Abscisic acid (ABA) induces formation of a set of proteins in the xerophilic liverwortExormotheca holstii. Some of them have immunological properties similar to the dehydrins of desiccated corn embryos and the desiccation-related proteins ofCraterostigma plantagineum. The fluctuations of endogenous ABA during cycles of desiccation and rehydration seem to be sufficiently high to indicate a role for ABA as a stress hormone and there by as an endogenous inductor of stress-related protein synthesis. Desiccation tolerance disappears when thalli are cultivated for a longer period under well-watered conditions; such thalli are not able to increase stress-dependent ABA biosynthesis sufficiently, or to form the desiccation-related proteins unless they are treated with external ABA. The formation of shoot and root meristems that ultimately give rise to all tissues of the plant body occurs for the first time during embryogenesis. Meristem formation has traditionally been defined in terms of the appearance of histological features of meristems; this approach has led to varying interpretations of the timing of meristem formation relative to other events in embryogenesis. Markers that would provide more objective criteria for the analysis of meristem formation have not been widely available. The maize homeobox gene, knotted1 (kn1), is expressed in shoot meristems throughout postembryonic stages of shoot development. In order to determine whether this gene is expressed in the shoot meristem from its earliest inception, we examined the expression of kn1 in embryos at a series of stages by in situ hybridization to kn1 mRNA and immunolocalization of KN1 protein. Our results show that the onset of kn1 expression is temporally and spatially coincident with the earliest histologically recognizable signs of shoot meristem formation in the embryo, and thus provides a valuable marker for this process. © 1995 Wiley-Liss, Inc.  相似文献

The primary shoot apical meristem of angiosperm plants is formed during embryogenesis. Lateral shoot apical meristems arise postembryonically in the axils of leaves. Recessive mutations at the PINHEAD locus of Arabidopsis interfere with the ability of both the primary shoot apical meristem as well as lateral shoot apical meristems to form. However, adventitious shoot apical meristems can form in pinhead mutant seedlings from the axils of the cotyledons and also from cultred root explants. In this report, the phenotype of pinhead mutants is described, and a hypothesis for the role of the wild-type PINHEAD gene product in shoot meristem initiation is presented. © 1995 Wiley-Liss, Inc.  相似文献

In cell-suspension cultures of Arabidopsis thaliana (L.) Heynh., transfer to auxin-free medium initiates regeneration leading to the formation of numerous rootlets around day 5. This process is promoted by continuous irradiation of the cell cultures with blue light (400–500 nm) while red light (600–700 nm) is ineffective in this respect. During the course of this process, two mRNA species, encoding, respectively, chalcone synthase and a plasmalemma channel protein, transiently accumulate. A second temporary increase in the steady-state level of these mRNAs is correlated with the onset of chloroplast development after 13–17 d of blue-light exposure of the cell cultures. During this cellular differentiation process a number of mRNAs start to accumulate which specify prominent plastid proteins: the small and the large subunits of ribulose-1,5-bisphosphate carboxylase/oxygenase (SSU and LSU), respectively the light-harvesting chlorophyll-a/b protein II (LHCPII). Albumins, globulins and gliadins were extracted from field-sprouted wheat grain var. Beta. The pure fractions were freeze-dried and then added to the residue after protein extraction, or to the ground wheat grain. The adults of Tribolium confusum Duv. and the larvae of T. confusum, Trogoderma granarium Ev. and Anagasta kuehniella Zell. were placed on the prepared food. The residue after protein extraction was inadequate food for complete development of larvae of all species in spite of adding of protein fractions. Larvae did not grow on such food although they remained alive very long time. Adults of T. confusum lived shorter times and laid less eggs on the residue after extraction than on ground wheat.
Zusammenfassung Aus den Körnern der Weizensorte Beta, die auf dem Felde ausgewachsen sind, wurden Albuminen, Globulinen und Gliadinen isoliert. Die reinen Eiweissfraktionen wurden lyophilisiert und zu den vermahlten Körnern oder zu den Extraktionsrückständen zugegeben. Diese auf solche Weisse vorbereitete Nahrung wurde den Käfern von Tribolium confusum und den Larven von T. confusum, Trogoderma granarium und Anagasta kühniella gegeben. Die Extraktionsrückstände erwiesen sich, trotz der Zugabe einzelner Fraktionen, als ungeeignete Nahrung für die Entwicklung der Larven aller drei Arten. Die von diesen Produkten ernährten Larven wurden nicht grösser und verpuppten sich nicht, obwohl sie sehr lange lebten.Die Käfer von Tribolium confusum, die von den Extraktionsrückständen ernährt wurden, lebten kürzer und legten weniger Eier als die, die von den vermahlten Körnern ernährt wurden.

