The neck organ of adult and immature forms of four marine gymnomeran Cladocera species (Podonidae) has been characterized morphologically by light-scanning- and electron microscopy. Although displaying some special features, a comparison with organs assuming a comparable regulating function in other organisms indicates that the neck organ of the adult Podonidae exhibits ultrastructural evidence of involvement in ion-transporting mechanisms. The extensive ventral plasma membrane infoldings closely connected with the chondriom and the peculiar organization of the cuticular site associated with the subjacent apical cell limits of the organ are among the chief structural specializations. The fact that similar structures are already present in the embryonic neck organ confirms previous indirect evidence that podonid embryos can hypo-osmoregulate as soon as the ion concentration of the brood chamber rises to that of the outside medium.  相似文献

Plectin is a multifunctional cytoskeletal linker protein with an intermediate filament-binding site and sequence elements with high homology to actin-binding domains. Mutations of the human plectin gene as well as the targeted inactivation of its murine analog cause a generalized blistering skin disorder and muscular dystrophy, thus implying its essential role in cells that are exposed to mechanical stress. In the present study we report the characterization of two new domain-specific plectin antibodies as well as ultrastructural localization of plectin in normal human skeletal muscle. Using immunogold electron microscopy, we localized plectin at three prominent sites: 1) Plectin is found at regularly spaced intervals along the cytoplasmic face of the plasma membrane. 2) It is distinctly localized at filamentous bridges between Z-lines of peripheral myofibrils and the sarcolemma and 3) at structures forming the intermyofibrillar scaffold. At the latter two locations, plectin and desmin were found to colocalize. Our ultrastructural analysis suggests that plectin may have a central role in the structural and functional organization of the intermediate filament cytoskeleton in mature human skeletal muscle.  相似文献

The nature of interactions between cells migrating through tissues and their structural surroundings are largely unknown. We have therefore examined the ultrastructural relationship between L5222 rat leukemia cells, moving through the loose connective tissue of the mesentery, and components of the extracellular matrix (ECM). Ultrathin tissue sections, fixed in the presence of ruthenium hexammine trichloride (RHT), revealed the following: Constitutents of fibrillar and nonfibrillar elements of the ECM are in contact with the plasma membrane of L5222 cells. Linear nonfibrillar ECM elements contact the plasma membrane at point-like sites, often associated with root-like structures present within the submembraneous microfilament mesh. Aggregates of ECM material are connected to patch-like cell membrane sites, associated with a condensed, plate-like part of the microfilament mesh. Point-like and patch-like contacts are more numerous at the anterior part of polarized migrating L5222 cells than on the posterior end. In round resting leukemia cells they are evenly distributed around the cell periphery. We suggest that the ECM-cell membrane contacts represent tissue adhesion sites. We therefore hypothesize that in migrating cells a coordinate interaction occurs between the contact sites and the continuous microfilament meshwork which results in a simultaneous backward movement of ECM-membrane contacts on the cell body and in a net forward movement of the whole cell. Since Dembo et al. (1981) present a similar mechanism for in vitro locomotion of granulocytes, we assume that blood cell locomotion in vivo and in vitro depends on similar molecular mechanisms: force generation by the cell, transmembraneous linkage between cytoskeletal and ECM elements, and membrane fluidity. The major difference in blood cell locomotion through a three-dimensional tissue or on a plane substratum would then be given by the distribution of contact sites, occurring around the cell periphery or limited to the ventral cell surface, respectively.  相似文献

Two P388 cell sublines with different levels of resistance to daunomycin (DNM), P388/20 and P388/100 cells (approximately 20- and 100-fold resistance, respectively), undergo a significant (approximately 2-fold) increase in the number of intramembrane particles (IMPs) present at their plasma membrane, as compared to that exhibited by the parental, drug-sensitive P388 (P388/S) cell line. Regardless of the level of resistance, incubation of drug-resistant cells with verapamil, a well known reverting agent of anthracycline resistance, restores the morphology of the plasma membrane in these cells, yielding a pattern in which the number and size distribution of IMPs at both leaflets of the bilayer, become undistinguishable from those displayed by drug-sensitive cells. Furthermore, verapamil did not affect the ultrastructural organization of the plasma membrane of drug-sensitive cells. It is possible that the alterations in the structural organization of the plasma membrane of the antineoplastic-resistant tumor cells, might represent a reliable 'marker' for early diagnosis of drug resistance.  相似文献

