The use of pulse-amplitude-modulated fluorometry (PAM) and rapid light curves (RLC) were evaluated for monitoring the physiological condition of the seagrass, Thalassia testudinum, at the landscape scale in Florida Bay, USA. PAM fluorometry provides rapid, non-invasive, and quantitative physiological information on the state of photosynthesis. Yet, previous studies of effective and maximum quantum yields have shown that problems arise when expanding measurements from the organismal scale to the landscape scale, mainly due to temporal and irradiance-induced changes in photophysiology. Here, the magnitude of diurnal and spatial variation of photosynthetic characteristics among 10 sample basins and between two sample years was investigated using RLCs. Because RLCs measure effective quantum yields over a range of changing actinic irradiances, we hypothesized that the response parameters might be less sensitive to diurnal light history effects. Our results indicate that the RLC parameters, alpha and ETRmax, significantly changed diurnally, as was previously found for both maximum and effective quantum yields, but the diurnal patterns were variable among the 10 basins. Both among-basin and between-year comparisons were confounded by diurnal variation and statistical analyses comparing morning, mid-day, and afternoon time periods were unable to definitively discern which time of day was best suited for assessing the relative photophysiological status of T. testudinum. However, pooling RLC data at the basin scale revealed among-basin differences and landscape scale trends that were consistent with basin-level morphometric variation in this seagrass. Thus, PAM fluorometry may be useful as a landscape scale monitoring tool within certain constraints. When using this approach over large spatial and temporal scales, diurnal variability must be considered.  相似文献

Turtlegrass, Thalassia testudinum, produces high concentrations of proanthocyanidins (condensed tannins) which we hypothesized are induced by grazing, as a component of a general wound response. To test this we quantified condensed tannins in a variety of turtlegrass tissues following simulated fish grazing, grazing by the urchin Lytechinus variegatus, and treatment with the natural plant wound hormone jasmonic acid. We observed that simulated fish grazing triggered rapid induction of condensed tannins by an average of 10 mg tannin g−1 dry mass (DM) after 5 days. Condensed tannin accumulations were correlated with a reduction of blade extension rates. Further, we observed that constitutive tannin levels in developing first-rank leaves were strongly correlated with the width of second-rank leaves on each shoot, with an increase of 7.7 mg tannin g−1 tissue dry mass per millimeter blade width on average. We propose that wider source leaves provide additional resources for phenolic biosynthesis. There was no induction of tannins in leaves, meristematic or sheath tissues in response to grazing by the urchin L. variegatus, the presence of potential waterborne cues from nearby grazed plants, or to treatment with 5 mM jasmonic acid. However, urchin grazing did induce tannin production in root/rhizome tissues, where they accumulated to levels 3–4 times higher than in blades (up to 350 mg tannin g−1 tissue dry mass). These results confirm the potential for rapid wound-induced condensed tannin accumulations in T. testudinum. The link between blade widths and the tannin content of new leaves indicates that leaf morphology may be a useful bioindicator for predicting herbivore and disease-resistance in the field.  相似文献

Northward expansion of Thalassia testudinum (turtle grass) in Laguna Madre is occurring faster than can be explained by rhizome growth. We hypothesized that seedling establishment can account for the measured rates of meadow expansion and that seedling carbohydrate reserves are utilized until the plant is photosynthetically self-sufficient. To address seedling establishment, we estimated seed output, seedling dispersal and survival. Carbon dynamics were calculated from measurements of biomass allocation, non-structural carbohydrate carbon reserves and photosynthetic parameters in relation to T. testudinum seedling age. Potential seed production calculated for 1996 was consistent with field observations and was estimated at 66±14 seeds m−2 bare area. Fruits can be positively buoyant for up to 10 days, while seeds were generally buoyant for <1 day. Water current measurements, made at about the time of seed release, indicate a positive net transport of 1.5 km d−1 to the north. Seedling survival in laboratory culture after 6 months was 96% compared to 11% in the field after 1 year. The average root:rhizome+seed:leaf ratio changed from 0:11:1 for a 1 week old plant to 1:3:1 for a 15 month old plant. Seedlings used to determine whole plant photosynthesis ranged in age from about 1 week (0.25 months) to 15 months. Gross Pmax increased from 80 to 220 μmol O2 gdw sht−1 h−1, while whole plant respiration decreased from 170 to 60 μmol O2 gdw sht−1 h−1. As the photosynthetic parameters changed, the average non-structural carbohydrate carbon (NSCC) reserves of the seeds decreased from 24 to 3.0 mg NSCC plant−1. Subsequent increases in NSCC were the result of rhizome development. Daily carbon balance, assessed using Hsat periods of 8–18 h d−1, predicts that T. testudinum seedlings become photosynthetically self-sufficient between 2 and 6 months. The unique characteristics of T. testudinum, including seed buoyancy, high seed production and survival rates, coupled with ontogenetic changes in carbon allocation and production imply that sexual reproduction can be important in the long distance dispersal and colonization for this species.  相似文献