The shoot apical meristem (SAM) is responsible for forming most of the above-ground portion of the plant. We sought to isolate regulatory genes expressed in the Arabidopsis SMA by screening a Brassica oleracea (cauliflower) meristem cDNA library with the homeobox fragment from the maize Knotted-1 (Kn1) gene. We isolated and characterized the corresponding clone, Merihb1, from Arabidopsis. Analysis shows that the predicted MERIHB1 protein exhibits strong homology to KN1 and RS1 from maize, SBH1 from soybean, and KNAT1 and KNAT2 from Arabidopsis. Merihb1 is highly expressed in mRNA from cauliflower meristems and also accumulates in stem and flower mRNA. Based on the similarity of the Merihb1 and Kn1 sequences, expression patterns, and in situ hybridizations, we suggest that Merihb1 represents an Arabidopsis homologue of the maize Kn1 gene.  相似文献

The yeast SKP1 gene and its human homolog p19 skp1 encode a kinetochore protein required for cell cycle progression at both the DNA synthesis and mitosis phases of the cell cycle. In orchids we identified a cDNA (O108) that is expressed in early stages of ovule development and is homologous to the yeast SKP1. Based on the orchid O108 cDNA clone, we identified and characterized an Arabidopsis thaliana (L.) Heynh. cDNA designated ATskp1 that also has high sequence similarity to yeast SKP1. The Arabidopsis ATskp1 is a single-copy gene that mapped to chromosome 1. The expression of the ATskp1 gene was highly correlated with meristem activity in that its mRNA accumulated in all of the plant meristems including the vegetative shoot meristem, inflorescence and floral meristems, root meristem, and in the leaf and floral organ primordia. In addition, ATskp1 was also highly expressed in the dividing cells of the developing embryo, and in other cells that become multinucleate or undergo endoreplication events such as the endosperm free nuclei, the tapetum and the endothelium. Zusammenfassung BeiSalamandra salamandra wurde der Einfluß desRamus palatinus (VII.) und der Lage des Vomer auf die sich während der Metamorphose bildende Vomerspange untersucht. Eine Durchtrennung des Nerven stört das Wachstum der Spange nicht, da der Nerr rasch regeneriert. Nach Durchtrennung und Verlagerung desRamus palatinus bildet sich jedoch keine Vomerspange, ebenso nicht nach Exstirpation eines Teiles des Nerven. Nach Entnahme von Nervenstücken nach Beginn der Metamorphose entwikkeln sich Teilspangen. Das Vorhandensein eines intaktenRamus palatinus ist offenbar Voraussetzung für das Auswachsen der neuen knöchernen Vomerspange. Die Richtung des Spangenwachstums ist nicht von einer bestimmten Lage des Larvenvomer abhängig.

Summary The influence of theramus palatinus (VII.) and the postion of the vomer on the vomerine bar, which develops during metamorphosis, was studied by various amputation procedures. Cutting off the nerve cord does not prevent the development of the vomerine bar, as the nerve soon regenerates, whereas cutting off and displacement as well as the removal of portions of theramus palatinus do prevent development. Removal of portions of the nerve cord during metamorphosis produces only parts of the vomerine bar. Obviously an intact ramus palatinus is necessary for the outgrowth of the novel osseous vomerine bar. The course of this outgrowth is independent of the position of the larval vomer.

Zusammenfassung Während eines zweiwöchigen Aufenthaltes im nordbulgarischen Küstengebiet (und bei Tyrnowo im Jantra-Tal) konnten 111 Arten festgestellt werden, von denen mindestens 91 offenbare Brutvögel sind. Für viele Arten konnten die Kenntnisse über die Verbreitung im Lande erweitert werden; eine Art,Locustella fluviatilis (Wolf), wurde erstmals für Bulgarien nachgewiesen.  相似文献

