Summary Calluna vulgaris possesses small roots called hair roots, which in natural conditions are colonized by symbiotic mycorrhizal fungi. A specialized cell surface-consisting of the cell wall and the overlaying mucilage-has been hypothesized to be important for the establishment of ericoid mycorrhizae. In this work the cell surface of hair roots of plants growing in sterile conditions has been characterized by using in situ techniques, integrated when possible, by biochemical analysis. The mucilage is abundant around the apex, while it becomes thinner and thinner on the differentiated parts. Sugar residues such as mannose, glucose and galactose are regularly distributed along the whole root length, while N-acetylglucosamine residues are limited to the differentiated part of the hair root. Cellobiohydrolase-gold complex, used to reveal -1, 4-glucans, regularly labels mucilage and cell walls of apical and differentiated regions. Polygalacturonic acids revealed by monoclonal antibodies are found at the surface of the cap cells and on the cell walls of the inner tissues in the differentiated zones, but never at the surface of the epidermal cells.The labeling continuity between mucilage and cell walls demonstrates that some molecules such as -1, 4-glucans are common to the two compartments, but probably have a different status of aggregation. In maize ( Zea mays L. cv. LG 11) roots cultured in humid air, the presence of hairs was not related to root growth. However, maximum hair length and length of the hair zone could be correlated to the elongation rate of the primary root. Under the growth conditions used, the emergence of root hairs always took place in the extending zone. In more basal regions, rhizodermal cells could not give rise to root hairs. Results were similar for roots preincubated in a buffer solution.  相似文献

Summary The method for determination of human aldehyde dehydrogenase (ALDH) phenotypes by isoelectric focusing of hair root extracts was re-examined. In order to cancel irregularities stemming from various factors which are not related to the ALDH abnormality and to obtain reproducible results, the sample size for isoelectric focusing should be adjusted using mitochondrial malate dehydrogenase or isocitrate dehydrogenase as an internal reference.  相似文献

We have developed a procedure for testing iduronate sulfatase, the enzyme deficient in Hunter syndrome, in single hair roots. Beta-Hexosaminidase was used as the reference enzyme. The ratio of iduronate sulfatase to beta--hexosaminidase, expressed in arbitrary units of activity, is near zero for Hunter patients and greater than 0.6 in almost all roots of normal individuals. Hair roots of Hunter heterozygotes show a characteristic continuum of activity ratios, ranging from totally deficient up to and including the normal range. The results are consistent with the origin of hair roots from a small number of progenitor cells which obey the Lyon hypothesis. The proportion of roots with low activity can be used to discriminate between normal and heterozygous individuals.  相似文献

Ericaceous dwarf shrubs including Calluna vulgaris and Vaccinium spp. occur both in open heathland communities and in forest ecosystems as understory vegetation. Ericaceous shrubs were once thought to form ericoid mycorrhizal associations with a relatively narrow range of ascomycetous fungi closely related to, and including, Rhizoscyphus ericae. However, perceptions have recently changed since the realization that a broader range of ascomycete fungi, and in some cases basidiomycete fungi, can also form associations with the roots of ericaceous plants. We used a combination of molecular approaches, including denaturing gradient gel electrophoresis, terminal restriction fragment length polymorphism, cloning and sequencing, to investigate the diversity of fungi associated with C. vulgaris roots collected across a heathland/native Scots pine forest vegetation gradient. We also determined differences in fungal community composition between roots of co-occurring C. vulgaris and Vaccinium myrtillus in the forest understory. Collectively, the data show that a large diversity of potentially ericoid mycorrhizal fungal taxa associate with roots of C. vulgaris and V. myrtillus, and that ascomycetes were about 2.5 times more frequent than basidiomycetes. The assemblages of fungi associated with C. vulgaris and V. myrtillus were different. In addition, the community of fungi associated with C. vulgaris hair roots was different for samples collected from the forest, open heathland and a transition zone between the two. This separation was partly, but not entirely, due to the occurrence of typical ectomycorrhizal basidiomycetes associated with the hair roots of C. vulgaris in the forest understory. These data demonstrate that forest understory ericaceous shrubs associate with a diverse range of ascomycete and basidiomycete taxa, including typical ectomycorrhizal basidiomycetes.  相似文献

Summary In interphase nuclei the Y-body can be stained by Giemsa after treating the preparations with diluted trypsin solution. The Y-chromosome as studied by Giemsa technique and by the fluorescence technique is found to be preferentially close to the nucleolus.

