The use of the moss Physcomitrella patens as a production system for heterologous proteins requires highly standardised culture conditions. For this purpose a semi-continuous photoautotrophic bioreactor culture of Physcomitrella was established. This culture grew stably for 7 weeks in a 5-l bioreactor with a dilution rate of 0.22/day. Enrichment of the air for aeration in a batch bioreactor culture with 2% (v/v) CO2 resulted in an increase in the specific growth rate to 0.57/day. Changes in the pH of the semi-continuous bioreactor culture medium between pH 4.5 and pH 7.0 influenced protonema differentiation; however it did not negatively affect the growth rate compared to uncontrolled pH. The advantages of Physcomitrella as a system for the production of heterologous proteins in plants are discussed.  相似文献

Male sterility mutations are an important tool in the investigation of anther and pollen development and for obtaining hybrid seeds in plant breeding. Cytological analysis of microsporo- and microgameto-genesis in sorghum plants with dominant mutation of male sterility (Mstc) derived from tissue culture has been carried out. Using substitution backcrosses, this mutation was introduced first into the nuclear background of the fertile sorghum line SK-723 and from this line into Volzhskoe-4w (V-4w). The mechanism of Mstc action on anther and pollen development differed in different nuclear backgrounds. In SK-723, phenotypic expression of Mstc began before the beginning of meiosis, which resulted in degeneration of sporogenous tissue in some anthers and in significant disturbances of anther morphology. In microsporocytes that did not degenerate, the frequency of non-specific meiotic abnormalities characteristic of the fertile line SK-723 significantly increased. In addition, in the mutant plants, a number of specific meiotic abnormalities--almost complete desynapsis, and formation of syncytial structures--were observed, apparently the consequence of Mstc action. In mono- or bi-nucleate microspores, degenerative processes resulting in formation of empty or anomalously coloured pollen grains led to almost complete male sterility. In the V-4w nuclear background, changes in anther structure and meiotic disturbances were infrequent. The degenerative processes began at the uni- or binucleate microspore stage and resulted in formation of empty or abnormally coloured pollen grains, and in partial pollen sterility. Thus, the same nuclear male sterility-inducing mutation in different nuclear backgrounds affects different stages of pollen development.  相似文献

The rice photoperiod sensitivity gene Hd3 was originally detected as a heading date-related quantitative trait locus localized on chromosome 6 of rice. High-resolution linkage mapping of Hd3 was performed using a large segregating population derived from advanced backcross progeny between a japonica variety, Nipponbare, and an indica variety, Kasalath. To determine the genotype of Hd3, we employed progeny testing under natural field and short-day conditions. As a result, two tightly linked loci, Hd3a and Hd3b, were identified in the Hd3 region. Nearly-isogenic lines for Hd3a and Hd3b were selected from progeny using marker-assisted selection. The inheritance mode of both Hd3a and Hd3b was found to be additive. Analysis of daylength response in nearly-isogenic lines of Hd3a and Hd3b showed that the Kasalath allele at Hd3a promotes heading under short-day conditions while that at Hd3b causes late heading under long-day and natural field conditions.  相似文献

To investigate the potential of heterologous transposons as a gene tagging system in broccoli (Brassica oleracea var. Italica), we have introduced a Ds-based two-element transposon system. Ds has been cloned into a 35S-SPT excision-marker system, with transposition being driven by an independent 35S-transposase gene construct (Tpase). In three successive selfed generations of plants there was no evidence of germinal-excision events. To overcome this apparent inability to produce B. oleracea plants with germinal excisions, we performed a novel tissue-culture technique to select for fully green shoots from seed with somatic-excision events. The results showed a very high efficiency of regeneration of fully green plants (up to 65%) and molecular analysis indicated that the plants genetically were like plants that contain a germinal-excision event. Further molecular analysis of these plants showed that 69% exhibited reinsertion of Ds back into the plant genome. Sequencing of donor-site footprints after Ds excision, revealed that there is an indication of more-severe deletions and rearrangements when higher concentrations of streptomycin are used in the tissue-culture selection process. Adapted versions of this regeneration technique have a high potential for providing germinal excision-like events in heterologous plants species which show low transposon activity. Alternatively, there is the potential to increase the proportion of 'germinal' plants in earlier generations of more-active plant species.  相似文献

