Spores of Monilinia fructicola, Colletotrichum coccodes, Botrytis cinerea, Alternaria alternata and Mucor mucedo were transferred to agar plates containing 0.5, 5 or 50 mM concentrations of various plant cell wall-related monosaccharides or glucose. The wall-related monosaccharides tested were arabinose, galactose, galacturonic acid, mannose, rhamnose and xylose. Germ tube length was measured after 24 h. Growth of germ tubes on media containing the different monosaccharides varied substantially among fungi. The most striking response was with M. mucedo; spores were only able to grow on media containing glucose or galactose.Increase in colony size was determined during a 7-day period after subcultures were transferred to media containing 5 mM arabinose, galactose, galacturonic acid, mannose, rhamnose and xylose, each with and without 5 mM glucose. Glucose, alone or in conjunction with any other monosaccharide, was the optimal carbon source for growth. In general, fungi were unable to grow as well on any of the wall-related monosaccharides without glucose. Galacturonic acid and rhamnose, major components of pectic polysaccharides of plant cell walls, were unable to support any growth of M. fructicola cultures. No wall-related monosaccharides supported more than 50% of the growth observed when M. mucedo cultures were grown on glucose.  相似文献

Regulation of endoglucanase formation by the brown-rot fungusGloeophyllum trabeum was investigated. This fungus produced endoglucanases in the presence of monosaccharides such as glucose or mannose as the sole carbon source, but the expression of these enzymes was four to five times higher in the presence of cellulose or cellobiose. In a lactose- or glucose-containing medium, endoglucanase production was induced by cellobiose. Glucose and glycerol did not repress enzyme production. We concluded that endoglucanase production by brown-rot fungi is inducible by cellulose and not subject to catabolite repression. Cellobiose is the most effective inducer of the system.  相似文献

The 13C and gas chromatography-mass spectrometry (13C-GC-MS)method was applied to determine the day-night changes in thecomposition of photosynthetic products of the natural phytoplanktonpopulation from Lake Biwa, Japan. Glucose is the most abundantmonosaccharide in acid-hydrolyzable carbohydrate. The contributionof glucose was high in incubatesd samples in daytime and decreasedduring the night. Other monosaccharides (rhamnose, fucose, ribose,arabinose, xylose, mannose and galactose) and amino acids tendedto be produced throughout both day- and night-time. These resultssuggested that the carbon flows from glucose, which might constitutereserve glucan, to other monosaccharides and amino acids duringnight-time. The disproportionate production of glucose (reservedglucan) during daytime was thus partly cancelled out at night.  相似文献

Due to catabolite repression in microorganisms, sugar mixtures cannot be metabolized in a rapid and efficient manner. Therefore, the development of mutant strains that avoid this regulatory system is of special interest to fermentation processes. In the present study, the utilization of sugar mixtures by an Escherichia coli mutant strain devoid of the phosphotransferase system (PTS) was characterized. This mutant can transport glucose (PTS- Glucose+ phenotype) by a non-PTS mechanism as rapidly as its wild-type parental strain. In cultures grown in minimal medium supplemented with glucose-xylose or glucose-arabinose mixtures, glucose repressed arabinose- or xylose-utilization in the wild-type strain. However, under the same culture conditions with the PTS- Glucose+ mutant, glucose and arabinose were co-metabolized, but glucose still exerted a partial repressive effect on xylose consumption. In cultures growing with a triple mixture of glucose-arabinose-xylose, the wild-type strain sequentially utilized glucose, arabinose and finally, xylose. In contrast, the PTS- Glucose+ strain co-metabolized glucose and arabinose, whereas xylose was utilized after glucose-arabinose depletion. As a result of glucose-arabinose co-metabolism, the PTS- Glucose+ strain consumed the total amount of sugars contained in the culture medium 16% faster than the wild-type strain. [14C]-Xylose uptake experiments showed that in the PTS- Glucose+ strain, galactose permease increases xylose transport capacity and the observed partial repression of xylose utilization depends on the presence of intracellular glucose.  相似文献

