In this study we examined hormonal responses of Black-legged Kittiwake (Rissatridactyla) chicks to experimental variations in energy content and nutritional quality (low or high lipid to protein ratio, LPR) of
their food. Starting at the age of 10 days, chicks were fed either high or low LPR fish at 30, 50, 70 and 100% of ad libitum
energy intake. After 20 days of treatment, chicks were exposed to a standardized acute handling and restraint stress protocol,
where a baseline sample was taken immediately after taking a chick from the nest, and three additional blood samples were
taken at intervals up to 50 min. Testosterone and corticosterone titres in plasma were measured via radioimmunoassay. We found
that baseline testosterone levels were not significantly affected by the experimental treatments. Food-restricted chicks had
elevated baseline and acute stress-induced levels of corticosterone compared to chicks fed ad libitum. An elevation of circulating
levels of corticosterone in energetically stressed individuals was further magnified by low nutritional quality of food. Baseline
and acute stress-induced corticosterone levels of chicks were negatively correlated with their fat reserves. We conclude that
the physiological condition of Black-legged Kittiwake chicks can be assessed reliably by measuring circulating levels of corticosterone.
We discuss short- and long-term effects of elevated corticosterone secretion in food-stressed nest-bound chicks.

The effects of the Met-enkephalin analog D-Ala2-Met-enkephalinamide (DALA) on basal and ACTH-stimulated corticosterone secretion from dispersed adrenal cells were investigated. Low doses (10?10 and 10?12 M) of DALA resulted in no apparent alteration in the response to ACTH (8×10?9, 3.2×10?8 or 1.6×10?7 M). High doses of DALA (10?8 and 10?6 M) produced a decline in the steroidogenic response to ACTH. The opiate receptor antagonist naloxone (10?4–10?10 M) did not influence the basal production of corticosterone or the stimulating action exerted by ACTH. However, the presence of naloxone reversed the blocking action on corticosterone production that was exerted by DALA. These findings indicate that enkephalins may decrease adrenocortical responsiveness to ACTH. 相似文献

Recent studies have shown a link between obesity and endoplasmic reticulum (ER) stress. Perturbations in ER homeostasis cause ER stress and activation of the unfolded protein response (UPR). Adipocyte differentiation contributes to weight gain, and we have shown that markers of ER stress/UPR activation, including GRP78, phospho-eIF2α, and spliced XBP1, are upregulated during adipogenesis. Given these findings, the objective of this study was to determine whether attenuation of UPR activation by the chemical chaperone 4-phenylbutyrate (4-PBA) inhibits adipogenesis. Exposure of 3T3-L1 preadipocytes to 4-PBA in the presence of differentiation media decreased expression of ER stress markers. Concomitant with the suppression of UPR activation, 4-PBA resulted in attenuation of adipogenesis as measured by lipid accumulation and adiponectin secretion. Consistent with these in vitro findings, female C57BL/6 mice fed a high-fat diet supplemented with 4-PBA showed a significant reduction in weight gain and had reduced fat pad mass, as compared with the high-fat diet alone group. Furthermore, 4-PBA supplementation decreased GRP78 expression in the adipose tissue and lowered plasma triglyceride, glucose, leptin, and adiponectin levels without altering food intake. Taken together, these results suggest that UPR activation contributes to adipogenesis and that blocking its activation with 4-PBA prevents adipocyte differentiation and weight gain in mice. 相似文献

在介绍性教科书、保护计划甚至基础著作中 ,常常将北极区和高山生物群系等同看待。高山和北极区生物群系无树的广阔区域 ,草本和非禾本科草本植物的初级生产力总量低 ,食物网单一。此外 ,除热带高山生境可能有例外 ,该两种环境都具有明显的季节性。但是 ,对于这些表面上相似的环境以前很少进行比较 ,而两者之间应具有明显的不同。我们假定 :如果环境相似 ,栖息在这些环境里的鸟类会对意外的应激刺激表现出相似的反应。相反 ,如果这些环境很不同 ,那么鸟类对剧烈应激的反应可进行调整以适应当地的不同环境 ,而不应一成不变。本文初步报道了青藏高原某些繁殖鸟类的肾上腺皮质反应 ,并与已发表的栖息于北极区苔原生态系统鸟类的数据进行了比较。白腰雪雀 (Onychostruthustaczanowskii)和棕颈雪雀 (Pyrgilaudaruficollis)被认为是高山代表物种 ,随着标准化捕捉 -处理 -抑制的应激刺激 ,它们表现出典型的血浆肾上腺皮质酮含量升高。但是 ,不同于许多北极区的代表物种 ,它们对应激的肾上腺皮质反应并无任何季节性变化。研究结果表示 :生活在高山和北极区环境下的鸟类物种可能具有激素反应以适应其生存环境所施加的生态挑战 [动物学报 49(1) :1~ 19,2 0 0 3]。 相似文献

