In alpine environments, flowering phenology can differ within local populations even at the same elevation. We assessed the effects of differences in flowering phenology due to snowmelt timing caused by local geographic heterogeneity on the genetic structure of a population of an alpine plant, Erythronium grandiflorum Pursh. We established a study plot of 250×70 m at 3,340 m above sea level in the Front Range of the Rocky Mountains, CO, USA. The flowering phenology was considerably influenced by snowmelt timing due to local geographic heterogeneity. Twenty-two patches of E. grandiflorum were recognized in the study plot and were classified into three phenological groups: early, middle, and late. To express the differentiation of flowering phenology among the patches, we defined phenological distance and analyzed the relationship between genetic and phenological distances. Additionally, since genetic distance is expected to co-vary with geographic distance, we also analyzed the relationship between genetic distance and geographic distance among patches. The results revealed not only that isolation by distance was present among patches, but also that the differences in snowmelt timing gave rise to phenologically distant patches of E. grandiflorum, which in turn determine the genetic structure caused by the limited pollen flow between patches.  相似文献

The timing of commitment during stenotele differentiation in Hydra was determined. Regeneration of isolated distal regions of the body column induces stenotele differentiation. The kinetics of appearance of committed stenotele precursors was determined in such regenerating pieces. Using [3H]thymidine labeling and hydroxyurea sensitivity, the G1/S and the S/G2 boundaries of the precursor population was also determined. Comparison of these results indicates that stenotele commitment is localized near the S/G2 boundary in the terminal cell cycle of nests of precursor cells.  相似文献

We examined the spatial genetic structure of Helianthus occidentalis Riddell ssp. occidentalis Riddell (western sunflower) to determine whether this species is highly clonal and whether the distance between prairie patches influences genetic differentiation. In the Edge of Appalachia Preserve System, Ohio, this species is restricted to prairie patches that have a clumped distribution in the forest matrix. Data from this insect-pollinated forb with gravity-dispersed seed were compared to data from the same patches for Asclepias verticillata, an insect-pollinated species with wind-dispersed seed. Allozyme electrophoresis was used to collect genetic data from H. occidentalis samples from eight patches in four regions. Genetic data from three polymorphic loci indicted that this species is not highly clonal. Genetic differentiation was greater among patches within a region than among regions, suggesting that gene flow among patches is more limited in H. occidentalis than in A. verticillata. Founder effect may also have contributed to observed genetic differences among patches as some of these populations may have re-established after release from human use. As habitat fragmentation is increasing in the preserve, it is also likely that genetic differentiation may be increasing. Therefore, monitoring of genetic structure is necessary to further assess the effect of fragmentation.  相似文献

Phyllonorycter mespilella (Hübner) is a leafmining moth with two stages of larval development: the initial sap-feeding (SF) stage followed by the tissue-feeding (TF) stage. Phenotypic selection by parasitoids on the duration of the SF stage (SF duration) was measured in artificial patches of larvae placed in the field during the diapausing generation. Pretreatment of larvae with different photoperiods allowed creation of patches that varied in the time-course of appearance of TF larvae. The shorter the photoperiod pretreatment, the sooner TF larvae tended to appear. Some patches were left exposed and others were caged to exclude parasitoids. Positive directional selection on SF duration was detected in exposed patches, and no selection was detected in caged patches. Directional selection in exposed patches was caused by both parasitoid oviposition and other unidentified sources of mortality. The other sources of mortality may have included host feeding by parasitoids on TF larvae. A larger proportion of parasitoid eggs were oviposited on TF larvae in patches where TF larvae appeared the earliest, but this variation in parasitoid oviposition did not result in significant differences in directional selection intensity among patches with early, intermediate and late appearance of TF larvae. Although the general form of the fitness function was very similar when compared among patch types, no significant directional selection could be detected in patches where TF larvae appeared late, and the causes of directional selection appeared to vary between patches where TF larvae appeared at early and intermediate dates.  相似文献