Since diurnal chloroplast movements in Halophila stipulacea were described by Drew in 1979, this phenomenon has not been studied further for seagrasses. In addition to an apparent photoprotective role, such movements may affect the measurements of photosynthetic rates based on pulse amplitude modulated (PAM) fluorometry. This is because calculations of electron transport rates (ETR) are directly affected by the light absorption of the leaves (or the so-called absorption factor, AF), the latter of which changes with the movements of the chloroplasts. In this work, we therefore determined chloroplast clumping and dispersal, and measured AFs, chlorophyll contents and PAM fluorescence diurnally for H. stipulacea grown under two irradiance regimes. Diurnal chloroplast clumping occurred in high-light grown (HL) plants (∼450 μmol photons m−2 s−1 during midday), which was accompanied by a decrease in AF values (from 0.56 in the early morning to 0.34 at midday) but not in the chlorophyll content. Also, non-photochemical quenching (measured as NPQ) increased during the day in these plants. No such chloroplast movements and, thus, no diurnal changes in AF values (0.60 ± 0.04 throughout the day), and no changes in NPQ, were found in low-light grown (LL) plants (∼150 μmol photons m−2 s−1 during midday). As a consequence of the chloroplast clumping in HL plants, and its effect on AF values, maximal ETRs did not differ significantly between HL and LL plants. This finding thus shows the importance of taking into account changing AF values along the day when calculating ETRs of H. stipulacea, and other seagrasses potentially featuring diurnally changing AFs, under high-irradiance conditions.  相似文献

Seagrasses are recognized as important plant communities in coastal estuaries and lagoons across both tropical and temperate climes; thus, large-scale seagrass die-off events worldwide are of general concern. In Florida Bay, at the southern terminus of the Florida peninsula, seagrass die-off events up to 4000 ha have been reported and smaller scale mortality events are noted annually. In the present study, we examined several hypothesized causative factors (high temperature, hypersalinity, sulfide toxicity) of seagrass (Thalassia testudinum) mortality in Florida Bay. To test sulfide effects, in situ sulfide production was stimulated by applying a labile carbon source (glucose) to sulfate reducers in the sediment at five sites across the bay (northeastern, northcentral, and southwestern basins). During the one year study, high temperature (32–36 °C) and salinity (> 50 psu) were recorded in the bay associated with a regional drought. We also experienced major seagrass die-off events at two of our southwestern bay sites. These field conditions provided an excellent opportunity to closely examine cause–effect relationships among stressors and die-off events in the field, and verify results of our previous mesocosm experiments. Even though glucose amendments stimulated porewater sulfides in bay sediments (4–8 mmol L− 1), no significant differences in biomass, short shoot density or final growth rates were found between control and glucose plots. In addition, the highest growth rates and shoot densities were concomitant with maximum water column salinity (> 50 psu) and temperature (32–36 °C), when porewater sulfides were also in the millimolar range. Large-scale seagrass mortality events, encompassing  50% of the entire meadow at one site, occurred at southwestern bay sites when plants were down regulating (slower growth and shoot density), probably in response to shorter day length and lower temperature (30–34 to 23–26 °C) from October, 2004 to January, 2005. Sulfate reduction rates (SRR) were also 2-fold higher in the southwestern (214–488 nmol cm− 3 d− 1) versus northcentral and northeastern (97–240 nmol cm− 3 d− 1) bay sites, possibly limited by labile carbon, which we found to stimulate SRR 3-fold in northeastern and northcentral bay sites (461–708 nmol cm− 3 d− 1) and 4-fold at southwestern bay sites (1211–2036 nmol cm− 3 d− 1). Based on a synthesis of the field data reported herein, our mesocosm experiments to date, and contributions by others, we present a conceptual model of seagrass die-off in Florida Bay outlining a cascade of stressors, stimulated by P enrichment, which leads to high O2 consumption in the system triggering a seagrass die-off event.  相似文献

