The action of T-activin on peritoneal macrophages of CBA mice after its introduction into the animals has been studied. In intact mice the phagocytic activity of macrophages and their resistance to the cytopathogenic action of Salmonella typhimurium live cells remains unchanged. The injection of corpuscular pertussis vaccine into mice leads to a decrease in the resistance of macrophages to the action of salmonellae. The simultaneous injection of T-activin into mice in doses of 0.1 and 1.0 microgram per animal abolishes the damaging action of the vaccine. The analysis of the in vitro action of T-activin on macrophages of intact mice revealed that the preliminary incubation of cells with the preparation sharply increases their resistance to the action of salmonellae, while its introduction simultaneously with bacteria or after them rapidly leads to the death of macrophages. The action of T-activin is supposed to be linked with triggering the biosynthetic processes mediating the resistance of macrophages to the cytopathogenic action of salmonellae.  相似文献

The effects of denervation on the electrical membrane properties of fish red muscle were investigated. Forty to fifty hours after denervation, miniature endplate potentials disappeared abruptly and field stimulation of the nerve within the muscle failed to evoke endplate potentials, indicating that transmission failure occurred at this time. The membrane resistance of the red muscle fibre increased after denervation. Normally innervated fish red muscles do not generate action potentials in response to either nerve or direct muscle stimulation. However, approximately 3 weeks after nerve sectioning, action potentials could be induced in the muscles. The action potential was sodium-dependent, and was sensitive to tetrodotoxin. Actinomycin D injected in the early phase after operation suppressed the induction of the action potential. These results indicate that RNA synthesis is preliminary to the induction of the action potential mechanism, and that this mechanism is under neural control.  相似文献

We investigated the actions of dantrolene Ca(2+)-induced on Ca(2+)-release (CICR) evoked by action potentials in cultured rat sensory neurons. The effect of dantrolene on action potential after-depolarization and voltage-activated calcium currents was studied in cultured neonatal rat dorsal root ganglion cells (DRG) using the whole-cell patch-clamp technique. Depolarizing current injection evoked action potentials and depolarizing after-potentials, which are activated as a result of CICR following a single action potential in some cells. The type of after-potentials was determined by inducing action potentials from the resting membrane potential. Extracellular application of dantrolene (10 microM) abolished after-depolarizations without affecting action potential properties. Furthermore, dantrolene significantly reduced repetitive action potentials after depolarizing current injection into these neurons, but had no significant effect on the steady-state current voltage relationship of calcium currents in these neurons. We conclude that dantrolene inhibits the induction of action potential after depolarizations by inhibiting CICR in cultured rat sensory neurons.  相似文献

The activity of neurones of the visual and sensorimotor areas of the neocortex was simultaneously recorded in rabbits under the action of conditioned, inhibitory stimuli (CS, IS) and at simultaneous presentation of CS and the unconditioned stimulus (US) after trans-switching of the positive and inhibitory conditioned reflexes. Mean time of conjugated reactions of simultaneously recorded pairs of neurons is similar under the action of CS, combined action of CS and US and under IS. During the IS action as compared with CS, the phasic activity of some neurones in different areas increased, and due to this the similarity of reactions also increased; in other pairs of neurones the reactions weakened and the responses similarity decreased. The frequency of the appearance of intervals of the conjugated excitation after CS corresponds to 4.3 Hz; under combined action of CS and US it increases up to 5.9 Hz, and after IS--it decreases to 3.6 Hz. The order of coming into conjugated action of different pairs of neurones changes depending on the signal significance of the stimuli.  相似文献

In the mixed venous blood of anaesthetized, heparinized cats prostacyclin de-aggregated platelet thrombi, which were formed on the surface of blood-superfused collagen strips or on the surface of blood-superfused aortic strips from atherosclerotic rabbits. The reversal of platelet aggregation by prostacyclin was still achieved 3 hrs after the formation of platelet clumps. After an intravenous injection of prostacyclin the ID50 for its de-aggregatory action was 7.5 microgram/kg. Theophylline ethyl-diamine (aminophylline), at a dose of 3 mg/kg i.v., did not reverse platelet aggregation but it enhanced the duration of the de-aggregatory action of prostacyclin; it had little effect on the hypotensive action of prostacyclin. It is concluded that prostacyclin disintegrates platelet clumps long after they are formed in heparinized blood in vivo and that its anti-platelet action, but not hypotensive action, is selectively potentiated by a phosphodiesterase inhibitor. The above experimental data indicate the possibility of the combined use of theophylline and prostacyclin in arterial thrombosis.  相似文献

A system for the generation of the repolarization phase of the ventricular action potential is described. The system is based on time-dependent changes in membrane conductance to sodium and potassium ions. However, the changes in conductance during an action potential retain a degree of voltage dependence through the initial conditions which depend on previous depolarizations of the membrane. The equations describing the system were solved with an analog computer and various action potential forms are reproduced. The effects of hyperpolarizing and depolarizing current applied during an action potential are investigated. The changes in shape of an action potential after a change in the rate of stimulation show partial agreement with previous experimental findings. The applicability of time-dependent and voltage-dependent systems for the generation of the repolarization phase of the ventricular action potential is discussed.  相似文献

