Effects of Acetylcholine on the Growth of the Vigna Seedling   总被引:2,自引:0,他引:2  
Acetylcholine applied to cotyledons influenced the growth patternof etiolated seedlings of Vigna sesquipedalis in a specificmanner. The reagent as well as red light suppressed hypocotylgrowth and simultanously promoted epicotyl growth. Such effectswere not found with choline. Neostigmine, a potent inhibitorof cholinesterases from plants, caused fundamentally the sameeffects as acetylcholine. Atropine, an acetylcholine antagonistin animals showed effects opposite to those of acetylcholineor neostigmine. Atropine also partially negated the red light-inducedeffects. Indole-3-acetic acid, kinetin and gibberellic acid could notproduce effects like those of cholinergic reagents. From these results, it was surmised that acetylcholine as agrowth substance in the Vigna seedling regulates the transitionfrom hypocotyl growth to subsequent epicotyl growth. Callus cultures were initiated from immature cotyledons of Vignaaconitifolia, V. mungo and V. radiata on MS medium supplementedwith NAA, picloram or 2, 4-D. On transfer to L-6 liquid mediumsupplemented with low concentrations of picloram, GA3 and cytokinins,large number of somatic embryos differentiated from the callus.The cells destined to become somatic embryos divided to formspherical or filamentous proembryos. From the filamentous proembryo,the embryo proper developed either at single or multiple sites.Development of somatic embryos from multiple sites resultedin several embryos connected by a common suspensor at the radicleend. Continued divisions of the proembryos led to globular,heart shaped and dicotyledonary stages of somatic embryogenesis.The somatic embryos of V. mungo and V. aconitifolia differentiatedinto tiny plantlets at low frequency (1%) in liquid suspensioncultures supplemented with zeatin, picloram and GA3. Levels of starch and soluble sugar in pods of Phaseolus vulgarisand Vigna mungo plants were analyzed during the course of maturationof fruits. The results suggest that the immature pods of P.vulgaris function to some extent as temporary reservoirs ofcarbohydrates for growth of seeds. Dynamics of the nitrogen relationships of Vigna mungo in pureand in mixed stands with Celosia argentea, a common weed ofleguminous crops in certain regions of India, were investigated.The weed significantly depressed nodulation and reduced thedry matter and nitrogen yield of the legume which was lowestat the highest density of the weed. A significant amount ofnitrogen transfer occurred from the legume in the weed mixtureswhich appeared to be taken up by the weed plants. Therefore,weed plants benefited from their association with the legumeby accumulating more dry matter and nitrogen in the legume mixtures.The possible competitive or biochemical interference of theweed with nitrogen relationships of the legume in mixtures isdiscussed. Glutamate dehydrogenase (E.C. 1.4.1.2 [EC] ) is usually assayed bythe disappearance of NAD(P)H from the assay medium. A new technique,in which the enzyme level in leaf tissue of Vigna mungo (L.)Hepper is estimated by the disappearance of 2-oxoglutarate,is described. By using the Giemsa C-banding technique, chromosome bandingpatterns on the somatic chromosomes of eight important pulsecrops, pea, lentil, guar (cluster bean), chick pea, pigeon pea,mung bean (green gram), urd (black gram) and cowpea have beenstudied. Each species has a characteristic C-banding pattern.The significance of such banding patterns which correlate withthe position of pachytene knobs, in chromosome identification,and in assigning relationships at the cytological level in thepulses of genus Vigna is stressed. The relation of the in vivo nitrate reductase (NR) activityto growth period was studied in the nodules and the leaves ofthe summer moong (Vigna radiata). The maximum NR activity wasobserved 31 days after sowing (DAS) in the leaves and 28 DASin the case of the nodules. In a pot experiment, the effectof the various nitrogen concentrations, namely 0, 3, 6, 9 and12 mg kg–1 was studied on NR activity at three growthstages. The maximum NR activity was observed at 6 mg kg–1N during the pre-flowering stage (26 DAS). Though the noduleshave higher NR activity, its expression was limited by substrateavailability. Striga gesnerioides(Scrophulariaceae) is an obligate hemiparasiticangiosperm, that infects the roots of cowpea (Vigna unguiculata).The development ofStrigaseedlings was characterized after transferto the surface of cowpea roots using cryo-scanning electronmicroscopy. Contact with the surface of the root caused distinctmorphological changes in theStrigaradicles. Radicle elongationceased 8 h after contact with host roots and radicle hairs developed.These hairs adhered to the surface of the host root. Early stagesof root penetration, i.e. prior to connection to the host vascularsystem were examined by light microscopy and transmission electronmicroscopy to determine the mechanisms by