菘蓝二倍体及其同源四倍体遗传差异的ISSR分析   总被引:1,自引:0,他引:1

以1个菘蓝二倍体及其10个同源四倍体株系为材料,利用19个随机扩增ISSR引物分析其遗传差异性,为菘蓝多倍体诱变的基因表达调控以及遗传改良提供依据。结果显示:菘蓝二倍体与其同源四倍体及四倍体之间的ISSR多态性有明显差异,除主要遗传位点相同外,有些四倍体株系扩增条带数多于二倍体,有些四倍体株系扩增条带数少于二倍体,四倍体株系间亦有多态位点;11个株系共扩增111条多态性条带,多态性达55.22%。聚类分析显示,不同四倍体株系与二倍体的遗传差异大小亦不同。研究表明,菘蓝二倍体与其同源四倍体具有中等偏高的遗传差异性。  相似文献

Vicia cracca diploids and autotetraploids are highly parapatric in Europe; tetraploids reside in western and northern part, whereas diploids occupy much drier south-eastern part. They meet together in a Central European contact zone. This distribution pattern raised questions about a transformative effect of polyploidization on plant performance and environmental tolerances. We investigated plant survival, growth, and seed production in two water regimes in a common garden experiment using seeds collected from five localities in the Central European contact zone where diploids and tetraploids occur in sympatry. Obtained data imply that tetraploids of V. cracca are not generally superior in performance to diploids. Significantly larger seeds from tetraploid mother plants collected in the field were not correlated with greater stature of the seedlings. Nonetheless, tetraploids might have a potential to out-compete diploids in the long run due to the tetraploids’ ability of greater growth which manifested in the second year of cultivation. Considering the response of diploids and tetraploids to water supply, drought stressed tetraploids but not diploids produced a higher proportion of aborted seeds than watered ones, which implies that tetraploids are more drought susceptible than diploids. On the other hand, decreased plant height in drought stresses tetraploids, which simultaneously increased total seed production, may suggest that tetraploids have a greater ability to avoid local extinction under unfavourable conditions by enhancing biomass allocation into production of seeds at the cost of lower growth. The significant interaction between ploidy level and locality in several traits suggests possible polyfyletic origin of tetraploids and the necessity to clarify the history of the tetraploids in Europe.  相似文献

Polyploid evolution is often considered a mechanism of instant speciation; yet the establishment of rare tetraploids within diploid populations may be constrained by a frequency-dependent mating disadvantage (minority cytotype exclusion principle). I tested this hypothesis using experimental populations of Chamerion angustifolium (Onagraceae) that contained different proportions of tetraploids and diploids. Fitness, measured as total seed production over the entire flowering season, was calculated from a census of flower number and estimates of ovule number per flower and proportion of seed set per fruit. The fitness of tetraploids relative to diploids was frequency dependent, increasing from 0.4, when tetraploids were rare, to 0.7 when at 50% and 1.15 when they were in the majority (67%). This pattern exists because of a negative relationship between tetraploid frequency and seed set per fruit in diploids. Seed set in tetraploids was independent of cytotype frequency. The frequency-independent effect in tetraploids reflects higher assortative mating, partly because of non-random patterns of bee visitation. Bees visited a disproportionately high number of diploid inflorescences; however, the proportion of successive flights between tetraploids increased above random expectations as the frequency of tetraploids decreased. These results provide the first experimental test of frequency-dependent fitness in diploid-polyploid mixtures and suggest an important role for more gradual, population processes governing the evolution of polyploidy in natural populations.  相似文献

This paper describes an efficient colchicine-mediated technique for the in vitro induction of hop tetraploids and its confirmation by flow cytometry. A window of conditions generated a high percentage (>20%) of tetraploid induction, with the highest induction (25.6%) achieved with 0.05% colchicine for 48 h. Colchicine-induced tetraploids remained stable after 6 months in soil. Leaf characteristics of diploid and tetraploid hops were compared, and it was determined that stomatal length and width are suitable parameters for identifying putative hop tetraploids. As well as generating tetraploids, this technique generates mixoploid hops. Calli, derived from mixoploid leaves, were induced to form shoot buds and shoots. Individual shoots were classed as diploid, mixoploid or tetraploid after screening by flow cytometry. This callus-based technique can be employed when a genome-doubling agent generates mixoploids but fails to generate tetraploids.  相似文献