. A multi-approach was used to study different aspects of self-incompatibility (SI) in almond (Prunus dulcis). First, a population of almond cultivars was characterised as to their individual S-allele combination using separation of stylar protein extracts (non-equilibrium pH gradient electrofocusing) followed by staining for RNase activity, which led to the identification of one putative new allele and several new S-allele combinations. Second, a field pollination scheme was designed to study pollen tube progression and to obtain a spatial and temporal characterisation of this reproductive stage in both incompatible and compatible crosses. In addition, an anti-serum was raised against a synthetic peptide designed from an almond S-protein (S8) and used for immunological in situ detection in pistil cryosections. S-RNases were found to accumulate intercellularly in the stylar transmitting tissue as previously reported for other rosaceous species. The results are discussed in view of the evolution of the gametophytic SI system and the models proposed for its mechanism. Gametophyte selection is also proposed as an important intraspecific barrier to fertilisation in this species.  相似文献

We have developed a new procedure for transforming a model legume, Lotus japonicus, that yields transformed plants from transverse cotyledon segments without contamination from the presence of non-transformants that survived the antibiotic selection. L. japonicus was transformed with the HPT gene as a selectable marker and the GUS reporter gene, both of which were driven by cauliflower mosaic virus 35S promoter. The efficacy of selection with hygromycin was tested using the assay of GUS activity in putative transformants. The integration of the GUS gene was also confirmed by polymerase chain reaction analysis of the genomic DNA. The integrated T-DNA was stable and inherited as a dominant trait. This procedure may have potential effectiveness and application in large-scale transformation for insertional mutagenesis or gene tagging.  相似文献

The physical characteristics of wood are not usually taken into account in the genetic improvement of perennial species cultivated for their fruits or seeds. Yet in coffee trees, stem breakage during harvesting, or lodging during their development are major defects in some cultivars. Such defects are linked to certain mechanical properties of the wood, such as stiffness, which can be characterized by the modulus of elasticity (Eb), a parameter used in the theory of mechanics of solids. The longitudinal Eb of several clones of the species Coffea canephora Pierre was assessed. The Eb was measured using a vibration analysis system developed at CIRAD, the "Bing" system. It is based on an analysis of vibrations caused by a blow to the tip of a piece of wood of known geometry. The Eb obtained in this way revealed substantial variability within the C. canephora study sample. Clone classification according to stiffness made it possible to identify the clones most susceptible to lodging. This technological characteristic of wood is highly heritable (0.64), meaning that effective improvement of this trait is feasible. This parameter could also be used as a predictor of other traits of agronomic interest, such as resistance to borer insects.  相似文献

Commercial barley cultivars are difficult to transform because of the lack of an efficient regeneration system. By modifying certain components in the standard culture medium, we have developed a reproducible and more efficient regeneration system. Herbicide-resistant transgenic plants from barley (Hordeum vulgare L. cv. Conlon) were obtained using this medium. Embryo-derived callus was bombarded with pAHC25, which contains the screenable marker gus (#-glucuronidase) and the selectable marker bar (bialaphos resistance gene), both driven by the maize ubiquitin promoter (Ubi1) and followed by the nos terminator. Following bombardment, callus was transferred to callus-induction medium supplemented with 5 mg/l bialaphos for selection. Resistant calli were subsequently transferred to maintenance medium containing 5 mg/l bialaphos for further selection and finally transferred to regeneration medium with 5 mg/l bialaphos. Green shoots that developed on the regeneration medium were transferred to rooting medium containing 3 mg/l bialaphos. Eighty-five transgenic plants were obtained from 13 independent transformation events. Progeny tests showed Mendelian inheritance for the transgenes. This is the first report of the production of large numbers of transgenic plants from a commercial cultivar adapted to Midwestern US barley production.  相似文献

