The effect of five constant relative humidity (RH) regimes (12%, 33%, 55%, 75% and 94%) on the preimaginal development and adult longevity and reproduction of Dichochrysa prasina Burmeister was studied at 26 ± 1 °C and a photoperiod of 16:8 (L:D) under laboratory conditions. For the experiments, larvae were fed individually on eggs of the flour moth Ephestia kuehniella (Zeller), whereas adults on a liquid diet consisting of water, yeast hydrolysate, sugar and honey. The experimental insects were maintained individually in sealed plastic containers with saturated water salt solutions at the base to control RH. Preimaginal developmental time from egg to adult for females and males was significantly shorter at 75% and 94% RHs than at other RHs in the range from 12% to 55%. Percentages of adult emergence were similar at all RHs tested and varied from 52.5% to 75.0%. Adult females that had no access to water and were maintained at 12%–75% RH lived on average less than two weeks and laid no eggs, whereas at 94% RH they lived longer and each laid on average 201 eggs. When the females had periodic access to a water source a significant increase in both longevity and egg production occurred and the estimated intrinsic rate of increase was high, irrespective of RH. These results could be useful for the mass-rearing of D. prasina and for better understanding its population dynamics under field conditions.  相似文献

Summary An efficient rapid and large-scale in vitro clonal propagation of the valuable medicinal herb Eclipta alba (Asteraceae) by enhanced axillary shoot proliferation in cotyledonary node segments was designed. The medium type, various carbon sources, plant growth regulators, and coconut water markedly influenced in vitro propagation of Eclipta alba. An in vitro plantlet production system has been investigated on Murashige and Skoog (MS) medium with the synergistic combination of benzyladenine (4.4μM), kinetin (4.6μM), 2-isopentenyladenine (4.9μM), gibberellic acid (1.4μM), 5% coconut water, and 3% sucrose which promoted the maximum number of shoots as well as beneficial shoot length: Subculturing of cotyledonary node segments on a similar medium enabled continuous production of healthy shoots with similar frequency. Rooting was highest (94.3%) on full strength. MS medium containing 9.8 μM indolebutyric acid. Micropropagated plants established in garden soil, farmyard soil, and sand (2∶1∶1) were uniform and identical to the donor plant with respect to growth characteristics as well as floral features. A mechanical technique was evaluated for releasing green lacewing eggs in liquid suspensions. Deposited eggs were enclosed within a circle of nonpoisonous adhesive to protect them from predation and to prevent escape of hatched larvae. Released eggs were monitored daily for 5 days after release by measuring three response variables: adhesion rate of eggs to foliage, hatch rate of eggs, and “yield” of larvae from discharged eggs; “yield” was the product of egg adhesion and egg hatch. Factors tested were: egg conditioning prior to release (incubated or refrigerated), carrier (distilled water or commercial carrier solution), application technique (mechanical or hand application), and row facing (North or South). Release technique did not significantly effect egg hatch on any day. Conditioning eggs prior to release had the greatest effect on hatch of eggs and resulting yield of larvae during the 5-day monitoring period. Carrier had a significant effect on adhesion of eggs to leaves and hatch of eggs. Commercial carrier solution increased egg adhesion but decreased egg hatch compared to water. Overall mean yield of larvae from incubated eggs distributed mechanically was not significantly different for eggs suspended in water (36.4% on day 5 post-release) and for eggs suspended in commercial carrier solution (36.1% on day 5 post-release). Hand-applied eggs had a higher hatch and subsequent yield of larvae than mechanically released eggs; however, the hand technique was labor intensive.  相似文献

