Improved laboratory methods are described in detail for mass rearing of various thrips species, such as Frankliniella occidentalis, Frankliniella intonsa, Thrips palmi, Thrips tabaci (Thysanoptera: Thripidae) and a thrips parasitoid, Ceranisus menes (Hymenoptera: Eulophidae), using various foods. In one method, plant pollen and honey solution are used as food sources. In a second method, germinated broad bean seeds are used. Eggs, produced in large numbers in water, are collected by a suction funnel onto a filter paper and incubated in a Petri dish. Large numbers of larvae that hatch are collected by using food traps (plant pollen). Larvae can be reared on pollen or on germinated broad bean seeds until adult emergence without additional water and food. This method has been found useful for producing even-aged thrips at different densities (up to 500 larvae in a cage of 80 mm diameter) with relatively low mortality rates. Evaluation of this rearing method for F. intonsa, shows that during 2 weeks at 20 °C per 100 females more than 4000 females could be produced in the next generation. About 5 min per day is required to achieve this productivity of mass production. The method is also suitable for producing large numbers of the solitary endoparasitoid of thrips larvae, C. menes.  相似文献

Methods are described for the in vivo production of the nematode Thripinema nicklewoodi (Siddiqi), an obligate parasite and potential biological control agent of western flower thrips Frankliniella occidentalis (Pergande). Nematode infection is not lethal but causes sterilization of adult female hosts. Both fertilization and horizontal transmission of T. nicklewoodi is achieved in 1.5-ml microcentrifuge tubes (infection arenas), in the presence of 100% humidity, a temporary food source and preferably a damp substrate. Following exposure to infection arenas, F. occidentalis are reared on excised bean leaves Phaseolus vulgaris (L.) in polypropylene containers for 2 wk at 25 degrees C to allow the reproduction and development of a single generation of nematodes within infected hosts's abdominal cavity. To identify infected hosts after this incubation period, thrips are isolated in microcentrifuge tubes and monitored for free-living nematodes being released along with frass. Infected thrips are reintroduced back into infection arenas to inoculate further thrips to maintain the culture. We documented the output of the rearing procedure using a standard method and following simple manipulation of several individual parameters of the infection technique. The standard method was the most efficient, and resulted in an increased (output/input) ratio of infected thrips of approximately 2; i.e., the number of infected thrips approximately doubles each generation. Monitoring infected thrips revealed that nematodes were first released between 12-14 d postinfection and for an average of 7.9 d at 25 degrees C; highlighting the potential to reuse infective thrips between infection arenas. The possibility of using T. nicklewoodi as an inoculative agent against F. occidentalis infesting floricultural crops is discussed.  相似文献

Rabbit immunoglobulin G (R‐IgG) was used successfully as an external mark for thrips. Females of both Thrips tabaci Lindeman and Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae) were marked with 1 mg ml?1 R‐IgG solution with 1%‘Tween 20’ by the contact exposure method. Determining the retention of the mark was by running the rinsing solution of individual thrips in an enzyme‐linked immunosorbent assay (ELISA). The sandwich ELISA method was used with an additional biotin–avidin step. The threshold for a positive marking score was defined as three times the mean optical density readings of the negative control thrips. Under laboratory conditions, on bean pods, all marked thrips scored positive up to 6 days after marking (DAM). When marked thrips were kept in the laboratory on marigold flowers for 2 days, they all scored positive. When marked and unmarked thrips were placed together on these flowers, the mark was transferred to 10–20% of the unmarked thrips and they became positive. Under field conditions, on sticky traps covered with water‐base glue, 100, 80, and 20% of the marked T. tabaci scored positive by the 3rd, 6th, and 9th DAM, respectively. Under the same conditions 100, 90, and 10% of the marked F. occidentalis scored positive by the 3rd, 6th, and 9th DAM, respectively. The retention of the R‐IgG decreased significantly under conditions of wetness and high humidity. After 6 days on chive plants kept at 80–100% r.h., all marked thrips scored negative while on plants kept at 40–60% r.h., 85% of the marked thrips scored positive. Rabbit IgG can be used as an external marker for thrips. The suitability of this marking method for dispersal studies of these important pests needs to be evaluated.  相似文献

