Monensin, a carboxylic ionophore, produces strong pressor, positive chronotropic effects and elevates the blood glucose level when injected intravenously (100 μg/kg) into pentobarbital anesthetized dogs or administered orally (2 mg/kg) to conscious dogs. Intravenously administered monensin disappeared from the blood rapidly with a t12 of ca. 2.5 min and, in the conscious dogs, ingested monensin showed a peak plasma level 90 min after feeding; this coincided with the time of maximum increase in arterial blood pressure and blood glucose. In conscious rabbits, although higher doses of monensin were administered, 200 μg/kg intravenously and 10 mg/kg orally, its cardiovascular effects were less than observed in the dog and were slower in onset. This correlated with slower clearing of injected monensin from the blood (t12 = 8 min) and slower entry of ingested monensin from the gut into the blood. Rabbit plasma and tissue levels were higher 17 hr after oral ingestion of monensin than six hr after ingestion.  相似文献

An investigation of the effect of PGF2α induced parturition alone or in combination with neostigmine (NEO) on subsequent productivity was conducted in a total confinement swine production unit. Forty-five crossbred gilts and 106 sows were randomly assigned to one of six treatments: (1) 0 mg PGF2 α0 mg NEO, (2) 10 mg PGF2 α0 mg NEO, (3) 0 mg PGF2 α5 mg NEO, (4) 10 mg PGF2 α5 mg NEO, (5) 0 mg PGF2 α10 mg NEO, (6) 10 mg PGF2 α10 mg NEO. PGF2α injections (IM) were given 2 days prior to the mean expected farrowing day for this farm (day 112). NEO injections (IM) were given after the fourth pig born in gilt litters and after the fifth pig born in sow litters. Females injected with 0 or 10 mg PGF2α farrowed 71.7 ± 3.7 and 31.4 ± 3.4 hours after treatment, respectively (P<0.01). No other parameters measured were significantly affected by injection of PGF2α. These data show that PGF2α effectively induced parturition and did not adversely affect subsequent litter productivity. NEO was found to be ineffective in reducing total farrowing time (P>0.05).  相似文献

We have utilized sodium butyrate-treated normal human diploid fibroblasts to study core histone hyperacetylation kinetics. We report a small, distinct population of core histone characterized by a very rapid rate of hyperacetylation (t12≈10–15 min for monoacetylated histone H4) compared to the slower rate (t12≈140–200 min for monoacetylated H4) observed for bulk histone. Two rates of core histone deacetylation were also detected and we demonstrated that the rapidly hypermodified histone H4 population was also rapidly deacetylated. The kinetics of histone H4 hyperacetylation and deacetylation in these cells were not significantly altered, regardless of whether cultures were exponentially growing, confluent or arrested in an essentially non-mitotic state.  相似文献

The in vitro incorporation of cytochrome b5 into purified plasma membranes was investigated by biochemical and immunological methods. Plasma membrane preparations incorporated three times less cytochrome b5 than did microsomal preparations; 60% of this cytochrome b5 could not be reduced by the NADH-cytochrome b5 reductase and was considered as being bound to the plasma membrane. The morphological observations made after the immunochemical labeling of cytochrome b5 clearly showed a good but asymmetrical distribution of the ferritin labeling: only the inner face of the plasma membrane incorporated cytochrome b5. These results are discussed with respect to theories which concern the subcellular membrane relationships in the cell.  相似文献

Initial rate, product inhibition, and isotope rate kinetic studies of pig heart mitochondrial and supernatant malate dehydrogenases, acting upon the nonphysiological substrates, meso-tartrate and 2-keto-3-hydroxysuccinate, are reported. The measured spontaneous keto-enol equilibrium for 2-keto-3-hydroxysuccinate in 0.05 m Tris-acetate (pH 8.0) at 25 °C favors the enol form, dihydroxyfumarate, with an apparent equilibrium constant of 0.036. The enzyme-catalyzed reaction favors meso-tartrate with an apparent equilibrium constant of 1.25 × 10?6, M?1 at pH 8.0. The mechanism apparently remains ordered bi bi for both enzymes when these nonphysiological substrates are used, and the chemical-converting hydride transfer step becomes more rate limiting for both enzymes. This conclusion is supported by VHVD and (VHKH)VDKD values of 2.6 and 3.1, respectively, for the mitochondrial enzyme and 1.9 and 2.9, respectively, for the supernatant enzyme.  相似文献