The current study examines the potential of the multicoloured Asian lady beetle Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae) to use pollen as a food to sustain development and reproduction in the absence of insect prey. Three populations of H. axyridis were used in this study: a long-term laboratory population (since 1998) and a melanic and non-melanic population originating from field collected individuals in Belgium. The insects were allowed to develop and reproduce on frozen eggs of Ephestia kuehniella Zeller (Lepidoptera: Phycitidae), frozen moist bee pollen or an even mixture of the two. Females of the field population offered the mixed diet initiated oviposition sooner than those fed only E. kuehniella eggs, but other developmental and reproductive traits were similar on these diets. A diet of pollen alone allowed 35–48% of the larvae of the field population of H. axyridis to successfully reach adulthood. However, developmental time for these individuals was prolonged by 31–49% and adult body weight was reduced by 37–68%, compared to individuals offered the diets containing E. kuehniella eggs. When fed exclusively on pollen in their larval and adult life, about 40% of the adult females of either field population were able to produce a small number of viable eggs. The laboratory and field strains differed in their response to diet for a number of developmental and reproductive traits. The exploitation of pollen and other plant foods at times when insect prey is scarce, may offer a further competitive advantage to the non-indigenous coccinellid H. axyridis over native European predatory lady beetles that share the same niche and are less capable of using pollen as an alternative food.  相似文献

The effect of yeast strain, the agave age and the cultivation field location of agave were evaluated using kinetic parameters and volatile compound production in the tequila fermentation process. Fermentations were carried out with Agave juice obtained from two cultivation fields (CF1 and CF2), as well as two ages (4 and 8 years) and two Saccharomyces cerevisiae yeast strains (GU3 and AR5) isolated from tequila fermentation must. Sugar consumption and ethanol production varied as a function of cultivation field and agave age. The production of ethyl acetate, 1-propanol, isobutanol and amyl alcohols were influenced in varying degrees by yeast strain, agave age and cultivation field. Methanol production was only affected by the agave age and 2-phenylethanol was influenced only by yeast strain. This work showed that the use of younger Agave tequilana for tequila fermentation resulted in differences in sugar consumption, ethanol and volatile compounds production at the end of fermentation, which could affect the sensory quality of the final product.  相似文献

Predation by a predator complex consisting of adults of the lygaeidGeocoris punctipes (Say), the nabidsNabis roseipennis Reuter and/orTropiconabis capsiformis Germar and the coccinelids,Hippodamia convergens Guerin-Meneville andColeomegilla maculata (DeGeer) onHeliothis virescens (F.) eggs, 1st-instar larvae or both on late pinhead square and early bloom stage cotton was evaluated in field cages in Mississippi. Prey densities of 4 (11, 512/ha) or 8 per cage and predator: prey (P:p) ratios of 1∶1, 2∶1, 3∶1, 4∶1 and 5∶1 were used. Prey were labeled with phosphorus-32 or carbon-14 to determine predator ingestion and effectiveness. Average percent egg predation as a function of P:p ratio ranged from 2.1 to 12.1 for a 48 h period. The average percent predation on larvae as a function of P:p ratio ranged from 5.3 to 22.0. The hemiptera fed more than the coleoptera on larvae, andG. punctipes was the best egg predator. For the range of predator densities used, the average area of discovery for the predator complex was 6.98 × 10−3 m/day and 2.34 × 10−2 m/day when exposed to eggs and larvae, respectively. This work tested if carbohydrates and proteins ingestion is regulated in the South American fruit fly, Anastrepha fraterculus, to optimize survival and reproduction. Adult food treatments were established by providing sugar and hydrolyzed yeast in various combinations either alone or mixed at a standard 3:1 ratio (sugar:hydrolyzed yeast). Individual food consumption was assessed and related to survival patterns. The effects of adult feeding on fecundity and fertility patterns were investigated in groups of flies. Sugar consumption was the lowest in the treatment where it was provided with hydrolyzed yeast at a fixed 3:1 ratio. Consumption of hydrolyzed yeast did not differ between this treatment and the one in which this solution was complemented with one solution of sugar. It seems that a mixture of sugar and hydrolyzed yeast at a fixed ratio of 3:1, respectively, restricts extra ingestion of sugar; most probably because of negative response of the fly to overconsumption of protein. Survival was affected by the treatments, being lower in those cases where protein was at the fixed ratio. Group experiments revealed that protein restriction expanded longevity and decreased egg production. In contrast, egg production was enhanced when flies were kept continuously with a mixture of yeast and sugar plus an extra source of sugar, and this was not in detriment of survival. Our results suggest that fixed sugar-protein ratios in which protein is in excess affects fitness components such as longevity and reproduction. These findings are discussed from a theoretical and applied perspective in the context of pest control by means of the sterile insect technique.  相似文献

