Using a retrograde tracer technique with horseradish peroxidase, we have revealed some afferent projections to the locus coeruleus complex from the contralateral pontine tegmentum, raphe nuclei, substantia nigra, nucleus of the solitory tract, dorsal motor nucleus of the vagus and other regions of the ponto-bulbar reticular formation as well as from hypothalamic and preoptic areas. 相似文献

Summary The central projections of the frontal organ of Rana pipiens are more widespread, and more similar to those of the epiphysis, than previously realized. Fibers labeled with horseradish peroxidase were traced to the amygdala, the epiphysis, the pretectal region, and several nuclear areas of the mesencephalic and diencephalic central gray. Neurons with projections into the vestibular receptor apparatus (efferent vestibular neurons) were identified in different medullary regions by retrograde labeling with horseradish peroxidase and transport-specific fluorochromes in the guinea pig. Two groups of efferent vestibular neurons could be distinguished, located dorsally and ventrally to the facial nerve fiber pathway. The dorsal group of efferent vestbular neurons consisted of small cells located close to the genu and the root of the facial nerve and the subependymal granular layer of the 4th ventricle floor. The ventral group was primarily composed of medium-sized cells, usually with only slight tracer accumulation; these were scattered over an extensive area of the lateral tegmental field within nucleus reticularis lateralis parvocellularis. The question of whether the test cells belong to the system of true vestibular efferents and satellite cells is discussed in the light of findings on cell location, morphology, and pattern of tracer accumulation.A. Synopsis Horseradish peroxidase (HRP) has been used as a tracer to study movements of solutions injected retrogradely via the duct of submandibular glands in rabbits. 0.1 ml of solution was injected either manually or by a constant hydrostatic pressure, and the subsequent distribution of HRP in the gland and duct at different times after injection has been examined histochemically at light and electron microscopical levels.Shortly after the injections, strong interstitial staining for peroxidase resulted from passage between acinar cells. Some sites of cellular uptake were observed and staining occurred in some ductal cells even when the duct had been cut at the hilum to minimize pressure effects. It is not known whether this diffuse uptake represents a physiological or pathological phenomenon. Some interstitial activity still remained 24 hr after injection but had disappeared by 48 hr. Inflammatory cells first appeared in the gland about 4 hr after the injection and slowly increased up to about 24 hr after injection.The results indicate that the HRP reaches the interstices of the gland principally by penetration between acinar cells, and that the junctional complexes between striated duct cells appear to be more resistant to disruption by luminal pressures. 相似文献

We have investigated the effects of taxol on the axonal transport of horseradish peroxidase (HRP) in dorsal root ganglia (DRG) cells and their neuronal cytoskeleton. The former were analysed by microinjection of HRP into single DRG cells and the latter was studied by means of immunohistochemistry and cryo-electron microscopy. In cultured and untreated DRG cells, microinjected HRP was typically transported anterogradely several hundred micrometres along their neurites. Different exposure periods (1, 2 and 3 days) to taxol were analysed. The axonal transport of HRP in DRG cells was time-dependently impeded by taxol. After the drug had been washed out, a recovery of the axonal transport of HRP was observed and confirmed by quantitative analysis. Cryo-electron microscopy revealed an abnormal aggregation of axonal and cytoplasmic microtubules, associated with a decreased amount of cross-linking structures, in taxol-treated DRG cell cultures. After 3 days of taxol exposure, microtubule-associated proteins and Tau-protein were restricted to the cellular somata but the neurofilament network and tubulin-proteins seemed to be unaffected. Our results demonstrate, for the first time, an inhibition of anterograde axonal transport of HRP in single neurons by taxol. This effect is reversible and seems not to be caused by cellular damage, but is rather a consequence of an altered organisation of microtubules and/or microtubule-associated proteins. 相似文献

