S-Adenosylmethionine decarboxylase activity in leaves of Pharbitisnil increased dramatically at lights-on and then gradually decreasedin the light. The enzymatic activity fell dramatically at lights-offthen increased slightly in darkness. A new L-amlnolactam-hydrolyzing enzyme was partially purifiedfrom cells of Cryptococcus laurentii which can grow on L-aminolactamas a carbon and nitrogen source. Laurie acid (1 mg/ml) sharply suppressed the cell division ofan acrA mutant strain of Escherichia coli K12. However, thewild type acrA$ strain was resistant to the fatty acid. Capricacid and myristic acid were not so toxic. Laurie acid inhibitedboth DNA and protein synthesis of the acrA mutant strain, withthe former being more sensitive than the latter. On the otherhand, DNA polymerase activity of toluene-treated cells was stimulatedrather than inhibited by the presence of 1 mg/ml of lauric acid.Fatty acid composition of phospholipids in the inner membranewas largely altered by the addition of lauric acid. These resultssuggest that addition of lauric acid to the medium causes adisorganization of the membrane lipids in the acrA mutant celland activities of DNA polymerase and other intramembranous enzymesare consequently inhibited. 1Present address: Osaka City Institute of Public Health andEnvironmental Sciences. Germ-tube emergence took place at almost the same time as DNAsynthesis during spore germination in the fission yeast, Schizosaccharomycespombe. Neither the emergence nor the elongation of the germ-tubeswas inhibited by hydroxyurea. Mesophyll chloroplasts were isolated from leaves of a Na+-requiringNAD-malic enzyme type, dicotyledonous C4 plant, Amaranthus tricolorL. Using vacuolar perfusion, which enabled us to replace the cellsap with a solution containing no k+, Na+ and Cl, theconcentrations of these ions in the protoplasm of three speciesof fresh water Characeae were determined. They were respectively,78, 2 and 27 mM in Nitella flexilis, 101, 9 and 31 mM in Nitellapulchella, and 112, 3 and 21 mM in Chara australis. 86Rb uptake was examined in two species of unicellular greenalgae, Chlamydomonas nivalis isolated from snow, and a cellwall-less mutant of the temperate freshwater Chlamydomonas reinhardii.In C. reinhardii cells grown at 20°C and cooled rapidlyto 0°C, 86Rb uptake was abolished. Cells cooled rapidlyto –5°C in the absence of ice accumulated 86Rb veryrapidly but the time course of this uptake suggested non-selectiveaccumulation through a damaged plasmalemma. Cells grown at 8°Cwere viable, able to divide and motile; they showed no signsof cold-shock and 86Rb uptake, albeit slow, was measurable at–5°C in the absence of extracellular ice. Cells ofC. nivalis grown at 20°C were damaged at sub-zero temperaturesalthough they did show an enhanced 86Rb uptake at 0°C. Cellsgrown at 5°C were able to accumulate 86Rb from media undercooledto -5°C in the absence of extracellular ice, and again showedenhanced uptake at 0°C. The inside-out patch-clamp technique was applied to the plasmolyzedplasmalemma of inter-nodes of Chara corallina without enzymatictreatment. We found two different types of channel activitythat were CP-sensitive. Both types of channel were Ca2+-dependent.However, the one that exhibited greater dependence on Ca2+ ionswas the focus of our studies, and we named it the Ca2+-dependentCP-sensitive anion channel. When the concentration ofCa2+ ions on the cyto-plasmic side was 1.0 µM, the Ca2+-dependentCP-sensitive channel opened most frequently between approximately–80 and –100 mV. At 10 µM Ca2+, it openedless frequently, and at 0.1 µM Ca2+ it scarcely openedat all. These observations indicate that the anion channel ofinterest is voltage-dependent over a restricted range of concentrationsof Ca2+ ions. The dependence on Ca2+ and voltage of the channelcan explain the behavior of the excitable Ca2+-activated Clchannel in the Chara plasmalemma. Rates of CO2 and HCC3 fixation in cells of various Chlorellaspecies in suspension were compared from the amounts of 14Cfixed during the 5 s after the injection of a solution containingonly 14CO2 or H14CO3. Results indicated that irrespectiveof the CO2 concentration during growth, Chlorella vulgaris 11h and C. miniata mainly utilized CO2, whereas C. vulgaris C-3,C. sp. K. and