The purpose of this study is to investigate the possible effect of an extremely low-frequency magnetic field (ELF-MF) on nitric oxide (NO) level. In this study, 27 male Sprague-Dawley rats were used. The rats were divided into three groups: two experimental and one control (sham-exposed). The first and second experimental group (n = 10) were exposed to 100 microT and 500 microT ELF-MF during 10 months, 2 h a day, respectively, and the third (n = 7) group was treated like an experimental group except for ELF-MF exposure in methacrylate boxes. After ELF-MF and sham exposure, serum nitrite levels were measured by Griess reaction. A significant reduction was observed in nitrite levels among the first and second experimental groups of rats and sham-exposed rats after exposure for 10 months, 2 h a day, to ELF-MF of 100 and 500 microT (p < 0.01). These results suggest that prolonged ELF-MF exposure at intensities of exposure limits, determined by ICNIRP for public and occupational, may reduce NO production probably affected by NO generation pathways.  相似文献

Interference of 50 Hz extremely low frequency magnetic fields (ELF-MF) with the known aneugen vinblastine (VBL) on micronucleus formation was tested with the in vitro cytokinesis block micronucleus assay in human lymphocyte cultures. Isolated lymphocyte cultures were prepared from 18 individuals. Three groups of quadruplicate cultures from six unrelated individuals were exposed to 50 Hz ELF-MF of background (bkg), 80 and 800 microT, respectively, during the complete incubation period (72 h). Twenty-four hours after culture initiation, one replicate culture from each individual within each ELF-MF group was exposed to 0, 5, 10, or 15 ng/ml VBL. The isolated lymphocyte cultures were scored for the presence of micronuclei, the nuclear division index (NDI), and apoptosis. As expected, increased VBL concentration resulted in an increased micronucleus and apoptosis frequency and in a decreased NDI. In the presence of VBL, there was a systematic tendency for increased micronucleus and apoptosis frequency in the ELF-MF exposed groups compared to the bkg group. In the absence of VBL, we observed no statistically significant effect of ELF-MF on micronucleus induction or apoptosis frequency, but the NDI was significantly higher in the 800 microT group compared to the other groups, suggesting an effect of ELF-MF on cell proliferation. An interaction between ELF-MF and VBL on NDI was observed. This interaction reflected the drastic decrease in NDI due to coexposure to VBL.  相似文献

A suppression in melatonin secretion is one of the mechanisms proposed to explain the possible adverse effects of extremely low frequency magnetic fields (ELF-MF), but the results of research are inconclusive. This study investigated the effect of occupational ELF-MF exposure on 6-sulfatoximelatonin (6-OHMS). Exposure was monitored for three complete work shifts in 59 workers using personal exposure meters. Environmental exposure was also evaluated. Urinary 6-OHMS in morning samples, an indicator of night-time melatonin production, was measured. Urine was collected twice on Friday and the following Monday. Workers were classified according to ELF exposure as low exposed (0.2 microT): 6-OHMS did not differ between groups (P > .05) in either Friday or Monday urine samples. In addition, 6-OHMS was not related to exposure under multivariate analysis. The ratio between 6-OHMS in Monday versus Friday samples was also calculated to test the hypothesis of a possible variation in pineal function after 2 days, interruption of occupational ELF-MF exposure: again no exposure-related difference was observed. Our results do not support the hypothesis that occupational exposure to ELF-MF significantly influences melatonin secretion.  相似文献

This study reports the continuous 8-h monitoring of data on extremely low-frequency magnetic fields (ELF-MF) relating to 14 children and 35 teachers in 11 elementary schools in Northern Taiwan. It was anticipated that the subjects in two of these campuses would have elevated exposure to ELF-MF as a result of their close proximity to high-voltage (161 kilo-Volt, kV) power lines. The results of our analysis reveal that in those schools with high-voltage power lines running through the campuses, the mean ELF-MF exposure level (0.38 +/- 0.51 micro-Tesla (microT), or 0.15, 0.25 and 0.44 microT at the respective 25th, 50th and 75th percentiles) was higher than the mean ELF-MF exposure level for campuses situated far away from such high-voltage power lines (0.14 +/- 0.27 microT, or 0.04, 0.06 and 0.10 microT at the respective 25th, 50th and 75th percentiles). The multi-level analytical technique, which takes individual measurements as the analytical unit, and which also takes into consideration the inter-correlation between measurements from the same individual and/or campus, was also applied to the analysis of the data. We conclude that individual-level and school-level clustering of the measurements, both of which were discernible in this study, should be taken into consideration in any future analysis of data obtained from the continuous monitoring of exposure to ELF-MF.  相似文献