介绍一种以甘蔗为饲料饲养红棕象甲Rhynchophorus ferrugineus Fab.的方法。将新鲜甘蔗切成段,置于培养皿中,放入恒温培养箱内,设置温度:(26±1)℃,RH:80%±10%,光周期:L∶D=0∶24。利用该方法饲养的红棕象甲世代历期130.80d,老熟幼虫平均体重5.51g,成虫单雌产卵量144.42粒,世代存活率21.67%。甘蔗饲养红棕象甲成本低,生物学特性恒定,可作为替代寄主扩繁虫源。  相似文献

Western flower thrips (WFT), Frankliniella occidentalis (Thysanoptera: Thripidae), is the most important insect pest for greenhouse flower crops, but chemical control is often difficult because of the thrips' location in flowers and buds. Thripinema nicklewoodi (Tylenchida: Allantonematidae) is an entomoparasitic nematode that attacks and sterilizes WFT, but its biology and impact on WFT are poorly understood. Methods to propagate and study T. nicklewoodi with a microscope slide arena for observation and a rolled bean leaf arena for rearing were developed. On average, 21.4 nematodes were excreted per day by a parasitized female WFT. The sex ratio of the excreted nematodes was 6.0/1.0 (female/male). After dissection of adult WFT, a maximum of 11 ovoid-shaped first-generation nematodes in a female thrips and 6 in a male thrips were found. There were more second-generation nematodes in the adult female WFT (192.6) than in the adult male WFT (93.7). When 50 healthy first instar WFT were exposed to 4 parasitized female WFT in a rolled bean leaf, a 75.3% mean parasitization rate in the adult stage of the thrips was obtained. In contrast to previous reports, male WFT can be parasitized as readily as females. Parasitism reduced the longevity of both adult female and adult male WFT, and the degree of reduction was higher in adult male WFT. T. nicklewoodi, when presented with various WFT life stages (first instar, second instar, prepupa, pupa, adult female, and adult male), achieved the highest attack rate in first and second instars and prepupa. The free-living nematodes excreted by the hosts actively migrated to a trichome on leaf disks in the observation arena and moved up this structure. Then, the nematode actively waved the anterior part of its body while attaching itself to the trichome with the posterior part of its body. After a nematode contacted the leg of a thrips, the nematode immediately moved up along the leg toward the abdomen of the host. Increased understanding of the biology of T. nicklewoodi is important to better assess its potential for biological control of WFT.  相似文献

In insect reverse genetics, dietary delivery of interfering RNAs is a practical approach in nonmodel species, such as thrips, whose small size, and feeding behavior restricts the use of other delivery methods. In a laboratory context, an unsuitable diet could confound the interpretation of an RNA interference (RNAi) phenotype, however well-formulated artificial diets can minimize experimental variability, reduce the need for insect handling, and can further be used for roles, such as delivering double-strand RNA (dsRNA)-expressing recombinant bacteria. In this study, artificial diets for oral delivery of dsRNA were developed for two important pest thrips species, western flower thrips (Frankliniella occidentalis) and onion thrips (Thrips tabaci), with the goal of (a) stimulating feeding behavior, (b) supporting optimal growth rates of dsRNA-expressing symbiotic bacteria, and (c) nutritionally supporting the thrips for sufficient periods to observe RNAi phenotypes. The efficacy of artificial diets for ingesting “naked” dsRNA or dsRNA-expressing symbionts and dsRNA delivery via host plant uptake was evaluated. Compared with previously published diet formulations, new combinations based on tryptone, yeast, and soy were superior for enhancing feeding and longevity. However, simply adding “naked” dsRNA to an artificial diet was an unreliable form of RNAi delivery in our hands due to dsRNA degradation. Delivery via host plants was more successful, and the new diet formulation was suitable for symbiont-mediated dsRNA delivery, which we believe is the most convenient approach for large-scale knockdown experiments. This study, therefore, provides alternative methodologies for thrips rearing, dietary RNAi delivery, and insights into the challenges of performing dietary RNAi in nonmodel insects.  相似文献