Two histone H4 cDNA clones were isolated from a tomato (Lycopersicon esculentum Mill.) shoot-tip cDNA library using a heterologous probe from barley (Hordeum vulgare L.). Both cDNAs, which are 81% identical in the coding region, are polyadenylated and belong to a small gene family in the tomato genome. Histone H4 message is abundant in young tissues and rare in older tissues. In the shoot apical meristem, the distribution of H4-expressing cells changes during development. In a juvenile vegetative apex, H4 message is detectable in the central region and the peripheral parts of the meristem. In a mature vegetative apical meristem, H4-expressing cells are localized in the peripheral zone extending into the provascular strands and the rib meristem whereas the central zone is almost devoid of H4 mRNA. After floral transition, H4 mRNA is found throughout the floral meristem, indicating a second change in the pattern of H4 expression. The observed changes in H4 expression are indicative of changes in the distribution of mitotic activity in the shoot apical meristem during plant development. In addition, H4-expressing cells were found to occur frequently in clusters, which may indicate a partial synchronization of cell divisions in the shoot apex.  相似文献

The function of the SHOOT MERISTEMLESS (STM) gene in shoot and floral meristems throughout Arabidopsis development has been analyzed. The results show that STM plays a major role in maintaining shoot and floral meristems. In an allelic series of stm mutants the shoot meristem was either reduced or completely absent in mature embryos and mutant seedling cotyledons showed partial fusion, indicating that the STM gene affects embryonic shoot meristem development and spacing of cotyledons. Postembryonically, stm mutants initiated adventitious shoot development at a position corresponding to the shoot meristem in wild-type. Repetitively initiated defective mutant shoot and floral meristems were consumed during primordia formation and typically terminated prematurely in fused ectopic primordia, indicating that STM is required for continuous shoot and floral meristem function. Analogous defects were observed in stm embryonic and postembryonic development suggesting that similar mechanisms are employed in embryonic and postembryonic organ primordia initiation. Allelic combinations suggest different thresholds for STM requirement during plant development. STM requirement could not be bypassed by standard growth factor regimes or by shoot regeneration from calli. The results suggest that STM functions by preventing incorporation of cells in the meristem center into differentiating organ primordia and that this role can completely account for all defects observed in stm mutants. Mutations in the WUSCHEL (WUS) and ZWILLE (ZLL) genes result in defective organization and premature termination of shoot meristems. Genetic interactions between STM, WUS and ZLL were analyzed and the results indicate that STM acts upstream of WUS and ZLL. Therefore, while STM appears to function in keeping central meristem cells undifferentiated, WUS and ZLL seem to be subsequently required for proper function of these cells.  相似文献

Ultraviolet light induces anthocyanin biosynthesis in cell cultures of an Afghan cultivar of Daucus carota (Daucus carota L. ssp. sativus). Simultaneous treatment with a fungal elicitor from Pythium aphanidermatum results in an inhibition of the catalytic activity of chalcone synthase (CHS), which in turn correlates with an inhibition of anthocyanin biosynthesis. On immunoblots, one isoenzyme (40 kDa) of CHS disappears upon elicitor treatment. On an mRNA level, only the mRNA for the 40-kDa-CHS is active after treatment with ultraviolet light. After inhibition of anthocyanin biosynthesis by the elicitor the enzyme protein disappears and the CHS mRNA is strongly diminished. This inhibition depends on the concentration of the elicitor. In addition, elicitor treatment leads to an induction of the general phenylpropanoid pathway as well as to the accumulation of 4-hydroxybenzoic acid which is covalently bound to wall polysaccharides of the carrot cells. Zusammenfassung Es ist der Beweis erbracht worden, dass die Kolonien vonVespa orientalis L. nicht ohne Larven bestehen können oder genauer gesagt, ohne Speichelabsonderung der Larven. Diese bilden die einzige Quelle von Rohstoffen für den Stickstoffmetabolismus der Arbeiterinnen und für die Königin, da diese selbst über keine Protease verfügen, welche für den Proteinabbau benötigt werden. Von besonderer Wichtigkeit ist dies für die Eiproduktion der Königin. Dieses Ergebnis legt nahe, dass das Gesellschaftsleben der Hornissen wahrscheinlich als eine Symbiose zwischen Erwachsenen und Larven aufzufassen ist.
Summary From the foregoing, it is clear that among the Oriental hornet there is absolute inter-dependence between the larvae and the adults, i.e. a symbiosis in whick the function of the adults is to collect food while that of the larvae is to digest it. At the beginning of the season, when the nector which serves as food for the adults is abundant in nature, the inter-dependence is chiefly with respect to protein digestion. With the advent of summer, the is in Israël and countries of a similar climate, a drop in the natural nectar supply, and the interdependence of the workers and larvae becomes obligatory also with respect to carbohydrates.
