The seedlings of two soybean genotypes, Al-tolerant PI 416937 (PI) and Al-sensitive Young, were cultured in the solution containing 0, 25 or 50 μM Al (AlCl3·6H2O) for 24, 36 or 48 h in the hydroponics, and the calluses induced from two genotypes were cultured in medium containing 0, 10, 50 or 100 μM Al for 5, 10 or 15 days, respectively. The effects of Al on growth of seedling roots and calluses, antioxidant enzyme activities of superoxide dismutase (SOD) and peroxidase (POD) and lipid peroxidation were investigated. Under Al stress, PI was more tolerant to Al toxicity than Young at both intact plant and tissue levels and lower concentrations of Al significantly stimulated the root and callus growth of PI. Al application enhanced the activities of SOD and POD and lipid peroxidation in both roots and calluses of two genotypes. Although the differences of SOD activities between two genotypes in response to Al toxicity depended on Al concentration and durations of treatment, SOD activities in the roots of PI were higher than those in the roots of corresponding Young in the presence of Al for 36 or 48 h. Meanwhile, the POD activities in PI roots increased as the Al levels and durations of treatment increased, significantly higher than those in the corresponding Young roots. Moreover, Al-treated PI had significantly lower lipid peroxidation than Young at both root and callus levels. These results suggest that the enhanced antioxidant-related enzyme activities and reduced lipid peroxidation in PI might be one of Al-tolerant mechanisms.  相似文献

Effects of exogenous nickel (Ni: 10 and 200 μM) on growth, mitotic activity, Ni accumulation, H2O2 content and lipid peroxidation as well as the activities of various antioxidative enzymes, such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and glutathione peroxidase (GSH-Px) were investigated in wheat roots. A considerable Ni accumulation in the roots occurred at both the concentrations. Although Ni at 10 μM did not have any significant effect on root growth, it strongly inhibited the root growth at 200 μM. Mitotic activity in the root tips was not significantly affected by exposure of the seedlings to 10 μM Ni; however, it was almost completely inhibited at 200 μM treatment. Ni stress did not result in any significant changes in CAT and APX activities as well as lipid peroxidation. However, H2O2 concentration increased up to 82% over the control in the roots of seedlings exposed to 200 μM Ni. There was a significant decline in both SOD (50%) and GSH-Px (20–30%) activities in the roots when the seedlings were treated with 200 μM Ni. The results indicated that a strong inhibition of wheat root growth caused by Ni stress was not due to enhanced lipid peroxidation, but might be related to the accumulation of H2O2 in root tissue.  相似文献

Imposition of NaCl-salinity stress induced oxidative reactions in root tissue of rice seedlings. A uniform accumulation of proline was marked with the increasing NaCl concentrations. Both peroxide content and lipid peroxidation level (MDA) increased with the salt treatment from the control. CAT, GPx and SOD activities decreased with the increasing NaCl concentrations suggesting a possible oxidative damage to root tissue.  相似文献

The seedlings of hybrid rice were exposed to low(l℃) and high (40℃) temperatures and the permeability of plasma membrane, the activities of SOD, catalase, and peroxidase were assayed and the contents of malondialdehyde (MDA), ascorbic acid and glutathione were estimated. The ability of the seedlings to scavenge activated oxygen progresively decreased with the time of exposure to low temperature, concurrently MDA gradually accumulated and the permeability of the plasma membrane decreased. The amounts of MDA increased were different in the two hybrid combinations and were not in accordance with the ability of cold resistance. Comparing the more resistant variety “Wei-you 287” with the less resistant variety “Wei-you 49”, it was found that the leakage of electrolytes, the activities of SOD, peroxidase and catalase and the content of glutathione were all higher in the more resistant variety, but the accumulation of MDA was more in the less resistant variety. Higher temperature was less harmful than lower one to the seedlings of hybrid rice. Both enzymic and non-enzymic systems scavenging activated oxygen were similarly important in protecting the seedlings from damage by such oxygen.  相似文献

