Separation of 2,3-butanediol from the fermentation broth is a difficult task that has become a bottleneck in industrial production. Aqueous two-phase systems composed of hydrophilic solvents and inorganic salts could be used to extract 2,3-butanediol from fermentation broth. The ethanol/ammonium sulfate system was investigated in detail, including phase diagram, effect of phase composition on partition, removal of cells and biomacromolecules from the broths and recycling of ammonium sulfate. The highest partition coefficient (7.10) and recovery of 2,3-butanediol (91.7%) were obtained by a system composed of 32% (w/w) ethanol and 16% (w/w) ammonium sulfate. The maximum selective coefficient of 2,3-butanediol to glucose was 30.74 in the experimental range. In addition, cells and proteins could be simultaneously removed from the fermentation broth. The removal ratio of cells and proteins reached 99.7% and 91.2%, respectively. The recovery of ammonium sulfate in the bottom phase reached 97.14% when two volumes of methanol were added to the salt-rich phase.  相似文献

The dehydration of fermentative 2,3-butanediol into methyl ethyl ketone   总被引:5,自引:0,他引:5  
A solid acid catalyst consisted of sulfonic groups covalently bound to an inorganic matrice was developed to dehydrate 2,3-butanediol into methyl ethyl ketone. Rate constant and apparent activation energy of the dehydration reaction were determined. The decay course of the catalyst was a two-stage curve. The catalyst was deactivated more rapidly in the first stage than in the second stage. The strategy of maintaining constant degree of dehydration was employed to lengthen the lifetime of catalyst. Treatment of the 2,3-butanediol containing fermentation broth with activated carbon greatly facilitated the subsequent dehydration reaction.  相似文献

Separation of 2,3-butanediol from the complex fermentation broths is a difficult task and becomes a bottleneck in industrial production. Aqueous two-phase systems composed of hydrophilic solvents and inorganic salts could be used to extract 2,3-butanediol from fermentation broths. Aqueous two-phase extraction of 2,3-butanediol from fermentation broths was studied by ethanol and dipotassium hydrogen phosphate system. The influences of phase composition on partition of 2,3-butanediol, removal of cells and biomacromolecules were investigated. The partition coefficient and recovery of 2,3-butanediol reached up to 28.34 and 98.13%, respectively, and the selective coefficient of 2,3-butanediol to glucose was 615.87 when the system was composed of 24% (w/w) ethanol and 25% (w/w) dipotassium hydrogen phosphate. Simultaneously, cells and proteins could be removed from the fermentation broths and the removal ratio reached 99.63 and 85.9%, respectively. This process is convenient and economic, furthermore, the operation is easy to scale-up, that is, this method provides a new possibility for the separation and refining of 2,3-butanediol.  相似文献

Acetoin (3-hydroxy-2-butanone), a very popular food spice is now used in many industries (pharmaceuticals, chemicals, paint, etc.). In this study, an acetoin high producing strain, numbered as JNA-310, was newly isolated and identified as Bacillus subtilis which is safe on food industry, based on its physiological, biological tests and 16S rDNA sequence analysis. When glucose was used as carbon source in fermentation, the fermentation characterizations of this strain were analyzed, and a new phenomenon of reverse transforming 2,3-butanediol which was synthesized from glucose in the fermentation broth to acetoin was detected. Before 96 h, glucose which was mainly transformed to 2,3-butanediol and acetoin was totally consumed, and the yield of the two products were 41.7 and 21.0 g/l respectively. Acetoin was only a by product in the fermentation broth at prophase of fermentation. At the end of fermentation, the yield of acetoin was greatly improved and the yield of 2,3-butanediol was declined and the yield of them were about 42.2 and 15.8 g/l, respectively. The results indicated that 2,3-butanediol was reversely transformed to acetoin.  相似文献

A novel aqueous two-phase system consisted of 2-propanol/ammonium sulfate was used for the extraction of 2,3-butanediol from fermentation broths. The maximum partition coefficient and recovery of 2,3-butanediol reached 9.9 and 93.7%, respectively, and more than 99% of the cells and about 85% of the soluble proteins were removed when 34% (w/w) 2-propanol and 20% (w/w) ammonium sulfate were used. The separated cells could be re-used as inocula for subsequent fermentations. The aqueous two-phase system described in this study may have potential application in the extraction of 2,3-butanediol produced by industrial fermentation processes.  相似文献

