Seven methods, including measurements of photosynthesis (A) and stomatal conductance (g(s)), carbon isotope discrimination, ecosystem CO2 and water vapour exchange using eddy covariance and the use of a multilayer canopy model and ecosystem Keeling plots, were employed to derive estimates of intercellular CO2 concentration (Ci) across a range of spatial and temporal scales in a low productivity rain forest ecosystem dominated by the conifer Dacrydium cupressinum Lamb. in New Zealand. Estimates of shoot and canopy Ci across temporal scales ranging from minutes to years were remarkably similar (range of 274-294 micromol mol(-1)). The gradual increase in shoot Ci with depth in the canopy was more likely attributable to decreases in A resulting from lower irradiance (Q) than to increases in g, due to changes in air saturation deficit (D). The lack of marked vertical gradients in A and g(s) at saturating Q through the canopy and the low seasonal variability in environmental conditions contributed to the efficacy of scaling Ci. However, the canopy Ci estimate calculated from the carbon isotope composition of respired ecosystem CO2 (delta13CR; 236 micromol mol(-1)) was much lower than other estimates of canopy Ci. Partitioning delta13CR into four components (soil, roots, litter and foliage) indicated root respiration as the dominant (> 50%) contributor to delta13CR. Variable time lags and differences in isotopic composition during photosynthesis and respiration make the direct estimation of canopy Ci from delta 13CR problematic.

Day-to-day variability in the carbon isotope composition of phloem sap (delta13Chd) and ecosystem respiratory CO2 (delta13CR) were measured to assess the tightness of coupling between canopy photosynthesis (delta13Chd) and ecosystem respiration (delta13CR) in two mature Nothofagus solandri (Hook. f.) forests in New Zealand. Abundant phloem-tapping scale insects allowed repeated, nondestructive access to stem phloem sap 1-2 m above ground. delta13Chd was compared with delta13C predicted by an environmentally driven, process-based canopy photosynthesis model. Keeling plots of within-canopy CO2 were used to estimate delta13CR. By including a lag of 3 d, there was good agreement in the timing and direction of variation in delta13Chd and predictions by the canopy photosynthesis model, suggesting that delta13Chd represents a photosynthesis-weighted, integrative record of canopy photosynthesis and conductance. Significant day-to-day variability in delta13CR was recorded at one of the two forests. At this site, delta13CR reflected variability in delta13Chd only on days with <2 mm rain. We conclude that the degree of coupling between canopy photosynthesis and ecosystem respiration varies between sites, and with environmental conditions at a single site.

The photosynthetic pathway composition (C3:C4 mixture) of an ecosystem is an important controller of carbon exchanges and surface energy flux partitioning, and therefore represents a fundamental ecophysiological distinction. To assess photosynthetic mixtures at a tallgrass prairie pasture in Oklahoma, we collected nighttime above-canopy air samples along concentration and isotopic gradients throughout the 1999 and 2000 growing seasons. We analyzed these samples for their CO2 concentration and carbon isotopic composition and calculated C3:C4 proportions with a two-source mixing model. In 1999, the C4 percentage increased from 38% in spring (late April) to 86% in early fall (mid-September). The C4 percentages inferred from ecosystem respiration measurements in 2000 indicate a smaller shift, from 67% in spring (early May) to 77% in mid-summer (late July). We also sampled daytime CO2 concentration and carbon isotope gradients above the canopy to determine ecosystem discrimination against 13CO2 during net uptake. These discrimination values were always lower than corresponding nighttime ecosystem respiration isotopic signatures would suggest. After accounting for the isotopic disequilibria between respiration and photosynthesis resulting from seasonal variations in the C3:C4 mixture, we estimated canopy photosynthetic discrimination. The C4 percentage calculated from this approach agrees with the percentage determined from nighttime respiration for sampling periods in both growing seasons. Isotopic imbalances between photosynthesis and respiration are likely to be common in mixed C3:C4 ecosystems and must be considered when using daytime isotopic measurements to constrain ecosystem physiology. Given the global extent of such ecosystems, isotopic imbalances likely contribute to global variations in the carbon isotopic composition of atmospheric CO2.