Hepatic insulin sensitizing substance (HISS) action accounts for 55% of the glucose disposal effect of a bolus of insulin in the fed state. To determine the effect of continuous versus pulsatile insulin delivery on HISS action in male Sprague-Dawley rats, insulin sensitivity was assessed using the rapid insulin sensitivity test (RIST) before and after a continuous, pulsatile, or bolus insulin (60 mU/kg i.v.) delivery. There was a significant difference in the RIST index after a continuous insulin infusion (247.9 mg/kg before, 73.2 mg/kg after) but not after 3 pulses where insulin action returned to baseline between pulses (211.6 mg/kg before, 191.0 mg/kg after) or single bolus (205.8 mg/kg before, 189.9 mg/kg after) insulin infusion. If a 3-pulse infusion was timed so that insulin action did not return to baseline between pulses, HISS action was suppressed. Continuous insulin infusion (10-30 min) showed progressive postinfusion blockade of HISS action. To maintain HISS-dependent insulin action, continuous insulin infusions should be avoided.  相似文献

The irreversibility of the damaging action of pulse electric discharges (PED) on microorganisms in the process of the disinfection of water was studied. The experiments with standard E. coli culture showed that bacterial cells remaining alive after being subjected to the action of PED eventually died the sooner, the higher the temperature of the medium was. In any of the "blind passages" made within 7 days no growth of the test microorganism was observed. The use of enriched fluid media and 10-day incubation did not ensure the preservation of proliferation ability of the cells after the action of PED with the parameters used in the experiment.  相似文献

The action of a high molecular weight peptidoglycan produced by Agrobacter radiobacter sp. on the functional activity parameters in leukocytes and macrophages i. e. chemotaxis and adhesion was studied. It was shown that the peptidoglycan had a stimulating action on the chemotaxis of cells of the peritoneal exudate. A marked stimulating action of the drug on the primary immune response to the tissue antigen of sheep erythrocytes was observed. The peptidoglycan stimulated the antibody titers and delayed hypersensitivity when administered in various periods after an antigenic stimulus. Multifactorial experiments on the protective action of the peptidoglycan in experimental infections were carried out. Second-order polynomial statistic models characterizing the animal survival rate were constructed and the dose-time parameters of the drug use were optimized.  相似文献

The paper presents the results of the experimental study of the action of rifampicin on the process of rabies infection in albino mice contaminated with 1-10 LD50 of the fixed rabies virus. Exposure to rifampicin in doses of 250 and 500 micrograms/mouse (35-70 mg/kg) resulted in survival of 66.7 and 83.4 per cent of the animals respectively while in the controls it did not exceed 16.6 and 25.0 per cent. The average life-span of the albino mice treated with the antibiotic increased 1.6-2.4-fold in comparison with the controls. The chemotherapeutic index of rifampicin representing the ratio of the maximum tolerance dose to the minimum dose providing the protective action was equal to 20. The protective action was observed either after administration of the antibiotic according to the treatment-and-prophylaxis scheme or after administration of its 2- or 3-fold dose once a day immediately after the contamination.  相似文献

Because of conflicting reports on the basic mechanisms responsible for the action potential changes produced by hypoxia or metabolic inhibitors, we investigated the effect of 2,4-dinitrophenol, substrate deprivation, and 2-deoxy-D-glucose on three preparations of rat myocardium: adult and newborn ventricle and cultured cells derived from neonatal rats. The latter exhibit slow action potentials in contrast to the other two, which show fast action potentials. Cultured cells were insensitive to 2,4-dinitrophenol and substrate deprivation but were markedly inhibited by blocking of glycolysis. The action potential of adult cells was shortened in the three conditions tested but was most sensitive to blocking of oxidative phosphorylation, which abolished propagated electrical activity after 15 min of exposure. The response of newborn ventricle was intermediate between the other two. Our data indicated that as far as maintenance of the membrane electrical properties is concerned, the relative importance of different metabolic pathways varies with development after birth or with time in culture.  相似文献

The extract of cells of ascitic Ehrlich's tumour (chaloun) and its cell-free fluid produced a marked inhibitory action on the cell proliferation of this tumour four hours after the administration. The effect is tissue-specific, more pronounced in the extract and depends on the dose of the antigen. Eight hours after the extract or the cell-free fluid administration the mitotic activity in the tumour proved to increase in comparison with control; this indicated the presence of a short-lived chaloun action on the G2-phase of the mitotic cycle and synchronization of cell division.  相似文献