采用克雷伯氏菌(Klebsiella pneumoniae CICC 10011)发酵生产2,3-丁二醇,并对2,3-丁二醇的盐析分离工艺进行了考察。通过实验确定了以葡萄糖为底物微氧批式流加发酵的条件,发酵液中2,3-丁二醇和3-羟基丁酮的质量浓度分别为90.98g/L和12.40g/L,2,3-丁二醇的摩尔转化率为82.7%,生产强度达到2.1g/(L·h)。对发酵液中2,3-丁二醇的盐析分离研究表明,K2HPO4和K3PO4对2,3-丁二醇的盐析效果优于K2CO3。当发酵液浓缩70%后,加入质量分数为45%的K,HPO4,2,3-丁二醇的分配系数达到9.10,回收率为79.37%;上相中2,3-丁二醇的质量浓度达到420g/L;此时3-羟基丁酮的分配系数和回收率分别为11.9和83.48%。  相似文献

Summary Hemicellulose-rich fractions from several agricultural residues were converted to 2,3-butanediol by a combined enzymatic hydrolysis and fermentation process. Culture filtrates from Trichoderma harzianum E58 were used to hydrolyze the substrates while Klebsiella pneumoniae fermented the liberated sugars to 2,3-butanediol. Approximately 50–60% of a 5% (w/v) xylan preparation could be hydrolyzed and quantitatively converted to 2,3-butanediol using this procedure. Although enzymatic hydrolysis was optimal at pH 5.0 and 50° C, the combined hydrolysis and fermentation was most efficient at pH 6.5 and 30° C. Combined hydrolysis and fermentation resulted in butanediol levels that were 20–40% higher than could be obtained with a separate hydrolysis and fermentation process. The hemicellulose-rich water-soluble fractions obtained from a variety of steam-exploded agricultural residues could be readily used by the combined hydrolysis and fermentation approach resulting in butanediol yields of 0.4–0.5 g/g of reducing sugar utilized.  相似文献

Klebsiella pneumoniae CGMCC 1.6366 is a bacterium isolated for 1,3-propanediol or 2,3-butanediol production previously. K. pneumoniae ΔbudA, a 2,3-butanediol synthesis pathway truncated mutant with the gene deletion of budA which encodes alpha-acetolactate decarboxylase, was found to execrate an unknown chemical at a high titer when grown in the broth using glucose as carbon source. Later this chemical was identified to be 2-ketogluconic acid, which was formed through the glucose oxidation pathway in K. pneumoniae. It was found that 2-ketogluconic can also be produced by the wild strain. The fermentation studies showed that the production of this metabolite is strictly pH dependent, when the fermenting broth was maintained at pH 6–7, the main metabolite produced by K. pneumoniae CGMCC 1.6366 was 2,3-butanediol, or some organic acids in the budA mutated strain. However, if the cells were fermented at pH 4.7, 2-ketogluconic acid was formed, and the secretion of all other organic acids or 2,3-butanediol were limited. In the 5L bioreactors, a final level of 38.2 and 30.2 g/L 2-ketogluconic acid were accumulated by the wild type and the budA mutant K. pneumoniae, respectively, in 26 and 56 h; and the conversion ratios of glucose to 2-ketogluconic acid reached 0.86 and 0.91 mol/mol for the wild and the budA mutant, respectively.  相似文献

Inulin could be converted to bio-based chemicals by an inulinase producer without external inulinase, and the production of 2,3-butanediol was less than 50 g/L. In this work, a novel inulinase producer of Klebsiella pneumoniae H3 was isolated, and inulinase catalytic properties as well as 2,3-butanediol fermentation were investigated. The enzyme was an intracellular inulinase with an optimal pH of 6 ∼ 7 and a temperature of 30 °C. The use of inulin by H3 was dependent on the degree of polymerization (DP), and the average DP of inulin in fermentation broth increased from 2.82 to 8.08 in 24-h culture of batch fermentation. Acidic pretreatment was developed to increase inulin utilization by adjusting medium pH to 3.0 prior to sterilization. In batch fermentation with optimized medium and fermentation conditions, the concentration of target product (2,3-butanediol and acetoin) was 80.4 g/L with a productivity of 2.23 g/(L⋅h), and a yield of 0.426 g/g inulin.  相似文献

