The objective of the current study was to determine the effects of hormonal treatments on ovarian follicular development and oocyte quality in anestrous ewes. Multiparous crossbred (RambouilletxTarghee) ewes were given melatonin implants (MEL) and/or controlled internal drug release (CIDR) devices in conjunction with follicle stimulating hormone (FSH) during anestrus (March-May). In Experiment 1, ewes (n=25) were assigned randomly to four groups (n=4-7/group) in a 2x2 factorial arrangement [+/-MEL and +/-CIDR], resulting in Control (no treatment), CIDR, MEL, and MEL/CIDR groups, respectively. Ewes received an implant containing 18 mg of melatonin (Melovine) on Day 42 and/or a CIDR from Days 7 to 2 (Day 0: oocyte collection). In Experiment 2, ewes (n=12) were assigned randomly to two groups (n=6/group; 1CIDR or 2CIDR) and received the same type of melatonin implant on Day 60. All ewes received a CIDR device from Days -22 to -17 and 2CIDR ewes received an additional CIDR device from Days -10 to -2. In both experiments, ewes were given FSH im twice daily (morning and evening) on Days -2 and -1 (Day -2: 5 units/injection; Day -1: 4 units/injection). On the morning of Day 0, ovaries were removed, follicles>or=1 mm were counted, and oocytes were collected. Thereafter oocytes were matured and fertilized in vitro. In Experiment 1, the number of visible follicles and the rates of oocyte recovery and in vitro maturation were similar (P>0.10) for Control, CIDR, MEL and MEL/CIDR (overall 29.7+/-2.9%, 89.9+/-7.1% and 95.0+/-2.0%, respectively). The rates of in vitro fertilization (IVF) were lower (P<0.01) for CIDR and MEL/CIDR than for Control and MEL groups (10.3% and 10.1% versus 20.0% and 18.5%, respectively). In Experiment 2, the number of visible follicles, and the rates of oocyte recovery and in vitro maturation were similar (P>0.10) for 1CIDR and 2CIDR groups (overall 27.3+/-3.2%, 92.1+/-2.7% and 90.2+/-1.9%, respectively). However, the rates of IVF were lower (P<0.01) for 2CIDR than 1CIDR group (30.2% versus 58.0%, respectively). In summary, when treatment with P4 commenced only 2 d before oocyte collection, rates of IVF were reduced in both experiments. Therefore, progestin treatment protocols used in ovine IVF programs should be carefully designed to minimize adverse effects on fertilization rates. In addition, melatonin treatment did not affect follicular development and oocyte quality for anestrous ewes.  相似文献

Experiments were conducted to determine the effects of porcine relaxin (pRLX) on cervical dilation and conception rates in postpartum ewes. In Experiment 1, ewes received medroxyprogesterone acetate (MAP) sponge on day 16 (day 0 = lambing) and 750 IU pregnant mare serum gonadotropin (PMSG) at sponge removal on day 30. Control ewes received saline and relaxin-treated (RLX) ewes received 0.5 mg pRLX (>/= 3000 U/mg) i.m. at 24 h and 1.0 mg pRLX at 36 h after PMSG. All ewes were inseminated (Al) at 55 h after PMSG with 0.4 ml fresh semen. The proportion of RLX treated ewes (6 6 ) in which the cervix was penetrated was greater (P < 0.05) than in Control ewes (0 5 ). However, ova recovery rate was lower (P < 0.05) for RLX ewes (1 6 ) than for control ewes (5 5 ). In Experiment 2, ewes between Days 90 to 120 post partum received MAP sponges for a period of 8 d and 750 IU PMSG at sponge removal. Control ewes (n = 9) received saline; RLX-1 ewes (n = 8) received 0.5 mg pRLX at 24 h and an additional 0.5 mg pRLX at 36 h after PMSG; and RLX-1.5 ewes (n = 9) received 0.5 mg pRLX at 24 h and an additional 1.0 mg pRLX at 36 h after PMSG. Ewes were mated to rams at estrus, and cervical dilation was checked at 55 h after PMSG. The cervix could not be penetrated in any of the ewes. Conception rates on Day 26 were 66, 56 and 63% for control, RLX-1 and RLX-1.5 groups, respectively. These results demonstrate that the effect of relaxin on cervical dilation and conception rate is dependent upon the postpartum stage of the ewes.  相似文献