Although many predatory insects appear to be opportunistic generalists in their selection of prey, only a subset of prey species may in fact serve as “essential foods” capable of supporting immature growth and adult reproduction. It has been suggested that other, “alternative foods” serve only to maintain the predator when essential foods are not available, but little research has evaluated the significance of a mixed diet of essential and alternative foods for predator growth or reproduction. Here we test the general hypothesis that although alternative prey may be inadequate to support reproduction when consumed alone by adult predators, consumption of such prey may enhance the predator's reproductive output when the predator also has access to essential prey. We compared egg production by two aphidophagous lady beetles, Coccinella septempunctata and C. transversoguttata, provided with diets of aphids (essential prey) and weevils (alternative prey). As predicted, female predators produced greater numbers of eggs when a diet of pea aphids in limited number was supplemented by alfalfa weevil larvae. The predators laid no eggs when provided only with weevils or only with sugar. But once aphids were added to the diet, females of C. transversoguttata (but not C. septempunctata) laid eggs in greater numbers when they had fed previously on weevils than on sugar. Females of both species also produced eggs in modest numbers when provided with both weevils in excess and sugar, but this diet supported a lower rate of egg production than did a diet of weevils in excess plus a limited number of aphids. Although C. septempunctata has a longer history of association with the alfalfa weevil than does C. transversoguttata, the former species was not more effective in exploiting this alternative prey in support of reproduction. The tendency of generalist predators such as adult lady beetles to consume alternative as well as essential prey probably enhances considerably their ability to capitalize on short-lived and scattered opportunities as they seek out suitable sites in which to reproduce. Summary The aim of this research was to develop methods to use low-cost carbon compounds for rhizobial inoculant production. Five raw starch materials; steamed cassava, sticky rice, fresh corn, dry corn and sorghum were tested for sugar production by an amylase-producing fungus. Streamed cassava produced the highest amount of reducing sugar after fermentation. Bradyrhizobium japonicum USDA110, Azorhizobium caulinodans IRBG23, Rhizobium phaseoli TAL1383, Sinorhizobium fredii HH103, and Mesorhizobium ciceri USDA2429 were tested on minimal medium supplemented with reducing sugar obtained from cassava fermentation. All strains, except B. japonicum USDA110, could grow in medium containing cassava sugar derived from 100 g steamed cassava per litre, and the growth rates for these strains were similar to those in medium containing 0.5 (w/v) mannitol. The sugar derived from steamed cassava was further used for production of glycerol using yeast. After 1 day of yeast fermentation, the culture containing glycerol and heat-killed yeast cells, was used to formulate media for culturing bradyrhizobia. A formulation medium, FM4, with a glycerol concentration of 0.6 g/l and yeast cells (OD600 = 0.1) supported growth of B. japonicum USDA110 up to 3.61 × 109 c.f.u./ml in 7 days. These results demonstrate that steamed cassava could be used to provide cheap and effective carbon sources for rhizobial inoculant production.  相似文献

Columbia agar base containing 0·001% acriflavine, 1·5% lithium chloride, 0·25% phenyl ethanol, 0·05% aesculin. 0·05% ferrous salt, 1% mannitol, 2·5% egg yolk emulsion and 0·008% phenol red (ALPAMY), recovered Listeria monocytogenes and some strains of Listeria seeligeri quantitatively, but suppressed Listeria ivanovii and virtually all other bacteria common in fresh foods. Thirteen yeasts isolated from the steep water of fermenting cassava tubers were screened for ethanol tolerance. Three strains which showed measurable growth in medium containing 10% (v/v) ethanol were also sugar-tolerant and grew well in medium containing 25% (w/v) glucose. One of the strains, YC3, was found to possess much higher invertase activity than the other two and could be of value in ethanol production from molasses. Further search for industrially useful yeasts in African fermented foods is suggested.  相似文献

To determine the preference of the hovering hawk mothMacroglossum stellatarum for different sugar concentrations, the foraging behavior of adults were analyzed under laboratory conditions. Six sucrose concentrations (range, 10–60%, w/w) were simultaneously offered in six artificial ab libitum feeders. The number of feeding bouts and the duration of each visit were automatically recorded and stored in a computer. Results showed that the frequency of visits to the feeders did not vary among the different solutions offered, but the gathered volume by the group attained a maximum at between 20 and 50% (w/w). Moths invested more time in front of the feeder with the more concentrated sugar solutions. It was assumed that factors different from maximizing energy intake, such as water balance and viscosity of concentrated nectars, have to be considered in order to understand the observed patterns of nectar choice.  相似文献

