Root rot severity of asparagus plants grown in sterilized field soil inoculated with Fusarium oxysporum f . sp . asparagi (Foa) was reduced by more than 50% when the soil was precolo nized by each of 13 non - pathogenic (np) isolates of F. oxysporum originating from asparagus roots or field soils . In a greenhouse experiment , application of six np isolates to naturally infested field soil was followed by a 23 - 49% decrease of disease severity , depending on the isolate . One of them , Fo47 originating from Fusarium suppressive soil in France , was applied to field plots infested with Foa . Foa root rot was not suppressed in asparagus plants grown for 1 year in these plots . Pathogenic and np isolates extensively colonized the root surface and isolates of both types infected the roots of asparagus plants grown in sterilized field soil , with significant differences among the np isolates . Inoculation of sterilized field soil with np isolates reduced germination of Foa chlamydospores by 43 - 64% depending on the isolate used . It is concluded that np isolates of F. oxysporum can suppress asparagus root rot caused by Foa in naturally infested field soil . The differences for root colonization capacity among the np isolates imply that selection for this trait might reveal isolates that perform better under field conditions .

Soybean root rot, caused primarily by Fusarium solani f. sp. phaseoli in a complex with F. oxysporum and Rhizoctonia solani, has become an increasing problem for soybeans, dry beans, and other rotation crops in central Minnesota due to soil conditions associated with reduced tillage. This study was conducted, in two field sites in central Minnesota located near Staples and Verndale, to develop methods for nondestructive assessment of root rot severity using plant radiometric properties. Soybean canopy reflectance was measured with a hand-held multi-spectral radiometer. Prior to the radiometer measurements, attempts were made to create differing root rot situations with moldboard or chisel tillage, and with or without a biological seed treatment. Root rot severity was estimated using a visual disease severity scale. Colony-forming units (CFU) were determined to estimate soil populations of pathogenic F. solani and F. oxysporum. Results from the Verndale site consistently showed significant treatment effects in the measured canopy radiometric parameters, and in the visual disease rating and yield (significant for seed treatment). Values of a simple ratio vegetation index from this site exhibited negative relationships with disease rating and F. oxysporum CFU, and a positive linear relationship with yield. Treatment effects were generally not significant at the Staples site because of low initial F. oxysporum populations. The results indicate that remote sensing is potentially a rapid, nondestructive means for assessment of root rot diseases in soybean.

A new isolate of Trichoderma harzianum (T-35) was isolated from the rhizosphere of cotton plants from a field infested with Fusarium. Under glasshouse conditions, the antagonist was applied to soil growing in a bran/peat mixture (1:1, v/v) or as a conidial suspension or used as a seed coating. When T. harzianum was tested against Fusarium oxysporum f. sp. vasinfectum, F. oxysporum f. sp. melonis or F. roseum‘Culmorum”, a significant disease reduction, was obtained in cotton, melon and wheat, respectively. Biological control of Fusarium wilt of cotton was achieved when tested at two inoculum levels of the pathogen (2 × 107 and 2 × 108 microconidia/kg soil), decreasing the Fusarium spp. soil population. The long term effect of T. harzianum on Fusarium wilt of cotton was studied using successive plantings. The antagonist persisted in soil throughout three consecutive plantings, reducing the Fusarium, wilt incidence in each growth cycle. At the first planting the largest amount of preparation was found superior, whereas at the third planting, no significant difference could be observed between the four rates of Trichoderma preparation. T. harzianum (T-35) controlled Fusarium wilt in cotton and muskmelon when applied in both naturally or artificially infested alluvial vertisol and sandy-loam soils, respectively. Soil or seed treatments with the antagonist provided a similar disease control of F. roseum‘Culmorum’ and of F. oxysporum f. sp. melonis.

