The influence of site fertility on soil microbial biomass and activity is not well understood but is likely to be complex because of interactions with plant responses to nutrient availability. We examined the effects of long-term (8 yr) fertilization and litter removal on forest floor microbial biomass and N and C transformations to test the hypothesis that higher soil resource availability stimulates microbial activity. Microbial biomass and respiration decreased by 20–30 % in response to fertilization. Microbial C averaged 3.8 mg C/g soil in fertilized, 5.8 mg C/g in control, and 5.5 mg C/g in litter removal plots. Microbial respiration was 200 µg CO2-C g–1 d–1 in fertilized plots, compared to 270 µg CO2-C g–1 d–1 in controls. Gross N mineralization and N immobilization did not differ among treatments, despite higher litter nutrient concentrations in fertilized plots and the removal of substantial quantities of C and N in litter removal plots. Net N mineralization was significantly reduced by fertilization. Gross nitrification and NO3 immobilization both were increased by fertilization. Nitrate thus became a more important part of microbial N cycling in fertilized plots even though NH4 + availability was not stimulated by fertilization.Soil microorganisms did not mineralize more C or N in response to fertilization and higher litter quality; instead, results suggest a difference in the physiological status of microbial biomass in fertilized plots that influenced N transformations. Respiration quotients (qCO2, respiration per unit biomass) were higher in fertilized plots (56 µg CO2-C mg C–1 d–1) than control (48 µg CO2-C mg C–1 d –1) or litter removal (45 µg CO2-C mg C–1 d–1), corresponding to higher microbial growth efficiency, higher proportions of gross mineralization immobilized, and lower net N mineralization in fertilized plots. While microbial biomass is an important labile nutrient pool, patterns of microbial growth and turnover were distinct from this pool and were more important to microbial function in nitrogen cycling.

Wet tropical forests play a critical role in global ecosystem carbon (C) cycle, but C allocation and the response of different C pools to nutrient addition in these forests remain poorly understood. We measured soil organic carbon (SOC), litterfall, root biomass, microbial biomass and soil physical and chemical properties in a wet tropical forest from May 1996 to July 1997 following a 7‐year continuous fertilization. We found that although there was no significant difference in total SOC in the top 0–10 cm of the soils between the fertilization plots (5.42±0.18 kg m?2) and the control plots (5.27±0.22 kg m?2), the proportion of the heavy‐fraction organic C in the total SOC was significantly higher in the fertilized plots (59%) than in the control plots (46%) (P<0.05). The annual decomposition rate of fertilized leaf litter was 13% higher than that of the control leaf litter. We also found that fertilization significantly increased microbial biomass (fungi+bacteria) with 952±48 mg kg?1soil in the fertilized plots and 755±37 mg kg?1soil in the control plots. Our results suggest that fertilization in tropical forests may enhance long‐term C sequestration in the soils of tropical wet forests.

The fate of soil organic carbon (SOC) is determined, in part, by complex interactions between the quality of plant litter inputs, nutrient availability, and the microbial communities that control decomposition rates. This study explores these interactions in a mesic grassland where C and nitrogen (N) availability and plant litter quality have been manipulated using both fertilization and haying for 7 years. We measured a suite of soil parameters including inorganic N, extractable organic C and N (EOC and EON), soil moisture, extracellular enzyme activity (EEA), and the isotopic composition of C and N in the microbial biomass and substrate sources. We use these data to determine how the activity of microbial decomposers was influenced by varying levels of substrate C and N quality and quantity and to explore potential mechanisms explaining the fate of enhanced plant biomass inputs with fertilization. Oxidative EEA targeting relatively recalcitrant C pools was not affected by fertilization. EEA linked to the breakdown of relatively labile C rich substrates exhibited no relationship with inorganic N availability but was significantly greater with fertilization and associated increases in substrate quality. These increases in EEA were not related to an increase in microbial biomass C. The ratio of hydrolytic C:N acquisition enzymes and δ13C and δ15N values of microbial biomass relative to bulk soil C and N, or EOC and EON suggest that microbial communities in fertilized plots were relatively C limited, a feature likely driving enhanced microbial efforts to acquire C from labile sources. These data suggest that in mesic grasslands, enhancements in biomass inputs and quality with fertilization can prompt an increase in EEA within the mineral soil profile with no significant increases in microbial biomass. Our work helps elucidate the microbially mediated fate of enhanced biomass inputs that are greater in magnitude than the associated increases in mineral soil organic matter.