Frankliniella zucchini transmits zucchini lethal chlorosis virus, causal agent of lethal chlorosis of zucchini squash. The characteristics of relationship between this virus with its vector have not been studied, one of the reasons being the lack of a method for rearing the thrips for laboratory studies. This work proposes a system for the rearing of F. zucchini on fresh virus free zucchini ‘Caserta’ fruits, offering a practical and efficient alternative for the supply of a large number of insects for later study of virus/vector relationship. In addition, to aid in the identification of this species of thrips, the immature and adult forms obtained from the colony were described.  相似文献

The sterile insect technique (SIT) has been widely used to suppress several fruit fly species. In southern Brazil, millions of sterile flies of the South American fruit fly, Anastrepha fraterculus Wiedemann (Dipetra: Tephritidae), will be produced in a mass‐rearing facility called MOSCASUL to suppress wild populations from commercial apple orchards. In spite of standard rearing conditions, the quality of pupal batches can be inconsistent due to various factors. The quantification of poor quality material (e.g. empty pupae, dead pupae or larvae) is necessary to track down rearing issues, and pupal samples must be taken randomly and evaluated individually. To speed up the inspection of pupal samples by replacing the manual testing with the mechanized one, this study assessed a multispectral imaging (MSI) system to distinguish the variations in quality of A. fraterculus pupae and to quantify the variations based on reflectance patterns. Image acquisition and analyses were performed by the VideometerLab4 system on 7‐d‐old pupae by using 19 wavelengths ranging from 375 to 970 nm. The image representing the near infrared wavelength of 880 nm clearly distinguished among high‐quality pupae and the other four classes (i.e. low‐quality pupae, empty pupae, dead pupae and larvae). The blind validation test indicated that the MSI system can classify the fruit fly pupae with high accuracy. Therefore, MSI‐based classification of A. fraterculus pupae can be used for future pupal quality assessments of fruit flies in mass‐rearing facilities.  相似文献

Brazilian peppertree, Schinus terebinthifolia Raddi (Sapindales: Anacardiaceae) (hereafter Schinus), is one of the worst invasive species in Florida and Hawaii. The thrips Pseudophilothrips ichini Hood (Thysanoptera: Phlaeothripidae) is being considered as a potential biological control agent of Schinus. Two populations of this thrips were collected in the weed's native range; one from central-east Brazil (Ouro Preto thrips) and a second from north-east Brazil (Salvador thrips). Temperature requirements, adult fecundity and impact on different plant haplotypes by P. ichini were examined in the laboratory. Complete development of thrips from both populations occurred at temperatures ranging from 20 to 30°C. Two approaches were used to model the predicted distributions of the thrips populations in the USA: the physiological model (NAPPFAST) based on cold tolerance and the ecological niche model based on climatic variables (MaxEnt). The physiological model predicted that both populations of P. ichini may establish in similar areas of the USA, overlapping with the distribution of Schinus. However, the niche model predicted that only the Ouro Preto thrips could establish in the USA. The difference in model predictions suggests an apparent preadaptation of the Salvador thrips to lower temperatures than those experienced at the locations they were collected in Brazil. The Ouro Preto thrips had similar fecundity on two Florida Schinus haplotypes, whereas lower fecundity on haplotype A was found for the Salvador thrips. Based on these results, the Ouro Preto population may be better adapted to the climatic conditions and plant haplotypes found in Florida. Moreover, greenhouse studies indicated that Schinus growth was greatly reduced by thrips feeding, which may result in lower weed reproduction and densities in the field.  相似文献