Summary Gene flow was investigated in a natural population of Lotus corniculatus L. (Fabaceae) using a combination of pollen and seed dispersal studies and a recombinant DNA technique. The population is spatially heterogeneous and grows with Empetrum nigrum. L. corniculatus is pollinated by the pollen-collecting bumblebee Bombus lapidarius L. Most pollinator flights occurred within patches, as bees usually visit nearest-neighbour plants, show no marked directionality, and forage mostly within patches. Gene flow by seeds is also limited, reinforcing the pattern of gene flow within patches. However, 2.6% of pollinator flights are between patches and considerable pollen carryover also occurs. Thus, gene flow between patches is potentially sufficient to retard or prevent genetic differentiation in spite of the patchy sub-structuring of the population. A sub-set of the population was analysed for restriction fragment length polymorphisms (RFLPs) to document the actual gene flow pattern of the population. The DNA analysis revealed significant levels of genetic differentiation between the patches. The level of gene flow that can be inferred from the distribution of genetic variation is surprisingly restricted, as compared to gene flow inferred from pollinator behaviour, and emphasizes that stochastic processes like genetic drift and founder effects may have a strong impact on the prevailing genetic structure.  相似文献

The sequence of events in posterior regeneration of the polychaete, Nephtys, has been examined in histological preparations from the fifth day to the end of the third week after amputation, that is from the time when wound healing is complete until several new segments are differentiated. The pygidium forms and begins to differentiate prior to segmentation. The first indication of each new segment is the appearance of a large pair of segmental blood vessels which arise from the vascular complex in the gut wall. Associated with these are fibroblasts, the anlagen of the new septum. Epidermal derivatives develop subsequently, appearing first ventrolaterally, close to the regenerating nerve cord. The mitotic rate appears to be highest prior to the period of maximum segment formation. Visible cell differentation follows, and subsequent growth of segments is primarily by cell enlargement. It appears likely that the blood-vascular system associated with the gut and the regenerating nerve cord, both of which are disproportionately large in the regenerate, are important for the initiation of new segments.  相似文献

We report the immunological differentiation of structures within the primary constriction. These include the kinetochore and the connecting strand, a structure which connects sister kinetochores. The location and temporal appearance of the connecting strand antigen suggest that it could play a role in the maintenance of sister chromatid pairing. In addition, we report the identification of a novel epitope that is localized to discrete patches along the entire length of the junction between sister chromatids at metaphase (the junction patch antigen). The patches on the inner surface of the euchromatic arms can be disrupted by Colcemid treatment while those found in the primary constriction remain intact. The apparent heterogeneity of the patches suggests that they may play different roles in the regulation of sister chromatid pairing. Because of their cytological localization and possible functional role, the junction patch and connecting strand antigens have provisionally been collectively termed CLiPs (Chromatid Linking Proteins'). All of these antigenic sites are shown to be distinct from centromeric heterochromatin, which can itself be immunologically differentiated from the euchromatic arms. The relationship between the antigenicity of the primary constriction and the unique manner in which chromatin is organized in this region is discussed.  相似文献

We conducted a population genetic analysis of the stalked kelp, Pterygophora californica, in the Santa Barbara Channel, California, USA. The results were compared with previous work on the genetic differentiation of giant kelp, Macrocystis pyrifera, in the same region. These two sympatric kelps not only share many life history and dispersal characteristics but also differ in that dislodged P. californica does not produce floating rafts with buoyant fertile sporophytes, commonly observed for M. pyrifera. We used a comparative population genetic approach with these two species to test the hypothesis that the ability to produce floating rafts increases the genetic connectivity among kelp patches in the Santa Barbara Channel. We quantified the association of habitat continuity and oceanographic distance with the genetic differentiation observed in stalked kelp, like previously conducted for giant kelp. We compared both overall (across all patches) and pairwise (between patches) genetic differentiation. We found that oceanographic transit time, habitat continuity, and geographic distance were all associated with genetic connectivity in P. californica, supporting similar previous findings for M. pyrifera. Controlling for differences in heterozygosity between kelp species using Jost's DEST, we showed that global differentiation and pairwise differentiation were similar among patches between the two kelp species, indicating that they have similar dispersal capabilities despite their differences in rafting ability. These results suggest that rafting sporophytes do not play a significant role in effective dispersal of M. pyrifera at ecologically relevant spatial and temporal scales.  相似文献