It was generally believed that butterflies and other holometabolous insects rely primarily on reserves accumulated during the larval stage for reproduction. Recent studies, however, highlight the often fundamental importance of adult nutrition to realize the full reproductive potential. While the importance of carbohydrates is fairly well understood, the role of most other adult-derived substances is only partially resolved. We here focus on the effects of dietary lipids (cholesterol, polyunsaturated fatty acids) and fruit decay (dietary yeast, ethanol) on female reproduction in the tropical, fruit-feeding butterfly Bicyclus anynana (Nymphalidae). We found that banana-fed control females outperformed all other groups fed on sucrose-based diets. Lipids, yeast or ethanol added to a sugar solution did not yield a similarly high reproductive output compared to fruit-fed females. Groups fed fresh or decaying banana showed no differences in reproductive performance. As we could not identify a single pivotal substance, we conclude that resource congruence (the use of nutrient types in a specified ratio) rather than any specific nutrient component is of key importance for maximum reproductive output. Further, dietary quality may affect egg hatching success in spite of no obvious effects on egg size and number. Thus, any implications about potential fitness effects of different diets need to consider egg (and hatchling) viability in addition to fecundity.  相似文献

The purpose of the present study was characterization of microparticles obtained by adsorption of poorly water soluble drug, meloxicam, on a porous silicate carrier Florite RE (FLR) and development of a tablet formulation using these microparticles, with improved drug dissolution properties. The study also reveals the use of FLR as a pharmaceutical excipient. Meloxicam was adsorbed on the FLR in 2 proportions (1∶1 and 1∶3), by fast evaporation of solvent from drug solution containing dispersed FLR. Drug adsorbed FLR microparticles were evaluated for surface topography, thermal analysis, X-ray diffraction properties, infrared spectrum, residual solvent, micromeritic properties, drug content, solubility, and dissolution studies. Microparticles showed bulk density in the range of 0.10 to 0.12 g/cm3. Dissolution of drug from microparticles containing 1∶3, drug∶FLR ratio was faster than microparticles containing 1∶1, drug∶FLR ratio. These microparticles were used for formulating directly compressible tablets. Prepared tablets were compared with a commercial tablet. All the prepared tablets showed acceptable mechanical properties. Disintegration time of prepared tablets was in the range of 18 to 38 seconds, and drug dissolution was much faster in both acidic and basic medium from prepared tablets as compared with commercial tablet. The results suggest that FLR provides a large surface area for drug adsorption and also that a reduction in crystallinity of drug occurs. I investigated the activity budget and diet of Yakushima macaques (Macaca fuscata yakui,)in warm temperate broad- leaved forest of Yakushima, Japan. Both time spent feeding and time spent moving varied considerably between half- months. However, total time spent in active behaviors— feeding time plus moving time— was stable. The composition of the diet also showed considerable variation between half- months. The macaques fed mainly on fruits, seeds,mature leaves, fallen seeds, flowers, and young leaves, each of which accounted for more than 30% of feeding time in at least 1 half- month. They also ate insects and fungi, but each of them comprised ≤ 25 and ≤ 8% of feeding time in any half- month, respectively. Time spent feeding on mature leaves, young leaves, flowers, or fallen seeds is positively correlated with total time feeding and is negatively correlated with time moving. In contrast, time feeding on fruits, seeds, insects or fungi is negatively correlated with time feeding and is positively correlated with time moving. Foraging on foods that have a low energy content, a high density, and a relatively even distribution— mature leaves— or that need much manipulation to be processed— flowers and fallen seeds— increased feeding time, while foraging on foods for which monkeys must search intensively in the forest— fruits, seeds, insects, and fungi— led to increased moving time. I examined foraging strategies of Yakushima macaques in terms of moving costs and the quality of food items. Regarding time feeding on fruits, which have more energy and may need less manipulation than other foods, as a benefit, and moving time as a cost, they seemed to employ a strategy that balanced the costs and benefits of foraging.  相似文献

