In studies of Trifolium repens nitrogen nutrition, the controlof nutrient solution pH using dipolar buffers, was evaluatedin tube culture under sterile conditions. Five buffers; MES,ADA, ACES, BES and MOPS with pK2s (20 °C) of 6.15, 6.60,6.90, 7.15 and 7.20 respectively, at a concentration of 2.0mol m–3, were provided to inoculated Trifolium repensgrowing in nutrient solution containing 7.13 mol m–3 nitrogenas (NH4)2SO4. Initial pH of each solution was adjusted to theappropriate buffer pK2 Two buffers, ADA and ACES completelyinhibited plant growth. The remaining buffers had little effectin limiting pH change, although plant dry matter was higherand nodule numbers lower in the presence of these buffers. MESand MOPS were supplied to nutrient solutions with and without7.13 mol m–3 (NH4)2SO4, at concentrations ranging from0–12 mol m–3. MES at 9 mol m–3 and 12 molm–3 reduced growth of plants reliant on the symbiosisfor providing nitrogen. The provision of MES to plants providedwith NH4+ significantly increased plant yield and reduced nodulenumber at all concentrations. MOPS did not affect plant yieldor nodule number. The use of dipolar buffers in legume nitrogennutrition studies is considered in terms of buffering capacity,and the side effects on plant growth and symbiotic development. Anoxia was imposed on 4–6-d-old, intact wheat seedlings,after the roots had first been exposed for 1 d to O2 concentrationsbetween 0·016 and 0·06 mol m–3. Apices ofthe main axis of the seminal roots were considered to have toleratedanoxia if elongation occurred after return from anoxia to air,hereafter called ‘retention of elongation potential’.During anoxia, elongation potential was retained longer in rootsof intact seedlings than in 0–5 mm excised root tips suppliedwith 50 mol m–3 glucose. In intact seedlings, elongation potential was retained longerat 15°C than at 25°C, and at pH 50 and 60 than at pH40. These differences between treatments were maintained inthe presence of exogenous glucose, and glucose supply prolongedthe retention of elongation potential in all anoxic treatments. Elongation potential was retained much longer at very low 02concentrations (0006 to 00l mol m–3) than under anoxia;this was established at pH 40. Anoxia inhibited the transport of sugars from the shoots and/orendosperm to the root by 79-97%, as assessed from experimentswith roots of intact plants exposed to anoxia at pH 60 and 15°C. Overall, the results demonstrate: (i) that the occurrence ofadverse effects of anoxia during waterlogging in the field mayinteract with other environmental factors and (ii) that thereare pronounced difficulties integrating data on tolerance toanoxia obtained in different laboratories. Bean plants (Phaseolus vulgaris L. cv. Fardenlosa Shiny) werelabelled with carbon-11 via their first trifoliate leaves when3-weeks-old and the transient inhibitions of translocation causedby the application of ammonium chloride solutions (10 mol m–3)to a peeled region of stem were studied. At pH 6·5 theammonium was without effect. At pH 11·0 even a briefapplication inhibited translocation for many minutes, whilelonger applications inhibited translocation for considerablylonger. Solutions of 10 mol m–3 sodium chloride were withouteffect at either pH. At pH 6·5 ammonium chloride solution contains predominantlyammonium ions (NH4+) and at pH 11·0 predominantly dissolvedammonia gas (NH3). Hence we conclude that phloem transport withinbean stems is inhibited by dissolved ammonia gas but not ammoniumions. Carbonic anhydrase (CA, EC. 4.2.1.1 [EC] ) activity in air-grown Characorallina was detected mainly in the intracellular fraction,most of which composed of chloroplasts and cytoplasmic gel,and not on the cell surface. Only minor levels of CA activity,on the basis of equivalent volumes, were detected in the cellsap and the cytoplasmic sol. The maximum rate of photosynthetic O2 evolution by air-grownChara corallina at pH 6.0 was twice that at pH 7.6, while theapparent Km for external inorganic carbon (Ci) at pH 7.6 wasabout three times that at pH 6.0. However, the apparent Km(CO2)was about