The earliest known diatom fossils with well‐preserved siliceous frustules are from Lower Cretaceous neritic marine deposits in Antarctica. In this study, we analyzed the cell wall structure to establish whether their cell and life cycles were similar to modern forms. At least two filamentous species (Basilicostephanus ornatus and Archepyrgus melosiroides) had girdle band structures that functioned during cell division in a similar way to present day Aulacoseira species. Also, size analyses of cell diameter indicated that the cyclic process of size decline and size restoration used to time modern diatom life cycles was present in five species from the Lower Cretaceous (B. ornatus, A. melosiroides, Gladius antiquus, Ancylopyrgus reticulatus, Kreagra forfex) as well as two species from Upper Cretaceous deposits (Trinacria anissimowii and Eunotogramma fueloepi) from the Southwest Pacific. The results indicate that the “Diatom Sex Clock” was present from an early evolutionary stage. Other ecological adaptations included changes in mantle height and coiling. Overall, the results suggest that at least some of the species in these early assemblages are on a direct ancestral line to modern forms.  相似文献

A new unicellular red alga, Corynoplastis japonica gen. et sp. nov., is described from Tobishima, Japan. Cells are spherical, 18–33 µm in diameter, pale purple to brownish red and surrounded by a mucilaginous sheath. A single chloroplast with many lobes extends from the cell periphery to the cell center. A peripheral thylakoid is present. A pyrenoid occurs at each innermost chloroplast lobe end and one or two thylakoids are present in the pyrenoid matrix. The nucleus is eccentric to peripheral and Golgi bodies are scattered throughout the cell and associated with endoplasmic reticulum. Cells have a slow random gliding motility. The low molecular weight carbohydrate mannitol is present in the cells. Molecular phylogenetic analysis indicates that this alga is closely related to members of the genus Rhodella. A new order, Dixoniellales, is established for Dixoniella, Neorhodella and Glaucosphaera based on molecular and ultrastructural evidence (Golgi bodies associated only with the nucleus). The redefined order Rhodellales in which Rhodella and Corynoplastis are placed is characterized ultrastructurally by Golgi bodies scattered throughout the cytoplasm and associated with endoplasmic reticulum.  相似文献

Regardin cell renewal of planarian epithelial tissue, little is known at present except for the epidermis (Tyler, 1984). According to Skaer (1965), the definitive epidermal cells in planarian embryos originate in the parenchyma where they develop as precursor cells which then migrate into the epidermis. This process of development is also reported in regenerating planarians (Spiegelman & Dudley, 1973; Morita & Best, 1974; Hori, 1978). Recently the validity of Skaer's conclusion has clearly been confirmed by Ishii & Sakurai (1998) using SEM and TEM.The present study deals with the origin and renewal of cells of another epithelium — that lining the pharyngeal cavity of a freshwater planarian Dugesia japonica. This epithelium is known to consist of cells with secretory granduls of fingerprint-like structure (Ishii, 1966; Bowen & Ryder, 1973; Gamo & Garcia-Corrales, 1988; Asai, 1991). The characteristic feature of these specific granules, as well as epitheliosomes (Tyler, 1984) of the epidermis, is very useful as a cell marker for distinguishing cell types and following cell differentiation. Gamo & Garcia-Corrales (1988) claimed the cell populations of the pharyngeal epithelium of Dugesia gonocephala is morphologically and functionally heterogeneous. The sequence of morphological alteration offers the oppurtunity to distinguish at least three functional stages. These represent different adaptive forms of a single cell type, and indicate transformation from one to the next induced by cell renewal. It suggests a beta cell (noeblast) origin of the epithelial cells.In the present study SEM observations revealed theat there are morphologically three distinct cell types which are considered to be juvenile, mature, and senile cells respectively, judging from the differences in cells and the many transitional forms among them (Fig. 1). When observed by TEM, the cytoplasmic patterns of these cell types are correspondingly different from one another, similarly the surface specializations of the plasma membranes of each cell type which facilitate their identification by TEM also vary. Apparently alterations of cell morphology are involved in ageing in a single cell type. Morphological features of each cell type are summarized. Juveniles (R) rarely occur, are small cells provided with prominent apical ruffles and considered to be phagocytic because the cells often contain newly formed phagosomes. Mature cells (F) predominate, are large, polygonal, flat cells, presumably with an intense secretory activity. The cells contain many profiles of granular ER and a small number of specific (secretory) granules. Senile cells (M) are intermittently distributed, irregularly contoured cells with heavy surface microvilli. The specific granules are dense in the apical cytoplasm, and profiles of granular ER decease in number.The precursor epithial cells that are seen in the parenchyma exhibit a considerable degree of differentiation before their translocation into the lining. The primordial cells observed are neoblast-like, with several cohromatoid bodies and many free ribosomes (Morita et al., 1969). These results obtain thus support the evidence and prediction of Gamo & Garcia-Corrales (1988). The observed mode of cell replacement appears to be the same as found in the epidermis (Lentz, 1967).  相似文献

