日本血吸虫胆碱酯酶的组织化学定位   总被引:4,自引:0,他引:4  
血吸虫的胆碱酯酶(ChE)与血吸虫神经介质的传递、肌肉活动以及与其它物质代谢均有密切关系,也是抗血吸虫药物作用的靶子之一。1959年沈美玲等进行了药物对日本血吸虫乙酰胆碱酯酶(AChE)活力的研究。最近,姚民一等(1981)采用等电聚焦电泳分离出日本血吸虫胆碱酯酶的同功酶。然而有关血吸虫胆碱酯酶的组织化学定位研究主要限于曼氏血吸虫(Pepler,1958;Lewert等,1965;Fripp,1967;Bueding等,1967;Bruckner等,1974;Diconza等,1975)除Bueding简单述及日本血吸虫成虫的AChE外,迄今国内外尚无日本血吸虫CbE的组化资料,而且日本血吸虫的神经系统也未被专门观察。由于ChE的组化定位能细致地显示出血吸虫的神经系统构造,并能进一步提  相似文献

Bacteria associated with the tegument of the tapeworm species Eubothrium rugosum (Cestoda, Pseudophyllidea) parasitizing the intestine of burbot (Lota lota) were studied using transmission and scanning electron microscopy. Three morphological types of the bacteria were revealed. Bacteria of the first type are localized between microtrichia and fit to them closely. Bacteria of the second (gram-positive) and third (gram-negative) types are localized over microtrichia of the tegument and do not attach to the surface. Physiological functions of the bacteria are discussed.  相似文献

Tedesco J. L. and Coggins J. R. 1979. Electron microscopy of the tumulus and origin of associated structures within the tegument of Eubothrium salvelini Schrank, 1790 (Cestoidea: Pseudophyllidea). International Journal for Parasitology10: 275–280. Validity of the tumulus, a new organ recently described on the tegument of Eubothrium salvelini, is confirmed. Spherical, dense inclusions approximately 0.23–0.29 μm were associated with the tumulus and with subtegumental cell bodies. Origin of these inclusions within subtegumental cell bodies and their transport via ducts to the tumulus is described. Inclusions are synthesized within granular endoplasmic reticulum and packaged by Golgi apparatus prior to transport. Inclusions were observed only in association with the tumulus within the tegumental distal cytoplasm.  相似文献

A phylogenetic analysis of tapeworms (Eucestoda) based on complete sequences of the 18S rRNA genes of 43 taxa (including new sequences of 12 species) was carried out, with the emphasis on the groups parasitising teleost fish and reptiles. Spathebothriidea and Trypanorhyncha (the latter group being paraphyletic) appeared as basal groups of the Eucestoda but their position was not stable. The tetrafossate orders (Litobothriidea, Lecanicephalidea, Tetraphyllidea, Proteocephalidea, Nippotaeniidea, Tetrabothriidea and Cyclophyllidea) were well separated from the remaining groups. Results supported polyphyly of the Pseudophyllidea formed by two distinct clades: one with diphyllobothriids (Diphyllobothrium, Schistocephalus, Spirometra and Duthiersia) and another including Abothrium, Probothriocephalus, Eubothrium and Bothriocephalus. The former pseudophyllidean clade formed a separate branch with the Caryophyllidea (Khawia and Hunterella) and Haplobothriidea (Haplobothrium), the latter taxon being closely related to either caryophyllideans or diphyllobothriids in different analyses. Proteocephalideans formed a monophyletic group in all analyses and constituted a clade within the Tetraphyllidea thus rendered paraphyletic. Within the Proteocephalidea, the Acanthotaeniinae (Acanthotaenia from reptiles in Africa) and Gangesiinae (Gangesia and Silurotaenia from silurid fish in the Palearctic Region) were separated from parasites of freshwater fish and mammals. The family Proteocephalidae was found to be paraphyletic due to the placement of a monticelliid species, Monticellia sp., in a clade within the former family. The genus Proteocephalus appeared as an artificial assemblage of unrelated taxa which is congruent with previous molecular analyses.  相似文献

Initial stages of the centralization of the nervous apparatus in the Turbellaria can be traced through a comparison of the structure of the nervous system in various representatives of the class. The most primitive state, found in the Acoela, is predominantly plexiform with a varying number of longitudinal trunks. Three, and in some cases four, longitudinal trunks are found in the Proseriata and Temnocephalida. Commissures appear in the Macrostomida and all higher orders and form an orthogon. Brain shape varies from ring-shaped in the Acoela to bilobed in the Neorhabdocoela. While the nervous system of the Polycladida is peculiar, having numerous lateral trunks and separation of dorsal and ventral parts of the nervous system, the development of the nervous system in Müller's larvae of polyclads shows it is of an orthogonal type comparable to other platyhelminths. Transition to parasitism is accompanied by some progressive transformations in the structure of the nervous system.  相似文献

