The effects of the antimicrobial tylosin on a methanogenic microbial community were studied in a glucose‐fed laboratory‐scale anaerobic sequencing batch reactor (ASBR) exposed to stepwise increases of tylosin (0, 1.67, and 167 mg/L). The microbial community structure was determined using quantitative fluorescence in situ hybridization (FISH) and phylogenetic analyses of bacterial 16S ribosomal RNA (rRNA) gene clone libraries of biomass samples. During the periods without tylosin addition and with an influent tylosin concentration of 1.67 mg/L, 16S rRNA gene sequences related to Syntrophobacter were detected and the relative abundance of Methanosaeta species was high. During the highest tylosin dose of 167 mg/L, 16S rRNA gene sequences related to Syntrophobacter species were not detected and the relative abundance of Methanosaeta decreased considerably. Throughout the experimental period, Propionibacteriaceae and high GC Gram‐positive bacteria were present, based on 16S rRNA gene sequences and FISH analyses, respectively. The accumulation of propionate and subsequent reactor failure after long‐term exposure to tylosin are attributed to the direct inhibition of propionate‐oxidizing syntrophic bacteria closely related to Syntrophobacter and the indirect inhibition of Methanosaeta by high propionate concentrations and low pH. The pH and hydraulic retention time (HRT) of an anaerobic sequencing batch reactor (ASBR) were varied to optimize the conversion of carbohydrate-rich synthetic wastewater into bio-hydrogen. A full factorial design using evolutionary operation (EVOP) was used to determine the effect of the factors and to find the optimum condition of each factor required for high hydrogen production rate. Experimental results from 20 runs indicate that a maximum hydrogen production rate of 4,460-5,540 mL/L/day under the volumetric organic loading rate (VOLR) of 75 g-COD/L/day obtained at an observed design point of HRT = 8 h and pH = 5.7. The hydrogen production rate was strongly dependent on the HRT, and the effect was statistically significant (P < 0.05). However, no significant effect (P > 0.05) was found for the pH on the hydrogen production rate. When the ASBR conditions were set for a maximum hydrogen production rate, the hydrogen production yield and specific hydrogen production rate were 60-74 mL/g-COD and 330-360 mL/g-VSS/day, respectively. The hydrogen composition was 43-51%, and no methanogenesis was observed. Acetate, propionate, butyrate, valerate, caproate, and ethanol were major liquid intermediate metabolites during runs of this ASBR. The dominant fermentative types were butyrate-acetate or ethanol-acetate, representing the typical anaerobic pathway of Clostridium species. This hydrogen-producing ASBR had a higher hydrogen production rate, compared with that produced using continuous-flow stirred tank reactors (CSTRs). This study suggests that the hydrogen-producing ASBR is a promising bio-system for prolonged and stable hydrogen production.  相似文献

The possibility of converting hydrogen to methane and simultaneous upgrading of biogas was investigated in both batch tests and fully mixed biogas reactor, simultaneously fed with manure and hydrogen. Batch experiments showed that hydrogen could be converted to methane by hydrogenotrophic methanogenesis with conversion of more than 90% of the consumed hydrogen to methane. The hydrogen consumption rates were affected by both (hydrogen partial pressure) and mixing intensity. Inhibition of propionate and butyrate degradation by hydrogen (1 atm) was only observed under high mixing intensity (shaking speed 300 rpm). Continuous addition of hydrogen (flow rate of 28.6 mL/(L/h)) to an anaerobic reactor fed with manure, showed that more than 80% of the hydrogen was utilized. The propionate and butyrate level in the reactor was not significantly affected by the hydrogen addition. The methane production rate of the reactor with H2 addition was 22% higher, compared to the control reactor only fed with manure. The CO2 content in the produced biogas was only 15%, while it was 38% in the control reactor. However, the addition of hydrogen resulted in increase of pH (from 8.0 to 8.3) due to the consumption of bicarbonate, which subsequently caused slight inhibition of methanogenesis. Biotechnol. Treatment of brewery slurry in a thermophilic anaerobic sequencing batch reactor (ASBR) was studied using conventional fully mixed semi-continuous digestion as a control. The process phases were adapted to fit the brewery slurry discharge schedule. ASBR experiments were conducted under different organic loading rates (OLR) from 3.23 to 8.57 kg of COD/m(3)day of reactor and control was conducted with OLR of 3.0 kg of COD/m(3)day. The ASBR COD degradation efficiency was from 79.6% to 88.9%, control experiment efficiency was 65%. ASBR VSS removal efficiency was from 78.5% to 90.5%, control experiment efficiency was 54%. The ASBR methane production yield was from 371 to 418 L/kg COD inserted, control experiment methane yield was 248 L/kg COD inserted. The ASBR process was superior to conventional fully mixed digestion, and is fully adaptable to brewery slurry discharge, needs no additional collection and settling pools and experiences no solids settling problems.  相似文献