Chromosome pairing and chiasma formation were studied in natural and induced tetraploids (2n = 28) of Lathyrus odoratus (induced), Lathyrus pratensis (natural and induced), Lathyrus sativus (induced), and Lathyrus venosus (natural), as well as in triploids of L. pratensis and diploids of L. odoratus, L. pratensis, and L. sativus. All natural tetraploids appeared to be autotetraploids and their meiotic metaphase I behaviour was very similar to that of the induced autotetraploids, with average numbers of pairing partner switches exceeding 4 or even 5. Multivalent frequencies were high, but the numbers of chiasmata were not much higher than necessary to maintain the configurations. Interstitial chiasmata were common, but not predominant. Fertility was reduced, but sufficient for predominantly vegetatively reproducing species. The triploids of L. pratensis had an even higher multivalent frequency than the tetraploids, but still produced some viable progeny at or close to the tetraploid level, suggesting that in mixed populations of diploids and tetraploids, triploids can contribute to gene flow between the ploidy levels. There was no significant correlation between chiasma frequency and ring bivalent frequency in the diploids and multivalent frequency in the corresponding tetraploids. In the tetraploids, chiasma frequency and multivalent frequency were negatively correlated.  相似文献

本文对分布于云南和四川金沙江河谷的云南山茶花C. reticulata及其两个近缘种(怒江山茶C.saluenensis和西南山茶C.pitardii)进行了细胞学研究。34个居群的云南山茶花中,21个居群是四倍体类型(2n=60),11个居群是六倍体类型(2n=90),另2个居群为二倍体(2n=30),云南山茶花的四倍体类型为首次发现,并且进行了核形态研究。四倍体和六倍体的花粉母细胞减数分裂染色体构型在大多数居群都为二价体(四倍体中30个二价体,六倍体中45个二价体),少数居群或是个体除二价体为主外还出现单价体和四价体,六倍体类型没有出现六价体构型。根据减数分裂的构型,我们认为,四倍体和六倍体分别为异源四倍体和异源六倍体,少数四价体的存在表明染色体有部分同源性。所有四倍体和六倍体的体细胞间期核特征和前期染色体形态特征基本相似。中海拔(1800m)以上的四倍体的云南山茶花外部形态特征与六倍体类型比较相似,而低海拔(1100~1800 m)的四倍体类型的外部形态特征则有些不同,但核形态结构是比较相似的。四倍体与六倍体类型地理分布是连续的,并与近缘的二倍体种怒江山茶、西南山茶重叠分布。  相似文献

The chromosome numbers of 889 plants from 222 British populationsof Ranunculus ficaria were counted. 66.1 per cent of the populationscontained diploids (49.1 per cent of plants), 4.1 per cent containedtriploids (5.5 per cent of plants) and 43.2 per cent tetraploids(45.4 per cent of plants); 13.1 per cent of the populationscontained two or more polyplotypes. At the fruiting stage, tetraploidplants can be reliably distinguished from diploids by the presenceof axillary bulbils in the tetraploids and their absence inthe diploids. The production of well-developed heads of achenes,which is more characteristic of the diploids than of the tetraploids,is a less reliable character. Diploid plants are widespreadthroughout the British Isles but tetraploids are more commonin the east than in the west; 17.6 per cent of the diploid plantshad B-chromosomes and these plants were virtually confined tosouthern England and the Midlands. The occurrence of triploidsand of mixed populations of diploids and tetraploids are bothshown to be more common than previously supposed.  相似文献

BACKGROUND AND AIMS: Polyploidy is a central force structuring genetic diversity in angiosperms, but its ecological significance and modes of origin are not fully understood. This work investigated the patterns of coexistence and molecular relatedness of polyploids in the perennial herb, Arnica cordifolia. METHODS: The local- and broad-scale distributions of cytotypes were analysed using flow cytometry. Samples were collected from both roadside and understorey habitats to test the hypothesis of niche separation between triploids and tetraploids. The nuclear rDNA internal transcribed spacer (ITS) and plastid rpl16 spacer, trnL intron plus trnL-trnF spacer and trnK 3' intron regions were sequenced. KEY RESULTS: Broad-scale sampling established that both triploids and tetraploids were common throughout the range of the species, pentaploids were rare, and diploids were not found. Local-scale sampling revealed coexistence of both triploids and tetraploids within the majority of sites. Triploids and tetraploids were equally represented in the understorey and roadside habitat. Triploids were more variable than tetraploids, but both cytotypes shared polymorphisms in ITS. CONCLUSIONS: Coexistence of cytotypes appears to be the norm in A. cordifolia, but habitat differentiation (roadside vs. understorey) is not supported as a coexistence mechanism. Molecular analyses supported multiple events creating triploids but revealed a lack of variation in the tetraploids. Additionally, sequence polymorphisms in ITS suggested a hybridization event prior to polyploidization.  相似文献

