these modifications and alter sperm function.  相似文献

Myostatin, which is also known as growth and differentiation factor 8 (GDF8), acts as a negative regulator of skeletal muscle growth. Variation in the myostatin gene (MSTN) has been associated with variation in muscularity in many animals including sheep. Polymerase chain reaction-single strand conformational polymorphism (PCR-SSCP) analysis was used to investigate MSTN in a diverse range of sheep breeds including the New Zealand (NZ) Romney, Coopworth, Corriedale, Dorper, Perendale, Suffolk, Merino, Dorset Down, Poll Dorset, Texel and other NZ cross-bred sheep. A total of 28 nucleotide substitutions were identified from nucleotide c.-1199 in the promoter region to c.*1813 (based on NCBI GenBank accession number DQ530260) and including the well-described substitution c.*1232G>A (MSTN g+6223G>A). Of these 28 substitutions, 3 were located in the promoter region, 3 in the 5′UTR, 11 in intron 1, 5 in intron 2 and 5 in the 3′UTR. One substitution in exon 1 (c.101G>A) potentially results in an amino acid substitution of glutamic acid (Glu) with glycine (Gly) at codon 34. Ten of these substitutions have not been reported previously. The genetic variation revealed in this study suggests this gene is more variable than hitherto reported and provides a foundation for future research into how this variation affects muscle and growth traits.  相似文献

1. Nucleotide sequences of a 280 base pair region of the cytochrome b gene were used to assess genetic diversity and to infer population histories in the New Zealand mayfly Acanthophlebia cruentata. 2. A hierarchial examination of populations from 19 streams at different spatial scales in the central and northern North Island of New Zealand found 34 haplotypes. A common haplotype was found in all central region streams and unique haplotypes in northern streams. Several central streams had region specific haplotypes with genetically differentiated populations at the 70–100 km scale. 3. Haplotype diversity was high (0.53–0.8) at most sites, but low (0–0.22) in some central sites. amova analyses found significant genetic diversity among regions (69%) and among catchments (58%). Most population pairwise FST tests were significant, with non‐significant pairwise tests among sites in the central region and pairs of sites between neighbouring streams. 4. The levels of sequence divergence are interpreted as the result of Pleistocene divergence in multiple refugia, leading to the evolution of regionally unique haplotypes. The low diversity in some central region populations may result from recent colonisation following local extinctions, associated with volcanic events.  相似文献

Abstract

Blood plasma from three populations of feral goats (Capra hircus L.) and from five domestic breeds was analysed by polyacrylamide gel electrophoresis. The albumin locus was monomorphic in all samples. The macroglobulin locus was variable but lacked resolution on the gel. The transferrin (Tf) locus was polymorphic. The only two Tf alleles observed (Tf A and Tf B) did not vary from those described by other authors. Gene frequencies of the two Tf alleles showed that one feral population could be distinguished from the other two. Gene frequencies of domestic breeds were variable and showed no definitive pattern. Domestic breed type could not be recognised in the feral populations on the basis of transferrin analysis alone.  相似文献

Off-season reproduction is a favorable economic trait for sheep industry.Hu sheep,an indigenous Chinese sheep breed,demonstrates a higher productivity of lambs and displays year-around oestrous behavior under proper nutrition and environment.The genetic basis behind these traits,however,is not well understood.In order to identify genes associated with the off-season reproduction,we constructed a suppression subtractive hybridization(SSH) cDNA library using pooled ovary mRNAs of 6 oestrous Hu females as a tester and the pooled ovary mRNAs of 6 non-oestrous Chinese Merino females as a driver.A total of 382 resulting positive clones were obtained after the SSH.We identified 114 differentially up-regulated genes in oestrous Hu sheep by using subsequent screening and DNA sequencing,of which 8 were previously known,93 were reported for the first time in sheep,and 13 were novel with no significant homology to any sequence in the DNA databases.Functions of the genes identified are related to cell division,signal transduction,structure,metabolism,or cell defense.To validate the results of SSH,6 genes(Ntrk2,Ppap2b,Htra1,Nid1,Serpine2 and Foxola) were selected for conformational analysis using quantitative real-time PCR(qRT-PCR),and two of them(Htral and Foxola) were verified by Northern blot.All of the 6 genes were differentially up-regulated in the ovary of oestrous Hu.It is obvious that off-season reproduction is a complex trait involving multiple genes in multiple organs.This study helps to provide a foundation for the final identification of functional genes involved in the sheep ovary.  相似文献