Fusarium oxysporum is one of the major pathogens causing root and crown rot in asparagus. Breeding of cultivars resistant to F. oxysporum would be the most efficient strategy for pathogen control. In this study, a bioassay was developed for screening seedling resistance. The non‐destructive bioassay comprises inoculation with a highly aggressive F. oxysporum isolate, incubation in a climate chamber and quantification of disease symptoms by a digital image analysing system and a PTA‐ELISA. This bioassay is simple to implement and demonstrated high reproducibility. Subsequently, it was used to determine the resistance behaviour of 16 asparagus genotypes to F. oxysporum. The asparagus cultivars revealed different levels of susceptibility, whereas the wild relative A. densiflorus was confirmed to be resistant.

Asparagus crown and root rot caused by Fusarium oxysporum f.sp. asparagi (Foa), F. proliferatum (Fp) and F. solani (Fs) result in early decline and loss of crop production. The role of several crop species on the survival of the Fusarium spp. was investigated. The root symptoms and plant weight of seven crop species were evaluated after inoculation with each of the three Fusarium spp. The number of colony‐forming units of the Fusarium spp. from root tissues was also determined. Garlic was shown to be a symptomatic host for Foa, Fp and Fs; Fs was also pathogenic to onion. Root colonization of garlic, onion, maize, wheat, potato and sunflower suggested that they are reservoirs of Foa, Fp and Fs from asparagus and demonstrated the importance of crop rotation on the development of this asparagus disease.

Sixteen endophytic actinobacteria isolated from roots of native plants were evaluated for their antagonistic potential against soil-borne phytopathogenic fungi. Among them, three strong antagonistic isolates were selected and characterised for in vitro plant-growth-promoting and biocontrol traits, including production of hydrogen cyanide, indole-3-acetic acid and siderophores, chitinase and β-1,3-glucanase activities, and inorganic phosphate solubilisation. In all trials, the strain Streptomyces sp. SNL2 revealed promising features. The selected actinobacteria were investigated for the biocontrol of Fusarium oxysporum f. sp. radicis lycopersici and for growth promotion of tomato (Solanum lycopersicum L. cv. Aïcha) seedlings in autoclaved and non-autoclaved soils. All seed-bacterisation treatments significantly reduced the root rot incidence compared to a positive control (with infested soil), and the isolate SNL2 exhibiting the highest protective activity. It reduced the disease incidence from 88.5% to 13.2%, whereas chemical seed treatment with Thiram® provided 14.6% disease incidence. Furthermore, isolate SNL2 resulted in significant increases in the dry weight, shoot and root length of seedlings. 16S rDNA sequence analysis showed that isolate SNL2 was related to Streptomyces asterosporus NRRL B-24328T (99.52% of similarity). Its interesting biocontrol potential and growth enhancement of tomato seedlings open up attractive uses of the strain SNL2 in crop improvement.

Bayoud, a vascular wilt of date palm caused by Fusarium oxysporum f. sp. albedinis (Foa), is the most devastating disease in palm groves of north Africa. Although Foa is able to induce resistance mechanisms in its host plant, no data are available on the early responses of the root cells. Fusaric acid (FA) is the main toxin found in culture filtrates from Foa aggressive strains. This phytotoxin induces modifications of membrane permeability or membrane potential in various cell types and could thus be involved in the early steps of signal exchange between the pathogen and the plant. We showed an early differential behaviour of the disease‐resistant and ‐susceptible cultivars from date palm when challenged by FA. This response could be due to a differential sensitivity of H+‐ATPases to FA.

The effects of Beauveria bassiana strains on coffee berry borers (CBB), which emerge from infested berries left on soil, and its impact on the infestation of coffee berries on tree branches were evaluated at two Experimental Stations (Naranjal-Caldas and Paraguaicito-Quindio) in the Colombian coffee zone. Using a completely randomized design with 10 repetitions, 50 coffee berries artificially infested with CBB were placed on the base of a coffee tree. Four treatments including B. bassiana strain Bb9205, a mixture of Cenicafé strains (Bb9001, Bb9024 and Bb9119), a commercial formulation of B. bassiana and a control (water) were sprayed with 1×109 conidia per tree. After 30 days, all fungal strains lowered the infestation levels of the coffee berries on the trees at both locations. The mixture of Cenicafé strains decreased the tree infestation between 50 and 30% at both locations. In the berries dissected from each treated tree, insect mortality was about 40% at both locations compared to 15% in the control. B. bassiana strains also decreased the insect population inside the newly infested berries on the trees by 55–75%. The mixture of Cenicafé strains was the most effective for decreasing insect populations. B. bassiana significantly decreased CBB populations that emerged from fallen, infested, coffee berries and reduced future insect generations.