whichStrigainvadesroot tissue. Cowpea roots were penetrated byS. gesnerioidesradiclesafter 48 h. Host cells immediately surrounding the invadingStrigawerecompressed, but other host cells were not affected. Nineteen accessions ofVigna luteola,five ofV. marinassp.oblonga,and two ofV. marinassp.marinawere analysed using variation ofisozymes and RAPD markers to obtain better insight into geneticrelationships within and between these taxonomic entities. Thirteenputative isozyme loci were scored, seven of which were polymorphic.Both species showed very low genetic diversity indices and mostof the variation was detected among populations. Pairwise Nei'sgenetic distances based on allozyme frequencies were also verylow and the accessions ofV. marinassp.marinawere the least relatedto the others. RAPD analysis was more discriminating and 66bands out of a total of 85 were polymorphic. Based on the presenceor absence of bands, Jaccard's similarity index was calculated.Similarity ranged from 0.476 to 0.98. Matrices derived fromboth isozyme and RAPD data were used to construct UPGMA dendrograms.In the tree obtained from Nei's genetic distance, based on allozymefrequencies, accessions belonging toV. marinassp.oblongaweremixed withV. luteolaaccessions; on the other hand, the twoV.marinassp.marinaclustered separately, with oneV. luteola.Thedendrogram derived from RAPD data showed three main groups correspondingto the three taxa analysed. A study was undertaken to define more clearly the role of theearly loss of apical dominance on yield of cowpea [Vigna unguiculata(L.) Walp. cv. Vita-5]. Decapitation at the fifth leaf stageresulted in an increase in branching components, yields andharvest indices, while vegetative d. wt accumulation was reduced.Foliar-applied sprays of 6-benzyladenine had no effect on branchingunless combined with decapitation and no significant effectson yield over that of controls were observed. The influence of temperature and photoperiod on phenologicaldevelopment of three bambara groundnut (Vigna subterranea) selectionsfrom Botswana, Zimbabwe and Mali was investigated in a semi-controlledenvironment experiment with factorial combinations of threeconstant temperatures (20.9, 23.4 and 26.2 °C) and fourconstant photoperiods (10.0, 12.5, 13.5 and 16.0 h d-1). Inall three selections, the onset of flowering was influencedby temperature but not by photoperiod, while the onset of pod-growth(‘podding’) of all three selections was influencedby both factors. The influence of temperature and photoperiodwas quantified by means of photothermal models, linking developmentrates to temperature and photoperiod with linear equations.The rate of progress from sowing to flowering of the three selectionscould be described very well (r2>95%) as a function of temperature;the rate of progress from flowering to podding was describedreasonably well as a function of both temperature and photoperiodby a combination of one to three response planes (r2for thedifferent selections ranging from 63 to 90%). Model testingwith independent data sets showed good agreement between observedand predicted times to flowering and podding. The effects of para-fluorophenylalanine (p-FPA) and azetidine-2-carboxylicacid (AZ) on acid phosphatase and peroxidase activity, and onL-leucine incorporation, in root segments of Triticum aestivumL. cv. Fenman and Vigna radial a (L.) Wilczek, were studied.Incubation with 50 mmol–3 AZ significantly reduced phosphataseand peroxidase activities in wheat roots, but with 20 mol m–3p-FPA, only the peroxidase activity was reduced. In mung beanroots, phosphatase activity was inhibited by both AZ and p-FPA.Effects of the ortho- and meta- isomers of FPA on wheat rootphosphatase and peroxidase, and on mung bean phosphatase, werealso examined. Leucine uptake and incorporation were not inhibitedby 5 h pre-incubation with either p-FPA or AZ, but were inhibitedafter 24 h of pre-incubation. The results support the view that,in the shorter term, the analogues inhibit enzyme activity bybecoming incorporated to produce non-functional protein and,in the longer term, metabolism is further affected by inhibitionof protein synthesis. The relationship between seed number per pod and senescenceof the leaf in its axil was examined in a determinate cowpea(Vigna unguiculata L. Walp) variety C.779. The seed number perpod was reduced at all fruiting nodes by surgical excision ofpart of the 4-d-old pod. Leaf senescence as measured by lossof leaf area, chlorophyll content and soluble protein was sloweddown in leaves supporting the development of an artificiallyreduced number of seeds. Diminished nitrogen mobilization fromthe leaf could not account for the reduced rate of leaf senescence.The result suggests the involvement of a senescence signal fromthe developing seeds to the leaf in its axil. Development ofthe basal half of the excised pod in the cowpea provides a uniquesystem for manipulating seed number per pod. Anatomical investigations were