Successive feeding of phenol at concentrations of less than 5.5 mM into a thick suspension of Trichosporon cutaneum WY 2-2 precultured in MPY-medium resulted in a high yield (approximately 28.7 g wet cells/liter) of intact cells capable of decomposing phenol actively (3.7 μmol/min/g of wet cells).

The effects of pH and additions of ethanol and 2-mercaptoethanol were tested on the stability of crude extracts from the strain grown on phenol. The crude extracts were stable at a pH range of 7.6 and 8.3, and were stable for 35 days when 10% ethanol and 5 mM 2-mercaptoethanol were added.

A highly purified preparation of catechol 1,2-oxygenase was obtained from strain WY 2-2 grown on phenol. The purified enzyme was homogeneous on polyacrylamide disc-gel electrophoresis. The enzyme had a molecular weight of about 105,000 and gave rise to subunits of molecular weight of 35,000 by SDS gel electrophoresis. Therefore, the enzyme appears to be a trimer of subunits with identical molecular weight. The Michaelis constants were 9.0 μM for catechol and 6.8 μM for 4-methylcatechol. The enzyme exhibited higher activities towards 4-methylcatechol and hydroxyquinol than towards catechol, and had essentially the same substrate specificity as the crude extracts. 4-Methylcatechol completely inhibited the enzyme activity towards catechol.  相似文献

Disc-electrophoresis of E. coli envelope proteins on SDS acrylamide gels reproducibly revealed up to 50 distinct polypeptide bands. Corresponding molecular weights ranged from 105,000 to 20,000 daltons or less. Major bands corresponded to molecular weights of 73,000, 48,000, 36,000 and 30,000 with the latter constituting up to 20% of the total envelope protein depending upon the method of isolation. Minimum levels of detection using stained gels equaled 0.25 μg protein or 1% of total sample analyzed; for a polypeptide of molecular weight 40,000 daltons this was calculated to be equivalent to 1,200 molecules per cell envelope. In envelopes from a cetB? mutant strain (refractory to colicin E2), an additional band, constituting up to 5% of the total envelope protein was present. The molecular weight of this protein, which was maximally present in wild type envelopes in only trace amounts, is 44,000 daltons, indicating a cellular concentration of approximately 6 × 103 molecules per envelope. This new band was not affected by heating envelope preparations to 100° prior to electrophoresis, but was largely eliminated by washing isolated envelopes in low ionic strength buffer, or by pre-incubating cells with trypsin prior to preparation of envelopes. Treatment of isolated envelopes with Triton X-100, which preferentially releases inner membrane proteins from the envelope (18), resulted in the extraction of a preponderance of the high molecular weight polypeptides, including the 44,000 dalton protein from envelopes of the mutant. The major polypeptides of the envelope and the low molecular weight components were not extracted by Triton X-100. The properties of the 44,000 dalton protein indicated that it is relatively loosely associated with the surface envelope and may be exposed on the external surface of the cytoplasmic membrane. Possible explanations for the appearance of this protein in mutant strains and its relationship to the inability of these to respond, specifically to surface bound colicin E2, will be discussed. Extensive analysis of envelopes from recA? mutants was also carried out and revealed an unusual amount of variation in polypeptide profiles obtained from different preparations. However, no consistent quantitative or qualitative difference between recA and rec+ strains was obtained. In recA, cetB double mutants, the increased level of the 44,000 dalton polypeptide was identical to that found in the rec+, cetB mutant.  相似文献

A latent form, mostly soluble, of polyphenoloxidase of La France pear fruit (Pyrus communis) was purified to homogeneity by ammonium sulfate fractionation and anion-exchange chromatography with DEAE-Sephadex A-25 and then DEAE-Toyopearl 650M, followed by gel permeation chromatography with Toyopearl HW-55s. The addition of 10% glycerol to the eluting buffer was needed for purification. The purified latent enzyme seemed to be a monomeric protein; the molecular weight was estimated to be 70,000 by gel permeation chromatography and 65,000 by SDS–polyacrylamide gel electrophoresis. The enzyme was activated by pressurization at 400 MPa or higher or by treatment with SDS. The highest activity was obtained by pressurization at 600 MPa and 20°C for 6 h.  相似文献

