From a monthly survey over a ten-year period (1983–1992) of the Ishitegawa Reservoir, Japan, two multiple regression equations describing the mean chlorophylla (Chl-a) concentration at 0.5 m depth during July, August and September (C S ) and the maximum Chl-a concentration in the photic zone, including its subsurface maximum during this period (C M ), were obtained. The conductivity at 0.5 m depth in May and the mean air temperature during May or during May and June were used as independent variables. These equations were calculated using seven years of data (1983–1988 and 1992). From 1989 to 1991 two impermeable plastic-coated sheet fences were installed across the upper end of the reservoir along the top 5 m of water column. The equations were used to determine the effectiveness of this flow regulation on the summer Chl-a concentration. In 1989 and 1991, when the fences were in place from June to August, no effects were found on the observed C S and C M values. In 1990, when the fences had been in place since October 1989, the observed values were lower than the predicted values.  相似文献

Langmuir-Blodgett (LB) films of two heteroacid phospholipids of biological interest 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoethanolamine (POPE) and 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC), as well as a mixed monolayer with χPOPC = 0.4, were transferred onto mica in order to investigate by a combination of atomic force microscopy (AFM) and force spectroscopy (FS) their height, and particularly, their nanomechanical properties. AFM images of such monolayers extracted at 30 mN m− 1 revealed a smooth and defect-free topography except for the POPE monolayer. Since scratching such soft monolayers in contact mode was proved unsuccessful, their molecular height was measured by means of the width of the jump present in the respective force-extension curves. While for pure POPC a small jump occurs near zero force, for the mixed monolayer with χPOPC = 0.4 the jump occurs at ∼ 800 pN. Widths of ∼ 2 nm could be established for POPC and χPOPC = 0.4, but not for POPE monolayer at this extracting pressure. Such different mechanical stability allowed us to directly measure the threshold area/lipid range value needed to induce mechanical stability to the monolayers. AFM imaging and FS were next applied to get further structural and mechanical insight into the POPE phase transition (LC-LC′) occurring at pressures > 36.5 mN m− 1. This phase transition was intimately related to a sudden decrease in the area/molecule value, resulting in a jump in the force curve occurring at high force (∼ 1.72 nN). FS reveals to be the unique experimental technique able to unveil structural and nanomechanical properties for such soft phospholipid monolayers. The biological implications of the nanomechanical properties of the systems under investigation are discussed considering that the annular phospholipids region of some transmembrane proteins is enriched in POPE.  相似文献

The linear dichroism of single monolayers of lutein, zeaxanthin and a mixture of lutein and synthetic phosphatidylcholine has been measured. The angle of orientation of the carotenoid molecules was found to lie between 45° and 51° relative to the plane of the solid support. Although the adsorbed monolayers were mostly in a monomeric state, microscopic observations, as well as the II-A isotherms, indicated the existence of crystalline islets. The results have been interpreted in connection with Haidinger's polarization brushes.  相似文献

A simple liquid/liquid partitioning procedure was developed which employed aqueous acetonitrile and hexane, for the isolation of chlorophyll and pheophytin. This procedure separated these pigments from other interfering pigmented compounds. The efficacy of this solvent separation method was evaluated using commercially available chlorophylla, b, pheophytina, b, carotenoids, and algal pigment extracts. The recovery efficiencies of this solvent partitioning process for chlorophyll a and pheophytina have been shown to be 95–98% and 93–96%, respectively, furthermore, the chlorophylla fraction was practically free of any contaminating pigments. It appears that a more accurate assessment of chlorophylla and pheophytina can be accomplished employing liquid/liquid partitioning than with the present standard method.  相似文献