The possibility of direct androgen determination of high unusual estrogen-binding protein (UEBP) level in female rat hepatocytes to the level typical to the males was studied. The direct effect of androgen (A) on the UEBP content in hepatocytes of ovariectomized female rats was shown by estimation in vivo and on hepatocyte culture. It has been revealed that the permissive action of growth hormone (GH) is necessary for normal expression of direct androgen effect. The latter has been stable after cessation of hormone action. High concentration of GH was found to possess positive regulatory effect on the UEBP in hepatocytes of ovariectomized rats. Effect of GH quickly disappeared after hormone withdrawal and was under negative regulation by physiological concentrations of A. It was concluded that A owing to its direct action together with permissive influence of GH is capable of determining stable expression of male phenotype of the UEBP level in hepatocytes of females.  相似文献

The modulation of the effects of sotalol (30 microM) by two sodium channel blockers, tetrodotoxin (0.07 microM) and lidocaine (50 microM), and by a potassium channel activator, nicorandil (30 microM), were examined. Sotalol alone greatly increased Purkinje fiber transmembrane action potential duration and, in some preparations, induced early after depolarizations. Concurrent with the changes in action potential duration, sotalol also increased isolated Purkinje strand developed force paced at slow rates (0.33 Hz). These sotalol-induced alterations of Purkinje strand electromechanical characteristics were similar to those produced by either veratrine (0.6 or 1.0 micrograms/mL) or by tetraethylammonium (10 mM). The effects of sotalol on action potential duration and force development were reversed by exposure to either tetrodotoxin or nicorandil. Lidocaine also reversed the effects of sotalol on action potential duration and developed force. The sotalol-induced increase in action potential duration and development of early after depolarizations may, therefore, be abated by combination with drugs that either block cardiac sodium channels or that increase membrane potassium conductance. Combination with such drugs may help prevent the adverse arrhythmogenic effects of sotalol.  相似文献

The persistence of cellular electropharmacologic effects of mexiletine on canine Purkinje fibers was studied utilizing standard microelectrode techniques and two different protocols. In the first, the tissue was exposed to hypoxic perfusion before and 30 min after perfusion with one of the following: mexiletine hydrochloride 6.25 microM solution, mexiletine hydrochloride 12.5 microM solution, or drug-free Tyrode's solution. With the higher concentration of mexiletine, depression of the maximal upstroke velocity (Vmax) persisted 30 min after drug washout and subsequent exposure to hypoxia did not result in the anticipated shortening of action potential duration but did prevent the restoration of normal Vmax. After perfusion with the lower concentration of mexiletine, Vmax was not depressed and hypoxic action potential duration shortening was not prevented. In the second protocol, Purkinje fibers were perfused with 12.5 microM mexiletine hydrochloride solution and then exposed to hypoxia after 15, 30, 45, or 60 min of perfusion with drug-free solution. Depression of maximal upstroke velocity and shortening of action potential duration persisted during washout, returning to control values by 45 min, although mexiletine was not detectable in the tissue bath after 10 min of washout. Hypoxia initiated at 15 or 30 min of washout failed to produce the anticipated shortening of action potential duration. At 45 and 60 min, action potential duration was shortened by hypoxia. We concluded that mexiletine depression of Vmax and shortening of action potential duration may persist in the absence of drug. Further shortening of action potential duration in response to hypoxia is prevented during this period. The persistence of Vmax depression is prolonged by hypoxia.  相似文献

It has been shown in chronic experiments on rats that electro-convulsive action applied in five to ten seconds after one-session conditioning completely precludes the reproduction of conditioned motor reactions, while conditioned bradycardiac reactions are clearly reproduced in tests one hour and 24 hours after elaboration. Unconditioned stimulus presented in another experimental situation six to seven days after elaboration and amnesic action, leads to a stable reproduction of the previously non-achieved conditioned motor reactions. The results obtained suggest that electrical seizures influence the reproduction of motor reactions, rather than fixation of temporary connection.  相似文献

The mechanism of action of ricin, an irreversible inhibitor of protein synthesis in eukaryotic ribosomes, has been studied with ethanol extracted 80S rat liver ribosomes. The results show that the irreversible action of the toxin is on a component of the ribosome which remains in the core ribosome after the removal of the ethanol soluble proteins. The acid phosphoproteins P1 and P2 are shown not to be the site of action of the toxin.  相似文献

We studied the clastogenic action of cyclophosphamide (CP) on bone marrow cells of the Armenian hamster (AH), Cricetulus migratorius. CP induced a dose-dependent linear increase in aberrant cells. The maximal cytogenetic action was observed 12 h after CP treatment. Male and female AHs were similarly sensitive to the clastogenic action of CP. We compared CP clastogenicity at a dose of 25 mg/kg on bone marrow cells of AHs, mice, rats, guinea pigs and Chinese hamsters 24 h after treatment. We observed that this dose of CP induced only 2.8% aberrant cells in bone marrow of AHs, but 42.8%, 32.2%, 25% and 14.6% aberrant cells in bone marrow of guinea pigs, rats, mice and Chinese hamsters respectively. AHs are much more resistant to the metaphase-arresting action of colchicine than other species of rodents (e.g., the colchicine dose for AHs is 100-fold more than for rats). Thus AHs are the most resistant of all rodent species studied to the clastogenic action of CP.  相似文献