Summary— When mouse peritoneal macrophages adherent to glass surface were removed by treatment with triethanolamine and Nonidet P-40, fine thread structures of unique loops were left behind on glass at the sites of cell adhesion. To examine the ultrastructural relationship between such looped threads and cytoskeletal components in glass-adherent macrophages, we successfully used the ‘zinc method’ to remove most of the cytoplasm including nuclei and to expose the cytoskeleton associated with the ventral plasma membrane. The cytoskeleton was seen to be mainly composed of actin filaments forming dense networks. The network contained scattered star-like foci from which actin filaments radiated. When the ventral plasma membrane-cytoskeleton complex was further treated with Nonidet P-40, the membrane was dissolved to expose the glass surface with actin foci persisting on glass. When the complex was removed by further treatment with Nonidet P-40 and DNase I, the looped threads became visible. Confocal laser microscopy of glass-adherent macrophages stained with fluorescent phalloidin showed the preferential distribution of F-actin in the ventral cytoplasm along the plasma membrane, where intense fluorescent spots were also scattered. Confocal interference reflection microscopy revealed densely populated dark dots and striae of focal contact, which corresponded in overall distribution to actin foci and looped threads. These observations suggest that actin cytoskeleton is closely associated with looped threads to reinforce cell adhesion to glass.  相似文献

By the freeze-fracture method it is shown that metabolic depletion of erythrocytes affects three levels of cell organization: the microstructural (erythrocyte form), microstructural (micro-relief of erythrocyte surface) and ultrastructural (ultrastructural state of erythrocyte plasma membranes) ones. As it is established, the size of spikes on the echinocyte surface and that of membrane vesicles budding from a cell coincide with each other. The structural modification of the membrane precedes the stage of erythrocyte crenation. The following model of vesicle budding process is suggested: reduction of ATP level and dephosphorylation of actin-spectrin network--structural modification of the protein and lipid membrane phases with the formation of regions disconnected from the spectrin framework--protrusion of these anomalous regions in the form of spikes--budding of spikes as spherical vesicles.  相似文献

The plasma membrane ultrastructural changes after the action of epidermal growth factor were studied in A-431 cells using freeze-fracture methods. The incubation with EGF (100 ng/ml, 0 degree C, 60 min) led to a decrease in density of intramembrane particles on the P surface of ventral cell membrane, while the number of coated pits increased there. The revealed effects of EGF may be related to direct consequences of EGF-receptor complex formation, because all the temperature dependent steps of its processing were blocked. The data obtained testify to an active involvement of the membrane ventral surface in the formation of cell response towards growth factors.  相似文献

ABSTRACT. The close contact of Giardia lamblia trophozoites with mucosal surfaces produces surface indentations of epithelial cells, that progress to areas of microvilli depletion. This interaction is mediated by the lateral crest, a specialized contractile structure in the ventral disc of the parasite. We have analyzed the plasma membrane of the ventral disc using freeze fracture electron microscopy to study the distribution of large integral proteins, and freeze fracture cytochemistry with filipin to reveal sterol-containing sites. A previously undetected structural specialization was found at the lateral crest as a horseshoe-shaped membrane domain characterized by a near absence of intramembrane particles, in sharp contrast to the remainder of the ventral disc plasma membrane. In addition, the lateral crest showed a striking paucity of cholesterol-containing complexes in replicas of trophozoites treated with filipin. These observations demonstrate that the plasma membrane of G. lamblia displays a microheterogeneity in the planar distribution of cholesterol and intramembrane particles. Both membrane components are noticeably less abundant at the outer rim of the ventral disc where contraction takes place.  相似文献

The fine structure of the brain and ventral nerve cord ganglia neurons has been studied in two subspecies of Chaetogaster lymnaei, one of them being a commensal and the other the kidney endoparasite. The nerve cells of parasitic forms are characterised by the presence of a whole set of structural simplifications. These are the poorer development of endoplasmic reticulum, the greater number of free ribosomes in the cell and the less number of mitochondria with certain alternations in the ultrastructure. The given features are suggested as criteria when studying the influence of parasitic mode of life upon the ultrastructural organization of the cells.  相似文献

The cytochemical and ultrastructural features of lamellar bodies in human skeletal muscle fibers were studied using tannic acid-glutaraldehyde, ruthenium red-glutaraldehyde fixation methods, conventional electron microscopy and the freeze fracture technique. The lamellar bodies consisted of concentric lamellae with a regular spacing of 6.5 +/- 0.2 nm. These structures were found preferentially at the cell periphery closely associated with the plasma membrane, near the nuclear poles and in the space between muscle fiber and satellite cell. The cytochemical and ultrastructural features of the lamellar bodies suggest they are largely composed of phospholipid. It is possible that these structures are involved in muscle membrane maintenance.  相似文献

