 The protein-storing cells (PSCs) in Hevea brasiliensis were studied by using light- and electron-microscopy and SDS-PAGE. The cells were found in stem and root where secondary phloem was developed. They are a special kind of phloem parenchyma cell which accumulate in their central vacuoles large amounts of protein, fibril-like under an electron microscope, and have few plastids with very small starch grains. Their distribution is strictly restricted to the secondary phloem axial system where they exactly sequestered in functional phloem or slightly over it. A 67 kDa protein was always found in the tissues where the PSCs were observed. During the first seasonal growth flush, the 67 kDa protein in the terminal branchlet exhibits marked quantitative fluctuation which is consistent with the change of the vacuole protein inclusion of the PSCs in the branchlet. These facts suggested that the 67 kDa protein might be the major part of the vacuole protein of the PSCs. Considering the differences between the PSCs in Hevea and the PSCs in the other trees studied, we define two types of PSCs: Hevea-type, which are the cells specialized for protein storage and Populus-type, which are ordinary parenchyma cells accumulating protein and starch. 以热研88-13×IAN873的94个F1群体为试材, 利用简单序列重复(Simple sequence repeat, SSR)标记, 采用FsLinkageMAP 1.0软件, 构建了巴西橡胶树热研88-13×IAN873的遗传连锁图谱。从441对SSR引物中筛选出160对具有多态信息的引物, 在分离群体中共检测到206个多态性位点, 176个位点用于遗传图谱的构建; χ2检验结果显示, 有147个位点符合1:1分离比例, 有12个符合1:2:1分离比例, 有17个符合1:1:1:1的分离比例, 共有13个偏分离位点, 偏分离率低(7.38%); 91个SSR位点被分为18个连锁群, 覆盖橡胶树基因组1 937.06 cM, 每个连锁群包含2~16个位点, 标记间的平均距离为21.29 cM。  相似文献

The hypothesis that root apical diameter may be used to evaluate root growth potential was tested. Temporal variations in the apical diameter of individual roots of rubber seedlings ( Hevea brasiliensis ) were studied together with their elongation patterns, using root observation boxes under controlled conditions. This study confirmed the overall positive correlation between apical diameter and growth rale. Moreover, the two parameters, varied in the same way during the life of a given root. For roots with short growth duration, there was a parallel quick decrease in both apical diameter and elongation rate, whereas roots that grew for longer periods showed synchronous fluctuations for both parameters. Since the mean values for the secondary roots within a root system exhibited the same trends, variations in apical diameter and elongation rates should depend on factors influencing the whole root system. When related to shoot rhythmic growth, both apical diameter and elongation rates were depressed during the periods of leaf growth. These effects were enhanced and/or prolonged by shading, hence reinforcing the hypothesis that this development depends on assimilate availability. Such results can be interpreted in terms of a source-sink relationship within the whole plant by considering the apical diameter, representing the size of the meristem related to the number of rneristematic cells, as an indicator of each root's growth potential.  相似文献

 The effect of the saprobe fungi Wardomyces inflatus (Marchal) Hennebert, Paecilomyces farinosus (Holm & Gray) A. H. S. Brown & G. Sm., Gliocladium roseum Bain., Trichoderma pseudokoningii Rifai and T. harzianum Rifai, isolated from sporocarps of Glomus mosseae, on arbuscular mycorrhizal (AM) colonisation and plant dry matter of soybean was studied in 2/3 and 1/5 diluted soils in a greenhouse trial. Soil dilution to 1/5 had no effect on shoot dry matter of soybean but decreased AM colonisation and root dry weight of plants. CFU of saprobe fungi, except T. harzianum, were higher in 1/5 than in 2/3 diluted soils. W. inflatus and Gliocladium roseum decreased the shoot dry weight of soybean plant when inoculated together with Glomus mosseae. The saprobe fungi P. farinosus and T. pseudokoningii increased the shoot dry weights of plants grown in 1/5 diluted soil. The shoot dry weight and AM colonisation in 1/5 diluted soil were also increased when T. harzianum was inoculated together with Glomus mosseae. Thus, saprobe fungi increased AM colonisation of soybean plants by indigenous endophytes. The AM colonisation of plants at both soil dilutions was increased by Glomus mosseae. The highest level of AM colonisation was observed when P. farinosus and T. pseudokoningii were inoculated together Glomus mosseae. The dilution of soils influenced the interaction between inoculated microorganisms and their effect on plant growth. Accepted: 7 June 1999  相似文献