【目的】研究实验室条件下韭蛆 Bradysia odoriphaga Yang et Zhang大量饲养的方法以解决试虫不足的问题。【方法】本实验在25℃,相对湿度75%,光周期16L:8D条件下,通过控制变量法筛选出人工饲料配方,运用两性生命表软件,分析比较人工饲料与韭菜Allium tuberosum的饲喂效果。【结果】对不同成分和不同含量的8种韭蛆人工饲料配方筛选,结果确定最优的人工饲料配方成分和含量为:平菇粉6 g,韭菜粉5 g,琼脂粉1.25 g,山梨酸0.04 g,苯甲酸0.05 g,酵母粉0.5 g,维生素C 2.5 g和水50 mL。该配方和韭菜饲养效果相当,不存在显著差异(P>0.05)。同时,两性生命表结果表明,人工饲料饲喂的韭蛆平均世代周期23.6800 d,韭菜饲喂的韭蛆世代周期为25.9000 d,两者之间差异显著(P<0.05)。生殖生物学特性分析发现:韭菜和人工饲料饲喂的韭蛆雌成虫净生殖率分别为16.3200和41.1800/d,内禀增长率分别为0.1078和0.1570/d。【结论】筛选的人工饲料和韭菜均可以稳定地完成韭蛆的世代繁殖,且人工饲料饲喂效果略优于韭菜,另外人工饲料较韭菜不但具有低成本省人工的优点,而且可解决韭菜反季节供应和农药残留问题,保证稳定的虫源供给。  相似文献

The searching strategies ofOrius insidiosus (Say) exposed to various densities of the soybean thrips,Sericothrips variabilis (Beach), were observed on soybean trifoliolates under laboratory conditions. The effects of prey density on the predator's searching speed and feeding time were evaluated. Predator searching speed and feeding time per soybean thrips declined with increases in prey density. The greatest number of predator-prey encounters occurred on the upper midrib of the soybean trifoliolate. Adult soybean thrips were captured in 45 and 56% of their total encounters with adult male and femaleO. insidiosus, respectively. Running was found to be the primary escape tactic used by soybean thrips in response to attack byO. insidiosus.
Résumé Les stratégies de recherche deOrius insidiosus (Say) exposé à des densités variées de thrips du soja,Sericothrips variabilis (Beach), furent observées sur du soja à 3 folioles en laboratoire. Les effets de la densité de la proie sur la vitesse de recherche et sur le temps nécessaire au prédateur pour se nourrir furent évalués. La vitesse de recherche et le temps de nourriture du prédateur sur le thrips du soja déclinèrent avec l'accroissement de la densité de la proie. Les rencontres prédateur-proie eurent lieu dans leur majorité dans la partie supérieure de la nervure du soja. Les thrips du soja adultes furent capturés respectivement dans 45 et 65% du total de leurs rencontres avec des males et des femelles adultesO. insidiosus. Courir s'est avéré être la tactique primaire de fuite utilisée par les thrips de soja en réponse aux attaques deO. insidiosus.

