Stored and cooked samples of pearl millet (Pennesetum typhoides), which is regularly consumed as food by the Paharia tribe in the hilly regions of Santhal Pargana, Bihar State, India, that were harvested in January 1989 were analyzed for mold flora, natural occurrence of Aspergillus flavus and A. parasiticus, and incidence and levels of aflatoxin B1. Of the 22 fungal species isolated, A. flavus and A. parasiticus were the predominant species (63.8%) during the rainy season, followed by other species of Aspergillus, Penicillium, Fusarium, Rhizopus, Helminthosporium, and Curvularia. Screening of 169 A. flavus and A. parasiticus strains showed that 59 of them were toxigenic, producing various combinations of aflatoxins B1, B2, G1, and G2. The amounts of aflatoxin B1 ranged between 4 and 30 mg/100 ml of liquid medium. Analysis of stored and cooked samples also revealed a high incidence and alarming levels of naturally produced aflatoxin B1. Forty-nine of 75 stored and 16 of 38 cooked samples contained various combinations of aflatoxins. The levels of aflatoxin B1 ranged between 17 and 2,110 ppb in stored samples and 18 and 549 ppb in cooked samples. The correlation of insect damage with A. flavus and A. parasiticus incidence and quantity of aflatoxin B1 was found to be insignificant.  相似文献

Fungi of 19 genera, 30 species, and one variety were isolated from 25 samples of sheep-, cattle- and camel feedstuffs collected from different farms in the Beida Governorate, Libya.Aspergillus, Penicillium andFusarium were the most common genera in the three substrates tested. TLC was used to establish the identity of aflatoxins in the chloroform extract of all samples and the ability to produce aflatoxins byAspergillus flavus in a synthetic liquid medium. Twenty samples out of 25 tested were naturally contaminated and 21 isolates ofA. flavus out of 30 produced at least one of the following aflatoxins: B1; B1, G1; and B1, B2, G1, G2. This is the first report about the natural occurrence of aflatoxins and aflatoxin-producers of the genusAspergillus in Libya.  相似文献

A survey of different types of cereal straw samples viz. paddy, maize and wheat, from Bihar State, India, was conducted in order to examine the mould flora and mycotoxin contamination. Out of 170 samples examined for mould flora,Aspergillus flavus group of fungi had highest level of incidence followed byA niger. Isolates ofA flavus, A ochraceus, Fusarium verticillioides andPenicillium citrinum were screened for their mycotoxins producing abilities. Out of 75, 63 and 68 isolates ofA flavus group obtained from stored straw of paddy, maize and wheat samples, respectively, 27 (36%), 14 (22%) and 24 (35%) were found to be toxigenic which produced different combinations of aflatoxins in different concentrations. The percentage toxigenicity was comparatively lower in the isolates of other mycotoxigenic fungi from all types of samples. Out of 222 samples of straw analysed for natural occurrence of different mycotoxins, besides the aflatoxins present, zearalenone, ochratoxin A and citrinin were also recorded alone or as co-contaminants. A conducive climate together with the socioeconomic conditions of this region are important determinants for the high incidence of mycotoxins in cereal straw samples.  相似文献

The response to delayed harvest of fungal and mycotoxin contamination of grain of the pearl millet hybrid HGM 100 was examined in 1992 and 1993. Samples of grain were assayed from seven plantings at locations near Tifton, Georgia, USA. Grain was harvested at 30, 40, and 50 days after anthesis and evaluated for infection byFusarium species andAspergillus flavus, and mycotoxin contamination. Mean isolation frequencies ofF. semitectum (35.6%) andF. chlamydosporum (17.2%) increased linearly with delayed harvest.Fusarium moniliforme andF. equisiti were infrequently isolated (<0.5%) and did not increase in the grain when harvest was delayed. Low mean concentrations of zearalenone (0.17 ppm), nivalenol (0.42 ppm), and deoxynivalenol (0.01 ppm) were detected but were not affected by delayed harvest. Isolation frequencies ofF. chlamydosporum andF. equiseti were correlated (P=0.07) with levels of nivalenol.Aspergillus flavus was not isolated from the grain, and aflatoxin concentrations averaged 1.9 ppb.  相似文献

Background

Soil-transmitted helminths (STHs) infect over a billion individuals worldwide. In India, 241 million children are estimated to need deworming to avert the negative consequences STH infections can have on child health and development. In February-April 2011, 17 million children in Bihar State were dewormed during a government-led school-based deworming campaign. Prior to programme implementation, a study was conducted to assess STH prevalence in the school-age population to direct the programme. The study also investigated risk factors for STH infections, including caste, literacy, and defecation and hygiene practices, in order to inform the development of complementary interventions.