C. ellipsoidea took up HCO3 in additionto CO2. Cells of C. pyrenoidosa that had been grown with 1.5%CO2 (high-CO2 cells) mainly utilized CO2, whereas those grownwith air (low-CO2 cells) utilized HCO3 in addition toCO2. Cells that utilized HCO3 had carbonic anhydrase(CA) on their surfaces. The effects of Diamox and CA on the rates of CO2 and HCO3fixation are in accord with the inference that HCO3 wasutilized after conversion to CO2 via the CA located on the cellsurface. CA was found in both the soluble and insoluble fractions;the CA on the cell surface was insoluble. Independent of the modes of utilization, the apparent Km (NaHCO3)for photosynthesis was much lower in low-CO2 cells than in high-CO2ones. Foliar application of NaCl to sodium-deficient Panicum coloratumstimulated photosynthesis, as did application via roots. Flowering in the short-day plant Lemna paucicostata 6746 canbe induced under continuous light by the addition of ferricyanie,ferrocyanide or KCN to M-sucrose medium. Each substance is nearly10 times more effective when the flasks are covered by glassbeakers than when cotton plugs are used. By contrast, when floweringis induced under continuous light by copper or by short-daytreatment, neither flowering nor growth are affected by whetherglass beakers or cotton plugs are used. Ferricyanide, ferrocyanideand KCN are also able to induce long-day flowering when theplants are grown on Msucrose medium in small beakers that areplaced in a covered storage dish that also contains a solutionof one of these compounds. Addition of a KOH trap to the storagedish completely blocks the flowering induced by these compounds.If [14C]ferrocyanide is added to the storage dish both the M-sucrosemedium and the plants contain significant amounts of radioactivity,the amount of radioactivity being proportional to the floweringresponse.   1. The sugars which induced gigantism of Chlorella cells wereglucose,fructose, galactose, mannose, xylose and arabinose.These sugarswere utilized as respiratory substrates by thealgal cells.
  2. The cellular division of Chlorella was stimulatedby glucoseand galactose, but suppressed by fructose, mannose,xylose andarabinose, while all these sugars evoked gigantism.No correlationwas found between cellular division and gigantism,
  3. The photosynthetic activity of giant Chlorella varied withthesorts of sugars added. It was decreased by glucose, fructoseand mannose, but was unaffected by other sugars such as galactose,xylose and arabinose.
  4. The respiratory activity of giant Chlorellacells as much higherthan that of control cells.
  5. The amountsof protein-N and dry weight per unit volume of giantChlorellawere much less than those of control cells.

Cytoplasm and cell sap of Lamprothamnium succinctum were analyzedseparately for the contents of free amino acids and sucroseto find whether they contribute to turgor regulation. In thevacuole, both amino acids and sucrose were found to be minorcomponents contributing to the generation of osmotic pressure.Their amounts were almost insensitive to changes in externalosmotic pressure. In the cytoplasm, both amino acids and sucrosein the cytoplasm contributed about 20% to the osmotic pressure.Hypotonic treatment did not affect the contents of either, buthypertonic treatment, while not affecting the amino acid contents,caused a significant increase in sucrose content. The methods of quantitative analysis of b-type haem in plantswere investigated. With an improved method developed was determinedthe haem content in the supernatant, mitochondrial, and microsomalfractions of sweet potato tissue. The activities of peroxidase,catalase, and cytochrome oxidase, as well as the contents ofb-type haem and acid-insoluble nitrogen in the cellular fractionswere determined at different incubation times after cuttingof sweet potato tissue. Peroxidase and catalase increased withtime in each celluler fraction, following a short lag phase.In the mitochondrial fraction, b-type haem, cytochrome oxidase,and acid insoluble nitrogen increased linearly with time. Inthe microsomal and supernatant fraction, b-type haem increasedwith time following a short lag phase. The increase in haemcontent of the supernatant fraction appeared to be associatedwith peroxidase formation. Time course