The microbial communities of high‐latitude ecosystems are expected to experience rapid changes over the next century due to climate warming and increased deposition of reactive nitrogen, changes that will likely affect microbial community structure and function. In moist acidic tundra (MAT) soils on the North Slope of the Brooks Range, Alaska, substantial losses of C and N were previously observed after long‐term nutrient additions. To analyse the role of microbial communities in these losses, we utilized 16S rRNA gene tag pyrosequencing coupled with community‐level physiological profiling to describe changes in MAT bacterial communities after short‐ and long‐term nutrient fertilization in four sets of paired control and fertilized MAT soil samples. Bacterial diversity was lower in long‐term fertilized plots. The Acidobacteria were one of the most abundant phyla in all soils and distinct differences were noted in the distributions of Acidobacteria subgroups between mineral and organic soil layers that were also affected by fertilization. In addition, Alpha‐ and Gammaproteobacteria were more abundant in long‐term fertilized samples compared with control soils. The dramatic increase in sequences within the Gammaproteobacteria identified as Dyella spp. (order Xanthomonadales) in the long‐term fertilized samples was confirmed by quantitative PCR (qPCR) in several samples. Long‐term fertilization was also correlated with shifts in the utilization of specific substrates by microbes present in the soils. The combined data indicate that long‐term fertilization resulted in a significant change in microbial community structure and function linked to changes in carbon and nitrogen availability and shifts in above‐ground plant communities.

The response of belowground biological processes to soil N availability in Larix gmelinii (larch) and Fraxinus mandshurica (ash) plantations was studied. Soil and root respiration were measured with Li-Cor 6400 and gas-phase O2 electrodes, respectively. Compared with the control, N fertilization induced the decreases of fine root biomass by 52% and 25%, and soil respiration by 30% and 24% in larch and ash plantations, respectively. The average soil microbial biomass C and N were decreased by 29% and 42% under larch stand and 39% and 47% under ash stand, respectively. While the fine root tissue N concentration under fertilized plots was higher 26% and 12% than that under control plots, respectively, the average fine root respiration rates were increased by 10% and 13% in larch and ash stands under fertilized plot, respectively. Soil respiration rates showed significantly positive exponential relationships with soil temperature, and a seasonal dynamic. These findings suggest that N fertilization can suppress fine root biomass at five branch orders (<2 mm in diameter), soil respiration, and soil microbial biomass C and N, and alter soil microbial communities in L. gmelinii and F. mandshurica plantations.

If microbial degradation of carbon substrates in arctic soil is stimulated by climatic warming, this would be a significant positive feedback on global change. With data from a climate change experiment in Northern Sweden we show that warming and enhanced soil nutrient availability, which is a predicted long-term consequence of climatic warming and mimicked by fertilization, both increase soil microbial biomass. However, while fertilization increased the relative abundance of fungi, warming caused only a minimal shift in the microbial community composition based on the phospholipid fatty acid (PLFA) and neutral lipid fatty acid (NLFA) profiles. The function of the microbial community was also differently affected, as indicated by stable isotope probing of PLFA and NLFA. We demonstrate that two decades of fertilization have favored fungi relative to bacteria, and increased the turnover of complex organic compounds such as vanillin, while warming has had no such effects. Furthermore, the NLFA-to-PLFA ratio for 13C-incorporation from acetate increased in warmed plots but not in fertilized ones. Thus, fertilization cannot be used as a proxy for effects on warming in arctic tundra soils. Furthermore, the different functional responses suggest that the biomass increase found in both fertilized and warmed plots was mediated via different mechanisms.