A study of diurnal variation over a 48 hour period was undertaken in July 1973 to ascertain the extent and timing of some major chemical, physical and biological variables in Loch Lomond. The phytoplankton population was dominated by the diatom Tabellaria fenestrata, with a maximum abundance between 04.00 and 06.00 h in surface waters on both days. A distinct diurnal variation in cell numbers was also recorded. Chlorophyll a values also showed a regular pattern of variation with a single peak between 10.00 and 14.00 h each day. Some chemical changes appeared to be a direct consequence of phytoplankton multiplication. Nitrate-nitrogen showed a decrease in concentration coinciding with the period of cell multiplication, whereas dissolved silica concentrations only fell on the completion of this process. Other common diatoms displayed less distinct patterns of variation although Fragilaria crotonensis attained its maximum abundance in surface waters. Considerable variation was recorded in the number of organisms and chlorophyll a levels at 25 cm intervals in the upper metre of the water column, with large variations in standing-crop and chemical parameters in the space of one hour. Diurnal oscillations in the position of the thermocline were recorded, with the thermal discontinuity being at its greatest depth in the early hours of the morning. The hypolimnion and thermocline regions clearly acted as a source of nutrient supply to the epilimnion. 干露胁迫强度决定着海草床在潮间带的分布,植物的生理特性是反映其环境胁迫效应的重要依据。为探索潮汐周期内不同时长的光热干露胁迫对海草生理特性的影响,该文以热带优势海草泰来草(Thalassia hemprichii)为对象,通过在日光温室内模拟高温(41.0~44.5 ℃)和强光(633~1 682 μmol·m-2·s-1)环境条件,研究不同露空时间(0、10、20、40、60、90 min·d-1,连续7 d)对泰来草叶片生长速率、光合色素含量和光合荧光参数,以及对叶片和根部抗氧化酶活性、非结构碳水化合物和次生代谢产物含量的影响。结果表明:(1)≤10 min·d-1的日周期光热干露对泰来草的生长速率和生理特性未产生显著影响(P>0.05)。(2)在光热干露条件下,随露空时间的延长,泰来草叶片损伤逐渐加重,颜色由绿色变为褐色且生长速率降低,光合色素含量、叶绿素a/叶绿素b、叶绿素荧光参数均呈下降趋势; 叶片和根的MDA、可溶性糖含量以及POD、SOD、CAT酶活性呈升高趋势; 叶片淀粉、总酚和总黄酮含量呈先升高后降低的趋势。(3)90 min·d-1的日周期光热干露是泰来草叶绿体PS Ⅱ光合反应中心不完全失活的承受极限。综上认为,潮汐周期内较长时间的光热干露胁迫会对泰来草的生长和生理特性产生负面影响,该研究结果为了解热带海草在潮间带的分布规律以及海草床生态修复时的位置选择提供了依据。  相似文献