Follicular dynamics and ovulation were compared in 3 groups of anestrous ewes: those treated with medroxyprogesterone acetate (MAP) sponges for 12 d, then with 750 IU PMSG at the time of sponge removal (P4 + PMSG, n = 6), or PMSG alone (n = 6) and untreated controls (n = 6). Waves of follicular activity were observed in all the animals. In the P4 + PMSG treatment group, MAP priming permitted more ovulatory follicles (P < 0.001) to be recruited without changing follicle growth rate; MAP priming also delayed the time of ovulation (P < 0.001) and the time of the LH surge (P < 0.01), which allowed for an increase in the size of ovulatory follicles (P < 0.05). Ovulation also resulted in normal luteal function after P4 + PMSG (P < 0.01) but not after PMSG alone, since premature luteal regression occurred in 80% of the cases and was related to the presence of follicles > 4 mm when P4 levels were < 1 ng/mL on the day following ovulation. The results showed that MAP priming increased the ovulation rate by increasing the number of follicles that responded to PMSG.  相似文献

A series of studies was designed to identify methods of improving out-of-season breeding success in ewes. In Experiment 1, 190 mature ewes were assigned to receive in April, either: (A) a control ration of 0.3 kg corn twice daily for 8 d before ram introduction (control ewes n=49), (B) the control ration containing 0.125 mg of melengestrol acetate (MGA) in 0.3 kg corn (MGA8a ewes n=46), (C) the control ration or 7.5 d followed by 1 feeding of 0.5 mg MGA in 0.3 kg corn (MGA1 ewes n=48), or (D) the control ration plus a 20 mg i.m. injection of progesterone on D 8 (P ewes n=47). Ewes were exposed to rams for 21 d. A greater percentage of MGA8a and P ewes lambed than did control ewes (P < 0.04). The lambing rate was greatest among MGA8a (P < 0.02 vs. control), intermediate among P ewes (P < 0.19 vs. control) and least among MGA1 and control ewes (P > 0.79). In Experiment 2, 70 mature ewes were assigned to receive in June, either: (A) a control ration of 0.3 kg of corn twice daily for 8 d before to ram introduction (control ewes n=25), (B) the control ration containing 0.125 mg of MGA per 0.3 kg corn (MGA8b ewes n=21), or (C) the control ration and simultaneous treatment of ewes with a progesterone-containing controlled internal-drug releasing device (CIDR ewes n=24). Ewes were exposed to rams for 21 d. Both CIDR and MGA8b ewes exhibited estrus earlier than did control ewes (P < 0.01). The CIDR ewes exhibited estrus earlier than did the MGA8b ewes (P < 0.05). A greater percentage of ewes treated with CIDR or MGA8b lambed than did control ewes (P < 0.01), with more CIDR ewes lambing than MGA8b ewes (P < 0.01). The lambing rate was greater in CIDR ewes than in control ewes (P < 0.04). These data provide evidence that several options exist to improve pregnancy success in ewes bred out of season and that success varies with method of treatment.  相似文献

Hair sheep ewes (St. Croix White and Barbados Blackbelly) were used to evaluate 3 methods of estrus synchronization for use with transcervical artificial insemination (TAI). To synchronize estrus, ewes (n = 18) were treated with PGF2alpha (15 mg, im) 10 d apart, with controlled internal drug release (CIDR) devices containing 300 mg progesterone for 12 d (n = 18), or with intravaginal sponges containing 500 mg progesterone for 12 d (n = 18). On the day of the second PGF2alpha injection or at CIDR or sponge removal, sterile rams were placed with the ewes. Jugular blood samples were collected from the ewes at 6-h intervals until the time of ovulation, and daily for 16 d after estrus (Day 0). Plasma was harvested and stored at -20 degrees C until LH, and progesterone concentrations were determined by RIA. There was no difference (P>0.10) in time to estrus among the CIDR-, PGF2alpha- or sponge-treated ewes. All of the ewes in the CIDR group and 94.4% of the sponge treated ewes exhibited estrus by 36 h after ram introduction, while only 72.2% of PGF2alpha-treated ewes showed signs of estrus by this time (P<0.06). The time from ram introduction to ovulation was not different (P>0.10) among the CIDR-, PGF2alpha- or sponge-treated ewes. The time to the preovulatory LH surge was similar (P>0.10) among CIDR, PGF2alpha and sponge treated ewes. Progesterone levels through Day 16 after the synchronized estrus were not different (P>0.10) among treatment groups. Hair sheep ewes (n = 23) were synchronized using PGF2alpha and bred by TAI using frozen-thawed semen 48 h after the second injection. The conception rate to TAI was 2/23 (8.7%) and produced 3 ram lambs. In a subsequent trial, 17 ewes were synchronized with CIDR devices and bred by TAI using frozen-thawed semen 48 h after CIDR removal, resulting in a conception rate of 52.9% (9/17). It is possible to synchronize estrus in hair sheep using either CIDRs, sponges or PGF2alpha. Even though there were no significant differences in the timing of ovulation or the LH surge among the treatment groups, a higher conception rate was achieved in ewes synchronized with CIDR devices during the second trial. This may reflect an increase in the skill level of the TAI technician.  相似文献