Hevein has been found to be an essential element in coagulation of rubber particles in latex of rubber trees. In a previous study, we cloned a 1 241-bp fragment of a 5' upstream region of the hevein gene by genome walking. This fragment was analyzed by a 5' end nested deletion method in the present study, fused with a uidA (gus) gene to produce a series of tested constructs, which were transferred into C-serum of latex and the Gus activities were detected. Results showed that the fragment from -749 to -292 was sufficient for expression of gus gene in latex, and the fragment from -292 to -168 was crucial in response to abscisic acid inducement. In a transient transgenic test of rubber leaf with particle bombardment, construct Hev749 conferred gus-specific expression in veins, in which the latex tubes mainly distributed. This implies that the fragment from -749 to -292 was laticiferous-specific.  相似文献

 Although Pinaceae and Betulaceae have been reported to contain Glomus–type root endophytes, its ecological importance and the conditions influencing this symbiosis are poorly understood. Seedlings of Abies lasiocarpa, Alnus rubra, Pinus contorta, Pinus ponderosa, Pseudotsuga menziesii, and Tsuga heterophylla were inoculated with Glomus intraradices to determine the vesicular-arbuscular mycorrhizae (VAM) development and responsiveness of these hosts. The role of companion VAM host plants on mycorrhizal colonization and nutrient uptake by Pseudotsuga menziesii was also examined by growing seedlings of Pseudotsuga menziesii in dual culture with VAM hosts Thuja plicata or Calamagrostis rubescens. After 8 weeks, no seedlings were colonized. At 16 weeks, 8 of 17 Thuja plicata seedlings grown with Pseudotsuga menziesii and all 18 inoculated Thuja plicata seedlings grown alone were colonized with vesicles and hyphae. Two of 17 inoculated Pseudotsuga menziesii seedlings grown in dual culture with Thuja plicata were colonized with abundant vesicles and hyphae. No ectomycorrhizal seedlings grown in monoculture were colonized. At 9 months, all 10 Calamagrostis rubescens and all 10 inoculated Pseudotsuga menziesii seedlings grown in dual culture were colonized by vesicles and hyphae. Two of 10 inoculated Pseudotsuga menziesiiand 1 of 10 inoculated Pinus ponderosa seedlings grown in monoculture were similarly colonized. The mean phosphorus content in the needles of colonized Pseudotsuga menziesii seedlings grown with Calamagrostis rubescens was about twice as high as in noncolonized Pseudotsuga menziesiiseedlings grown with Calamagrostis rubescens. Tissue nitrogen did not differ between these treatments. The results show that Glomus intraradices colonization of Pinaceae is most successful when a VAM host is present, although some vesicular colonization of Pinaceae occurred in the absence of a VAM host. 对与死皮病有密切关系的HbMybl基因进行了克隆和结构分析。该基因含有一个344bp的内含子和2个外显子。利用Genome Walker的方法获得了1.2kb的5′前年序列核心启动子,分析表明该启动子可能是富含GC和依赖于起始子(Inr)的非典型启动子。Southern杂交分析表明,HbMybl在橡胶树基因组中为2个拷贝。HbMybl在树皮中表达最强,在叶片中最弱。  相似文献

How tropical trees flower synchronously near the equator in the absence of significant day length variation or other meteorological cues has long been a puzzle. The rubber tree (Hevea brasiliensis) is used as a model to investigate this phenomenon. The annual cycle of solar radiation intensity is shown to correspond closely with the flowering of the rubber tree planted near the equator and in the subtropics. Unlike in temperate regions, where incoming solar radiation (insolation) is dependent on both day length and radiation intensity, insolation at the equator is due entirely to the latter. Insolation at the upper atmosphere peaks twice a year during the spring and autumn equinoxes, but the actual solar radiation that reaches the ground is attenuated to varying extents in different localities. The rubber tree shows one or two flowering seasons a year (with major and minor seasons in the latter) in accordance with the solar radiation intensity received. High solar radiation intensity, and in particular bright sunshine (as distinct from prolonged diffuse radiation), induces synchronous anthesis and blooming in Hevea around the time of the equinoxes. The same mechanism may be operational in other tropical tree species.  相似文献

用茉莉酸(JA)处理三叶橡胶,分别在JA处理1、24、48、72、96、120小时后,分析三叶橡胶光合作用能力和抗氧化系统活性以及单萜类物质的变化。结果发现,与对照相比,用JA处理的三叶橡胶的叶片最大净光合速率和气孔导度显著下降,光系统II潜在最大光化学效率和内在光化学效率也明显下降。光合作用能力的下降可能是由于JA处理导致的叶绿素、类胡萝卜素、可溶性蛋白质含量逐渐下降的结果。然而,JA处理却导致过氧化氢含量逐渐增加,过氧化氢含量的增加激活了抗氧化系统,结果导致超氧化物歧化酶、过氧化氢酶、过氧化物酶活性上升,抗坏血酸-谷胱甘肽循环加强。但是,α-蒎烯、β-蒎烯、桧烯以及总单萜这些潜在的抗氧化物的含量却先增加,而后逐渐下降。  相似文献