The uptake of radiolabeled carnitine and butyrobetaine has been studied in human heart cells (CCL 27). The uptake of carnitine is 3–10-fold higher in heart cells than in fibroblasts (pmol · μg DNA?1). The uptake of carnitine increases with temperature coefficient KT of 1.6 in the interval 10–20° C and with a negligible uptake at 4 and 10° C. The uptake of carnitine follows Michaelis-Menten kinetics with a KM of 4.8 ± 2.2 μM and V = 8.7 ± 3.2 pmol · μg DNA?1 · h?1. Carnitine uptake is suppressed 90% by NaF (24 mM). Butyrobetaine is taken up into heart cells to the same extent as carnitine with a KM of 5.7–17.3 μM and V = 8.7–9.3 pmol · μg DNA?1 · h?1. Butyrobetaine inhibits competitively the uptake of carnitine and carnitine inhibits the uptake of butyrobetaine to the same extent. No conversion of radiolabeled butyrobetaine to carnitine, or carnitine to methyl choline was observed intra- or extracellulary during incubation. These data are compatible with a selective transport mechanism for carnitine which is also responsible for the uptake of butyrobetaine.  相似文献

Respiration (O), ammonium (NH4), phosphate (PO4), total nitrogen (NT) and phosphorus (PT) excretions were measured on mixed zooplankton during 3-, 6-, 9-, 12-, 21-, and 24-h incubation periods at 20–23 C. The excretion rates of PO4, NT. and PT decrease during a 21-h period, while rates of respiration and excretion of NH{IN4} are constant. The percentage of inorganic nitrogen excreted increases regularly from 3 h (30–40% of total nitrogen) to 21 h (70–80%) and it could be either due to a bacterial activity which was measured or to a decrease with time of organic nitrogen excreted because of starvation. ONT, OPO4, OPT, and NH4PO4 ratios increase during the first 9 h of incubation; the percentage of inorganic phosphorus excreted is higher at the very beginning and then remains constant from 6 to 24 h. ONH4 and NTPT ratios are constant during a 24-h term, which makes them useful metabolic indexes.  相似文献

In the redheaded bunting Emberiza bruniceps, thyroidectomy inhibited premigratory fattening and nocturnal restlessness—two characteristics of avian migration—observed in caged birds during the premigratory period (March/April). Thyroxine (T4) and triiodothyronine (T3) administration in thyroidectomized birds stimulated locomotor activity and restored the loss in body weight. Annual variations in circulating thyroid hormone concentrations revealed a significant rise in T3T4 ratio prior to spring migration in both years studied. This increase in circulating T3T4 ratio may be associated with the development of migratory disposition in this bird. There was no increase in circulating T3T4 ratio prior to autumnal migration, however, plasma T4 increased significantly. Different thyroidal mechanisms are most likely involved in spring and fall migratory periods. While T3 remained low throughout, apart from the characteristic spring rise, high T4 levels in E. bruniceps were associated with periods of reproduction and molting, the latter coinciding partly with autumnal migration.  相似文献

Stable isotope labeled methadone (pentadeuteromethadone) has been used in conjunction with gas chromatography-chemical ionization mass spectrometry to study plasma disappearance rates and urinary excretion of pharmacologically active R-(?)-methadone (l-methadone) and inactive S-(+)-methadone (d-methadone) in three adult methadone maintenance patients. In all three cases, the analgesically active enantiomeric form of the drug had a significantly longer elimination half-life (t12β) when studied in a steady state than did the inactive form (t12β for active R-(?)-methadone, 51.7 to 61.8 hours; t12β for inactive S-(+)-methadone, 31.8 to 37.0 hours). The ratio of drug elimination half-lives } R-(?)-/S-(+)- ranged between 1.40 and 1.94. In the two cases so studied, slower plasma disappearance of active R-(?)-enatiomer than the inactive S-(+)-enantiomer was also observed (t1 R-(?)-, 42.8 and 52.5 hours; t1 S-(+)-, 38.3 and 41.3 hours).  相似文献