Fusarium vascular wilt (caused by Fusarium oxysporum f. sp. passiflorae) is a limiting factor in the cultivation of yellow passion fruit (Passiflora edulis). Since there is no effective and economically viable control available, development of resistant or at least tolerant cultivars are in demand. A number of procedures have been used for the initial selection of plant genotypes resistant to various fungal pathogens by means of a fungal culture filtrate or purified toxin. In this study, seeds and in vitro-grown plantlets of passion fruit were screened with different concentrations of either Fusarium oxysporum f. sp. passiflorae (FOP) culture filtrate (0, 20, 30, 40 or 50%, v/v) or fusaric acid (0.10, 0.20, 0.30 or 0.40 mM) supplemented in Murashige and Skoog (MS) basal media. Subsequently, selected plants were inoculated with a conidial suspension of FOP to assess correlation between in vivo and in vitro responses. In vitro sensitivity to the selective agents and the resistance response to the pathogen were also compared. Root growth was markedly influenced by FA, culture filtrate, and conidial suspension culture treatments. Observations indicated that roots were primary targets for attack by F. oxysporum. Successful in vitro selection of resistant genotypes by both FA and culture filtrate treatments suggested that this strategy was viable for accelerating breeding of passion fruit for resistance to the Fusarium vascular wilt.  相似文献

. Tufts of multiple shoots were produced from dormant, axillary buds of pineapple in vitro. Tiny shoots (2-5 mm) isolated from the tuft of multiple shoots were encapsulated in 3% sodium alginate prepared using hormone-free Murashige and Skoog's basal medium, Murashige and Skoog's vitamins, 0.56 mM myo-inositol and 0.06 M sucrose. The encapsulated shoots represented synthetic seeds that germinated and formed roots in vitro after subculture onto one of the following media solidified with 0.8% agar: (1) hormone-free Murashige and Skoog's basal medium, Murashige and Skoog's vitamins, 0.56 mM myo-inositol and 0.06 M sucrose (Pin1), (2) Murashige and Skoog's basal medium, Murashige and Skoog's vitamins, 0.56 mM myo-inositol, 0.06 M sucrose, 9.67 µM 1-naphthalene acetic acid, 9.84 µM indole-3-butyric acid and 9.29 µM kinetin (Pin2), and (3) White's basal medium, White's vitamins, 0.56 mM myo-inositol, 0.03 M sucrose, 0.54 µM 1-naphthalene acetic acid and 1.97 µM indole-3-butyric acid (Pin3). Pretreatment of shoots in either liquid Pin3 or Pin4 medium (White's basal medium, White's vitamins, 0.56 mM myo-inositol, 0.03 M sucrose, 10.8 µM 1-naphthalene acetic acid and 39.4 µM indole-3-butryic acid) was required for development into plantlets with roots after culture on either Pin1, Pin2 or Pin3 media. One hundred percent germination of synthetic seeds to plantlets occurred after pretreatment of shoots in liquid Pin4 medium for 12 h followed by culture of synthetic seeds on Pin2 medium. Synthetic seeds stored at 4°C remained viable without sprouting for up to 45 days. Plantlets produced in vitro from synthetic seeds were successfully established in soil. The protocol provides an easy and novel propagation system for pineapple, an otherwise vegetatively propagated fruit crop.  相似文献

Doubled haploid lines, which have high levels of genetic uniformity, are suitable for physiological or genetic studies. We established a transformation system using a doubled haploid line of cabbage. Hypocotyl explants that had been precultured for 3 days on Murashige and Skoog medium containing 50 µM acetosyringone were inoculated and cocultured with Agrobacterium tumefaciens strain LBA4404 (pIG121Hm) for 3 days. Kanamycin-tolerant shoots were regenerated onto shoot induction medium 3 months after Agrobacterium inoculation. The transformation efficiency was about 3% under optimal conditions. The segregation pattern of the self-pollinated transformant that carried one copy of the introduced transgene revealed that the #-glucuronidase gene was inherited in a 3:1 ratio.  相似文献