A lectin has been purified from the seeds ofMimosa invisa L. by gel filtration and preparative Polyacrylamide gel electrophoresis. The purified lectin was homogeneous as judged by analytical Polyacrylamide gel electrophoresis, immunodiffusion and Immunoelectrophoresis. The apparent molecular weight is 100,000; the protein is a tetramer with two types of subunits (molecular weight 35,000 and 15,000). The lectin is a glycoprotein with approximately 21% carbohydrate and interacts with Sephadex and concanavalin A-Sepharose. It agglutinates erthrocytes non-specifically, does not agglutinate leucocytes and is not mitogenic, agglutinates Mimosa-nodulatingRhizobium and is a panagglutinin; the agglutination is not inhibited by several simple sugars. Two enzymes containing thiosulfate sulfur transferase activity were purified fromChlorobium vibrioforme f.thiosulfatophilum by ion exchange chromatography, gel filtration and isoelectrofocusing. Enzyme I is a basic protein with an isoelectric point at pH 9.2 and has a molecular weight of 39,000. TheK m-values for thiosulfate and cyanide of the purified basic protein were 0.25 mM (thiosulfate) and 5 mM (cyanide). Enzyme II is an acidic protein. The enzyme has an isoelectric point at pH 4.6–4.7 and a molecular weight of 34,000. TheK m-values of the acidic protein were found to be 5 mM for thiosulfate and 125 mM for cyanide.In addition to thiosulfate sulfur transferase activity, cellfree extracts ofChlorobium vibrioforme f.thiosulfatophilum also contained low thiosulfate oxidase activity and negligible thiosulfate reductase activity. An esterase with excellent stereoselectivity for (+)-trans-ethyl chrysanthemate was purified to homogeneity from Arthrobacter globiformis SC-6-98-28. The purified enzyme hydrolyzed a mixture of ethyl chrysanthemate isomers stereoselectively to produce (+)-trans-acid with 100% stereoisomeric purity. The apparent molecular weight of the purified enzyme was 43,000 on SDS–polyacrylamide gel electrophoresis, and 94,000 on gel filtration chromatography. The optimum conditions for the ester hydrolysis were pH 10.0 at 45°C. The purified esterase hydrolyzed short-chain fatty acid esters, but did not have detectable activity on long-chain water-insoluble fatty acid esters. The enzyme activity was inbibited by diisopropyl fluorophosphate and phenylmethylsulfonyl fluoride.  相似文献

Rabbit serum paraoxonase/arylesterase has been purified to homogeneity by Cibacron Blue-agarose chromatography, gel filtration, DEAE-Trisacryl M chromatography, and preparative SDS gel electrophoresis. Renaturation (Copeland et al., 1982) and activity staining of the enzyme resolved by SDS gel electrophoresis allowed for identification and purification of paraoxonase. Two bands of active enzyme were purified by this procedure (35,000 and 38,000). Enzyme electroeluted from the preparative gels was reanalyzed by analytical SDS gel electrophoresis, and two higher molecular weight bands (43,000 and 48,000) were observed in addition to the original bands. This suggested that repeat electrophoresis resulted in an unfolding or other modification and slower migration of some of the purified protein. The lower mobility bands stained weakly for paraoxonase activity in preparative gels. Bands of each molecular weight species were electroblotted onto PVDF membranes and sequenced. The gas-phase sequence analysis showed that both the active bands and apparent molecular weight bands had identical amino-terminal sequences. Amino acid analysis of the four electrophoretic components from PVDF membranes also indicated compositional similarity. The amino-terminal sequences are typical of the leader sequences of secreted proteins. Human serum paraoxonase was purified by a similar procedure, and ten residues of the amino terminus were sequenced by gas-phase procedures. One amino acid difference between the first ten residues of human and rabbit was observed.  相似文献