A portion of the hindgut of Xenopsylla cheopis larvae, the rectal sac, is extraordinarily enlarged and differentiated into a specialized organ displaying well-elaborated transporting epithelia. This organ is unique among insect recta because of gutter-like arrangement of two ultrastructurally distinct cell types with an asymmetrical position of the membranemitochondria complexes. The cells of the dorsal gutter have highly folded apical plasma membranes on the lumen side. The cells of the ventral gutter show deep infoldings of the basal plasma membrane on the haemolymph side. Less elaborate folds originate from the opposite sides of the cells. The nuclei of the dorsal epithelial cells are located in the basal part; those of the ventral cells in the apical portion of the cell. The ultrastructural features of the rectal sac, its ventilation synchronized with rhythmic opening of the anus during phases of active water vapour uptake, as well as the reversible arrest of uptake by experimental occlusion of the anus, reveal that condensation and uptake of atmospheric water take place in this organ.  相似文献

The presence and pattern of pituitary adenylate cyclase activating polypeptide (PACAP) type I (PAC1) receptors were identified by means of pre- and post-embedding immunocytochemical methods in the ventral nerve cord ganglia (VNC) of the earthworm Eisenia fetida. Light and electron microscopic observations revealed the exact anatomical positions of labeled structures suggesting that PACAP mediates the activity of some interneurons, a few small motoneurons and certain sensory fibers that are located in ventrolateral, ventromedial and intermediomedial sensory longitudinal axon bundles of the VNC ganglia. No labeling was located on large interneuronal systems such as dorsal medial and lateral giant axon systems and ventral giant axons. At the ultrastructural level labeling was mainly restricted to endo- and plasma membranes showing characteristic unequal distribution in various neuron parts. An increasing abundance of PAC1 receptors located on both rough endoplasmic reticulum and plasma membranes was seen from perikarya to neural processes, indicating that intracellular membrane traffic might play a crucial role in the transportation of PAC1 receptors. High number of PAC1 receptors was found in both pre- and postsynaptic membranes in addition to extrasynaptic sites suggesting that PACAP acts as neurotransmitter and neuromodulator in the earthworm nervous system.  相似文献

Summary In the epithelium lining the ventral prostate of the rat, complex carbohydrate-containing structures have been studied by means of both light and electron microscopic histochemical methods. According to light microscopy, the free surface and granules of different sizes in the distal cytoplasm of the epithelial cells were found to exhibit positive reactions for complex carbohydrates with 1,2-glycol and acidic groups and sialic acid residues. In addition, secretory substances within the glandular lumen were shown to exhibit positive reactions for similar groups and saccharide residues of complex carbohydrates. In electron microscopy, the surface coat of the plasma membrane, certain elements of the Golgi apparatus and lysosomal dense bodies were found to exhibit positive reactions for glycoproteins with 1,2-glycol groupings. The histophysiological significances of the carbohydrate-containing structures have been discussed with special reference to the known physiological functions of the prostate in the rat.  相似文献

Spatial organization of membranes into domains of distinct protein and lipid composition is a fundamental feature of biological systems. The plasma membrane is organized in such domains to efficiently orchestrate the many reactions occurring there simultaneously. Despite the almost universal presence of membrane domains, mechanisms of their formation are often unclear. Yeast cells feature prominent plasma membrane domain organization, which is at least partially mediated by eisosomes. Eisosomes are large protein complexes that are primarily composed of many subunits of two Bin-Amphiphysin-Rvs domain-containing proteins, Pil1 and Lsp1. In this paper, we show that these proteins self-assemble into higher-order structures and bind preferentially to phosphoinositide-containing membranes. Using a combination of electron microscopy approaches, we generate structural models of Pil1 and Lsp1 assemblies, which resemble eisosomes in cells. Our data suggest that the mechanism of membrane organization by eisosomes is mediated by self-assembly of its core components into a membrane-bound protein scaffold with lipid-binding specificity.  相似文献

In response to calcium influx, synaptic vesicles fuse very rapidly with the plasma membrane to release their neurotransmitter content. An important mechanism for sustained release includes the formation of new vesicles by local endocytosis. How synaptic vesicles are trafficked from the sites of endocytosis to the sites of release and how they are maintained at the release sites remain poorly understood. Recent studies using fast freezing immobilization and electron tomography have led to insights on the ultrastructural organization of presynaptic boutons and how these structural elements may maintain synaptic vesicles and organize their exocytosis at particular areas of the plasma membrane.  相似文献