A new species of semi-endophytic coralline alga, Lithophyllum cuneatum (Corallinaceae: Lithophylloideae), is described from Fiji. The species is characterized by a wedge-like thallus that is partially buried in the thallus of the host coralline, Hydrolithon onkodes (Heydrich) Penrose et Woelkerling or occasionally Neogoniolithon sp., and that appears at the surface of the host as a small pustule that is usually paler in color than the host. The thallus consists of erect filaments that are derived from a single cell. The basal cell, when visible, is non-palisade, and areas of bistratose margin are absent. Cells of contiguous erect filaments are joined by secondary pit connections. Epithallial cells are present in 2–3 layers, and individual trichocytes are common. Gametangial plants are dioecious. Male conceptacles have simple spermatangial systems that are confined to the floors of their elliptical chambers. Carposporangial conceptacles contain 5–8 celled gonimoblast filaments that are borne at the margin of a more-or-less discoid fusion cell, and so occupy the periphery of the elliptical conceptacle chambers. Tetrasporangial conceptacles are uniporate, with roofs formed from peripheral filaments, and chambers lack a central columella of sterile filaments. Despite its semi-endophytic nature, haustorial cells are absent, and plastids and pigmentation are present.  相似文献

The ultrastructure, morphology and life cycle of a new chlorarachniophyte alga collected from Okinawa in Japan have been studied. The life cycle of this alga consists of amoeboid, wall‐less round, coccoid and flagellated cells in culture condition; however, the coccoid and flagellate cells are very rare. The pyrenoid ultra‐structure of this alga is the same as that of a previously described species, Lotharella globosa. Since pyrenoid ultrastructure has been adopted as the main criterion for the generic classification of the chlorarachniophytes, the present alga is placed in Lotharella. However, the present alga has a dominant amoeboid cell stage and a reduced walled‐cell stage in the life cycle, while in L. globosa, the walled‐cell stage is dominant and there is no amoeboid cell stage. Therefore the present alga is described as a new species of Lotharella: Lotharella amoeboformis Ishida et Y. Hara sp. nov.  相似文献

Abstract: Rhesus monkeys show a high proliferative T cell response to the bacterial exotoxin SLO without prior immunization. The present study was undertaken to characterize this naturally present SLO-responsiveness with particular emphasis on CD4+ve reactive T cells. It is demonstrated that the frequency of SLO-reactive cells in the circulation ranges between 1 in 75 and 1 in 610 CD4+ve T cells as determined with limiting dilution analysis. It is also shown that induction of a good proliferative response requires Mhc-DR matching between T cell and the antigen presenting cells (APC). Stable and DR-restricted SLO-specific CD4+ve T cell lines were generated from CD8 depleted peripheral blood mononuclear cells (PBMC). The SLO-reactive CD4+ve cell lines are tentatively characterized as Thl-like based on the predominant production of interferon-gamma (IFN-γ) over IL-4, although this seems contradicted by the IL-4 dependent growth of the lines.  相似文献