The spore coat protein of Clostridium perfringens type A was solubilized from intact spores by treatment with a mixture of sodium dodecyl sulfate (SDS) and dithiothreitol (DTT) at alkaline pH. About 35% of the total dry weight of spores was extracted with this treatment. The extracted protein was partially purified by gel filtration. The major component (Fr-Bl) is rich in glutamic acid and aspartic acid, as well as half-cystine. SDS-polyacrylamide gel electrophoresis analysis of the Fr-Bl showed a major polypeptide band of a molecular weight of 17,000.  相似文献

A pectin-releasing enzyme produced by Kluyveromyces wickerhamii IFO 1675 (PPase-W) was purified to homogeneity from a culture filtrate by cation-exchange and size-exclusion chromatographies. This enzyme had a molecular weight of 35,000 determined by both size exclusion chromatography and ultracentrifugal analysis, and of 40,000 by SDS-PAGE. It contained 2.4% sugar, and its isoelectric point was at pH 5.2. PPase-W catalyzed the release of highly polymerized pectin from various protopectins, and also showed endopolygalacturonase (endo-PGase) activity. The purified enzyme had optimum PGase activity at about pH 5.2 and 50°C and was stable in the range of pH from 4.0 to 7.0 and up to 50°C. The properties of PPase-W were compared with those of PPase-F from Kluyveromyces fragilis IFO 0288, and some differences were found. Also, some preliminary data dealing with the relationship between enzyme activities (PPase and endo-PGase) and protein structure are discussed.  相似文献

Abstract

Atlantic salmon (Salmo salar) serum albumin was purified from plasma and its N‐terminal sequence determined. Atlantic salmon albumin is the predominant plasma protein, negatively charged, at pH 8.6. Albumin was purified to >95% purity which yielded a single band on SDS‐PAGE and agarose gel electrophoresis. The molecular weight of the purified albumin was approximately 6,5 kDa. The N‐terminal sequence of Atlantic chinook salmon albumin was consistent with that predicted from its previously determined cDNA sequence and was identical to that of salmon (Oncorhynchus tshawytscha) albumin through the first 15 residues. However, the fact that the actual N‐terminus was different from that predicted from cDNA sequence indicates that Atlantic salmon albumin, like chinook salmon albumin, lacks a propeptide.  相似文献

A zinc-binding protein was purified to homogeneity for the first time from the gonad parts of scallops, Patinopecten yessoensis, kept in filtered seawater to which no heavy metals were added. Based upon the elution profiles in two chromatographic systems, spectrophotometric analysis, and amino acid composition of the purified preparation, the protein met the criteria for classification as a metallothionein; i.e., low molecular weight (about 9000), paucity of both aromatic amino acid residues and absorbance at 280 nm, and abundance of both cysteinyl residues (> 25%) and absorbance at 215 and 254 nm. Furthermore, the results of chromatographies on a Sephacryl S-300 column and electrophoresis with or without SDS suggested that the protein molecules would be in several polymeric forms in vivo. The antiserum prepared with the purified protein as the antigen was shown to have immunocross-reactivity to neither an extract of the surf clam, Pseudocardium sybillae, nor the whelk, Neptunea arthritica, indicating the heterogeneity of the proteins in marine shellfishes. These results suggested that the Zn-binding protein purified in this study was characteristic of scallops and involved in zinc storage of this organism.  相似文献