three times larger at pH 6.0 than at pH 7.6. The Km(Ci)-valueat pH 7.6 increased severalfold in the presence of acetazolamide(AZA), an inhibitor of CA, but no inhibition was observed atpH 6.0. The pH-dependence may be due to differences in the permeabilityof AZA at the given pH values. Fixation of 14CO2 at 20 µMand of H14CO3 at 200 µM over the course of 5 swas very similar at pH 7.4. Addition of CA significantly suppressedthe photosynthetic 14CO2-fixation but it stimulated the H14CO3-fixation.This result indicates that free CO2 is an active species ofCi that is incorporated into the cell during photosynthesis. These results together suggest the following: (1) Free CO2 isutilized for photosynthesis, (2) CA is mainly located insidethe cell and functions to increase the affinity for CO2 in photosynthesisby facilitating the supply of CO2 from the plasmalemma to thesite of CO2-fixation. 3Present address: Biological Laboratory, The University of theAir, Wakaba 2-11, Chiba, 260 Japan. The carbon dioxide compensation point of the unicellular greenalga, Chloretla saccharophila, was determined in aqueous mediumby a gas chromatographic method. Compensation points decreasedmarkedly from 63 cm3 m–3 at an external pH of 4.0 to 3.2cm3 m–3 at pH 8.0 and were not affected by the O2 concentrationof the medium. The calculated CO2 concentration required tosupport the half-maximum photosynthetic rate of the algal cellsranged from 6.0 mmol m–3 at an external pH of 60 to 1.5mmol m–3 at pH 8.0 and these values were not affectedby O2 concentration. The Km(CO2) of nbulose-l,5-bisphosphatecarboxylase isolated from cells grown either at pH 4.0 or pH8.0 was determined to be 64 mmol m–3. These results indicatethat loss of CO2 by photorespiration does not occur in C. saccharophilacells at acid pH and the disparity between the apparent affinityfor CO2 of the intact cells and that of the carboxylase indicatesthe operation of a ‘CO2 concentrating mechanism’in this alga at acid pH. Salinity-induced Malate Accumulation in Chara   总被引:3,自引:0,他引:3  
Ion absorption by Chara corallina from solutions containingpredominantly KC1 or RbCl at up to 100 mol m–3 resultedin accumulation of salts and turgor regulation. Turgor regulationdid not occur in solutions containing Na+ or Li+salts. Duringion absorption from various salts of K+ and Rb+ vacuolar cationconcentration exceeded Cl concentration. This differencewas shown to be balanced by the synthesis and accumulation ofmalate. Vacuolar malate concentration reached 48 mol m3,with accumulation occurring at rates of up to 0.45 mol m–3h–1. Malate accumulation was inhibited by low externalpH and was dependent upon external HCO3 concentration.The synthesis of malic acid and its subsequent dissociationimposed a severe acid load on the cell. Biophysical regulationof cellular pH was achieved by a H+efflux at a rate of about40 nmol m–2 s–1from the cell. The results presentedargue against cytoplasmic Cl, HCO3 or pH regulatingmalate accumulation in Chara and it is suggested that malatetransport across the tonoplast may regulate malate accumulation. Effects of removal of external Ca2+ on the cytoplasmic pH (pHc)of Chara corallina have been measured with the weak acid 5,5-dimethyl-oxazolidine-2,4-dione(DMO) as a function of external pH (pH0) and of the externalconcentration of K+. Removal of Ca2+ always decreased pHc whenpH0 was below about 6.0; the decrease was about 0.2–0.4units at pH0 5.0, increasing to about 0.5 units at pH0 4.3.When pH0 was 6.0 or higher the removal of Ca2+ had little orno effect on pHc. This situation was not altered by changingthe concentration of K+, though in some experiments at pH0 5.0–5.2there was a slight decrease in pH0 (about 0.2 units) when K+was increased from 0.2 to 2.0 mol m–3, an effect apparentlyreversed when K+ was higher (5.0 or 10.0 mol m–3). Theresults suggest that H+ transport continues in the absence ofexternal Ca2+, despite previous suggestions to the contrary,and that the H+ pump does not necessarily run near thermodynamicequilibrium with its chemical driving reaction. They indicate,rather, that the H+ pump is under