The Lake Casitas (LC) mouse population located in south western Ventura county in California is unusual insofar as 85% of these mice are persistently viremic with congenitally transmitted murine leukemia virus (MuLV). The virus has been identified as the etiological agent responsible for lymphoma and neuromotor paralysis in large numbers of the mice. The majority of other wild mouse populations are generally free of infectious MuLV despite the presence of endogenous cellular DNA sequences homologous to infectious virus isolated from wild mice. Electrophoretic variation in 46 gene-enzyme systems was surveyed using mice from Lake Casitas and from a virus-negative population located in Bouquet Canyon (BC) approximately 40 miles from Lake Casitas. The LC and BC populations are genetically very similar to each other and to feral mouse populations previously studied in California and Europe. In the LC population 24% of the loci are polymorphic compared to 17% in the BC population. The average heterozygosities for the LC and Bc populations are 0.094 and 0.073, respectively. The large amount of genic variation in LC fails to support the concept of the derivation of the colony from a small number of founders. Tests for linkage disequilibrium and/or selective association of viremia and polymorphism at 15 loci located on nine mouse chromosomes did not reveal any nonrandom assortments. The viremic LC population, then, appears indistinguishable within the limits of experimental resolution from the virus-negative BC population in its population genetic structure.  相似文献

We investigated intraspecific transmission of Mycobacterium bovis (etiologic agent of bovine tuberculosis [Tb]) among penned pigs (Sus scrofa) in New Zealand. Between 2002 and 2006, we conducted two trials, each involving two 1-ha pens in natural habitat in which uninfected sentinel pigs were kept with wild-caught feral pigs, many of which were infected with M. bovis. The rate of M. bovis transmission to sentinels was used to assess whether intraspecies transmission between live pigs could explain the high levels of infection in the wild population. In trial 1, no new infection was detected in 18 penned sentinels after total exposure of 3.9 yr (2.6 mo/sentinel); three of 11 sentinels in the wild became infected. In trial 2, a more heavily infected source pig population (94% prevalence compared with 42% in the first trial) was used, and one (4%) of 25 penned sentinels became infected. However, 75% of the eight sentinels released to the wild became infected. Combining trials, the difference in apparent annual incidence was significant (mean ± 95% confidence interval = 0.03 ± 0.12 for penned sentinels vs. 1.06 ± 6.74 for released sentinels; t=-3.51, P=0.04). In the pens, infected pigs were kept in contact with susceptible sentinels for 7 yr in total, but only one transmission event was detected. Taking pig longevity into account, the R(0) value (the basic reproductive rate of disease) for intraspecies infection between live pigs seems unlikely to exceed 0.25, even under highly conducive conditions. We suggest that live pigs are unlikely to transmit M. bovis to wild conspecifics often enough to sustain Tb in pigs by this route of transmission alone. The high prevalences seen in some New Zealand feral pig populations are likely to result from transmission from another host or route.  相似文献

The Caatinga biome is one of the largest areas of the South American seasonally dry tropical forest that has been severely affected by unsustainable natural resource use. Furthermore, the biome has been identified as an ecologically sensitive region that is particularly susceptible to climate changes. One of the most economically important native palm tree for traditional communities from the semi-arid Caatinga is the carnauba palm, Copernicia prunifera, which offers diverse natural resources, yet its natural populations suffer intense exploitation. To inform conservation and population management strategies, we sought to determine if remaining natural populations of this species in an intensively exploited area in Northeast Brazil displayed evidence of negative genetic impacts because of exploitation and how this might interact with expected environmental changes. Mantel’s test revealed a positive and significant correlation between geographic and genetic distances, suggesting natural populations are structured by isolation by distance, while also experiencing genetic barriers as identified through Monmonier's algorithm. The studied populations showed evidence of genetic bottlenecks, while future climate scenarios suggest that potentially suitable habitats for C. prunifera within its native range will be reduced. Significant genetic differentiation among populations resulted in three distinct genetic groups which are consistent with ecological niche modelling. In addition to the need for in situ conservation of C. prunifera populations to minimize the loss of important alleles, the creation of germplasm banks for ex situ conservation and strategies for developing planted productive forests are urgently required to maintain natural populations and ensure sustainability resources for traditional communities.