Methods

A cross-sectional survey was conducted among children in 20 schools in Bihar. In addition to providing stool samples for identification of STH infections, children completed a short questionnaire detailing their usual defecation and hand-hygiene practices. Risk factors for STH infections were explored.

Results

In January-February 2011, 1279 school children aged four to seventeen provided stool samples and 1157 children also completed the questionnaire. Overall, 68% of children (10-86% across schools) were infected with one or more soil-transmitted helminth species. The prevalence of ascariasis, hookworm and trichuriasis was 52%, 42% and 5% respectively. The majority of children (95%) practiced open defecation and reported most frequently cleansing hands with soil (61%). Increasing age, lack of maternal literacy and certain castes were independently associated with hookworm infection. Absence of a hand-washing station at the schools was also independently associated with A. lumbricoides infection.

Conclusions

STH prevalence in Bihar is high, and justifies mass deworming in school-aged children. Open defecation is common-place and hands are often cleansed using soil. The findings reported here can be used to help direct messaging appropriate to mothers with low levels of literacy and emphasise the importance of water and sanitation in the control of helminths and other diseases.  相似文献

In the Sahel, short periods of intra-seasonal drought, caused by unfavourable rainfall distribution, often have stronger effect on crop growth than fluctuations in annual rainfall. The interactive effects of nutrient deficiency and water shortage (during panicle initiation, flowering and grain filling) on yield and yield components of pearl millet (Pennisetum glaucum (L.) R. Br.), were studied on-farm along a cultivated slope, during three years with close to average annual rainfall. Grain yield was correlated to plant nutrient availability but not to annual rainfall, which was explained by the capacity of the crop to compensate for damage caused by water shortage during early growth phases. The performance of each yield component was positively correlated to cumulative rainfall during the growth phase when it was formed. Leaf area index (LAI) was very low, and leaf development followed rainfall distribution. Water and nutrients interacted during each growth phase for all fertility levels. Fertilised millet suffered less during water shortage at panicle initiation and at grain filling compared to non-fertilised millet. However, compared to favourable soil water conditions yield components were systematically lower for all treatments, indicating the synergistic effect of water and nutrients. The results suggest that water availability plays an exclusive role during flowering. Grain number dropped significantly due to water shortage and was similar for all treatments. Despite extremely high spatial variability in yields (varying with a factor 46 within the field), a significant slope effect was observed, of progressively increasing yields when moving downslope. Spatial redistribution of surface runoff resulting in higher soil water availability on lower slope positions, contributed to the yield gradient, which was reinforced for fertilised millet. For each drought period, yield components suffered systematically more upslope than downslope. This slope effect was smoothed out for manured millet, which indicates that manure increased soil infiltrability on crusted zones upslope. The slope interaction observed here – indicating that downslope (i) the risk for crop failure during droughts is lower and (ii) the response to fertilisers is greater – suggests that farmers can benefit relatively more from fertilisers applied in lower parts of the watershed. Taking advantage of spatial soil and water variability is an interesting system of low technology precision farming, which combined with water harvesting systems to master droughts, can constitute options for increased crop yields in the Sahel.  相似文献

The floodplain of the Himalayas is a land formed and destroyed incessantly by the water of its rivers. Measures intended for flood control, aimed at separating productive land from river water through earth levees, have instead worsened the inundations, disrupting the beneficial flow of soil through floodwaters, obstructing water drainage, and resulting in enormous waterlogged areas. This article proposes that the failure of flood control in Bihar, India, is due to misunderstanding the river as a matter of water only, hence attempting the conceptual naturalization of an otherwise relative ontological distinction between water and land. Local knowledge of water reveals that neither water nor land can even be named, let alone understood, without the other. Informed by ethnographic fieldwork and multidisciplinary research in North Bihar, this article presents land and water as being in intimate correspondence with each other. By virtue of comparison, the ethnographic encounter is held to defy other ontologies of water that see the two substances as being in opposition. As a result, this article posits ontologies of natural substances as ‘watertight’, sclerotic, mutually exclusive, unable to adapt, and prone to be caught in a semiotic conflict.  相似文献

Summary Dwarf varieties have had virtually no impact on the production of pearl millet, in contrast to the case of wheat, rice, and sorghum. This research compared tall and dwarf near-isogenic F1 hybrids to attempt to determine if there were deleterious effects of the d 2 dwarfing gene that might account for the lack of release/cultivation of dwarf pearl millet cultivars. Dwarf isohybrids on average yielded less than the tails, because of a smaller average seed size combined with a similar grain number per unit area. There was, however, a larger contribution of background genetic variation (pollinator, male-sterile, and interaction effects) to hybrid variation for nearly all characters measured, including seed size, than there was of the dwarfing gene. The effects of developmental stage of the anther, ageing of the plant, inbreeding and season on meiotic segregation of an interchange chain quadrivalent were studied in pearl millet, Pennisetum americanum (L.) Leeke. The frequency of adjacent orientation (and segregation) increased with developmental stage of the anther, independent of other factors. Plant age and degree of inbreeding had no demonstrable effects, but there was an indication that high temperature favoured adjacent orientation. Chromosome contraction as measured by change in chromosome length appeared to be negligible during metaphase-anaphase. Therefore, increased adjacent orientation cannot readily be explained by metaphase reorientation resulting from the straightening of chromosomes caused by an increase in their rigidity. It is probable that the unoriented or mal-oriented quadrivalents observed regularly at early metaphase I continue to straighten out prior to their delayed orientation. When they finally orientate late in metaphase, their orientation will more likely be adjacent than alternate.  相似文献