kinetic control and providefurther evidence for the inadequacy of present models for theoperation of the H+ pump in charophyte cells, especially inrelation to its proposed role in regulating pHc. DNA polymerases were purified several hundred-fold from the10 000 x g soluble (polymerase I) and particulate (polymeraseIII) fractions prepared from virus PBCV-1 infected ChlorellaNC64A extracts. Both DNA polymerases exhibited optimal activitywith activated calf thymus DNA at pH 8.5. DNA polymerase I required3.0 mol m–3 MgSO4 and 150 to 250 mol m–3 KCl foroptimum activity whereas, DNA polymerase III required 2.0 molm–3 MgSO4 and 150 mol m–3 KCl. Both enzymes wereinhibited by pyrophosphate, actinomycin D, ethidium bromide,dideoxythymidine triphosphate, and N-ethylmaleimide but wererelatively insensitive to aphidicolin. DNA polymerase I differedfrom DNA polymerase III in its response to cations (particularlyNH4Cl), elution from a DEAE cellulose column, and molecularweight. The effects of excess salinity and oxygen deficiency on growthand solute relations in Zea mays L. cv. Pioneer 3906 were examinedin greenhouse experiments. The roots of plants 14 d old growingin nutrient solution containing additions of NaCl in the range1.0–200 mol m–3 were either exposed to a severedeficiency of O2 by bubbling with nitrogen gas (N2 treatment),or maintained with a supply of air (controls), for a periodof 1–7 d. The threshold NaCl concentration resulting inappreciable inhibition of leaf extension, and shoot f. wt gainin controls was between 10 and 25 mol m–3. At 25 mol m–3NaCl the ratio of Na+/K+ transported to shoots was about 20times greater than in plants in 1.0 mol m–3 NaCl. Theeffect of addition of NaCl to the nutrient solution was to enhanceNa+ movement but simultaneously depress the rate of K+ transportto shoots (per g f. wt roots). Interactions between NaCl levels and aeration treatment wereshown by analyses of variance to be statistically significantfor leaf extension, shoot and root f. wt gains, Na+ and K+ concentrationsin shoots and roots. When roots were N2-treated, shoot and rootgrowth were depressed, the effect of aeration treatment beinggreatest at NaCl concentrations of 50 mol m–3 or less.Additionally, N2-treatment greatly accelerated Na- transportto shoots while depressing K+ transport still further, so thatat 10 mol m–3 NaCl the ratio Na+/K+ acquired by the shootswas 230 times greater than in controls. Over the concentrationrange 1.0 to 50 mol m–3 NaCl, the ratio Na+/K+ transportedto shoots by anoxic roots increased by a factor of 860. Mechanisms controlling changes in solute flux to the shoot,and the significance in relation to plant tolerance of excesssalts or oxygen deficiency are discussed. Seedlings of cotton (Gossypium hirsutum L. cv. Acala SJ-2) weregrown in modified Hoagland nutrient solution with various combinationsof NaCl and CaCl2. Marking experiments and numerical analysiswere conducted to characterize the spatial and temporal patternsof cotton root growth at varied Na/Ca ratios. At 1 mol m–3Ca, 150 mol m–3 NaCl reduced overall root elongation rateto 60% of the control, while increasing Ca to 10 mol m–3at the same NaCl concentration restored the elongation rateto 80% of the control. Analysis of the spatial distributionof elongation revealed that the presence of 150 mol m–3NaCl in the medium shortened the growth zone by about 2 mm fromthe approximate 10 mm in the control and also reduced the relativeelemental elongation rate (i.e. the longitudinal strain rate,defined as the derivatives of displacement velocity of a cellularparticle with respect to position on root axis). Supply of 10mol m–3 Ca at the high salt condition restored partiallythe relative elemental elongation rate, but not the length ofthe growth zone. Compared to the control, the growth trajectoriesshowed that at 1 mol m–3 CaCl2 it took more time for acellular particle to move through the growth zone at 150 molm–3 NaCl, while at 10 mol m–3 CaCl it took lesstime and there was no difference between the NaCl treatments Key words: Gossypium hirsutum, salinity stress, root growth kinematics  相似文献