analysis showed that 59Fe-incorporation into b-typehaem of the supernatant fraction increased with time and thatincorporation was markedly inhibited by blasticidin S. The incorporationof 59Fe into mitochondrial haem did not increase with time andwas not inhibited by blasticidin S. Blasticidin S inhibited59Fe-incorporation into microsomal haem. Proline accumulation was determined in a facultative halophyte,Mesembryanthemum crystallinum and glycophytes, barley (Hordeumvulgare L.) and wheat (Triticum aestivum L.) Proline accumulationpreceded the shift of CAM in M. crystallinum and did not occurin the continuous darkness. The novel light-dark change of prolinelevel (high in the light and low in the dark) was observed inleaves of all three plants. Proline levels of shoots in barleyand wheat also showed the same light-dark change, suggestingthat proline accumulated in the leaves in the light was nottranslocated to other tissues in the dark period. These resultssuggest that proline has a bifunctional role in the acclimationto high salt stress; an osmoregulant role in the light, anda substrate for dark respiration to supply energy to compartmentationof ions into vacuole in the dark. 1Present address: Kyoto Biological Res. The cortical cytoplasm of the alga Nitella contains reticulateactin that does not survive perfusion fixation with glutaraldehydeunless prestabilized with the cross-linker 3-maleimidobenzoyl-N-hydroxysuccinimidester (MBS). Cytochalasin D remodels thiscortical actin into short rods which are more stable, survivingaldehyde fixation without MBS pre-treatment. The overall alignmentof these actin rods correlates with that of cortical microtubules(transverse in young cells, random in old cells) but probablydoes not involve one-to-one correspondence. The time course,dose dependence and reversibility of these structural changesbroadly resemble those for streaming inhibition by cytochalasinbut the cortical actin responds to concentrations that do notslow streaming. Because the structural changes concern the corticaland not the subcortical actin, they seem unlikely to directlyinhibit streaming. Formation of cortical rods is not a responseto streaming inhibition per se since it does not occur whentwo other inhibitors of streaming (2,4-dinitrophenol (DNP) andNethyl maleimide (NEM)) are used. The electon transport systems of Rhizobium japonicum were studied,comparing cells harvested from effective nodules with thosefrom artificial culture. Participation of the cytochrome systemwas confirmed in both forms of cells. Absorption peaks of thecytochromes of cultured cells were a, b, c type, resemblingthose of Bacillus subtilis, yeast and mammalian tissue. Cytochromea could not be detected in the absorption spectrum of symbioticcells, although the CO binding difference spectrum showed apeak at about 438 mµ, which can be attributed to a componenta3 or a1. CO difference spectrum also showed a shoulder at about416 mµ. Cells cultivated under the insufficient supply of oxygen showedthe cytochrome absorption spectrum closely resembled that ofsymbiotic cells. Diaphorase activity was lower in symbioticcells. These results are considered to be due to the insufficientsupply of oxygen within nodule tissue. Succinate oxidation bythe symbiotic cell paniculate was shown to be carbon monooxideresistant. Effects of Boron Deficiency on the Chemical Composition of a Marine Diatom   总被引:2,自引:0,他引:2  
Cells of Cylindrotheca fusiformis, a marine pennate diatom,multiplied in complete nutrient medium (containing 0?5 mg l–1B) with a generation time of 11 h; in a boron-deficient culturemedium (0?02 mg l–1 B), the generation time was 90 h.The chemical composition of logarithmically growing cells harvestedfrom each of the two culture conditions was compared in orderto assess the effects of boron deficiency on the various chemicalfractions. Under conditions of boron deficiency, the concentrationof protein, carbohydrate, and RNA was depressed below that ofcontrol cells; DNA showed no change, while lipids, phenoliccompounds, and unaccounted-for organic fractions were all increasedin concentration. The greatest effect was on the phenolic fraction.These results are discussed and related to the published literatureon effects of boron deficiency on higher plants.  相似文献