Abstract. A cross‐over fertilization experiment was carried out in Dutch floating fens to investigate effects on biomass production in the same and the following years. In total 16 fertilizer treatments were applied, combining four treatments in 1999 with four treatments in 2000 (addition of 20 g.m?2 N, 5 g.m?2 P, both elements and unfertilized control). The above‐ground biomass production of vascular plants was co‐limited by N and P in both years. However, in plots that were only fertilized in 1999 the effects of individual nutrients differed between the two years: N‐fertilization slightly increased the amount of biomass produced in the same year (1999), whereas P‐fertilization did so in the following year (2000). Fertilizer applied in 1999 also influenced the effects of fertilizer applied in 2000. One year after N‐fertilization vascular plant growth was still co‐limited by N and P, but one year after P‐fertilization, vascular plant growth was only limited by N. Bryophyte biomass responded weakly to fertilization. Nutrient concentrations in plant biomass, nutrient standing crops and measurements of N and P availability in the soil indicated that one year after fertilization, the N‐fertilizer had mostly ‘disappeared’ from N‐fertilized plots, whereas the availability of P remained markedly enhanced in P‐fertilized plots. In addition, P‐fertilization enhanced the uptake of N by plants the following year. The time‐dependence of fertilizer effects was probably caused by (1) higher addition of P than of N relative to the requirements of plants; (2) longer retention of P than of N in the system; (3) positive effect of P‐fertilization on the availability of N; (4) contrasting effects of N‐ and P‐fertilization on nutrient losses by plants and/or on their responses to subsequent nutrient addition; (5) changing interactions between vascular plants and mosses (mainly Sphagnum spp.); (6) nutrient export through the repeated harvest of above‐ground biomass. To determine which nutrient limits plant growth fertilization experiments should be short, avoiding that indirect effects of a non‐limiting nutrient influence results. To indicate how changed nutrient supply will affect an ecosystem longer‐term experiments are needed, so that indirect effects have time to develop and be detected.

We studied long-term effects of fertilization with wood ash on biomass, vitality and mycorrhizal colonization of fine roots in three conifer forest stands growing in Vacciniosa turf. mel. (V), Myrtillosa turf. mel. (M) and Myrtillosa turf. mel./Caricoso-phragmitosa (MC) forest types on peat soils. Fertilization trials amounting 5 kg/m2 of wood ash were established 12 years prior to this study. A total of 63 soil samples with roots were collected and analysed. Ectomycorrhizal (ECM) fungi in roots were identified by morphotyping and sequencing of the fungal internal transcribed spacer (ITS) region. In all forest types, fine root biomass was higher in fertilized plots than in control plots. In M forest type, proportion of living fine roots was greater in fertilized plots than in control plots, while in V and MC, the result was opposite. Fifty ECM species were identified, of which eight were common to both fertilized and control plots. Species richness and Shannon diversity index were generally higher in fertilized plots than in control plots. The most common species in fertilized plots were Amphinema byssoides (17.8 %) and Tuber cf. anniae (12.2 %), while in control plots, it was Tylospora asterophora (18.5 %) and Lactarius tabidus (20.3 %). Our results showed that forest fertilization with wood ash has long-lasting effect on diversity and composition of ECM fungal communities.