The dominant seagrass in Florida Bay, Thalassia testudinum Banks ex König, is a stenohaline species with optimum growth around marine salinity (30-40 PSU). Previous studies have examined the responses of mature short shoots of T. testudinum to environmental stresses. Our goal was to assess responses of seedlings to changes in water chemistry in Florida Bay that might occur as part of the Comprehensive Everglades Restoration Plan (CERP). Specifically, we examined seedling survival, growth, photosynthesis, respiration and osmolality in response to hypo- and hyper-salinity conditions, as well as possible synergistic effects of depleted and elevated ammonium concentrations. The study was conducted in mesocosms on T. testudinum seedlings collected during August 2003 near Florida Bay. Hyper- and hypo-saline conditions were detrimental to the fitness of T. testudinum seedlings. Plants at 0 and 70 PSU exhibited 100% mortality and a significant decrease in survival was observed in the 10, 50 and 60 PSU treatments. Increased levels of ammonium further decreased growth in the lower salinity treatments. Seedlings in 30 and 40 PSU had the greatest growth. Quantum yield and relative electron transport rate, measured using PAM fluorometry, showed a decrease in photosynthetic performance on either side of the 30-40 PSU optimum. Tissue osmolality decreased significantly with decreased salinity but tissue remained consistently hyperosmotic to the media across all salinity treatments. Maintaining negative water potential and allocating more energy to osmoregulation may decrease the productivity of this species in salinity-stress conditions. Our results suggest that the salinity-tolerance limits of this seagrass at the seedling stage are not as broad as those reported for mature plants. Increased fresh water inflow, especially if co-occurring with an increase in water-column ammonium, could negatively affect successful recruitment of T. testudinum seedlings in northern regions of Florida Bay.  相似文献

为了解纳帕海流域高山环境中优势乔木树种光合速率特征及其对环境因子的响应规律,运用LI-6400便携式光合测定仪,分别测定了香格里拉纳帕海流域5种优势乔木树种净光合速率(Pn)、蒸腾速率(Tr)、水分利用效率(WUE)等主要光合生理参数日变化,并对其与环境因子的相关性进行统计分析。结果表明:(1)所测定的5种优势树种中,除川滇高山栎以外,其他4个树种的Pn均呈双峰型;5种树种Pn峰值大小依次为云杉高山松山楂清溪杨川滇高山栎,分别为21.58、21.57、15.21、14.18、11.87μmol·m~(-2)·s~(-1)。(2)树种气孔导度(Gs)和蒸腾速率(Tr)在一天之内都呈现出早晚低、中午高的规律,胞间CO2浓度(Ci)与Pn基本呈相反趋势。(3)树种WUE和LUE日均值与Pn值表现出基本一致的顺序特征,云杉和高山松相对较高,WUE分别为3.27和5.82 mmol·m~(-2)·s~(-1),LUE分别为4.42%和2.99%,川滇高山栎最低,仅为0.92 mmol·m~(-2)·s~(-1)和0.74%。(4)树种Pn对PAR和Gs的变化具有显著的响应特征。该区域树种的Pn最大值高于低海拔区域的同类树种;区域内针叶树种的WUE和LUE显著大于阔叶树种,对高山区域内相对较高的辐射环境具有更高效的适应策略;当地环湖面山上种植的大面积云杉树对区域植被的恢复具有高效的现实意义。  相似文献

Summary The diurnal patterns of twig xylem water potential, net photosynthesis rate, water use efficiency of photosynthesis, and stomatal and mesophyll conductance to CO2 in tamarack, black spruce and swamp birch growing in a natural peatland in central Alberta, Canada, were examined. The relationships of photosynthesis to other ccophysiological parameters were investigated. Data were collected on three days with different weather and soil moisture conditions in the 1988 growing season. Day 1 was clear and warm and the ground water table was 7 cm above the average peat surface. Day 2 was clear and hot. Day 3 was cloudy but warm. On day 2 and day 3, the water tables were in the normal range for that season. Major findings were: 1) Soil flooding depressed photosynthesis in tamarack and black spruce. 2) Swamp birch was better adapted to flooding than tamarack or black spruce. 3) The trees experienced water stress in the afternoons of the two days with lower water table. 4) Changes in photosynthesis of the three species were primarily affected by changes in mesophyll conductance (gm) and the response of photosynthesis to changes in gm was similar for all three species.  相似文献

