Fourteen plant species exhibiting a wide range of salt resistance as halophytes, semi-resistant glycophytes and sensitive glycophytes, have been grown in nutrient solution culture under low and high salt conditions. Inorganic analyses and shoot sap osmotic pressure values of these plants confirm that osmotic compensation at high salt levels is largely achieved by the accumulation of Na salts. Choline was found in shoots and roots in the range 1.0-0.2 μmol g fr. wt?1 and varied little following salt stress. Trigonelline was found in some of the sensitive glycophytes and did not increase significantly in stressed plants. Betaine levels were high (10 μmol g fr. wt?1) in the shoot of the halophytes at low salt conditions, lower values (1–10 μmol g fr. wt?1) were found in the semi-resistant glycophytes and none detected in the sensitive glycophytes. In the two resistant groups betaine accumulated to higher levels following NaCl stress. Shoot betaine levels always exceeded root levels. Proline occurred in all plants and in all cases was accumulated following NaCl stress. 相似文献

Quaternary ammonium compounds (QAC) are widely used as disinfectants in both medical and food environments. Microbial contaminants are, therefore, regularly exposed to their action and the isolation from clinical and food sources of resistant bacteria continues to be reported in many countries. Resistance to QAC in clinical strains of staphylococci is encoded by one of at least three resistance genes, designated qacA, qacB and qacC. Using hybridisation analysis, we have shown that these QAC resistance genes are also distributed among staphylococcal strains in the food industry. In addition, we have discovered two new resistance determinants in these food isolates, which are now being characterised and sequenced. Although the general level of resistance of pure cultures is low, the resistant strains have originally been isolated after exposure to the recommended user concentration of a commercial brand of QAC.We have also studied resistance to QAC in pseudomonads isolated from the food industry. Their level of resistance is much higher than that found in staphylococci. About 30% of the collected strains were able to grow in 200 μg·ml−1 benzalkonium chloride, the lowest recommended use concentration for this commonly used type of QAC. 相似文献

AIMS: The aim of this work was to establish if the response to tetradecyltrimethylammonium (TDTMA), a representative quaternary ammonium compound (QAC), involves changes in the phospholipid (PL) composition of Pseudomonas putida A ATCC 12633. METHODS AND RESULTS: Pseudomonas putida was exposed to 50 mg l(-1) of TDTMA for 15 min, and PL composition was analysed. With respect to control values, phosphatidic acid and phosphatidylglycerol increased by 140% and 120%, respectively; cardiolipin decreased about 60%. In TDTMA-adapted bacteria, the most significant change was a 380% increase in phosphatidic acid. Accompanying this change was a 130% increase in phosphatidylglycerol and a 70% decrease in cardiolipin. The changes in adapted cells were reverted after two subcultures without biocide. CONCLUSIONS: Pseudomonas putida responded to TDTMA through quantitative changes in PLs with specific variations in the content of phosphatidic acid, phosphatidylglycerol and cardiolipin. These modifications indicated that these PLs are involved in cellular responses to QACs, utilizing phosphatidic acid principally to neutralize the high positive charge density given for the ammonium quaternary moiety from TDTMA. SIGNIFICANCE AND IMPACT OF THE STUDY: The changes in PL composition give a new insight about the response inflicted by Ps. putida when these bacteria are exposed to QACs. 相似文献