Chemical 2,3-butanediol is an important platform compound possessing diverse industrial applications. So far, it is mainly produced by using petrochemical feedstock which is associated with high cost and adverse environmental impacts. Hence, finding alternative routes (e.g., via fermentation using renewable carbon sources) to produce 2,3-butanediol are urgently needed. In this study, we report a wild-type Klebsiella sp. strain XRM21, which is capable of producing 2,3-butanediol from a wide variety of carbon sources including glucose, sucrose, xylose, and glycerol. Among them, fermentation of sucrose leads to the highest production of 2,3-butanediol. To maximize the production of 2,3-butanediol, fermentation conditions were first optimized for strain XMR21 by using response surface methodology (RSM) in batch reactors. Subsequently, a fed-batch fermentation strategy was designed based on the optimized parameters, where 91.2 g/L of 2,3-butanediol could be produced from substrate sucrose dosing in 100 g/L for three times. Moreover, random mutagenesis of stain XMR21 resulted in a highly productive mutant strain, capable of producing 119.4 and 22.5 g/L of 2,3-butanediol and ethanol under optimized fed-batch fermentation process within 65 h with a total productivity of 2.18 g/L/h, which is comparable to the reported highest 2,3-butanediol concentration produced by previous strains. This study provides a potential strategy to produce industrially important 2,3-butanediol from low-cost sucrose.  相似文献

End-product conversion, low product concentration and large volumes of fermentation broth, the requirements for large bioreactors, in addition to the high cost involved in generating the steam required to distil fermentation products from the broth largely contributed to the decline in fermentative products. These considerations have motivated the study of organic extractants as a means to remove the product during fermentation and minimize downstream recovery. The aim of this study is to assess the practical applicability of liquid–liquid extraction in 2,3-butanediol fermentations. Eighteen organic solvents were screened to determine their biocompatibility, and bioavailability for their effects on Klebsiella pneumoniae growth. Candidate solvents at first were screened in shake flasks for toxicity to K. pneumoniae. Cell density and substrate consumption were used as measures of cell toxicity. The possibility of employing oleyl alcohol as an extraction solvent to enhance end product in 2,3-butanediol fermentation was evaluated. Fermentation was carried out at an initial glucose concentration of 80 g/l. Oleyl alcohol did not inhibit the growth of the fermentative organism. 2,3-Butanediol production increased from 17.9 g/l (in conventional fermentation) to 23.01 g/l (in extractive fermentation). A rapid and sample procedure was developed to determine, by gas-liquid chromatography, the concentrations of C2---C4 alcohols, C2---C6 volatile fatty acids (VFA) and lactic acid or 2,3-butanediol in fermentation liquids. both lactic acid and 2,3-butanediol are oxidized to acetaldehyde by periodic acid and acetaldehyde was eluted before ethanol. A complete separation of the alcohols and acids was performed in <15 min on a column packed with 80/100 Chromosorb WAW, having GP 10% SP-1200/1% H3PO4 as the liquid phase. The method was suitable for the analysis of rumen fluid and fermentation products from microbial cultures. The detection limits for all compounds were <0.13 nmol · injection−1.  相似文献