Flatworm growth patterns are interesting and the guiding principles behind them have long been sought. Epidermal growth factor immunoreactivity (EGF-IR) was detected in a dispersed population of nerve cells of the constantly growing adult gull-tapeworm Diphyllobothrium dendriticum (Cestoda, Pseudophyllidea). The EGF-IR cells are located close to regions characterized by active growth and the presence of mitotically competent cells. As no EGF-IR was observed in the non-growing plerocercoid larva a correlation with the growth rate of the worm and the presence of EGF-IR is suggested.  相似文献

Patterns of tracheation in the abdominal central nervous system and the cerci of Acheta domesticus are described from whole mounts, and light and electron microscopy. The tracheal supply of the ganglia is derived from ventral longitudinal tracheal trunks which have segmental connections to the spiracels. Each abdominal ganglion is served by a single pair of tracheal trunks, except the terminal ganglion, which has two pairs. Within the ganglia, tracheoles occur principally in association with glia-rich areas of the neuropile. We suggest that the respiratory exchange may be concentrated in the cell bodies of neurons and glia. Each cercus has a tracheal supply in paralle with a large air sac which, it is suggested, serves to lighten the cercus, functions as a resonator for sound reception, or facilitates tidal flow of hemolymph and postecdysial expansion of the cercus. No tracheae run continuously between ganglia or between the terminal ganglion and the cerci, and they do not appear to have a potential role as a contact guidance pathway for cercal nerve growth.  相似文献

Rastrognathia macrostoma gen. et sp.n. is described as belonging to Rastrognathiidae fam.n. The ultrastructure is described, especially the nervous and digestive systems and the epidermal specializations. In the epidermis the single cilia belong to the choanocyte-like cell type. An epidermal sensory cell with several cilia is described. The intraepithelial nervous system shows an orthogon pattern. Two ventral nerve trunks unite into a buccal ganglion. The jaws are secreted by the buccal epidermis. The endoderm consists of a single cell type. The food consists of bacteria. The mesodermal elements consist of six main longitudinal muscles.  相似文献

The host specificity and distribution of Eubothrium crassum (Bloch, 1779) and Eubothrium salvelini (Schrank, 1790), morphologically fairly similar pseudophyllidean tapeworms parasitizing salmonid fish, were critically assessed on the basis of morphological and genetic evaluation of extensive material collected from different definitive hosts and geographical regions in Europe. Eubothrium crassum occurs in fish of the genera Salmo, i.e. salmon (S. salar - both freshwater and marine), sea trout (S. trutta trutta), brown trout (S. trutta fario), and lake trout (S. trutta lacustris), and also in Danubian salmon (Hucho hucho) and vendace (Coregonus albula). Eubothrium salvelini parasitizes Arctic char (Salvelinus alpinus) and brook trout (Salvelinus fontinalis) in Europe, and also whitefish (Coregonus wartmanni). Rainbow trout (Oncorhynchus mykiss), which is not a native European fish species, was found to be a suitable definitive host for both Eubothrium species, which may occur simultaneously in the same fish. Previous records of E. crassum in Arctic char and brook trout, and those of E. salvelini in fish of the genus Salmo were most probably misidentifications. Most studies of Eubothrium have involved salmonids from the northern part of Europe, with few records from southern and south-eastern Europe. This study also confirmed the reliability of the morphology of the apical disc for the discrimination of E. crassum and E. salvelini.  相似文献

Abstract. Freshly released eggs from four species of the cestode Eubothrium ( Eubothrium crassum, Eubothrium fragile, Eubothrium rugosum , and Eubothrium salvelini ) were subjected to morphological and morphometric analysis. The eggs of the two freshwater species, E. rugosum and E. salvelini , were ovoid with a lobed embryophore whereas the eggs of the two marine species, E. crassum and E. fragile , were more circular with a smooth embryophore. However, the morphological differences between species were not readily evident to permit their clear distinction from one another. To discriminate species, a forward stepwise linear discriminant analysis, using six of the seven measured metric characters made on the eggs, was used, which gave 100% correct classification of two species, E. rugosum and E. salvelini , and a high proportion of correct classification for E. crassum (98%) and E. fragile (83%). Of the latter two species, one specimen of E . crassum and five specimens of E. fragile were misclassified between the respective groups. The principal characters used in the classification of the species were the width of the egg, the length of the mediolateral hooks, and the width of the oncosphere. To provide more information on the life cycle of each species, the eggs were used in a series of infection trials to identify appropriate intermediate hosts. Experimental infections with freshwater copepods were successful when exposed to the eggs of E . salvelini , partially successful when exposed to the eggs of marine E. crassum with 10% of the copepods becoming infected, but no infections were obtained when the eggs of E. fragile were used.  相似文献