Seagrass photosynthesis is usually measured in laboratory experiments, following oxygen evolution in closed chambers. Pulse amplitude modulated (PAM) fluorometry constitutes an alternative and non-intrusive method of measuring photosynthesis in the field. Validation of electron transport rate (ETR) measurements as reliable estimators of actual photosynthetic production requires that a significant linear relationship between oxygen production and ETR is demonstrated, and also that a 0.25 molar ratio between these two measures is verified. In this work, both parameters were measured simultaneously in laboratory experiments, over a range of light intensities, under well-defined and controlled conditions. A linear relationship was observed between the average rates of oxygen production and the electron transport rates for Zostera noltii (Hornemann) obtained at several irradiances. The molar ratio found between oxygen production and ETR was 0.15±0.02, lower than the theoretically expected value of 0.25. The use of PAM fluorescence as a valid proxy for photosynthetic production was validated for the range of 35-490 μmol photons m−2 s−1 (PAR), under the assumption that the electron sinks responsible for the molar ratio deviation remain constant in similar experimental conditions.  相似文献

Pulse-amplitude modulated (PAM) fluorometry allows instantaneous estimates of photosynthetic rates, but may well produce variable measurements of photosynthetic activity depending on time of day, recent light history, internal fluctuations, and environmental variability. To investigate this, we compare estimates of diurnal variability in relative photosynthetic performance for the giant kelp, Macrocystis pyrifera (L.) C. Agardh, obtained from PAM fluorometry at three depths during 3 days characterized by different light conditions, and for two different blade ages. Sampling in the mid morning, late morning, early afternoon and late afternoon, we examined diurnal changes in relative photosynthetic performance in meristematic tissue and older blades occurring near the bottom, in the mid water, and at the water surface. Measures of maximum relative electron transport rates (rETRmax), minimum saturating irradiance (Ek), photosynthetic efficiency (α) and maximum quantum yield (Fv/Fm) show that giant kelp blades in the mid water and near the bottom exhibit little to no photosynthetic changes during the day. Near the surface, however, blades exhibit photosynthetic characteristics similar to light-adapted species in that they begin the day acclimated to low light, acclimate to increasing irradiance during the day, and end the day acclimated to low light. Consequently, while estimates of rETRmax were highest during the midday for all sample depths and days, they were also always highest near the surface for both old blades (112.16 ± 8.7, 98.6 ± 14.7, 70.16 ± 5.7) and meristematic tissue (109.0 ± 9.0, 86.9 ± 1.9, 59.2 ± 11.6, surface, mid water and bottom, respectively). Similar patterns were observed for Ek for both old blades (169.2 ± 5.4, 88.0 ± 11.2, 83.8 ± 5.2) and meristematic tissue (138.4 ± 11.5, 96.6 ± 4.69, 68.4 ± 10.6). In contrast, estimates of Fv/Fm were lowest near the surface during the midday for both old blades (0.6 ± 0.02, 0.73 ± 0.69, 0.75 ± 0.01) and meristematic tissue (0.58 ± 0.02, 0.69 ± 0.05, 0.74 ± 0.01, surface, mid water and bottom, respectively). These patterns coincided with similar patterns in ambient light, which was most variable and reached its greatest values near the surface during the midday.  相似文献

Seagrass grazing is an intrinsic disturbance in primarily tropical and subtropical areas. While there is a general parabolic response in seagrass growth to grazing intensity, there is less knowledge on the role of grazing frequency, as well as potential interactions between grazing intensity and frequency. This study experimentally investigated physiological responses in Thalassia hemprichii to simulated (leaf cutting) grazing regimes with different intensities (25% vs. 75%) and frequencies (1 times vs. 3 times) over 35 days in Chwaka Bay (Zanzibar, Tanzania). The results showed that the two high-intensity treatments (75% removal) had 37–41% lower growth rate than the low-intensity/low-frequency treatment, and rhizome sugar and starch content were both affected in a similar way. A 36% lower starch content in the simulated low-intensity/high-frequency regime (25% × 3) compared to the one of low-intensity/low-frequency (25% × 1) also shows an interaction between grazing intensity and frequency. This suggests that high-intensity (and to some extent frequency) grazing regimes, in comparison to low-intensity regimes, could negatively affect T. hemprichii growth, energy reserves, and thereby the ability to deal with additional stress like light limitation or grazing.  相似文献

