In Cayuga Lake water amended with 30 g of glucose or amino acids per ml, an added strain of Pseudomonas fluorescens and indigenous bacteria grew extensively, Pseudomonas sp. B4 and two rhizobia multiplied at a moderate extent, and introduced Escherichia coli and Klebsiella pneumoniae multiplied but to only a slight degree. The amendments did not enhance growth of Micrococcus flavus and Arthrobacter citreus, and an asporogenous strain of Bacillus subtilis decreased in numbers. The pseudomonads, rhizobia, E. coli and K. pneumoniae multiplied extensively when inoculated into sterile lake water amended with amino acids, A. citreus grew to a slight extent, but the numbers of M. flavus and B. subtilis did not change appreciably. In nonsterile lake water amended with 30 g of Trypticase soy broth per ml, the indigenous bacteria greatly increased in abundance, the pseudomonads, rhizobia, and E. coli developed to a lesser extent, the numbers of K. pneumoniae, A. citreus and M. flavus showed little increase, and B. subtilis decreased in numbers. Tests in pure culture containing 2 to 64 g of Trypticase soy broth per ml demonstrated good growth of P. fluorescens, Pseudomonas sp. B4, and the rhizobia at all concentrations; an initial decline followed by growth of E. coli, K. pneumoniae and R. leguminosarum biovar phaseoli at low concentrations; little or no growth or decline at the low levels but multiplication at the high levels by A. citreus and B. subtilis; and decline of M. flavus. It is proposed that the apparent Ks value, max value, length of lag phase and resistance to stress can be used to predict behavior of bacteria in lake water receiving low levels of organic nutrients.  相似文献

Changes in populations of microorganisms around germinating bean (Phaseolus vulgaris L.) seeds, in the rhizosphere of bean, and in a model rhizosphere were studied. Strains of Rhizobium phaseoli that were resistant to streptomycin and thiram were used, and as few as 300 R. phaseoli cells per g of soil could be enumerated with a selective medium that was devised. A direct role was not evident for bacterial competitors, lytic bacteria, antibiotic-producing microorganisms, bacteriophages, and Bdellovibrio in the suppression of R. phaseoli around germinating seeds and in the rhizosphere. Protozoa increased in numbers in the soil upon planting of the seeds. The extent of colonization of soil by R. phaseoli was inversely related to the presence of large numbers of bacteria and protozoa. Colonization of R. phaseoli was improved upon suppression of protozoa with thiram and also when the soil was amended with other protozoan inhibitors and mannitol to simulate seed and root exudation. The data support the view that the decrease in numbers of R. phaseoli is caused by an increase in protozoan predation, the protozoa increasing in number because they prey on bacteria that proliferate by using seed and root exudates as nutrients.  相似文献

Fluorescent Pseudomonas sp. strain 267 promotes growth of nodulated clover plants under gnotobiotic conditions. In the growth conditions (60 M FeCl3), the production of siderophores of the pseudobactin-pyoverdin group was repressed. Plant growth enhancement results from secretion of B vitamins by Pseudomonas sp. strain 267. This was proven by stimulation of clover growth by naturally auxotrophic strains of Rhizobium leguminosarum bv. trifolii and marker strains E. coli thi- and R. meliloti pan- in the presence of the supernatant of Pseudomonas sp. strain 267. The addition of vitamins to the plant medium increased symbiotic nitrogen fixation by the clover plants.  相似文献

Alfalfa (Medicago sativa L.) was grown in hydroponic culture to investigate adaptation to Fe-deficiency. Root exudates released into the nutrient solution from Fe-deficient plants were trapped and condensed on an amberlite XAD-4 resin column. The diethyl ether fraction of these exudates dissolved ferric phosphate remarkably. The dissolving capability was about 62 times higher than that of root exudates obtained from Fe-sufficient plants in complete nutrient solution. The Fe-dissolving compound was separated and identified. It was a new natural compound with molecular formula C14H10O5 and was identified as 2-(3,5-dihydroxyphenyl)-5,6-dihydroxybenzofuran by means of mass spectrometry and 1H-nuclear magnetic resonance. This new compound worked as a phytoalexin and inhibited completely the fungal growth of Fusarium oxysporum f. sp. phaseoli.  相似文献