By utilizing a combination of several ultrastructural techniques, we have been able to demonstrate differences in filament organization on the adherent plasma membranes of spreading and mobile PMN as well as within the extending lamellipodia. To follow the subplasmalemmal filaments of this small amoeboid cell during these kinetic events, we sheared off the upper portions of cells onto glass and carbon surfaces for 30 s--5 min. The exposed adherent membranes were immediately fixed and processed for high-resolution SEM or TEM. Whole cells were also examined by phase contrast microscopy, SEM, and oriented thin sections. Observed by SEM, the inner surface of nonadherent PMN membranes is free of filaments, but within 30 s of attachment to the substrate a three-dimensional, interlocking network of globular projections and radiating microfilaments--i.e., a subplasmalemmal filament complex--is consistently demonstrable (with or without postfixation in OsO4). Seen by TEM, extending lamellipodia contain a felt of filamentous and finely granular material, distinct from the golbule/filament complex of the adjacent adherent membrane. In the spread cell, this golbule-filament complex covers the entire lower membrane and increases in filament-density over the next 2--3 min. By 3--5 min after plating, as the PMN rounds up before the initiation of amoeboid movements, another pattern emerges--circumferential bands of anastomosing filament bundles in which thick, short filaments resembling myosin are found. This work provides structural evidence on the organization of polymerized contractile elements associated with the plasma membrane during cellular adherence.  相似文献

There is mounting evidence for the organization and compartmentation of signaling molecules at the plasma membrane. We find that hormone-sensitive adenylyl cyclase activity is enriched in a subset of regulatory G protein-containing fractions of the plasma membrane. These subfractions resemble, in low buoyant density, structures of the plasma membrane termed caveolae. Immunofluorescence experiments revealed a punctate pattern of G protein α and β subunits, consistent with concentration of these proteins at distinct sites on the plasma membrane. Partial coincidence of localization of G protein α subunits with caveolin (a marker for caveolae) was observed by double immunofluorescence. Results of immunogold electron microscopy suggest that some G protein is associated with invaginated caveolae, but most of the protein resides in irregular structures of the plasma membrane that could not be identified morphologically. Because regulated adenylyl cyclase activity is present in low-density subfractions of plasma membrane from a cell type (S49 lymphoma) that does not express caveolin, this protein is not required for organization of the adenylyl cyclase system. The data suggest that hormone-sensitive adenylyl cyclase systems are localized in a specialized subdomain of the plasma membrane that may optimize the efficiency and fidelity of signal transduction.  相似文献

The body muscle cells of the nematode Ascaris lumbricoides are characterized by massive amounts of intermediate filaments (IF). These occur in all three regions of this giant cell type. They traverse the cytoplasm of the balloon-like belly, which houses the nucleus, and occur as bundles in the arm-like extensions to the nerve. The organization of IF in the third region, the contractile fiber, was analyzed further by serial sections and three-dimensional reconstruction. IF bundles traverse the glycogen-rich lumina of the fiber and reach as baskets around the sarcomeres. Together with numerous dense bodies they form the Z-band-like arrangements. IF bundles reach the plasma membrane at hemidesmosome-like specializations often situated at deep membrane invaginations filled with a fibrillar component of the extracellular matrix. The ultrastructural appearance of IF bundles is connected to the contractional state of the sarcomeres. They appear straight in extended muscle but coil up upon contraction. In the pharynx massive IF bundles are oriented longitudinally. A second type of IF bundles follows the radially oriented sarcomeres. These reveal pronounced Z-band type structures with massive disks. IF surround the sarcomeres and seem to terminate at these disks. We discuss possible functions of the complex IF organization in body muscle and pharynx.  相似文献

A procedure for the isolation of the plasma membranes from zoospores ofBlastocladiella emersonii, which involves the rupturing of the plasma membrane, is described. The ultrastructural organization of the zoospore then permits the removal of the major cytoplasmic organelles as a single unit. Analysis of the membrane lipids indicated that the membrane is composed of glycolipids and neutral lipids, rather than phospholipids. The glycolipid fraction contained only diglycosyldiglycerides, and ≅ 90% of the glycolipid head groups were glucose, indicating that diglycosyldiglycerides comprise the largest single group of lipids in the plasma membrane preparations (≅ 47% of total lipid by weight). The membrane preparation was also characterized by the absence of triglycerides, although it did contain sterols. The preponderance of glycolipids in the plasma membrane preparations can account for the physical-chemical changes previously observed in zoospore plasma membranesin vivo, and in isolated plasma membrane preparations. The presence of glycolipids in both the plasma membrane and γ-particles, which directly interact with each other, via vesicle fusion, to form the chitinous cell wall, and the biochemical role of glycolipids in chitin synthesis in this organism suggest that the glycolipids may be involved in the regulation of zoospore encystment.  相似文献