According to the present practice of Hungarian greenhouse sweet pepper production, only exotic agents are used for biological control purposes against thrips pests. The suitability for biological control of the second instars of an indigenous species, the common crab spider, Xysticus kochi Thorell (Araneae: Thomisidae) was tested on thrips species, Frankliniella occidentalis (Pergande) and Frankliniella intonsa (Trybom) (Thysanoptera: Thripidae) using a cage set-up on greenhouse sweet pepper plants. Effects of introducing second instars of X. kochi on thrips infested plants were judged by assessing the degree of aesthetical damage, the commercial value of the fruits (degree of economic loss) and the quality composition of the harvested peppers. The estimated damaged surface unit was significantly lower in control plants than in plants infested by F. occidentalis. A significant effect of the spider introduction was observed in all of the pepper quality indicators applied. We suspect that direct Xysticus predation or any other predator-induced effect resulted in reduced levels of damage on harvested peppers. However, further investigation is needed to detect the origin (e.g., predation and non-predation effects) of the suppression of thrips damage. Our results suggest that X. kochi could be an effective component of greenhouse antagonistic fauna and the application of mulch may encourage the effectiveness of spiderlings. Cucurbit fly is an important pest of cucurbit plants (Cucumber, melon and watermelon) in most mid-east countries including, Iran. This insect causes a high damage so that several sprayings are required to control the pest. Using male strile is another physical method for controlling this pest. For this purpose it is necessary to rear many male insects. Therefore an investigation was carried out to compare different artificial diets for rearing the melon fly. First, pupae were collected from cucurbit field and transferred to laboratory. In this experiment five diet formulations were compared and the following was used for rearing: Wheat bran (14 g), soybean lees (3 g) sugar (50 g), yeast extract (2.7 g) nipagin (0.1 g). sodiumbenzoate (0.1 g) Hydrochloric acid 3.5% (4.0 ml) and distilled water 70.5 mil. For rearing the adult, the best diets contained: Brower yeast (1 part), Honey (5 parts), water (94 parts) and a slice of cucumber. The ecosystems supporting Pacific salmon (Oncorhynchus spp.) are changing rapidly as a result of climate change and habitat alteration. Understanding how—and how consistently—salmon populations respond to changes at regional and watershed scales has major implications for fisheries management and habitat conservation. Chinook salmon (O. tshawytscha) populations across Alaska have declined over the past decade, resulting in fisheries closures and prolonged impacts to local communities. These declines are associated with large‐scale climate drivers, but uncertainty remains about the role of local conditions (e.g., precipitation, streamflow, and stream temperature) that vary among the watersheds where salmon spawn and rear. We estimated the effects of these and other environmental indicators on the productivity of 15 Chinook salmon populations in the Cook Inlet basin, southcentral Alaska, using a hierarchical Bayesian stock‐recruitment model. Salmon spawning during 2003–2007 produced 57% fewer recruits than the previous long‐term average, leading to declines in adult returns beginning in 2008. These declines were explained in part by density dependence, with reduced population productivity following years of high spawning abundance. Across all populations, productivity declined with increased precipitation during the fall spawning and early incubation period and increased with above‐average precipitation during juvenile rearing. Above‐average stream temperatures during spawning and rearing had variable effects, with negative relationships in many warmer streams and positive relationships in some colder streams. Productivity was also associated with regional indices of streamflow and ocean conditions, with high variability among populations. The cumulative effects of adverse conditions in freshwater, including high spawning abundance, heavy fall rains, and hot, dry summers may have contributed to the recent population declines across the region. Identifying both coherent and differential responses to environmental change underscores the importance of targeted, watershed‐specific monitoring and conservation efforts for maintaining resilient salmon runs in a warming world.  相似文献

In Turkey, the western thrips Frankliniella occidentalis (Pergande) is a key pest affecting eggplants grown in greenhouses for which an appropriate control strategy is under investigation. It was observed in a previous study that the release of the beneficial predatory mite Amblyseius swirskii (Athias‐Henriot) alone did not result in an effective control of thrips on eggplants. Since pollen is known to improve control efficiency of predators, this study was undertaken to investigate if provision of pollen to eggplants can greatly improve the efficiency of A. swirskii in controlling thrips. The experiments were carried out in both greenhouse and low tunnel. The provision of pollen led to a significant increase in the predator population density on the eggplants but did not result in an effective control of the thrips populations. In this paper, various factors are discussed that could have affected the efficiency of the predatory mite in controlling F. occidentalis on eggplants.  相似文献

The effects of larval density on the wing form determination of female tobacco thrips, Frankliniella fusca, were investigated by rearing thrips on leaf disks at 27.5 °C. The developmental period, head width, body length, and forewing length of individuals in each wing morph were determined to assess the relationships among larval density, growth, and wing form. Data showed that higher rearing densities increased the production of female F. fusca brachypters. There was no consistent difference in the mean developmental periods between the two wing morphs or among all 5 density treatments. The body length of females tended to decrease with increasing rearing density, but there was no significant difference in body size between the two wing morphs when they were reared under the same density level.  相似文献