The systematic position of Polygordiidae is still under debate. They have been assigned to various positions among the polychaetes. Recent molecular analyses indicate that they might well be part of a basal radiation in Annelida, suggesting that certain morphological characters could represent primitive character traits adopted from the annelid stem species. To test this hypothesis, an investigation of the muscular and nervous systems by means of immunological staining and confocal laser scanning microscopy and transmission electron microscopy was conducted. With the exception of the brain, the nervous system is entirely basiepidermal and consists of the brain, the esophageal connectives, the subesophageal region, the ventral nerve cord and several smaller longitudinal nerves. These are connected by a considerable number of ring nerves in each segment. The ventral nerve cord is made up of closely apposed longitudinal neurite bundles, a median and two larger lateral ones. Since distinct ganglia are lacking, it represents a medullary cord. The muscular system mainly consists of longitudinal fibers, regularly distributed oblique muscles and strong septa. The longitudinal fibers form a right and a left unit separated along the dorsal midline, each divided into a dorsal and ventral part by the oblique muscles. Anteriorly, the longitudinal musculature passes the brain and terminates in the prostomium. There is no musculature in the palps. In contrast to earlier observations, regularly arranged minute circular muscle fibers are present. Very likely, a basiepithelial and non-ganglionic organization of the ventral nerve cord as well as an orthogonal nervous system represent plesiomorphic characters. Four species of the genus Eubothrium (E. crassum, E. fragile, E. rugosum and E. salvelini) were subjected to morphometric comparison. Discriminant analysis was conducted utilising 17 characters measured on the scolex and strobila of 101 specimens. Univariate statistics were first used to detect features that were useful for separating individual Eubothrium species and two different host populations of E. salvelini. Subsequent multivariate discriminant analysis, combining all the measured variables, made it possible to separate all four species. A comparison of the four taxa revealed that (1) E. fragile is the most distinct species, possessing a much smaller scolex than the other congeners, and its similarity with the other marine species E. crassum is not proven; (2) the two freshwater taxa, E. rugosum and E. salvelini are the most similar; (3) the characters most suitable for species differentiation are the length of the scolex, the width of the apical disc, the width of the neck and its area, the width of eggs and the number of testes; (4) the width of the apical disc was confirmed to be the most stable character at the intraspecific level (within E. salvelini host populations) and is therefore considered to be a trait of the highest discriminative power in the subset of four Eubothrium species.  相似文献

A total of 129 sea trout smolts from 10 locations on the west coast of Ireland were examined for metazoan parasites. Of 11 species identified only four had a prevalence >20%. Two species Hysterothylacium aduncum and Hemiuris communis have a confirmed marine origin; Eubothrium crassum has been shown to exist in both the sea and fresh water. The remaining species were fresh water. Overall the parasite community was dominated by autogenic species with only Diphyllobothrium dendriticum having allogenic transmission. Infracommunity values for mean species richness, and Brillouins index of diversity were low, indicating the presence of an impoverished helminth community with little potential for interspecific interaction. The relationship between marine helminths and sea lice was also examined.  相似文献

Histochemical reaction with glyoxylic acid has revealed a specific fluorescence caused by the presence of indolamine (apparently serotonin) in the nervous system of larva and mature cestode. The internal transcribed spacers (ITS-1 and ITS-2) and 5.8S subunit of the ribosomal RNA gene of Eubothrium crassum and Eubothrium salvelini, cestode parasites of salmonid fish, were sequenced and compared. The lengths and GC contents of the regions sequenced were 1119 bp and 50.1% for E. crassum, and 1117 bp and 50.5% for E. salvelini, respectively. Sequence alignment and comparison of both taxa showed 97.9% similarity. Within the ITS-1 and ITS-2 of both species, the restriction enzymes MspI and Bsh1236I possessed different cleavage sites and could be employed for rapid differentiation of both species of Eubothrium by PCR-RFLP.  相似文献

Using supravital Methylene blue staining, development of elements of the peripheral nervous system was traced in the 1st–5th instar larvae of the locust Locusta migratoria L. Data were obtained on sensory cells of the I and II types in larvae of different instars, as well as on character of their changes in the course of larval development of the insect. The studied elements of the peripheral nervous system have been shown to be essentially transformed in the course of larval ontogenesis. The data are presented about the sensory cells of the proximal parts of the nerve trunks; they indicate that in the course of larvae development the cell number in these trunks near the ganglia of the nerve chain decreases, which might be due to their partial degeneration. With growth of muscle, changes in their sensory innervation take place. Subepithelial nerve plexus in larvae is largely represented in areas with hard and soft cuticles.  相似文献