Dark fermentative hydrogen production from glucose by a thermophilic culture (33HL), enriched from an Icelandic hot spring sediment sample, was studied in two continuous-flow, completely stirred tank reactors (CSTR1, CSTR2) and in one semi-continuous, anaerobic sequencing batch reactor (ASBR) at 58 degrees C. The 33HL produced H2 yield (HY) of up to 3.2 mol-H2/mol-glucose along with acetate in batch assay. In the CSTR1 with 33HL inoculum, H2 production was unstable. In the ASBR, maintained with 33HL, the H2 production enhanced after the addition of 6 mg/L of FeSO4 x H2O resulting in HY up to 2.51 mol-H2/mol-glucose (H2 production rate (HPR) of 7.85 mmol/h/L). The H2 production increase was associated with an increase in butyrate production. In the CSTR2, with ASBR inoculum and FeSO4 supplementation, stable, high-rate H2 production was obtained with HPR up to 45.8 mmol/h/L (1.1 L/h/L) and HY of 1.54 mol-H2/mol-glucose. The 33HL batch enrichment was dominated by bacterial strains closely affiliated with Thermobrachium celere (99.8-100%). T. celere affiliated strains, however, did not thrive in the three open system bioreactors. Instead, Thermoanaerobacterium aotearoense (98.5-99.6%) affiliated strains, producing H2 along with butyrate and acetate, dominated the reactor cultures. This culture had higher H2 production efficiency (HY and specific HPR) than reported for mesophilic mixed cultures. Further, the thermophilic culture readily formed granules in CSTR and ASBR systems. In summary, the thermophilic culture as characterized by high H2 production efficiency and ready granulation is considered very promising for H2 fermentation from carbohydrates.  相似文献

Granular sludge from an upflow anaerobic sludge blanket reactor treating synthetic waste water containing a mixture of volatile fatty acids and nitrate showed a removal efficiency of nearly 100% for both nitrogen and carbon. This activity was achieved by a combined process of denitrification and methanogenesis under conditions of surplus carbon. Under batch conditions the two processes proceeded clearly separated in time with first denitrification dominating and excluding methanogenesis. However, as soon as nitrate was depleted, methane production was initiated, showing that the inhibition of methanogenesis by nitrate was reversible. Of the volatile fatty acids supplied to the reactor, i.e. acetate, propionate, and butyrate, the denitrifying population clearly preferred butyrate and propionate even though acetate could also be metabolized. Consequently, growth of syntrophic volatile fatty acid degraders was suppressed by the denitrifiers in cases of low C:N ratios in the medium, leaving acetate as the major substrate for methanogenesis.Abbreviations UASB upflow anaerobic sludge blanket - COD chemical oxygen demand - VFA volatile fatty  相似文献