Inhibition of root elongation and modification of membrane properties are sensitive responses of plants to aluminium. The present paper reports on the effect of AI on lipid peroxidation and activities of enzymes related to production of activated oxygen species. Soybean seedlings (Glycine max L. cv. Sito) were precultured in solution culture for 3–5 days and then treated for 1–72 h with Al (AICI3) concentrations ranging from 10 to 75 μM at a constant pH of 4.1. In response to Al supply, lipid peroxidation in the root tips (< 2 cm) was enhanced only after longer durations of treatment. Aluminium-dependent increase in lipid peroxidation was intensified by Fe2+ (FeSO4). A close relationship existed between lipid peroxidation and inhibition of root-elongation rate induced by Al and/or Fe toxicity and/or Ca deficiency. Besides enhancement of lipid peroxidation in the crude extracts of root tips due to Al, the activities of superoxide dismutase (EC 1.15.1.1) and peroxidase (EC 1.11.1.7) increased, whereas catalase (EC 1.11.1.6) activity decreased. This indicates a greater generation of oxygen free radicals and related tissue damage. The results suggest that lipid peroxidation is part of the overall expression of Al toxicity in roots and that enhanced lipid peroxidation by oxygen free radicals is a consequence of primary effects of Al on membrane structure.  相似文献

The effect of exogenously applied glycinebetaine (GB) on the alleviation of damaging effects of NaCl treatment was studied in view of relative water content (RWC), malondialdehyde content, and the activity of some antioxidant enzymes in two rice (Oryza sativa L.) cultivars differing in salt tolerance (salt-tolerant Pokkali and--sensitive IR-28), comparatively. Both cultivars took up exogenously applied GB through their roots and accumulated it to considerable levels. Leaf RWC of both cultivars under salt treatment showed an increase with GB application. The activities of superoxide dismutase (SOD), ascorbate peroxidase (AP), catalase (CAT), and glutathione reductase (GR) increased in leaves of Pokkali, but peroxidase (POX) activity decreased under salinity. In IR-28, the activities of SOD, AP and POX increased, whereas CAT and GR decreased upon exposure to salt treatment. When compared to the salt-treated group alone, GB application decreased the activities of SOD, AP, CAT, and GR in Pokkali, whereas it increased the activities of CAT and AP in IR-28 under salinity. However, the activity of POX in IR-28 under salinity showed a decrease with GB application compared to the NaCl group. In addition, lipid peroxidation levels of both cvs. under salt treatment showed a decrease with GB treatment. Therefore, we conclude that GB protects both rice seedlings from salinity-induced oxidative stress.  相似文献

Thirty-day-old seedlings of tomato (Lycopersicon esculentum Mill.) were treated with various Hg concentrations (0, 10, and 50 μM) for up to 20 days, and the hypothesis that Hg induces oxidative stress leading to the reduction of biomass and chlorophyll content in leaves was examined. The accumulation of Hg in seedlings increased with external Hg concentration and exposure time, and Hg content in roots exposed to 50 μM Hg for 20 days was about 27-fold higher than that in shoots. Furthermore, Hg exposure not only reduced biomass and chlorophyll levels in leaves but also caused an overall increase of endogenous H2O2, lipid peroxidation products (malondialdehyde), and antioxidant emzymes activities such as superoxide dismutase, catalase, and peroxidase in leaves and roots. Our results suggest that the suppression of growth and the reduction of chlorophyll levels in tomato seedlings exposed to toxic Hg levels may be caused by an enhanced production of active oxygen species and subsequent high lipid peroxidation.  相似文献