Eight species of bifidobacteria were tested for their abilities to grow on a range of monosaccharides (glucose, arabinose, xylose, galactose and mannose). In contrast to the other sugars, glucose and galactose were utilized by all species and, in general, specific growth rates were highest on these sugars. Different substrate preferences were observed between species when the bacteria were grown in the presence of all five monosaccharides. For example, glucose and xylose were coutilized by Bifidobacterium longum, whereas glucose repressed uptake of all other sugars in B. bifidum and B. catenulatum. Galactose was the preferred substrate with B. pseudolongum. In B. angulatum, glucose and galactose were utilized simultaneously. B. breve did not grow on arabinose when this sugar provided the sole source of energy. However, glucose and arabinose were preferentially taken up during growth on sugar mixtures.  相似文献

Hemicellulose was extracted from horse bean and wheat straws in a yield of 5 and 9% respectively. The whole hemicellulose was hydrolysed and the molar ratio of the component monosaccharides was determined. Uronic acid, galactose, glucose, arabinose and xylose were found in both hemicelluloses. The molar ratio of the monosaccharides was determined in each of 4 fractions derived from the saccharide. The main fractions (B and C) were partially hydrolysed and an oligosaccharide containing arabinose and xylose (1:1) was isolated from both hemicelluloses. Another oligosaccharide containing xylose and glucose (2:1) was also isolated from wheat straw hemicellulose. Periodate oxidation was carried out on fractions B and C. The formic acid and the consumed periodate were determined. Each hemicellulose was subjected to Smith's degradation. Glycerol, erythrytol and compounds containing xylose and glycerol (1:1), and xylose and erythrytol (1:1) were isolated.  相似文献

In the process of producing ethanol from lignocellulosic materials such as wheat straw, compounds that can act inhibitory to enzymatic hydrolysis and to cellular growth may be generated during the pretreatment. Ethanol production was evaluated on pretreated wheat straw hydrolysate using four different recombinant Saccharomyces cerevisiae strains, CPB.CR4, CPB.CB4, F12, and FLX. The fermentation performance of the four S. cerevisiae strains was tested in hydrolysate of wheat straw that has been pretreated at high dry matter content (220 g/L dry matter). The results clearly showed that F12 was the most robust strain, whereas the other three strains were strongly inhibited when the fraction of hydrolysate in the fermentation medium was higher than 60% (v/v). Furthermore, the impact of different lignin derivatives commonly found in the hydrolysate of pretreated wheat straw, was tested on two different enzyme mixtures, a mixture of Celluclast 1.5 L FG and Novozym 188 (3:1) and one crude enzyme preparation produced from Penicillium brasilianum IBT 20888. From all the potential inhibiting compounds that were tested, formic acid had the most severe influence on the hydrolysis rate resulting in a complete inactivation of the two enzyme mixtures.  相似文献

Abstract Growth studies of Clostridium thermohydrosulfuricum Rt8.B1 demonstrated that glucose and xylose were used simultaneously when supplied together at nonlimiting concentrations in pH-controlled batch culture. Under conditions of hyperbolic growth, both catabolite repression and inducer exclusion were absent. Glucose did not repress xylose metabolism (i.e. xylose permease and xylose isomerase genes were expressed in the presence of glucose and were not subject to catabolite inhibition when glucose was added to cultures growing on high concentrations of xylose). The kinetics of glucose and xylose utilisation indicated that separate systems were present for the uptake of these substrates when supplied together. Glucose utilisation was biphasic, indicating high- and low-affinity systems for glucose uptake. Xylose utilisation was directly proportional to the xylose concentration, suggesting a facilitated diffusion mechanism was operative for uptake.  相似文献

The ability of a recombinant Saccharomyces yeast strain to ferment the sugars glucose, xylose, arabinose and galactose which are the predominant monosaccharides found in corn fibre hydrolysates has been examined. Saccharomyces strain 1400 (pLNH32) was genetically engineered to ferment xylose by expressing genes encoding a xylose reductase, a xylitol dehydrogenase and a xylulose kinase. The recombinant efficiently fermented xylose alone or in the presence of glucose. Xylose-grown cultures had very little difference in xylitol accumulation, with only 4 to 5g/l accumulating, in aerobic, micro-aerated and anaerobic conditions. Highest production of ethanol with all sugars was achieved under anaerobic conditions. From a mixture of glucose (80g/l) and xylose (40g/l), this strain produced 52g/l ethanol, equivalent to 85% of theoretical yield, in less than 24h. Using a mixture of glucose (31g/l), xylose (15.2g/l), arabinose (10.5g/l) and galactose (2g/l), all of the sugars except arabinose were consumed in 24h with an accumulation of 22g ethanol/l, a 90% yield (excluding the arabinose in the calculation since it is not fermented). Approximately 98% theoretical yield, or 21g ethanol/l, was achieved using an enzymatic hydrolysate of ammonia fibre exploded corn fibre containing an estimated 47.0g mixed sugars/l. In all mixed sugar fermentations, less than 25% arabinose was consumed and converted into arabitol.  相似文献