carried out on the structureand development of the mature secondary haustorium in Alectravogelii growing on Arachis hypogaea or Vigna unguiculata. Followingthe formation of the young secondary haustorium, both the cambiumand pericycle of the host root directly opposite the young secondaryhaustorium are stimulated to divide and form new tissues andorgans including haustorial roots. Further proliferations ofthe host root pericycle and the haustorial cortex give riseto a large, tuberous and complex mature secondary haustoriumwithin which the tissues of the host and parasite remain inintimate contact forming a perfect graft union with a wide zoneof contact. Apart from the haustorial axial xylcm strand whichnormally connects the xylem of the parasite secondary root withthat of the host, direct xylary connections are also establishedbetween the axial xylem of the haustorium and the xylem of thehaustorial roots. The entire surface of the mature secondaryhaustorium of Alectrais covered with these haustorial rootsas was previously observed in its mature primary haustorium. Piperazine, a chemical used as buffer component, greatly promotedadventitious root formation in cuttings of sunflower (Helianthusannuus L.), pea (Pisum sativum L.), mung bean (Vigna radiataL.) and to a lesser extent in bean (Phaseolus vulgaris L.) seedlings.Piperazine was more effective in acidic pH. The studies withpiperazine analogues showed that any substantial modificationof the structure caused the chemical to be less effective, oreven inhibitory. Histological studies in sunflower hypocotylsdemonstrated that piperazine did not alter the timing of theinitial cell division. In the presence of piperazine, sunflowerhypocotyls failed to develop primary phloem fibres. Piperazineat the concentrations that promote rooting did not kill or damagethe tissue at the base of the hypocotyl. Compared to controls,piperazine treatment did not alter the proportion of primordiathat eventually developed into actively elongating roots. Sixdays after treatment 45% of the control roots in the basal sectionwere actively growing, compared to 51% in the piperazine. Structural, biochemical, and immunological comparisons of nodulesfrom ten species of plants were made to determine if a correlationexists between nodule structure, ureide production, urate oxidaseactivity, and antigenic similarity in urate oxidase. In specieswith high urate oxidase activity and cross-reaction with soybeananti-urate oxidase [soybean (Glycine max), green bean (Phaseolusvulgaris), mung bean (Vigna radiata), cowpea (Vigna unguiculata)],the nodules were determinate and contained numerous interstitialcells, interspersed among the infected cells. Within the interstitialcells of the ureide producing nodules numerous peroxisomes werenoted and these peroxisomes appear to be structurally similar,viz. a large electron opaque core surrounded by a less electronopaque rim. Although hemp sesbania (Sesbania exaltata) noduleswere similar in ultrastructure to other ureide producers withdetectable urate oxidase activity, no cross-reactivity was observedwith anti-soybean urate oxidase. Amide producing nodules eithercontained no interstitial cells [e.g. Indian jointvetch (Aeschynomeneindica), showy crotalaria (Crotalaria spectabilis)} or interstitialcells with few peroxisomes [e.g. alfalfa (Medicago saliva),broad bean (Vicia faba), pea (Pisum sativum)] with little urateoxidase activity, exhibiting no cross-reaction with soybeananti-urate oxidase. These data indicate that the urate oxidasein most ureide producing nodules is very similar and, structurally,ureide producing nodules are organized in a specialized wayto carry out ureide assimilation in the uninfected interstitialcells. A single copy of the a-amylase gene, composed of three intronsand four exons, was found in Vigna mungo. Examination of levelsof a-amylase and its mRNA in detached cotyledons indicated thatattachment of the embryonic axis is not required for expressionof the gene in cotyledons of germinating seeds. Prat, R. 1985. A comparative study of some methods for measuringcell growth potentials in Vigna radiata hypocotyl, in situ andafter excision.—J. exp. Bot. 36: 1150–1158. Accurate estimations of growth rates are required to understandthe development of physiological and biochemical parametersinvolved in growth processes. Growth measurements can be madeeither ‘in situ’ on whole seedlings by marking successivelevels or ‘in vitro’, after excision of successivehypocotyl segments. These different methods have been comparedin order to determine which measurements allow one to assessaccurately the normal growth characteristics. In vitro measurementsperformed with excised segments required a prolonged incubationperiod before stable growth rates were achieved. The growthpotentials of the successive hypocotyl levels remain unchangedafter excision. These in vitro measurements can therefore beused to assess the growth characteristics of cells in hypocotyls.