Rice (Oryza sativa L.) roots were fed with L-ascorbic acid (AsA) and its putative precursors to observe AsA and oxalate concentrations and the resistance of rice to chilling, water stress, and Al toxicity. AsA concentration was significantly enhanced in both shoots and roots of rice seedlings by feeding with D-glucose or L-galactono-gamma-lactone. AsA or L-galactono-gamma-lactone treatment increased accumulation of oxalate mainly in soluble form, while these treatments decreased electrolyte leakage from root cells, H2O2 and lipid peroxidation level in rice seedlings subjected to chilling, water stress, and Al toxicity. They also alleviated the inhibition on root growth by Al. These results indicated that AsA and its immediate precursor protected plants against the oxidative damages induced by various stresses. However, 0.5 mM AsA and 10 mM L-galactono-gamma-lactone treatment had no significant effect on superoxide dismutase and catalase activity and ascorbate-peroxidase activities. Enhanced Al resistance caused by AsA and L-galactono-gamma-lactone may possibly be resulted from increased level of oxalate, which acts as metal chelator. Thus it is proposed that manipulation of AsA and oxalate biosynthesis through enhancement of L-galactono-gamma-lactone level in plants could be a strategy for improving abiotic stress tolerance.  相似文献

Seedlings of rice (Oryza sativa L.) cv. Pant-12 grown in sand cultures containing 200 and 400 μM NiSO4, showed a decrease in length and fresh weight of roots and shoots. Nickel was readily taken up by rice seedlings and the concentration was higher in roots than shoots. Nickel-treated seedlings showed increased rates of superoxide anion (O2 •− ) production, elevated levels of H2O2 and thiobarbituric acid reactive substances (TBARS) demonstrating enhanced lipid peroxidation, and a decline in protein thiol levels indicative of increased protein oxidation compared to controls. With progressively higher Ni concentrations, non-protein thiol and ascorbate (AsA) increased, whereas the level of low-molecular-weight thiols (such as glutathione and hydroxyl-methyl glutathione), the ratio of these thiols to their corresponding disulphides, and the ratio of AsA to dehydroascorbic acid declined in the seedlings. Among the antioxidant enzymes studied, the activities of all isoforms of superoxide dismutase (Cu-Zn SOD, Mn SOD and Fe SOD), guaiacol peroxidases (GPX) and ascorbate peroxidase (APX) increased in Ni-treated seedlings, while no clear alteration in catalase activity was evident. Activity of the ascorbate-glutathione cycle enzymes monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR)—significantly increased in Ni-treated seedlings. However such increase was apparently insufficient to maintain the intracellular redox balance. Results suggest that Ni induces oxidative stress in rice plants, resulting in enhanced lipid peroxidation and decline in protein thiol levels, and that (hydroxyl-methyl) glutathione and AsA in conjunction with Cu-Zn SOD, GPX and APX are involved in stress response.  相似文献

In excess, iron can induce the production and accumulation of reactive oxygen species (ROS), causing oxidative stress. The objective of this work was to evaluate the impact of toxic concentrations of iron (Fe) on the antioxidative metabolism of young Eugenia uniflora plants. Forty-five-day-old plants grown in Hoagland nutrient solution, pH 5.0, were treated with three Fe concentrations, in the form of FeEDTA, during three periods of time. At the end of the treatment, the plants were harvested and relative growth rate, iron content, lipid peroxidation and enzymes and metabolites of the antioxidative metabolism were determined. Iron-treated plants showed higher iron contents, reduced relative growth rates and iron toxicity symptoms in both leaves and roots. There was an increase in lipid peroxidation with increasing Fe, only in the leaves. The enzymatic activities of superoxide dismutase (SOD) and glutathione reductase (GR) increased with increasing Fe concentration and treatment exposure time. The activities of catalase (CAT), peroxidase (POX) and ascorbate peroxidase (APX) also increased with increasing Fe concentration but decreased with increasing treatment exposure time. Glutathione peroxidase activity (GPX) decreased with increasing Fe concentration and exposure time. The ascorbate (AA) and reduced glutathione (GSH) contents and the AA/DHA and GSH/GSSG ratios, in general, increased with increasing Fe concentration and treatment exposure time. The results indicate that under toxic levels of Fe, young E. uniflora plants suffer increased oxidative stress, which is ameliorated through changes in the activities of antioxidative enzymes and in the contents of the antioxidants AA and GSH.  相似文献