Maize (Zea mays L.) and pearl millet (Pennisetum americanum (L.) Leeke) seedlings were exposed to [15N]nitrate for 1-h periods at eight times during a 24-h period (16–8 h light-dark for maize; 14–10 h for millet). Influx of [15N]nitrate as well as its reduction and translocation were determined during each period. The efflux of previously absorbed [14N]nitrate to the uptake solution was also estimated. No marked diurnal changes in [14N]nitrate efflux or [15N]nitrate influx were evident in maize. In contrast, [14N]nitrate efflux from millet increased and eventually exceeded [15N]nitrate influx during the late dark and early light periods, resulting in net nitrate efflux from the roots. The dissimilarity of their diurnal patterns indicates that influx and efflux are independently regulated. In both species, [15N]nitrate reduction and 15N translocation to shoots were curtailed more by darkness than was [15N]nitrate influx. In the light, maize reduced 15% and millet 24% of the incoming [15N]nitrate. In darkness, reduction dropped to 11 and 17%, respectively. Since the accumulation of reduced-15N in shoots declined abruptly in darkness, whereas that in roots was little affected, it is suggested that in darkness [15N]nitrate reduction occurred primarily in roots. Aflatoxin contamination in five varieties of pearl millet (ICMH-451, ICMP-50I, ICTP-8203, WCC-75 and ICMV-155) was studied from field and storage conditions in three districts of Andhra Pradesh State, India and the inter-relationships between various parameters such as stage of grain maturation in the field and insect pest infestation in storage in relation to aflatoxin production were evaluated. Aflatoxin contamination was more frequent in the seed samples collected from the fields during rainy season than winter season. All major aflatoxins were isolated from one or the other varieties of pearl millet, whereas aflatoxin G2 was not commonly observed in the seed samples collected during winter. Among all the varieties tested, ICMH-451 was vulnerable to aflatoxin contamination whereas ICMV-155 was the least susceptible variety. The higher amount of aflatoxins was observed in the matured seed samples followed by pre-matured and milky stage. Among all the toxins reported in the field, aflatoxin B1 was found in higher concentration (185 (μg/kg) followed by B2 (105 μg/kg). The four major types of aflatoxins with higher levels (35, 40, 140, 190 μg/kg of G1, G2, B2, B1 were reported in the rainy season seed samples after six months of storage, whereas aflatoxin G1 was not observed in any variety of stored seed sample from winter. Statistical analysis revealed that the aflatoxin incidence in relation to different parameters studied was significantly different for each factor. The relationship between aflatoxin contamination and insect damaged-grain clearly indicated that the seed samples with 16-40% of insect damage contained higher amounts of aflatoxins (758 μg/kg). Summary The synergids and other cells of mature, unpollinated pearl millet ovaries were investigated using: (1) freeze-substitution fixation in conjunction with scanning electron microscope observations and energy-dispersive X-ray microanalysis to localize total calcium (Ca) and other elements, and (2) antimonate precipitation to selectively localize loosely sequestered, exchangeable calcium (Ca++). In freeze-fixed ovaries, the synergid cells, ovary wall, nucellus, and other regions of the ovary displayed, respectively and relatively, extremely high, high, moderate, and low levels of Ca. In antimonate-fixed ovaries, Ca-containing antimonate precipitates exhibited similar distribution patterns. In ovaries fixed using the conventional 2% (w/v) antimonate in fixatives, the synergids were disrupted due to precipitate overload. In the ovary wall, precipitates were mainly located in the intercellular spaces. Some precipitates were observed at the micropyle and along the outer ovule integument, associated with diffuse extracellular material, and in the cell walls of nucellar cells proximal to the micropyle. Examination of precipitate distribution inside the synergids was possible in ovaries fixed using 0.5% (w/v) antimonate in the fixatives. Cytoplasmic organelles of all synergids examined exhibited variable states of disintegration. The amount of precipitates associated with the degenerated organelles appeared to be proportional to the degree of their degeneration. Distinct precipitates were localized in contiguous regions of the nucellar cells fused with the embryo sac, the micropylar half of the embryo sac wall, and the filiform apparatus. Summary LDH variants were tested for in 392 bloods samples taken from Christian and non-Christian Munda groups, of Ranchi, Bihar by the filter paper technique. Six cases of LDH Cal-1 variant were detected (1.53%). This result is compared with that obtained in Hindu upper caste and Hindu lower caste of West Bengal, and the Munda population was found to be closer to the lower caste people than the upper caste (t=1.752, 0.05> P>0.025).
Zusammenfassung 392 Blutproben von Mundas aus christlichen und nichtchristlichen Gruppen, aus Ranchi (Bihar) wurden mit Hilfe der Filterpapiertechnik gesammelt und auf LDH-Varianten gestestet. 6 Fälle (1,53%) der Variante LDH Cal-1 wurden entedeckt. Dieser Wert wird verglichen mit dem von Hindus höherer und niederer Kasten aus West-Bengalen. Die Mundas ähneln den unteren Kasten mehr als den höheren (t=1,752; 0,05> P>0,025).