Characteristics of the vacuolar-type (V-type) H+-ATPase fromguard cell protoplasts of Commelina communis L. were investigatedusing a linked enzyme assay and nitrate inhibition as a diagnosticindicator of the enzyme activity. ATPase activity was completelyinhibited by about 50 mol m–3 nitrate and activity wasoptimal near pH 8.0. The temperature optimum for activity wasabout 37 C and an Arrhenius plot indicated changes in activationenergy for the ATPase at 15C and possibly at about 30 C. Theenzyme was stimulated by Cl while Ca2+ inhibited activity(l50 = 1.5 mol m–3). The apparent Km (MgATP) was 0.62mol m–3. Incubation of guard cell protoplasts for up to 5 h in 50 µMabscisic acid (ABA) or 25µM fusicoccin (FC) did not affectsubsequent ATPase activity. In vitro assays with FC or ABA alsodid not affect enzyme activity. Activity was not affected bylight or potassium ferricyanide, two factors which are knownto influence stomatal activity. Beticoline was a potent inhibitorof activity (l50 = 50 µM) while DCCD was less effective(l50 = 90µM). On chlorophyll, protein and protoplast bases, V-type ATPaseactivity was greater in guard cell protoplasts than mesophyllcell protoplasts by 66, 13.9 and 1.9, respectively. A procedure is described for the purification of phosphoenolpyruvatecarboxylase (EC 4.1.1.31 [EC] ) and NADP-dependent malic enzyme (EC1.1.1.40 [EC] ) from sugar cane leaves. Each enzyme was purified tohomogeneity as judged by sodium dodecyl sulfate-polyacrylamidegel electro-phoresis, with about 30% yield. Phosphoenolpyruvatecarboxylase was purified 54-fold. A molecular weight of 400,000and a homotetrameric structure were determined for the nativeenzyme. The purified carboxylase had a specific activity of20.0 {diaeresis}mol (mg protein)–1 min–1, and wasactivated by glucose-6-phosphate and inhibited by L-malate.Km values at pH 8.0 for phosphoenolpyruvate and bicarbonatewere 0.25 and O.l0 mM, respectively. NADP-malic enzyme, 356-foldpurified, exhibited a specific activity of 71.2 {diaeresis}mol(mg protein)–1 min–1 and was characterized as ahomotetramer with native molecular weight of 250,000. Purifiedmalic enzyme showed an absolute specificity for NADP+ and requireda divalent metal ion for activity. Km values of 0.33 and 0.008mM for L-malate and NADP+, respectively, were determined. Thisenzyme was inhibited by several organic acids, including ketoand amino acids; while succinate and citrate increased the enzymeactivity when assayed with 10{diaeresis}M L-malate. We have devised conditions which produced isolated protoplastsof non-spherical shape and which, therefore, affected the mechanismsthat control the exchange of membrane material between the plasmamembrane and an intracellular membrane reservoir. Non-sphericalprotoplasts of Avena sativa were obtained if protoplasts weretreated with hypertonic shock in the presence of 1.0 mol m–3LaCl3 at pH 8.3. This indicated that their ability to removeplasma membrane material via endocytotic vesiculation was suppressed.Non-spherical protoplasts were obtained under isotonic conditionsif protoplasts were incubated with 1.0 mol m-3 LaCl3 at pH 8.3and the proton carrier CCCP (12 mmol m–3) was added. Thenon-spherical protoplasts had intact membranes as judged bystaining with fluorescein diacetate. The loss of the sphericalshape was reversible. On addition of EDTA protoplasts resphericulatedimmediately. Incubation in isotonic solution at pH 8.3 containingeither only 1.0 mol m–3 LaCl3 or only CCCP did not influencethe protoplast shape. We conclude that the membrane hyperpolarizationinduced by CCCP at high pH acted to stimulate the incorporationof membrane material into the plasma membrane and, subsequently,produced nonspherical protoplasts if the removal of membranematerial was simultaneously suppressed. This demonstrates thatmembrane incorporation and removal are two largely independentprocesses.  相似文献