Iduronate sulfatase, the enzyme deficient in Hunter syndrome, can be readily measured in individual hair roots. Samples from Hunter syndrome hemizygotes had activities at or near the limits of detection. Samples from two mothers of Hunter syndrome patients, one an obligate heterozygote, had lower average iduronate sulfatase activity than the normal mean, and a significant number of hair roots had activity in the pathognomic range. A third mother showed a normal distribution of enzyme activity, and no hair roots were in the range of those from an affected individual. These results are similar to studies on the distribution of other X-linked enzymes in individual hair root samples from heterozygotes. This suggests that hair root iduronate sulfatase assessment is useful in the detection of Hunter syndrome carrier status, but further refinement of the test system is necessary.  相似文献

Emerging evidence suggests that protein phosphatases play an important role in the growth and development of higher plants. We report here on the effects of okadaic acid and calyculin-A, two specific and potent inhibitors of the type-1 and type-2A families of serine/ threonine protein phosphatases, on the growth and development ofArabidopsis thaliana L. roots. Application of these drugs in nanomolar ranges arrested root hair growth, severely affected the shape of cells within the zone of elongation and inhibited root growth rates. Root hair elongation was inhibited by concentrations of okadaic acid and calyculin-A as low as 3 nM. The pleiotropic effects of okadaic acid and calyculin-A point to multiple functions for type-1 and -2A protein phosphatases in controlling root growth and development.We thank Kelly Aldrich for excellent technical assistance. Summary Root hairs have been shown to enhance P uptake by plants growing in low P soil. Little is known of the factors controlling root hair growth. The objective of this study was to investigate the influence of soil moisture and P level on root hair growth of corn (Zea mays L.). The effect of volumetric soil moistures of 22% (M0), 27% (M1), and 32% (M2) and soil (Raub silt loam, Aquic Argiudoll) P levels of, 0.81 (P0), 12.1 (P1), 21.6 (P2), 48.7 (P3), and 203.3 (P4) mol P L–1 initially in the soil solution, on shoot and root growth, P uptake, and root hair growth of corn was studied in a series of pot experiments in a controlled climate chamber. Root hair growth was affected more by soil moisture than soil P. The percentage of total root length with root hairs and the density and length of root hairs on the root sections having root hairs all increased as soil moisture was reduced from M2 to M0. No relationship was found between root hair length and soil P. Density of root hairs, however, was found to decrease with an increase in soil P. No correlation was found between root hair growth parameters and plant P content, further suggesting P plays a secondary role to moisture in regulating root hair growth in soils. The increase in root hair growth appears to be a response by the plant to stress as yield and P uptake by corn grown at M0 were only 0.47 to 0.82, and 0.34 to 0.74, respectively, of that measured at M1 across the five soil P levels. The increase in root hair growth at M0, which represents an increase of 2.76 to 4.03 in root surface area, could offset, in part, the reduced rate of root growth, which was the primary reason for reduced P uptake under limited soil moisture conditions.Journal Paper No. 10,066 Purdue Univ. Germination of surface-disinfected resting spores ofPlasmodiophora brassicae and its infection of turnip hairy root hairs were studied. Surface-disinfected resting spores showed higher germination than non-disinfected resting spores. Root hair infection was most frequent in the section of root formed 1 d before inoculation. Root hair infection began 4 d after inoculation, increased up to 6 d, and continued to increase more slowly until 10 to 12 d after inoculation. Growth ofP. brassicae in the root hair of hairy roots was observed serially. Most primary plasmodia differentiated to mature zoosporangia 8–10 d after inoculation. 植物根系与真菌形成菌根, 在自然生态系统的物质能量循环中具有重要的生态功能。作者在四川省的中国杜鹃园选取银叶杜鹃(Rhododendron argyrophyllum)和繁花杜鹃(R. floribundum), 通过直接扩增杜鹃花根部真菌rDNA-ITS区片段, 来揭示该地区杜鹃花属植物根部真菌的多样性。ITS序列分析结果表明: 从两种杜鹃的根部共检测到41个真菌分类单元, 分别属于子囊菌纲的柔膜菌目(Helotiales)、散囊菌目(Eurotiales)、盘菌目(Pezizales)、假球壳目(Pleosporales)和担子菌纲的蜡壳耳目(Sebacinales)、伞菌目(Agaricales)、Erythrobasidiales、线黑粉菌目(Filobasidiales)。银叶杜鹃和繁花杜鹃根部真菌种类丰富, 包括了杜鹃花类菌根真菌、外生菌根真菌和其他类型真菌, 其中担子菌纲的蜡壳耳目和子囊菌纲的柔膜菌目占有较大比例。  相似文献

利用rDNA ITS序列对锦绣杜鹃菌根真菌的16个菌株进行了分类分析。根据菌株ITS序列全长计算各菌株间序列相似度和遗传距离,并与GenBank中最相似菌株序列构建系统发育树。结果表明:16个菌株在系统树上聚为3个大分支。其中7个菌株在支持率为100%的基础上与树粉孢属真菌Oidiodendron sp.聚为一类;2个菌株与未鉴定的杜鹃花科植物根系真菌unidentified root associated fungi聚为一类,支持率为100%;其他7个菌株在98%的支持率上与几种未命名的欧石楠类菌根真菌  相似文献