The present paper concerns a morphological and cytological comparison between the naturally occurring diploid and the colchicine induced tetraploid ofTorenia fournieri Lind.Treatment of growing points of young seedlings with 0.20% aqueous colchicine for 10 hours on two successive days was most effective in inducing tetraploidy. The induced tetraploids showed the usual gigas characters associated with autotetraploidy. There was a significant reduction in pollen fertility and seed setting of tetraploids as compared to their diploid progenitors.The chromosome numbers of the diploids and the induced tetraploids were determined as n=9 and n=18, respectively. The diploids showed normal meiosis whereas tetraploids were characterized by a high frequency of unequal distribution of chromosomes at anaphase. The induced tetraploids possessed larger flowers. The variability in flower size of the tetraploids offers a good scope for evolving a better horticultural variety of this species.  相似文献

Experimental crosses between diploids, triploids and tetraploids ofHieracium echioides were made to examine mating interactions. Specifically, cytotype diversity in progeny from experimental crosses, intercytotype pollen competition as a reproductive barrier between diploids and tetraploids, and differences in seed set between intra- and intercytotype crosses were studied. Only diploids were found in progeny from 2x × 2x crosses. The other types of crosses yielded more than one cytotype in progeny, but one cytotype predominated in each cross type: diploids (92%) in 2x × 3x crosses, tetraploids (88%) in 3x × 2x crosses, triploids (96%) in 2x × 4x crosses, triploids (90%) in 4x × 2x crosses, tetraploids (60%) in 3x × 3x crosses, pentaploids (56%) in 3x × 4x crosses, triploids (80%) in 4x × 3x crosses and tetraploids (88%) in 4x × 4x crosses. No aneuploids have been detected among karyologically analyzed plants. Unreduced egg cell production was detected in triploids and tetraploids, but formation of unreduced pollen was recorded only in two cases in triploids. Triploid plants produced x, 2x and 3x gametes: in male gametes x (92%) gametes predominated whereas in female gametes 3x (88%) gametes predominated. Cytotype diversity in progeny from crosses where diploids and tetraploids were pollinated by mixture of pollen from diploid and tetraploid plants suggested intercytotype pollen competition to serve as a prezygotic reproductive barrier. No statistically significant difference in seed set obtained from intra- and intercytotype crosses between diploids and tetraploids was observed, suggesting the absence of postzygotic reproductive barriers among cytotypes.  相似文献

Polyploids are overrepresented in invasive species. Yet, the role of genetic diversity and drift in colonization success of polyploids remains unclear. Here, we investigate genetic diversity, genetic differentiation and small-scale genetic structure in our model system, the three geo-cytotypes of Centaurea stoebe: monocarpic diploids and polycarpic (allo)tetraploids coexist in the native range (Eurasia), but only tetraploids are reported from the invasive range (North America). For each geo-cytotype, we investigated 18–20 populations varying in size and habitat type (natural vs. ruderal). Population genetic analyses were conducted at eight microsatellite loci. Compared to diploids, tetraploids revealed higher genetic diversity and lower genetic differentiation, whereas both were comparable in tetraploids between both ranges. Within spatial distances of a few meters, diploid individuals were more strongly related to one another than tetraploids. In addition, expected heterozygosity in diploids increased with population size and was higher in natural than in ruderal habitats. However, neither relationship was found for tetraploids. The higher genetic diversity of tetraploid C. stoebe may have enhanced its colonization abilities, if genetic diversity is correlated with fitness and adaptive capabilities. Furthermore, the inheritance of a duplicated chromosome set as well as longevity and frequent gene flow reduces drift in tetraploids. This counteracts genetic depletion during initial introductions and in subsequent phases of small or fluctuating population sizes in ruderal habitats. Our findings advocate the importance of studying colonization genetic processes to gain a more mechanistic understanding of the role of polyploidy in invasion dynamics.  相似文献