Specific growth rates of Bacteroides thetaiotaomicron NCTC 10582 with either glucose, arabinose, mannose, galactose or xylose as sole carbon sources were 0.42/h, 0.10/h, 0.38/h, 0.38/h and 0.16/h respectively, suggesting that hexose metabolism was energetically more efficient than pentose fermentation in this bacterium. Batch culture experiments to determine whether carbohydrate utilization was controlled by substrate-induced regulatory mechanisms demonstrated that mannose inhibited uptake of glucose, galactose and arabinose, but had less effect on xylose. Arabinose and xylose were preferentially utilized at high dilution rates (D > 0.26/h) in carbon-limited continuous cultures grown on mixtures of arabinose, xylose, galactose and glucose. When mannose was also present, xylose was co-assimilated at all dilution rates. Under nitrogen-limited conditions, however, mannose repressed uptake of all sugars, showing that its effect on xylose utilization was strongly concentration dependent. Studies with individual D-ZU-14C]-labelled substrates showed that transport systems for glucose, galactose, xylose and mannose were inducible. Measurements to determine incorporation of these sugars into trichloroacetic acid-precipitable material indicated that glucose and mannose were the principal precursor monosaccharides. Xylose was only incorporated into intracellular macromolecules when it served as growth substrate. Phosphoenolpyruvate:phosphotransferase systems were not detected in preliminary experiments to elucidate the mechanisms of sugar uptake, and studies with inhibitors of carbohydrate transport showed no consistent pattern of inhibition with glucose, galactose, xylose and mannose. These results indicate the existence of a variety of different systems involved in sugar transport in B. thetaiotaomicron.  相似文献

The cellulases that strains of Streptomyces albogriseolus, S. nitrosporeus, and Micromonospora melanosporea produce when grown on untreated ballmilled bagasse were investigated. Optimum conditions for extracellular cellulase production and activity were determined to be growth at pH 6.7-7.4 and 25-35 degrees C for 4-5 days and assay at pH 5.0-6.0 and 45-55 degrees C, respectively. The endoglucanases were thermally stable at 50 degrees C, but the Avicelases had a half-life of approximately 24 h at this temperature. Nearly half of the endoglucanases and almost all of the Avicelases were absorbed on ballmilled bagasse after 15 min incubation at 50 degrees C. The beta-glucosidases were found to be mainly intracellular or cell wall bound. These mesophilic actinomycetes concomitantly produced xylanases and beta-xylosidases with cellulases that, apart from cellobiose and glucose, also release xylose from bagasse. This feature may be advantageous in the commerical application of the enzymes of mesophilic actinomycetes for the saccharification of natural cellulosic substrates.  相似文献

In ruminant diets, soluble sugar is an important factor in the digestive process. The objective of this study was to evaluate the effects of the source and dose of soluble sugars, under controlled pH conditions, on the in vitro digestibility of DM, fibre fractions (NDF and ADF) and cell wall neutral monosaccharides of corn silage. Silage was collected from several points in a silage mass from a bunker silo, oven-dried at 55°C and ground through a 1-mm screen. Sub-samples were combined with sugars to compose the treatments, in a 5 × 5 factorial arrangement, as a combination of five soluble sugar sources (glucose, fructose, arabinose, xylose and sucrose) and five sugar doses (0, 100, 200, 300 and 400 g/kg sugar in DM corn silage), respecting the following proportions of sugar : corn silage, 0 : 100, 10 : 90, 20 : 80, 30 : 70, 40 : 60 represented by the sugar doses, respectively. An in vitro test was performed to determine the true digestibility (D) of the chemical entities (DM, NDF and ADF) and cell wall monosaccharides (glucose = gluc, arabinose = arab and xylose = xyl). During the first 12 h of incubation, the pH was maintained above 6.0 by the addition of 2.5 N NaOH. The concentrations of neutral monosaccharides (arabinose, xylose and glucose) were determined by GLC. The soluble sugars decreased the digestibility of corn silage followed by pH reduction, especially at doses higher than 200 g/kg sugar. Overall, xylose, followed by sucrose, fructose and arabinose, had greater impacts on DM digestibility, whereas fibre digestibility was impaired by sucrose at all doses. Xylose and fructose had greater impacts on NDF digestibility at 300 and 400 g/kg sugar. Although xylose impaired the Dgluc in the cell wall in all doses. All doses of glucose improved the Dgluc and Dxyl in the cell wall.  相似文献