Many ant partners of tropical ant-plants prune the leaves and shoot tips of other plants growing around their hosts. According to the hypothesis proposed by Davidson et al. (Ecology 69:801-808), this specialized behaviour not only protects the host plants against overgrowth, but it also conveys a direct benefit to the ant colony as it removes contact points to the neighbouring vegetation where invasions of enemy ants could occur. Here we test this hypothesis by comparing pruning intensity in five closely related Crematogaster (subgenus Decacrema) plant-ant species (and one species of Technomyrmex) that differ in their exposure to competition by other ants. Pruning intensity was quantified by measuring the area loss of paper tape pieces wrapped around the stems of Macaranga host plants. All Crematogaster (Decacrema) ants tested but not Technomyrmex sp. pruned, but the intensity of the behaviour varied strongly between and within species. Pruning was significantly weaker in the three tested Crematogaster species inhabiting Macaranga host plants with a slippery, waxy stem surface, which functions as a mechanical barrier protecting the specific ant partners against generalist competitors. Pruning was generally stronger on more densely ant-populated trees. Even though the number of ants per twig length was lower in associations of ants with glaucous Macaranga hosts, only part of the variation of pruning activity could be explained by "ant density". When corrected for ant density, "wax-running" Crematogaster (Decacrema) ants still pruned more weakly than their congeners inhabiting non-glaucous Macaranga hosts. Pruning is obviously most important when an ant-plant is potentially accessible to intruders, but less necessary when the ant colony is isolated by a protective wax barrier. Our results support the hypothesis that "selfish" defence against invasions is the major selective pressure that has led to the development and maintenance of pruning behaviour in weakly competitive plant-ants.  相似文献

Mature seeds of Salix alba L. and Salix matsudana Koidz. are exendospermous and consist of an embryo and a seed coat. Ultrastructural studies show the presence of protein bodies, lipid bodies, chloroplasts, and a nucleus in the cells of most of the embryo tissues. Protein bodies always contain two or more globoid crystals. Energy-dispersive X-ray analysis of globoid crystals revealed the presence of P, K, Mg and Ca as the main constituents in all tissues. The chloroplasts present well-developed grana and, frequently, starch grains in the stroma. In cells of apical meristems, plastid endomembranes are non-organised in grana and deposits of phytoferritin are present in the stroma. Some cells of the subdermal layers of the cotyledons and hypocotyl-radicle axis present a large central vacuole and a narrow peripheral band of cytoplasm within which the protein bodies are scarce. Seeds of the two species studied here have recently been characterised as orthodox with short viability. The present study was carried out in an attempt to advance in the characterisation of these seeds as part of a comprehensive study of Salicaceae seeds.  相似文献

An earliness per se gene, designated Eps-Am1, was mapped in diploid wheat in F2 and single-seed descent mapping populations from the cross between cultivated (DV92) and wild (G3116) Triticum monococcum accessions. A QTL with a peak on RFLP loci Xcdo393 and Xwg241, the most distal markers on the long arm of chromosome 1Am, explained 47% of the variation in heading date (LOD score 8.3). Progeny tests for the two F2:3 families with critical recombination events between Xcdo393 and Xwg241 showed that the gene was distal to Xcdo393 and linked to Xwg241. Progeny tests and replicated experiments with line #3 suggested that Eps-Am1 was distal to Xwg241. This gene showed a large effect on heading date in the controlled environment experiments, and a smaller, but significant, effect under natural conditions. Eps-Am1 showed significant epistatic interactions with photoperiod and vernalization treatments, suggesting that the different classes of genes affecting heading date interact as part of a complex network that controls the timing of flowering induction. Besides its interactions with other genes affecting heading date, Eps-Am1 showed a significant interaction with temperature. The effect of temperature was larger in plants carrying the DV92 allele for late flowering than in those carrying the G3116 allele for early flowering. Average differences in heading date between the experiments performed at 16 °C and 23 °C were approximately 11 days (P < 0.001) for the lines carrying the Eps-Am1 allele for early flowering but approximately 50 days (P < 0.0001) for the lines carrying the allele for late flowering. The large differences in heading time (average 80 days) observed between plants carrying the G3116 and DV92 alleles when grown at 16 °C, suggest that it would be possible to produce very detailed maps for this gene to facilitate its future positional cloning.  相似文献

Filtration was studied in two Arctic clams, Hiatella arctica and Mya sp., collected in Young Sound, Northeast Greenland. Clearance rates were determined as a function of ambient temperature and algal cell concentration, using the clearance method and feeding with a unicellular flagellate. For both species, clearance rates increased with increasing temperature from <у up to 4-8°C. At higher temperatures, filtration ceased and the clams closed their valves. Clearance rates were also determined in temperate specimens of H. arctica collected on the west coast of Sweden. For these specimens, clearance rates increased with increasing temperature from 0 to 18-20°C. When weight-specific clearance rates were compared between the two populations and between species, there were no differences at 1°C. Clearance rates in Arctic H. arctica were maximal at algal cell concentrations corresponding to 2.5-8 µg chlorophyll a l-1. Temperature compensation in Arctic bivalves is discussed and it is concluded that adaptations to constant low temperatures consist of a lower minimum temperature, for active filtration. Low clearance rates due to low temperatures did not seem to limit growth, under the prevailing conditions in Young Sound.  相似文献