Camellia reticulata and its two allied species, C. saluenensis and C. pitardii,all from the Jinshajiang Valley of Yunnan and Sichuan, were cytologically studied in this work. Among 34 populations of C.reticulata studied here, 21 were found to be tetraploids (2n=60), 11 hexaploids (2n = 90), and the remaining two diploids (2n = 30). Tetraploid was reported in C. reticulata for the first time and its karyomophology was studied. In meiosis of pollen mother cells of the tetraploids and the hexaploids, only bivalents were observed in most of the populations, although in several populations or individuals a few univalents or tetravalents appeared. In the hexaploids, hexavalents were not observed. Thus, the meiotic chromosome cofigurations showed that the tetraploids and hexaploids in C.reticulata allopolyploids, and the appearance of a few tetravalents in some populations might indicatehomology of chromosomes in different genomes to some extent. The characteristics of the resting nuclei and the prophase chromosomes of all the tetraploids and hexaploids were quite similar to each other. The tetraploids of C.reticulata from the localities with the altitude of over 1800 m were gross-morphologically similar to the hexaploids, but those growing between 1100 ~1800 m were different to some extent from the hexaploids. In geographical distribution, the tetraploids and the hexaploids were continuous, and overlapped with C. saluenensis and C. pitardii.  相似文献

A survey of restriction fragment polymorphism in mitochondrial DNA of three subspecies of Carassius auratus throughout four provinces in China was undertaken using 17 restriction enzymes. Two carp, Cyprinus carpio rubbrofuscus and Cyprinus carpio carpio, were included as the outgroup. A total of 16 haplotypes was observed: 5 in tetraploids of C. auratus auratus; 8 in hexaploids of C. auratus auratus; and 2 in C. auratus gibelio and C. auratus cuvieri, respectively. The tetraploids and hexaploids share three common haplotypes as I, V, and VI. C. a. Cuvieri may have diverged first among the three subspecies. Interestingly, C. a. auratus and C. a. cuvieri did not form monophyletic clades, which indicated that the classification of Carassius auratus required further studies. The current hypothesis, that hexaploids originated from tetraploids by a polyploidy event, is less favorable, based on the distribution of haplotypes and the lower diversity in tetraploids than in hexaploids. Our data also indicate that divergence of hexaploids and tetraploids might be recent and mtDNA polymorphism existed before the divergence. Meanwhile, genetic isolation exists between the hexaploids and the tetraploids.  相似文献

Citrus limon (L.) Osbeck cultivated all over the world is a valuable source of aromatic essential oil. To develop tetraploids of C. limon, four different concentration of colchicine (0.025, 0.05, 0.1 and 0.2%; w/v) and three varied exposure time (12, 24 and 48 h) were employed. The ploidy level of diploids (2n = 2x = 18) and tetraploids (2n = 4x = 36) were determined by direct chromosome count and confirmed by flow cytometric analyses. Successful result with maximum tetraploidy frequency was observed in plantlets developed from seeds treated with 0.025% colchicine for 24 h. Morphological and stomatal characteristics indicated that tetraploids were taller with increased leaf and root length. On the other side, the leaves of tetraploids had fewer and larger stomata with a greater number of chloroplasts in guard cells in contrast with diploids. GC–GC/MS analyses showed cyclic monoterpene, limonene had increased significantly in tetraploids and was further confirmed by HPLC quantification. RT-PCR analyses revealed unaltered expression of monoterpene synthase, sesquiterpene synthase and flavone synthase and remarkable upregulation of genes such as limonene synthase, chalcone synthase and phenylalanine ammonia lyase in tetraploids. Antioxidant activity of essential oil of tetraploids was higher than diploids in all the five test systems studied. Overall, the findings of the present study prove that colchicine induced tetraploidy in C. limon could be a greater source of essential oil with improved composition and of economic significance.  相似文献

以多胚系山金柑(Fortunella hindsii Swingle)为材料,采用“观根辨叶看油胞”形态初选法,从1289株实生后代筛选出疑似四倍体(双二倍体)8株,流式细胞仪检测和SSR分子鉴定表明它们均为同源四倍体,初选准确率100%,群体自然发生率0.62%。对其形态和初生代谢物进行检测,结果显示:山金柑四倍体株高、茎粗、节间数、节间长、气孔密度均显著低于二倍体,而叶片厚度、气孔大小显著高于二倍体;GC-MS分析鉴定到24种初生代谢物,四倍体叶片奎宁酸含量显著高于二倍体,肌醇、4-氨基丁酸和1-棕榈酸单甘油酯含量显著低于二倍体。  相似文献