Studies on reconstituted mixtures of extensively purified cellobiohydrolases I and II and the five major endoglucanases of the fungus Penicillium pinophilum have provided some new information on the mechanism by which crystalline cellulose in the form of the cotton fibre is rendered soluble. It was observed that there was little or no synergistic activity either between purified cellobiohydrolases I and II, or, contrary to previous findings, between the individual cellobiohydrolases and the endoglucanases. Cotton fibre was degraded to a significant degree only when three enzymes were present in the reconstituted enzyme mixture: these were cellobiohydrolases I and II and some specific endoglucanases. The optimum ratio of the cellobiohydrolases was 1:1. Only a trace of endoglucanase activity was required to make the mixture of cellobiohydrolases I and II effective. The addition of cellobiohydrolases I and II individually to endoglucanases from other cellulolytic fungi resulted in little synergistic activity; however, a mixture of endoglucanases and both cellobiohydrolases was effective. It is suggested that current concepts of the mechanism of cellulase action may be the result of incompletely resolved complexes between cellobiohydrolase and endoglucanase activities. It was found that such complexes in filtrates of P. pinophilium or Trichoderma reesei were easily resolved using affinity chromatography on a column of p-aminobenzyl-1-thio-beta-D-cellobioside.  相似文献

Lignocellulosic biomass is the most abundant bioresource on earth containing polymers mainly consisting of d ‐glucose, d ‐xylose, l ‐arabinose, and further sugars. In order to establish this alternative feedstock apart from applications in food, we engineered Pseudomonas putida KT2440 as microbial biocatalyst for the utilization of xylose and arabinose in addition to glucose as sole carbon sources. The d ‐xylose‐metabolizing strain P. putida KT2440_xylAB and l ‐arabinose‐metabolizing strain P. putida KT2440_araBAD were constructed by introducing respective operons from Escherichia coli. Surprisingly, we found out that both recombinant strains were able to grow on xylose as well as arabinose with high cell densities and growth rates comparable to glucose. In addition, the growth characteristics on various mixtures of glucose, xylose, and arabinose were investigated, which demonstrated the efficient co‐utilization of hexose and pentose sugars. Finally, the possibility of using lignocellulose hydrolysate as substrate for the two recombinant strains was verified. The recombinant P. putida KT2440 strains presented here as flexible microbial biocatalysts to convert lignocellulosic sugars will undoubtedly contribute to the economic feasibility of the production of valuable compounds derived from renewable feedstock.  相似文献

Abstract Butyrivibrio fibrisolvens is a fibrolytic ruminal bacterium that degrades hemicellulose and ferments the resulting pentose sugars. Washed cells of strain D1 accumulated radiolabelled xylose ( K m= 1.5 μ M) and arabinose ( K m= 0.2 μ M) when the organism was grown on xylose, arabinose, or glucose, but cultures grown on sucrose or cellobiose had little capacity to transport pentose. Glucose and xylose inhibited transport of each other non-competitively. Both sugars were utilized preferentially over arabinose, but since they did not inhibit transport of arabinose, it appeared that the preference was related to an internal metabolic step. Although the protonmotive force was completely abolished by ionophores, cells retained some ability to transport pentose. In contrast, the metabolic inhibitors iodoacetate, arsenate, and fluoride had little effect on protonmotive force but caused a large decrease in intracellular ATP and xylose and arabinose uptake. These results suggested that high-affinity, ATP-dependent mechanisms were responsible for pentose transport and hexose sugars affected the utilization of xylose and arabinose.  相似文献