Alfalfa (Medicago sativa) roots were treated with mercuric ions in a concentration- and time-dependent manner, and lipid peroxidation was studied biochemically as well as histochemically along with other physiological responses. Histochemical staining with Schiff's reagent and Evans blue revealed that the peroxidation of membrane lipids and loss of plasma membrane integrity in Hg-treated roots occurred in the meristem and the elongation zone. The histochemical observations were supported by the quantitative determinations of thiobarbituric acid reactive substances (TBARS). However, under the mercuric ions stress, the alfalfa plants showed no significant alteration of hydrogen peroxide in roots. Analysis of lipoxygenase activity by non-denaturing polyacrylamide gel electrophoresis (PAGE) showed that there were two isoforms in the root of alfalfa plants, but they showed quite different patterns under the Hg exposure. Also, using non-denaturing PAGE, activities of superoxide dismutase (SOD) and peroxidase (POD) were determined in roots after treatment with Hg ions. The total activities of SOD and POD increased in roots after Hg treatment of roots. Activity of ascorbate peroxides (APX) was stimulated at relatively high concentration of Hg (40microM), and after prolonged Hg exposure (20microM, 24h). In contrast, glutathione reductase activity was depressed at higher concentrations of Hg (10-20microM). Treatments of seedlings with 10-40microM Hg decreased the ascorbate and glutathione amounts but increased total non-protein thiols. The above results indicated that Hg exerted its toxic effect on the root growth of alfalfa by induction of oxidative stress.  相似文献

Nitric oxide (NO) is a key signal molecule involved in many physiological processes in plants. To study the mechanisms of exogenous NO contribution to alleviate the aluminum (Al) toxicity, roots of rice (Oryza sativa) seedlings pre-treated with sodium nitroprusside (SNP, a NO donor) were used to investigate the effect of Al in this study. Results indicated that NO alleviated the lipid peroxidation induced by Al and promoted the root elongation, whereas butylated hydroxyanisole (BHA), an efficient lipophilic antioxidant, alleviated the lipid peroxidation only. Rice seedling roots pre-treated with SNP followed by Al treatment had lower contents of pectin and hemicellulose, lower Al accumulation in root tips and cell walls, higher degree of methylation of pectin and lower wall Al-binding capacity than the roots with Al treatment only. Therefore, the decreased Al accumulation in the cell walls of rice roots is likely to be the reason for the NO-induced increase of Al tolerance in rice, and it seems that exogenous NO enhanced Al tolerance in rice roots by decreasing the contents of pectin and hemicellulose, increasing the degree of methylation of pectin, and decreasing Al accumulation in root cell walls.  相似文献

We studied how the relationship between cadmium (Cd) toxicity and oxidative stress influenced the growth, photosynthetic efficiency, lipid peroxidation, and activity of ntioxidative enzymes in the roots and leaves of rice(Oryza sativa L Dongjin). Plants were exposed to Cd for 21 d. Both seedling growth and photosynthetic efficiency decreased gradually with increasing cadmium concentrations. Lipid peroxidation increased slowly in both roots and leaves, causing oxidative stress. However, each tissue type responded differently to Cd concentrations with regard to the induction/ inhibition of antioxidative enzymes. The activity of Superoxide dismutase (SOD) increased in both roots and leaves. Ascorbate peroxidase (APX) activity increased in leaves treated with up to 0.25 μM Cd, then decreased gradually at higher concentrations. In contrast, APX activity in roots increased and remained constant between 0.25 and 25 μM Cd. Enhanced peroxidase (POD) activity was recorded for treatments with up to 25/M Cd, gradually decreasing at higher concentrations in the leaves but remaining unchanged in the roots. Catalase (CAT) activity increased in the roots, but decreased in the leaves, whereas the activity of glutathione reductase (GR) was enhanced in both roots and leaves, where it remained elevated at higher Cd concentrations. These results suggest that rice seedlings tend to cope with free radicals generated by Cd through coordinated, enhanced activities of the antioxidative enzymes involved in detoxification.  相似文献