The goal of this study was to examine the effect of feeding strategy on the capability for treatment and the stability of an anaerobic sequencing batch reactor (ASBR) under increasing organic loading. The lab-scale ASBR systems were operated at 35 degrees C using synthetic organic wastewater under both batch and fed-batch operational modes with different feed to cycle time (F:C) ratios. Experimental studies were conducted over a wide range of volumetric organic loading rates (VOLRs) (1.524 g COD/l/d) by varying the hydraulic retention time (HRT) (1.25, 2.5, and 5d) and the feed wastewater's COD (3750-30,000 mg/l). With an F:C ratio greater than or equal to 0.42, the fed-batch mode operation showed higher system efficiency in COD removal, volumetric methane production rate (VMPR), and specific methane production rate (SMPR) as compared to those in the batch mode with identical VOLR and HRT. In the fed-batch mode, the COD removals reached 86-95% with VOLR up to 12 g COD/l/d. The maximums for VMPR of 3.17 l CH4/l/d and for SMPR of 1.63 g CH4-COD/g VSS/d were achieved with a VOLR of 12 g COD/l/d at HRTs of 2.5 and 1.25 d, respectively. The fed-batch operation presented a lower concentration of volatile fatty acids (VFAs) than those in the batch operation. A lower concentration of VFAs confirmed the stability and efficiency of the fed-batch mode operation. The specific methanogenic activity (SMA) analysis showed that the VFA-degrading activity of the biomass in the fed-batch mode was higher for acetate and butyrate, and lower for propionate. Determined biomass yield and bacterial decay coefficients in the fed-batch operational mode were 0.05 g VSS/g COD rem and 0.001 d(-1), respectively.  相似文献

The effect of vinyl acetate in acetoclastic methanogenesis   总被引:1,自引:0,他引:1
The influence of vinyl acetate (VA) in the methanogenesis was evaluated, by using an upflow anaerobic sludge blanket reactor of 1.5L. The reactor was operated at 33.5 g/L volatile suspended solids to 30±2 °C, a hydraulic residence time of 1 day, an organic loading rate of 1 kgCOD/m3/d of two different mixtures of VA and glucose. The VA was methanized to 81% when its proportion was of 10% into reactor loading rate, when VA proportion increased to 25%, the methane production rate decreased to 62% and the acetate production rate increased almost 8 times. These results indicated that VA was only hydrolyzed and glucose was not used as a co-substrate. The effect of glucose on VA methanogenic degradation was evaluated through batch reactors of 60 mL, concluding that the glucose supported the methanogenesis without favoring the VA elimination. On the other hand, the results of the sludge from the reactor in the presence of VA demonstrated that VA caused an irreversibly inhibition of acetoclastic methanogenesis when the anaerobic sludge was exposed to this compound.  相似文献

Inhibitory effects of nitrogen oxides on a mixed methanogenic culture   总被引:3,自引:0,他引:3
The effect of nitrate, nitrite, nitric oxide (NO), and nitrous oxide on a mixed, mesophilic (35 degrees C) methanogenic culture was investigated. Short-term inhibition assays were conducted at a concentration range of 10-350 mg N/L nitrate, 17-500 mg N/L nitrite, 0.02-0.8 mg N/L aqueous NO, and 19-191 mg N/L aqueous nitrous oxide. Simultaneous methane production and N-oxide reduction was observed in 10 and 30 mg N/L nitrate and 0.02 mg N/L aqueous NO-amended cultures. However, addition of N-oxide resulted in immediate cessation of methanogenesis in all other cultures. Methanogenesis completely recovered subsequent to the complete reduction of N-oxides to nitrogen gas in all N-oxide-amended cultures, with the exception of the 500 mg N/L nitrite- and 0.8 mg N/L aqueous NO-amended cultures. Partial recovery of methanogenesis was observed in the 500 mg N/L nitrite-amended culture in contrast to complete inhibition of methanogenesis in the 0.8 mg N/L aqueous NO-amended culture. Accumulation of volatile fatty acids was observed in both cultures at the end of the incubation period. Among all N-oxides, NO exerted the most and nitrate exerted the least inhibitory effect on the fermentative/methanogenic consortia. The effect of multiple additions of nitrate (300 mg N/L) on the same methanogenic culture was also investigated. Long-term exposure of the methanogenic culture to nitrate resulted in an increase of N-oxide reduction rates and decrease of methane production rates, which was attributed to changes in the microbial community structure due to nitrate addition.  相似文献

