The mouth, pharynx and oesophagus of Calicotyle are lined by syncytial epithelia, and there are numerous unicellular glands associated with the oesophagus. An infolding of unmodified external tegument lines the mouth cavity and is connected by discrete cytoplasmic processes to subjacent perikarya. It contains two types of secretory body and its luminal surface is invested with a finely filamentous coating. The pharynx and oesophagus are lined by irregularly-folded epithelia that are interconnected by a septate desmosome. Membranous inclusions distinguish the pharynx epithelium and there is a well developed basal lamina for insertion of the pharyngeal muscles. The oesophagus epithelium is perforated by the openings of the oesophageal glands. These lie in the surrounding parenchyma and produce a dense, membrane-bound secretion which is conveyed by duct-like extensions of the glands to the oesophagus lumen. The ducts are supported in places by microtubules and are anchored to the oesophageal epithelium by septate desmosomes. A septate desmosome also marks the junction between the epithelium and the gut caeca.  相似文献

建立一种用蛋白质微阵列法可同时检测血清中艾滋病毒(HIV), 丙型肝炎病毒(HCV)和梅毒螺旋体(TP)抗体的方法.将基因工程HIV、HCV和TP 3种融合抗原共价结合于固相载体玻片上,制成蛋白质微阵列,血清样本经稀释、加样、孵育、洗涤后,加上Cy3荧光标记二抗,洗涤,激光共聚扫描,将获得的图片用Bio-discover公司的Imagene专用分析软件进行分析,所获数据在经过自行编写的软件,根据Cutoff值自动生成判断结果.用此蛋白质微阵列系统检测了400例阴性血清,确定了Cutoff值,检测中国药品生物制品检定研究所的HIV, HCV和TP 3种参比品,并与3种ELISA试剂盒的结果进行了比较.蛋白质微阵列的艾滋病毒阳性和阴性符合率均为100%(20/20); 丙型肝炎病毒阳性和阴性符合率均为95%(38/40).梅毒螺旋体参比品阳性符合率为100%(10/10),阴性符合率为100%(20/20),蛋白质微阵列与三种ELISA试剂检测国家HIV、TP、HCV参比品(共150份血清样本),结果具有高度的符合率.  相似文献

An overview is presented of the formation of organic radical cations in zeolites. Attention is paid firstly to their deliberate production by radiolysis and then to the spontaneous oxidation by activated H-exchanged zeolites of adsorped organic substrates. The nature of the oxidation site arising from thermal pretreatment in oxygen is considered, and possible mechanistic details are outlined for the frequently observed oligomerisations of simple substrates. The broad conclusions are that radical cations are formed from that component of the organic product mixture with the lowest ionisation potential, and that the dominant molecular transformations are driven by proton (Brønsted) catalysis: it appears that there is little evidence that radical cations are involved particularly in heterogeneous catalysis by zeolites, but they may well precede the formation of neutral radicals which are implicated as reaction intermediates. The reorientational dynamics of alkene radical cations in zeolites, as determined by ESR spectroscopy, are also considered.  相似文献

The equivalence of the early mammalian cells, of importance in assisted reproductive technologies (ART), is considered. It is suggested that this controversial topic can be settled by finding whether the cells are distinguished by the Turing-Child (TC) field, as expressed for example by patterns of mitochondrial activity. The division of the pronuclear embryo is driven by a symmetrical bipolar TC pattern whose experimental shape and chemical nature is predicted by TC theory. This bipolar pattern drives the subsequent cell divisions too, and according to present experimental results all cells are equivalent until compaction since they are not distinguished by the TC field in normal development. Interphase cells exhibit homogeneous mitochondrial activity, or perinuclear, or perinuclear and cortical activity, and these patterns too and the rotational symmetry observed are predicted by TC theory. The first differentiation, into an inner mass cell and the trophectoderm, as well as the formation of cell polarity in the trophectoderm are considered. It is suggested that these two events are driven by a peripheral spherical shell of high energy metabolism in the morula; such a shell is predicted by TC theory in a compacted multicellular sphere whose cells are connected by gap junctions. The experimental patterns of mitochondrial activity in unfertilized oocytes exhibit rotational symmetry or polarity. The shape and the chemical nature of these patterns also are predicted and explained by TC theory in a sphere. The change in the spatial pattern of mitochondrial activity with development is attributed to a change in the spatial pattern of mitochondrial activity and not to physical translocation of mitochondria. The experimental finding that these spatial patterns of mitochondrial activity are observed only in live and not in dead biological material is explained by the TC pattern being biology's unique and universal dissipative structure that requires ongoing specific biochemical reactions and energy dissipation.  相似文献