In order to test the question if a pool of lipophilic ions may exist in black lipid membranes which cannot be detected by electrical relaxation measurements we have performed simultaneously measurements of the optical absorption of a lipophilic ion. The absorbance of membrane-bound dipicrylamine at 410 nm was measured with a sensitive spectrophotometer which can detect absorbance changes ? 4 · 10?5. A minimal concentration of about 6 · 1011 dipicrylamine ions per cm2 of the membrane could be detected with this instrument. The dipicrylamine concentration in the membrane obtained with the optical method Ntopt is compared with the concentrations Ntel obtained from simultaneous electrical relaxation measurements. Ntopt and Ntel agreed at low dipicrylamine concentrations (10?8–10?7 M in the aqueous phase) and showed saturation at higher concentrations (up to 5 · 10?6 M). In the saturation range Ntopt was maximally four times higher than Ntel. The significance of this difference is discussed together with general aspects of the saturation phenomenon.  相似文献

Infinite cis uptake of cyclic AMP into red blood cell ghosts has been measured. The Kicoi is calculated from two different integrated rate equations that are applicable when the substrate concentration is unsufficient to cause volume changes. Values of 0.69 mM and 0.66 mM are obtained for the infinite cisKm at 30°C using these procedures. These values are only slightly higher than that predicted from zero trans net flux experiments.Lowering the temperature reduces Kicoi from 0.69 mM at 30°C to 0.478 mM at 20°C, 0.108 mM at 10°C and 0.072 mM at 4°C (Q10 = 2.4). The Q10 for activation of influx permeability of 10?5 M cyclic AMP is 1.55.  相似文献

Phosphate transporter of bovine heart mitochondria was purified by solubilization of submitochondrial particles with octylglucoside and fractionation of the extract with ammonium sulfate. After reconstitution into liposomes the purified protein catalyzed phosphate transport which was sensitive to mersalyl and other SH reagents. Transport measured either as PiOH or PiPi exchange was proportional to protein concentration and time. The PiOH but not the PiPi exchange was stimulated several fold by valinomycin plus nigericin in the presence of K+. The reconstituted system provides a suitable assay during purification of the mitochondrial phosphate transporter.  相似文献

The apparent Arrhenius energy of activation (Ea) of the water osmotic permeability (Posc) of the basolateral plasma cell membrane of isolated rabbit proximal straight tubules has been measured under control conditions and after addition of 2.5 mM of the sulfhydryl reagent, para-chloromercuribenzenesulfonic acid (pCMBS), of mersalyl and of dithiothreitol. Ea (kcal/mol) was 3.2 ± 1.4 (controls) and 9.2 ± 2.2 (pCMBS), while Posc decreased with pCMBS to 0.26 ± 0.17 of its control value. Mersalyl also decreased Posc both in vitro and in vivo (using therapeutical doses). These actions of pCMBS and mersalyl were quickly reverted with 5 mM dithiothreitol and prevented by 0.1 M thiourea. Ea for free viscous flow is 4.2 and greater than 10 for non-pore-containing lipid membranes. By analogy with these membranes and with red blood cells, where similar effects of pCMBS on Pos are observed, it is concluded that cell membranes of the proximal tubule are pierced by aqueous pores which are reversibly shut by pCMBS. Part of the action of mercurial diuretics can be explained by their action on Posc.  相似文献