Fertilized white leghorn eggs were exposed to a 4 micro-Tesla (microT) 60 Hz horizontal magnetic field for 15, 18, 23 and 28 h. After exposure to the magnetic field, the embryos were isolated and assayed for ornithine decarboxylase (ODC) activity. ODC activity in magnetic field-exposed embryos was compared to ODC activity in sham-exposed embryos. ODC activity in magnetic field-exposed embryos was not statistically elevated above sham-exposed embryos.  相似文献

The extremely low-frequency (ELF) magnetic fields (MFs) originating from equipment used for assisted reproduction, umbilical cord-blood and peripheral-blood stem cell transplantation, transfusion, and hemodialysis were measured. The ELF-MF values were 0.1-1.2 microT on clean benches, <0.1-8.0 microT on inverted microscopes, <0.1-13.6 mmicroT in CO2 incubators, 4.3-11.5 microT in centrifuges, 0.4-18.8 microT in programmed freezers, <0.1-0.3 microT in deep freezers, 0.3-3.1 microT on cell separators, and 0.2-0.9 microT in hemodialysers. Frequencies of MFs were nominally 60 Hz, but some devices showed non-sinusoidal 120 Hz. Such MFs can be reduced by shielding the sources or altering the protocols employed.  相似文献

Photosynthetic CO2 uptake rate and early growth parameters of radish Raphanus sativus L. seedlings exposed to an extremely low frequency magnetic field (ELF MF) were investigated. Radish seedlings were exposed to a 60 Hz, 50 microT(rms) (root mean square) sinusoidal magnetic field (MF) and a parallel 48 microT static MF for 6 or 15 d immediately after germination. Control seedlings were exposed to the ambient MF but not the ELF MF. The CO2 uptake rate of ELF MF exposed seedlings on day 5 and later was lower than that of the control seedlings. The dry weight and the cotyledon area of ELF MF exposed seedlings on day 6 and the fresh weight, the dry weight and the leaf area of ELF MF exposed seedlings on day 15 were significantly lower than those of the control seedlings, respectively. In another experiment, radish seedlings were grown without ELF MF exposure for 14 d immediately after germination, and then exposed to the ELF MF for about 2 h, and the photosynthetic CO2 uptake rate was measured during the short-term ELF MF exposure. The CO2 uptake rate of the same seedlings was subsequently measured in the ambient MF (control) without the ELF MF. There was no difference in the CO2 uptake rate of seedlings exposed to the ELF MF or the ambient MF. These results indicate that continuous exposure to 60 Hz, 50 microT(rms) sinusoidal MF with a parallel 48 microT static MF affects the early growth of radish seedlings, but the effect is not so severe that modification of photosynthetic CO2 uptake can observed during short-term MF exposure.  相似文献

The objective was to investigate the effects of developmental stage (fully-expanded or expanding blastocysts) and/or age (harvested on Days 7 or 8) on post-vitrification in vitro survival of bovine blastocysts derived from intracytoplasmic sperm injection (ICSI) or in vitro fertilization (IVF). Post-warming survival (re-expansion of blastocoele within 24 h) of ICSI-derived fully-expanded blastocysts (80%) was similar to that of their IVF-derived counterparts (88%). However, the ability of ICSI-derived expanding blastocysts to survive vitrification procedures (61%) was lower than that of IVF-derived blastocysts (85%; P < 0.05), although the ICSI- and IVF-derived fresh blastocysts were of similar quality. The age of the blastocysts before vitrification did not affect cryotolerance for either ICSI-derived (73 and 59% for Days 7 and 8 embryos, respectively) or IVF-derived blastocysts (86% for both Days 7 and 8 embryos). At 24 h of post-warming culture, ICSI-derived blastocysts surviving vitrification contained a higher proportion of dead cells than their IVF-derived counterparts (5-13% vs. 2-4%; P < 0.05), but these proportions were not different from those of fresh control embryos. There was an adverse effect of vitrification on the ability of blastocysts to hatch within 72 h of culture only in IVF-derived Day 8 blastocysts (41 and 70% in vitrified and fresh control groups, respectively). In conclusion, the proportion of blastocysts that survived vitrification procedures was similar for ICSI- and IVF-derived bovine blastocysts if the former were cultured to the fully-expanded stage prior to vitrification, with no significant difference between embryos harvested on Day 7 versus Day 8.  相似文献