Importance of N source on heat stress tolerance due to the accumulation of proline and quaternary ammonium compounds in tomato plants 总被引:1,自引:0,他引:1
Proline and quaternary ammonium compounds (QAC), in addition to being N-rich, are known to accumulate in plants under different environmental stress conditions. The accumulation of N-rich compounds in plants has been shown to confer stress resistance. The aim of our work is two-fold: first, to study the influence of temperature on proline, QAC, and choline metabolism in tomato leaves; and second, to investigate the relationship between N source applied (NO3- or NH4+) and thermal stress resistance in these plants. To do this, experiments were conducted at three different temperatures (10 degrees C, 25 degrees C, 35 degrees C); at each temperature half of the plants received NO3-, and the other half received NH4+. At 35 degrees C the plants had the lowest biomass production with respect to 25 degrees C (optimal temperature) and 10 degrees C (cold stress), suggesting that tomato plants were most affected by heat stress. At 35 degrees C, there were also high levels of choline and proline due to the activation of Delta1-pyrroline-5-carboxylate synthetase (P5CS) and ornithine aminotransferase (OAT), and simultaneous inhibition of proline dehydrogenase (PDH) and proline oxidase (PO). However, plants with NH4+ as the N source exhibited reduced growth with respect to the plants fed with NO3-. This is interesting because, under heat stress (35 degrees C), biomass production, as well as proline and choline accumulation, in NH4+ fed plants was higher than in NO3- fed plants. From this, we concluded that tomato plants fed with NH4+ as the N source show higher tolerance to heat stress (35 degrees C) than plants fed with NO3-. 相似文献

AIMS: The aims of this study were to evaluate the antimicrobial activity of chlorhexidine diacetate (CHX) and benzalkonium chloride (BZK) for strains of Pseudomonas aeruginosa exhibiting increased minimum inhibitory concentrations (MIC) for CHX, and to determine whether residues of chlorhexidine digluconate (CHG) and Hibiscrub (Hib, a formulation containing CHG) affect the susceptibility of P. aeruginosa to these biocides and a number of antibiotics. METHODS AND RESULTS: The bactericidal activity of CHX and BZK was evaluated for strains of P. aeruginosa exhibiting increased MIC for CHX with established suspension and surface disinfection tests. None of the strains of P. aeruginosa exhibiting raised MIC for CHX was less sensitive than the parent strain to CHX or BZK in either method. A test was designed to investigate the effects of dried CHG and Hib residues on P. aeruginosa cells. Exposure of P. aeruginosa to dried residues of CHG or Hib did not result in the organism becoming less sensitive to either biocide or a number of antibiotics. CONCLUSIONS: Pseudomonas aeruginosa strains with raised MIC to CHX were no less sensitive than the parent strain to CHX and BZK in bactericidal investigations. Exposure to dried residues of CHG and Hib did not render P. aeruginosa less sensitive to either of these agents or a number of antibiotics. SIGNIFICANCE AND IMPACT OF THE STUDY: An increase in the MIC for a biocide in a micro-organism does not necessarily result in a failure of the biocide to effectively kill the organism. The residue that remains after the use of an antimicrobial agent can be at a far lower concentration than that initially applied and this study highlights the necessity for further investigations into the effect of residues, at low concentration, on bacterial populations and their role, if any, in the continued problem of antibiotic resistance. 相似文献

The contents of polyamines and quaternary ammonium compounds (QAC), substances involved in several kinds of stress phenomena, were tested in abscisk acid (ABA)-treated barley leaves (Hordeum vulgare L. cv. Georgie) accumulating pro-line. The characteristic parameters of the accumulation of proline induced by ABA [i.e. kinetics of accumulation, synergistic interaction hormone-K(Na)Cl, enhancing effect of Cl-, inhibiting effect of tetraethylammonium chloride (TEA), D-mannose and glucosamine] were tacking as far as polyamine and QAC content was concerned. Moreover: i) ABA slightly decreased the level of spermine and spermidine, slightly increased that of putrescine but did not influence the level of QAC; ii) the content of polyamines was reduced by KCl; iii) treatment with sorbitol increased the level of polyamines and prevented proline accumulation induced by ABA. These results indicate that there is no relationship between ABA-induced proline accumulation, polyamine level and QAC level; furthermore, accumulation of proline by ABA treatment is possible without increasing the levels of polyamines and QAC. 相似文献