In Egypt, sesame cultivation is subject to attack by wilt and root-rot diseases caused by Fusarium oxysporum f.sp. sesami (Zap) Cast. and Macrophomina phaseolina (Maubl) Ashby causing losses in quality and quantity of sesame seed yield. Bacillus subtilis and Trichoderma viride isolates which were isolated from sesame rhizosphere were the most effective to antagonise fungal pathogens, causing high reduction of hyphal fungal growth. Trichoderma viride was found to be mycoparasitic on Fusarium oxysporum f.sp. sesami and M. phaseolina causing morphological atternation of fungal cells and sclerotial formation. In general, Bacillus subtilis, T. viride, avirulent Fusarium oxysporum isolate and Glomus spp. (Amycorrhizae) significantly reduced wilt and root-rot incidence of sesame plants at artificially infested potted soil by each one or two pathogens. Data obtained indicate that Glomus spp significantly reduced wilt and disease severity development on sesame plants followed by T. viride. Meanwhile, avirulent Fusarium oxysporum isolate followed by Glomus spp. were effective against root-rot disease incidence caused by M. phaseolina. Glomus spp. followed by B. subtilis significantly reduced wilt and root-rot disease of sesame plants. All biotic agents significantly reduced F. oxysporum f.sp. sesami and M. phaseolina counts in sesame rhizosphere at the lowest level. Glomus spp. and the avirulent isolate of F. oxysporum eliminated M. phaseolina in sesame rhizosphere. Meanwhile T. viride was the best agent at reducing F. oxysporum at a lower level than other treatments. Application of VA mycorrhizae (Glomus spp.) in fields naturally infested by pathogens significantly reduced wilt and root-rot incidence and it significantly colonised sesame root systems and rhizospheres compared to untreated sesame transplantings.

Pathogenicity tests of twenty-six fungal isolates were tested on peanut plants (Giza 5 cv.) and the results revealed that, Fusarium oxysporum isolate (No. I) followed by F. solani (No. II) then F. moniliforme (No III) significantly caused highest incidence of root rot disease. Also, F. moniliforme (No III) followed by F. solani (No II) then F. oxysporum (No I) gave the highest incidence of pod rot disease. The effectiveness of vescular arbuscular-mycorrhiza (VAM) at different application rates on the incidence of root rot, pod rot diseases and plant growth parameters of peanut was studied. All soil treatments with each rate of VAM significantly reduced root and pod rot diseases compared with control (rate 0%). The best reduction in the severity of both diseases with VAM was found at the rate of 3%. Application of rhizobacterin, microbin and cerialin biofertilisers at the different concentrations decreased the severity of both root rot and pod rot severity diseases compared with non-treated seeds. The greatest reduction in both diseases was achieved at a concentration of 8/100?g seeds. The highest number of pods and fresh weight (g) was achieved in seed supplemented with each biofertiliser at concentration of 8/100?g seed.

Solarization of soil, (potting mix = coarse sand:Eucalyptus marginata fines = 1∶1) infested with 3 fungi pathogenic to gerbera (Phytophthora cryptogea, Fusarium oxysporum andRhizoctonia solani), for 3 to 4 weeks within transparent polyethylene bags controlled root rot of gerbera. Solarization for 2 weeks however, was less effective. All plants grown in the infested potting mix which had been kept in shade for 2, 3 or 4 weeks were severely attacked. Solarization of soil within plastic bags for 4 weeks also increased availability of nutrients such as NH4 +-N, PO4 and K+ in comparison to bagged soil kept in the shade for the same period.