The possible effects of repeated night-time exposure to an extremely low frequency magnetic field (ELF-MF) on melatonin were investigated in nine healthy male subjects aged 23-37 yr. The 16-week experiment consisted of 3 weeks of pre-exposure, 11 weeks of night-time exposure to MF generated from a nonheated electric sheet (ES), and 2 weeks of post-exposure recovery observation. The average MF intensity (rms, mainly 50 Hz AC) on the surface of the sheet was 0.7 microT at the head, 8.3 microT at the waist, and 3.5 microT at the feet of the subject. For each of the urine samples collected 5 times a day on scheduled sampling days, the urinary excretion rate (ng/h) of melatonin was determined, and 24 h rhythms were extracted for each subject and each experimental period (pre-exposure, first half and latter half exposure, and post-exposure periods) by the method of complex cosine curve fitting. Although estimates of the peak height, acrophase, and total daily amount of melatonin were characterized by significant variations among individual subjects, they did not reveal any statistically significant difference between exposure periods and nonexposure periods. Thus, the present study indicates that any profound effect of the MF originating from an ES on nocturnal melatonin production and its circadian rhythm is unlikely.  相似文献

The purpose of this study was to reproduce and extend an earlier investigation of the effects of human exposure to combined, 60-Hz electric and magnetic fields. This paper presents the neurobehavioral results. Thirty men participated in one training session and four testing sessions. Subjects were randomly assigned to two groups. The 18 subjects in Group I were exposed (9 kV/m, 20 microT) and sham exposed in two counterbalanced orders. In Group II, half of 12 subjects were exposed (9 kV/m, 20 microT) every session, and the remaining half were sham exposed every session. The study was doubly blinded. Measures of cardiac interbeat interval, event-related brain potentials, and performance were obtained before, during, and after exposures. As in the earlier study, exposure to the combined field resulted in a statistically significant slowing of heart rate, in changes in late components of event-related brain potentials, and in decreased errors on a choice reaction-time task. In addition, field effects on several other measures approached statistical significance. The physiological measures obtained during exposure indicated that effects were greatest soon after the field was switched on, and again when it was switched off. The data indicate that changes in exposure level may be more important than duration of exposure for producing effects in human beings.  相似文献

The aim of the present work is to evaluate the effect of caffeine, the world’s most popular psychoactive drug, on the electric activity of the rat’s brain that exposed to extremely low-frequency magnetic field (ELF-MF), during 15 days. The obtained results showed that administration of caffeine in a group of rats by dose of 10 mg/kg (equivalent to human daily consumption) caused a reduction in the mean power amplitude of electroencephalogram (EEG) trace for almost all frequency bands especially α (8–12 Hz). It was observed that the influence of caffeine was more evident in motor cortex than in visual cortex. While the exposure of another group to ELF-MF of intensity 0.2 mT during the same period caused an enhancement in the mean power amplitude of most EEG frequency bands; this was more observed in the right hemisphere of the brain than that of the left hemisphere. The administration of caffeine while rats were exposed to ELF-MF, led, after 5 days of exposure, to a great increase in the mean power amplitude of α band at all places of recording electrodes. It may be concluded that caffeine administration was more effective in reducing the hazardous of ELF-MF in motor cortex than in visual cortex.  相似文献

Zebrafish embryos were exposed intra ovo to a 50 Hz AC magnetic field of 1000 microT rms, and the progress of asynchronous hatching was monitored. A statistically significant delay was observed when field exposure started 48 h after fertilization. In contrast, when exposure started 2 h after fertilization, no statistically significant effect was seen. When field exposure was administered together with submaximal doses of progesterone at 48 h postfertilization, the two treatments appeared to delay hatching in an additive manner. Evaluating the progress of hatching in zebrafish embryos seems relevant for exploration of EMF effects on reproduction.  相似文献

The purpose of this study was to reproduce and extend two earlier studies of the effects of human exposure to 50 Hz magnetic fields (MF). In a recent paper, we described results of two double-blind investigations performed to examine effects of 100 microT(rms) 50 Hz MF exposure on psychological parameters in the same group of healthy human volunteers. In each exposure session, at 1 week intervals, with sham, continuous, and intermittent (15 s ON/OFF cycles) MF conditions, mood ratings, performance measures, and electrophysiological measures were taken. In the first study, significant amplitude changes were observed in the event-related brain potentials (ERP) recorded during a dichotic listening task. In the second study, latency and reaction time (RT) slowing were seen on a visual discrimination task (P(300) paradigm). Although these results were little related to the number of parameters analysed, they indicate that low level 50 Hz MF might have a slight influence on ERP and RT under specific circumstances of sustained attention. Before concluding that moderately strong MF exposure can influence cognitive function, previous results should be replicated, using the same paradigms with another group of healthy volunteers. In the present study, 18 healthy subjects were exposed to three experimental sessions of 30 min each, given at 1 week intervals. The sessions consisted of continuous 100 microT(rms) 50 Hz MF exposure, sham condition, and bright light (5000 lux) exposure. The study was performed double-blind, with the exposure order counter-balanced. The data on mood, ERP, RT, and other performance measures did not show any differences among the sham exposure, light exposure, and MF exposure conditions. The results of this study do not support the hypothesis that extremely low frequency (ELF) MF exposure affects the brain's electrical activity or cognitive function at field strength (100 microT(rms)) similar to that found in very close proximity of some household and industrial electrical appliances and well in excess of the average MF strength (c. 0.1 microT) found in homes. The sensitivity of the experiment was possibly not sufficient to detect an effect at this relatively low MF, and larger sample sizes would be required in further studies.  相似文献