The oxygen isotope composition of CO(2) respired by Ricinus communis leaves (delta(18)O(R)) was measured under non-steady-state conditions with a temporal resolution of 3 min using a tunable diode laser (TDL) absorption spectrometer coupled to a portable gas exchange system. The SD of delta(18)O measurement by the TDL was +/- 0.2 per thousand and close to that of traditional mass spectrometers. Further, delta(18)O(R) values at isotopic steady state were comparable to those obtained using traditional flask sampling and mass spectrometric techniques for R. communis grown and measured in similar environmental conditions. As well as higher temporal resolution, the online TDL method described here has a number of advantages over mass spectrometric techniques. At isotopic steady state among plants grown at high light, the "one-way flux" model was required to accurately predict delta(18)O(R). A comparison of measurements and the model suggests that plants grown under low-light conditions have either a lower proportion of chloroplast CO(2) that isotopically equilibrates with chloroplast water, or more enriched delta(18)O of CO(2) in the chloroplast that has not equilibrated with local water. The high temporal resolution of isotopic measurements allowed the first measurements of delta(18)O(R) when stomatal conductance was rapidly changing. Under non-steady-state conditions, delta(18)O(R) varied between 50 and 220 per thousand for leaves of plants grown under different light and water environments, and varied by as much as 100 per thousand within 10 min for a single leaf. Stomatal conductance ranged from 0.001 to 1.586 mol m(-2) s(-1), and had an important influence on delta(18)O(R) under non-steady-state conditions not only via effects on leaf water H(2) (18)O enrichment, but also via effects on the rate of the one-way fluxes of CO(2) into and out of the leaf.

Contributions of C3 and C4 plants to respiration of C3-C4 ecosystems can be estimated on the basis of their contrasting 13C discrimination. But accurate partitioning requires accurate measurements of the isotope signature of whole system respiratory CO2 (deltaR), and of its members (delta3 and delta4). Unfortunately, experimental determination of representative delta3 and delta4 values is virtually impossible in nature, generating a need for proxies (surrogates) of delta3 and delta4 values (e.g., the delta of leaf biomass). However, recent evidence indicates that there may be systematic differences among the delta of respiratory and biomass components. Thus, partitioning may be biased depending on the proxy. We tested a wide range of biomass- and respiration-based delta proxies for the partitioning of respiration of mixed Lolium perenne (C3) - Paspalum dilatatum (C4) stands growing at two temperatures inside large 13CO2/ 12CO2 gas exchange chambers. Proxy-based partitioning was compared with results of reference methods, including (i) the delta of whole plant respiratory CO2 (delta3 and delta4) or (ii) respiration rate of intact C3 and C4 plants. Results of the reference methods agreed near perfectly. Conversely, some proxies yielded erroneous partitioning results. Partitioning based on either the delta of shoot or root respiratory CO2 produced the worst bias, because shoot respiratory CO2 was enriched in 13C by several per thousand and root respiratory CO2 was depleted by several per thousand relative to whole plant respiratory CO2. Use of whole plant or whole shoot biomass delta gave satisfactory partitioning results under the constant conditions of the experiments, but their use in natural settings is cautioned if environmental conditions are variable and the time scales of respiration partitioning differ strongly from the residence time of C in biomass. Other biomass-based proxies with faster turnover (e.g., leaf growth zones) may be more useful in changing conditions.

Perennial species with the C(4) pathway hold promise for biomass-based energy sources. We have explored the extent that CO(2) uptake of such species may be limited by light in a temperate climate. One energetic cost of the C(4) pathway is the leakiness () of bundle sheath tissues, whereby a variable proportion of the CO(2), concentrated in bundle sheath cells, retrodiffuses back to the mesophyll. In this study, we scale from leaf to canopy level of a Miscanthus crop (Miscanthus x giganteus hybrid) under field conditions and model the likely limitations to CO(2) fixation. At the leaf level, measurements of photosynthesis coupled to online carbon isotope discrimination showed that leaves within a 3.3-m canopy (leaf area index = 8.3) show a progressive increase in both carbon isotope discrimination and as light decreases. A similar increase was observed at the ecosystem scale when we used eddy covariance net ecosystem CO(2) fluxes, together with isotopic profiles, to partition photosynthetic and respiratory isotopic flux densities (isofluxes) and derive canopy carbon isotope discrimination as an integrated proxy for at the canopy level. Modeled values of canopy CO(2) fixation using leaf-level measurements of suggest that around 32% of potential photosynthetic carbon gain is lost due to light limitation, whereas using determined independently from isofluxes at the canopy level the reduction in canopy CO(2) uptake is estimated at 14%. Based on these results, we identify as an important limitation to CO(2) uptake of crops with the C(4) pathway.