The transport of sucrose by selected mutant and wild-type cells of Streptococcus mutans was studied using washed cocci harvested at appropriate phases of growth, incubated in the presence of fluoride and appropriately labelled substrates. The rapid sucrose uptake observed cannot be ascribed to possible extracellular formation of hexoses from sucrose and their subsequent transport, formation of intracellular glycogen-like polysaccharide, or binding of sucrose or extracellular glucans to the cocci. Rather, there are at least three discrete transport systems for sucrose, two of which are phosphoenolpyruvate-dependent phosphotransferases with relatively low apparent Km values and the other a non-phosphotransferase (non-PTS) third transport system (termed TTS) with a relatively high apparent Km. For strain 6715-13 mutant 33, the Km values are 6.25·10?5 M, 2.4·10?4 M, and 3.0·10?3 M, respectively; for strain NCTC-10449, the Km values are 7.1·10?5 M, 2.5·10?4 M and 3.3·10?3 M, respectively. The two lower Km systems could not be demonstrated in mid-log phase glucose-adapted cocci, a condition known to repress sucrose-specific phosphotransferase activity, but under these conditions the highest Km system persists. Also, a mutant devoid of sucrose-specific phosphotransferase activity fails to evidence the two high affinity (low apparent Km) systems, but still has the lowest affinity (highest Km) system. There was essentially no uptake at 4°C indicating these processes are energy dependent. The third transport system, whose nature is unknown, appears to function under conditions of sucrose abundance and rapid growth which are known to repress phosphoenolpyruvate-dependent sucrose-specific phosphotransferase activity in S. mutans. These multiple transport systems seem well-adapted to S. mutans which is faced with fluctuating supplies of sucrose in its natural habitat on the surfaces of teeth.  相似文献

High avidity antisera against β-endorphin h-EP) were obtained in two of five rabbits immunized with unconjugated synthetic human βh-EP. One of these antisera (K-7762) cross-reacted 1.5% on a molar basis with β-lipotropin h-LPH) and did not recognize leucine-enkephalin in a concentration as high as 0.2 mmol/l. The cross-reaction with methionine-enkephalin h-LPH 61–65) was 9%, while that with α-endorphin (βh-LPH 61–76) was 69%. This implied that the specific recognition site was in the amino-terminal region of βh-EP. Although this sequence is present in βh-LPH it was poorly recognized by the antiserum, suggesting that the free amino-terminal is essential. This interpretation was supported by the finding that α-N-acetylh-EP was equally poorly recognized by the antiserum. The sensitivity of the radioimmunoassay was 1.9 pmol/l. βh-EP was not detectable (< 3 pmol/l) in 26 of 27 extracted plasma samples in healthy blood donors, in one it was 5 pmol/l. In five of six patients with an enlarged sella turcica, but without clinical and laboratory evidence of pituitary dysfunction, βh-EP was detectable (5 ± 3 pmol/l; mean ± S.D.) after metyrapone. βh-EP was elevated in Addison's disease (23, 54 and 76 pmol/l), Nelson's syndrome (37, 39 and 109 pmol/l), ectopic ACTH production (27, 59 and 76 pmol/l), but only detectable in one of three samples from patients with Cushing's disease (7 pmol/l). Gel chromatography of extracts of porcine pituitary revealed only one immunoreactive peak co-eluting with synthetic human βh-EP. The specificity of the antiserum K-7762 was such that the βh-EP concentration in plasma extracts could be reliably estimated by radioimmunoassay without prior chromatography.  相似文献

To determine whether changes in unsaturation of fatty acids in rat liver plasma membranes might alter activities of membrane-associated enzymes, liver plasma membranes were prepared from rats fed purified diets lacking or supplemented with essential fatty acids. Two methods of membrane purification were used. A similar degree of purification was obtained with both methods for both depleted and control membranes, as indicated by marker enzyme purification. The proportion of essential fatty acids of the linoleate series was significantly lower in phospholipids from depleted rats. The specific activity of 5′-nucleotidase was lower, and the activity, V and apparent Km for total (Na++K++Mg2+)-ATPase were higher in the depleted liver plasma membranes. Arrhenius plots of total ATPase activity showed a discontinuity at the same temperature for both the depleted and control membranes. Activity with the depleted membranes was higher at all temperatures tested. Supplementation of deficient rats with a source of essential fatty acids (corn oil) restored V and apparent Km values to normal. Adenylate cyclase activity in the presence of fluoride, glucagon or glucagon plus GTP was significantly lower in the depleted plasma membranes.  相似文献