从巴西橡胶树Hevea brasiliensis差减cDNA文库中分离到微管相关蛋白(Microtubule-associated protein,MAPs)基因片段,根据该基因片段序列信息,设计特异引物,采用cDNA末端快速扩增技术RACE(Rapid Amplification ofcDNA Ends)进行差异片段的5'和3'端的扩增,获得了长度为788bp的全长cDNA,该基因在GenBank中的登录号为AY461412.序列分析表明该基因包含完整的开放阅读框,编码144个氨基酸,与微管相关蛋白基因家族具有很高的同源性,推测该基因是微管相关蛋白基因.半定量RT-PCR检测证实它在胶乳中的表达强于叶中,胁迫处理(伤害及乙烯处理)使其表达上调.  相似文献

Tapping panel dryness (TPD) syndrome affecting rubber tree (Hevea brasiliensis) is known to reduce natural latex production. Its aetiology remains ambiguous despite long years of research. A low molecular weight RNA similar to viroid RNA was isolated from TPD-affected samples of rubber trees. In the present study, a modified return-polyacrylamide gel electrophoresis procedure was standardised. The viroid-like low molecular weight (LMW) RNA was found associated with leaf, bark and root tissues and rubber seedlings. The technique was employed to detect LMW RNA in different clones of rubber planted in different locations and in bud-grafted plants. The LMW RNA isolated from TPD-affected trees was found infectious on seedlings of tomato cv Pusa Ruby. The LMW RNA was reisolated from symptomatic tomato leaves but not from control plants. This is for the first time that a biotic agent, a viroid RNA, is found consistently associated with the syndrome. The technology developed can be useful to demonstrate the onset of TPD in untapped trees in the absence of other methods such as nucleic acid hybridisation.  相似文献

橡胶树死皮病胶乳C-乳清差异表达蛋白质的筛选与鉴定   总被引:9,自引:0,他引:9  
橡胶树死皮病(Tapping Panel Dryness,TPD)在世界各橡胶种植园普遍发生,给橡胶种植业带来严重的危害。为了更好地了解和阐明死皮病发生、发展的分子机制,本研究应用双向凝胶电泳技术(2-DE)比较橡胶树死皮株与健康株胶乳C-乳清蛋白质组表达的差异。采用固相pH梯度双向凝胶电泳分离橡胶树死皮株与健康株C-乳清的总蛋白质,凝胶经考染显色后,用PDQuest7.40图像分析软件进行比较分析,识别差异表达的蛋白质。这些点经过胶内酶切后进行基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)分析获取肽质指纹图谱(PMF),Mascot软件搜索SWISS-PROT和NCBInr数据库鉴定蛋白质。结果:①橡胶树死皮株与健康株C-乳清凝胶的平均蛋白质点数分别为1075±35和1134±27,其平均匹配的点数分别为982±38和1008±22,组内图像匹配率达91.89﹪和88.72﹪。②橡胶树死皮株与健康株C-乳清组间的平均匹配蛋白点数为970±25。利用MALDI-TOF-MS质谱技术对40个差异明显的蛋白点进行分析鉴定,通过查询数据库鉴定了27个蛋白质。本研究建立了分辨率高且重复性较好的橡胶树死皮株与 健康株胶乳C-乳清的双向凝胶电泳图谱,并应用质谱技术鉴定了其中表达差异的蛋白质点,这些差异表达的蛋白质可能参与了死皮发生和发展的过程。  相似文献

Plantago lanceolata L. and Trifolium repens L. were grown for 16 wk in ambient (360 μmol mol−1) and elevated (610 μmol mol−1) atmospheric CO2. Plants were inoculated with the arbuscular mycorrhizal (AM) fungus Glomus mosseae (Nicol. & Gerd.) Gerdemann & Trappe and given a phosphorus supply in the form of bonemeal, which would not be immediately available to the plants. Seven sequential harvests were taken to determine whether the effect of elevated CO2 on mycorrhizal colonization was independent of the effect of CO2 on plant growth. Plant growth analysis showed that both species grew faster in elevated CO2 and that P. lanceolata had increased carbon allocation towards the roots. Elevated CO2 did not affect the percentage of root length colonized (RLC); although total colonized root length was greater, when plant size was taken into account this effect disappeared. This finding was also true for root length colonized by arbuscules. No CO2 effect was found on hyphal density (colonization intensity) in roots. The P content of plants was increased at elevated CO2, although both shoot and root tissue P concentration were unchanged. This was again as a result of bigger plants at elevated CO2. Phosphorus inflow was unaffected by CO2 concentrations. It is concluded that there is no direct permanent effect of elevated CO2 on mycorrhizal functioning, as internal mycorrhizal development and the mycorrhizal P uptake mechanism are unaffected. The importance of sequential harvests in experiments is discussed. The direction for future research is highlighted, especially in relation to C storage in the soil.  相似文献