Previously, a Saccharomyces cerevisiae strain was engineered for xylose assimilation by the constitutive overexpression of the Orpinomyces xylose isomerase, the S. cerevisiae xylulokinase, and the Pichia stipitis SUT1 sugar transporter genes. The recombinant strain exhibited growth on xylose, under aerobic conditions, with a specific growth rate of 0.025 h−1, while ethanol production from xylose was achieved anaerobically. In the present study, the developed recombinant yeast was adapted for enhanced growth on xylose by serial transfer in xylose-containing minimal medium under aerobic conditions. After repeated batch cultivations, a strain was isolated which grew with a specific growth rate of 0.133 h−1. The adapted strain could ferment 20 g l−1 of xylose to ethanol with a yield of 0.37 g g−1 and production rate of 0.026 g l−1 h−1. Raising the fermentation temperature from 30°C to 35°C resulted in a substantial increase in the ethanol yield (0.43 g g−1) and production rate (0.07 g l−1 h−1) as well as a significant reduction in the xylitol yield. By the addition of a sugar complexing agent, such as sodium tetraborate, significant improvement in ethanol production and reduction in xylitol accumulation was achieved. Furthermore, ethanol production from xylose and a mixture of glucose and xylose was also demonstrated in complex medium containing yeast extract, peptone, and borate with a considerably high yield of 0.48 g g−1.  相似文献

Fed-batch culture is the mode of operation of choice in industrial baker’s yeast fermentation. The particular mode of culture, operated at stable glucose and maltose concentration levels, was employed in this work in order to estimate important kinetic parameters in a process mostly described in the literature as batch or continuous culture. This way, the effects of a continuously falling sugar level during a batch process were avoided and therefore the effects of various (stable) sugar levels on growth kinetics were evaluated. Comparing the kinetics of growth and the inhibition by the substrate in cultures grown on glucose, which is the preferential sugar source for Saccharomyces cerevisiae, and maltose, the most common sugar source in industrial media for baker’s yeast production, a milder inhibition effect by the substrate in maltose-grown cells was observed, as well as a higher yield coefficient. The observed sugar inhibition effect in glucostat cultures was taken into account in modeling substrate inhibition kinetics. The inhibition coefficient K i increased with increasing sugar concentration levels, but it appeared to be unaffected by the type of substrate and almost equal for both substrates at elevated concentration levels.  相似文献

The influence of feeding, egg load and stage of ovarian development on the sexual receptivity of some insect species has been reported. This work investigated the effects of different brewer's yeast concentrations in the diet offered to females of West Indian fruit fly, Anastrepha obliqua, on their sexual receptivity. Wild females were fed on artificial diets containing 2.0 g brewer's yeast per 100 mL of water (low‐yeast diet, LY diet), 6.5 g yeast per 100 mL of water (high‐yeast diet, HY diet) or only sucrose (Su diet) and then placed with the males. Female sexual receptivity tests were carried out in acrylic boxes in the laboratory under controlled conditions (temperature, humidity and light). Females fed on the HY diet were more sexually receptive to male courtship than the other females. Additionally, when fed on the same diet (HY diet), those females with higher egg load were more sexually receptive. The data suggest that the ingestion of a protein source is essential for increasing sexual receptivity and that egg load is the most important factor affecting sexual receptivity of A. obliqua females.  相似文献

In this study, a characterization of cell wall polysaccharide composition of three yeasts involved in the production of agave distilled beverages was performed. The three yeast strains were isolated from different media (tequila, mezcal and bakery) and were evaluated for the β(1,3)-glucanase lytic activity and the β-glucan/mannan ratio during the fermentation of Agave tequilana juice and in YPD media (control). Fermentations were performed in shake flasks with 30 g l−1 sugar concentration of A. tequilana juice and with the control YPD using 30 g l−1 of glucose. The three yeasts strains showed different levels of β-glucan and mannan when they were grown in A. tequilana juice in comparison to the YPD media. The maximum rate of cell wall lyses was 50% lower in fermentations with A. tequilana juice for yeasts isolated from tequila and mezcal than compared to the bakery yeast.  相似文献