The effect of the placement of inoculum of Fusarium oxysporum at two soil depths, and the sequences of inoculations with Meloidogyne arenaria and Fusarium oxysporum on root growth and development of root disease in Trifolium subterraneum L. (subterranean clover) were investigated. The timing of infection and the proximity of root tips of the host root system to infection by M. arenaria and F. oxysporum appeared to be the major determining factors of root growth and of disease development in plants exposed to the pathogens. Immediate contact of roots with F. oxysporum (where the fungus was placed at seed level of 10 mm depth) appeared to result in more severe effects on roots in the presence of the nematode than later infection by the fungus placed at 30 mm depth. The production of galls by the nematode and early infection by F. oxysporum at 10 mm depth resulted in a severe inhibition of root growth, particularly of the lateral roots. But no such growth inhibition was evident when F. oxysporum and M. arenaria were introduced together at the lower depth of 30 mm. The lowest density of M. arenaria inoculum was sufficient to cause severe root rot if F. oxysporum was present at the host seed level. With the fungus at 30 mm depth, however, the expression of root rot appeared to be influenced by the inoculum level of the nematode. In sequential inoculation with F. oxysporum or M. arenaria, the organism added 2 weeks later had little or no effect on root development. The first organism (M. arenaria or F. oxysporum) to infect the germinated seedlings was the main cause of root growth inhibition. The organism that came into contact with the roots 2 weeks later had little or no effect on the roots. Concurrent infection by F. oxysporum and M. arenaria resulted in less M. arenaria gall production on the tap root system than those added with the nematode alone or in advance of the fungus.

Seed sample of lentil collected from the Swabi district were treated with NaCl and KCl at 0.01, 0.1 and 1.0% (w/w). Seed-borne mycoflora was observed at 0, 20, 40, 60 and 80 day intervals. Seed treatment with both the salts was found to be effective against storage fungi; however, KCl was more efficient against storage mycoflora such as Aspergillus species when compared with NaCl. With the passage of time, the incidence of deep-seated fungi was observed in salt-treated seed samples while untreated seed sample showed heavy infestation by the species of Aspergillus. Seed viability also remained unaffected in storage, except in seeds heavily infested with seed-borne mycoflora. Aspergillus spp. were the main cause of seed rot. Surface sterilisation of seeds with 1% Na(OCl)2 reduced the fungal infestation of seeds. Among various concentrations of salts, 0.1% (w/w) of both salts were better in controlling seed-borne mycoflora.

Pseudomonas solanacearum was transmitted from contaminated seed to the cotyledons of capsicum (Capsicum frutescens) at 92, 73 and 60% r. h. and to the cotyledons of tomato (Lycopersicon esculentum) seedlings at 92% r. h. Subsequent epiphytic colonisation of the true leaves of capsicum occurred at 92 and 73% r. h. An increase in the population on capsicum cotyledons was detected at 92 and 73% r. h. but only at the higher r. h. was an increased population of P. solanacearum associated with the colonisation of the true leaves. Lesions developed on the true leaves of capsicum at 92% r. h. Transmission of P. solanacearum from capsicum seeds was affected by inoculum concentration, occurring at an infestation level of ∽ 103 propagules seed-1 but not at 50 propagules seed-1. Pseudomonas solanacearum was detected on the cotyledons of capsicum held at 98% r. h. after germination of seed in soil infested with 2 × 108 propagules g-1 soil; lesions were detected on cotyledons 11 days after planting and invasion occurred in 10 stems and one root of the 20 seedlings sampled. The movement of the pathogen from the soil to the seedlings was affected by the level of soil infestation. Pseudomonas solanacearum was detected in four pairs of cotyledons, two stems and one root of the 36 seedlings sampled from soil infested with 107 propagules g-1 soil but it was not isolated from seedlings sampled from soil infested with 105 or 4 × 103 propagules g-1 soil. Leaf and stem prints demonstrated the epiphytic nature of this organism on the cotyledons and stems of seedlings.