A cell-free model for the transfer of endocytosed material to lysosomes is described. Rat liver late endosomes, loaded in vivo with radiolabeled ligand by intravenous injection shortly before killing the animal, showed a specific interaction with lysosomes when incubated at 37 degrees C in the presence of cytosol and an ATP regenerating system. The location of the ligand, generally asialofetuin, was analyzed by isopycnic centrifugation on Nycodenz gradients. Appearance of radiolabel in the lysosomal position on such gradients was maximal after approximately 30 min at 37 degrees C and required the provision of undamaged cytosol, lysosomes, and an ATP regenerating system. It could not be accounted for by nonspecific bulk aggregation of membranes. Transfer occurred only from late endosomes; radiolabel in early endosomes was unaffected. Digestion of the asialofetuin, as shown by the appearance of TCA-soluble radioactivity, occurred on incubation at 37 degrees C and was increased by the provision of an ATP regenerating system.  相似文献

The mechanisms affecting forest regeneration in human-modified landscapes are attracting increasing attention as tropical forests have been recognized as key habitats for biodiversity conservation, provision of ecosystem services, and human well-being. Here we investigate the effect of the leaf-cutting ants (LCA) Atta opaciceps on regenerating plant assemblages in Caatinga dry forest. Our study encompassed 15 Atta opaciceps colonies located in landscape patches with a gradient of forest cover from 8.7% to 87.8%, where we monitored regenerating individuals (seedlings and saplings of woody and herbaceous plants) in different habitats (nests, foraging areas, and control areas) over one year. We recorded 2,977 regenerating plant individuals, distributed among 55 species from 23 families. Herbaceous plants represented 82.1% and 58.2% of the total number of individuals and species, respectively. Species richness of both the whole and herbaceous plant assemblages increased along the forest cover gradient, but without difference between the habitats. Total plant abundance was highest in control areas followed by foraging areas and nests and this pattern held for both woody and herbaceous plants. Although forest cover did not influence the abundance of herbaceous plants and the whole plant assemblage, it positively affects woody plant abundance across control areas. Forest cover and habitat changed species composition of both the entire regenerating and the herbaceous assemblages. These results together indicate that LCA negatively impact regenerating plant assemblages, particularly in those sites with increased forest cover. As LCA proliferate in human-modified landscapes, they may prevent plant regeneration of disturbed areas.  相似文献

The recent detection of dermal ridge configurations on the volar pads of the rat (Rattus norvegicus) has created opportunities for experimental studies of dermatoglyphics. In the present work, the palmar and plantar surfaces of the rat were studied to establish the feasibility of comparative rat and human dermatoglyphic investigations. The studied features included the volar pads and flexion creases. The number and location of the palmar and plantar pads in the rat were found to be similar to those of humans. The exception was a previously unrecognized small pad on the palms and soles of the rat, located on the radial and tibial side, respectively, of the proximal component of the first interdigital pad. This pad has no parallel in human embryos. Rats were found to have flexion creases in the non-pad areas between the neighboring pads, similar in location and appearance to those of humans. Unlike humans, however, rats also have boundary creases, separating the pad and non-pad areas. The marked similarities in the morphology of the volar areas between rats and humans make the rat ideally suitable for experimental studies of dermatoglyphics and flexion creases. Results of such studies should be applicable to human developmental dermatoglyphics, including those pertaining to medical disorders. © 1994 Wiley-Liss, Inc.  相似文献

The differentiation of somatotropocytes was studied in the Leghorn chick embryos during 11 to 18 days of incubation and in chickens during the first week of life using the immunohistochemical method of nonlabelled antibodies with PAP complex and antiserum against human somatotropic hormone (STH). Unlike in humans, the fixation of pituitaries in the Carnoy mixture is optimal for STH to be immunohistochemically estimated in the chickens and chick embryos. STH was found in adenohypophysis from 12-13 days of development. Besides predominant localization of somatotropocytes in the adenohypophysis caudal lobe, individual STH-positive cells are also present in the cephalic lobe of chickens and chick embryos. The results obtained suggest a relatively late appearance of STH during histotypical development of adenohypophysis in chick embryos.  相似文献

A histological evaluation of the effects of hypophysectomy and throxine therapy in young tail regenerates was carried out in the small iguanid lizard, Anolis c. carolinensis. Hypophysectomy caused a delay but did not inhibit blastema formation. The growth of the ependyma into the wound region was delayed in hypophysioprivic regenerates by about a week. Growth and differentiation of hypophysioprivic regenerates after blastema formation was variable, ranging from virtually no growth to the formation of a differentiated but very small protuberance. However, actual tail elongation was inhibited by hypophysectomy. In those hypophysioprivic regenerates that did show signs of differentiation, muscle groups were poorly defined, scanty in appearance and not as well differentiated as the cartilage tube. Thyroxine treatment in the young hypophysioprivic regenerates stimulated normal growth and normal appearance and differentiation of promuscle and procartilage aggregates as well as the growth of the ependymal tube into the blastema.  相似文献