Recent (13) CO(2) canopy pulse chase labeling studies revealed that photosynthesis influences the carbon isotopic composition of soil respired CO(2) (δ(13) C(SR)) even on a diel timescale. However, the driving mechanisms underlying these short-term responses remain unclear, in particular under drought conditions. The gas exchange of CO(2) isotopes of canopy and soil was monitored in drought/nondrought-stressed beech (Fagus sylvatica) saplings after (13) CO(2) canopy pulse labeling. A combined canopy/soil chamber system with gas-tight separated soil and canopy compartments was coupled to a laser spectrometer measuring mixing ratios and isotopic composition of CO(2) in air at high temporal resolution. The measured δ(13) C(SR) signal was then explained and substantiated by a mechanistic carbon allocation model. Leaf metabolism had a strong imprint on diel cycles in control plants, as a result of an alternating substrate supply switching between sugar and transient starch. By contrast, diel cycles in drought-stressed plants were determined by the relative contributions of autotrophic and heterotrophic respiration throughout the day. Drought reduced the speed of the link between photosynthesis and soil respiration by a factor of c. 2.5, depending on the photosynthetic rate. Drought slows the coupling between photosynthesis and soil respiration and alters the underlying mechanism causing diel variations of δ(13) C(SR).

Changes in the concentration and stable isotope ratio of atmospheric CO(2) can be used to study variations in the net exchange of carbon dioxide in terrestrial ecosystems (net difference between total photosynthesis and respiration). Changes in the timing of seasonal fluctuations in atmospheric CO(2) concentration have suggested that net uptake of carbon dioxide has been increasing in northern latitude ecosystems in association with warmer temperatures and a lengthening of the growing season. Stable isotope techniques allow a more detailed separation of differences between ecosystem photosynthesis and respiration because these two processes have contrasting effects on both the carbon and oxygen isotope ratio of atmospheric CO(2). Future applications of stable isotope analyses include documenting and monitoring the influence of global environmental change on ecosystem CO(2) exchange at regional scales (10-1000km(2)).

Elements of a dynamic systems model of canopy photosynthesis
Improving photosynthesis throughout the full canopy rather than photosynthesis of only the top leaves of the canopy is central to improving crop yields. Many canopy photosynthesis models have been developed from physiological and ecological perspectives, however most do not consider heterogeneities of microclimatic factors inside a canopy, canopy dynamics and associated energetics, or competition among different plants, and most models lack a direct linkage to molecular processes. Here we described the rationale, elements, and approaches necessary to build a dynamic systems model of canopy photosynthesis. A systems model should integrate metabolic processes including photosynthesis, respiration, nitrogen metabolism, resource re-mobilization and photosynthate partitioning with canopy level light, CO(2), water vapor distributions and heat exchange processes. In so doing a systems-based canopy photosynthesis model will enable studies of molecular ecology and dramatically improve our insight into engineering crops for improved canopy photosynthetic CO(2) uptake, resource use efficiencies and yields.

The oxygen isotope composition of atmospheric CO(2) is an important signal that helps distinguish between ecosystem photosynthetic and respiratory processes. In C(4) plants the carbonic anhydrase (CA)-catalyzed interconversion of CO(2) and bicarbonate (HCO(3)(-)) is an essential first reaction for C(4) photosynthesis but also plays an important role in the CO(2)-H(2)O exchange of oxygen as it enhances the rate of isotopic equilibrium between CO(2) and water. The C(4) dicot Flaveria bidentis containing genetically reduced levels of leaf CA (CA(leaf)) has been used to test whether changing leaf CA activity influences online measurements of C(18)OO discrimination (Delta(18)O) and the proportion of CO(2) in isotopic equilibrium with leaf water at the site of oxygen exchange (theta). The Delta(18)O in wild-type F. bidentis, which contains high levels of CA relative to the rates of net CO(2) assimilation, was less than predicted by models of Delta(18)O. Additionally, Delta(18)O was sensitive to small decreases in CA(leaf). However, reduced CA activity in F. bidentis had little effect on net CO(2) assimilation, transpiration rates (E), and stomatal conductance (g(s)) until CA levels were less than 20% of wild type. The values of theta determined from measurements of Delta(18)O and the (18)O isotopic composition of leaf water at the site of evaporation (delta(e)) were low in the wild-type F. bidentis and decreased in transgenic plants with reduced levels of CA activity. Measured values of theta were always significantly lower than the values of theta predicted from in vitro CA activity and gas exchange. The data presented here indicates that CA content in a C(4) leaf may not represent the CA activity associated with the CO(2)-H(2)O oxygen exchange and therefore may not be a good predictor of theta during C(4) photosynthesis. Furthermore, uncertainties in the isotopic composition of water at the site of exchange may also limit the ability to accurately predict theta in C(4) plants.