Yoldia hyperborea is a deposit-feeding circumpolar protobranch that also inhabits muddy sediments of the cold water boreal system of Conception Bay, Newfoundland, Canada. Little is known about this species, despite its wide distribution and frequent high density in the benthos. The present work deals with oxygen consumption and ammonia excretion under cold ambient conditions. Y. hyperborea showed low basal metabolism [0.051 ml O2 hу·(g dry weight)у, T=у°C] and low ammonia excretion rates [4.212 µg·NH4-N·hу·(g dry weight)у, T=у°C]. Low metabolic activity could prove a useful strategy during periods of low food availability. In addition, Y. hyperborea was able to regulate its O2 consumption rate at very low pO2 levels, which may be advantageous for a species that may experience periods of hypoxia.  相似文献

A highly efficient and reproducible transformation system for orchardgrass (Dactylis glomerata L. cv. Rapido, 2n=42=28) was established using microprojectile bombardment of highly regenerative, green tissues derived from mature seeds. These tissues, induced from embryogenic callus, were bombarded with a mixture of three plasmids containing the hygromycin phosphotransferase (hpt), phosphinothricin acetyltransferase (bar) and #-glucuronidase (uidA; gus) genes. From 147 individual explants bombarded, 11 independent hygromycin-resistant lines (7.5%) were obtained after an 8- to 16-week selection period using 30-50 mg/l hygromycin B. Of the 11 independent lines, ten (91%) were regenerable. The presence and integration of the transgene(s) were assessed using PCR and DNA blot hybridization. Coexpression frequency of the three transgenes (hpt/bar/uidA) in T0 plants was 20%, and of two transgenes, either hpt/bar or hpt/uidA, 45-60%. Due to greenhouse conditions optimized for the growth of other species, T1 seed has not been obtained from these plants. While the inability to analyze progeny plants precludes the conclusive demonstration of stable transformation, the results of all molecular and biochemical analyses of T0 plants are consistent with the production of stably transformed plants. Frequent change in ploidy level was observed in transformed T0 orchardgrass plants. Plants from only three of the ten independent lines analyzed had the normal tetraploid number of chromosomes (2n=42=28), while plants from seven lines (70%) were octaploid (2n=82=56). The octaploid plants had abnormal morphological features, such as narrower, thicker and more upright leaves.  相似文献

A protocol for the in vitro germination and the establishment of tissue cultures of Bulbine caulescens Linn., Kniphofia pumila Kunth, and K. uvaria Hook has been developed. The members of the family Asphodelaceae contain interesting substances with antiplasmodial activity against several strains of Plasmodium falciparum. With respect to species of Bulbine, this is the first time that in vitro germination has been achieved. For seeds of K. pumila and K. uvaria, a facilitated germination was attained by an optimized pretreatment of the seeds. After in vitro germination, root explants of Kniphofia and bulb explants of B. caulescens were successfully transformed into callus cultures. From these, liquid cultures of the two species of Kniphofia were easily produced. Studies on shoot multiplication of Kniphofia in vitro were also performed.  相似文献

Glycinebetaine is an important quaternary ammonium compound that is produced in response to salt and other osmotic stresses in many organisms. Its synthesis requires the catalysis of betaine aldehyde dehydrogenase encoded by BADH gene that converts betaine aldehyde into glycinebetaine in some halotolerant plants. We transformed the BADH gene, cloned from Atriplex hortensis and controlled by two 35S promoters of the cauliflower mosaic virus, into a salt-sensitive tomato cultivar, Bailichun, using Agrobacterium tumefaciens strain LBA4404 carrying a binary vector pBin438, and using a leaf regeneration system. Polymerase chain reaction and Southern hybridization analyses demonstrated that the BADH gene had integrated into the genome of tomato. Transgenic tomato plants showed significantly higher levels of mRNA and BADH enzyme activity than wild-type plants. Observations on rooting development and relative electronic conductivity suggested that the transgenic plants exhibited tolerance to salt stress, with these plants growing normally at salt concentrations up to 120 mM.  相似文献