Summary A homolog of the Edinger-Westphal nucleus of other vertebrates is described in two species of serranid basses of the genusParalabrax, a group possessing a wide range of ocular accommodation but lacking a pupillary reflex to light. The nucleus was found by retrograde labeling from the ciliary ganglion and lies dorsolateral to the ipsilateral oculomotor nucleus. The nucleus consists of 60 to 100 neurons with an average soma diameter of about 20 m in animals weighing 70 to 150 g. Electrophysiological experiments support the identification. Microstimulation of the nucleus evokes contraction of the ipsilateral lens retractor muscle and slight constriction of the caudal ipsilateral iris. Multi- and single-unit recordings in the nucleus reveal spontaneous firing (about 30 spikes/s in single units), the rate of which decreases during visually-evoked lens retractor relaxations (accommodation to near stimuli). Recordings of muscle fiber activity in the lens retractor show essentially the same behavior, which suggests that the ciliary ganglion and neuromuscular junctions simply relay impulses with little if any synaptic integration. The existence of a discrete Edinger-Westphal nucleus devoted largely to accommodation makesParalabrax a good model system for the further tracing of central accommodation control pathways.Abbreviations

Detailed differential scanning calorimetry (DSC), steady-state tryptophan fluorescence and far-UV and visible CD studies, together with enzymatic assays, were carried out to monitor the thermal denaturation of horseradish peroxidase isoenzyme c (HRPc) at pH 3.0. The spectral parameters were complementary to the highly sensitive but integral method of DSC. Thus, changes in far-UV CD corresponded to changes in the overall secondary structure of the enzyme, while that in the Soret region, as well as changes in intrinsic tryptophan fluorescence emission, corresponded to changes in the tertiary structure of the enzyme. The results, supported by data about changes in enzymatic activity with temperature, show that thermally induced transitions for peroxidase are irreversible and strongly dependent upon the scan rate, suggesting that denaturation is under kinetic control. It is shown that the process of HRPc denaturation can be interpreted with sufficient accuracy in terms of the simple kinetic scheme N -->k D where k is a first-order kinetic constant that changes with temperature, as given by the Arrhenius equation; N is the native state, and D is the denatured state. On the basis of this model, the parameters of the Arrhenius equation were calculated. 相似文献

Experiments using the retrograde transport of horseradish peroxidase were performed in order to identify the cells of origin the ascending projections from different brainstem regions to the area hypothalamica dorsalis (aHd) in the cat. The afferent inputs to this area originate mainly from the midbrain and medulla oblongata regions. The main afferent source of the area hypothalamica dorsalis arises from the substantia grisea centralis, where a large number of labeled cells were observed bilaterally, although more abundant on the ipsilateral side. Substantial afferents reach the aHd from the nuclei vestibularis medialis and inferior and the formatio reticularis mesencephali. A modest number of peroxidase-labeled neurons were observed in the nuclei ruber, interpeduncularis, substantia nigra, reticularis gigantocellularis, vestibularis lateralis, cuneatus and gracilis. From the pons, the nucleus raphe magnus sends a weak projection to the aHd. These anatomical data suggest that such area could be involved in visceral, sexual, nociceptive somatosensorial, sleep-waking and motor mechanisms. 相似文献

The projections of four anatomically distinct groups of putative neurosecretory cells found within the supra-oesophageal ganglion of the leech Macrobdella decora were studied by intracellular injection of horseradish peroxidase. All four groups have their own characteristic branching pattern while sharing the common feature of possessing primary branches that project into the dorsal commissure. Numerous secondary processes extend from these primary branches to terminate within the neural lamella, as well as within the neuropile. Electron microscopy of the regions into which these secondary processes project reveals numerous neurosecretory terminals. The data suggests that the midregion of the dorsal commissure constitues a neurohemal complex. These observations strengthen the argument that the four groups of identified cells are indeed neurosecretory. 相似文献