The present-day Sable Island horse population, inhabiting an island off the eastern coast of Canada, is believed to have originated mainly from horses confiscated from the early French settlers in Nova Scotia in the latter half of the 18th century. In 1960, the Sable Island horses were given legal protected status and no human interference has since been allowed. The objective of this study was to characterize the current genetic diversity in Sable Island horses in comparison to 15 other horse breeds commonly found in Canada and 5 Spanish breeds. A total of 145 alleles from 12 microsatellite loci were detected in 1093 horses and 40 donkeys. The average number of alleles per locus ranged from 4.67 in the Sable Island horse population to 8.25 in Appaloosas, whereas the mean observed heterozygosity ranged from 0.626 in the Sable Island population to 0.787 in Asturcons. Various genetic distance estimates and clustering methods did not permit to support that the Sable Island horses originated from shipwrecked Spanish horses, according to a popular anecdote, but closely resemble light draft and multipurpose breeds commonly found in eastern Canada. Based on the Weitzman approach, the loss of the Sable Island horse population to the overall diversity in Canada is comparable or higher than any other horse breed. The Sable Island horse population has diverged enough from other breeds to deserve special attention by conservation interest groups.  相似文献

The aim of this study was to analyze, through the use of microsatellite molecular markers, the genetic diversity and variability of nine Alpine sheep breeds reared in Italy (Bergamasca, Biellese, Schwarzbraunes Bergschaf, Tiroler Bergschaf, and Schnalserschaf), Germany (Brillenschaf and Weisses Bergschaf) and Slovenia (Bovška and Jezerzkǒ-Solčavska), and one Italian crossbreed population. Allelic richness was rather high in each breed highlighting a considerable genetic diversity. However, the study evidenced a significant departure from Hardy–Weinberg equilibrium in all analyzed breeds caused by a heterozygote deficiency. Such lack seems to be caused both to the presence of population substructure within breed and to a rather high level of inbreeding. The genetic differentiation among breed was rather low (FST = 0.057) but significant. The clustering analysis performed with STRUCTURE detected the presence of eight clusters; Schwarzbraunes Bergschaf and Tiroler Bergschaf were grouped together and Biellese and Bergamasca as well, evidencing a high similarity of their genetic make up. Reynolds’ genetic distance estimates confirmed the close relationship between these pairs of breeds. Moreover, Bovška, Jezerzkǒ-Solčavska and Brillenschaf resulted rather similar as expected according to their region of origin and to their common ancestors. Molecular coancestry confirmed as well such findings, suggesting that the studied breeds had origin from different ancestral populations. Concluding, microsatellite resulted a useful tool to investigate breed variability and to characterize Alpine sheep breeds. Obtained findings suggest the need to set up a conservation plan aiming to safeguard and increase the genetic variability of the studied breeds compromised by the high level of inbreeding. Microsatellites genotyping could help to monitor breed variability and to organize matings.  相似文献

Cats are important in the epidemiology of Toxoplasma gondii because they are the only hosts that excrete environmentally resistant oocysts in feces. In the present study, 115 viable T. gondii isolates from tissues of cats from Egypt were genotyped using 10 PCR-restriction fragment length polymorphism markers (SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico) and DNA from tachyzoites. Seven genotypes were recognized including the clonal Type II, Type III (2 genotypes), and 4 atypical genotypes. Ninety percent (103 of 115) of isolates were clonal, i.e., Type II (n = 61) and Type III (n = 42) strains. Of the 61 Type II strains, all had the Type II alleles at all loci, except for 2 strains that had allele I at Apico. Eight isolates were divided into 4 atypical genotypes. One of these genotypes (with 4 isolates) was previously reported in dogs from Sri Lanka and in sand cats from the United Arab Emirates. Four isolates had mixed infections. These results revealed a strong clonal population structure with the dominance of clonal Type II and III lineages of T. gondii in feral cats from Egypt.  相似文献