Summary A series of five discrete virus replicating insect cell lines were isolated from the ovarian and fat body tissues ofHeliothis zea pupae. Two of these cell lines (IPLB-HZ-1075 and-HZ-1079) were studied in depth as to their growth and virus replication responses to specific nutrients (acetyl-β-methylcholine, fresh glutamine) in a number of media. The same two cell lines were identified to species by serological (microimmunodiffusion) and isozyme (phosphoglucoisomerase and peptidase:glycyl-leucine) techniques. Distinguishing comparisons were made with other cell lines that have been confused with the present lines in the literature and with cell line and host pupal extracts from the same and other lepidopteran species studied concurrently in this laboratory. Sterility culture tests were negative for mycoplasmas. The present fiveH. zea lines were the first insect cell lines to replicate polyhedra from a unicapsid multiple embedded nuclear polyhedrosis virus (Baculovirus Group A), in this case the homologous virus obtained from larvae ofH. zea.  相似文献

The cortical development during cell division and the interphase ultrastructure of the marine interstitial hypotrich Certesia quadrinucleata is described using light microscopy and both scanning and transmission electron microscopy. Membranelles are paramembranelles; postciliary microtubules from rightmost membranellar kinetosomes line the buccal cavity and separate parallel arrays of pharyngeal discs that border the cytopharynx. A large paroral membrane is present; an endoral membrane is absent. Alveolar plates lie within alveolar membranes except in regions where organelles and organellar complexes (cirri, the condylopallium, dorsal bristles, membranelles, and the paroral membrane) emerge from the cortex. Muciferous-like bodies attach to the plasma membrane in these regions. Dorsal bristles possess transverse and postciliary microtubules as well as kinetodesmal fiber like those of other hypotrichs. Lasiosomes are present. A unique bulbous structure—the condylopallium—protrudes from the anterior right of the cell. The morphogenetic pattern is euplotine in that cortical development begins in one latitudinal zone, and the oral primordium of the opisthe develops within a subsurface pouch apart from the frontal primordia. Microtubular bundles appear beside (later attached to) developing frontal anlagen; they disappear after cirri are in final interphase locations. Although possessing unique characters, Certesia shares a close phylogenetic relationship with Euplotes.  相似文献

Culex tritaeniorhynchus rhabdovirus (CTRV) is a mosquito virus that establishes persistent infection without any obvious cell death. Therefore, occult infection by CTRV can be present in mosquito cell lines. In this study, it is shown that NIID‐CTR cells, which were derived from Cx. tritaeniorhynchus, are persistently infected with a novel strain of CTRV. Complete genome sequencing of the infecting strain revealed that it is genetically similar but distinct from the previously isolated CTRV strain, excluding the possibility of contamination. These findings raise the importance of further CTRV studies, such as screening of CTRV in other mosquito cell lines.  相似文献

Morphological and ultrastructural changes during the growth of embryogenic cell suspension cultures of Panicum maximum and Pennisetum purpureum have been studied. The suspensions consist almost entirely of cell aggregates of 50–75 embryogenic cells. The cell aggregates vary in size from 90–400 μm in P. maximum and from 70–340 μm in P. purpureum. Following the period of exponential growth starch grains gradually disappear and vacuolation increases. Ten to 16 days after subculture, P. maximum cells enlarge and separate from each other, and organized embryo-like structures appear. Ultrastructural studies show that the cell aggregates are made up of discrete, individual groups of 2–6 cells each. Each cell group appears to arise from a single cell and breaks away from the ‘mother group’ as cell divisions continue. The embryogenic cells are small (20 μm), isodiametric with many starch grains and contain a large nucleus with a prominent nucleolus. Extensive profiles of endoplasmic reticulum, many small peripheral vacuoles, and several amyloplasts are present. Plasmodesmatal connections exist only between cells within a cell group but not between cells of different cell groups in the large cell aggregates.  相似文献