The proposal to implement a phylogenetic nomenclatural system governed by the PhyloCode), in which taxon names are defined by explicit reference to common descent, has met with strong criticism from some proponents of phylogenetic taxonomy (taxonomy based on the principle of common descent in which only clades and species are recognized). We examine these criticisms and find that some of the perceived problems with phylogenetic nomenclature are based on misconceptions, some are equally true of the current rank-based nomenclatural system, and some will be eliminated by implementation of the PhyloCode. Most of the criticisms are related to an overriding concern that, because the meanings of names are associated with phylogenetic pattern which is subject to change, the adoption of phylogenetic nomenclature will lead to increased instability in the content of taxa. This concern is associated with the fact that, despite the widespread adoption of the view that taxa are historical entities that are conceptualized based on ancestry, many taxonomists also conceptualize taxa based on their content. As a result, critics of phylogenetic nomenclature have argued that taxonomists should be free to emend the content of taxa without constraints imposed by nomenclatural decisions. However, in phylogenetic nomenclature the contents of taxa are determined, not by the taxonomist, but by the combination of the phylogenetic definition of the name and a phylogenetic hypothesis. Because the contents of taxa, once their names are defined, can no longer be freely modified by taxonomists, phylogenetic nomenclature is perceived as limiting taxonomic freedom. We argue that the form of taxonomic freedom inherent to phylogenetic nomenclature is appropriate to phylogenetic taxonomy in which taxa are considered historical entities that are discovered through phylogenetic analysis and are not human constructs.  相似文献

Integrins are heterodimeric membrane-spanning adhesion receptors that are essential for a wide range of biological functions. Control of integrin conformational states is required for bidirectional signalling across the membrane. Key components of this control mechanism are the transmembrane and cytoplasmic domains of the α and β subunits. These domains are believed to interact, holding the integrin in the inactive state, while inside-out integrin activation is accompanied by domain separation. Although there are strong indications for domain interactions, the majority of evidence is insufficient to precisely define the interaction interface. The current best model of the complex, derived from computational calculations with experimental restraints, suggests that integrin activation by the cytoplasmic protein talin is accomplished by steric disruption of the α/β interface. Better atomic-level resolution structures of the α/β transmembrane/cytoplasmic domain complex are still required for the resting state integrin to corroborate this. Integrin activation is also controlled by competitive interactions involving the cytoplasmic domains, particularly the β-tails. The concept of the β integrin tail as a focal adhesion interaction ‘hub’ for interactions and regulation is discussed. Current efforts to define the structure and affinity of the various complexes formed by integrin tails, and how these interactions are controlled, e.g. by phosphorylation and localization, are described.  相似文献

This paper develops a model of transport of quantum dot (QD) nanoparticles in membrane nanotubes (MNTs). It is assumed that QDs are transported inside intracellular organelles (called here nanoparticle-loaded vesicles, NLVs) that are propelled by either kinesin or dynein molecular motors while moving on microtubules (MTs). A vesicle may have both types of motors attached to it, but the motors are assumed to work in a cooperative fashion, meaning that at a given time the vesicle is moved by either kinesin or dynein motors. The motors are assumed not to work against each other, when one type of motors is pulling the vesicle, the other type is inactive. From time to time the motors may switch their roles: passive motors can become active motors and vice versa, resulting in the change of the vesicle’s direction of motion. It is further assumed that QDs can escape NLVs and become free QDs, which are then transported by diffusion. Free QDs can be internalized by NLVs. The effects of two possible types of MT orientation in MNTs are investigated: when all MTs have a uniform polarity orientation, with their plus-ends directed toward one of the cells connected by an MNT, and when MTs have a mixed polarity orientation, with half of MTs having their plus-ends directed toward one of the cells and the other half having their plus-ends directed toward the other cell. Computational results are presented for three cases. The first case is when organelles are as likely to be transported by kinesin motors as by dynein motors. The second case is when organelles are more likely to be transported by kinesin motors than by dynein motors, and the third case is when NLVs do not associate with dynein motors at all.  相似文献