The aim of this study was to evaluate the activity of the antioxidant enzymes mitochondrial and cytosolic superoxide dismutase (EC 1.15.1.1), glutathione peroxidase (POX, EC 1.11.1.9) and glutathione S-transferase (EC 3.1.2.7), as well as the concentration of malone dialdehyde (MDA), as an indicator of lipid peroxidation rate in the liver tissue homogenates and blood serum of male rats exposed to extremely low-frequency magnetic field (ELF-MF) in order to improve the healing process of an experimental cut wound on the back of each animal. The exposure to ELF-MF with frequency 40 Hz and magnetic flux density 10 mT induced an increase in POX serum activity and a decrease in MDA contents in the liver tissue, which suggests the inhibition of phospholipid peroxidation and subsequent stabilization of cellular membranes, as a result of ELF-MF action. Based on the results obtained, it seems that ELF-MF could be a useful supplement in the complex treatment of prolonged wound healing, due to the activation of endogenous enzymatic antioxidant system.  相似文献

alpha-Solanine and alpha-chaconine are two naturally occurring steroidal glycoalkaloids in potatoes (Solanum tuberosum), and solanidine-N-oxide is a corresponding steroidal aglycone. The objective of this research was to screen potential cyto-toxicity of these potato glycoalkaloids using bovine oocyte maturation, in vitro fertilization techniques and subsequent embryonic development as the in vitro model. A randomized complete block design with four in vitro oocyte maturation (IVM) treatments (Experiment 1) and four in vitro embryo culture (IVC) treatments (Experiment 2) was used. In Experiment 1, bovine oocytes (n=2506) were matured in vitro in medium supplemented with 6 microM of alpha-solanine, alpha-chaconine, solanidine-N-oxide or IVM medium only. The in vitro matured oocytes were then subject to routine IVF and IVC procedures. Results indicated that exposure of bovine oocytes to the steroidal glycoalkaloids during in vitro maturation inhibited subsequent pre-implantation embryo development. Potency of the embryo-toxicity varied between these steroidal glycoalkaloids. In Experiment 2, IVM/IVF derived bovine embryos (n=2370) were cultured in vitro in medium supplemented with 6 microM of alpha-solanine, alpha-chaconine, solanidine-N-oxide or IVC medium only. The results showed that the pre-implantation embryo development is inhibited by exposure to these glycoalkaloids. This effect is significant during the later pre-implantation embryo development period as indicated by fewer numbers of expanded and hatched blastocysts produced in the media containing these alkaloids. Therefore, we conclude that in vitro exposure of oocytes and fertilized ova to the steroidal glycoalkaloids from potatoes inhibits pre-implantation embryo development. Furthermore, we suggest that ingestion of Solanum species containing toxic amounts of glycoalkaloids may have negative effects on pre-implantation embryonic survival.  相似文献

We compared the developmental ability and gene expression of in vivo- and IVM/IVF-derived porcine embryos following microinjection with SV40-LacZ. A total of 412 IVM/IVF-derived and 129 in vivo-collected zygotes was used to examine developmental ability and gene expression following DNA microinjection. When either DNA injected or noninjected zygotes were cultured for 4 d in NCSU 23 followed by 5 d in Eagle's minimal essential medium (EMEM), the percentages of zygotes developing to blastocysts and hatched blastocysts were higher (P < 0.05) compared with groups cultured in NCSU 23 alone. The percentages of injected embryos reaching the morula and blastocyst stages were significantly lower (P < 0.05) than that of noninjected control embryos whether in vivo or IVM/IVF derived. The percentages of morula and blastocyst stage embryos expressing the gene were higher in the in vivo-derived embryos than in IVM/IVF-derived embryos. A lower proportion of (67 to 77%) mosaicism was observed in the in vivo-derived embryos than in IVM/IVF (90 to 100%) derived embryos. The total cell number of blastocysts cultured in both NCSU 23 and EMEM media was significantly higher than that of blastocysts cultured continuously in NCSU 23. Our results suggest that this dual culture system enhanced embryo viability following microinjection of foreign DNA.  相似文献