In an accompanying publication by Duckwitz-Peterlein, Eilenberger and Overath ((1977) Biochim. Biophys. Acta 469, 311–325) it is shown that the exchange of lipid molecules between negatively charged vesicles consisting of total phospholipid extracts from Escherichia coli occurs by the transfer of single lipid monomers or small micelles through the water. Here a kinetic interpretation is presented in terms of a rate constant, k?, for the escape of lipid molecules from the vesicle bilayer into the water. The evaluated rate constants are k?P = (0.86 ± 0.05) · 10?5s?1 and k?E = (1.09 ± 0.13) · 10?6s?1 for phospholipid molecules with trans-Δ9-hexadecenoate and trans-Δ9-octadecenoate, respectively, as the predominant acyl chain component. The rate constants are discussed in terms of the acyl chain and polar head group composition of the lipids.  相似文献

Delocalized chemiosmotic coupling of oxidative phosphorylation requires that a single-value correlation exists between the extent of Δ\?gmH+ and the kinetic parameters of respiration and ATP synthesis. This expectation was tested experimentally in nigericin-treated plant mitochondria in single combined experiments, in which simultaneously respiration (in State 3 and in State 4) was measured polarographically, FΔψ (which under these conditions was equivalent to Δ\?gmH+) was evaluated potentiometrically from the uptake of tetraphenylphosphonium+ and the rate of phosphorylation was estimated from the transient depolarization of mitochondria during State 4-State 3-State 4 transitions. The steady-state rates of the different biochemical reactions were progressively inhibited by specific inhibitors active with different modalities on various steps of the energy-transducing process: succinate respiration was inhibited competitively with malonate or noncompetitively with antimycin A, or by limiting the rate of transport into the mitochondria of the respiratory substrate with phenylsuccinate; Δ\?gmH+ was dissipated by uncoupling with increasing concentrations of valinomycin; ADP phosphorylation was limited with oligomycin. The results indicate generally that when the rate of respiratory electron flow is decreased, a parallel inhibition of the rate of phosphorylation is also observed, while very limited effects can be detected on the extent of Δ\?gmH+. This behavior is in marked contrast to the effect of uncoupling where the decreased rate of ATP synthesis is clearly due to energy limitation. Extending previous observations in bacterial photosynthesis and in respiration by animal mitochondria and submitochondrial particles the results indicate, therefore, that respiration tightly controls the rate of ATP synthesis, with a mechanism largely independent of Δ\?gmH+. These data cannot be reconciled with a delocalized chemiosmotic coupling model.  相似文献

The effect of cold exposure caused by shearing on serum thyroid hormone (TH) concentrations in sheep kept at an ambient temperature of 8.5°C was studied. While the deep body temperature fell to the lowest level 4 h after shearing the concentration of triiodothyronine (T3) increased to a peak value at that time. Thyroxine (T4) and metabolically inactive reverse triiodothyronine (rT3) levels reached their peak value after 24 h. The T3T4 ratio reached a maximum at about 4 h and rT3T4 and rT3T3 ratios rose to maximum values about 24 h after shearing. This sequence of events suggest a biphasic response to cold—an immediate secretion of TH from the thyroid gland, followed by adaptive alteration in T3 and rT3 generation in the extrathyroidal tissues.  相似文献

In the presence of the Na+-channel blocker amiloride, the short-circuit current across the skins of bullfrog tadpoles in metamorphic stages XIX–XXIV was subjected to fluctuation analysis. The resulting power spectra contained a Lorentzian component of which the plateau value (S0) decreased while the corner frequency (fc) increased as the mucosal amiloride concentration was increased from 0.5 to 24 μM. From the linear relationship between the fc values and the amiloride concentrations it was possible to determine the binding (k′01) and unbinding (k10) constants for amiloride to its receptor on the Na+ channel. With these parameters as well as short-circuit current and S0 values, the current through the individual Na+ channels (i) was calculated (average 0.58 pA). It did not increase significantly during late metamorphosis. The density of Na+ channels (M) in the apical membrane, on the other hand, increased significantly. It would appear that the increase in short-circuit current which occurs at this time is due primarily to an increase in amiloride-blockable Na+ channels. Unexpectedly, a Lorentzian component could be fitted to power spectra in amiloride-treated skins (stages XIX–XXI) which showed no amiloride-sensitive short-circuit current. Moreover, the typical increase in fc with the amiloride concentration did not occur in these animals.  相似文献