Extrinsic allergic alveolitis is caused by the inhalation of small organic allergen particles by non-atopic subjects which provoke an allergic reaction, thought to be chiefly due to a type III mechanism, in the peripheral respiratory tissues. The clinical features are determined by the nature of exposure, the immunopathological mechanism(s) involved and the site of reaction in the lung. When the exposure is intermittent and intensive, febrile episodes with respiratory symptoms beginning after four to six hours are prominent, but when it is more continuous and less intensive they are not and the features are those of a chronic fibrosing lung disease. The diagnosis is important to make because management by the avoidance of exposure is followed by improvement. It is made by recognizing the clinical presentation, by identifying the source of allergen exposure and by obtaining supportive evidence from precipitin and skin tests, or from allergen inhalation tests or lung biopsy.  相似文献

Free peritoneal mast-cells of the rat are stimulated in vitro by molecular oxygen as well as by hydrogen peroxide. Histamine release is also observed in vivo when molecular oxygen or diluted solutions of hydrogen peroxide are injected into the peritoneal cavity of the rat. Inflammatory lesions are produced (vascular congestion, oedema, exudate) which are suppressed by pretreatment with anti-H1 antihistaminics. When hydrogen peroxide solutions are more concentrated, inflammation is also provoked but antihistaminics are no more inhibitory. Mast-cells of the skin and of the lungs are not stimulated neither by molecular oxygen, nor by hydrogen peroxide.  相似文献

Molting processes in crustaceans are regulated by ecdysteroids produced in the molting gland (Y-organ), and molting is indirectly controlled by circulating factors that inhibit the production of these polyhydroxylated steroids. Two of these regulatory factors are the neuropeptides molt-inhibiting hormone (MIH) and crustacean hyperglycemic hormone (CHH). CHH appears to inhibit ecdysteroidogenesis in the Y-organ through the activation of a receptor guanylyl cyclase. The signaling pathway activated by MIH, however, remains a subject of controversy. It is clear that neuropeptides inhibit ecdysteroidogenesis by simultaneously suppressing ecdysteroid biosynthetic processes, protein synthesis, and uptake of high density lipoproteins. Data demonstrate that cAMP is the primary regulator of critical catabolic, anabolic, and transport processes, which ultimately support the capacity for ecdysteroid production by the Y-organ. While cAMP also regulates acute ecdysteroidogenesis to some extent, data indicate that cGMP is the primary signaling molecule responsible for acute inhibition by neuropeptides. It is clear that the regulatory roles filled by cAMP and cGMP are conserved among decapod crustaceans. It is unknown if these complementary second messengers are linked in a single signaling pathway or are components of independent pathways activated by different factors present in extracts of eyestalk ganglia.  相似文献

Photophosphorylation by spinach chloroplasts is inhibited after they have been incubated in the dark with either phenylglyoxal or butanedione. Inhibition by phenylglyoxal is strongest when N-ethylmorpholine is the buffer used during the incubation; that by butanedione requires the presence of borate as buffer. The inhibitions are not reversed by simply washing out the inhibitor, suggesting that a covalent modification of one or more arginine residues is responsible. This is supported by the reversibility of the butanedione inhibition if both the inhibitor and borate buffer are removed. ATPase of the chloroplasts, and of extracted protein, is inhibited, whether activated by trypsin or by heating. This indicates that arginine residues of the coupling factor are the probable major site(s) for attack by these modifiers, leading to the observed inhibitions.  相似文献

We describe octopamine responses of 3 large buccal neurons of Lymnaea and test the hypothesis that these are cAMP-dependent. The B1 neuron is excited by octopamine and the depolarisation is significantly enlarged (P < 0.05) by application of the blocker of cAMP breakdown, 3-isobutyl-1-methylxanthine (IBMX). The B1 neuron is also depolarised by forskolin, an activator of adenylyl cyclase. The B2 and B3 neurons are inhibited by octopamine, and the response is not affected by IBMX. Both cells are excited by forskolin. We conclude that the B1 neuron response to octopamine is likely to be mediated by cAMP, while the B2 and B3 responses are cAMP-independent.  相似文献