To investigate the temporal sequence of physiological reactions of garlic (Allium sativum) to cadmium (Cd) treatment, seedlings developed from cloves were grown in increasing concentrations of CdCl2, ranging from 1-10 mM, for up to 8 days in sand. Analysis of Cd uptake indicated that most Cd accumulated in roots, but some was also translocated and accumulated in leaves at longer exposure time (after 12h) and higher concentrations (5 and 10mM) of CdCl2. Changes in activities of antioxidative enzymes, including superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), were characterized in leaves of garlic seedlings. Cd (5 and 10 mM) initially inhibited the activities of SOD and CAT but thereafter recovered or even increased compared with control plants. POD activities at 5 and 10 mM of Cd increased more than 3-4 times over control plants within 12 h and then dropped, but were still higher than controls at the end of the experiment. Otherwise lipid peroxidation enhanced with the increasing of incubation time and concentrations of external Cd. Leaves exposed to 1 mM CdCl2 showed a less pronounced response and only a small reduction in shoot growth. These results suggested that in leaves of garlic seedlings challenged by CdCl2 at higher concentrations, induction of these various enzymes is part of a general defense strategy to cope with overproduction of reactive oxygen. The possible mechanism of antioxidative enzymes changing before Cd accumulation in leaves of garlic seedlings is discussed.  相似文献

Two triticale cultivars ZC 237 (Al-resistant) and ZC 1890 (Al-sensitive) were used to investigate the effects of 30 to 100 μM Al on antioxidative enzyme activity, lipid peroxidation and cell wall composition. In ZC 1890, the root elongation was significantly inhibited after 1-h exposure to 50 μM Al, the changes in hemicellulose fraction were clearly detected after 2-h Al exposure, while the peroxidase (POD) and superoxide dismutase (SOD) activities significantly increased after 6-h exposure, and the malondialdehyde (MDA) content after 12-h exposure. The similar patterns were also found in ZC 237. Treatment of ZC 1890 with 1 mM citrate for 30 min after 3-h exposure to Al resulted in significant decrease of Al bound to cell-wall and recovery of root elongation. These results suggested that Al affected cell wall before the damage of plasma membrane, but this was not the primary cause of root elongation inhibition.  相似文献

用含有不同浓度(0~400μmol/L)Cd(NO3)2的Hoagland营养液处理砂培的菊芋。处理50d后,测定植物体内镉积累量以及过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,并对POD同工酶进行电泳分析。发现在Cd50~100μmol/L浓度内,随着镉浓度的升高,菊芋根和叶中镉的积累量显著增加,而随后积累量的增加有所减少。根和叶中MDA含量显著上升,说明镉引起了膜脂过氧化。0~100μmol/LCd处理,根和叶中POD活性随Cd浓度增加而增强,而在200~400μmol/LCd处理下有所减弱。根和叶SOD活性在50~200μmol/LCd处理下随Cd浓度增加而增强,而在400μmol/LCd处理下SOD活性明显受到抑制。根和叶CAT活性随Cd浓度升高而增强。电泳结果显示,POD同工酶变化明显,镉诱导出一条新酶带LP10。菊芋POD同工酶可以作为镉污染的土壤的生物指示剂。  相似文献