Efferent duct (ED) differentiation was examined histologically during female-to-male sex change in the honeycomb grouper Epinephelus merra . During natural sex change, ED differentiation began with the appearance of slit-like structures between the stromal tissue and the tunica ovary and small oval-shaped spaces within the wall of the ovarian cavity, accompanied by oocyte degeneration and the initiation of spermatogenesis, i.e. the early transitional phase. In the late transitional phase, ED structure formation expanded and further ED differentiation occurred, including the rapid multiplication of spermatogonial germ cells. In sex-changed males, the slit-like structures increased in size, fused with each other and finally formed a well-developed ED. The oval-shaped spaces also increased in size and fused to form an ED. In contrast, during artificial sex change, induced by aromatase inhibitor (AI, 1 mg kg−1), ED differentiation in E. merra was first observed as the appearance of slit-like structures and small oval-shaped spaces in the restructuring gonads in the third week after AI treatment. These were accompanied by oocyte degeneration and the proliferation of gonial germ cells into spermatogonia. In the fifth week, the rapid multiplication of spermatogonial germ cells, increases in 11-ketotestosterone (11-KT), and further differentiation of EDs were observed. Sex-changed males had testes containing sperm in the completely differentiated EDs; the significantly highest levels of 11-KT were observed in the sixth week. Simultaneous increases in 11-KT and initiation of ED differentiation were observed, suggesting a role of 11-KT in ED differentiation during sex change. There were no basic differences in the mechanisms of natural and artificially induced ED differentiation. Two types of structure led to the formation of EDs in two different areas of the newly formed testis during sex change.  相似文献

Purpose

To investigate light transmission and preference for six eye patches for occlusion therapy.

Methods

Six patches were examined, including; Ortopad Fun Pack, Ortopad Flesh, Kawamoto A-1, Kawamoto A-2, 3M Opticlude, and Everade Eye Guard. The size and the presence of a light blocking pad of patches were investigated. The amount of light transmitted through the patches was evaluated, using a digital light meter and a model eye, in three different environments; indoors with fluorescent light, outdoors on a sunny day, and strong light from illuminator. After patching the normal eye, the flash visual evoked potential (VEP) was measured. Thirty patients with amblyopia or horizontal strabismus, who received occlusion therapy as initial treatment, were included. After using all six patches, patients completed a 7-item questionnaire regarding the patch preference for size, color and shape, adhesive power, pain with removal, skin irritation after removing patch, parent’s preference and overall opinion.

Results

All patches had a light-blocking pad, except the 3M Nexcare. Ortopad had the strongest light blocking power in the three environments, and the 3M Nexcare had the weakest power. In flash VEP, Ortopad and Kawamoto patches showed flat, but 3M Nexcare and Everade Eye Guard showed normal response. There were significant preferential differences among the patches in all the items of the questionnaire (P<0.05). In comparison between the patches respectively, 3M Nexcare received the lowest satisfaction in pain when removing a patch and skin irritation after removing a patch. Kawamoto A-2 received the lowest score in the overall satisfaction.

Conclusions

We found differences in the light-blocking power and in the preference of the various patches for the occlusion treatment. This is a pilot study regarding only characteristics and preferences of patches. Further clinical studies regarding the relationship between characteristics or preferences of patches and outcomes of occlusion treatment are needed.  相似文献

The developmental increase in the level of NGF mRNA in mouse maxillary process/whisker pad is paralleled in vivo by the biochemical and morphological differentiation of whisker pad epidermis, i.e., changes in the keratin expression pattern and the appearance of hair follicles. In cultures of maxillary processes, however, depending on the age of explanted tissue, the increase in NGF mRNA levels either precedes or follows the appearance of epithelial differentiation markers. In addition, we found that prevention of epithelial differentiation by retinoic acid did not affect the increase in NGF mRNA levels. Only in explants from E11.5 embryos was the timing of NGF mRNA production comparable to that of the in vivo situation, whereas at earlier stages (E10/10.5) NGF mRNA levels increased slowly but never reached in vivo levels, even after extended culture periods. However, the amount of NGF mRNA in E10/10.5 maxillary processes was strongly increased in the presence of medium conditioned by E11.5 explants. This effect was not mimicked by the factors IL-1 beta and TGF-beta 1 known to induce NGF mRNA in other systems. It is concluded that the developmental increase in NGF mRNA levels in developing mouse whisker pad is not linked to epidermal differentiation. Interestingly, it is strongly stimulated by a soluble factor(s) produced within the tissue.  相似文献