We describe an open leaf gas exchange system coupled to a tunable diode laser (TDL) spectroscopy system enabling measurement of the leaf respiratory CO(2) flux and its associated carbon isotope composition (delta(13)C(Rl)) every 3 min. The precision of delta(13)C(Rl) measurement is comparable to that of traditional mass spectrometry techniques. delta(13)C(Rl) from castor bean (Ricinus communis L.) leaves tended to be positively related to the ratio of CO(2) produced to O(2) consumed [respiratory quotient (RQ)] after 24-48 h of prolonged darkness, in support of existing models. Further, the apparent fractionation between respiratory substrates and respired CO(2) within 1-8 h after the start of the dark period was similar to previous observations. In subsequent experiments, R. communis plants were grown under variable water availability to provide a range in delta(13)C of recently fixed carbohydrate. In leaves exposed to high light levels prior to the start of the dark period, CO(2) respired by leaves was up to 11 per thousand more enriched than phloem sap sugars within the first 10-15 min after plants had been moved from the light into the dark. The (13)C enrichment in respired CO(2) then decreased rapidly to within 3-7 per thousand of phloem sap after 30-60 min in the dark. This strong enrichment was not observed if light levels were low prior to the start of the dark period. Measurements of RQ confirmed that carbohydrates were the likely respiratory substrate for plants (RQ > 0.8) within the first 60 min after illumination. The strong (13)C enrichment that followed a high light-to-dark transition coincided with high respiration rates, suggesting that so-called light-enhanced dark respiration (LEDR) is fed by (13)C-enriched metabolites.

We investigate the utility of an improved isotopic method to partition the net ecosystem exchange of CO2 (F) into net photosynthesis (FA) and nonfoliar respiration (FR). Measurements of F and the carbon isotopic content in air at a high‐elevation coniferous forest (the Niwot Ridge AmeriFlux site) were used to partition F into FA and FR. Isotopically partitioned fluxes were then compared with an independent flux partitioning method that estimated gross photosynthesis (GEE) and total ecosystem respiration (TER) based on statistical regressions of night‐time F and air temperature. We compared the estimates of FA and FR with expected canopy physiological relationships with light (photosynthetically active radiation) and air temperature. Estimates of FA and GEE were dependent on light as expected, and TER, but not FR, exhibited the expected dependence on temperature. Estimates of the isotopic disequilibrium D , or the difference between the isotopic signatures of net photosynthesis (δA, mean value ?24.6‰) and ecosystem respiration (δR, mean value ?25.1‰) were generally positive (δAR). The sign of D observed here is inconsistent with many other studies. The key parameters of the improved isotopic flux partitioning method presented here are ecosystem scale mesophyll conductance (gm) and maximal vegetative stomatal conductance (gcmax). The sensitivity analyses of FA, FR, and D to gcmax indicated a critical value of gcmax (0.15 mol m?2 s?1) above which estimates of FA and FR became larger in magnitude relative to GEE and TER. The value of D decreased with increasing values of gm and gcmax, but was still positive across all values of gm and gcmax. We conclude that the characterization of canopy‐scale mesophyll and stomatal conductances are important for further progress with the isotope partitioning method, and to confirm our anomalous isotopic disequilibrium findings.

An increase in atmospheric CO(2) concentration ( [CO(2)]) is generally expected to enhance photosynthesis and biomass. Rice plants (Oryza sativa L.) were grown in ambient CO(2) (AMB) or free-air CO(2)-enrichment (FACE), in which the target [CO(2)] was 200 micromol mol(-1) above AMB. (13)CO(2) was fed to the plants at different stages so we could examine the partitioning of photosynthates. Furthermore, canopy photosynthesis and respiration were measured at those stages. The ratio of (13)C content in the whole plant to the amount of fixed (13)C under FACE was similar to that under AMB at the vegetative stage. However, the ratio under FACE was greater than the ratio under AMB at the grain-filling stage. At the vegetative stage, plants grown under FACE had a larger biomass than those grown under AMB owing to enhancement of canopy photosynthesis by the increased [CO(2)]. On the other hand, at the grain-filling stage, CO(2) enrichment promoted the partitioning of photosynthate to ears, and plants grown under FACE had a greater weight of ears. However, enhancement of ear weight by CO(2) enrichment was not as great as that of biomass at the vegetative stage. Plants grown under FACE did not necessarily show higher canopy photosynthetic rates at the grain-filling stage. Therefore, we concluded that the ear weight did not increase as much as biomass at the vegetative stage owing to a loss of the advantage in CO(2) gain during the grain-filling period.