The outer membrane proteins of Vibrio vulnificus including isolates from humans, seawater and an asari clam were examined by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. A major outer membrane protein with an apparent molecular weight of 48,000 (48K protein) was common to all the strains grown in 3% NaCl-nutrient broth; however this 48K protein was not produced in any of the strains grown in chemically defined medium. Other major outer membrane proteins with molecular weights ranging from 33,000 to 40,000 varied in number, relative amount and molecular weight depending on the strain. One to three new outer membrane proteins with molecular weights ranging from 74,000 to 85,000 were produced in the cells grown in iron-deficient medium. The 48K protein and one or two major proteins with molecular weights ranging from 35,000 to 37,000 in the cells grown in 3% NaCl-nutrient broth were not solubilized by 2% SDS at 60 C for 30 min and were resistant to trypsin, indicating that they are porins. On the other hand, in cells grown in chemically defined medium, one or two major outer membrane proteins with molecular weights ranging from 33,000 to 40,000 might be porins.  相似文献

Cytokinin binding protein from tobacco leaves was isolated and purified to a single protein by means of affinity chromatography on benzyladenine-linked Sepharose column combined with polyacrylamide gel electrophoresis. In vitro binding of this protein to [14C] benzyladenine was inhibited remarkably by cold benzyladenine and kinetin and slightly by adenine, but not adenosine. The molecular weight of the protein was determined to be about 4,000 daltons by gel filtration and SDS polyacrylamide gel electrophoresis.  相似文献

The psychrotolerant Pseudoalteromonas issachenkonii PAMC 22718 was isolated for its high exo-acting chitinase activity in the Kara Sea, Arctic. An exo-acting chitinase (W-Chi22718) was homogeneously purified from the culture supernatant of PAMC 22718, the molecular weight of which was estimated to be approximately 112?kDa. Due to its β-N-acetylglucosaminidase activity, W-Chi22718 was able to produce N-acetyl-D-glucosamine (GlcNAc) monomers from chitin oligosaccharide substrates. W-Chi22718 displayed chitinase activity from 0 to 37°C (optimal temperature of 30°C) and maintained activity from pH 6.0 to 9.0 (optimal pH of 7.6). W-Chi22718 exhibited a relative activity of 13 and 35% of maximal activity at 0 and 10°C, respectively, which is comparable to the activities of previously characterized, cold-adapted bacterial chitinases. W-Chi22718 activity was enhanced by K+, Ca2+, and Fe2+, but completely inhibited by Cu2+ and SDS. We found that W-Chi22718 can produce much more (GlcNAcs) from colloidal chitin, working together with previously characterized cold-active endochitinase W-Chi21702. Genome sequencing revealed that the corresponding gene (chi22718_IV) was 2,856?bp encoding a 951?amino acid protein with a calculated molecular weight of approximately 102?kDa.  相似文献

An extracellular alkaline serine protease has been purified from Aspergillus terreus (IJIRA 6.2). The purification procedure involved chromatography on DEAE-Sephadex A25, phosphocellulose, hydroxyapatite, casein-Sepharose, gel filtration on Sephacryl-S-300 and by glycerol density gradient centrifugation. The enzyme was further purified to apparent homogeneity through a combination of electrophoresis in polyacrylamide gel containing 0.1% sodium dodecyl sulfate (SDS) with or without protease substrate (gelatin) and subsequent regeneration of its activity in situ by removal of SDS. The active enzyme was visualized in a zymogram or on the basis of protease activity exhibited on an X-ray film. The protein in the unstained segment of the gel was electroeluted. The eluted protein with protease activity exhibited a molecular mass of 37,000-daltons on electrophoresis in SDS-polyacrylamide gel. A sedimentation coefficient of 3.2S was obtained by glycerol density gradient contrifugation. Maximum activity of protease was observed at pH 8.5 and at 37°C. Purified protease was active between pH 5.5 and 9.5 and was found to be stable up to 60°C. With Na-caseinate, the K m of the purified protease was found to be 0.055 mM. Antipain, phenylmethane sulfonyl fluoride, and chymostatin served as non-competitive inhibitors. Substrate specificity was determined by using a synthetic chromogenic peptide containing N-P-Tosyl-Gly-Pro-Arg-p-nitroanilide. —Acetyl-CoA:choline-O-acetyltransferase (ChAc, EC 2.3.1.6) was purified from rat cerebral cortex and its half-life determined. The molecular weight of the enzyme under non-denaturing conditions was estimated by gel filtration to be in the range of 60,000–65,000. On SDS acrylamide gels, the purified enzyme migrated as a single band with a molecular weight estimated as 62,000. The turnover rate of ChAc in the mature rat was determined by the double label method, employing l -[1-14C]leucine and l -[4,5-3H]leucine. Its half-life under steady-state conditions was estimated to be 5.2 days. As a control, tubulin was isolated from the same preparation and its half-life measured. Under these conditions tubulin exhibited a half-life of 3.8 days.  相似文献