The effects of different volatile fatty acids (VFA, formate, acetate, propionate and butyrate), ammonium (NH (4) (+)) and agitation on methane (CH(4)) production were determined in 120-mL serum bottles. We showed that the addition of formate did not lead to an inhibition of methanogenesis until a concentration of 120 mmol/L. A complete inhibition of methanogenesis was detected in variants containing 360 mmol/L formate or propionate until day 3 but the production started afterwards within next 2 days. This might indicate a kind of adaptation to the higher volatile fatty acid concentrations. Increasing NH (4) (+) concentrations led to higher initial CH(4) production, with an optimum at 120 mmol/L. The addition of 720 mmol/L NH (4) (+) led to a complete inhibition until day 3; subsequently, CH(4) production started again on day 5 though it was still significantly lower compared to the other variants. Finally, also the speed of agitation showed significant effects on methanogenesis. The CH(4) production from complex carbon sources was most favourable at a moderate agitation of 150 rpm of the lab-scale serum bottles. A lower or higher speed brought about a distinct reduction of CH(4) production.  相似文献

Tylosin-producing Streptomyces fradiae was cultured on a synthetic medium with a high glutamate-glucose ratio. Tylosin batch fermentations with this medium were characterized by a high initial specific production rate of tylosin (q(tylosin), mg/g h) that decreased as the fermentation progressed. Continuous feeding of glutamate, glucose, and methyloleate at a constant feed rate initiated during the period of high q(tylosin) had been shown to produce some increase in tylosin productivity. By using a cyclic feeding strategy, it was possible to increase tylosin productivity further. Tylosin fed-batch fermentations with glutamate and glucose being fed to the culture in cyclic square-wave profiles with methyloleate in excess showed several-fold increase in final q(tylosin) and tylosin titers. By varying cycle amplitudes and period of the substrates, it was found that maximum tylosin productivity occurred when the glutamate cycle amplitude was 600 mg/L and that of glucose was 42.5 mg/L per cycle period of 24 h. With these cycle amplitudes of glutamate and glucose, the tylosin cyclic fed-batch culture also showed high cellular uptake of methyloleate. Decreasing or increasing glucose cycle amplitude at fixed glutamate amplitude lowered tylosin production, and no further stimulation of tylosin synthesis was observed when alpha-ketoglutarate was supplemented to the cyclic substrate feeds. Under optimum cyclic conditions it was possible to maintain linear tylosin accretion and a constant value of q(tylosin) up to 240 h.  相似文献

The use of a down-flow fluidized bed (DFFB) reactor for the treatment of a sulfate-rich synthetic wastewater was investigated to obtain insight into the outcome of sulfate reduction in a biofilm attached to a plastic support under a down-flow regime. Fine low-density polyethylene particles were used as support for developing a biofilm within the reactor. The reactor treated a volatile fatty acids mixture of acetate or lactate, propionate, and butyrate at different chemical oxygen demand (COD) to sulfate ratios ranging from 1.67 to 0.67 (g/g). Organic loading rate changed from 2.5 to 5 g COD/L x day and sulfate loading rate increased from 1.5 to 7.3 g SO(4) (2-)/L x day. At the beginning of continuous operation, methanogenesis was the predominant process; however, after 187 days, sulfate reduction became the main ongoing biological process. After 369 days, a COD removal of 93% and a sulfate removal of 75% were reached. Total sulfide concentrations in the reactor ranged from 105, when the reactor was mainly methanogenic, to around 1,215 mg/L at the end of the experiment. The high sulfide concentrations did not affect the performance of the reactor. Results demonstrated that the configuration of the DFFB reactor was suitable for the anaerobic treatment of sulfate-rich wastewater.  相似文献

Brewery wastewater was treated in a pilot-scale anaerobic sequencing batch reactor (ASBR) in which a floating cover(@) was employed. Long time experiments showed that the reactor worked stably and effectively for COD removal and gas production. When the organic loading rate was controlled between 1.5 kg COD/m3 d and 5.0 kg COD/m(3)d, and hydraulic retention time one day, COD removal efficiency could reach more than 90%. Sludge granulation was achieved in the reactor in approximately 60 days, which is much less than the granulation time ever reported. In addition, high specific methanogenic activity (SMA) for formate was observed. The study suggests that the ASBR technology is a potential alternative for brewery wastewater treatment.  相似文献