The effect of selenium (Se) on rape (Brassica napus) seedlings subjected to cadmium (Cd) stress was studied in vitro by investigating plant growth and changes in fatty acid composition, activity of antioxidative enzymes and DNA methylation pattern. Physiological experiments were carried out on seedlings cultured for 2 weeks on Murashige-Scoog (MS) media with Cd concentrations of 0, 400 and 600 μM, and on corresponding media supplied with Se (2 μM). Exposure to increasing Cd concentrations reduced the fresh weight of the upper part (hypocotyls+cotyledons) of the seedlings more strongly than that of the root system, which was accompanied by higher Cd accumulation in these tissues. In the upper part, Cd exposure led to significant changes in the biochemical parameters: fatty acid unsaturation of plasmalemma decreased, the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPOX) diminished and that of ascorbate peroxidase (APX) increased. In contrast, the roots showed an increase in fatty acid unsaturation and in the activity of antioxidative enzymes. In both parts of rape seedlings H2O2 level and lipid peroxidation increased. Se addition to medium considerably reversed the Cd-induced decrease in fresh mass as well as the changes in lipid unsaturation and peroxidation. Se applied separately or in combination with Cd did not significantly affect the activity of antioxidative enzymes in the roots, but diminished it in the upper part. Moreover, the presence of Se in medium prevented changes in the DNA methylation pattern triggered in rape seedlings by high Cd concentrations. Two possible mechanisms for the action of Se were considered: (1) removal of Cd from metabolically active cellular sites, and (2) reduction of oxygen radicals.  相似文献

用20%聚乙二醇(PEG 6000)研究了渗透胁迫对黑麦(Secale cereale L.)幼苗活性氧(reactive oxygen species, ROS)和主要抗氧化酶—— 超氧化物歧化酶(superoxide dismutase, SOD)、过氧化氢酶(catalase, CAT)、抗坏血酸过氧化物酶(ascorbate peroxidase, APX)和谷胱甘肽还原酶(glutathione reductase, GR)活性的影响。结果表明, 与对照相比, PEG处理明显提高了叶子和根中丙二醛(malondialdehyde, MDA)的含量、ROS的水平和以上4种抗氧化酶的活性。渗透胁迫下,叶子和根中MDA和ROS水平变化的规律基本相似, 但抗氧化酶活性在2种器官中表现不完全相同, 叶子中CAT的活性在对照和处理中无显著差异, 但在根中差异明显, 表明叶子中SOD、APX和GR在植物应答渗透胁迫中起重要作用, 而根中这4种抗氧化酶都参与植物对胁迫的反应。GR活性随PEG处理变化幅度显著高于其它抗氧化酶, 表明GR在黑麦应答渗透胁迫中所起作用可能强于其它抗氧化酶。  相似文献

Pigeonpea [Cajanus cajan (L.) Millsp.] is a waterlogging-sensitive legume crop. We studied the effect of waterlogging stress on hydrogen peroxide (H2O2) content, lipid peroxidation and antioxidant enzyme activities in two pigeonpea genotypes viz., ICPL-84023 (waterlogging resistant) and MAL-18 (waterlogging susceptible). In a pot experiment, waterlogging stress was imposed for 6 days at early vegetative stage (20 days after sowing). Waterlogging treatment significantly increased hydrogen peroxide accumulation and lipid peroxidation, which indicated the extent of oxidative injury posed by stress conditions. Enzyme activities of peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX), superoxide dismutase (SOD) and polyphenol oxidase (PPO) increased in pigeonpea roots as a consequence of waterlogged conditions, and all the enzyme activities were significantly higher in waterlogged ICPL-84023 than in MAL-18. POX activity was the maximum immediately after imposing stress, therefore, it was suggested to be involved in early scavenging of H2O2, while rest of the enzymes (CAT, APX, SOD and PPO) were more important in late responses to waterlogging. Present study revealed that H2O2 content is directly related to lipid peroxidation leading to oxidative damage during waterlogging in pigeonpea. Higher antioxidant potential in ICPL-84023 as evidenced by enhanced POX, CAT, APX, SOD and PPO activities increased capacity for reactive oxygen species (ROS) scavenging and indicated relationship between waterlogging resistance and antioxidant defense system in pigeonpea.  相似文献