Genetic analysis of five in vitro characters was made through a 5 × 5 diallel analysis using callus derived from immature inflorescence segments of pearl millet (Pennisetum glaucum). The characters studied were: — volume of total callus, — frequency of embryogenic calli, — embryogenic callus volume, — growth rate in terms of increase in fresh weight, and — frequency of regeneration. High heritability values and heterosis were noticed for all these characters except for E callus frequency. Additive gene action was predominant for callus growth rate and frequency of regeneration. Of the five inbreds, IP 1346 (= P5) was found to be the best genetic background for embryogenic callus volume, embryogenic growth rate and frequency of regeneration. Summary Combining ability studies with respect to grain quality characteristics viz., beta-Carotene, total carotenoids, protein content, 250-grain weight, grain hardness and grain yield were carried out from a 13x13 diallel cross set in pearl millet [Pennisetum typhoides (Burm S&H)].The parents versus hybrids comparison indicated significant heterosis for all the traits under study. In general the hybrids having higher grain yield had bold hard grains with more carotene but low protein content, although a few hybrids combined high yield with an average protein percentage. The relative proportions of the general and specific combining ability variances indicated predominance of non-additive genetic variance with respect to all the traits. The per se performance of parents provided a fairly good indication of their combining ability in most cases. Parents possessing desirable grain quality characteristics were identified. Prometaphase I orientation, reorientation and anaphase I segregational behaviour of a chain-forming interchange quadrivalent involving one of the long chromosomes and the long arm of the seventh (nucleolar) chromosome was studied during anther development in pearl millet. The data obtained from 34 anthers showed that by early prometaphase I about 90% of the bivalents have attained stable bipolar orientation but about 48% of the quadrivalents are mal-oriented. There seems to be an interaction between bivalents and quadrivalents during mal-orientation and reorientation. The mal-oriented bivalents reoriented before the quadrivalents. For quadrivalent mal-orientation four types, 4/0, 3/1, 2/1/1/1 and 2/2 (adjacent 1), were distinguished in addition to the regular types, adjacent 2 and alternate. Based on their potential to reorient, the order of the mal-oriented quadrivalent types was 4/0 > 3/1 > 2/1/1; 2/2 led to anaphase I disjunction as for an adjacent 1 segregation. The data from 36 anthers at anaphase I showed alternate segregation of chromosomes in nearly 50% of pollen mother cells (PMCs) up to a developmental index of about 65. In late anthers about 35% PMCs showed alternate segregation. This suggests that the PMCs that reached metaphase I later had more adjacent 2 orientations since mal-oriented configurations delay meiotic development, and implies preferential reorientation behaviour of the maloriented quadrivalent types.  相似文献

The mycoflora of a range of mixed ruminant feed and forage samples collected in Eastern India and Bangladesh were determined. Most fungi isolated from the samples were those associated with low moisture contents. Several potentially mycotoxigenic fungi and thermotolerant species were present particularly in the rice straw. The most frequently isolated fungi were: Aspergillus versicolor (Vuill.) Tiraboschi, Penicillium citrinum Thorn, Eurotium species, Wallemia sebi (Fr.) v. Arx, Aspergillus penicilloides Speg. and yeasts. Fusarium moniliforme Sheldon and Cladosporium species were prevalent in straw but not other feeds. Levels of certain individual fungi, particularly thermotolerant types, were higher in Bangladesh rice straw although overall counts were similar to those of the Indian samples. Mycotoxin analyses for aflatoxins, zearalenone and sterigmatocystin were carried out. Aflatoxins were detected in significant amounts in some feed samples from Eastern India but not in rice straw from India and Bangladesh. Zearalenone occurred in a Paspalum straw sample and three other feed samples from India and sterigmatocystin was detected in three rice straw samples from Bangladesh.  相似文献