Incidence of root rot and foliar yellowing, rhizome rot, panicle wilt and stem rot diseases of small cardamom (Elettaria cardamomum Maton) are caused by Fusarium oxysporum Schlecht., and were surveyed in the high ranges of Idukki district, Kerala during 2010–2011. The diseases were noticed in different areas to varying degrees. Root rot was found to be most severe, followed by pseudostem rot, rhizome rot and panicle wilt. The Fusarium infections were prevalent throughout the year (January–December) and varied from 1.5 to 10.6%. Even though the pathogen was isolated from different plant parts, during pathogenicity studies, all the isolates could cross-infect other plant parts too. Twenty different isolates of F. oxysporum were obtained from diseased samples, and five morphologically distinct isolates were analysed with Randomly Amplified Polymorphic DNA (RAPD) markers to study the genetic variability, if any, among them. PCR amplification of total genomic DNA with random oligonucleotide primers generated unique banding patterns, depending upon primers and isolates. Nine oligunucleotide primers were selected for the RAPD assays, which resulted in 221 bands for the five isolates of F. oxysporum. The number of bands obtained was entered into an NTSYS, and the results showed moderate genetic variability among F. oxysporum isolates causing root rot, rhizome rot, panicle wilt and pseudostem rot, collected from different locations. The dendrogram of different isolates into groups resulted in one major cluster at 0.61 similarity index comprising of four isolates (CRT 3, CRR 3, CPW 2 and CSR 1) and one isolate (CRT 5) formed in a separate cluster. Among the five isolates of F. oxysporum, CRT 5 was entirely different from the other four isolates. The isolates also differ according to the geographical area, as revealed from the genetic variability observed in different root rot isolates (CRT 3 and CRT 5). It is inferred that despite moderate variability, F. oxysporum, infecting small cardamom in Idukki district of Kerala, consists of a single clonal lineage.

ABSTRACT

The effect of three rates of a commercial formulation of Beauveria bassiana Strain GHA was evaluated against the coffee berry borer (CBB) Hypothenemus hampei Ferrari (Coleoptera: Curculionidae: Scolytinae), at three commercial coffee farms located at different altitudes on the island of Hawaii. H. hampei infestation and natural prevalence of B. bassiana increased with the elevation. At 145 metres above sea level (Farm 1), beetle infestation was 3.9%; at 538?m (Farm 2), beetle infestation was 12.2%; and at 768?m (Farm 3) infestation was 22.3%. The prevalence of natural B. bassiana killing CBB was 5.5% on Farm 1, 3.3% on Farm 2 and 23.1% on Farm 3. Monthly applications of B. bassiana resulted in no significant differences in levels of CBB infestation among treatments. Similarly, rates of infested berries with visually detectable signs of B. bassiana were similar among the B. bassiana treatments, ranging from 0.44% to 4.24%, and those percentages were larger than the treatments without B. bassiana. The percentage of females killed by Beauveria ranged from 69% to 95%. Effect of dose of BotaniGard® ES was reduced when beetles were in C position compared to A and B positions. B. bassiana can be an important component of an integrated pest management program for CBB.

In naturally infested soil containingPythium ultimum, P. acanthicum andPhytophthora megasperma, onlyP. ultimum was associated with root rot and damped-off seedlings. Damping-off was promoted by low soil temperatures and by flooding. Seedling stands were markedly reduced when seed was pre-incubated in soil at 12°C but not at 25°C or 35°C. Dusting carrot seed with metalaxyl significantly increased seedling stands in the field at rates from 1.5–6 g kg−1 seed and in both flooded and unflooded, naturally infested soil at 3.15 g kg−1. In greenhouse experiments using artifically infested soil,P. ultimum andP. paroecandrum caused damping-off of carrot seedlings andRhizoctonia solani reduced root and shoot weights.R. solani caused damping-off in nutrient-enriched soil.P. acanthicum andP. megasperma were not pathogenic to seedlings, although both fungi colonized roots. Soil populations of allPythium spp., particularlyP. ultimum, increased during growth of seedlings and population growth ofP. megasperma was promoted by periodic flooding. Infestation of soil withP. acanthicum did not reduce damping-off of carrot seedlings byP. ultimum orP. paroecandrum, but significantly increased root and shoot weights and decreased root colonization byR. solani P. acanthicum has potential as a biocontrol agent againstR. solani.