Administration of FSH increases the number of developing follicles, and affects oocyte health and cleavage rate. To determine the optimal level of FSH treatment, studies were conducted during the normal breeding season and seasonal anestrus. In Experiment 1, ewes were implanted with SyncroMate-B (SMB; norgestomet) for 14 days during the breeding season. Beginning on day 12 or 13 after SMB implantation, ewes were treated with saline (control; n=10), or treated with FSH for two days (2D; n=9) or three days (3D; n=10). In Experiment 2, conducted during seasonal anestrus, ewes were implanted with SMB for 14 days (n=23) or were not implanted (n=26). The SMB-implanted and nonimplanted ewes were assigned to one of three treatments as in Experiment 1: control (n=13), 2D (n=21) or 3D (n=15). In Experiments 1 and 2, ewes were laparotomized to count the number of follicles < or = 3 mm and > 3 mm and to retrieve oocytes. Healthy oocytes from each treatment were used for IVF. In Experiment 3, ewes (n=6) were implanted twice with SMB for 14 days during seasonal anestrus. Ewes were injected with FSH for 2 days, and the oocytes were collected and fertilized as in Experiments 1 and 2. In Experiment 1, FSH-treatment increased (P < 0.05) the number of follicles > 3 mm, the number of oocytes retrieved from follicles < or = 3 mm and > 3 mm, the proportion of healthy oocytes, and the number of oocytes used for IVF. Oocytes from control and 2D ewes had greater (P < 0.01) cleavage rates than 3D ewes (68% and 71% vs. 42%). In Experiment 2, implanted and nonimplanted ewes had similar (P > 0.05) numbers of follicles, total oocytes, and healthy oocytes; therefore, data were combined. The FSH treatment increased (P < 0.01) the number of follicles > 3 mm, and the number of oocytes recovered from follicles > 3 mm. The recovery rate of oocytes and the percentage of healthy oocytes were similar for control and FSH-treated ewes. The cleavage rate in Experiment 2 ranged from 4 to 16%. In Experiment 3, the cleavage rate for ewes treated twice with SMB was 27% which tended to be greater (P < 0.07) than for the 2D ewes that received one SMB implant in Experiment 2. These data indicate that FSH increased the number of developing follicles and the number of healthy oocytes retrieved from ewes during the breeding season and seasonal anestrus. However, cleavage rates during seasonal anestrus were lower than during the normal breeding season in both FSH-treated and control ewes. Treatment of ewes for 2 days with FSH resulted in a greater cleavage rate than treatment of ewes for 3 days.  相似文献

The use of either 1 or 3 controlled internal drug release (CIDR) devices for progesterone priming in ewes (n=11) superovulated with 1500 IU pregnant mare serum gonadotrophin (PMSG) at 28 hours prior to CIDR device withdrawal was investigated in relation to the stages of development and viability of the ova produced. Progesterone levels in the ewes (n=6) treated with 3 CIDR devices were significantly higher (P<0.01) during the 11 days of insertion than in those (n=5) treated with 1 CIDR device (7.3 vs 3.3 ng/ml) over the same period. However, following superovulation, the mean (+/-SEM) ovulation rates were similar for both groups (8.2 +/- 1.7 vs 10.2 +/- 1.5). The number of ova (M+/-SEM) recovered by laparoscopy 5 days after insemination was 4.2 +/- 1.0 for ewes treated with 3 CIDR devices and 7.0 +/- 1.1 for those treated with 1 CIDR device (P<0.10). The respective ovum recovery rates (M+/-SEM) were 55+/-9.8 and 74+/-13.2%. There was no effect of progesterone concentration in the priming phase on either the stages of development of the recovered ova or on their ability to develop during in vitro culture. It was concluded, therefore, that progesterone concentrations within the range 3.3 +/- 0.1 to 7.3 +/- 0.3 ng/ml during the priming phase and 2.4 +/- 0.3 to 6.5 +/- 0.2 ng/ml at the time of PMSG administration did not affect the ovulation rate or the viability of ova recovered from superovulated ewes.  相似文献