Responses of wheat (Triticum aestivum L.) to various concentrations of NaCl and levels of drought were followed. With the rise of NaCl or drought, or NaCl and drought together, growth was retarded. The water content of shoots and roots was mostly unchanged. The chlorophyll and carotenoid contents were increased in plants subjected to salinity or drought or both. Only high salinity level induced a considerable decrease in net photosynthetic rate (PN) and dark respiration rate (RD). PN and RD were decreased with the decrease of soil moisture content. The content of Na+ in the shoots and roots of wheat plants increased with increasing salinity or decreasing soil moisture content or both treatments. Considerable variations in the content of K+, Ca2+ or Mg2+ were induced by the NaCl, drought or both treatments.  相似文献

The efficiency of 9:1 acetone-water, DMSO and boiling 9:1 ethanol-water in extracting chlorophyll and carotenoid pigments from benthic cyanobacterial mats from Antarctica for HPLC (high performance liquid chromatography) analysis was examined. Considerable breakdown of chlorophylla was observed after 5 min extraction in boiling ethanol and 2 h extraction in DMSO. Over 50% of the chlorophylla was degraded to chlorophyllidea and there was substantial loss of carotenoids after a 15 h exposure of ground cells to cold 9:1 acetone-water.Mild sonication of ground mat material in 9:1 acetone-water followed by a 4 h extraction at 4 ° C was found to minimise chlorophylla breakdown and dramatically improved the extraction efficiency of chlorophylla, myxoxanthophyll and -carotene.  相似文献

This paper describes the primary structure of two visual pigment opsins (DfRh1 and DfRh2) in the regionalized compound eye of a dragonfly,Sympetrum frequens. The amino acid sequences were deduced from the nucleotide sequences of cDNAs isolated from a cDNA library of the dragonfly retina. The two opsins both consist of 379 amino acids with 81.3% identity. Analysis of hydropathy indicated that the sequences have seven transmembrane domains like those of previously described opsins. Expression analysis using RT-PCR revealed that DfRh1 was present only in the dorsal region whereas DfRh2 was detected in both the dorsal and the ventral regions of the eye.  相似文献

Blue light stimulates the accumulation of beta-carotene (photocarotenogenesis) in the fungus Phycomyces blakesleeanus. To be effective, light must be given during a defined period of development, which immediately precedes the cessation of mycelial growth and the depletion of the glucose supply. The competence periods for photocarotenogenesis and photomorphogenesis in Phycomyces are the same when they are tested in the same mycelium. Photocarotenogenesis exhibits a two-step dependence on exposure, as if it resulted from the additon of two separate components with different thresholds and amplitudes. The low-exposure component produces a small beta-carotene accumulation, in comparison with that of dark-grown mycelia. The high-exposure component has a threshold of about 100 J· m–2 blue light and produces a large beta-carotene accumulation, which is not saturated at 2·106 J·m–2. Exposure-response curves were obtained at 12 wavelengths from 347 to 567 nm. The action spectra of the two components share general similarities with one another and with those of other Phycomyces photoresponses. The small, but significant differences in the action spectra of the two components imply that the respective photosystems are not identical. Light stimulates the carotene pathway in the carB mutants, which contain the colourless precursor phytoene, but not beta-carotene. Carotenogenesis is not photoinducible in carA mutants, independently of their carotene content. This and other observations on various car mutants indicate that light prevents the normal inhibition of the pathway by the carA and carS gene products. 通过盆栽试验,模拟蓝桉(Eucalyptus globulus)凋落叶在土壤中分解对受体植物菠菜(Spinacia oleracea)生长及光合特性的影响,设置L30(30 g/盆)、L60(60 g/盆)、L90(90 g/盆)和L120(120 g/盆)4个凋落叶施用水平,对照(CK)不加凋落叶。结果表明:(1)在分解30 d时,低量的凋落叶(L30)未抑制菠菜生物量的积累,而中量和高量处理(L60、L90和L120)下其生物量显著降低(P0.05);在凋落叶分解50 d后,蓝桉凋落叶的化感抑制效应已经减弱,仅L120仍然抑制菠菜生物量的积累;(2)一定量的凋落叶(L30和L60)对菠菜叶绿素的合成有促进作用,而凋落叶超过一定量时(L120)转为抑制作用,在凋落叶分解28 d时相对不明显,40 d后逐渐显现;(3)经凋落叶处理的菠菜叶片胞间CO2浓度(Ci)、气孔导度(Gs)及蒸腾速率(Tr)均显著高于对照(CK)(P0.05),而净光合速率(Pn)在L30处理下最高,L60和L90处理与CK差异不显著,仅L120处理显著低于CK(P0.05);(4)光响应与CO2响应曲线的特征参数表观量子效率(AQY)、最大净光合速率(Pmax)、光饱和点(Lsp)、光补偿点(Lcp)、暗呼吸速率(Rd)、Ru BP表观羧化效率(CE)和光呼吸速率(Rp)在L30处理下高于CK,其余凋落叶处理均低于CK,而CO2饱和点(Csp)、CO2补偿点(Ccp)随凋落叶量的增加而升高。综合各指标可见,少量的蓝桉凋落叶分解(L30)可通过增加叶绿素含量,提高对光和CO2的利用能力促进菠菜的光合作用,进而促进其生长,而凋落叶超过一定量时(L90—L120)则起到相反的作用。  相似文献