The effect of oleate on the anaerobic digestion process was investigated. Two thermophilic continuously stirred tank reactors (CSTR) were fed with mixtures of cattle and pig manure with different total solid (TS) and volatile solid (VS) content. The reactors were subjected to increasing pulses of oleate. Following pulses of 0.5 and 1.0 g oleate/L, the most distinct increase in volatile fatty acid (VFA) concentrations were observed in the reactor with the lowest TS/VS content. This suggests a higher adsorption of oleate on the surfaces of biofibers in the reactor with the highest TS/VS and a less pronounced inhibition of the anaerobic digestion process. On the other hand, addition of 2.0 g oleate/L severely inhibited the process in both reactors, and a significant increase in all VFA concentrations combined with an immediate drop in methane production was noticed. However, 20 days after the reactors had been exposed to oleate both reactors showed a lower VFA concentration along with a higher methane production than before the pulses. This indicates that oleate had a stimulating effect on the overall process. The improved acetogenic and methanogenic activity in the reactors was confirmed in batch activity tests. In addition to this, toxicity tests revealed that the oleate pulses induced an increase in the tolerance level of acetotrophic methanogens towards oleate. When evaluating the usability of different process parameters (i.e., VFA and methane production) as indicators of process recovery, following the inhibition by oleate, propionate was found to be most suitable.  相似文献

Anaerobic digestion of animal waste is a technically viable process for the abatement of adverse environmental impacts caused by animal wastes; however, widespread acceptance has been plagued by poor economics. This situation is dismal if the technology is adapted for treating low strength animal slurries because of large digester-volume requirements and a corresponding high energy input. A possible technology to address these constraints is the anaerobic sequencing batch reactor (ASBR). The ASBR technology has demonstrated remarkable potential to improve the economics of treating dilute animal waste effluents. This paper presents preliminary data on the effects of temperature and frequency-cycle on the operation of an ASBR at a fixed hydraulic retention time (HRT). The results suggest that within the parameter range under consideration, temperature did not affect the biogas yield significantly, however, higher cycle-frequency had a negative effect. The biogas quality (%CH(4)) was not significantly affected by temperature nor by the cycle-frequency. The operating principle of the ASBR follows four phases: feed, react, settle, and decant in a cyclic mode. To improve the biogas production in an ASBR, one long react-phase was preferable compared to three shorter react-phases. Treatment of dilute manure slurries in an ASBR at 20 degrees C was more effective than at 35 degrees C; similarly more bio-stable effluents were obtained at low cycle-frequency. The treatment of dilute swine slurries in an ASBR at the lower temperature (20 degrees C) and lower cycle-frequency is, therefore, recommended for the bio-stabilization of dilute swine wastewaters. The results also indicate that significantly higher VFA degradation occurred at 20 degrees C than at 35 degrees C, suggesting that the treatment of dilute swine slurries in ASBRs for odor control might be more favorable at the lower than at the higher temperatures examined in this study. Volatile fatty acid reduction at the two reactor temperatures and cycle-frequencies, from a high of 639+/-75 mg/L to a low of 92+/-23 mg/L, greatly reduced the odor and the odor-generation potential in post-treatment storage. The nutrients (both N and P) in the waste influent were conserved in the effluents.  相似文献

The effects of terramycin (OTC) and monensin on methanogenesis were studied using batch activity tests and continuous-flow reactor experiments. Methanogenesis was not adversely affected by OTC but was inhibited by monesin in downflow stationary fixed film reactors at concentrations commonly found in manure. At relatively higher concentrations, monensin stopped methane production.  相似文献

Recycle batch fermentations using immobilized cells of Propionibacterium acidipropionici were studied for propionate production from whey permeate, de-lactose whey permeate, and acid whey. Cells were immobilized in a spirally wound fibrous sheet packed in a 0.5-L column reactor, which was connected to a 5-L stirred tank batch fermentor with recirculation. The immobilized cells bioreactor served as a breeder for these recycle batch fermentations. High fermentation rates and conversions were obtained with these whey media without nutrient supplementation. It took approximately 55 h to ferment whey permeate containing approximately 45 g/L lactose to approximately 20 g/L propionic acid. Higher propionate concentrations can be produced with various concentrated whey media containing more lactose. The highest propionic acid concentration obtained with the recycle batch reactor was 65 g/L, which is much higher than the normal maximum concentration of 35 to 45 g/L reported in the literature. The volumetric productivity ranged from 0.22 g/L . h to 0.47 g/L . h, depending on the propionate concentration and whey medium used. The corresponding specific cell productivity was 0.033 to 0.07 g/L . g cell. The productivity increased to 0.68 g/L . h when whey permeate was supplemented with 1% (w/v) yeast extract. Compared with conventional batch fermentation, the recycle batch fermentation with the immobilized cell bioreactor allows faster fermentation, produces a higher concentration of product, and can be run continually without significant downtime. The process also produced similar fermentation results with nonsterile whey media. (c) 1995 John Wiley & Sons, Inc.  相似文献