The objective of this work was to investigate the effect of progesterone (P) and gonadotropin-releasing hormone (GnRH) treatment on estrogen receptor (ER) and P receptor (PR) concentrations in the pituitary gland and uterus of anestrous ewes. Ewes were either not treated (group C, n = 4); were treated with 0.33 g P-controlled internal drug release (P-CIDR) for 10 days (group P, n = 4), with GnRH, 6.7 ng i.v. injections every 2 h for 18 h followed by a 4 microg bolus administration of Receptal at 20 h (group GnRH, n = 4), or with a combination of the P and GnRH treatment (group P + GnRH, n = 3). Ewes were humanely killed either at the beginning of the experiment (group C), when the CIDR was removed (group P), or 24 h after the GnRH bolus treatment (groups GnRH and P + GnRH). Progesterone treatment increased serum P concentrations, indicating that the treatment was effective. All GnRH treated ewes had similar luteinizing hormone (LH) surges, which lasted 8 h. At slaughter, estradiol (E2) concentrations in the GnRH group were higher than in groups C, P, and P + GnRH. Treatment with GnRH increased more than 10-fold the content of ER and PR in the pituitary gland without altering steroid receptor concentrations in the uterus. When GnRH was combined with P the uterine receptor contents were higher than with P treatment alone. The treatment with P decreased ER and PR content in the uterus, but had no effect on the pituitary gland. The results show that regulation by P and GnRH of ER and PR content in anestrous ewes is tissue-specific.  相似文献

Mature nonlactating Altamurana ewes (n = 168) were synchronized in the seasonal anestrus period with FGA-impregnated intravaginal pessaries for 12 d. In Experiment 1, 48 ewes were divided into a 3 x 4 factorial design for anti-PMSG monoclonal antibody (AP) bioassay test. Concomitant injections of PMSG (1000, 1500, 2000 IU) and AP (0, 1, 2, 3 microl/IU PMSG) were given, and ovarian response was evaluated by laparoscopy. In Experiment 2, 120 ewes were divided into 8 experimental groups (n = 15 per group). The ewes treated with 1000 or 1500 IU PMSG at -24 h from sponge removal were given AP intravenously at 50 h after pessary withdrawal, 12 or 24 h after the onset of estrus, while the controls did not receive AP. Blood samples were collected from ewes (n = 6) treated with 1500 IU PMSG with or without anti-PMSG. Ovarian response and embryo production were evaluated on Day 7 after sponge removal upon laparotomy. It was found that 1 microl AP was effective in neutralizing 1 IU PMSG. No significant differences in serum concentrations of progesterone were observed among the groups of superovulated ewes. Estradiol-17 beta levels were reduced following AP treatment 12 h after the onset of estrus. At a lower dosage of superovulatory treatment (1000 IU PMSG), AP injected at 12 or 24 h after the onset of estrus significantly lowered large follicles (P < 0.01) and increased the rate of ovulation (P < 0.05). Moreover, embryo production showed a more than two-fold increase (P < 0.01) of viable embryos following AP injection at 12 or 24 h after the onset of estrus (3.2 to 3.3 vs 1.3, with vs without anti-PMSG). It is concluded that superovulatory treatment with 1000 IU PMSG plus AP administered at a fixed time after the onset of estrus may improve ovarian response and the yield of viable embryos in ewes.  相似文献

Experiments were conducted to evaluate a controlled internal drug-release (CIDR) dispenser containing progesterone to control the estrous cycle of ewes. After insertion of CIDR dispensers into the vaginae of ovariectomized ewes (Experiment 1; n = 11), the mean plasma progesterone rose from 0.74 ± 0.2 ng/ml to a peak of 5.5 ± 1.0 ng/ml within 2 h and then declined to 3.0 ± 0.5 ng/ml by 48 h. This was followed by a more gradual decline to 1.7 ± 0.3 ng/ml at the time of removal 12 or 14 d later. Following removal, the levels declined to baseline within 4 h. In Experiment 2, a 12- or 14-d treatment with CIDR dispensers was initiated in ewes 2, 9 and 16 d after synchronization of the estrous cycle with fluorogestone acetate (FGA)-impregnated intravaginal sponges. An intramuscular (i.m.) injection of 500 IU pregnant mare serum gonadotropin (PMSG) was given at the time of removal of the FGA sponge or CIDR dispenser. Based on plasma progesterone profiles, CIDR dispensers inserted 9 or 16 d after FGA sponge removal delayed the onset of a new estrous cycle until they were withdrawn. Following withdrawal, ovulation was effectively synchronized in all treatment groups and accompanied by development of functionally active corpora lutea with a normal lifespan. In Experiment 3, comparison of the mating response of ewes after treatment with CIDR dispensers (n = 192) or FGA sponges (n = 194) showed that 92% and 91% of the treated ewes, respectively, were marked by the ram within 72 h. Fertility and litter size of ewes bred at the synchronized and followup estrus were similar for both treatments. These results indicate that treatment of ewes with CIDR dispensers containing progesterone maintains plasma levels of progesterone within the range found during the normal estrous cycle. The CIDR dispenser is effective in synchronizing the estrous cycle of adult ewes and offers a promising alternative to the FGA-impregnated intravaginal sponge.  相似文献