Loss of a blue-light photoreceptor in the hy4 mutants of Arabidopsis thaliana (L.) Heynh substantially delayed flowering (>100 d to flower vs. 40–50 d), especially with blue light exposure from lamps lacking much red (R) and/or far-red (FR) light. Red night breaks were promotory but flowering was still later for the hy4-101 mutant. However, with exposure to light from FR-rich lamps, flowering of all mutants was early and no different from the wild type. Thus, flowering of Arabidopsis involves a blue-light photoreceptor and other, often more effective photoreceptors. Balance trials were performed to investigate the effects of experimental Eimeria bovis coccidiosis on the metabolism of water, sodium and potassium in calves. Non-infected pair-fed controls and controls fed according to plan were included in the study to allow differentiation between the effects due to infection and due to changes in feed intake. Primary infection with 5 × 104 (group A) or 1 × 105 (group B) oocysts caused mild diarrhoea in three out of four group A calves and mild to severe haemorrhagic diarrhoea in all five group B calves. Losses of sodium and potassium via faeces tended to increase in the infected calves during patency and apparent digestibility (AD) of these minerals was comparably low. In the urine of the infected calves the Na/K-ratio decreased due to a reduced urinary excretion of sodium. The retention (RT) of sodium was particularly high in the calves that had received the higher oocyst dose. Potassium RT did not underlie significant changes during the course of coccidiosis. In the infected calves the plasma level of sodium was reduced transiently while the level of potassium remained fairly stable. Infections with the higher oocyst dose caused a distinct reduction of fluid excretion via urine which compensated for the increased faecal water losses during severe diarrhoea. Reinfection of the group A calves with 1 × 105 oocysts did not cause any significant metabolic impairment. The results of this study indicate that although acute sublethal bovine coccidiosis alters electrolyte and water metabolism the overall balance of electrolytes and water is largely maintained by physiologic adaptation.  相似文献