The ongoing search for neurotrophic factors for motoneurons has led to the identification of a number of molecules which regulate
motoneuron survival and function. Among these factors, the neurotrophins brain derived neurotrophic factor (BDNF), neurotrophin-3
(NT-3) and NT-4/5 but not nerve growth factor (NGF), can prevent embryonic and postnatal motoneuron cell death in a variety
of experimental paradigms. Analysis of expression of p75, trkB and trkC—components of the neurotrophin receptors—supports
a potential physiological role for these factors as muscle- and glial-derived trophic factors for motoneurons. However, the
survival of motoneurons during embryonic development is not reduced in the absence of BDNF, NT-3 or NT-4, as revealed by gene
knockout experiments. This points to the involvement of additional trophic factors in the regulation of embryonic and postnatal
motoneuron survival. The purpose of this review is to bring together the often prophetic observations from earlier studies—prior
to the identification and characterization of these neurotrophins—with more recent results.

Oxanine (Oxa), generated from guanine (Gua) by NO- or HNO2-induced nitrosative oxidation, has been thought to cause mutagenic problems in cellular systems. In this study, the response of Oxa to different enzymatic functions was explored to understand how similarly it can participate in biomolecular reactions compared to the natural base, Gua. The phosphorylation efficiency of the T4 polynucleotide kinase was highest when Oxa was located on the 5′-end of single stranded DNAs compared to when other nucleobases were in this position. The order of phosphorylation efficiency was as follows; Oxa > Gua > adenine (Ade) ∼ thymine (Thy) > cytosine (Cyt). Base-pairing of Oxa and Cyt (Oxa:Cyt) between the ligation fragment and template was found to influence the ligation performance of the T4 DNA ligase to a lesser degree compared to Gua:Cyt. In addition, EcoRI and BglII showed higher cleavage activities on DNA substrates containing Oxa:Cyt than those containing Gua:Cyt, while BamHI, HindIII and EcoRV showed lower cleavage activity; however, this decrease in activity was relatively small. 相似文献

Summary The response of a pollutant-degrading bacterium P. putida CP1 to stresses was investigated. The growth on the mono-chlorophenols resulted in a decrease in dry weight of the organism,
although there was an increase in cell number. There was a change of bacterial shape from rod to round as well as the reduction
of cell size when grown on phenol and chlorophenols. Changes in cell shape and size were also evident in glucose-free medium,
which suggested that alteration of cell shape from rod to round as well as reduction of cell size were due to nutritional
stress. The increase in cell number but a drop in dry weight correlated with the reduction of cell size and shape. The organism
flocculated with chlorophenols but not with phenol. The cause of flocculation was due to the toxicity of chlorophenol. Isomerization
of cis to trans forms of the unsaturated fatty acids in P. putida CP1 occurred under conditions of environmental stress. Trace amounts of the polyunsaturated fatty acid linoleic acid (cis-9, cis-12-octadecadienoic acid) rarely found in bacterial membranes and oleic acid (cis-9-octadecanoic acid), which is a typical product of aerobic fatty acid synthesis, were found in P. putida CP1. 相似文献

To further examine the structural and thermodynamic basis of hydrophobic stabilization in proteins, all of the bulky non-polar residues that are buried or largely buried within the core of T4 lysozyme were substituted with alanine. In 25 cases, including eight reported previously, it was possible to determine the crystal structures of the variants. The structures of four variants with double substitutions were also determined. In the majority of cases the "large-to-small" substitutions lead to internal cavities. In other cases declivities or channels open to the surface were formed. In some cases the structural changes were minimal (mainchain shifts < or = 0.3 A); in other cases mainchain atoms moved up to 2 A. In the case of Ile 29 --> Ala the structure collapsed to such a degree that the volume of the putative cavity was zero. Crystallographic analysis suggests that the occupancy of the engineered cavities by solvent is usually low. The mutants Val 149 --> Ala (V149A) and Met 6 --> Ala (M6A), however, are exceptions and have, respectively, one and two well-ordered water molecules within the cavity. The Val 149 --> Ala substitution allows the solvent molecule to hydrogen bond to polar atoms that are occluded in the wild-type molecule. Similarly, the replacement of Met 6 with alanine allows the two solvent molecules to hydrogen bond to each other and to polar atoms on the protein. Except for Val 149 --> Ala the loss of stability of all the cavity mutants can be rationalized as a combination of two terms. The first is a constant for a given class of substitution (e.g., -2.1 kcal/mol for all Leu --> Ala substitutions) and can be considered as the difference between the free energy of transfer of leucine and alanine from solvent to the core of the protein. The second term can be considered as the energy cost of forming the cavity and is consistent with a numerical value of 22 cal mol(-1) A(-3). Physically, this term is due to the loss of van der Waal''s interactions between the bulky sidechain that is removed and the atoms that form the wall of the cavity. The overall results are consistent with the prior rationalization of Leu --> Ala mutants in T4 lysozyme by Eriksson et al. (Eriksson et al., 1992, Science 255:178-183). 相似文献