Arabidopsis thaliana grows efficiently on GABA as the sole nitrogen source, thereby providing evidence for the existence of GABA transporters in plants. Heterologous complementation of a GABA uptake-deficient yeast mutant identified two previously known plant amino acid transporters, AAP3 and ProT2, as GABA transporters with Michaelis constants of 12.9±1.7 and 1.7±0.3 mM at pH 4, respectively. The simultaneous transport of [1-14C]GABA and [2,3-3H]proline by ProT2 as a function of pH, provided evidence that the zwitterionic state of GABA is an important parameter in substrate recognition. ProT2-mediated [1-14C]GABA transport was inhibited by proline and quaternary ammonium compounds. 相似文献

Controlling and monitoring the residual activity of quaternary ammonium compounds (QACs) are critical for maintaining safe yet effective levels of these agents in the environment. This study investigates the utility of bromophenol blue (BPB) as a safe, rapid and user-friendly indicator to detect in situ residual QACs dried on hard, non-porous surfaces, as well a means to assess their antimicrobial efficacy. At pH 7, BPB has a purple colour which turns blue upon its complexation with QACs such as didecyldimethylammonium chloride (DDAC). BPB itself has no antimicrobial properties up to 400 ppm. Within the range of 0–400 ppm, BPB colour change was tied to specific DDAC antimicrobial performances with a detection threshold of 100 ppm. BPB concentration and application volume could be adjusted such that a colour shift from purple to blue correlated with a set percent reduction (>99·9%) in test bacteria (Staphylococcus aureus and Klebsiella aerogenes). The BPB solutions developed in this study yielded similar colour shifts on polycarbonate and stainless steel surfaces and did not cross-react with chemical ingredients commonly found in sanitizers and disinfectant products. Overall, this study suggests that BPB provides a simple solution to safely monitor the post-application level and biocidal activity of residual dried QACs on surfaces. 相似文献

AIMS: To determine the susceptibility to disinfectants and cross-resistance to antibiotics in Listeria monocytogenes strains isolated from fish products and the fish-processing environment. METHODS AND RESULTS: Minimal inhibitory concentration assessment, using the agar dilution method, showed 108 of 255 L. monocytogenes isolates with low susceptibility to benzalkonium chloride (BC), commonly used in food industries. Most of them are from raw products of farmed fish during processing, while the remaining resistant isolates were mainly from the environment and finished products irrespective of the fish species. Two BC-resistant isolates were resistant to ethidium bromide (EB). The conservation of resistance after plasmid curing suggested that the resistance genes are not plasmid associated. EB accumulation assays demonstrated that the two BC(R) EB(R) isolates used an efflux pump to expel these substrates whereas a different mechanism was probably used by the majority of the strains with BC(R) EB(S) pattern. No cross-resistance was found with antibiotics. CONCLUSIONS: This study highlights the difference in susceptibilities to BC for L. monocytogenes strains isolated from fish-processing plants and in resistance mechanisms to BC developed by these bacteria. SIGNIFICANCE AND IMPACT OF THE STUDY: The presence of BC resistant L. monocytogenes strains could contribute to their adaptation and so explained their survival and persistence in the fish-processing environment. 相似文献

The tissues of the quiescent wheat grain contained free amino acids and quaternary nitrogen compounds. During germination the amino acid levels increased several fold. In the aleurone tissue and starchy endosperm glutamine was the predominant amino acid. Asparagine was predominant in the seedling tissues. Choline and glycine betaine were the principal quaternary nitrogen compounds present. The aleurone tissue and the embryo/seedling contained large quantities of glycine betaine. The increase in free amino acid levels in the aleurone tissue during the first 2 days of germination occurred independently of the embryo. After the second day, the further increase in levels was dependent upon the presence of the embryo and of gibberellic acid (GA). Estimation of the individual amino acids and quaternary nitrogen compounds released from incubating aleurone layers into aqueous media revealed a selective release of some compounds and retention of others. The process was regulated by GA. Possible mechanisms for the release of amino acid and its control by GA are discussed. 相似文献