With more than 50 genes in human, keratins make up a large gene family, but the evolutionary pressure leading to their diversity remains largely unclear. Nevertheless, this diversity offers a means to examine the evolutionary relationships among organisms that express keratins. Here, we report the analysis of keratins expressed in two cyprinid fishes, goldfish and carp, by two-dimensional polyacrylamide gel electrophoresis, complementary keratin blot binding assay, and immunoblotting. We further explore the expression of keratins by immunofluorescence microscopy. Comparison is made with the keratin expression and catalogs of zebrafish and rainbow trout. The keratins among these fishes exhibit a similar range of molecular weights and isoelectric points, with a similar overall pattern on two-dimensional gels. In addition, immunofluorescence microscopy studies of goldfish and carp tissues have revealed the expression of keratins in both epithelial and mesenchymally derived tissues, as reported previously for zebrafish and trout. We conclude that keratin expression is qualitatively similar among these fishes, with goldfish and carp patterns being more similar to each other than to zebrafish, and the cyprinid fishes being more similar to each other than to the salmonid trout. Because of the detected similarity of keratin expression among the cyprinid fishes, we propose that, for certain experiments, they are interchangeable. Although the zebrafish distinguishes itself as being a developmental and genetic/genomic model organism, we have found that the goldfish, in particular, is a more suitable model for both biochemical and histological studies of the cytoskeleton, especially since goldfish cytoskeletal preparations seem to be more resistant to degradation than those from carp or zebrafish. This article presents the distribution and status of bustards, which are listed as first-category protected animals according to the survey results during 1990–2002 in China. The Chinese populations of Otis tarda dybowskii are breeding in south-west of Heilongjiang Province, western Jilin Province, east and middle Inner Mongolia, north Ningxia Hui Autonomous Region, and Gansu Province. A few can winter in the south breeding-range. Its winter-range lies from the south to the Yellow River, as far as to Guizhou Province and Jiangxi Province. Its population number is about 200–300 or 500–800. The Chinese populations of O. t. tarda are breeding in the north and west of Xinjiang. It is unclear about its winter-range, which is presumed to be in south Asia. Recently we found individuals wintering in Chabuchaer and west Xinjiang. The population number is about 2000–3000. The habitat in breeding range includes steppe, grassland, desert grassland, and farmland. The habitat in winter range is the beach of rivers and lakes, meadows, meadow-grassland, and wheatland. The Chinese populations of Chlamydotis undulata macqueeni are breeding in the fringe of the Jungar Basin, the banks of the Ulungur River, Balikun and south Turpan Basin in Xinjiang, west Inner Mongolia, and west Gansu. NortheastMulei in eastern Jungar Basin of Xinjiang is the main breeding-range in the world. The bird uses desert and desert grassland as its habitat. Its winter-range is west Asia and south Asia. Its population number is about 2000. The Chinese populations of Tetrax tetrax are breeding in north Xinjiang, and China is located on the east border of its breeding-range. Its habitat is grassland and semi-desert, and its winter-range lies in south Asia. Its population in China is very scarce. Translocation of [14C]gibberellic acid into antheridial cells of Chara vulgaris L. was investigated in relation to the presence of symplasmic connections between the antheridium and the thallus. It was found that manubria, capitular cells, and antheridial filaments were about three-fold more strongly labelled in young antheridia connected to the thallus by plasmodesmata than in older antheridia in which spontaneous symplasmic isolation had occurred. Plasmolytically induced symplasmic isolation of young antheridia severely diminished the radioactivity of all the cells, down to the level characteristic for spontaneously isolated antheridia. It is concluded that plasmodesmata are the main channel of gibberellin transport into antheridia. Microspore-derived embryos ofBrassica napus cv. Topas that survived salt stress, were obtained after selection against otherwise lethal doses (0.6 and 0.7%) of NaCl after mutagen treatment. A total of 10 salt-surviving embryos were obtained out of a possible 834 000 embryos that were mutagenized. One embryo out of a possible 845 000 obtained from nonmutagenized controls survived but failed to develop into a plant. Visual assessment after salt stress indicated that both the putative salt-tolerant plants and plants from control seeds behaved similarly. However, based on individual characteristics related to salt tolerance, one of the lines (PST-2) accumulated less sodium and retained more potassium, and hence was able to maintain a more favorable Na:K ratio as compared to the controls under salt stress. Also chlorophylla fluorescence induction and quenching signals indicated a high energetic state of the thylakoid membranes in PST-2 under salt stress. The other putative salt-tolerant line (PST-1) had a higher background level of proline that may have enabled it to survive salt stress during initial screening, although its later performance was no better than the control plants.  相似文献