Plant traits are known to control litter decomposition rates through afterlife effects on litter quality. Land-use practices that modify plant traits, e.g. livestock grazing and soil fertilization, also have cascading effects on litter decomposition. However, almost all studies of these afterlife effects ignored the role of soil detritivores in the decomposition processes. We explored how the feeding activities of a macroarthropod modify microbial activity in leaf litter. Dead leaves from two grassland species, Bromopsis erecta and Potentilla verna, were collected in fertilized or unfertilized grazed plots and fertilized or unfertilized ungrazed plots. We determined how intraspecific variation in litter quality in response to sheep grazing and soil fertilization (i) influences the consumption and assimilation of leaf litter by the millipede Glomeris marginata, and (ii) affects the activity of microbial decomposers, assessed by substrate-induced respiration (SIR), in leaf litter before consumption and in faecal pellets and litter remains processed by Glomeris under all treatments. In the absence of millipedes, microbial activity was significantly higher in leaf litter from fertilized plots. Glomeris consumed larger amounts of leaf litter from fertilized grazed plots, owing to increased consumption of the otherwise poorly palatable Bromopsis, and produced larger amounts of faecal pellets when fed on this food. However, irrespective of the food consumed, SIR in faecal pellets was found to be similar in all treatments. Moreover, SIR in litter remains unconsumed at the end of the experiment was reduced to low and similar levels in all treatments. Overall, homogenization of microbial activity by Glomeris suppressed differences in SIR between leaf litter from fertilized and unfertilized plots, in both Bromopsis and Potentilla. Our results suggest that studies that assess afterlife effects of plant traits on decomposition using methods that exclude soil macrofauna may prove inadequate in ecosystems with abundant populations of detritivores.

Nitrogen transformations in the soil, and the resulting changes in carbon and nitrogen compounds in soil percolate water, were studied in two stands of Norway spruce (Picea abies L.). Over the last 30 years the stands were repeatedly limed (total 6000 kg ha–1), fertilized with nitrogen (total about 900 kg ha–1), or both treatments together. Both aerobic incubations of soil samples in the laboratory, and intact soil core incubations in the field showed that in control plots ammonification widely predominated over nitrification. In both experiments nitrogen addition increased the formation of mineral-N. In one experiment separate lime and nitrogen treatments increased nitrification, in the other, only lime and nitrogen addition together had this effect. In one experiment immobilization of nitrogen to soil microbial biomass was lower in soil only treated with nitrogen. Soil percolate water was collected by means of lysimeters placed under the humus layer and 10 cm below in the mineral soil. Total N, NH4-N and NO3-N were measured, and dissolved organic nitrogen was fractioned according to molecular weight. NO3-N concentrations in percolate water, collected under the humus layer, were higher in plots treated with N-fertilizer, especially when lime was also added. The treatments had no effect on the N concentrations in mineral soil. A considerable proportion of nitrogen was leached in organic form.

Based on lipid analyses, 16S rRNA/rRNA gene single-strand conformation polymorphism fingerprints and methane flux measurements, influences of the fertilization regime on abundance and diversity of archaeal communities were investigated in soil samples from the long-term (103 years) field trial in Bad Lauchst?dt, Germany. The investigated plots followed a gradient of increasing fertilization beginning at no fertilization and ending at the 'cattle manure' itself. The archaeal phospholipid etherlipid (PLEL) concentration was used as an indicator for archaeal biomass and increased with the gradient of increasing fertilization, whereby the concentrations determined for organically fertilized soils were well above previously reported values. Methane emission, although at a low level, were occasionally only observed in organically fertilized soils, whereas the other treatments showed significant methane uptake. Euryarchaeotal organisms were abundant in all investigated samples but 16S rRNA analysis also demonstrated the presence of Crenarchaeota in fertilized soils. Lowest molecular archaeal diversity was found in highest fertilized treatments. Archaea phylogenetically most closely related to cultured methanogens were abundant in these fertilized soils, whereas Archaea with low relatedness to cultured microorganisms dominated in non-fertilized soils. Relatives of Methanoculleus spp. were found almost exclusively in organically fertilized soils or cattle manure. Methanosarcina-related microorganisms were detected in all soils as well as in the cattle manure, but soils with highest organic application rate were specifically dominated by a close phylogenetic relative of Methanosarcina thermophila. Our findings suggest that regular application of cattle manure increased archaeal biomass, but reduced archaeal diversity and selected for methanogenic Methanoculleus and Methanosarcina strains, leading to the circumstance that high organic fertilized soils did not function as a methane sink at the investigated site anymore.