The primary in-water emergency treatment method for mussel fouling of internal seawater systems of Royal Australian Navy vessels is to flush with a 1% detergent solution containing quaternary ammonium compounds (QAC). Parameters for application of this treatment are based on previous research; however, much of the research has been conducted at small-scales under controlled laboratory conditions. This study examined the efficacy of QAC solutions for treating mussel biofouling under realistic field conditions using experimental seawater piping systems. The efficacy of QAC solutions was highly dependent on the size of mussels present. Chemical treatments comprising 1, 2 and 5% v v–1 QAC solution were effective at killing large (50–92 mm) mussels in the pipework and sea chest of the system following 24 h exposure. In contrast, small mussels (10–30 mm) appeared resilient to the majority of treatment regimes. Differences in water temperature, DO and pH during dosing had no discernible impact on treatment efficacy. 相似文献

Abstract Isolates of Pseudomonas spp. collected from the rhizosphere of sugarcane and cane stalks were screened for their antagonistic activity against Colletotrichum falcatum causing red rot disease in sugarcane. Talc formulations of the selected Pseudomonas spp. isolates improved the sugarcane vegetative sett germination and sugarcane growth under field conditions. Optimal talc formulations were assessed for their effect on induction of systemic resistance against the pathogen in the canes under artificial inoculation. All the four isolates CHAO, EP1, KKM1 and VPT4 were effective in inducing systemic resistance against C. falcatum in two seasons. In other studies, the bacterial formulations were assessed to induce resistance in sugarcane in a sick plot situation. In pathogen-infested soil the isolates KKM1 and CHAO suppressed the red rot disease development in susceptible sugarcane cultivar. Pseudomonas strains also protected sugarcane in a disease-endemic location. Pseudomonas spp treatment substantially improved the cane juice quality parameters affected by the pathogen infection. Standardization of talc formulations and application methods in the field offers potential for large-scale application of biocontrol formulations for the management of red rot disease in sugarcane growing regions. 相似文献

目的 了解我院铜绿假单胞菌获得性肺炎患者病原菌的分布及耐药情况。方法 选取2016年7月至2017年12月我院送检的获得性肺炎患者痰液培养标本,采用VITEK2-COMPACT全自动微生物分析仪进行菌株鉴定和药敏分析。结果 总共检出358株铜绿假单胞菌,主要分布于呼吸内科和ICU。患者主要基础疾病为慢性阻塞性肺疾病、慢性支气管炎、脑梗死、脑出血等。检出的铜绿假单胞菌对复方新诺明耐药率最高,对阿米卡星耐药率最低。铜绿假单胞菌耐药率由低到高依次为妥布霉素、哌拉西林/他唑巴坦、庆大霉素、头孢他啶、头孢哌酮/舒巴坦、头孢唑肟钠、头孢曲松钠、亚胺培南和左氧氟沙星。结论 我院需要加强对碳青霉烯类和含β-内酰胺酶抑制剂药物的管控,减少这些药物的使用强度,各临床科室尤其是呼吸内科和ICU需及时监控铜绿假单胞菌的耐药情况,合理使用抗生素,从而逐步降低病原菌耐药状况。 相似文献

AIMS: To examine the causes for variations in sensitivity and intrinsic tolerance of Pseudomonas aeruginosa to plant volatile compounds. METHODS AND RESULTS: Minimum inhibitory concentrations were determined for a selection of volatile phytochemicals against P. aeruginosa using a microdilution assay. Effects on growth were also assessed in 100-ml broth cultures. The two strains of P. aeruginosa included in the study exhibited intrinsic tolerance to all compounds, with the exception of carvacrol and trans-cinnamaldehyde. The protonophore carbonyl cyanide m-chlorophenylhydrazone increased P. aeruginosa sensitivity to all compounds except trans-cinnamaldehyde, implicating an ATP-dependent efflux mechanism in the observed tolerance. Outer membrane integrity following treatment with test compounds was assessed by measuring sensitization to detergents. Only carvacrol caused damage to the outer membrane of P. aeruginosa. CONCLUSIONS: The intrinsic tolerance of P. aeruginosa strains to plant volatile compounds is associated with an active efflux mechanism and the barrier function of the outer membrane. SIGNIFICANCE AND IMPACT OF THE STUDY: These findings offer an explanation for the intrinsic tolerance to plant volatile compounds exhibited by P. aeruginosa. The study also confirms that the outer membrane-permeabilizing action of carvacrol, previously reported for the gram-negative bacteria Escherichia coli and Salmonella, extends to monoterpene-tolerant strains of P. aeruginosa. 相似文献