Diurnal variation in progesterone and LH during the luteal phase and the temporal relationships between oscillations of the two hormones were studied in 10 heifers by collection of blood samples at 0100, 0700, 1300, and 1900 h each day, beginning on Day 1 (Day 0 = ovulation). Concentration of LH on Days 5-9, but not on Days 10-14, was lower (P < 0.05) at 0700 h (0.25 ± 0.02 ng/mL) than at each of the other three hours (combined, 0.32 ± 0.02 ng/mL). An oscillation was defined as an uninterrupted increase and decrease in concentrations. The number of LH oscillations/heifer with the peak at 1900 h (6.1 ± 0.7) throughout the luteal phase was greater (P < 0.01) than for each of the other hours (combined, 4.0 ± 0.2). Diurnal variation in progesterone was not detected. Only statistically defined LH oscillations were used to determine the temporal association between the peak of an LH oscillation and various components of a progesterone oscillation. On Days 5-14, the frequency of the peak of an LH oscillation occurring at the same hour as the peak of a progesterone oscillation (26/48, 54%) was greater (P < 0.0001) than at the progesterone nadir (3/48, 6%). The frequency of the LH peak occurring during increasing (11/34, 32%) and decreasing (8/25, 32%) progesterone concentrations was intermediate (P < 0.05). Results indicated the following: 1) diurnal variation occurred in LH as determined by concentration and by the hour of the peak of an oscillation; and 2) LH oscillations were temporally and positively related to progesterone oscillations.  相似文献

从SPAC理论出发,建立了一个冬小麦光合和蒸散的耦合模型.感热通量和潜热通量采用Shuttleworth-Wallace的双层模型计算,并通过冠层阻力的参数化,将光合作用与蒸腾作用耦合起来.用涡度相关方法,观测了感热通量和潜热通量,对模型进行了验证.结果表明,模拟值与观测值比较一致,模型可以很好地模拟感热通量和潜热通量的日变化过程.对模型的敏感性分析发现,冬小麦蒸腾比较敏感的参数有凋萎点、气孔导度参数、叶对红外辐射的反射率和光响应曲线凸度;土壤蒸发只对土壤阻力参数的敏感性较强.本模型对水热通量与环境因子作用过程的理论研究和指导农田的灌溉制度等有一定的意义.  相似文献

Steady state and dynamic responses of two tree fern species of contrasting origins, Dicksonia antarctica (of Gondwanan origin) and Cyathea australis (Pan-tropical), were studied over two consecutive years under field conditions in a wet sclerophyll forest of south-east Australia. Irrespective of their different origins, there were no significant differences in photosynthetic performance between the two species. Growth irradiance and leaf temperature, but not plant water status, was significantly related to photosynthetic and morphological traits. At a common leaf temperature, maximum light-use efficiency of photosystem II (Fv/Fm) was significantly lower in winter than in summer, suggesting some limitation to PSII efficiency potentially associated with cold winter mornings. Both species displayed seasonal acclimation in a number of measured photosynthetic parameters and frond traits (i.e. Fv/Fm, Asat, gs, NA, total chlorophyll, SLA). Acclimation of stomatal density to spatial variation in growth irradiance seemed limited in both species, although stomatal pattern differed between species. Because there were no significant differences between the two species in photosynthetic parameters, both species can be described by common carbon gain and water use models at the leaf scale.  相似文献

在秋季对粤西乡土香根草的光合等生理特性日动态变化进行了测定.结果表明,香根草的净光合速率(Pn)日变化呈双峰型,蒸腾速率(Tr)和气孔导度(Gs)变化趋势与Pn相似,胞间CO2浓度(Ci)变化方向则与其相反.Pn与光合有效辐射(PAR)、Tr与PAR、Ta和大气相对湿度(RH)的相关程度都达到了极显著水平(P<0.01),Gs则与空气温度(Ta)相关程度达到了显著水平(P<0.05).同一环境因子对Pn、Tr和Gs的影响程度不同,其中PAR是影响香根草光合作用最为强烈的环境因子.Tr是受环境因子控制最为显著的生理参数.  相似文献