Barley (Hordeum vulgare L. cv. Digger) was grown for 22 d inenclosed chambers with a CO2 enrichment of 35, 155, 400 or 675µmol CO2 mol1. CO2 enrichment increased photosyntheticcapacity in the plants grown at either of the two highest levelsof pCO2. A CO2 enrichment of 675µmol CO2 caused a significantincrement of shoot dry weight, whereas no changes were observedin fresh weight, chlorophyll or protein levels. At a light intensityof 860µmol m–2s–1 CO2 enrichment caused photosyntheticcapacity to increase by 250%, whereas no effect was observedat 80 µmol m–2 s–1. Over time, photosynthesisdecreased by 70% independent of CO2. A time-dependent increasein the level of extractable fructose was observed whereas totalextractable carbohydrate only changed slightly. The effects of a range of applied nitrate (NO3) concentrations(0–20 mol m3) on germination and emergence percentageof Triticum aestivum L. cv. Otane were examined at 30, 60, 90and 120 mm sowing depths. Germination percentage was not affectedby either sowing depth or applied NO3 concentration whereasemergence percentage decreased with increased sowing depth regardlessof applied NO3 concentration. Nitrate did not affectemergence percentage at 30 mm sowing depth, but at 60 to 120mm depth, emergence percentage decreased sharply with an increasedapplied NO3 concentration of 0 to 1·0 mol m–3then decreased only slightly with further increases in appliedNO3 of about 5·0 mol m–3. Root and shoot growth, NO3 accumulation and nitrate reductaseactivity (NRA) of plants supplied with 0, 1·0 and 1·0mol m–3 NO3 at a sowing depth of 60 mm were measuredprior to emergence. The coleoptile of all seedlings opened withinthe substrate. Prior to emergence from the substrate, shootextension growth was unaffected by additional NO3 butshoot fr. wt. and dry wt. were both greater at 1·0 and1·0 mol m–3 NO3 than with zero NO3.Root dry wt. was unaffected by NO3. Nitrate concentrationand NRA in root and shoot were always low without NO3.At 1·0 and 10 mol m3 NO3, NO3 accumulatedin the root and shoot to concentrations substantially greaterthan that applied and caused the induction of NRA. Regardlessof the applied NO3 concentration, seedlings which failedto emerge still had substantial seed reserves one month afterplanting. Coleoptile length was substantially less for seedlingswhich did not emerge than for seedlings which emerged, but wasnot affected by NO3. It is proposed that (a) decreasedemergence percentage with increased sowing depth was due tothe emergence of leaf I from the coleoptile within the substrateand (b) decreased emergence percentage with additional NO3was due to the increased expansion of leaf 1 within the substrateresulting in greater folding and damage of the leaf. The mechanism of uptake of inorganic carbon and its accumulationhas been studied in the freshwater diatom Navicula pelliculosa.No external carbonic anhydrase could be detected, although itwas detected in cell extracts. The rate of photosynthetic O2evolution, in media in the range pH 7.5–8.5, exceededthe calculated rate of CO2 supply 2- to 5-fold, indicating thatHCO3 was taken up by the cells. At an external pH of7.5, the internal pH, measured by 14C-dimethyloxazolidine-2,4-dione distribution between the cells and the medium, was pH7.6 in the light and pH 7.4 in the dark. Accumulation of inorganiccarbon was determined by the silicone oil centrifugation methodand inorganic carbon pools of 23.5 mol m–3 were found,a concentration 21.6-fold that in the external medium. The resultsindicate an active accumulation of inorganic carbon againstpH and concentration gradients in this diatom, probably by activeHCO3 uptake. Potassium-Ammonium Uptake Interactions in Tobacco Seedlings   总被引:6,自引:0,他引:6  