The hypothalamic-pituitary-adrenal (HPA) axis becomes less responsive to some types of repeated stress over time, a process termed habituation. Many facets of the stressful stimulus can modify such HPA responses to stressors, such as predictability and controllability. However, the physical context in which the stressor occurred may also provide a discriminative stimulus that can affect the HPA response to that stressor. In the present study, we examined whether a change in the context in which stress exposure occurs can alter HPA responses to a subsequent [corrected] homotypic stressor. Three separate contexts were produced by manipulating odor cues. Rats housed in the 3 context rooms exhibited similar HPA responses to acute 30-min restraint or repeated (8th) 30-min restraint in their home environments. However, rats that were restrained for 30 min per day for 7 days in a room in one context and then restrained on day 8 in a novel context exhibited attenuated habituation compared to rats restrained on day 8 in the familiar context. These results provide evidence that repeated stress-induced HPA activity depends, in part, on the context in which the stress is experienced. 相似文献

Glutathione (L-γ-Glutamyl-L-Cysteinylglycine) appears as the major nonprotein thiol compound in yeasts. Recent advances have shown that glutathione (GSH) seems to be involved in the response of yeasts to different nutritional and oxidative stresses. When the yeast Saccharomyces cerevisiae is starved for sulfur or nitrogen nutrients, GSH may be mobilized to ensure cellular maintenance. Glutathione S-transferases may be involved in the detoxification of electrophilic xenobiotics. Vacuolar transport of metal derivatives of GSH ensure resistance to metal stress. Growth of methylotrophic yeasts on methanol results in the formation of an excess formaldehyde that is detoxified by a GSH-dependent formaldehyde dehydrogenase. Growth of yeasts on glycerol results in the accumulation of methylglyoxal detoxified by the glyoxalase pathway. Glutathione per se can react with oxidative agents or is involved in the oxidative stress response through glutathione peroxidase. 相似文献

Short-term and long-term effects of plant water deficits on stomatal response to humidity in Corylus avellana L. 总被引:2,自引:0,他引:2
Short-term (hours) changes in plant water status were induced in hazel (Corylus avellana L.) by changing the evaporative demand on a major portion of the shoot while maintaining a branch in a constant environment. Stomatal conductance of leaves on the branch was influenced little by these short-term changes in water status even with changes in leaf water potential as great as 8 bars. Long-term (days) changes in plant water status were imposed by soil drying cycles. Stomatal conductance progessively decreased with increases in long-term water stress. Stomata still responded to humidity with long-term water stress but the range of the conductance response decreased. Threshold responses of stomata to leaf water potential were not observed with either short-term or long-term changes in plant water status even when leaves wilted. It is suggested that concurrent measurements of plant water status may not be sufficient for explaining stomatal and other plant responses to drought. 相似文献

Oxidative stress and low grade chronic inflammation are increased in accumulating fat. Our objective was to test whether 4-hydroxynonenal (4-HNE), an end-product of lipid peroxidation, affects cyclooxygenases in 3T3-L1 adipose cells. 4-HNE increased COX-2 mRNA and protein expression and p38MAP-kinase phosphorylation in a dose-dependent manner. Pretreatment of 3T3-L1 cells by a selective inhibitor of p38MAPK (PD 169316) abolished 4-HNE and glucose oxidase induced COX-2 expression. Our results show that oxidative stress induces COX-2 expression through the production of 4-HNE which activates p38MAPKinase, suggesting that 4-HNE links oxidative stress and chronic inflammation through the activation of cyclooxygenase. 相似文献