Peripheral nerves travel to their targets along precise routes, and it is likely that different cues provide guidance at different stages of the journey. In a developing chick limb, the cutaneous nerve fibres follow at first deep mixed nerve trunks, in company with motor axons; they branch from these trunks at predictable points and approach the skin; they then ramify profusely to form a plexus at a precisely defined depth beneath the ectoderm, at exactly the same level as the blood vascular plexus. To analyse the role of signals from the target patch of skin in regulating cutaneous nerve development, we have ablated patches of dorsal wing ectoderm using short-wave ultraviolet irradiation at E4 (embryonic day 4), approximately one day before nerves grow into the limb bud. The irradiated patches remain denuded of ectoderm for more than a week, by which time the cutaneous nerve plexus on the contralateral control side is well developed and can be revealed by whole-mount silver staining. Where the ectoderm has been ablated, no cutaneous nerve plexus forms, and the nerve branches that normally would have diverged from the neighbouring mixed nerve trunk to innervate the missing patch of skin are absent - ab initio, apparently. The routes of the mixed nerve trunks are not affected. Partial ablation of the territory of a cutaneous nerve branch often leads to loss of the whole nerve branch; the intact skin territory thus left vacant is invaded by ramifications from the remaining cutaneous branches, as expected if the normal extent of a cutaneous nerve's territory is regulated by competition. Where there is an ectodermal lesion, cutaneous innervation stops precisely at its boundary, even though the vascular plexus extends for some distance beyond this margin, beneath the denuded surface. The data suggest that the embryonic skin is required firstly to trigger divergence of cutaneous nerve branches from the mixed nerve trunks, and secondly, once the nerve fibres have reached the skin, to supply a trophic cue (probably NGF) encouraging growth of a plexus; at the same time, the embryonic skin generates a signal inhibiting nerves from approaching closer than about 70 microns to the surface.  相似文献

A tendency for increase both of numerical and of ecological factors of infestation in pseudophyllid cestodes is realized due to morphological peculiarities of the structure of their reproductive system. In the species E. rugosum with the closed bursiform uterus and intrauterine development of eggs, there are revealed the same degree of maturity of oocytes in ovary, the lack of sphincter or filtering cell at the base of the ovicapt infundibulum, and the presence of a large vitelline reservoir. By analogy with trematodes, an interconnection is traced between the degree of egg maturation in the uterus and morphology of uterus and vittelicles. The thickening of the egg membrane has been shown to occur not only by deposits of individual vitelline globules at the internal superficial egg layer, but also by binding of lipid droplets excreted from the surrounding parenchyma to the uterine tube along its entire length to the external egg surface. The male reproductive system of Eubothrium rugosum is characterized by the presence of a small cirrus pouch, by the cirrus supplied with microtrichia, which corresponds to the similar vagina structure, and by localization of prostate glands outside the cirrus pouch. Comparison is presented with other ultrastructural peculiarities of organization of the reproductive system in the earlier studied species of pseudophyllid cestodes, Diphyllobothrium latum.  相似文献

On the basis of esterase localization the complete nervous system has been visualized in Olveria indica Thapar & Simha, 1945, a paramphistome from the rumen of cattle, recorded for the first time from the north-east region of India. The basic number of posterior longitudinal nerves (i.e., 3 pairs) is the same as in other paramphistomes; four pairs of anterior longitudinal nerves are present, there being an extra pair of pharyngeal nerves. Conspicuous differences in the innervation patterns were observed, mainly in the innervation of the genital atrial region and acetabulum. The presence of prominent longitudinal connections, joining the anterior and posterior dorsal nerves and the anterior and posterior ventral nerves on either side, is a feature not hitherto found in other paramphistome species. The inbuilt intrinsic cholinergic nervous apparatus of the gastric wall of the cat was studied by using two thiocholine methods for mapping the acetylcholinesterase-positive nerves and nerve cells. A rich distribution of acetylcholinesterase-positive nerves was observed in all layers of the gastric wall, except the superficial half of the lamina propria (with the epithelium), which was completely devoid of acetylcholinesterase activity, and the submucosa, in which a scarce distribution of large nerve fascicles and nerve trunks was observed. Acetylcholinesterase-positive ganglia were observed both in the subserous layer and in the myenteric plexus of Auerbach, whereas none were recognized in the submucous plexus of Meissner. This obviously fits well to the results of some electrophysiological experiments which indicate that the submucous plexus of Meissner includes an important intramural pathway from the extrinsic vagus nerves to the antrum region; so the submucous plexus of Meissner seems to be mainly involved in direct rapid conduction of nerve impulses without integrative activities, like a cable. Certain clear differences exist in the pattern of organization of the cholinergic intrinsic nervous apparatus within the different layers of the gastric wall in the fundic and pyloric regions. These differences seem to correspond quite logically to the different types of motor, secretory and neurohumoral activities of these main regions of the stomach. The activity of the non-specific cholinesterases was localized both in the neural elements and the smooth muscle, as well as in some epithelial cells.  相似文献