Abstract. Vegetation maps serve as the basis for spatial analysis of forest ecosystems and provide initial information for simulations of forest landscape change. Because of the limitations of current remote sensing technology, it is not possible to directly measure forest understory attributes across large spatial extents. Instead we used a predictive vegetation mapping approach to model Tsuga heterophylla and Picea sitchensis seedling patterns in a 3900‐ha landscape in the Oregon Coast Range, USA, as a function of Landsat TM imagery, aerial photographs, digital elevation models, and stream maps. Because the models explained only moderate amounts of variability (R2 values of 0.24–0.56), we interpreted the predicted patterns as qualitative spatial trends rather than precise maps. P. sitchensis seedling patterns were tightly linked to the riparian network, with highest densities in coastal riparian areas. T. heterophylla seedlings exhibited complex patterns related to topography and overstory forest cover, and were also spatially clustered around patches of old‐growth forest. We hypothesize that the old growth served as refugia for this fire‐sensitive species following wildfires in the late 19th and early 20th centuries. Low levels of T. heterophylla regeneration in hardwood‐dominated forests suggest that these patches may succeed to shrublands rather than to conifer forest. Predictive models of seedling patterns could be developed for other landscapes where georeferenced inventory plots, remote sensing data, digital elevation models, and climate maps are available.  相似文献

The patchy local distribution of the common milkweed, Asclepias syriaca, organizes populations of a beetle that feeds on it, Tetraopes tetraophthalmus, into numerous local demes. Genetic and ecological characteristics of demes of adult milkweed beetles occupying two naturally occurring size classes of patches, defined as large and small, were studied in order to describe the effect of patch size variation on local population structure. Allele frequency variance in two of three protein polymorphisms was significantly greater in collections of beetles from an array of 13 small patches when compared to collections from an array of 11 large populations. A multivariate measure of variance using information from all 3 genetic markers confirmed that the small patches displayed greater overall genetic differentiation. This was further quantified by computing an Fst value, combined across loci, of 0.018 for the small patches and 0.004 for the large patches. No significant difference between patch size classes in mean allele frequency was detected. Mark and recapture studies of the adults found in five small and four large patches showed the residence times of adults in small patches to be less than half of those found in large patches. This was interpreted as resulting from higher emigration rates from small patches. It is proposed that greater genetic differentiation is found among demes from smaller patches because smaller patches support smaller population sizes and further because smaller patches act as net exporters of migrants while larger patches act as net migrant importers.  相似文献

Binding sites for Concanavalin A have been located in the ciliate Stentor coeruleus by utilizing FITC-Con A and fluorescence microscopy. When both nonregenerating and regenerating Stentor are fixed prior to FITC-Con A exposure, FITC-Con A binds intensely to the cilia of the membranellar band and to the somatic cilia that cover much of the cell surface. No binding is observed between the ciliary rows. The FITC-Con A also binds to the developing oral primordia of regenerating cells. Binding of FITC-Con A in the early stages of regeneration (prior to stage 4) appears to be less intense than that in the later stages. Additional FITC-Con A binding appeared as a granular fluorescence in the area of the developing buccal cavity beginning at about stage 4 and disappearing around stages 6–7. The presence of α-D-methyl mannoside prevented the binding of FITC-Con A to either regenerating or nonregenerating cells. If nonregenerating Stentor are exposed to FITC-Con A prior to fixation, the binding pattern is entirely different with the fluorescence primarily in the form of random, granular patches spread over much of the cell but with no binding to either type of cilia. These results demonstrate that membrane glycoproteins capable of binding Con A are located primarily in the membranellar and somatic cilia and in the developing oral primordia during oral regeneration in Stentor. Concanavalin A binding to these sites may be involved in the Con A-induced inhibition of oral regeneration observed in earlier studies.  相似文献