A mathematical model of growth and competitive interaction of microorganisms in the chemostat is analyzed. The growth-limiting nutrient is not in a form that can be directly assimilated by the microorganisms, and must first be transformed into an intermediate product by cell-bound extracellular enzymes. General monotone functions, including Michaelis-Menten and sigmoidal response functions, are used to describe nutrient conversion and growth due to consumption of the intermediate product. It is shown that the initial concentration of the species is an important determining factor for survival or washout. When there are two species whose growth is limited by the same nutrient, three different modes of competition are described. Competitive coexistence steady states are shown to be possible in two of them, but they are always unstable. Phosphatidylserine (PS) and phosphatidylethanolamine (PE) are two aminophospholipids whose metabolism is interrelated. Both phospholipids are components of mammalian cell membranes and play important roles in biological processes such as apoptosis and cell signaling. PS is synthesized in mammalian cells by base-exchange reactions in which polar head groups of preexisting phospholipids are replaced by serine. PS synthase activity resides primarily on mitochondria-associated membranes and is encoded by two distinct genes. Studies in mice in which each gene has been individually disrupted are beginning to elucidate the importance of these two synthases for biological functions in intact animals. PE is made in mammalian cells by two completely independent major pathways. In one pathway, PS is converted into PE by the mitochondrial enzyme PS decarboxylase. In addition, PE is made via the CDP-ethanolamine pathway, in which the final reaction occurs on the endoplasmic reticulum and nuclear envelope. The relative importance of these two pathways of PE synthesis has been investigated in knockout mice. Elimination of either pathway is embryonically lethal, despite the normal activity of the other pathway. PE can also be generated from a base-exchange reaction and by the acylation of lyso-PE. Cellular levels of PS and PE are tightly regulated by the implementation of multiple compensatory mechanisms.  相似文献

Advanced glycation end-products (AGEs) are formed from the so-called Amadori products by rearrangement followed by other reactions giving rise to compounds bound irreversibly. The structure of some of them is shown and the mechanism of formation is described. Several AGE binding molecules (Receptors for AGE, RAGE) are known and it is thought that many of the effects caused by AGEs are mediated by RAGE. Some of these were shown to be toxic, and called TAGE. The mechanism of detoxification of glyoxal and methylglyoxal by the glyoxalase system is described and also the possibility to eliminate glycated proteins by deglycation enzymes. The pollination of the orchid Microtis parviflora by ants, insouthern Victoria, is described. The ants are attracted to theinflorescence by a sweet perfume and feed on an abundance ofnectar secreted by the labellum. The pollinarium is depositedon the frons of the ant and is so structured that layers ofpollen are stripped off by contact with a sticky stigma. The activation of DNase I by Mg, Mn, Co, Ni, Fe, Cd, Zn, Ba, Sr, Ca, and Cu ions has been studied by several methods, at different pH and salt concentrations. Mg, Mn, and Co are the best activators for initial stages of degradation. A synergistic effect is shown only by the pair Mg-Ca. Optimal pH of action is always situated at 6.5. DNase II is activated to about the same degree by alkaline earths and Mn ions. Cd and Cu are strong inhibitors. Optimal pH is always 4.6. By titration of liberated secondary phosphate groups, two stages in the hydrolysis of DNA by DNase I are evidenced: a rapid phase activated most by Mg and a slow phase activated by Ca. Some possible mechanisms of action of both enzymes are outlined and the general influence of metal ions is discussed.  相似文献

Data are presented on the type-material representing the species described by Linnaeus in his genera Sphex, Chrysis, Vespa, Apis and Mutilla , The names here considered total 158; six are currently applied in the Hymenoptera Parasitica. Of the balance of 147, three are emendations and five are unavailable homonyms or names proposed in the synonymy of other species. Fifteen Linnaean names are here placed as synonyms. One name is attributed to an author other than Linnaeus, and live names remain species incertae sedis. The remainder, 118, are applied as valid names in die Hymenoptera Aculeata. Holotype specimens in London, Uppsala or Stockholm represent 60 names; 55 names are based on lectotype specimens of which 49 are here designated, two by other authors. One name is represented by a neotype specimen in Lund, and one by a lectotype figure. One non-aculeate species is based on syntypes. Specimens appear to be lost with respect to 25 names; three specimens in Uppsala may be the holotypes of a further three species.
Eight new combinations are made, and twelve new synonyms established; three further new combinations and three further new synonymies are suggested. A systematically arranged summary of species treated and of nomenclatural changes made is given. Nomenclatural changes affecting non-Linnaean names are included where relevant; lectotypes are designated for two non-Linnaean species.  相似文献