Summary Subsequent to the injection of horseradish peroxidase into the parietal eye of adult Lacerta sicula, the course of the parietal nerve and its projections were determined.The parietal nerve enters the left habenular ganglion where it branches into a medial and a lateral route. Some nerve fibers decussate within the habenular commissure. Whereas this pathway exhibits a striking asymmetry at the level of the habenular ganglia, its projections to the dorsolateral nucleus of the thalamus, the periventricular hypothalamic area, the preoptic hypothalamic and telencephalic regions, and the pretectal area are arranged in a strictly symmetric manner. A possible innervation of tegmental areas could not be proven due to the presence of endogenous peroxidase within these regions. No parietal nerve fibers were observed in the optic tectum.In a few animals investigated, scattered labeled perikarya were located in the periventricular hypothalamic gray indicating a parietopetal innervation in Lacerta sicula.
The injection of horseradish peroxidase into one of the lateral eyes revealed terminal areas of the optic nerve within the preoptic region, and the thalamic and pretectal nuclei, displaying partial overlapping with the projections of the parietal nerve to these areas.From the present investigation further evidence is obtained that the pineal complex of lower vertebrates is a component of the photoneuroendocrine system. The present study compared the relative effectiveness of wheatgerm agglutinin--horseradish peroxidase (WGA--HRP) and olfactory marker protein (OMP) in detecting the presence of intact olfactory axons in glomeruli of the main olfactory bulb (MOB) in the rat. The olfactory epithelium was damaged by i.p. injections of the toxin 3-methyl indole and, after 5 or 6 days, the olfactory sac was injected with a 1% WGA--HRP solution. No anterograde labeling was observed in the dorsal and ventromedial quadrants of the MOB in the WGA--HRP material. However, in the same cases OMP immunostaining was observed throughout the MOB. In other rats the rostral olfactory epithelium was aspirated unilaterally and after 3, 11 and 16 days the olfactory sacs were injected with WGA--HRP and rats were perfused 1 day later. In these cases WGA--HRP reaction product was absent in the dorsolateral quadrant of the MOB on the aspirated side in all animals, but OMP immunostaining could be detected in the 4 and 12 day survival animals but not in the 17 day survival rat. These findings indicate that anterograde transport of WGA--HRP accurately reflects the presence of intact axons en route to the MOB. In contrast, OMP immunostaining persists in axon terminals severed from their parent cell body for at least 12 days and is a less useful marker of intact olfactory axons in experiments using short survival times. 相似文献

Synopsis Horseradish peroxidase (HRP) administered close-arterially, has been found to enter rabbit submandibular saliva elicited by parasympathetic nerve stimulation. Adrenalin, superimposed on parasympathetic nerve stimulation, increased the passage of HRP into the saliva. Use of - and -adrenoceptor agonists, either separately or together, and use of - or -adrenoceptor antagonists together with adrenalin indicate that both - and -receptor stimulation is necessary for this increase in glandular permeability to occur. Histochemical assessment showed that HRP had permeated the interstitial spaces of the gland and entered the spaces between adjacent parenchymal cells. However, in unstimulated glands it had only reached the lumina of striated ducts, but after adrenalin administration, peroxidase was also observed within acinar lumina. This work indicates that the predominant pathway taken by the HRP was via intercellular spaces and it is suggested that the permeability between junctional complexes of parenchymal cells is capable of being modifiedin vivo. 相似文献

The organization of the projection from the pretectal region to the inferior olive in the cat was studied with autoradiographic and horseradish peroxidase (HRP) methods. After injections of HRP into the olive in six cats, cells were labeled ipsilaterally in the anterior pretectal nucleus (NPA), the posterior pretectal nucleus (NPP), the nucleus of the optic tract (NOT), and the dorsal terminal nucleus of the accessory optic tract (DTN). In three experiments, tritiated amino acids were injected into those parts of the pretectal region which contained labeled cells in the HRP experiments, and the projections to the olive were plotted. Both NPA and NPP projected to the rostral half of the dorsal accessory olive, the rostromedial margin of the ventral lamella, and the lateral part of the ventrolateral outgrowth. NOT projected to the caudal half of the dorsal cap, while DTN projected to both the dorsal cap and nucleus beta. The projections are entirely ipsilateral. 相似文献