Summary This paper is concerned with physical and chemical changes occurring in the taste buds due to the presence of sapid substances, and not with the nervous mechanism transmitting impulses to the brain. From this standpoint taste phenomena are closely allied with general cell physiology or pharmacology. It has been shown that the great majority of anesthetic or toxic substances have a bitter taste and it is considered that the same structural changes occur in the cell membranes in taste buds as in other cells of the body when such substances are present. From consideration of a large amount of experimental data on the taste of organic compounds it is shown that thesweet and bitter substances are chemically very closely related and there is no line of demarcation between them. In many homologous series there is a continuous (rather than discontinuous) transition from sweet to bitter. Sweet substances having a bitter after-taste are also common.It has been pointed out that it is a general property of anesthetics to be stimulants (i. e. to accelerate cell activities)when in low concentrations. The continuous transition from stimulant to narcotic action is correlated with the sweet-bitter transition. FollowingClowes, Lillie and others the cell membrane is looked upon as a balanced emulsion partly of the water in oil type and partly of the reverse type.The phenomena cited above have been explained in terms of adsorption and surface tension action of substances on this emulsion system and the consequent changes in cell membrane permeability due to changes in the phase ratio of the emulsion types.  相似文献

There are only a few proteins identified at the cell surface that could directly regulate plant cell wall functions. The cell wall-associated kinases (WAKs) of angiosperms physically link the plasma membrane to the carbohydrate matrix and are unique in that they have the potential to directly signal cellular events through their cytoplasmic kinase domain. In Arabidopsis there are five WAKs and each has a cytoplasmic serine/threonine protein kinase domain, spans the plasma membrane, and extends a domain into the cell wall. The WAK extracellular domain is variable among the five isoforms, and collectively the family is expressed in most vegetative tissues. WAK1 and WAK2 are the most ubiquitously and abundantly expressed of the five tandemly arrayed genes, and their messages are present in vegetative meristems, junctions of organ types, and areas of cell expansion. They are also induced by pathogen infection and wounding. Recent experiments demonstrate that antisense WAK expression leads to a reduction in WAK protein levels and the loss of cell expansion. A large amount of WAK is covalently linked to pectin, and most WAK that is bound to pectin is also phosphorylated. In addition, one WAK isoform binds to a secreted glycine-rich protein (GRP). The data support a model where WAK is bound to GRP as a phosphorylated kinase, and also binds to pectin. How WAKs are involved in signaling from the pectin extracellular matrix in coordination with GRPs will be key to our understanding of the cell wall's role in cell growth.  相似文献

Cell wall chemistry in the coencocytic green seaweed Codium vermilara (Olivi) Delle Chiaje (Bryopsidales, Chlorophyta) is well understood. These cell walls are composed of major amounts of neutral β‐(1→4)‐D‐ mannans (Mn), sulfated polysaccharides (SPs), which include pyranosic arabinan sulfates (ArpS), pyruvylated galactan sulfates (pGaS), and mannan sulfates (MnS); also minor amounts of O‐glycoproteins are present. In this study, cell wall samples of C. vermilara were investigated with regard to their monosaccharide composition and infrared spectra (using Fourier transform infrared spectroscopy coupled to principal component [FTIR‐PC] analysis). Samples from three different populations of C. vermilara from the Argentine coast showed: (i) an important variation in the relative arabinan content, which increases from north to south, and (ii) a measurable degree of cell wall variability in the sulfate distribution between the different sulfated polysaccharides, independent of the amount of each polysaccharide present and of total sulfate content. When cell wall composition was analyzed over three consecutive years in a single geographic location, the quantity of Mn and overall sulfate content on SPs remained constant, whereas the pGaS:ArpS molar ratio changed over the time. Besides, similar cell wall composition was found between actively growing and resting zones of the thallus, suggesting that cell wall composition is independent of growth stage and development. Overall, these results suggest that C. vermilara has developed a mechanism to adjust the total level of cell wall sulfation by modulating the ArpS:pGaS:MnS molar ratio and also by adjusting the sulfation level in each type of polymer, whereas nonsulfated Mn, as the main structural polysaccharide, did not change over the time or growing stage.  相似文献