An extracellular enzyme that produces di-D-fructofuranose 2′,1;2,1′-dianhydride (difructose anhydride I= DFA I) from inulin was purified from the culture broth of Streptomyces sp. MCI-2524. The purification enhanced the specific activity 7-fold with an overall yield of 17%. The purified enzyme, when electrophoresed on a SDS polyacrylamide gel, gave a single band corresponding to a molecular weight of 36 kDa. Gel filtration chromatography gave a single peak that eluted at a position corresponding to 70 kDa. The enzyme was active from pH 3.0 to pH 9.0, and at temperatures up to 65°C. Maximal activity was observed at pH 6.0, at 55°C. The enzyme was inhibited by Cu2+.  相似文献

The enzyme 3,4-dihydroxyphenylacetate:oxygen 2,3-oxidoreductase (decyclizing) (homoprotocatechuate 2,3-dioxygenase) was purified from the thermophilic organism Bacillus stearothermophilus, grown with j-hydroxyphenylacetic acid as a source of carbon. The enzyme appeared to be homogeneous as judged by disc-gel electrophoresis and sedimentation equilibrium measurements. The average molecular weight determined by three independent procedures was 106,000; the protein was globular and was dissociated in sodium dodecyl sulfate to give a species of molecular weight 33,000 to 35,000. The enzyme was fairly stable on heating and showed maximal activity at about 57 degrees C. An Arrhenius plot of Km for homoprotocatechuate was concave upward, with a break at 32 degrees C; an increase in delta H above this temperature was compensated by lower values of --delta S. Several properties of this enzyme are contrasted with those reported for homoprotocatechuate 2,3-dioxygenase purified by other workers from Pseudomonas ovalis.  相似文献

Phytase from Nocardia sp. MB 36 was purified (9.65-fold) to homogeneity by acetone precipitation, ion exchange, and molecular sieve chromatography. Native polyacrylamide gel electrophoresis (PAGE) and zymogram analysis showed a single active protein in the purified enzyme preparation. Sodium dodecyl sulfate (SDS)-PAGE analysis showed that phytase was a monomeric protein with a molecular weight of approximately 43 kDa. Phytase exhibited activity and stability over a broad pH range (2–8) and elevated temperatures (50–80°C), and utilized several phosphate compounds as substrates. Phytase was extremely resistant to pepsin and trypsin. Various metal ions viz. Fe2+, Co2+, and Mn2+, and NH4+, ethylenediaminetetraacetic acid or EDTA and phenylmethylsulfonyl fluoride or PMSF had no influence on activity, while Ca2+ and Zn2+ enhanced activity by 15 % and 3.58 %, respectively. SDS caused significant reduction in enzyme activity (41.8 %), while 2,3-butanedione did so moderately (15.9 %). Features of Nocardia sp. MB 36 phytase suggest a potential for animal feed applications.  相似文献

Summary The nucleotide sequence of the entire nusB gene of Escherichia coli has recently been determined and the amino acid sequence of its product deduced (Ishii et al. 1984; Swindle et al. 1984). The NusB protein was purified by chromatography on Sephadex G-100, phosphocellulose and hydroxylapatite. Purification of the protein was monitored using 14C-labelled NusB protein, which was synthesized in a maxicell containing an nusB plasmid as a marker. The final product, which was at least 95% pure as judged by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate, had a molecular weight of about 16,000 and an isoelectric point of about 7.3.