An experiment was designed to evaluate a) the effect of a progesterone-estradiol combined treatment on ovarian follicular dynamics in postpartum beef cows, and b) ovulation and the subsequent luteal activity after short-term calf removal and GnRH agonist treatment. Multiparous Angus cows (25 to 40 d after calving) were assigned to the following treatments: untreated (Control, n = 9); short term calf removal (CR, n = 8); progesterone (CIDR, n = 9) and progesterone plus estradiol-17 beta (CIDR + E-17 beta, n = 9). Progesterone treatment (CIDR) lasted 8 d and the day of device insertion was considered as Day 0. Cows in the CIDR + E-17 beta group also received an i.m. injection of 5 mg of E-17 beta on Day 1. On Day 8, calves were removed for 48 h (CR, CIDR and CIDR + E-17 beta groups) and 6 h before the end of calf removal these cows also received an i.m. injection of 8 micrograms of Busereline (GnRH). Anestrus was confirmed in all cows by the absence of luteal tissue and progesterone concentrations below 1 ng ml-1 at the beginning of the experiment. Although mean (+/- SEM) interval from the beginning of the experiment (Day 0) to wave emergence did not differ (P > 0.05) among treatment groups (Control, 1.9 +/- 1.0, range -2 to 7 d; CR, 3.9 +/- 0.7, range 0 to 6 d; CIDR, 2.8 +/- 0.5, range 0 to 4 d and CIDR + E-17 beta, 4.1 +/- 0.2, range 3 to 5), the variability was less (P < 0.05) in the CIDR + E-17 beta group. The proportion of cows ovulating 24 to 48 h after GnRH administration tended (P = 0.08) to be higher in cows from CIDR + E-17 beta group (8/9) than in those of CR (5/8) or CIDR (6/9) groups, respectively and was associated with a higher proportion (P < 0.05) of CIDR + E-17 beta treated cows (9/9) that had a dominant follicle in the growing/early static phase at the time of GnRH treatment compared to the other GnRH treated groups (5/8, and 4/9 for CR and CIDR groups, respectively). Two CR cows ovulated 0-24 h after GnRH and only one Control cow ovulated the day before the time of GnRH administration. Cows pretreated with progesterone had longer (P < 0.05) luteal lifespan (CIDR, 14.5 +/- 0.7, CIDR + E-17 beta, 13.9 +/- 0.6 d) than those not treated with CIDR (Control, 5, CR, 4.0 +/- 0.4). We conclude that progesterone plus estradiol treatment results in tightly synchronized wave emergence and high GnRH-induced ovulation rate with normal luteal activity in postpartum beef cattle.  相似文献

Factors affecting superovulation in heifers treated with PMSG   总被引:1,自引:0,他引:1  
In this study we determined 1) if the immunoneutralization of PMSG affected the ovulatory response, the number of large follicles and embryo yield compared with that of PMSG alone or pFSH, and 2) whether the stage of the estrous cycle at which PMSG was injected affected the ovulatory response and yield of embryos in superovulated heifers. Estrus was synchronized in 99 (Experiment 1) and 71 (Experiment 2) heifers using prostaglandin F2alpha (PG) analogue, cloprostenol, given 11 d apart in replicate experiments over 2 yr. In Experiments 1 and 2, heifers were randomly allocated to 1 of 3 treatments (initiated at mid-cycle): Treatment 1--24 mg of pFSH (Folltropin) given twice daily for 4 d; Treatment 2--a single injection of 2000 IU PMSG; Treatment 3--2000 IU PMSG followed by 2000 IU of Neutra-PMSG at the time of first insemination. In Experiment 3, 116 heifers were given 2000 IU PMSG on Day 2 (n = 28), Day 3 (n = 27), Day 10 (n = 41) or Day 16 (n = 20) of the estrous cycle. The PG was given at 48 h (500 microg cloprostenol) and 60 h (250 microg cloprostenol) after the first gonadotropin treatment. Heifers were inseminated twice during estrus, and embryos were recovered on Day 7, following slaughter and graded for quality. The numbers of ovulations and large follicles (> or =10 mm) were also counted. There was no effect of treatment on ovulation rate in Experiment 1, but in Experiment 2 it was greater (P < 0.002) in heifers given PMSG (14.7 +/- 1.5) than pFSH (7.5 +/- 1.4) or PMSG-neutra-PMSG (8.7 +/- 1.5). The number of large follicles was higher following PMSG than pFSH treatment in Experiment 1, and it was higher (P < 0.004) in heifers given PMSG (5.5 +/- 0.8) than pFSH (1.12 +/- 0.7) or PMSG-neutra-PMSG (2.7 +/- 0.8) in Experiment 2. The use of Neutra-PMSG did not affect the numbers of embryos recovered or numbers of Grade 1 or 2 embryos, but it did decrease the number of Grade 3 embryos in both experiments. In Experiment 3, the ovulation rate decreased (P < 0.004) when PMSG was given on Day 3 (5.7 +/- 1.46) of the cycle rather than on Day 2 (12.3 +/- 1.64), Day 10 (13.4 +/- 1.45) or Day 16 (12.5 +/- 1.87). There was no effect of day of treatment on the numbers of large follicles. The mean numbers of embryos recovered were lower (P < 0.01) in heifers treated on Day 3 (2.1 +/- 0.67) than on Day 2 (6.8 +/- 1.0), Day 10 (6.4 +/- 0.86) or Day 16 (7.8 +/- 1.87). It is concluded that Neutra-PMSG given to heifers treated with PMSG did not improve embryo yield or quality and that treatment with PMSG early in the cycle can result in acceptable embryo yields provided sufficient time elapses between treatment and luteolysis.  相似文献