Antimicrobial resistance (AMR) has been detected in the microbiota of wildlife, yet little is known about the origin and impact within the ecosystem. Due to the shortage of nonepizootic surveillance, there is limited understanding of the natural prevalence and circulation of AMR bacteria in the wild animal population, including avian species. In this surveillance study, feces from wild birds in proximity to the River Cam, Cambridge, England, were collected and Pseudomonas spp. were isolated. Of the 115 samples collected, 24 (20.9%; 95% CI, 12.6%‒29.2%) harbored Pseudomonas spp. of which 18 (75%; 95% CI, 58%‒92%) had a multiple antibiotic resistance (MAR) index greater than 0.2. No Pseudomonas spp. isolate in this study was pansusceptible. Resistance was found among the 24 isolates against ciprofloxacin (87.5%; 95% CI, 74.3%‒100%) and cefepime (83.3%; 95% CI, 68.4%‒98.2%), both of which are extensively used to treat opportunistic Pseudomonas spp. infections. The prevalence of Pseudomonas spp. in the wild bird feces sampled during this study is greater than previous, similar studies. Additionally, their multidrug resistance profile provides insight into the potential risk for ecosystem contamination. It further highlights the importance of a One Health approach, including ongoing surveillance efforts that help to develop the understanding of how wildlife, including avifauna, may contribute and disperse AMR across the ecosystem. 相似文献

K.A. ALBASHERI AND W.J. MITCHELL. 1995. Maltose metabolism in the obligate anaerobe Clostridium acetobutylicum was studied. The sugar is accumulated via an energy-dependent transport process which is not a phosphotransferase. Cell extracts were incapable of phosphorylating maltose in the presence or absence of phosphoenolpyruvate or ATP, but exhibited hydrolytic activity against a range of glucoside substrates. The activity was predominantly in the soluble fraction of cell extracts, indicating a cytoplasmic location in the cell. Gel filtration on Sephadex G100 indicated the presence of at least two α-glucosidases. One enzyme (maltase) was active with maltose and maltotriose, while the other (pNPGase) hydrolysed isomaltose and several glucoside analogues, but neither showed activity against starch. Both glucosidases were induced by isomaltose, maltose, glucose and starch, but not by xylose, sucrose or cellobiose. In the presence of both glucose and maltose, growing cells showed a preference for glucose, apparently due to regulation of maltose transport, which did not occur in glucose-grown cells. 相似文献

Cationic biocides are widely utilized for surface disinfection. Photosensitizers such as toluidine blue O (TBO) produce reactive oxygen species following light excitation and are being investigated as novel biocides for similar applications. Aspergillus brasiliensis conidia contain melanin which protects against environmental stressors. The negative charge and antioxidant properties of melanin may confer resistance to photosensitizers and other biocides. In this study, the yeasticidal and fungicidal activity benzalkonium chloride (BZC), sodium dichloroisocyanurate (NaDCC) and TBO with red light were examined using quantitative suspension tests. All three biocides were highly effective against Candida albicans and > 5·0 log10 reductions in viability were attainable within 5 minutes. Wild-type A. brasiliensis conidia were highly tolerant to treatment and 0·4 log10 reductions in viability were observed within the same time frame when treated with TBO or BZC. NaDCC was markedly more effective. Inhibition of melanin biosynthesis by culturing with 100 μg ml−1 kojic acid resulted in a hypopigmented phenotype with significantly increased sensitivity to all three biocides. These observations indicate that melanin is a significant contributor towards A. brasiliensis tolerance of biocides and photosensitizers and demonstrate that cationic biocides are poorly suited to applications where the control of A. brasiliensis is required. 相似文献