Short-term (< 12 h) uptake experiments were conducted with6–7-week-old tobacco (Nicotiana tabacum L. cv. Ky 14)seedlings to determine absorption interactions between K+ andNH4+. At equal solution concentrations (0.5 mol m–3) netK+ uptake was inhibited 30–35% by NH4+ and NH4+ uptakewas decreased 9–24%. Removal of NH4+ resulted in completerecovery in K+ uptake rate, but NH4+ uptake rate did not recoverwhen K+ was removed. In both cases, inhibition of the uptakerate of one cation saturated as the concentration of the othercation was increased up to 0.5 mol m–3. The relative effectof K+-NH4+ interactions was not altered when Cl- was replacedwith SO42–, but the magnitudes of the uptake rates wereless in the absence of Cl-. The Vmax for NH4+ uptake was reducedfrom 128 to 105 µmol g–1 dry wt. h–1 in thepresence of 0.5 mol m–3 K+ and the Km for NH4+ doubledfrom 12 to 27 mmol m–3 in the presence of K+. The resultsof these K+-NH4+ experiments are interpreted as mixed-noncompetitiveinteractions. The acidophilic alga Dunaliella acidophila exhibits optimalgrowth at pH 1. We have investigated the regulation of phosphateuptake by this alga using tracer techniques and by performingintracellular phosphate measurements under different growthconditions including phosphate limitation. In batch culturewith 2·2 mol m–3 phosphate in the medium the uptakeof phosphate at micromolar phosphate concentrations followeda linear time dependence in the range of minutes and rates werein the range of 1 µmol phosphate mg–1 chl h–1,only. However, under discontinuous phosphate-limited growthconditions, tracer influx revealed a biphasic pattern at micromolarphosphate concentrations: An initial burst phase resulted ina 104-fold internal phosphate accumulation and levelled offafter about 10 s. A double reciprocal plot of the initial influxrates obtained for phosphate-limited and unlimited algae exhibitedMichaelis-Menten kinetics. Phosphate limitation caused a significantactivation of the maximum velocity of uptake, yielding Vmaxup to 1 mmol mg–1 chl h–1 as compared to valuesin the order of 50 µmol phosphate mg–1 chl h–1for the second phase (this magnitude is also representativefor non-limited batch cultures). Concomitantly the Michaelisconstant was altered from 4 mmol m–3 to 0·7 mmolm–3. The rapid uptake of phosphate was inhibited by arsenateand FCCP and was not stimulated by Na+. The pH dependence oftracer accumulation and measurements of the intracellular phosphatepool under different growth conditions indicate that at lowpH and low external phosphate concentrations the high protongradient present under these conditions is utilized for a H3PO4uptake or a H+/H2PO4 cotransport. However, when the externalphosphate concentration was increased to levels sufficientlyhigh for transport to be driven by the positive membrane potential(10 mol m–3 phosphate), the pH dependence of phosphateuptake was more complex, but could be explained by the uptakeof H3PO4 or a H+/H2PO4-cotransport at low pH and a differenttype H2PO4-transport (with unknown type of ion coupling)at high pH-values. It is suggested that this flexible couplingof phosphate transport is of essential importance for the acidresistance of Dunaliella acidophila. Guard cell protoplasts (GCP) were prepared from leaves of Commelinacommunis L. and phosphoenolpyruvate carboxylase (PEPc) activityrecorded after injection of the protoplasts directly into theassay medium. The GCP were lysed immediately by the presenceof Triton X-100 and a lowered osmotic concentration in the assaycuvette enabling PEPc activity to be measured with ‘nascent’enzyme. There was no light activation of the enzyme with KmPEP (about 3.7 mol m–3) and Vmax being similar in light-ordark-treated protoplasts. Illumination of the GCP in the presenceof CO2-free air and KCI, a treatment which is known to swellGCP, did not change the kinetics. PEPc activity at saturating PEP was very sensitive to malateinhibition, 20 mmol m–3 (the I50 value) inhibiting activityby about 50%. Inhibition was similar in light- or dark-treatedprotoplasts. Malate inhibition was, however, much less (I50= 500 mmol m–3) if the enzyme source was a protoplastextract kept in the absence of glycerol. Inclusion of 20% glycerolin the extraction medium maintained the enzyme in the malate-sensitiveform as occurred in the in vivo assays. The high apparent KmPEP and the high sensitivity to malate inhibition of GCP PEPcare features unlike those observed with PEPc from leaf tissuesof C4 and CAM plants and from GCP extracts. PEPc activity increased slightly in the presence of KCI in theassay medium up to about 10 mol m–3 and thereafter activityslowly declined as KCI concentrations increased further.