 Well-supplied and K-deficient 4-year-old clonal Norway spruce trees were exposed to combinations of two levels of ozone (20 and 80 nl l –  1 O3) and carbon dioxide (350 and 750 μl l –  1 CO2) to study the effects of possible future climate factors on gas exchange characteristics. The fumigation was performed in environmental chambers for a complete growing season. After the exposure, plants were cultivated outdoors to investigate possible recovery and delayed effects. During the exposure 1-year-old needles responded to the 80 nl l –  1 O3 treatment by a sharp but transient decrease of both apparent carboxylation efficiency (CE) and maximum photosynthetic capacity (A2500). Elevated CO2 also reduced CE and A2500. The effect became stronger in the course of the exposure and was accompanied by decreases of N and P as well as chlorophyll contents. In case of K deficiency, the acclimation response of current-year needles was even more pronounced reflecting lower sink capacities for carbon metabolites. The joint application of elevated O3 and CO2 resulted in the lowest values of gas exchange parameters and chlorophyll contents. At the beginning of the growing season after the exposure and under outdoor conditions, all these treatment effects disappeared in the needles which had developed during the fumigation. In the course of the development of the new flush, however, the well-supplied 1-year-old needles which had been treated with 80 nl l –  1 O3 and 350 μl l –  1 CO2 in the year before, exhibited a sharp decline of CE and A2500. Simultaneously, chlorotic mottle and bands developed. These delayed symptoms are discussed in the context of the previously published “memory” effect for O3 (Sandermann et al. 1989). The objective of the work was to determine differences in plant response to geographic isolates of a vesicular-arbuscular mycorrhizal (VAM) fungus, and to demonstrate the need for such determinations in the selection of desirable host-endophyte combinations for practical applications. Soybean ( Glycine max (L.) Merr.) plants were inoculated with Bradyrhizobium japonicum and isolates of the VAM-fungal morphospecies Glomus mosseae (Nicol. & Gerd.) Gerd. and Trappe, collected from an arid (AR), semiarid (SA) or mesic (ME) area. Inoculum potentials of the VAM-fungal isolates were determined and the inocula equalized, achieving the same level of root colonization (41%, P >0.05) at harvest (50 days). Plants of the three VAM treatments (AR, SA and ME) were evaluated against von VAM controls. Significant differences in plant response to colonization were found in dry mass, leaf K, N and P concentrations, and in root/shoot, nodule/root, root length/leaf area and root length/root mass ratios. The differences were most pronounced and consistent between the AR and all other treatments. Photosynthesis and nodule activity were higher ( P <0.05) in all VAM treatments, but only the AR plants had higher ( P <0.05) photosynthetic water-use efficiency than the controls. Nodule activity, evaluated by H2 evolution and C2H2 reduction, differed significantly between treatments. The results are discussed in terms of nutritional and non-nutritional effects of VAM colonization on the development and physiology of the tripartite soybean association in the light of intraspecific variability within the fungal endophyte.  相似文献

The purposes of this investigation were to quantify the noise component of child breath-by-breath data, investigate the major determinants of the breath-to-breath noise, and to characterise the noise statistically. Twenty-four healthy children (12 males and 12 females) of mean (SD) age 13.1 (0.3) years completed 25 min of steady-state cycle ergometry at an exercise intensity of 50 W. Ventilatory and gas exchange variables were computed breath-by-breath. The mean (SD) oxygen consumption (VO2) ranged from 0.72 (0.16) to 0.92 (0.26) l x min(-1); mean (SD) carbon dioxide production (VCO2) ranged from 0.67 (0.20) l x min(-1) to 0.85 (0.16) l x min(-1); and mean (SD) minute ventilation ranged from 17.81 (3.54) l x min(-1) to 24.97 (5.63) l x min(-1). The majority of the breath-to-breath noise distributions differed significantly from Gaussian distributions with equivalent mean and SD parameters. The values of the normalised autocorrelation functions indicated a negligible breath-to-breath correlation. Tidal volume accounted for the majority of the VO2 (43%) and VCO2 (49%) variance. The breath-to-breath noise can be explained in terms of variations in the breathing pattern, although the large noise magnitude, together with the relatively small attainable response amplitudes in children reduces the certainty with which ventilatory and gas exchange kinetics can be measured.  相似文献