Coastal marshes represent an important transitional zone between uplands and estuaries. One important function of marshes is to assimilate nutrient inputs from uplands, thus providing a buffer for anthropogenic nutrient loads. We examined the effects of nitrogen (N) and phosphorus (P) fertilization on biogeochemical and microbial processes during the summer growing season in a Spartina patens (Aiton (Muhl.)) marsh in the Narragansett Bay National Estuarine Research Reserve on Prudence Island (RI). Quadruplicate 1 m2 plots were fertilized with N and P additions, N-only, P-only, or no additions. N-only addition significantly stimulated bacterial production and increased pore water NH4+ and NO3 concentrations. Denitrification rates ranged from 0 to 8 mmol m−2 day−1. Fertilization had no apparent effect on soil oxygen consumption or denitrification measured in the summer in intact cores due to high core-to-core variation. P fertilization led to increased pore water dissolved inorganic phosphorus (DIP) concentrations and increased DIP release from soils. In contrast the control and N-only treatments had significant DIP uptake across the soil-water interface. The results suggest that in the summer fertilization has no apparent effect on denitrification rates, stimulates bacterial productivity, enhances pore water nutrient concentrations and alters some nutrient fluxes across the marsh surface.

According to the singular hypothesis of plant diversity, different plant species are expected to make unique contributions to ecosystem functioning. Hence, individual species would support distinct microbial communities. It was hypothesized that microbial community dynamics in the respective rhizospheres of, two floristically divergent species, Agrostis capillaris and Prunella vulgaris that were dominant in a temperate, upland grassland in northern Greece, would support distinct microbial communities, in agreement to the singular hypothesis. Phospholipid lipid fatty acid (PLFA) profiles of the rhizosphere soil microbial community were obtained from the grassland which had been subjected to factorial nitrogen (N) and phosphorus (P) fertilization over five plant growth seasons. The soil cores analyzed were centered on stands of the two co-occurring target plant species, sampled from five blocks in all four factorial N and P fertilization combinations. Distinct PLFA clustering patterns following principle component analysis of PLFA concentrations revealed that, in the absence of P fertilization, soils under the two plant species supported divergent microbial communities. In the P fertilized plots, however, no such distinction could be observed. Results reveal that nutrient fertilization may mask the ability of plant species to shape their own rhizosphere microbial community.

This study examines the effect of four tree species on nitrogen (N) retention within forested catchments of the Catskill Mountains, New York (NY). We conducted a 300-day 15N field tracer experiment to determine how N moves through soil, microbial, and plant pools under different tree species and fertilization regimes. Samples were collected from single-species plots of American beech (Fagus grandifolia Ehrh.), eastern hemlock (Tsuga canadensis L.), red oak (Quercus rubra L.), and sugar maple (Acer saccharum Marsh). Using paired plots we compared the effects of ambient levels of N inputs (11 kg N/ha/y) to additions of 50 kg N/ha/y that began 1.5 years prior to and continued throughout this experiment. Total plot 15N recovery (litter layer, organic and mineral soil to 12 cm, fine roots, and aboveground biomass) did not vary significantly among tree species, but the distribution of sinks for 15N within the forest ecosystem did vary. Recovery in the forest floor was significantly lower in sugar maple stands compared to the other species. 15Nitrogen recovery was 22% lower in the fertilized plots compared to the ambient plots and red oak stands had the largest drop in 15N recovery as a result of N fertilization. Aboveground biomass became a significantly greater 15N sink with fertilization, although it retained less than 1% of the tracer addition. These results indicate that different forest types vary in the amount of N retention in the forest floor, and that forest N retention may change depending upon N inputs.