Nineteen Corriedale ewes were treated with an im dose of a PGF2alpha during the luteal phase to synchronize estrus. After ovulation had been detected by using ultrasonography (Day 0); the ewes were randomly assigned to 2 different groups. In 11 ewes a CIDR, which had previously been used for 10 d, was inserted on the fourth day after ovulation. The ewes then received a dose of PGF2alpha on Day 5 to induce luteolysis. The CIDR remained in place until the end of the experiment (Day 9). Control ewes (n = 8) received no treatment. Blood samples were taken daily for estradiol, progesterone and FSH determinations. In the untreated ewes, 2 follicular waves were detected in all of the animals throughout the monitoring period, with a mean wave interval of 4.5 d. The total number of follicles which were > or =2 mm decreased from Day 0 to Day 4 (8.8+/-1.0 to 5.3+/-0.6; P< or =0.05) and then increased at Day 7 (7.5+/-0.9; P< or =0.05). The growth profiles of both the largest and the second largest follicles of Wave 1 showed significant divergence, while no divergence was observed in Wave 2. Serum estradiol concentrations decreased significantly from the day before to the day of ovulation and then increased again during the growing phase of the largest follicle of Wave 1. Concentrations of FSH were high on the day of emergence of both waves, but while a significant decline was observed after emergence in Wave 1, the levels remained high in Wave 2. In 8 of the 11 treated ewes, the largest follicle of Wave 1 was still present on the ninth day after ovulation (persistent follicle). In the other 3 ewes, the largest follicle of Wave 1 was already regressing on the day that the treatment was administered, and the largest follicle that was present on Day 9 originated from Wave 2 (nonpersistent follicle). In persistent follicle ewes, the largest follicle of Wave 1 prolonged its lifespan significantly, attaining the maximum diameter (Day 8.1+/-0.8) later than in untreated (Day 3.0+/-0.4) and nonpersisted follicle ewes (Day 2.0+/-0.6). The total number of follicles decreased in persistent follicle ewes between Day 0 and Day 4 (7.9+/-1.5 to 4.5+/-0.5, respectively; P< or =0.05) and remained low until the end of the experiment. Progesterone concentrations (nmol/L) between Days 6 and 9 were significantly different between untreated and persistent follicle ewes (12.8+/-1.0 vs. 9.4+/-1.0, P< or =0.02). The present study confirms that the largest follicle of Wave 1 is dominant in the ewe and that subluteal progesterone concentrations can prolong its lifespan and extend this dominance.  相似文献

In a previous study in our laboratory, treatment of non-prolific Western White Face (WWF) ewes with PGF(2 alpha) and intravaginal sponges containing medroxyprogesterone acetate (MAP) on approximately Day 8 of a cycle (Day 0 = first ovulation of the interovulatory interval) resulted in ovulations during the subsequent 6 days when MAP sponges were in place. Two experiments were performed on WWF ewes during anestrus to allow us to independently examine if such ovulations were due to the direct effects of PGF(2 alpha) on the ovary or to the effects of a rapid decrease in serum concentrations of progesterone at PGF(2 alpha)-induced luteolysis. Experiment 1: ewes fitted with MAP sponges for 6 days (n = 12) were injected with PGF(2 alpha) (n = 6; 15 mg im), or saline (n = 6) on the day of sponge insertion. Experiment 2: ewes received progesterone-releasing subcutaneous implants (n = 6) or empty implants (n = 5) for 5 days. Six hours prior to implant removal, all ewes received a MAP sponge, which remained in place for 6 days. Ewes from both experiments underwent ovarian ultrasonography and blood sampling once daily for 6 days before and twice daily for 6 days after sponge insertion. Additional blood samples were collected every 4 h during sponge treatment. Experiment 1: 4-6 (67%) PGF(2 alpha)-treated ewes ovulated approximately 1.5 days after PGF(2 alpha) injection; these ovulations were not preceded by estrus or a preovulatory surge release of LH, and resulted in transient corpora hemorrhagica (CH). The growth phase was longer (P < 0.05) and the growth rate slower (P < 0.05) in ovulating versus non-ovulating follicles in PGF(2 alpha)-treated ewes. Experiment 2: in ewes given progesterone implants, serum progesterone concentrations reached a peak (1.7 2 ng/mL; P < 0.001) on the day of implant removal and decreased to basal concentrations (<0.17 ng/mL; P < 0.001) within 24 h of implant removal. No ovulations occurred in either the treated or the control ewes. We concluded that ovulations occurring after PGF(2 alpha) injection, in the presence of a MAP sponge, could be due to a direct effect of PGF(2 alpha) at the ovarian level, rather than a sudden decline in circulating progesterone concentrations.  相似文献