Inhibition by secondary fermentation products may limit the ultimate productivity of new glucose to ethanol fermentation processes. New processes are under development whereby ethanol is selectively removed from the fermenting broth to eliminate ethanol inhibition effects. These processes can concentrate minor secondary products to the point where they become toxic to the yeast. Vacuum fermentation selectively concentrates nonvolatile products in the fermentation broth. Membrane fermentation systems may concentrate large molecules which are sterically blocked from membrane transport. Extractive fermentation systems, employing nonpolar solvents, may concentrate small organic acids. By-product production rates and inhibition levels in continuous fermentation with Saccharomyces cerevisiae have been determined for acetaldehyde, glycerol, formic, lactic, and acetic acids, 1-propanol, 2-methyl-1-butanol, and 2,3-butanediol to assess the potential effects of these by-products on new fermentation processes. 1,3-Propanediol (1,3-PD) can be used for the industrial synthesis of a variety of compounds, including polyesters, polyethers, and polyurethanes. 1,3-PD is generated from petrochemical and microbial sources. 1,3-Propanediol is a typical product of glycerol fermentation, while acetate, lactate, 2,3-butanediol, and ethanol also accumulate during the process. Substrate and product inhibition limit the final concentration of 1,3-propanediol in the fermentation broth. It is impossible to increase the yield of 1,3-propanediol by using the traditional whole-cell fermentation process. In this study, dhaD and dhaK, the genes for glycerol dehydrogenase and dihydroxyacetone kinase, respectively, were inactivated by homologous recombination in Klebsiella pneumoniae. The dhaD/dhaK double mutant (designated TC100), selected from 5,000 single or double cross homologous recombination mutants, was confirmed as a double cross by using polymerase chain reaction. Analysis of the cell-free supernatant with high-performance liquid chromatography revealed elimination of lactate and 2,3-butanediol, as well as ethanol accumulation in TC100, compared with the wild-type strain. Furthermore, 1,3-propanediol productivity was increased in the TC100 strain expressing glycerol dehydratase and 1,3-PDO dehydrogenase regulated by the arabinose PBAD promoter. The genetic engineering and medium formulation approaches used here should aid in the separation of 1,3-propanediol from lactate, 2,3-butanediol, and ethanol and lead to increased production of 1,3-propanediol in Klebsiella pneumoniae.  相似文献

The fermentation of glucose to 2,3-butanediol by Bacillus polymyxa was improved by increasing the amount of yeast extract in the culture medium. A level of 1.5% (w/v) resulted in optimal 2,3-butanediol production. A comparable fermentation could be achieved with 0.5% yeast extract if the phosphate level of the medium was increased from 0.0026 to 0.078 M and the medium was supplemented with 40 M iron and 1.7 M manganese.NRCC #23497  相似文献

Microalgal biomass was hydrolyzed using a solid acid catalyst with the aid of liquid acid. The use of solid acid as the main catalyst instead of liquid acid was to omit subsequent neutralization and/or desalination steps, which are commonly required in using the resulting hydrolysates for microbial fermentation. The hydrolysis of 10 g/L of lipid-extracted Chlorella vulgaris containing 12.2% carbohydrates using 7.6 g/L Amberlyst 36 and 0.0075 N nitric acid at 150°C resulted in 1.08 g/L of mono-sugars with a yield of 88.5%. For hydrolysis of higher concentrations of the biomass over 10 g/L, the amount of Amberlyst 36 needed to be increased in proportion to the biomass concentration to maintain similar levels of hydrolysis performance. Increasing the solid acid concentration protected the surface of the solid acid from being severely covered by cell debris during the reaction. A hydrolysate of lipid-extracted C. vulgaris 50 g/L was used, with no post-treatment of desalination, for the cultivation of Klebsiella oxytoca producing 2,3-butanediol. Cell growth in the hydrolysate was found to be almost the same as in the conventional medium with the same monosaccharide composition, confirming its fermentation compatibility. 选用实验室自行筛选的Klebsiella pneumoniae ECU-15,进行了玉米秸秆水解液发酵联产氢气和2,3-丁二醇的初步研究。结果表明:以葡萄糖为碳源时,两目标产物随培养条件的改变呈现相同的变化趋势,且最佳发酵温度为37℃,最佳pH为6.0,最佳初始糖浓度为30 g/L;不同比例葡萄糖/木糖为混合碳源时,均能实现氢气和2,3-丁二醇的联产过程,但随着木糖含量的增加,细胞产量、氢气产量和2,3-丁二醇的产量都有所下降,并且木糖的存在会降低葡萄糖的消耗速率;实验最后以玉米秸秆水解液和同比例模拟合成培养基为底物,初步探明了该菌株利用水解液发酵联产氢气和2,3-丁二醇的可行性,最终氢气产量为0.65 v/v,产氢得率为0.43 mol/mol sugar;2,3-丁二醇产量为5.05 g/L,得率为0.82 mol/mol sugar。  相似文献