A Panax notoginseng cell culture was successfully scaled up from shake flask to 1.0-L bubble column reactor and concentric-tube airlift reactor. High-density bioreactor batch cultivation was carried out using a modified MS medium. The maximum cell density in batch cultures reached 20.1, 21.0 and 24.1 g/L in the shake flask, bubble column and airlift reactors, respectively, and their corresponding biomass productivity was 950, 1140 and 1350 mg/(L x d) for each. The productivity of ginseng saponin was 70, 96 and 99 mg/(L x d) in the flask, bubble column and airlift reactors, respectively; and the polysaccharide productivity reached 104, 119 and 151 mg/(L x d) for each. Furthermore, a fed-batch cultivation strategy was developed on the basis of specific oxygen uptake rate (SOUR), i.e., sucrose feeding before a sharp decrease of SOUR, and the highest cell density of 29.7 g/L was successfully achieved in the airlift bioreactor on day 17 with a very high biomass productivity of 1520 mg/(L x d). The concentrations of ginseng saponin and polysaccharide reached about 2.1 and 3.0 g/L, respectively, and their productivity was 106 (saponin) and 158 mg/(L x d) (polysaccharide). This work successfully demonstrated the high-density bioreactor cultivation of P. notoginseng cells in pneumatically agitated bioreactors and the reproduction of the shake flask culture results in bioreactors. The cell density, biomass productivity, production titer and productivity of both ginseng saponin and polysaccharide obtained here were the highest that have been reported on a reactor scale for all the ginseng species.  相似文献

The production of tylosin and related compounds by Streptomyces fradiae NRRL 2702 was studied in batch and chemostat cultures using a soluble synthetic medium. In batch culture, a trophophase–idiophase kinetic pattern was observed with tylosin, macrocin, and relomycin accumulating in the idiophase. When the organism was grown in chemostat culture, the specific rate of production of tylosin and related compounds (qtylosin) was found to be a function of the growth rate. The maximum value of (qtylosin) was observed when D = 0.017 hr?1. At this growth rate only tylosin and relomycin accumulated in the medium. By varying the concentration of glucose in the ingoing medium it was possible to study the effects of glucose on tylosin synthesis in chemostat cultures. At a growth rate of 0.017 hr?1, the maximum value of qtylosin was 0.71 mg tylosin/g dry weight (DW)/hr when the glucose uptake rate was 7 mg glucose/g DW-hr. This value of qtylosin was 40% greater than the maximum qtylosin observed in batch culture. When glycerol was substituted for glucose in the medium, it was possible in chemostat culutures to get values of qtylosin approximately 20% greater than those obtained with glucose at the same uptake rate. By varying the concentration of sodium glutamate in the ingoing medium it was possible to show that increasing the specific uptake rate of sodium glutamate increased the values of qtylosin obtained. Similar chemostat experiments where the inorganic phosphate concentration in the ingoing medium was varied showed that increased the uptake of phosphate decreased the values of qtylosin obtained. Also increasing the uptake rate of phosphate increased the relomycin-to-tylosin ratio. By taking into consideration the suppressing effects of glucose and the stimulating effects of sodium glutamate on tylosin synthesis, it was possible to formulate a medium that resulted in a value of qtylosin of 1.1 mg/g/hr being obtained at a growth rate of 0.03 hr?1. Batch fermentations with this medium did not follow a trophophase–idiophase kinetic pattern, but instead tylosin was actively synthesized during a period of rapid mycelial growth.  相似文献