The extent of decline in the population density of Rhizobium sp. exposed to Bdellovibrio was markedly reduced in the presence of montmorillonite, kaolinite or vermiculite but not by a soil clay fraction. Increasing levels of montmorillonite reduced the numbers of vibrios that appeared in a two-membered culture and allowed for greater survival of the rhizobia. Bdellovibrio and not Rhizobium sp. was retained when mixed with the three clay minerals, but no appreciable retention was evident with the soil clay fraction. Suspensions of colloidal soil organic matter protected the hosts from parasitism, although aqueous extracts of soil did not affect the relationship. Cells from old Rhizobium sp. cultures were attacked only after a lag phase, but rhizobia that had been stored were more rapidly lysed than cells tested immediately after removal from the growth medium. The possible significance of these findings to the survival of rhizobia in soils containing Bdellovibrio is discussed.  相似文献

Twelve strains of algicidal bacteria were isolated from the surfacewater of the pond Shinobazu and the moat Ote-bori (Tokyo, Japan). Nine of thesestrains were considered to be in the Pseudomonas group byanalyses of 16S rDNA sequences. The methanol extract ofPseudomonas sp. K44-1 showed marked antialgal activity bythe paper disk method. Harmane (1-methyl--carboline) was isolated fromtheethyl acetate extract of the whole culture broth ofPseudomonas sp. K44-1 by using silica gel columnchromatography and high performance liquid chromatography (HPLC). Harmaneshowedantagonistic activities against several cyanobacterial strains at aconcentration of 30 g disk–1.  相似文献

The number and weight of pods and the weight and nitrogen content of the tops of beans (Phaseolus vulgaris) derived from seeds inoculated with a thiram-resistant strain of Rhizobium phaseoli were increased if the seeds were treated with thiram before sowing in soil. A greater percentage of the nodules on 21-day-old plants were derived from the resistant strain, more nodules were formed, and these nodules were more effective in the presence of the fungicide than in its absence. These differences in nodule numbers were no longer present in 56-day-old plants, and only a small percentage of the nodules contained the resistant strain. The abundance of the fungicide-tolerant R. phaseoli increased rapidly soon after planting the seed and subsequently fell markedly, but the rate of decline was less if the seeds had been treated with the chemical. Protozoa also proliferated if thiram had not been applied to the seed, but their numbers were deleteriously influenced by thiram. Bdellovibrio, bacteriophages, and lytic micro-organisms acting on R. phaseoli were rare under these conditions. Ciliates and flagellated protozoa were initially suppressed by planting thiram-coated bean seeds in nonsterile soil, but the former were inhibited longer than the latter and the ciliate numbers never fully recovered if the seeds were treated with the fungicide. The resistant strain grew well in sterile soil also inoculated with a protozoa-free mixture of soil microorganisms whether thiram was added or not, but after an initial rise in numbers, its abundance fell if the mixture contained protozoa; the rate of this fall was delayed by the fungicide. The numbers of R. phaseoli were consistently less in sterile soil inoculated with the rhizobium plus a mixture of soil microorganisms containing ciliates and other protozoa than if the inoculum contained other protozoa but no ciliates. These results suggest that a suppression of protozoa, and possibly especially the ciliates, accounts for the enhanced growth of beans and the greater initial frequency of nodules formed by the thiram-resistant R. phaseoli in the presence of this fungicide. Thiram applied to uninoculated seed enhanced bean growth if thiram-resistant R. phaseoli were present in soil.  相似文献