The production of 1,3-propanediol, 2,3-butanediol and ethanol was studied, during cultivations of strain Klebsiella oxytoca FMCC-197 on biodiesel-derived glycerol based media. Different kinds of glycerol feedstocks and experimental conditions had an important impact upon the distribution of metabolic products; production of 1,3-propanediol was positively influenced by stable pH conditions and by the absence of N2 gas infusions throughout the fermentation. Thus, during batch bioreactor fermentations conducted at increasing glycerol concentrations, 1,3-propanediol at 41.3 g/L and yield ~47% (w/w) was achieved at initial glycerol concentration ~120 g/L. At even higher initial glycerol media (150 and 170 g/L), growth was not ceased, but 1,3-propanediol production declined. During fed-batch fermentation under optimal experimental conditions, 126 g/L of glycerol were converted into 50.1 g/L of 1,3-propanediol. In this experiment, also 25.2 g/L of ethanol (conversion yield ~20%, w/w) were formed. A batch-bioreactor culture was performed under non-sterilized conditions and the 1,3-propanediol production was almost equivalent to the sterilized process. Concerning 2,3-butanediol formation, the most detrimental parameter was the absence of N2 sparging and as a result, no 2,3-butanediol was produced. The presence of glucose as co-substrate seriously enhanced 2,3-butanediol production; when commercial glucose was employed as sole substrate, 32.1 g/L of 2,3-butanediol were formed.  相似文献

Lake Averno sediment was used to isolate the facultative anaerobic bacteria having the potential for H2 production. Twenty-five out of 35 isolates recovered from the sediment sample produced hydrogen under anaerobic conditions from glucose with yields ranging from 0.1 to 0.49 mol H2/mol glucose. Identification based on 16S rRNA gene sequence analysis revealed that most of them belong to the Firmicutes group, with a prevalence of the Paenibacillus polymyxa species. Seven distinct genomic fingerprints among the 11 P. polymyxa isolates were obtained using the random amplified polymorphic DNA (RAPD) technique. Glucose fermentation by P. polymyxa isolates was investigated. Glucose was totally consumed after 3 days of fermentation. The fermentation products were hydrogen (0.18–0.47 mol H2/mol glucose), ethanol (0.1–0.5 mol ethanol/mol glucose), and 2,3-butanediol (0.1 mol 2,3-butanediol/mol glucose). Lower amounts of acetic, butyric, formic, lactic, and propionic acids were detected. All metabolic data concerning P. polymyxa isolates were analyzed by cluster analysis to reveal similarities and/or differences with clustering based on RAPD profiles. Despite the high metabolic similarity among almost all P. paenibacillus isolates, results of cluster analyses of metabolic and genetic data do not match completely.  相似文献

2,3-Butanediol is an important compound that can be used in many areas, especially as a platform chemical and liquid fuel. But traditional 2,3-butanediol producing microorganisms, such as Klebsiella pneumonia and K. xoytoca, are pathogens and they can only ferment sugars at 37°C. Here, we reported a newly developed Bacillus licheniformis. A protoplast transformation system was developed and optimized for this organism. With this transformation method, a marker-less gene deletion protocol was successfully used to knock out the ldh gene of B. licheniformis BL1 and BL3. BL1 was isolated earlier from soil for lactate production and it was further evolved to BL3 for xylose utilization. Combined with pH and aeration control, ldh mutant BL5 and BL8 can efficiently ferment glucose and xylose to D-(-) 2,3-butanediol at 50°C, pH 5.0. For glucose and xylose, the specific 2,3-butanediol productivities are 29.4 and 26.1 mM/h, respectively. The yield is 0.73 mol/mol for BL8 in xylose and 0.9 mol/mol for BL5 and BL8 in glucose. The D-(-) 2,3-butanediol optical purity is more than 98%. As far as we know, this is the first reported high temperature butanediol producer to match the simultaneous saccharification and fermentation conditions. Therefore, it has potential to further lower butanediol producing cost with low cost lignocellulosic biomass in the near future.  相似文献