The estrus — ovulation time relationships was examined in Romney ewes treated with progestogen (intravaginal sponge) and gonadotropins (PMSG + HCG or PMSG alone) prior to (January) and during (April) the breeding season. The conception rate of ewes inseminated at predetermined times after treatment was also investigated.Ewes exhibited estrus sooner after sponge removal in April than in January (34.9 v 38.9 hrs, P < 0.001). The interval from sponge removal to ovulation was also shorter in April than in January (56.3 – 62.1 hrs, P < 0.01). There were no significant differences between treatments or season on the mean interval from estrus to ovulation. Types of gonadotropin treatment had no effect on the estrus — ovulation time relationships. There were no significant effects of season, hormone treatment or time of insemination on lambing rate.  相似文献

The objectives of the present study were to evaluate the induction of estrus and fertility in yak cows treated with Co-Synch regimens or progesterone (P(4)). In Experiment 1, postpartum suckled yaks were assigned to three treatments: (1) A (n=28), insertion of an intravaginal device containing P(4) (CIDR) on Day 0, PGF(2alpha) (i.m.) on Day 6 and PMSG (i.m.) at the time of CIDR removal on Day 7 (P(4)-PGF(2alpha)-PMSG); (2) B (n=21), PGF(2alpha) (i.m.) on Day 6 and PMSG on Day 7; (3) C (n=26), control group. Seven yak bulls were grazed with the cows for natural breeding. Rate of estrus within 96h of the end of treatment was greater (P<0.05) in A (100.0%) than in B (28.6%) or C (0.0%). First service conception rate (CR) determined by serum P(4) on Day 21 after breeding was greater (P<0.05) in A (78.6%) than in B (22.2%). Also, pregnancy rate (PR) during the breeding season was greater (P<0.05) in A (82.1%) than in B (19.0%) and C (7.7%). In Experiment 2, non-suckled yaks that calved in previous years but not in the current year were assigned to three treatments: (1) A (n=31), GnRH (i.m.) on Day 0, followed by PGF(2alpha) on Day 7 and timed artificial insemination (TAI) concurrently with GnRH treatment on Day 9 (Co-Synch regimen); (2) B (n=50), a CIDR device for 7 days plus PGF(2alpha) and PMSG at the time of CIDR withdrawal on Day 7 and TAI on Day 9 (P(4)-PGF(2alpha)-PMSG); (3) C (n=50), yak cows were artificially inseminated at spontaneous estrus. Frozen semen of Holstein and Jersey were used for insemination in Experiment 2. The CR assessed by rectal palpation 35 days after TAI was not different in A (22.6%), B (30.0%) and C (33.3%), but PR was greater in A and B than in C, when based on those cows presented for estrous synchronization programs. It is concluded that P(4)-PGF(2alpha)-PMSG protocol could efficiently induce estrus and result in an acceptable pregnancy rate in postpartum suckled yak cows. This technique and Co-Synch regimen can be applied successfully for TAI of non-suckled yak cows.  相似文献

The objective of this study was to evaluate the efficacy of combinations of PMSG treatment and ram contact on the reproductive performance of progestagen-treated ewes during three different times of the year, Febraury (early anestrus), July (late anestrus) and October (breeding season). A total of 109 multiparous Dorset ewes was used. Ewes were treated with intravaginal progestagen pessaries for 12 days, injected with 500 IU PMSG at pessary removal and either isolated from rams prior to mating (n = 12, February; n = 12, July; n = 8, October) or exposed to rams during pessary treatment (n = 17, February; n = 12, July; n = 8, October). A third treatment group (n = 18, February; n = 6, July; n = 8, October) received pessaries and ram exposure but no PMSG. An additional treatment of progestagen pessaries alone was included in October (n = 8). There were no differences among treatments in their ability to induce estrus at different times of the year, but incidence of estrus tended (P < 0.10) to be lower for PMSG treatment during the July breeding. During February, the use of pessaries with PMSG treatment increased (P < 0.05) conception and lambing rates, whereas ram contact was without any beneficial effects. The trend was reversed during July breeding, when ram contact increased (P < 0.05) fertility of progestagen-treated ewes compared with other treatment combinations. Pessaries alone were sufficient to attain acceptable levels of fertility and fecundity in October.  相似文献