We report diurnal variations in 18O discrimination (18 delta) during photosynthesis (18 delta A) and respiration (18 delta R) of Picea sitchensis branches measured in branch chambers in the field. These observations were compared with predicted 18 delta (18 delta pred) based on concurrent measurements of branch gas exchange to evaluate steady state and non-steady state (NSS) models of foliage water 18O enrichment for predicting the impact of this ecosystem on the Delta 18O of atmospheric CO2. The non-steady state approach substantially improved the agreement between 18 delta pred and observed 18 delta (18 delta obs) compared with the assumption of isotopic steady state (ISS) for the Delta 18O signature of foliage water. In addition, we found direct observational evidence for NSS effects: extremely high apparent 18 delta values at dusk, dawn and during nocturnal respiration. Our experiments also show the importance of bidirectional foliage gas exchange at night (isotopic equilibration in addition to the net flux). Taken together, neglecting these effects leads to an underestimation of daily net canopy isofluxes from this forest by up to 30%. We expect NSS effects to be most pronounced in species with high specific leaf water content such as conifers and when stomata are open at night or when there is high relative humidity, and we suggest modifications to ecosystem and global models of delta 18O of CO2.

Stable Isotopes and Carbon Cycle Processes in Forests and Grasslands
Abstract: Scaling and partitioning are frequently two difficult challenges facing ecology today. With regard to ecosystem carbon balance studies, ecologists and atmospheric scientists are often interested in asking how fluxes of carbon dioxide scale across the landscape, region and continent. Yet at the same time, physiological ecologists and ecosystem ecologists are interested in dissecting the net ecosystem CO2 exchange between the biosphere and the atmosphere to achieve a better understanding of the balance between photosynthesis and respiration within a forest. In both of these multiple-scale ecological questions, stable isotope analyses of carbon dioxide can play a central role in influencing our understanding of the extent to which terrestrial ecosystems are carbon sinks. In this synthesis, we review the theory and present field evidence to address isotopic scaling of CO2 fluxes. We first show that the 13C isotopic signal which ecosystems impart to the atmosphere does not remain constant over time at either temporal or spatial scales. The relative balances of different biological activities and plant responses to stress result in dynamic changes in the 13C isotopic exchange between the biosphere and atmosphere, with both seasonal and stand-age factors playing major roles influencing the 13C biosphere-atmosphere exchange. We then examine how stable isotopes are used to partition net ecosystem exchange fluxes in order to calculate shifts in the balance of photosynthesis and respiration. Lastly, we explore how fundamental differences in the 18O isotopic gas exchange of forest and grassland ecosystems can be used to further partition terrestrial fluxes.

The C balance of a tall fescue sward grown under different ratesof N fertilization in summer, autumn, and spring was calculatedusing models derived from measurements of shoot growth, canopygross photosynthesis, shoot respiration and of C partitioningto the roots. Under the diverse growing conditions associatedwith the seasons and the N fertilization, C utilization forabove- and below-ground biomass accumulation never exceeded39 and 14% of the canopy gross photosynthesis, respectively.Carbon losses attributed to root respiration and exudation, which were estimated by difference between canopy net photosynthesisand total growth, ranged between 3 and 30% of canopy gross photosynthesis.Seasonal differences in shoot growth could be attributed tothe amount of intercepted radiation, the radiation-use efficiencyand the C partitioning to the roots. The effect of N deficiencyon shoot growth can be attributed to its effects on canopy photosynthesis(principally resulting from changes in intercepted photosyntheticallyactive radiation) and C partitioning. In comparison with theeffect on shoot growth, the effect of the N deficiency on thecanopy gross photosynthesis per unit of light intercepted overthe regrowth cycle was limited. It is concluded that most ofthe effect of N fertilization on shoot growth is due to changesin C partitioning which result in faster leaf area developmentand greater light interception.Copyright 1994, 1999 AcademicPress Tall rescue, Festuca arundinacea Schreb., carbon balance, nitrogen, grass, fertilization  相似文献