The aim of this study was to determine whether exposure to extremely low frequency magnetic field (ELF-MF) affects the normal diurnal rhythm of the pain threshold in mice. Pain thresholds were evaluated in mice using the hot plate test. A significant increase of pain threshold during night was observed compared to that during day. This rhythm was attenuated by both constant exposure to light (LL) and constant exposure to darkness (DD) for 5 days. Under DD exposure, the diurnal rhythm in pain threshold was restored when mice were exposed to ELF-MF (60 Hz, 1.5 mT for 12 h daily, from 08:00 to 20:00 h) for 5 days. The diurnal rhythm was not reversed under dark with reversed ELF-MF cycle (exposure to 1.5 mT from 20:00 to 08:00 h, next day) for 5 days, although pain threshold in the ELF-MF exposed period of night was slightly decreased. The diurnal rhythm of melatonin analgesic effect related to pain threshold was also observed under DD by the exposure of ELF-MF for 5 days, but not for 5 nights. The present results suggest that ELF-MF may participate in the diurnal rhythm of pain threshold by acting on the system that is associated with environmental light-dark cycle.  相似文献

Birth of large calves that developed from in vitro-derived bovine embryos   总被引:2,自引:0,他引:2  
High birth weights were observed in calves that developed from bovine embryos produced by in vitro maturation (IVM) and in vitro fertilization (IVF) procedures. After IVM and IVF, embryos were either co-cultured in vitro with oviductal epithelial cells or transferred into the sheep oviduct for development to the blastocyst stage. Blastocysts were transferred to the reproductive tracts of recipient heifers and cows for development to term. Birth weights and gestation periods were compared between calves that developed from in vitro-derived embryos and calves born after artificial insemination (AI) of cows in the herd from which recipient females were selected. Gestation periods were not different among the groups (P > 0.05), but calves that developed from IVM/IVF-derived embryos co-cultured in vitro were larger at birth than calves born from IVM/IVF-derived embryos that developed into blastocysts in the sheep oviduct and calves born from AI (P < 0.001). Dystocia and calf mortality were associated with large calf size at birth. These data were collected from an experiment designed for other purposes, and confounding variables and small sample size could have influenced the observed differences in birth weights. Nevertheless, the extreme birth weights of some calves suggest that abnormal prenatal growth occurs in some IVM/IVF-derived bovine embryos and that conditions for co-culture to the blastocyst stage may exacerbate the problem.  相似文献

The principal objective of this study was to assess the DNA damage in a normal cell line system after exposure to 60 Hz of extremely low frequency magnetic field (ELF-MF) and particularly in combination with various external factors, via comet assays. NIH3T3 mouse fibroblast cells, WI-38 human lung fibroblast cells, L132 human lung epithelial cells, and MCF10A human mammary gland epithelial cells were exposed for 4 or 16 h to a 60-Hz, 1 mT uniform magnetic field in the presence or absence of ionizing radiation (IR, 1 Gy), H2O2 (50 μM), or c-Myc oncogenic activation. The results obtained showed no significant differences between the cells exposed to ELF-MF alone and the unexposed cells. Moreover, no synergistic or additive effects were observed after 4 or 16 h of pre-exposure to 1 mT ELF-MF or simultaneous exposure to ELF-MF combined with IR, H2O2, or c-Myc activation.  相似文献

Some epidemiological studies suggest that exposure to 50 or 60 Hz magnetic fields might increase the risk of leukemia, especially in children with a comparable high residential exposure. To investigate this possibility experimentally, the influence of 50 Hz magnetic-field exposure on lymphoma induction was determined in a mouse strain that is genetically predisposed to this disease. The AKR/J mouse genome carries the AK virus, which leads within 1 year to spontaneous development of thymic lymphoblastic lymphoma. Beginning at an age of 4-5 weeks, groups of 160 female mice were sham-exposed or exposed to 50 Hz magnetic fields at 1 or 100 microT for 24 h per day, 7 days per week, for 38 weeks. Animals were checked visually daily and were weighed and palpated weekly. There was no effect of magnetic-field exposure on body weight gain or survival rate, and lymphoma incidence did not differ between exposed and sham-exposed animals. Therefore, these data do not support the hypothesis that chronic exposure to 50 Hz magnetic fields is a significant risk factor for developing hematopoietic malignancy.  相似文献