Two experiments were conducted during mid-gestation to examine effects in ewes of propylthiouracil (PTU) treatment alone or with melatonin on serum thyroid hormones, postpartum reproduction, and lamb performance. In the first experiment, beginning on day 0 (first day of treatment when all animals were 72.2+/-0.9 days of gestation), ewes received daily treatments (gavage) consisting of either 0mg (n=6) or 40 mg (n=6) PTU/kg body weight/day for 15 days. After 15 days, the 40 mg dosage was decreased to 20mg/kg body weight for an additional 20 days (35 days of PTU). Serum thyroxine (T4) did not differ (P>0.10) between groups through day 4; but on day 5, control females had a serum value of 67 ng/ml compared with 46 (+/-5)ng/ml for PTU-treated ewes (P=0.02). On the last day that 40 mg of PTU was administered, serum T4 averaged 67 and 7 (+/-5)ng/ml (P<0.001) in the two respective groups. Serum T4 remained low and was 80 and 1 ng/ml (P<0.001) in control and treated ewes on day 34. Serum T4 rose gradually after PTU but remained different from that observed in control ewes through day 48. Lambs from control and treated ewes had similar (P=0.46) T4 values at birth but lambs from PTU-treated ewes had lower (P=0.03) birth weights than did those from control ewes. Serum progesterone (P4) after parturition indicated a lack of cyclicity in all ewes. In the second experiment, beginning on day 0 (76.8+/-4.7 days of gestation), ewes received PTU as in Experiment 1. In addition, after 15 days of PTU, melatonin was given (i.m. injections at 5mg/day) for 30 days. Propylthiouracil decreased (P0.60) for lambs born to control and treated ewes. Female offspring of PTU+melatonin-treated dams reached puberty, became anestrus, and returned to cyclicity at similar (P>0.10) times to contemporary ewe lambs. Results indicate that 40/20mg PTU alone or with melatonin does not induce cyclicity after lambing in spring lambing ewes and has little effect on offspring performance.  相似文献

Follicles were obtained from the ovaries of four groups of 15 ewes. Ewes in the control group were ovariectomized on the 12th day of the oestrous cycle. The other ewes were all given PMSG on the 12th day of the cycle; some were ovariectomized 24 or 40 h later, the others were given prostaglandin followed by hCG and were ovariectomized 6 or 12 h after the hCG injection. All follicles greater than 2 mm in diameter were measured and examined macroscopically for signs of atresia. Some were subjected to detailed morphological examination, the pattern of steroid secretion was determined in others. All the evidence from these three approaches suggested that, in vivo, reversal of the atretic process ('rescue') plays no part in the increase in the number of follicles observed following administration of PMSG.  相似文献

The present study was designed to study follicular growth and its interactions with the corpus luteum of pregnancy in sheep during early, middle and late pregnancy and during postpartum anestrus. Ewes with 1 or 2 corpora lutea in one ovary were selected from a larger group of Serres ewes. All pregnant ewes were randomly allocated to two groups, with 10 to 12 ewes per group. Ewes of Group I were treated with 750 IU hCG at Day 25 or 45 or 70 or 100 or 125 of pregnancy. In Group II, ewes were treated with a combination of 1000 IU PMSG + 750 IU hCG either at Day 25 or 45 or 70 or 100 of pregnancy. The results demonstrated the presence of gonadotrophin-responsive follicles during early pregnancy (Days 25 to 45), reduction of their number during mid-pregnancy (Days 70 to 100), and their disappearance during late pregnancy (Day 125). Administration of hCG to Serres ewes at 10 and 20 days postpartum induced ovulation of a high proportion of ewes at 10 days postpartum (62%) with a further increase observed at 20 days postpartum (75%). During pregnancy, as well as during the postpartum period, there was no significant difference in the number of ovulations induced according to the location of the corpus luteum of pregnancy. These data demonstrate that the presence of the corpus luteum of pregnancy does not affect the number of gonadotrophin-responsive follicles until Day 100 of pregnancy. However, during late pregnancy such follicles were no longer present in the ovaries. Gonadotrophin-responsive follicles were again present as soon as Day 10 postpartum.  相似文献