Activation and proliferation of T cells are tightly regulated during the immune response. We show here that kinetics of proliferation of PHA activated T cells follows the expression of cMyc. Expression of p53 is also elevated and remains high several days after activation. To investigate the role of p53 in activated T cells, its expression was further elevated with nultin-3 treatment, a small molecule that dissociates the E3 ubiquitin protein ligase MDM2 from p53. Concomitantly, cMyc expression and proliferation decreased. At the other end of the cMyc-p53 axis, inhibition of cMyc with 10058-F4 led to down regulation of p53, likely through the lower level of cMyc induced p14ARF, which is also known to dissociate the p53-MDM2 complex. Both compounds induced cell cycle arrest and apoptosis. We conclude that the feedback regulation between cMyc and p53 is important for the T cell homeostasis. We also show that the two compounds modulating p53 and cMyc levels inhibited proliferation without abolishing the cytotoxic function, thus demonstrating the dichotomy between proliferation and cytotoxicity in activated T cells. 相似文献

Quiescent 3T3 cells grown in media containing 4% foetal calf serum showed different responses to insulin and to serum repletion (to 12%). Insulin stimulated protein synthesis within 1 h and this early response was insensitive to actinomycin D. The later insulin response showed progressive sensitivity to actinomycin D. The serum response was slower, not occurring until 1 h, and was inhibited by actinomycin D. Depletion of cell protein kinase C by pre-treatment with phorbol ester caused a total block of the immediate response to insulin but had little effect on the response to serum or the later response to insulin. Acute phorbol ester treatment stimulated protein synthesis. 相似文献

The winter‐flowering succulent Aloe marlothii provides nectar for many opportunistic avian nectarivores in southern African savannas. We assessed the importance of A. marlothii nectar sugar for opportunistic nectarivores by analysing temporal changes in stable carbon isotope ratios (δ13C) in the tissues of birds in Suikerbosrand Nature Reserve, South Africa. The blood of the 11 most common non‐granivorous opportunistic nectarivores at our site was enriched in 13C by 3.4 ± 1.5‰ during the flowering period of A. marlothii, reflecting the enriched crassulacean acid metabolism (CAM) isotopic signature of nectar (?12.6 ± 0.5‰). This relatively small contribution of A. marlothii nectar to assimilated carbon in whole blood contrasted with that of exhaled CO2 in African Red‐eyed Bulbuls Pycnonotus nigricans and Cape White‐eyes Zosterops capensis. In both these species, the δ13C of breath samples was significantly enriched compared with blood and feathers, and closely resembled that of the nectar, revealing combustion of ingested nectar rather than assimilation. Although our analysis was complicated by the presence of C4 grasses, whose δ13C values are similar to those of CAM photosynthesizers, when considered with previously published feeding observations our data reveal that opportunistic nectarivores feeding on A. marlothii nectar obtain a relatively small fraction of their assimilated carbon, but most of their metabolized carbon, from this seasonally available carbohydrate food resource. Because the δ13C values of insects associated with C3 plants also became enriched during the flowering season, some insect‐eating opportunistic nectarivores may have assimilated A. marlothii carbon indirectly from insects. This study highlights the importance of understanding isotopic routing when assessing the nutritional significance of specific dietary items to consumer communities. 相似文献

Ralstonia solanacearum gram-negative phytopathogenic bacterium exerts its virulence through a type III secretion system (T3SS) that translocates type III effectors (T3Es) directly into the host cells. T3E secretion is finely controlled at the posttranslational level by helper proteins, T3SS control proteins, and type III chaperones. The HpaP protein, one of the type III secretion substrate specificity switch (T3S4) proteins, was previously highlighted as a virulence factor on Arabidopsis thaliana Col-0 accession. In this study, we set up a genome-wide association analysis to explore the natural diversity of response to the hpaP mutant of two A. thaliana mapping populations: a worldwide collection and a local population. Quantitative genetic variation revealed different genetic architectures in both mapping populations, with a global delayed response to the hpaP mutant compared to the GMI1000 wild-type strain. We have identified several quantitative trait loci (QTLs) associated with the hpaP mutant inoculation. The genes underlying these QTLs are involved in different and specific biological processes, some of which were demonstrated important for R. solanacearum virulence. We focused our study on four candidate genes, RKL1, IRE3, RACK1B, and PEX3, identified using the worldwide collection, and validated three of them as susceptibility factors. Our findings demonstrate that the study of the natural diversity of plant response to a R. solanacearum mutant in a key regulator of virulence is an original and powerful strategy to identify genes directly or indirectly targeted by the pathogen. 相似文献