The population of a strain ofEscherichia coli that was resistant to nalidixic acid and streptomycin declined rapidly in samples of sterile and nonsterile Cayuga Lake water and reached an undetectable level in nonsterile water at 24 and 72 hours when counted on eosin-methylene blue (EMB) agar and half-strength trypticase soy agar (TSA), respectively. In sterile lake water amended with 10g amino acids per ml or 0.1 M phosphate,E. coli multiplied exponentially for more than 24 hours. The addition ofRhizobium leguminosarum biovarphaseoli to unamended sterile lake water prevented the decline ofE. coli, and its addition to amended sterile lake water preventedE. coli multiplication. The cell density of this strain ofE. coli declined in the first 8 hours after its introduction into an inorganic salts solution, but the bacterium then grew extensively. This increase in abundance was not observed in the presence ofR. phaseoli, andE. coli counts on half-strength TSA remained unchanged between 8 hours and 6 days. When counted on EMB agar, the abundance of the antibiotic-resistant strain ofE. coli and a strain not selected for resistance increased in solutions containing phosphate and amino acids but declined in the presence of high densities ofR. phaseoli. Many of the cells of the antibiotic-resistantE. coli strain failed to grow on antibiotic-amended EMB agar after introduction of the organism into nonsterile or sterile lake water or into an inorganic salts solution containingR. phaseoli, although colonies appeared on TSA. The data suggest thatE. coli cells grown on rich media suffer a shock when introduced into lake water because of low hypotonicity, the indigenous competing flora, or both. This shock is prevented by either phosphate buffer or by amino acids at low concentration. The shocked bacteria formed colonies on half-strength TSA. Depending on environmental conditions, the presence of a second organism either has no effect or results in an increase or decrease inE. coli numbers.  相似文献

Natural mycoflora associated with fumonisins were analyzed in 150 samples of freshly harvested corn from Central-Southern, Central-Western and Northern regions of the State of Paraná, Brazil and correlated to climatic conditions. The corn samples were frequently contaminated with Fusarium sp.(98.7 to 100%) and Penicillium sp. (93 to 100%), when compared to Aspergillus sp. (not detected to 27.7%). The highest contamination with potentially mycotoxigenic fungi occurred in corn harvested in the Central-Western region, where total mould and yeast counts ranged from 5.5 × 103 to 5.2 × 106 CFU/g, with 98.7% contaminated byFusarium sp. and 93% by Penicillium sp. In this region F. moniliforme (F. verticillioides) was the predominant Fusariumsp., and was isolated in 85.9% of the samples. Aspergillus sp. was isolated from 27.7% samples. FB1 was detected in 100% of the samples (mean of 2.39 g/g) and FB2 in 97.7% (mean of 1.09 g/g). Fumonisins were also detected in all samples from Northern region, with mean of 4.56 g/g (FB1) and 2.20 g/g (FB2).Considering 1.0 g/g as the threshold, 72% of the corn samples from the Central-West and 92% from the North were contaminated with concentrations above this value, in contrast to a 18.5% contamination rate from Central-Southern samples. Between corn planting to harvesting season, the average maximum temperature and relative humidity were 26 °C and 77.1%(Central-Southern), 27 °C and 69% (Northern)and 29.9 °C and 89.1% (Central-Western).Therefore, the higher fumonisins contamination of corn from Northern region when compared to the Central-South were due to the differences in rainfall levels (92.8 mm in Central-Southern, 202 mm in Northern) during the month preceding harvest.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Oily sludge degradation by bacteria from Ankleshwar, India   总被引:7,自引:0,他引:7  
Three bacterial strains, Bacillus sp. SV9, Acinetobacter sp. SV4 and Pseudomonas sp., SV17 from contaminated soil in Ankleshwar, India were tested for their ability to degrade the complex mixture of petroleum hydrocarbons (such as alkanes, aromatics, resins and asphaltenes), sediments, heavy metals and water known as oily sludge. Gravimetric analysis showed that Bacillus sp. SV9 degraded approx. 59% of the oily sludge in 5 days at 30 °C whereas Acinetobacter sp. SV4 and Pseudomonas sp. SV17 degraded 37% and 35%. Capillary gas chromatographic analysis revealed that after 5 days the Bacillus strain was able to degrade oily sludge components of chain length C12–C30 and aromatics more effectively than the other two strains. Maximum drop in surface tension (from 70 to 28.4 mN/m) was accompanied by maximum biosurfactant production (6.7 g l−1) in Bacillus sp. SV9 after 72 h, these results collectively indicating that this bacterial strain has considerable potential for bioremediation of oily sludge.  相似文献

The susceptibility ofPhialophora gregata ff.sp.adzukicola andsojae to antimicrobial chemicals was investigated. The minimum inhibitory concentrations (MICs) of benomyl, chloramphenicol, CuSO4, cycloheximide and perchlorate for mycelial growth were the same for the two formae speciales. The MIC of hygromycin against f.sp.adzukicola was slightly lower than that against f.sp.sojae, and the latter was more resistant to iprodion than the former. Susceptibility to nystatin was markedly different: ff.sp.adzukicola andsojae had relative growth values of 3–20% and 59–93% at 100 µg/ml, respectively, and this difference could be used to differentiate the two formae speciales.  相似文献

Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

In the area of Jumla region in Western Nepal, measurements of saturated leaf net photosynthetic rate (Psat), nitrogen content, leaf fluorescence, carbon isotopic composition, and water status were performed on woody coniferous (Pinus wallichiana, Picea smithiana, Abies spectabilis, Juniperus wallichiana, Taxus baccata), evergreen (Quercus semecarpifolia, Rhododendron campanulatum), and deciduous broadleaved species (Betula utilis, Populus ciliata, Sorbus cuspidata) spreading from 2 400 m up to the treeline at 4 200 m a.s.l. With the exception of J. wallichiana, Psat values were lower in coniferous than broadleaved species. Q. semecarpifolia, that in this area grows above the coniferous belt between 3 000 and 4 000 m, showed the highest Psat at saturating irradiance and the highest leaf N content. This N content was higher and Psat lower than those of evergreen oak species of tempe forests at middle and low altitudes. For all species, Psat and N content were linearly correlated, but instantaneous nitrogen use efficiency was lower than values measured in lowland and temperate plant communities. The values of carbon isotopic composition, estimated by 13C, showed the same range reported for temperate tree species. The ranking of 13C values for the different tree types was conifers < evergreen broadleaved13C were found along the altitudinal gradient. Quantum yield of photochemistry at saturating irradiance, measured by leaf fluorescence (F/Fm), was highest in J. wallichiana and lowest in T. baccata. Overall, photochemical efficiency was more strongly related to species than to altitude. Interestingly, changes of .F/Fm along the altitudinal gradient correlated well with the reported altitudinal distribution of the species.This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

The structure of a cellulose-decomposing bacterial association was described using phenotypic and phylogenetic characteristics. Based on their morphological, physiological, and biochemical characteristics, the bacteria isolated from the association were identified as Sporocytophaga sp., Xanthomonas sp, and Pseudomonas sp. The phylogenetic analysis based on comparison of 16S rRNA gene fragments obtained from the association revealed six bacterial species belonging to the clusters of Alcaligenes sp., Ochrobactrum sp., Sphingomonas sp., Achromobacter sp., Pseudomonas sp., and Flexibacteriaceae (Sporocytophaga).  相似文献

Many microorganisms fail to degrade pollutants when introduced in different natural environments. This is a problem in selecting inocula for bioremediation of polluted sites. Thus, a study was conducted to determine the success of four inoculants to degradep-nitrophenol (PNP) in lake and industrial wastewater and the effects of organic compounds on the degradation of high and low concentrations of PNP in these environments.Corynebacterium strain Z4 when inoculated into the lake and wastewater samples containing 20 µg/ml of PNP degraded 90% of PNP in one day. Addition of 100 µg/ml of glucose as a second substrate did not enhance the degradation of PNP and the bacterium utilized the two substrates simultaneously. Glucose used at the same concentration (100 µg/ml), inhibited degradation of 20 µg of PNP in wastewater byPseudomonas strain MS. However, glucose increased the extent of degradation of PNP byPseudomonas strain GR. Phenol also enhanced the degradation of PNP in wastewater byPseudomonas strain GR, but had no effect on the degradation of PNP byCorynebacterium strain Z4.Addition of 100 µg/ml of glucose as a second substrate into the lake water samples containing low concentration of PNP (26 ng/ml) enhanced the degradation of PNP and the growth ofCorynebacterium strain Z4. In the presence of glucose, it grew from 2×104 to 4×104 cells/ml in 3 days and degraded 70% of PNP as compared to samples without glucose in which the bacterium declined in cell number from 2×104 to 8×103 cells/ml and degraded only 30% PNP. The results suggest that in inoculation to enhance biodegradation, depending on the inoculant, second organic substrate many play an important role in controlling the rate and extent of biodegradation of organic compounds.Abbreviations PNP p-nitrophenol  相似文献