Summary In the polyandrous honey bee, Apis mellifera, workers can potentially increase their inclusive fitness by rearing full-sister queens. If the mother queen dies suddenly, workers feed a few larvae in worker cells with royal jelly and rear them into queens (emergency queen rearing). Using DNA microsatellite markers we determined the patriline of emergency queens reared in two colonies headed by naturally-mated queens before being made queenless. We found that some patrilines were reared more than others in one colony, but not in the other. These differences between colonies suggest that selective rearing is not always present and this might explain the mixed results of previous nepotism studies in the honey bee.Received 10 February 2003; revised 7 March 2003; accepted 17 March 2003.  相似文献

Rearing insects in controlled conditions is a prerequisite to supply high-quality specimens for bioassays. However, while artificial diets and standardized rearing methods have been developed for many phytophagous insects, especially Lepidoptera, there are limited published diets for root-feeding Coleoptera which are commonly fed either grass roots or pieces of vegetables as a simple alternative to artificial diets during bioassays. These feeding options, while convenient, can be considered suboptimal as they do not maximise the insects' development and health. Additionally, it is also important to develop standardised screening methods designed to test sublethal effects of control agents which may have repellent, antifeedant, antimetabolic and/or delayed mortality effects. The greyback canegrub (Dermolepida albohirtum, Waterhouse) is the most damaging native pest of Australian sugarcane, but no rearing method or artificial diet has ever been developed for this species. Our objectives were to improve bioassay methodology for D. albohirtum by describing and developing standard rearing and health assessment protocols. We describe a successful rearing method to raise healthy D. albohirtum larvae with a total of 48.8% of first instars successfully moulting to the second instar. We also tested a modified artificial diet which increased the weight, size and food uptake of larvae compared to traditional methods (i.e., pieces of carrots). For example, the average weight increase of larvae fed with the modified diet was 3.4 times higher than for carrot-fed larvae while modified diet-fed larvae were 2.1 times wider than if they were fed with carrots. Finally, we developed a method to measure larval activity which can be used to identify sublethal effects of control agents such as effects on activity level. Our methods may also be applied to improve bioassay methodology for other root-feeding Coleoptera.  相似文献

Western flower thrips (WFTs), Frankliniella occidentalis Pergrande, and onion thrips (OTs), Thrips tabaci Lindeman, are two cosmopolitan insect pests of agricultural and horticultural plants. Understanding the occurrence and development of thrips on plants is crucial for identifying suitable plants that can be used for developing a “push-pull” strategy against thrips. In this study, the dynamics of WFTs and OTs on plants (Allium fistulosum L., Medicago sativa L., Luffa cylindrica (L.) Roem., Ocimum basilicum L., and Schizonepeta tenuifolia (Benth.) Briq.) were investigated for two consecutive years (2018–2019). Throughout the survey, the abundances of both thrips species were strongly associated with plant species and plant phenology; both thrips species were present at relatively high densities on M. sativa but very low densities on O. basilicum and S. tenuifolia. Populations of both thrips species greatly increased during plant flowering. A Y-tube olfactory test was used to study the effects of plant volatiles in mediating thrips behaviour and showed that volatiles of M. sativa were attractive to both thrips species whether emitted by the plant in the vegetative or flowering stage, while volatiles of O. basilicum and S. tenuifolia were repellent to thrips. Additionally, because of the presence of a high number of floral chemicals, both thrips species exhibited a greater preference for volatiles emitted by plants in the flowering period over those emitted by plants in the vegetative period. Our observations indicate that plant species and flowering status play an important role in the abundance dynamics of thrips and that the volatiles of flowering plants attract thrips more strongly than volatiles emitted by vegetative plants. These findings can facilitate the screening of attractive/unattractive plants for developing push-pull strategies to control thrips.  相似文献