Background  

Embryonic stem cells (ESC) are pluripotent cells obtained from the inner cell mass (ICM) of blastocysts derived from in vitro culture associated with reproductive endocrinology therapy. Human ESCs are regarded as highly significant since they retain the capacity to differentiate into any of approximately 200 unique cell types. Human ESC research is controversial because to acquire such cells, the ICM of human blastocysts must be manipulated in a way that renders embryos nonviable and unsuitable for transfer in utero. Techniques to yield competent ESCs with conservation of source blastocysts would satisfy many objections against ESC research, but at present such approaches remain largely untested.  相似文献

The septal pores in uredial mycelium of Puccinia graminis and P. recondita lack the septal swelling and septal pore cap (dolipore-parenthosome configuration) typically associated with the pores of previously investigated Homobasidiomycetidae and the Tremellales among the Heterobasidiomycetidae. The pores in young hyphae of these two species of Puccinia are characterized by the presence of a cytoplasmic matrix which apparently occludes the pore and acts as a plug, thus preventing the migration of organelles from cell to cell. Large vesicles are typically present at the periphery of the pore matrix and the matrix may be very incompletely bounded by a membrane. Nuclei and other cytoplasmic structures migrate from cell to cell through an opening in the septum lateral to the pore. The available evidence indicates that this peripheral gap in the septum results from a breakdown of a portion of an initially complete septum rather than from incomplete septum formation. In addition to the centripetally formed septa, the hyphae of P. graminis and P. recondita are further compartmentalized by shallow infoldings of the lateral wall and limited unilateral septum formation. There is apparent free passage of cellular material between adjacent compartments.  相似文献

The association of the two sperm cells inBrassica napus pollen following the generative cell division was investigated. The generative cell during division is located in the center of the pollen grain, within the vegetative cell. The space present between the two cells is slightly irregular as seen following standard glutaraldehyde fixation. After completion of mitosis vesicles appear in the equatorial plane, coalescing centripetally to form a cell plate which fuses with the membrane of the generative cell, dividing it in two sperm cells. They are isolated from the vegetative cell by the space between the two cell membranes and are separated from each other by a similar space resulting from the cell plate formed during cytokinesis.  相似文献

Electrophysiological recordings were made from the maxillary sensilla styloconica of fifth instars of the African Armyworm, Spodoptera exempta (Wlk.) (Lep.: Noctuidae), and the borers (Lep.: Pyralidae) Eldana saccharina Wlk. and Maruca testulalis (Geyer), and fifth and sixth instars of the borer Chilo partellus (Swinhoe). Stimuli were different concentrations of 13 carbohydrates and 11 phenolic substances. It appeared that a sucrose sensitive cell is present in the lateral styloconic sensilla of Spodoptera, Maruca and Chilo, and in both the lateral and medial sensilla of Eldana. The sucrose cell of fifth-instar Chilo is more sensitive than that of sixth instars. The additional responses of the medial cells to the phagostimulant sucrose are supposed to have contributed to the widespread infestation of sugarcane by Eldana. The medial sensilla of the Armyworm contain a cell type responding to both meso-inositol and D-ribose. Chilo appeared to be the only one of the three borer species able to perceive phenolic substances. It has a cell responsive to chlorogenic acid in its medial sensilla, which is a bit more sensitive in fifth- than in sixth-instar larvae. The presence of this cell may account for the positive correlation between total phenols content and resistance of sorghum cultivars to this borer. Spodoptera can perceive four phenolic acids. Its lateral sensilla contain a cell responsive to chlorogenic acid and its medial sensilla a cell responsive to 4-hydroxybenzoic acid, gallic acid as well as vanillic acid. Feeding experiments showed that the latter substances inhibit feeding in the Armyworm.So far, in the borers adequate stimuli could be determined for only tow or three of the four taste cells present in each sensillum styloconicum. This indicates that we are still unaware of chemicals that may be important to these caterpillars. On the other hand, for the sensilla styloconica of Spodoptera, electrophysiology suggests the presence of five or six taste cell types, whereas only four taste cells are known from morphological studies.  相似文献