After a pre-treatment with red light, hair formation at the growing tip of the siphonaceous green alga Acetabularia mediterranea Lamour. (= A. acetabulum (L.) Silva) can be induced by a pulse of blue light. Red light is needed again after the inductive blue-light pulse if the new whorl of hairs is to develop within the next 24 h. In order to investigate the role of this red light, the duration of the red irradiation was varied and combined with periods of darkness. The response of hair-whorl formation was dependent on the total amount of red light, regardless of whether the red irradiation followed the blue pulse immediately or was separated from it by a period of darkness. Furthermore, periods of exposure to the photosynthesis inhibitor 3-(3,4-dichlorophenyl)-1-1dimethylurea had a similar effect to darkness. Both observations indicate that this red irradiation acts as a light source for photosynthesis. Whether or not the red light had an additional effect via phytochrome was tested in another type of experiment. The dependence of hair-whorl formation on red-light irradiance in the presence of simultaneous far-red irradiation was determined for the pre-irradiation period as well as for the irradiation period after the blue pulse. In both experiments, far-red light caused a small promotion of hair-whorl formation when low irradiances of red light were used. However, these differences were attributable to a low level of photosynthetic activity (which in fact was measurable) caused by red light reflected in the growth chamber. Furthermore, lowering the proportion of active phytochrome by far-red light would be expected to suppress hair-whorl formation. The influence of far-red light was also tested in a strain of Acetabularia mediterranea that developed hair whorls in about 20% of cells even when kept in complete darkness after the blue-light pulse. Far-red irradiation had no effect. These results strongly indicate that phytochrome is not involved in hair-whorl formation. In this first comprehensive review of negative effects of introduced rodents on insular small mammals, the focal species Rattus rattus, R. norvegicus, R. exulans and Mus musculus are implicated in at least 11 extinctions. Furthermore, removal experiments, eradication campaigns and control programmes provide evidence for negative effects on extant populations. While data are currently insufficient for meaningful generalisation with regard to the most threatening rodents, the most threatened small mammals, and the true extent of the problem, it is interesting that R. rattus is implicated in the majority of impacts. This may be explained by its extensive distribution and ecological plasticity. I conclude with methodological recommendations to guide data collection for impact quantification and the study of impact mechanism. This information should facilitate the prioritisation and justification of eradication campaigns, control programmes and biosecurity measures while ensuring that much-needed attention is paid to the conservation of insular small mammals.  相似文献

Sporangia were accumulated in autotrophically and mixotrophically growing cultures of the Chlamydomonas reinhardtii mutant strain ls entering the stationary phase. Such an accumulation of sporangia was never observed in stationary-phase cultures of wildtype strains. Sporangia harvested from stationary-phase cultures of the mutant strain ls released their zoospores after being resuspended in fresh culture medium. Liberation of zoospores was also observed during fixation of these sporangia with glutaraldehyde and OsO4. Release of zoospores during fixation was prevented by pretreatment with 3 mol·l–1 LiCl. Ultrastructural analyses of these LiCl-pretreated sporangia revealed that they contained abnormal sporangial walls: sporangia containing sporangia and sporangia surrounded by additional multilayered cell walls have been observed. Similar abnormal cell-wall structures were found in sporangia accumulated at the end of the dark period, when the mutant strain ls was grown photoautotrophically under a 12 h light-12 h dark regime with suboptimal aeration. Functional chloroplasts from photoheterotrophic Euglena gracilis can be isolated in isoosmotic gradients of 10–80% Percoll. The chloroplasts display rates of CO2 dependent O2 evolution and CO2 fixation of 30–50 mol mg-1 chlorophyll h-1 or 25–35% of the net O2 evolution by the whole cells and appear to be strikingly different from spinach chloroplasts in several respects: 1. tolerance to high concentration of orthophosphate in the assay medium; 2. inability to support oxaloacetate-dependent O2 evolution; 3. ability to support only low to moderate rates of 3-phosphoglycerate-dependent O2 evolution; 4. an apparent absence of a phosphate translocator in the terms described by Heldt and Rapley ([1970] FEBS Lett. 10, 143–148).University of California, Dept. of Plant and Soil Biology, 108 Hilgard Hall, Berkeley, CA 94720 USA  相似文献