以4个不同烤烟品种为研究材料,采用盆栽试验,运用高效毛细管区带电泳法测定各品种根际土、非根际土、根系及叶片中的单糖组分及含量,并分析其相关关系,探究根系分泌物中糖类的分泌特性。结果表明:在各样品中,共检出木糖、葡萄糖、半乳糖、核糖、阿拉伯糖和鼠李糖6种糖;不同品种根际土、非根际土、根系及叶片中检出的糖组分及含量均存在差异;同一品种中,叶片最高,根系次之,根际土和非根际土最低;相关性分析表明,木糖、阿拉伯糖、葡萄糖、鼠李糖和半乳糖总量在根际土、非根际土、叶片和根系间呈正相关关系,各单糖组分间均呈正相关关系,部分组分呈显著或极显著相关关系。研究表明,不同烤烟品种根系分泌这些单糖存在品种差异,且根系分泌单糖可能是一个沿浓度梯度的扩散过程。  相似文献

An endogenous wheat (Triticum aestivum) flour endoxylanase was purified to homogeneity from a crude wheat flour extract by ammonium sulfate precipitation and cation-exchange chromatography. The 30-kD protein had an isoelectric point of 9.3 or higher. A sequence of 19 amino acids at the NH2 terminus showed 84.2% identity with an internal sequence of 15-kD grain-softness protein, friabilin. High-performance anion-exchange chromatography and gel-permeation analysis of the hydrolysis products indicated the preferential hydrolysis of highly branched structures by the enzyme; wheat arabinoxylan and rye (Secale cereale) arabinoxylan (high arabinose to xylose ratios) were hydrolyzed more efficiently by this enzyme than oat (Avena sativa) spelt xylan (low arabinose to xylose ratios). The release of the hydrolysis products as a function of time suggested that the endoxylanolytic activity was associated with the release of arabinose units from the polysaccharides, suggesting that the enzyme action is similar to that by endoxylanases from Ceratocystis paradoxa, Aspergillus niger, and Neurospora crassa. Although the enzyme released arabinose from arabinoxylan, it did not hydrolyze p-nitrophenyl-alpha-L-arabinofuranoside. From the above, it follows that the enzyme, called arabinoxylanase, differs from most microbial endoxylanases and from an endoxylanase purified earlier from wheat flour.  相似文献

糖类(即碳水化合物)是土壤有机质的重要组成部分, 经生物化学降解形成不同结构的单糖。土壤中的中性单糖也叫中性糖, 主要包括木糖、核糖、阿拉伯糖、葡萄糖、半乳糖、甘露糖、岩藻糖和鼠李糖。其中, 植物来源的糖主要为五碳糖, 如木糖和阿拉伯糖; 微生物来源的糖主要包括半乳糖、甘露糖、岩藻糖、鼠李糖等六碳糖。研究中常利用六碳糖和五碳糖的比例指示微生物和植物对土壤有机碳的相对贡献。中性糖是微生物重要的碳源和能量来源, 在团聚体的形成过程中扮演着重要角色。该文整合了近30年土壤中性糖的研究进展, 对比了提取中性糖的常用方法, 分析了不同土地利用类型和不同土壤组分中中性糖的含量、来源和周转特征, 综述了影响中性糖含量和分布的主要环境因素。结果表明, 中性糖在耕地土壤中的绝对含量和相对含量均显著低于针叶林、阔叶林、草地和灌丛4种土地利用类型。(半乳糖+甘露糖)/(阿拉伯糖+木糖)(GM/AX)在不同土地利用间差异不显著, 而(鼠李糖+岩藻糖)/(阿拉伯糖+木糖)(RF/AX)则表明草地土壤中的微生物来源的中性糖含量高于针叶林和耕地。不同密度的土壤组分中, 轻质组分中中性糖的含量比重质组分高, 重质组分中微生物来源的中性糖较多; 就不同粒径(或团聚体)而言, 黏粒(或微团聚体)中微生物来源的中性糖含量更丰富。有关影响土壤中性糖含量和分布的因素的研究, 目前主要集中在人为活动(如耕种和放牧等), 而有关温度、降水等自然环境因素影响的研究较少。  相似文献