在检测到鲫鲤F2产生3种不同大小(直径分别为0.13 cm,0.17cm和0.2 cm)类型的卵子基础上,进行了F2(♀)×红鲫(♂)及F2(♀)×四倍体鲫鲤(♂)的交配实验.通过染色体计数和流式细胞仪分析,在F2(♀)×红鲫(♂)后代中获得了四倍体、三倍体、二倍体鱼;在F2(♀)×四倍体鲫鲤(♂)后代中获得了四倍体和三倍体鱼.这两个交配组合后代中出现的不同倍性的鱼类为证明鲫鲤F2能产生三倍体、二倍体和单倍体卵子提供了进一步证据.F2(♀)×红鲫(♂)中雄性四倍体鱼的存在说明在四倍体后代中存在基因型为XXXY的个体.对上述两个交配组合后代的四倍体鱼和三倍体鱼的性腺结构观察表明四倍体鱼是可育的,而三倍体鱼是不育的.作者认为鲫鲤F2能够产生二倍体和三倍体卵子与核内复制机制和生殖细胞的融合有关.  相似文献

Tetraploid induction by inhibiting mitosis I with heat shock (32, 35, and 38°C), cold shock (1, 4, and 7°C), and nocodazole (0.02 to 1.6 mg/L) was investigated in the hard clam Mercenaria mercenaria. All treatments were applied to fertilized eggs about 5 min before the first cell division at 22 to 23°C, and lasted for 10, 15, and 20 min. Three replicates were produced for each treatment with different parents. The ploidy of resultant larvae and juveniles was determined with flow cytometry. Heat shock of 35 and 38°C was effective in inhibiting mitosis I, producing 54% to 89% tetraploid larvae. Heat shock of 32°C accelerated embryonic development without inhibiting mitosis or producing tetraploids. In all heat-shock groups, the survival to D-stage larvae was lower than in controls, suggesting that heat-shock treatments and tetraploidy were detrimental to larval development. At the juvenile stage, survivors from heat-shock groups contained no tetraploids. Cold shocks suspended the first cell division during the treatment, but produced no tetraploids in the 4°C and 7°C treatment groups. Cold shock of 1°C produced 31% tetraploid larvae in one replicate, with none surviving to juvenile stage. Nocodazole inhibited mitosis I at concentrations of 0.04 mg/L or higher, but did not produce tetraploids. This study indicates that heat shock is most effective in inducing tetraploids through mitosis I inhibition, although none of the induced tetraploids survived to juvenile stage.  相似文献

Polyploidization is a common route to plant diversification. Polyploids often differ from their progenitors in size, flower number, flower size and flowering phenology. Such differences may translate into differences in the intensity of interactions with animals. Here we investigated the impact of the ploidy-related differences in tetraploids and octoploids of the perennial herb Cardamine pratensis on pre-dispersal seed predation by the butterfly Anthocharis cardamines . The probability of escaping attack was lower for octoploids than for tetraploids, even after accounting for the fact that octoploids were larger and had fewer flowers than tetraploids. Flower shoot size was correlated with probability of attack in tetraploids but not in octoploids. Differences in plant traits associated with polyploidization can alter interactions with animals, and animal-mediated differences in trait selection between ploidy types can contribute to their further divergence.  相似文献

采用染色体压片技术对云贵高原17个多星韭居群,共412株个体进行染色体数目和核型研究。结合已有的细胞学资料,对云贵高原多星韭多倍体分布格局进行了研究。9个二倍体居群、6个二倍体与四倍体混生居群分布于云南中部至西北部,12个四倍体居群分布于整个云贵高原。二倍体除香格里拉d和保山居群核型类型为3A型外,其余均为2A型,四倍体除会泽和赫章居群为2 B型外,其余均为2A型。云南西北部四倍体核型不对称指数低于其它分布区,推测该地区四倍体可能起源较早。多星韭二倍体与四倍体核型类型和形态并无明显差异,表明多星韭四倍体可能为同源多倍体。  相似文献