Previous studies have shown that fertilization with nitrogen depresses overall microbial biomass and activity in soil. In the present study we broaden our understanding of this phenomenon by studying the seasonality of responses of specific microbial functional groups to chronic nitrogen additions in alpine tundra soils. We measured soil enzyme activities, mineralization kinetics for 8 substrates, biomass of 8 microbial functional groups, and changes in N and carbon pools in the soil. Our approach allowed us to compare the ability of the soil microbial biomass to utilize various substrates in addition to allowing us to estimate changes in biomass of microbial functional groups that are involved in carbon and nitrogen cycling. Overall microbial activity and biomass was reduced in fertilized plots, whereas pools of N in the soil and microbial biomass N were higher in fertilized plots. The negative effects of N were most prominent in the summer. Biomass of the dominant microbial functional groups recovered in fertilized soils during the winter and nitrogen storage in microbial biomass was higher in fertilized soils in the autumn and winter than in the summer. Microbial immobilization of N may therefore be a significant sink for added N during autumn and winter months when plants are not active. One large microbial group that did not recover in the winter in fertilized soils was phenol mineralizers, possibly indicating selection against microbes with enzyme systems for the breakdown of phenolic compounds and complex soil organic matter. Overall, this work is a step towards understanding how chronic N additions affect the structure and biogeochemical functioning of soil microbial communities.

The effects of preplant phosphorus fertilization on fine root (2 mm) distribution were examined in an 11-year-old stand of loblolly pine (Pinus taeda L.) located on the lower Coastal Plain of North Carolina. Root auger cores were collected from the planting bed and interbed areas from two depths (0–10 and 10–20 cm) from fertilized (45 kg P ha–1 at time of planting) and nonfertilized plots. Root samples were collected and aboveground growth measured during the 11th year after fertilization. Despite significant increases in aboveground volume and biomass due to fertilization, fine root biomass was unaffected. No differences in rooting density (root length per volume of soil) due to phosphorus additions were detected. However, the ratio of fine root biomass to aboveground (shoot) biomass (R:S) was significantly smaller on plots receiving phosphorus fertilization.operated by Martin Marietta Energy Systems, Inc., under Contract No. DE-AC05-840 R21400 with the U.S. Department of Energy  相似文献

In grassland ecosystems, N and P fertilization often increase plant productivity, but there is no concensus if fertilization affects soil C fractions. We tested effects of N, P and N+P fertilization at 5, 10, 15 g m−2 yr−1 (N5, N10, N15, P5, P10, P15, N5P5, N10P10, and N15P15) compared to unfertilized control on soil C, soil microbial biomass and functional diversity at the 0–20 cm and 20–40 cm depth in an alpine meadow after 5 years of continuous fertilization. Fertilization increased total aboveground biomass of community and grass but decreased legume and forb biomass compared to no fertilization. All fertilization treatments decreased the C:N ratios of legumes and roots compared to control, however fertilization at rates of 5 and 15 g m−2 yr−1 decreased the C:N ratios of the grasses. Compared to the control, soil microbial biomass C increased in N5, N10, P5, and P10 in 0–20 cm, and increased in N10 and P5 while decreased in other treatments in 20–40 cm. Most of the fertilization treatments decreased the respiratory quotient (qCO2) in 0–20 cm but increased qCO2 in 20–40 cm. Fertilization increased soil microbial functional diversity (except N15) but decreased cumulative C mineralization (except in N15 in 0–20 cm and N5 in 20–40 cm). Soil organic C (SOC) decreased in P5 and P15 in 0–20 cm and for most of the fertilization treatments (except N15P15) in 20–40 cm. Overall, these results suggested that soils will not be a C sink (except N15P15). Nitrogen and phosphorus fertilization may lower the SOC pool by altering the plant biomass composition, especially the C:N ratios of different plant functional groups, and modifying C substrate utilization patterns of soil microbial communities. The N+P fertilization at 15 g m−2 yr−1 may be used in increasing plant aboveground biomass and soil C accumulation under these meadows.