为了探究从何种类型的自然生境中更易分离得到溶藻微生物,采用高氏1号培养基分别从水库底泥、湖泊底泥、农田土壤、林地土壤等四种来源共36份样品中分离了7 600株菌,并最终从中筛选得到了5株溶铜绿微囊藻(Microcystis aeruginosa)的溶藻菌,其中4株为假单胞菌(Pseudomonas sp.),1株为黄杆菌(Flavobacterium sp.),5株菌溶藻效率的变化范围为62%~95%。结果表明,当采用高氏1号培养基作为分离培养基时,湖泊底泥和水库底泥中的成功筛选概率最高,农田土壤次之,而林地土壤中则难以筛选得到,假单胞菌是较容易筛选得到的溶藻菌。  相似文献

Eucalyptus camaldulensis Dehnh. seedlings inoculated with Pisolithus tinctorius (Pers.) Coker & Couch and Thelephora terrestris Ehrl. per Fr. were grown in well watered soil (s –0.03 MPa) or subjected to a long-term soil water stress of up to –1.0 MPa over 13-week period in a glasshouse. After 13 weeks, all seedling containers were watered to field capacity and then water was withheld from the E. camaldulensis seedlings to induce a short-term drought. Diurnal measurements of seedling photosynthesis rate (A), leaf stomatal conductance (g) and leaf water potential (p) were completed before, during, and after the short term drought. Although they were growing in an equal soil volume, photosynthesis rate (A), leaf stomatal conductance and leaf water potential (p) of larger seedlings with P. tinctorius ectomycorrhizae were similar to those of smaller seedlings colonized with T. terrestris during the short-term drought period. Seedlings inoculated with Pisolithus tinctorius maintained higher photosynthesis rates over the course of the short-term drought. Thus, P. tinctorius ectomycorrhizae appear to be more efficient than those of T. terrestris in assisting seedlings to maintain gas exchange and photosynthesis under limited soil moisture conditions.  相似文献

Summary Activated sludge from a sewage treatment plant in Kanpur, India, was screened for bacterial strains metabolizing p-cresol exclusively under aerobic conditions. One such isolate was identified to be belonging to the genus Pseudomonas based on morphological and physiological criteria as well as 16S ribosomal RNA gene sequence analysis. Two intermediates were identified from the culture medium during the growth phase of Pseudomonas sp. strain A that indicated that the strain degraded p-cresol via the protocatechuate (PCA) pathway. p-Cresol was rapidly converted into p-hydroxybenzaldehyde (PHB) during early growth phase, which was later utilized after p-cresol depletion. p-Hydroxybenzoate (PHBA) accumulation was observed during the later stages of exponential growth phase. Kinetic constants for the degradation of p-cresol were determined using Haldane’s model. High μmax and inhibitory constant (KI) values along with the observed accumulation of significant amounts of PHB in culture filtrates seem to indicate that the isolated Pseudomonas sp. strain A may be of potential use in biotransformations.  相似文献

We compared responses of maize, tomato, and bean plants to water stress. Maize reached a severe water deficit (leaf water potential –1.90 MPa) in a longer period of time as compared with tomato and bean plants. Maize stomatal conductance (g s) decreased at mild water deficit. g s of tomato and bean decreased gradually and did not reach values as low as in maize. The protein content was maintained in maize and decreased at low water potential (w); in tomato it fluctuated and also decreased at low w; in bean it gradually decreased. Ribulose-1,5-bisphosphate carboxylase/oxygenase activity remained high at mild and moderate stress in maize and tomato plants; in bean it remained high only at mild stress.  相似文献

O2 protection of nitrogenase in a cultured Frankia isolate from Alnus rubra (HFPArI3) was studied in vivo. Evidence for a passive gas diffusion barrier in the vesicles was obtained by kinetic analysis of in vivo O2 uptake and acetylene reduction rates in response to substrate concentration. O2 of NH 4 + -grown cells showed an apparent K m O2 of approximately 1M O2. In N2-fixing cultures a second K m O2 of about 215 M O2 was observed. Thus, respiration remained unsaturated by O2 at air-saturation levels. In vivo, the apparent K m for acetylene was more than 10-fold greater than reported in vitro values. These data were inter oreted as evidence for a gas diffusion barrier in the vesicles but not vegetative filaments of Frankia sp. HFPArI3.  相似文献
